At psychrophilic temperatures (<20 °C), anaerobic digestion produces less methane (CH4). For psychrophilic anaerobic digestion (PAD) to be successful, investigation of cold-adapted microbial consortia involved in methane production is critical. This study aimed to investigate the microbial community driving enhanced methane production from the cold-adaptation process and bioaugmentation of PAD with cold-adapted inoculum (BI). Microbial consortia in cattle manure (CM) and food waste (FW) were adapted and applied during batch PAD of CM and FW to bioaugment methane production at 15 °C. Cold adaptation and PAD with BI resulted in cumulative specific methane yields of 0.874 ± 0.231 and 0.552 ± 0.089 L CH4 g−1 volatile solids, respectively, after 14 weeks, while the absence of BI (control) led to acidification and no methane production during PAD. Following 16S rRNA V4–V5 amplicon sequencing and metagenomic analyses, Methanosarcina was revealed as a key driver of methanogenesis during cold adaptation and PAD bioaugmentation. Furthermore, based on the predictive functional and metabolic analysis of the communities, possible synergies were proposed in terms of substrate production and utilization by the dominant microbial groups. For instance, during methane production, Bacteroides and Methanobrevibacter were possibly involved in a syntrophic relationship, which promoted methanogenesis by Methanosarcina. These findings provide insight into the prospective microbial synergies that can be harnessed and/or regulated in cold-adapted inoculum for the improvement of methane production during PAD. Classical theories predict that relatively constant environments should generally favour specialists, while fluctuating environments should be selected for generalists. However, theoretical and empirical results have pointed out that generalist organisms might, on the contrary, perform poorly under fluctuations. In particular, if generalism is underlaid by phenotypic plasticity, performance of generalists should be modulated by the temporal characteristics of environmental fluctuations. Here, we used experiments in microcosms of Tetrahymena thermophila ciliates and a mathematical model to test whether the period or autocorrelation of thermal fluctuations mediate links between the level of generalism and the performance of organisms under fluctuations. In the experiment, thermal fluctuations consistently impeded performance compared with constant conditions. However, the intensity of this effect depended on the level of generalism: while the more specialist strains performed better under fast or negatively autocorrelated fluctuations, plastic generalists performed better under slow or positively autocorrelated fluctuations. Our model suggests that these effects of fluctuations on organisms' performance may result from a time delay in the expression of plasticity, restricting its benefits to slow enough fluctuations. This study points out the need to further investigate the temporal dynamics of phenotypic plasticity to better predict its fitness consequences under environmental fluctuations. To investigate how and to what extent there are differences in the photosynthetic plasticity of trees in response to different light environments, six species from three successional groups (late successional, mid‐successional, and pioneers) were exposed to three different light environments [deep shade – DS (5% full sunlight – FS), moderate shade – MS (35% FS) and full sunlight – FS].Maximum net photosynthesis (Amax), leaf N partitioning, stomatal, mesophile, and biochemical limitations (SL, ML, and BL, respectively), carboxylation velocity (Vcmax), and electron transport (Jmax) rates, and the state of photosynthetic induction (IS) were evaluated.Higher values of Amax, Vcmax, and Jmax in FS were observed for pioneer species, which invested the largest amount of leaf N in Rubisco. The lower IS for pioneer species reveals its reduced ability to take advantage of sunflecks. In general, the main photosynthetic limitations are diffusive, with SL and ML having equal importance under FS, and ML decreasing along with irradiance. The leaf traits, which are more determinant of the photosynthetic process, respond independently in relation to the successional group, especially with low light availability.An effective partitioning of leaf N between photosynthetic and structural components played a crucial role in the acclimation process and determined the increase or decrease of photosynthesis in response to the light conditions. The life history, distribution and diversity of fishes are largely influenced by environmental salinity. Changes in salinity affect a range of physiological processes including metabolism, nutrition, reproduction and growth. Therefore, fish can be conditioned to environmental parameters most suitable for production, where distinct traits are optimised through species‐specific manipulation of salinities. The primary purpose of this review is to summarise the existing literature on the salinity tolerance of aquacultured fish. The various experimental approaches for determining salinity tolerance are compared, along with summarised information for key species employed in aquaculture, including their native distributions, life history stage and optimal salinity for survival and growth. Plant transcriptomes are complex entities shaped spatially and temporally by a multitude of stressors. The aim of this review was to summarize the most relevant transcriptomic responses to selected abiotic (UV radiation, chemical compounds, drought, suboptimal temperature) and biotic (bacteria, fungi, viruses, viroids) stress conditions in a variety of plant species, including model species, crops, and medicinal plants. Selected basic and applicative studies employing RNA-seq from various sequencing platforms and single-cell RNA-seq were involved. The transcriptomic responsiveness of various plant species and the diversity of affected gene families were discussed. Under stress acclimation, plant transcriptomes respond particularly dynamically. Stress response involved both distinct, but also similar gene families, depending on the species, tissue, and the quality and dosage of the stressor. We also noted the over-representation of transcriptomic data for some plant organs. Studies on plant transcriptomes allow for a better understanding of response strategies to environmental conditions. Functional analyses reveal the multitude of stress-affected genes as well as acclimatory mechanisms and suggest metabolome diversity, particularly among medicinal species. Extensive characterization of transcriptomic responses to stress would result in the development of new cultivars that would cope with stress more efficiently. These actions would include modern methodological tools, including advanced genetic engineering, as well as gene editing, especially for the expression of selected stress proteins in planta and for metabolic modifications that allow more efficient synthesis of secondary metabolites.

IntroductionUpside-down jellyfish (Cassiopea sp.) are highly tolerant to multiple abiotic stressors, including fluctuating temperatures associated with shallow marine habitats. This resilience may underlie the ability of Cassiopea sp. to inhabit a wide variety of tropical habitats across the globe. Additionally, Cassiopea sp. are marked by a conspicuous array of appendage coloration; individual medusae vary in the hue and number of oral appendages, which are often strikingly blue. The function of this coloration is not understood. We aimed to understand how extrinsic (temperature, location) and intrinsic (host color) factors may shape thermal tolerance.MethodsAdult Cassiopea xamachana were collected from two sites that vary in daily temperature range within the Florida Keys and were subjected to acute lethal heat stress. To quantify a whole-organism response to heat, we measured changes in bell pulsation, which likely plays a role in feeding, oxygen exchange, and symbiont uptake. Finally, color morphs were acclimated at either ambient (26°C) or elevated (33°C) temperatures.ResultsC. xamachana from two locations that vary in thermal range do not exhibit different responses to heat, suggesting that temperature fluctuations do not prime individuals for higher thermal tolerance. Additionally, C. xamachana with blue appendages survived significantly higher temperatures and exhibited less change in bell pulsation rates compared to non-blue individuals. We found that acclimation at 33°C, as well as appendage color in each treatment, led to higher survival under acute heat stress.DiscussionThese findings highlight the importance of temperature and coloration in Cassiopea xamachana resilience during heat stress.

Salty-alkaline waters are widely distributed globally, and how to effectively develop saline-alkaline water to improve the utilization rate of water resources has become a global challenge. This study examined the survival performance of juvenile silver crucian carp (Carassius gibelio) under alkaline gradient acclimation conditions and explored the regulatory mechanisms for their adaptation to high alkalinity. Compared to directly immersing in high concentrations, alkaline gradient acclimation significantly improved the survival rate of juvenile silver crucian carp. Long-term high alkaline stress caused significant reduction in the growth performance of juvenile silver crucian carp. After alkaline stress, the enzyme activities of Na+-K+-ATP (NKA) and Ca2+-Mg2+-ATP (CMA) in the gills were significantly decreased. Meanwhile, histopathology of the gills showed structural changes, indicating that the physiological functions of juvenile silver crucian carp were impaired under alkaline conditions. Transcriptomic analysis found that the expression of genes related to metabolism and immune response pathways in the gill and kidney underwent significant changes. Genes related to carbohydrate metabolism were upregulated while those related to protein metabolism were downregulated, indicating that alkaline stress caused the organism to utilize carbohydrates rather than proteins as much as possible to meet enhanced metabolism. The HIF pathway that maintains the body's survival was significantly up-regulated, at the expense of high energy consumption. Dysregulation of genes related to the formation of neutrophil extracellular traps (NETosis) pathway indicated that the immune function of the fish was suppressed. Overall, this study revealed the physiological and molecular responses of juvenile silver crucian carp to alkaline stress, deepening our understanding of their adaptation strategies and the potential impact of alkaline environments on aquatic life.

Climate change is predicted to increase atmospheric vapor pressure deficit, exacerbating soil drought, and thus enhancing tree evaporative demand and mortality. Yet, few studies have addressed the longer-term drought acclimation strategy of trees, particularly the importance of morphological versus hydraulic plasticity. Using a long-term (20 years) irrigation experiment in a natural forest, we investigated the acclimation of Scots pine (Pinus sylvestris) morpho-anatomical traits (stomatal anatomy and crown density) and hydraulic traits (leaf water potential, vulnerability to cavitation (Ψ50), specific hydraulic conductivity (K s), and tree water deficit) to prolonged changes in soil moisture. We found that low water availability reduced twig water potential and increased tree water deficit during the growing season. Still, the trees showed limited adjustments in most branch-level hydraulic traits (Ψ50 and K s) and needle anatomy. In contrast, trees acclimated to prolonged irrigation by increasing their crown density and hence the canopy water demand. This study demonstrates that despite substantial canopy adjustments, P. sylvestris may be vulnerable to extreme droughts because of limited adjustment potential in its hydraulic system. In the Mediterranean region, a reduction of annual precipitation and a longer and drier summer season are expected with climate change by the end of the century, eventually endangering forest survival. To cope with such rapid changes, trees may modulate their morpho-anatomical and physiological traits. In the present study, we focused on the variation in leaf gas exchange and different leaf morpho-anatomical functional traits of Quercus pubescens Willd. in summer using a long-term drought experiment in natura consisting of a dynamic rainfall exclusion system where trees have been submitted to amplified drought (AD) (~−30% of annual precipitation) since April 2012 and compared them with trees under natural drought (ND) in a Mediterranean forest. During the study, we analyzed net CO2 assimilation (An), stomatal conductance (gs), transpiration (E), water-use efficiency (WUE), stomatal size and density, density of glandular trichomes and non-glandular trichomes, thickness of the different leaf tissues, specific leaf area and leaf surface. Under AD, tree functioning was slightly impacted, since only An exhibited a 49% drop, while gs , E and WUE remained stable. The decrease in An under AD was regulated by concomitant lower stomatal density and reduced leaf thickness. Trees under AD also featured leaves with a higher non-glandular trichome density and a lower glandular trichome density compared with ND, which simultaneously limits transpiration and production costs. This study points out that Q. pubescens exhibits adjustments of leaf morpho-anatomical traits which can help trees to acclimate to AD scenarios as those expected in the future in the Mediterranean region. Summary: A specific, robust threshold for drought‐induced tree mortality is needed to improve the prediction of forest dieback. Here, we tested the relevance of continuous measurements of stem diameter variations for identifying such a threshold, their relationship with hydraulic and cellular damage mechanisms, and the influence of growth conditions on these relationships.Poplar saplings were grown under well‐watered, water‐limited, or light‐limited conditions and then submitted to a drought followed by rewatering. Stem diameter was continuously measured to investigate two parameters: the percentage loss of diameter (PLD) and the percentage of diameter recovery (DR) following rewatering. Water potentials, stomatal conductance, embolism, and electrolyte leakage were also measured, and light microscopy allowed investigating cell collapse induced by drought.The water release observed through loss of diameter occurred throughout the drought, regardless of growth conditions. Poplars did not recover from drought when PLD reached a threshold and this differed according to growth conditions but remained linked to cell resistance to damage and collapse.Our findings shed new light on the mechanisms of drought‐induced tree mortality and indicate that PLD could be a relevant indicator of drought‐induced tree mortality, regardless of the growth conditions. Simple Summary: Antarctic marine invertebrates live in the constant of the Southern Ocean and are characterised by sensitivity to small increases in temperature. We conducted a series of aquarium experiments that tested this ability and found species-specific responses to warming. We found that some species were able to survive for many months at up to 10 °C, a temperature which is up to 4 °C warmer than previously recorded. We found that the survivors of three species had adjusted their biological systems (acclimated) and were better able to survive additional rapid warming, but one anemone species did not elevate its upper temperature limit, even though it survived for 270 days at 6 °C. There were also species-specific effects of increasing oxygen concentration on long-term survival to elevated temperatures, with extended, no change, or reduced survival duration all found in different species. Thermal sensitivity is clearly the product of multiple ecological and physiological capacities, and this diversity of response needs further investigation and interpretation to improve our ability to predict future patterns of biodiversity. Antarctic marine ectotherms live in the constant cold and are characterised by limited resilience to elevated temperature. Here we tested three of the central paradigms underlying this resilience. Firstly, we assessed the ability of eight species, from seven classes representing a range of functional groups, to survive, for 100 to 303 days, at temperatures 0 to 4 °C above previously calculated long-term temperature limits. Survivors were then tested for acclimation responses to acute warming and acclimatisation, in the field, was tested in the seastar Odontaster validus collected in different years, seasons and locations within Antarctica. Finally, we tested the importance of oxygen limitation in controlling upper thermal limits. We found that four of 11 species studied were able to survive for more than 245 days (245–303 days) at higher than previously recorded temperatures, between 6 and 10 °C. Only survivors of the anemone Urticinopsis antarctica did not acclimate CTmax and there was no evidence of acclimatisation in O. validus. We found species-specific effects of mild hyperoxia (30% oxygen) on survival duration, which was extended (two species), not changed (four species) or reduced (one species), re-enforcing that oxygen limitation is not universal in dictating thermal survival thresholds. Thermal sensitivity is clearly the product of multiple ecological and physiological capacities, and this diversity of response needs further investigation and interpretation to improve our ability to predict future patterns of biodiversity. In this review, we discuss some of the recent advances in our understanding of the physiology of the air pollution and exercise. The key areas covered include the effect of exercise intensity, the effects of pre‐exposure to air pollution, acclimation to air pollution, and the utility of masks during exercise. Although higher intensity exercise leads to an increase in the inhaled dose of pollutants for a given distance traveled, the acute effects of (diesel exhaust) air pollution do not appear to be more pronounced. Second, exposure to air pollution outside of exercise bouts seems to have an effect on exercise response, although little research has examined this relationship. Third, humans appear to have an ability to acclimate to ground level ozone, but not other pollutants. And finally, masks may have beneficial effects on certain outcomes at low intensity exercise in pollution with significant levels of particles, but more study is required in realistic conditions. In northern temperate zones, peach trees are vulnerable to cold temperature injury in the fall, particularly as climate change prolongs warm weather in the fall and potentially delays the onset of cold acclimation. Four experiments evaluated how cold acclimation of flower buds and shoot phloem, cambium, and xylem is affected by exposure to varying temperatures in the fall. One-year-old peach shoots from trees grown in Maine, USA, were collected from October through November, exposed to 1, 3, or 6 days of low, high, and freezing temperatures, and subjected to stepwise controlled freezing to about −30 °C. Injury was visually quantified as oxidative browning of flower buds and shoot tissues. High temperature exposure, even of a single day, decreased cold tolerance of flower buds and shoot tissues until late November, when high temperatures only minimally decreased cold hardiness. In mid-November, increasing the duration of high temperature exposure from 1 to 3 days decreased cambium and phloem hardiness, but hardiness in flower buds was not further decreased by the longer duration of 3 days. By late November, hardiness in flower buds, cambium, and phloem was less responsive to high temperature, and was increased by prior exposure to 6 days of freezing. After high temperature, xylem lost hardiness to a small degree in mid-October and late November, but in mid-November this occurred in only one experiment. SUMMARY: The development of photosynthetically competent seedlings requires both light and retrograde biogenic signaling pathways. The transcription factor GLK1 functions at the interface between these pathways and receives input from the biogenic signal integrator GUN1. BBX14 was previously identified, together with GLK1, in a core module that mediates the response to high light (HL) levels and biogenic signals, which was studied by using inhibitors of chloroplast development. Our chromatin immunoprecipitation‐Seq experiments revealed that BBX14 is a direct target of GLK1, and RNA‐Seq analysis suggests that BBX14 may function as a regulator of the circadian clock. In addition, BBX14 plays a role in chlorophyll biosynthesis during early onset of light. Knockout of BBX14 results in a long hypocotyl phenotype dependent on a retrograde signal. Furthermore, the expression of BBX14 and BBX15 during biogenic signaling requires GUN1. Investigation of the role of BBX14 and BBX15 in GUN‐type biogenic (gun) signaling showed that the overexpression of BBX14 or BBX15 caused de‐repression of CA1 mRNA levels, when seedlings were grown on norflurazon. Notably, transcripts of the LHCB1.2 marker are not de‐repressed. Furthermore, BBX14 is required to acclimate plants to HL stress. We propose that BBX14 is an integrator of biogenic signals and that BBX14 is a nuclear target of retrograde signals downstream of the GUN1/GLK1 module. However, we do not classify BBX14 or BBX15 overexpressors as gun mutants based on a critical evaluation of our results and those reported in the literature. Finally, we discuss a classification system necessary for the declaration of new gun mutants. Significance Statement: A picture is emerging in which BBX proteins have multifaceted functions in plant development, acting as receivers of chloroplast signals as well as playing a role in responses to acute stress. Thermal acclimation is often considered critical in organismal responses to novel thermal conditions. Our understanding of the physiological implications of acclimation is largely derived from lab studies that either manipulate daytime basking availability or use of constant thermal regimes. In contrast, the importance of the nocturnal thermal environment (e.g. the extent of thermal respite) is often overlooked yet could play a vital role in thermal acclimation in the wild.To fill this gap, we acclimated lizards (Amphibolurus muricatus) under three thermal regimes (Hot Constant, Cold Constant and Alternating) and compared their physiological responses (Metabolic Rate, Sprint Speed, Thermal Preferences and Thermal Limits).We found that animals maintained constantly at hot temperatures (preferred temperature, 35°C) gained sprint performance increases and exhibited shifts in thermal optima not seen in those maintained constantly at cold temperatures (20°C), yet suffered costs to growth (in smaller animals) and maintenance (mass loss in larger animals). Animals maintained at alternating temperatures (12 h 20°C; 12 h 35°C) had performance benefits similar to animals in the hot treatment, without experiencing reductions in juvenile growth and adult mass.Animals acclimated under hot temperatures showed a significant lower preferred and voluntary maximum temperatures compared to animals acclimated under a cold temperature regime.We found no impact of acclimation treatment on behavioural thermal limits or standard metabolic rate.Overall, we show that alternating between access to preferred temperatures and having periods of energetic rest confer the greatest benefits for our animals. These results highlight the importance of natural body temperature variation for enhancing overall ectotherm performance and physiology, and the costs of novel thermal environments that fail to provide this variation. Large-scale afforestation on the Loess Plateau has been subjected to more frequent and severe drought due to climate change. However, there is a lack of definite evidence elucidating the adjustments in water supply-demand relationships under prolonged drought. In this study, we investigated the responses of plant characteristics related to plant water supply and demand in 2-year-old Robinia pseudoacacia seedlings to soil water availability (SWA). We systemically analyzed the acclimation of plant water use to prolonged drought. A logistic function was used to fit relationships between normalized net photosynthetic rate (P n ), transpiration rate (T r ), stomatal conductance (G s ), soil-root hydraulic conductance (K sr ), root-leaf hydraulic conductance (K rl ), and whole plant hydraulic conductance (K plant ) with SWA. The results revealed significant difference in responses of these parameters to SWA (p  < .001). The sensitivities of P n , T r , and G s to drought increased from 60-day to 120-day drought, contributing to reduce water consumption. Meanwhile, the relative stable sensitivities of hydraulic conductance (K sr , K sr , and K plant ) promoted a consistent water supply at different ecological levels. Additionally, coordination between G s with hydraulic conductance helped maintain normal physiological activities under drought. These findings enhance our understanding of adjustments in plant water use in response to prolonged drought in Robinia pseudoacacia. Study Implications: Our findings have implications for better understanding the acclimation of Robinia pseudoacacia to prolonged drought. First, our results suggest decreased water demand (water consumption) by Robinia pseudoacacia under prolonged drought because the sensitivities of net photosynthetic rate (P n ), transpiration rate (T r ), and stomatal conductance (G s ) to drought increased from 60-day to 120-day drought treatments. Second, our study indicates a relative stable water supply (soil-root, root-leaf, and whole plant hydraulic conductance; K sr , K rl , and K plant ) of Robinia pseudoacacia at different ecological levels under prolonged drought. Critical thermal maximum (CTmax) is widely used to measure upper thermal tolerance in fish but is rarely examined in embryos. Upper thermal limits generally depend on an individual's thermal history, which molds plasticity. We examined how thermal acclimation affects thermal tolerance of brook trout (Salvelinus fontinalis) embryos using a novel method to assess CTmax in embryos incubated under three thermal regimes. Warm acclimation was associated with an increase in embryonic upper thermal tolerance. However, CTmax variability was markedly higher than is typical for juvenile or adult salmonids. Red macroalga Pyropia yezoensis is a high valuable cultivated marine crop. Its acclimation to cold stress is especially important for long cultivation period across winter in coasts of warm temperate zone in East Asia. In this study, the response of P. yezoensis thalli to low temperature was analyzed on physiology and transcriptome level, to explore its acclimation mechanism to cold stress. The results showed that the practical photosynthesis activity (indicated by FPSII and qP) was depressed and pigment allophycocyanin content was decreased during the cold stress of 48 h. However, the Fv/Fm and non-photochemical quenching increased significantly after 24 h, and the average growth rate of thalli also rebounded from 24 to 48 h, indicating a certain extent of acclimation to cold stress. On transcriptionally, the low temperature promoted the expression of differentially expressed genes (DEGs) related to carbohydrate metabolism and energy metabolism, while genes related to photosynthetic system were depressed. The increased expression of DEGs involved in ribosomal biogenesis and lipid metabolism which could accelerate protein synthesis and enhance the degree of fatty acid unsaturation, might help P. yezoensis thallus cells to cope with cold stress. Further co-expression network analysis revealed differential expression trends along with stress time, and corresponding hub genes play important roles in the systemic acquired acclimation to cold stress. This study provides basic mechanisms of P. yezoensis acclimation to cold temperature and may aid in exploration of functional genes for genetic breeding of economic macroalgae. Asparagus (Asparagus officinalis L.) is an important vegetable crop in southern Ontario, Canada, where winter air and soil temperatures below 0°C are common. Consequently, cultivars growing in this area must possess winterhardiness and freezing tolerance for survival. Asparagus acquires freezing tolerance in the fall through cold acclimation and loses freezing tolerance in the spring through deacclimation. To understand the molecular bases of these processes, transcriptomic analysis (RNA-Seq) was conducted on two cultivars, one adapted, ‘Guelph Millennium’ (GM), and one unadapted, ‘UC157’ (UC), to the winter conditions of southern Ontario. RNA extracted from bud and rhizome tissues, sampled on three dates during early spring and late fall, was subjected to sequencing. In the fall, the numbers of differentially expressed (DE) genes at the second and third harvests increased, relative to the first harvest, in dormant buds and rhizomes as freezing tolerance of cultivars increased, and the majority of DE genes were downregulated. In spring, freezing tolerance decreased as plants deacclimated and most genes DE at second and third harvests were upregulated in both cultivars. GM had lower LT50 (lethal temperature at which 50% of plants die) values and hence higher freezing tolerance than UC on specific sampling dates during both spring and fall, and expression patterns of specific genes were correlated with LT50 differences. Functional analysis revealed that these genes were involved in carbohydrate metabolic process, plant hormone signal transduction (auxin and gibberellin), proline metabolism, biosynthesis of secondary metabolites, circadian rhythm, and late embryogenesis abundant proteins and could be associated with cold acclimation and deacclimation processes. These findings will help researchers understand the molecular mechanisms of freezing tolerance in asparagus, leading to breeding and genetic strategies to improve the trait.

Abstract The persistence of plant populations depends crucially on successful regeneration. Yet, little is known about the effects of consecutive winter and spring frost events on the regeneration stage of trees from different seed sources, although this will partly determine the success of climate warming‐driven poleward range shifts. In a common garden experiment with European beech (Fagus sylvatica) seedlings from winter 2015/2016 to autumn 2017, we studied how simulated successive spring and winter frost events affect leaf‐out dates, growth performance, and survival rates of 1‐ to 2‐year‐old seedlings from provenances differing in climate at origin. We further investigated the combined effects of successive frost events. The first spring frost after germination led to a mortality rate up to 75%, resulting in reduced seedling numbers but better frost tolerance of the survivors, as reflected in a weaker impact of the following winter frost event in the survivors compared to the non‐acclimated control. Final plant height was most strongly reduced by the spring frost in the second year. The winter frost event delayed leaf‐out by up to 40 days, leading to severe growth impairment in 2017. Our results indicate partly successful frost acclimation and/or the selection of frost‐hardier individuals, because the negative growth effects of consecutive frost events did not add up after exposure to more than one event. Both mechanisms may help to increase the frost tolerance of beech offspring. Nevertheless, mortality after the first spring frost was high, and frost exposure generally caused growth reductions. Thus, achieving higher frost tolerance may not be sufficient for beech seedlings to overcome frost‐induced reductions in competitive strength caused by winter frost damage and delayed leaf enfolding.

Abstract Phenotypic plasticity can buffer organisms against short‐term environmental fluctuations. For example, previous exposure to increased temperatures can increase thermal tolerance in many species. Prior studies have found that acclimation to higher temperature can influence the magnitude of transcriptional response to subsequent acute thermal stress (hereafter, “transcriptional response modulation”). However, mechanisms mediating this gene expression response and, ultimately, phenotypic plasticity remain largely unknown. Epigenetic modifications are good candidates for modulating transcriptional response, as they broadly correlate with gene expression. Here, we investigate changes in DNA methylation as a possible mechanism controlling shifts in gene expression plasticity and thermal acclimation in the reef‐building coral Acropora nana. We find that gene expression response to acute stress is altered in corals acclimated to different temperatures, with many genes exhibiting a dampened response to heat stress in corals pre‐conditioned to higher temperatures. At the same time, we observe shifts in methylation during both acclimation (11 days) and acute heat stress (24 h). We observed that the acute heat stress results in shifts in gene‐level methylation and elicits an acute transcriptional response in distinct gene sets. Further, acclimation‐induced shifts in gene expression plasticity and differential methylation also largely occur in separate sets of genes. Counter to our initial hypothesis no overall correlation between the magnitude of differential methylation and the change in gene expression plasticity. We do find a small but statistically significant overlap in genes exhibiting both dampened expression response and shifts in methylation (14 genes), which could be candidates for further inquiry. Overall, our results suggest transcriptional response modulation occurs independently from methylation changes induced by thermal acclimation.

IntroductionOlive cultivation, like other evergreen fruit crops worldwide, is limited by the occurrence of frost episodes in different times of the year, mainly in winter or early spring. Some contradictory results are reported about cultivars’ response to frost, which depends on the physiological stage of the tissues (acclimated or not acclimated) when the cold or frost episode occurs. This work aimed to implement a user-friendly and reliable lab method for discerning frost tolerance.MethodsOur methodology considered both detached leaves and potted plantlets. The optimal temperature at which damage differentiated between cultivars was evaluated, as well as the time of exposure to cold and the recovery time for the correct evaluation of the symptoms. Furthermore, a comparative analysis of damage on both young and mature leaves was conducted. To validate the efficacy of the methodology, assessments were conducted on the cultivars ‘Arbequina’ (tolerant), ‘Picual’ (moderately tolerant), and ‘Frantoio’ (susceptible) under acclimated and non-acclimated conditions.Results and discussionThe results indicated that, when detached leaves were used for frost evaluation, a temperature of -10°C ± 1°C for 30 min and a recovery time at 26°C for 24–48 h after exposure to cold are enough to induce damages on the leaves and discriminate between cultivar susceptibility. Under these conditions, a precise assessment of symptoms can be made, facilitating the categorization of frost tolerance level in various olive cultivars. Notably, no significant differences were observed between young and mature leaves during the evaluation process. On the other hand, the critical temperature to assess damages on potted plantlets was determined to be -7°C ± 1°C. In addition, it was observed that acclimated plants exhibited fewer symptoms compared to non-acclimated ones, with ‘Frantoio’ being the most affected alongside ‘Picual’ and ‘Arbequina’.ConclusionThe implemented methodology will allow the assessment of frost tolerance in several olive cultivars within a short timeframe, and it is proven to be user-friendly and reliable.

Nitrogen (N) limitation has been considered as a constraint on terrestrial carbon uptake in response to rising CO2 and climate change. By extension, it has been suggested that declining carboxylation capacity (Vcmax ) and leaf N content in enhanced-CO2 experiments and satellite records signify increasing N limitation of primary production. We predicted Vcmax using the coordination hypothesis and estimated changes in leaf-level photosynthetic N for 1982-2016 assuming proportionality with leaf-level Vcmax at 25°C. The whole-canopy photosynthetic N was derived using satellite-based leaf area index (LAI) data and an empirical extinction coefficient for Vcmax , and converted to annual N demand using estimated leaf turnover times. The predicted spatial pattern of Vcmax shares key features with an independent reconstruction from remotely sensed leaf chlorophyll content. Predicted leaf photosynthetic N declined by 0.27% yr-1 , while observed leaf (total) N declined by 0.2-0.25% yr-1 . Predicted global canopy N (and N demand) declined from 1996 onwards, despite increasing LAI. Leaf-level responses to rising CO2 , and to a lesser extent temperature, may have reduced the canopy requirement for N by more than rising LAI has increased it. This finding provides an alternative explanation for declining leaf N that does not depend on increasing N limitation.

Abstract Background GOLDEN-like (GLK) transcription factors are central regulators of chloroplast biogenesis in Arabidopsis and other species. Findings from Arabidopsis show that these factors also contribute to photosynthetic acclimation, e.g. to variation in light intensity, and are controlled by retrograde signals emanating from the chloroplast. However, the natural variation of GLK1-centered gene-regulatory networks in Arabidopsis is largely unexplored. Results By evaluating the activities of GLK1 target genes and GLK1 itself in vegetative leaves of natural Arabidopsis accessions grown under standard conditions, we uncovered variation in the activity of GLK1 centered regulatory networks. This is linked with the ecogeographic origin of the accessions, and can be associated with a complex genetic variation across loci acting in different functional pathways, including photosynthesis, ROS and brassinosteroid pathways. Our results identify candidate upstream regulators that contribute to a basal level of GLK1 activity in rosette leaves, which can then impact the capacity to acclimate to different environmental conditions. Indeed, accessions with higher GLK1 activity, arising from habitats with a high monthly variation in solar radiation levels, may show lower levels of photoinhibition at higher light intensities. Conclusions Our results provide evidence for natural variation in GLK1 regulatory activities in vegetative leaves. This variation is associated with ecogeographic origin and can contribute to acclimation to high light conditions.

The American lobster fishery is the most economically significant commercial fishery in Atlantic Canada and takes place in waters that are warming due to climate change. Lobster are poikilotherms that tolerate a wide range of seasonal temperatures with an optimal range of 12–18 °C. Lobster in the Canadian Maritimes may be naturally acclimated to a wide range of temperatures and thus, could have a wide range of thermal tolerance that may be distinct across regions. The present study used non-invasive open-source tools to explore differences in thermal tolerance in real time between geographically separated lobster populations from around the Canadian Maritimes. Lobsters were acquired from six lobster fishing areas in the Canadian Maritimes and acclimated to either warm (15 °C) or cold (5 °C) water for two weeks before the onset of thermal trials. Geographic origin was not a significant predictor of estimated thermal maximum, while acclimation temperature was a significant predictor. These results suggest that thermal tolerance is more strongly linked to acclimation temperature than to geographic region.

The aim of this study was to investigate the effect of acclimation at different applied voltages on the electricity generation and composition of anodic biofilm in microbial fuel cell (MFC). Single-chamber air-cathode 1.2-L MFCs were acclimated by different applied voltages (i.e., 0.5, 1.0, 1.5 and 2.0 V). The maximum power density of 1587 mW/m2 and maximum current density of 32.2 ± 0.3 A/m2 were obtained in the MFC after the acclimation by 1.0 V, respectively, which were 10.5 and 1610 times that of the unacclimated MFC. The MFC acclimated by 2.0 V could remain in stable operation with an average maximum current density of 7.8 ± 0.1 A/m2 and a maximum power density of 1439 ± 30 mW/m2 within 40 cycles (∼20 d). The average biomass of the anodic biofilms increased from 3.8 ± 0.9 mg/g before the acclimation to 11.3 ± 1.6 mg/g after the acclimation by 2.0 V. Correspondingly, the anode bacterial viability increased from 0.74 ± 0.02 to 0.91 ± 0.02. The relative abundance of Geoalkalibacter decreased from 18.7 % to 36.1 % before the acclimation to 1.3 %–18.0 % after the acclimation by 2.0 V in the bacterial communities of the anodic biofilms in the MFC. Results from this study should provide an easy way to enhance electrochemically active bacteria acclimation and improve electricity generation in the MFC.

Abstract Background Heat stress (HS) is an increasing threat for pig production with a wide range of impacts. When submitted to high temperatures, pigs will use a variety of strategies to alleviate the effect of HS. While systemic adaptations are well known, tissue-specific changes remain poorly understood. In this study, thirty-two pigs were submitted to a 5-day HS at 32 °C. Results Transcriptomic and metabolomic analyses were performed on several tissues. The results revealed differentially expressed genes and metabolites in different tissues. Specifically, 481, 1774, 71, 1572, 17, 164, and 169 genes were differentially expressed in muscle, adipose tissue, liver, blood, thyroid, pituitary, and adrenal glands, respectively. Regulatory glands (pituitary, thyroid, and adrenal) had a lower number of regulated genes, perhaps indicating an earlier sensitivity to HS. In addition, 7, 8, 2, and 8 metabolites were differentially produced in muscle, liver, plasma, and urine, respectively. The study also focused on the oxidative stress pathway in muscle and liver by performing a correlation analysis between genes and metabolites. Conclusions This study has identified various adaptation mechanisms in swine that enable them to cope with heat stress (HS). These mechanisms include a global decrease in energetic metabolism, as well as changes in metabolic precursors that are linked with protein and lipid catabolism and anabolism. Notably, the adaptation mechanisms differ significantly between regulatory (pituitary, thyroid and adrenal glands) and effector tissues (muscle, adipose tissue, liver and blood). Our findings provide new insights into the comprehension of HS adaptation mechanisms in swine.

Summary: The threat of rising global temperatures may be especially pronounced for low‐latitude, lowland plant species that have evolved under stable climatic conditions. However, little is known about how these species may acclimate to elevated temperatures. Here, we leveraged a strong, steep thermal gradient along a natural geothermal river to assess the ability of woody plants in the Amazon to acclimate to elevated air temperatures.We measured leaf traits in six common tropical woody species along the thermal gradient to investigate whether individuals of these species: acclimate their thermoregulatory traits to maintain stable leaf temperatures despite higher ambient temperatures; acclimate their photosynthetic thermal tolerances to withstand hotter leaf temperatures; and whether acclimation is sufficient to maintain stable leaf thermal safety margins (TSMs) across different growth temperatures.Individuals of three species acclimated their thermoregulatory traits, and three species increased their thermal tolerances with growth temperature. Mounting evidence suggests that ectotherms are already living close to their upper physiological thermal limits. Phenotypic plasticity has been proposed to reduce the impact of climate change in the short-term providing time for adaptation, but the tolerance-plasticity trade-off hypothesis predicts organisms with higher tolerance have lower plasticity. Empirical evidence is mixed, which may be driven by methodological issues such as statistical artefacts, nonlinear reaction norms, threshold shifts or selection. Here, we examine whether threshold shifts (organisms with higher tolerance require stronger treatments to induce maximum plastic responses) influence tolerance-plasticity trade-offs in hardening capacity for desiccation tolerance and critical thermal maximum (CTMAX) across Drosophila species with varying distributions/sensitivity to desiccation/heat stress. We found evidence for threshold shifts in both traits; species with higher heat/desiccation tolerance required longer hardening treatments to induce maximum hardening responses. Species with higher heat tolerance also showed reductions in hardening capacity at higher developmental acclimation temperatures. Trade-off patterns differed depending on the hardening treatment used and the developmental temperature flies were exposed to. Based on these findings, studies that do not consider threshold shifts, or that estimate plasticity under a narrow set of environments, will have a limited ability to assess trade-off patterns and differences in plasticity across species/populations more broadly. Simple Summary: In order to help horses perform better and stay healthy in hot climates during equestrian competitions, it is recommended to get them acclimated to heat. The effects of training in a heated indoor arena on four Olympic horses (13.3 ± 2.2 years; three eventers, one para-dressage horse) were studied. The horses were trained in a heated indoor arena (32 °C with 50–60% humidity) for 14 days following their normal training routine in preparation for the Tokyo Olympic Games. Before and after this acclimation period, standardized exercise tests were performed to measure various factors like heart rate, lactate concentrations, rectal temperature, sweat loss, and sweat composition. After the 14-day training period in the heat, the horses showed positive changes. Their body temperature and heart rate decreased, and it took longer before they reached their highest body temperature, meaning they were better able to dissipate heat and experienced reduced stress. In conclusion, training for 14 days in a heated indoor arena helped reduce the thermal strain on elite sport horses, making it easier for them to compete in hot weather. This study showed that top-level horses can acclimate to heat while following their regular training routines, which is important for participating in competitions like the Olympics. To optimise the performance and welfare of horses during equestrian competitions in hot climates, it is advised to acclimate them to the heat. The effects of training in a heated indoor arena were studied. Four Olympic horses (13.3 ± 2.2 years; three eventers, one para-dressage horse) were trained for 14 consecutive days in a heated indoor arena (32 ± 1 °C; 50–60% humidity) following their normal training schedule in preparation for the Tokyo Olympic games. Standardised exercise tests (SETs) were performed on Day 1 and Day 14, measuring heart rate (HR; bpm), plasma lactate concentration (LA; mmol/L), deep rectal temperature (Trec; °C), sweat loss (SL; L), and sweat composition (K+, Cl− and Na+ concentration). The data were analysed using linear mixed models. The Trec and HR were significantly decreased after acclimation (estimate: −0.106, 95% CI −0.134, −0.078; estimate: −4.067, 95% CI −7.535, −0.598, respectively). Furthermore, for all the horses, the time taken to reach their peak Trec and heat storage increased, while their LA concentrations decreased. The SL, Cl−, and Na+ concentrations decreased in three out of the four horses. Conclusions: Fourteen days of normal training in a heated indoor arena resulted in a reduction in cardiovascular and thermal strain. Long-term ecological research of deep Lake Mondsee covers over more than six decades of phytoplankton observation. According to our study, phytoplankton groups of various phenological traits are suitable to address the impact of two major environmental stressors: nutrient surplus by eutrophication from 1968 to 1998 and warming by climate change. Here, we focus on phytoplankton biovolume, phytoplankton assemblage structure, net changes rates, and phytoplankton biodiversity. Biweekly net change-dependent persistence of phytoplankton assemblages followed a dome-shaped relationship of observations. A short-term persistence of taxonomic traits along weeks is predominant and contributes to the structural stabilization of phytoplankton assemblages. This short-term persistence is interpreted by the benefit of lifetime adjustment of phytoplankton organisms. The long-term development phytoplankton structure is discussed as an alignment of organisms over generations. Single key taxa, as Planktothrix rubescens (De Candolle ex Gomont) Anagnostidis &Komárek 1988, which are omnipotent players in phytoplankton assemblages and occur during different environmental scenarios in the long-term, are most suitable for long-term ecological research. Our findings interpret that phytoplankton taxa are excellent organisms to track the impact of environmental constraints due to their short generation time (1), their lifetime adjustment (2), and the re-occurrence in the long-term over generations (3). A total of fourteen diverse, interspecific hybrid grapevines (Vitis spp.) were evaluated for their adaptability to North Dakota winter conditions using differential thermal analysis (DTA) of low-temperature exotherms (LTE) and bud cross-sectional assessment of survival techniques. This research was conducted in two vineyard locations in eastern North Dakota. This work demonstrates the use of DTA for monitoring and selecting cultivars capable of withstanding sub-zero temperatures. These results were assessed for quantitative genetic traits. High heritability was observed for bud LTE traits and may thus be a useful target for cold hardiness breeding programs; however, it is necessary to ensure that variance is reduced when pooling multiple sample events. After DTA sampling, grapevines were assessed for survival of primary and secondary dormant buds using cross-sectional visual evaluation of death. 'Valiant' had the greatest primary bud survival (68%), followed by 'Frontenac gris', 'Crimson Pearl', and 'King of the North'. These varieties are among those with potential for production in eastern North Dakota's environment. The newly evaluated relationships between traits and the heritability of DTA results provide valuable tools to grapevine breeders for the development of cold-tolerant genotypes for future climatic challenges. We examined photosynthetic traits of pre-existing and newly developed flag leaves of four wheat genotypes grown in controlled-environment experiments. In newly developed leaves, acclimation of the maximum rate of net CO2 assimilation (A n) to warm nights (i.e. increased A n) was associated with increased capacity of Rubisco carboxylation and photosynthetic electron transport, with Rubisco activation state probably contributing to increased Rubisco activity. Metabolite profiling linked acclimation of A n to greater accumulation of monosaccharides and saturated fatty acids in leaves; these changes suggest roles for osmotic adjustment of leaf turgor pressure and maintenance of cell membrane integrity. By contrast, where A n decreased under warm nights, the decline was related to lower stomatal conductance and rates of photosynthetic electron transport. Decreases in A n occurred despite higher basal PSII thermal stability in all genotypes exposed to warm nights: T crit of 45–46.5 °C in non-acclimated versus 43.8–45 °C in acclimated leaves. Pre-existing leaves showed no change in A n–temperature response curves, except for an elite heat-tolerant genotype. These findings illustrate the impact of night-time warming on the ability of wheat plants to photosynthesize during the day, thereby contributing to explain the impact of global warming on crop productivity. There is great interest in exploring epigenetic modifications as drivers of adaptive organismal responses to environmental change. Extending this hypothesis to populations, epigenetically driven plasticity could influence phenotypic changes across environments. The canonical model posits that epigenetic modifications alter gene regulation and subsequently impact phenotypes. We first discuss origins of epigenetic variation in nature, which may arise from genetic variation, spontaneous epimutations, epigenetic drift, or variation in epigenetic capacitors. We then review and synthesize literature addressing three facets of the aforementioned model: (i) causal effects of epigenetic modifications on phenotypic plasticity at the organismal level, (ii) divergence of epigenetic patterns in natural populations distributed across environmental gradients, and (iii) the relationship between environmentally induced epigenetic changes and gene expression at the molecular level. We focus on DNA methylation, the most extensively studied epigenetic modification. We find support for environmentally associated epigenetic structure in populations and selection on stable epigenetic variants, and that inhibition of epigenetic enzymes frequently bears causal effects on plasticity. However, there are pervasive confounding issues in the literature. Effects of chromatin-modifying enzymes on phenotype may be independent of epigenetic marks, alternatively resulting from functions and protein interactions extrinsic of epigenetics. Associations between environmentally induced changes in DNA methylation and expression are strong in plants and mammals but notably absent in invertebrates and nonmammalian vertebrates. Given these challenges, we describe emerging approaches to better investigate how epigenetic modifications affect gene regulation, phenotypic plasticity, and divergence among populations. As methane fermentation is inhibited by ammonia derived from organic waste, anaerobic microbial communities tolerant to enriched wastewater with high concentrations of ammonia and salt must be obtained for methane fermentation. Therefore, acclimation cultures were prepared in bottles for 60–80 weeks with artificial wastewater medium added every 2 weeks, using three types of sludge from wastewater treatment plants in food factories. These cultures were maintained without substantially decreasing methanogenesis and gradually increasing NH4-N and salt concentrations to 5 and 34 g/L, respectively, via the accumulation of ammonia and salt through anaerobic digestion and direct addition. The culture did not show the severe inhibition of methanogenesis or the accumulation of volatile fatty acids (VFAs) such as acetic and propionic acids. The analysis of bacterial consortia in the acclimated sludge based on the 16S rRNA sequence showed that hydrogenotrophic methanogenic bacteria of the genus Methanoculleus were dominant among archaea, whereas bacteria from the orders Clostridiales and Bacteroidales were dominant among eubacteria. Further, VFA-assimilating bacteria, including synthetic acetate-oxidizing bacteria coupled with hydrogenotrophic Methanoculleus to convert methane from acetate, were present to prevent the excessive accumulation of VFAs in the acclimation culture. Plant temperature acclimation is closely related to maintaining a positive carbon gain under future climate change. However, no systematic summary of the field has been conducted. Based on this, we analyzed data on plant temperature acclimation from the Web of Science Core Collection database using bibliometric software R, RStudio and VOSviewer. Our study demonstrated that a stabilized upward trajectory was noted in publications (298 papers) from 1986 to 2011, followed by a swift growth (373 papers) from 2012 to 2022. The most impactful journals were Plant Cell and Environment, boasting the greatest count of worldwide citations and articles, the highest H-index and G-index, followed by Global Change Biology and New Phytologist, and Frontiers in Plant Science which had the highest M-index. The USA and China were identified as the most influential countries, while Atkin was the most influential author, and the Chinese Academy of Sciences was the most influential research institution. The most cited articles were published in the Annual Review of Plant Biology in 1999. “Cold acclimation” was the most prominent keyword. Future plant temperature acclimation research is expected to focus on thermal acclimation and photosynthesis, which have important significance for future agricultural production, forestry carbon sequestration, and global food security. Many published coral disease pathogen transmission and heat stress studies lack consistent acclimation periods in their experimental design, making it difficult to interpret or compare results between studies. To determine if condition of coral tissue changes during laboratory acclimation, thus potentially influencing susceptibility to stressors, we conducted a histological analysis on fragments of the Caribbean staghorn coral, Acropora cervicornis, during a 9-day acclimation period. We found a significant decline in overall condition between clippings taken initially and on day 2, and significant improvements in two key surface body wall parameters from day 2 to day 9. These findings indicate that A. cervicornis has a delayed response to new environmental conditions and therefore needs at least a 4-day acclimation period, and when possible, 9 days or more prior to experimentation. Background: Heat stress (HS) is an increasing threat for pig production with a wide range of impacts. When submitted to high temperatures, pigs will use a variety of strategies to alleviate the effect of HS. While systemic adaptations are well known, tissue-specific changes remain poorly understood. In this study, thirty-two pigs were submitted to a 5-day HS at 32 °C. Results: Transcriptomic and metabolomic analyses were performed on several tissues. The results revealed differentially expressed genes and metabolites in different tissues. Specifically, 481, 1774, 71, 1572, 17, 164, and 169 genes were differentially expressed in muscle, adipose tissue, liver, blood, thyroid, pituitary, and adrenal glands, respectively. Regulatory glands (pituitary, thyroid, and adrenal) had a lower number of regulated genes, perhaps indicating an earlier sensitivity to HS. In addition, 7, 8, 2, and 8 metabolites were differentially produced in muscle, liver, plasma, and urine, respectively. The study also focused on the oxidative stress pathway in muscle and liver by performing a correlation analysis between genes and metabolites. Conclusions: This study has identified various adaptation mechanisms in swine that enable them to cope with heat stress (HS). These mechanisms include a global decrease in energetic metabolism, as well as changes in metabolic precursors that are linked with protein and lipid catabolism and anabolism. Notably, the adaptation mechanisms differ significantly between regulatory (pituitary, thyroid and adrenal glands) and effector tissues (muscle, adipose tissue, liver and blood). Chemosensory science, the study of how organisms produce and assess olfactory information, is central to our understanding of how organisms interact and gain information about their environment. Signaling cue identification in aquatic systems lags behind our knowledge in terrestrial insects due to analytical challenges in aqueous environments. Unambiguous, reliable, and fast behavioral assays to evaluate the biological activity and function of a chemosensory cue are critical to understand aquatic signaling systems and enable research into their ecology, evolution, and threats in a changing environment. Yet, a range of anthropomorphic assumptions made in this research field create additional challenges to interpret data generated. Here, we evaluate common challenges including assumed readiness of individuals to respond, lack of information on the animals' physiological and social status, their pre-experimental cue exposure, the innate or learned character of the responses, the animals' acclimation and habituation status, and the impact of the animals upon their own environment. These factors lead to significant variability in animals' responses in bioassays, both in the field and in laboratory setups. In the light of our limited knowledge of aquatic chemosensory cues' chemical structure, active concentrations in samples, and undetermined response thresholds, we evaluate methods of mitigation to minimize differences between studies. We conclude that currently it is nearly impossible to compare results from chemosensory behavioral studies undertaken in different ecosystems, laboratories, and time points. There is an urgent need for the standardization of behavioral methods, recording of environmental conditions, and individuals' physiology, physical, and social status, to avoid conflicting and contradicting results when comparing studies. Including these parameters in experimental design and data interpretation will provide a deeper understanding of chemosensory communication, reduce unconscious bias in studies, and can help to explain the substantial individuality in animals' responses to chemosensory cues and their acclimation to environmental stress. Climate projections predict higher temperatures and more frequent hurricanes in the tropics. Tropical plants subjected to these stresses may respond by acclimating their physiology. We investigated tropical forest root respiration in response to in situ experimental warming and hurricane disturbance in eastern Puerto Rico. We measured mass-normalized root specific respiration, root biomass, and root traits at the Tropical Responses to Altered Climate Experiment (TRACE), where understory vegetation is warmed + 4 °C above ambient. Our measurements span 5 years, including before and after two major hurricanes, to quantify root contributions to ecosystem carbon fluxes. Experimental warming did not affect root specific respiration at a standard temperature of 25° (RSR25, mean = 3.89 nmol CO2 g−1 s−1) or the temperature sensitivity of root respiration (Q10, mean = 1.75), but did result in decreased fine-root biomass, thereby decreasing area-based estimations of ecosystem-level root respiration in warmed plots by ~ 35%. RSR25 of newer roots, which increased with increasing root nitrogen, showed greater rates 6 months after the hurricanes, but subsequently decreased after 12 months. Root specific respiration did not acclimate to higher temperatures, based on lack of adjustments in either Q10 or RSR25 in the warmed plots; however, decreased root biomass indicates the root contribution to soil carbon dioxide efflux was overall lower with warming. Lower root biomass may also limit nutrient and water uptake, having potential negative effects on carbon assimilation. Our results show that warming and hurricane disturbance have strong potential to affect tropical forest roots, as well as ecosystem carbon fluxes. [ABSTRACT FROM AUTHOR]

Summary: Efficient plant acclimation to changing environmental conditions relies on fast adjustments of the transcriptome, proteome, and metabolome. Regulation of enzyme activity depends on the activity of specific chaperones, chemical post‐translational modifications (PTMs) of amino acid residues, and changes in the cellular and organellar microenvironment. Central to carbon assimilation, and thus plant growth and yield, Rubisco activity is regulated by its chaperone Rubisco activase (Rca) and by adjustments in the chloroplast stroma environment. Focused on crops, this review highlights the main PTMs and stromal ions and metabolites affecting Rubisco and Rca in response to environmental stimuli. Rca isoforms differ in regulatory properties and heat sensitivity, with expression changing according to the surrounding environment. Much of the physiological relevance of Rubisco and Rca PTMs is still poorly understood, though some PTMs have been associated with Rubisco regulation in response to stress. Ion and metabolite concentrations in the chloroplast change in response to variations in light and temperature. Some of these changes promote Rubisco activation while others inhibit activation, deactivate the enzyme, or change the rates of catalysis. Understanding these regulatory mechanisms will aid the development of strategies to improve carbon fixation by Rubisco under rapidly changing environments as experienced by crop plants. This review is the final contribution to a four-part, historical series on human exercise physiology in thermally stressful conditions. The series opened with reminders of the principles governing heat exchange and an overview of our contemporary understanding of thermoregulation (Part 1). We then reviewed the development of physiological measurements (Part 2) used to reveal the autonomic processes at work during heat and cold stresses. Next, we re-examined thermal-stress tolerance and intolerance, and critiqued the indices of thermal stress and strain (Part 3). Herein, we describe the evolutionary steps that endowed humans with a unique potential to tolerate endurance activity in the heat, and we examine how those attributes can be enhanced during thermal adaptation. The first of our ancestors to qualify as an athlete was Homo erectus, who were hairless, sweating specialists with eccrine sweat glands covering almost their entire body surface. Homo sapiens were skilful behavioural thermoregulators, which preserved their resource-wasteful, autonomic thermoeffectors (shivering and sweating) for more stressful encounters. Following emigration, they regularly experienced heat and cold stress, to which they acclimatised and developed less powerful (habituated) effector responses when those stresses were re-encountered. We critique hypotheses that linked thermoregulatory differences to ancestry. By exploring short-term heat and cold acclimation, we reveal sweat hypersecretion and powerful shivering to be protective, transitional stages en route to more complete thermal adaptation (habituation). To conclude this historical series, we examine some of the concepts and hypotheses of thermoregulation during exercise that did not withstand the tests of time. [ABSTRACT FROM AUTHOR]

Climate change presents a challenge for plants to acclimate their water relations under changing environmental conditions, and may increase the risks of hydraulic failure under stress. In this study, maize plants were acclimated to two different CO2 concentrations ([CO2]; 400 ppm and 700 ppm) while under either water stress (WS) or soil salinity (SS) treatments, and their growth and hydraulic traits were examined in detail. Both WS and SS inhibited growth and had significant impacts on hydraulic traits. In particular, the water potential at 50% loss of stem hydraulic conductance (P 50) decreased by 1 MPa in both treatments at 400 ppm. When subjected to elevated [CO2], the plants under both WS and SS showed improved growth by 7–23%. Elevated [CO2] also significantly increased xylem vulnerability (measured as loss of conductivity with decreasing xylem pressure), resulting in smaller hydraulic safety margins. According to the plant desiccation model, the critical desiccation degree (time×vapor pressure deficit) that the plants could tolerate under drought was reduced by 43–64% under elevated [CO2]. In addition, sensitivity analysis showed that P 50 was the most important trait in determining the critical desiccation degree. Thus, our results demonstrated that whilst elevated [CO2] benefited plant growth under WS or SS, it also interfered with hydraulic acclimation, thereby potentially placing the plants at a higher risk of hydraulic failure and increased mortality. One of life's decisive innovations was to harness the catalytic power of metals for cellular chemistry. With life's expansion, global atmospheric and biogeochemical cycles underwent dramatic changes. Although initially harmful, they permitted the evolution of multicellularity and the colonization of land. In land plants as primary producers, metal homeostasis faces heightened demands, in part because soil is a challenging environment for nutrient balancing. To avoid both nutrient metal limitation and metal toxicity, plants must maintain the homeostasis of metals within tighter limits than the homeostasis of other minerals. This review describes the present model of protein metalation and sketches its transfer from unicellular organisms to land plants as complex multicellular organisms. The inseparable connection between metal and redox homeostasis increasingly draws our attention to more general regulatory roles of metals. Mineral co-option, the use of nutrient or other metals for functions other than nutrition, is an emerging concept beyond that of nutritional immunity. [ABSTRACT FROM AUTHOR]
