Abstract

Objective We investigated the genetic diversity, molecular epidemiology and evolutionary dynamics of Staphylococcus aureus (SA) isolated from SLE patients by means of phylogenetic analysis. Methods Consecutive SLE patients (ACR 1997 criteria) were enrolled: clinical/laboratory data were collected and nasal swab for SA identification was performed. On the basis of the translation elongation factor (tuf) gene, a phylogenetic analysis was performed to investigate relationships and to assess significant clades. Selective pressure analysis was used to investigate the evolution of the SA tuf gene. The gene sequences from non-SLE individuals, downloaded from the GenBank database, were compared through phylogenetic analysis with the tuf gene from SLE patients. Results We enrolled 118 patients [M/F 10/108; median (interquartile range (IQR)) age 45.5 (13.2) years; median (IQR) disease duration 120 (144) months]. Twenty-four patients (20.3%) were SA carriers (SA+), three of them MRSA. SA+ SLE showed significantly higher SLEDAI-2k values [SA+: median (IQR) 2 (3.75); SA−: 0 (2); P = 0.04]. The phylogenetic analysis, restricted to 21 non-MRSA SA+, revealed a statistically supported larger clade (A, n  = 17) and a smaller one (B, n  = 4). Patients located in clade A showed a significantly higher prevalence of joint involvement (88.2%) in comparison with clade B (50.0%, P  < 0.0001) and SA− (62.7%, P  < 0.0001). Haematological manifestations were significantly more frequent in clade A (64.7%) compared with B (50.0%, P  = 0.004). Conclusion We suggest a possible role of SA nasal carriage status in SLE disease activity. Abstract

Environmental status and conservation values of Croatian rivers have never been comprehensively assessed. We present a methodology for the identification of the most pristine rivers of high conservation value which are important for the protection of freshwater biodiversity in Croatia. The methodology is a combination of ecoregional conservation approach where representation of all distinct biodiversity features is a key goal, and assessment of river reach condition. Main units of analysis, the Evaluated River Reaches (ERRs), were identified after all river types were distinguished. The condition of each ERR was assessed using a combination of five criteria: hydrology, channel morphology, riparian vegetation, water quality and land use. The final condition of ERR was scored (max. 50) by summing scores for the five criteria. The distribution of endangered freshwater f ish species and protected areas was used for prioritizing the ERRs in high and good condition. Altogether 82 rivers and 4522 km of river length were analyzed in three ecoregions: Pannonian, Dinaric‐Continental and Dinaric‐ Mediterranean. The total length of very high priority ERRs was 1607 km (35%), while additional ERRs comprising 998 km (22%) of river length were included to assure representation of all river types in each ecoregion. The selected ERRs represent priority river reaches whose protection should ensure the long term persistence of the most important features of freshwater biodiversity in Croatia.

81. SNCA and mTOR Pathway Single Nucleotide Polymorphisms Interact to Modulate the Age at Onset of Parkinson's Disease.

Abstract

Background: Single nucleotide polymorphisms (SNPs) in the α-synuclein (SNCA) gene are associated with differential risk and age at onset (AAO) of both idiopathic and Leucine-rich repeat kinase 2 (LRRK2)-associated Parkinson's disease (PD). Yet potential combinatory or synergistic effects among several modulatory SNPs for PD risk or AAO remain largely underexplored.Objectives: The mechanistic target of rapamycin (mTOR) signaling pathway is functionally impaired in PD. Here we explored whether SNPs in the mTOR pathway, alone or by epistatic interaction with known susceptibility factors, can modulate PD risk and AAO.Methods: Based on functional relevance, we selected a total of 64 SNPs mapping to a total of 57 genes from the mTOR pathway and genotyped a discovery series cohort encompassing 898 PD patients and 921 controls. As a replication series, we screened 4170 PD and 3014 controls available from the International Parkinson's Disease Genomics Consortium.Results: In the discovery series cohort, we found a 4-loci interaction involving STK11 rs8111699, FCHSD1 rs456998, GSK3B rs1732170, and SNCA rs356219, which was associated with an increased risk of PD (odds ratio = 2.59, P < .001). In addition, we also found a 3-loci epistatic combination of RPTOR rs11868112 and RPS6KA2 rs6456121 with SNCA rs356219, which was associated (odds ratio = 2.89; P < .0001) with differential AAO. The latter was further validated (odds ratio = 1.56; P = 0.046-0.047) in the International Parkinson's Disease Genomics Consortium cohort.Conclusions: These findings indicate that genetic variability in the mTOR pathway contributes to SNCA effects in a nonlinear epistatic manner to modulate differential AAO in PD, unraveling the contribution of this cascade in the pathogenesis of the disease. © 2019 International Parkinson and Movement Disorder Society. 82. Lavoisier and Music: An Analysis of Notes sur la Musique (1778) within Its Cultural Context.

Abstract

Notes sur la Musique are a set of notes devoted by the French chemist Antoine Laurent Lavoisier (1743–1794) to music theory and in particular to the rules for correct composition. Given its subject matter, Notes constitutes a unicum in Lavoisier's corpus that has been little studied and never published. This essay will present the initial results of an in-depth analysis of this manuscript, followed by a transcription of the text. It will be argued that a large part of the Notes were derived from the writings of Alexandre Théophile Vandermonde (1735–1796), a mathematician who collaborated with Lavoisier on his scientific experiments, but who also developed his own system of harmony. After a brief examination of Vandermonde's contributions to music theory, various passages from Lavoisier's Notes will be evaluated in order to show the link between the two scientists' perspectives on music. Some of Vandermonde's unpublished papers will also be taken into consideration. On the basis of this analysis, hypotheses will be advanced regarding the circumstances that could have led Lavoisier to take an interest in music theory. Overall, this paper will shed light on a little-known side of Lavoisier's cultural interests and activities and provide elements that could contribute to a better understanding of his Notes sur la Musique. Abstract

This paper reports on DNA DSB induction in human fibroblasts by iron ions of different energies, namely 5, 1 GeV/u, 414 and 115 MeV/u, in absence or presence of different shields (PMMA, Al and Pb). Measure of DNA DSB was performed by calibrated Pulsed Field Gel Electrophoresis using the fragment counting method. The RBE–LET relationships for unshielded and shielded beams were obtained both in terms of dose average LET and of track average LET. Weak dependence on these parameters was observed for DSB induction. The shielding efficiency, evaluated by the ratio between the cross sections for unshielded and shielded beams, depends not only on the shield type and thickness, but also on the beam energy. Protection is only observed at high iron ions energy, especially at 5 GeV/u, where PMMA shield gives higher protection compared to Al or Pb shields of the same thickness expressed in g/cm2.

Abstract

Taking risks is a natural human response, but, in some, risk taking is compulsive and may be detrimental. The subthalamic nucleus (STN) is thought to play a large role in our ability to inhibit responses. Differences between individuals' ability to inhibit inappropriate responses may underlie both the normal variation in trait impulsivity in the healthy population, as well as the pathological compulsions experienced by those with impulse control disorders (ICDs). Thus, we review the role of the STN in response inhibition, with a particular focus on studies employing imaging methodology. We also review the latest evidence that disruption of the function of the STN by deep brain stimulation in patients with Parkinson's disease can increase impulsivity.

Abstract

Tau and amyloid-β (Aβ) aggregates have been suggested to play a role in the development of dementia in Parkinson's disease (PD). Positron emission tomography (PET) with [18F]FDDNP and the determination of cerebrospinal fluid (CSF) levels of these proteins constitute a means to visualize in vivo Aβ and tau brain accumulation. Information about longitudinal changes of these CSF and PET biomarkers in PD with regard to progression to dementia is lacking. We assessed the cross-sectional and longitudinal associations of CSF and PET biomarkers of tau and Aβ with PD-related cognitive dysfunction in 6 healthy-controls (HC), 16 patients with PD without dementia (PDND), and 8 PD with dementia (PDD). All subjects underwent comprehensive neuropsychological testing, [18F]FDDNP PET, and CSF Aβ-tau determination. After 18 months, the PDND group was re-assessed clinically and by neuropsychological, PET, and CSF determinations. Cross-sectionally, PDD had higher [18F]FDDNP binding in lateral temporal regions and lower levels of CSF Aβ levels compared to PDND, with a congruent correlation between the [18F]FDDNP binding and CSF Aβ levels. Longitudinally, higher baseline lateral temporal [18F]FDDNP binding was associated to longitudinal worsening in cognitive performances and progression to dementia among subjects classified as PDND at baseline, who additionally disclosed at follow-up an increase in lateral-temporal FDDNP binding, as well as a reduction in CSF Aβ and an increase in CSF tau levels. These results confirm the relevance of these CSF and PET biomarkers to PDD, being specifically the first to show [18F]FDDNP PET as a dementia risk biomarker in PD, along with longitudinal CSF and PET changes over time. Abstract

Protons and high energy heavy ions (HZE particles ) are the components of space radiation of major concern. Their biological consequences are expected to be related to DNA lesions. Since DNA double strand breaks (DSB) and other lesions produced by these charged particles occurs in a correlated manner along the particle track, the current working hypothesis is that spatial correlation of damage, that depends on the particle type and energy, affects its reparability. Several characteristic distances for damage correlation can be considered, related to various levels of chromatin organization. We have investigated, in AG1522 human fibroblasts, the DNA fragmentation patterns in the size range 23 kbp - 5.7 Mbp, characteristic of the loop structure of chromosomes, using calibrated Pulsed Field Gel electrophoresis (PFGE). We have compared various beams i.e., low -energy protons of 0.88 MeV (representative of stopping protons at the Bragg peak) and Fe ions of 1050 and 414 MeV/u, using g-rays as reference. Irradiations with protons were performed at the Laboratori Naz ionali di Legnaro (Padova, Italy), those with 1050 MeV/u Fe ions at the Brookhaven National Laboratory (USA) and those with 414 MeV/u Fe ions at the National Institute for Radiological Sciences (Chiba, Japan). DSB yields were evaluated from the number of DNA fragments induced in the size range studied, at doses between 40 and 200 Gy. The results until now obtained show the general trend of a linear, or almost linear, increase of DSB with the dose. 1050 MeV/u Fe ions are the most effective particles in terms of both unit dose and unit fluence. The frequency distributions of fragments induced by the charged particles are shifted towards smaller sizes with respect to that induced by comparable doses of g-rays. These distributions also suggest that at doses of the order of 100 Gy most of the fragments induced by protons have size > 23 kbp, while for Fe ions it is expected a significant amount of fragments smaller than that size, not revealed by the mentioned PFGE analysis. This is in agreement with recent experimental measurements of DNA fragments induced by 1050 MeV/u Fe ions in the size range 2 - 23 kbp. The occurrence of such small fragments (and even smaller) further increases the effectiveness for DSB induction of HZE particles compared to protons., 34th COSPAR Scientific Assembly

Abstract

Carbapenems resistant Enterobacteriaceae infections are increasing worldwide representing an emerging public health problem. The application of phylogenetic and phylodynamic analyses to bacterial whole genome sequencing (WGS) data have become essential in the epidemiological surveillance of multi-drug resistant nosocomial pathogens. Between January 2012 and February 2013, twenty-one multi-drug resistant K. pneumoniae strains, were collected from patients hospitalized among different wards of the University Hospital Campus Bio-Medico. Epidemiological contact tracing of patients and Bayesian phylogenetic analysis of bacterial WGS data were used to investigate the evolution and spatial dispersion of K. pneumoniae in support of hospital infection control. The epidemic curve of incident K. pneumoniae cases showed a bimodal distribution of cases with two peaks separated by 46 days between November 2012 and January 2013. The time-scaled phylogeny suggested that K. pneumoniae strains isolated during the study period may have been introduced into the hospital setting as early as 2007. Moreover, the phylogeny showed two different epidemic introductions in 2008 and 2009. Bayesian genomic epidemiology is a powerful tool that promises to improve the surveillance and control of multi-drug resistant pathogens in an effort to develop effective infection prevention in healthcare settings or constant strains reintroduction.
