Objective: To examine the fauna of rodents as zoonotic cutaneous leishmaniasis reservoir hosts in Zarqan County, Fars Province, south of Iran, during 2012. Methods: During 2012, wild rodents from different parts of this region were caught by Sherman traps and checked by the examination of liver and spleen smears, for Leishmania infection, to see which species were acting as reservoir hosts; the slides were then processed to extract DNA for molecular test using PCR assay. Results: From 108 rodent species caught, 63% were male and 37% identified as female. Meriones libycus was the most abundant species caught (80.5%) and 5.7% of them were found to be smear-positive for Leishmania amastigotes. The other species were Rattus rattus (14.8%) and Mus musculus (4.7%), but none of them were found positive. Leishmania infection was observed in male and female samples microscopically. Moreover, molecular results revealed Leishmania major in three male and two female specimens. Conclusions: Based on our knowledge, Meriones libycus is incriminated as the main reservoir hosts of Leishmania major in the rural area of Zarqan.

Background and purpose: Cutaneous leishmaniasis is a global health problem. The discovery of new and highly efficient anti-leishmanial treatments with lower toxicity is globally needed. The current study was carried out to evaluate the anti-leishmanial effects of artemether (ART) and ART-loaded nanostructured lipid carriers (ART-NLCs) against promastigotes and amastigotes of Leishmania major. Experimental approach: Solvent diffusion evaporation technique was applied to prepare ART-NLCs. These nanoparticles were characterized using a particle size analyzer (PSA), transmission electron microscopy (TEM), and dynamic light scattering (DLS). The antiparasitic activity on amastigote was assessed in J774 cell culture. The drug cytotoxicity on promastigote and macrophage was assessed using the MTT technique after 24 and 48 h and compared with NLCs, ART, and amphotericin B, as the control agents. The selectivity index was calculated for the agents. Findings/Results: The DLS and PSA techniques confirmed that ART-NLCs were homogenous in size with an average diameter of 101 ± 2.0 nm and span index of 0.9. The ART-NLCs significantly heighten the anti-leishmanial activity of ART (P < 0.001). The IC50 values of ART and ART-NLCs on promastigotes after 24 and 48 h were 76.08, 36.71 and 35.14, 14.81 μg/mL, respectively while they were calculated 53.97, 25.43 and 20.13, 11.92 for amastigotes. Also, ART-NLCs had the lowest cytotoxicity against macrophages. Furthermore, among the agents tested, ART-NLCs had the highest selectivity index. Conclusion and implications: ART-NLCs had lower cytotoxic effects than ART and amphotericin B, also its selectivity index was significantly higher. Background and purpose: The aim of this study was to evaluate the in vitro and in vivo anti-Toxoplasma gondii (T. gondii) effect of 5-oxo-hexahydroquinoline compounds. Moreover, molecular docking study of the compounds into the active site of enoyl-acyl carrier protein reductase (ENR) as a necessary enzyme for the vitality of apicoplast was carried out. Experimental approach: A number of 5-oxo-hexahydoquinoline derivatives (Z1-Z4) were synthesized. The T. gondii tachyzoites of RH strain were treated by different concentrations (1-64 μg/mL) of the compounds. The viability of the encountered parasites with compounds was assessed using flow cytometry and propidium iodide (PI) staining. Due to the high mortality effect of Z3 and Z4 in vitro, their chemotherapy effect was assessed by inoculation of tachyzoites to four BALB/c mice groups (n = 5), followed by the gavage of various concentrations of the compounds to the mice. Molecular docking was done to study the binding affinity of the synthesized 5-oxo-hexahydroquinolines into ENR enzyme active site byusing AutoDock Vina® software. Docking was performed by a Lamarckian Genetic Algorithm with 100 runs. Findings / Results: Flow cytometry assay results indicated compounds Z3 and Z4 had relevant mortality effect on parasite tachyzoites. Besides, in vivo experiments were also performed and a partial increase of mice longevity between control and experiment groups was recorded. Molecular docking of Z3 and Z4 in the binding site of ENR enzyme indicated that the compounds were well accommodated within the binding site. Therefore, it could be suggested that these compounds may exert their anti-T. gondii activity through the inhibition of the ENR enzyme. Conclusion and implications: Compounds Z3 and Z4 are good leads in order to develop better anti-T. gondii agents as they demonstrated both in vitro and in vivo inhibitory effects on tachyzoites viability and infection. Further studies on altering the route of administration along with additional pharmacokinetics evaluations are needed to improve the anti-T. gondii impacts of 5-oxo-hexahydroquinoline compounds. Background: We aimed to detect Toxoplasma gondii in ovine aborted fetuses and evaluate its genetic variations in the southwest of Iran. Methods: This cross-sectional study was performed on 100 aborted ovine fetuses collected from the different region of Kohgiluyeh and Boyer-Ahmad Province, Iran, in lambing season during 2017 and 2018. DNA was extracted from the brain samples of all of the aborted fetuses and PCR amplified, targeting a 529 bp repetitive element gene of T. gondii. Moreover, to find out the heterogeneity of the positive samples, PCR-DNA amplification of the two main genetic markers, B1 and GRA6, of T. gondii were performed. Nucleotide sequencing and phylogenetic analysis were performed, using the BLAST program and MEGA-X software. Results: The 529 bp gene of T. gondii was detected in 2 out of 100 (2%) of the ovine aborted samples. The sequences analysis of GRA6 and B1 genes revealed that both isolates from the aborted fetuses of sheep belonged to type I of T. gondii. Intra divergence was more seen in GRA6 gene whereas less divergence was ob-served in B1 gene. Conclusion: Congenital infection with Type I of T. gondii during the neonatal period is associated with abortion in ovine. Evaluation of more aborted samples from broader geographical areas is needed to elucidate the molecular epidemiology and also the genotypes of T. gondii associated with abortion. Background: Toxoplasmosis is a global zoonotic disease that causes critical medical complications in neonates and immunocompromised persons. Infection rates in cats, specifically stray cats, are believed to be the best sentry of the level of Toxoplasma gondii in the environment. Therefore, in this study, we surveyed T. gondii infection in stray cats of Shiraz, one of the metropolises of Iran. Methods: The appearance of antibodies and DNA of T. gondii in samples from 145 stray cats was determined in order to appraise the prevalence of T. gondii infection, by MAT and Nested-PCR. Results: The rate of T. gondii infection in the cats was 69% by PCR and 82.8% by MAT. Besides, the highest rate of infection was discerned in diaphragm (37.9%) and intercostal muscle (34.5%), while the lowest rate was related to ileum (6.9%). Moreover, the similarity between MAT with titers 1:20, 1:40 and PCR were 79.2% and 86.2%, respectively (P=0.02 and P=0.0001). Conclusion: Nested-PCR and MAT are valuable techniques for molecular and serological detection of T. gondii. The prevalence of T. gondii infection in stray cats in Shiraz is high. Objective: To explore the co-detection of natural infection of Trypanosomatidae parasites such as Leishmania and Crithidia in reservoir hosts of leishmaniasis. Methods: Rodent populations were monitored in two endemic foci of cutaneous leishmaniasis of Fars province, southern Iran from March to October 2016. Rodents were trapped alive in several parts of Shiraz and Kharameh cities. Afterwards, their organs were prepared for detection of Leishmania and Crithidia species by molecular, microscopic, and culture methods. Results: Totally, 115 rodents of five species; Tatera indica (T. indica) (85), Rattus rattus (12), Meriones libycus (9), Mus musculus (7), and Rattus norvegicus (2), were trapped alive and their tissue samples were examined using microscopic, cultivation, and molecular assays. Overall, 59 (51.3%) rodents were positive for Leishmania or Crithidia parasites. The highest rate (61.2%; 52/85) of Leishmania infection was related to the T. indica population. The cultivation, and molecular observations showed that two (2.4%; 2/85) of T. indica (foot-pad, and spleen samples) were positive to Crithidia. Conclusions: This is the first report of Crithidia infection in T. indica in Iran. Consequently, more epidemiological and ecological studies are needed to understand the role of Crithidia and Leishmania in T. indica. [ABSTRACT FROM AUTHOR]

Introduction: Almost 1.5-2 million people are infected with leishmaniasis and 60,000 die due to the disease every year. The treatment of cutaneous leishmaniasis with the existing medications is not effective and accompanied with various side effects. Expensiveness and not being available in different forms are other limitations associated with these drugs. In addition to, parasite resistance restricts the use of these medications; therefore, identification of new treatment methods is highly essential. Humans have always used medicinal plants for the treatment of diseases. This study aimed to assess the impact of quercetin and luteolin on leishmaniasis wounds created on mice's tails. Materials and Methods: A total of 56 infected Balb/C mice were divided into eight groups, each containing seven animals. The first three groups received 3.5 mg/kg luteolin through oral, intradermal, and intraperitoneal routes. The other three groups received 14 mg/kg quercetin through oral, intradermal, and intraperitoneal routes. The treatments were administered twice a week for 4 weeks in comparison to meglumine antimonite as control groups. Results: The results showed that there was no statistically significant difference between the recovery of mice in the luteolin and quercetin groups and those in the meglumine antimonite group. However, the ratio of recovered Balb/C mice in the luteolin and quercetin group is 1.75-2 times more than meglumine group, respectively. Conclusion: Quercetin and luteolin seems to be candidate medications with fewer side effects for recovery from cutaneous leishmaniasis. They can also be used as complementary medications together with other standard drugs, such as meglumine, antimonite, and cryotherapy. [ABSTRACT FROM AUTHOR]
