Abstract

Reproduction involves multiple complex behaviours, and the effects of familiarity on such social interactions are seldom described in fish. This is particularly true for sound production and communication within aggressive or non-aggressive context. This study explores the effects of a common garden rearing without parental care of two closely related cichlid species (Nile tilapia Oreochromis niloticus and blackchinned tilapia Sarotherodon melanotheron) on their sound production features and social interactions. After 9 months in common garden rearing, from embryonic stage to first maturity, sound production and associated behaviours were recorded on specimens of the two species in intraspecific and interspecific pairings. The authors found that fish were able to produce the same kind of sounds as those recorded in similar context for their parents. Drum sounds were associated to chasing, lateral attack and courtship in O. niloticus and only to fleeing or avoidance in S. melanotheron. Specific grunts were produced in chasing, after biting and in nest building by O. niloticus, and specific rolling sounds were associated to courtship in S. melanotheron. Sound production and behaviours were not correlated to sex steroid levels, but the number of sounds recorded in aggressive context was correlated to dominance in O. niloticus. The authors conclude that one generation of common garden rearing does not modify sound features, which remain specific and innate in the two cichlids. Despite the familiarity, O. niloticus remained dominant on S. melanotheron, but the aggressiveness between the two species decreased. [ABSTRACT FROM AUTHOR]

Abstract

International audience; Introduction En raison d’une forte pression anthropique, les zones estuariennes sont touchées par une importante pollution aux microplastiques (MP) et les espèces inféodées à ces zones y sont particulièrement exposées. L’objectif de cette étude, financée par le programme Seine-Aval, est d’évaluer les effets de MP environnementaux collectés en estuaire de Seine sur une espèce emblématique de nos côtes, la sole commune (Solea solea) à travers une exposition directe ou par voie trophique. Exposition Deux types d’expositions ont été réalisés. La première est une exposition directe de larves de sole à plusieurs plastiques industriels de différentes tailles et densités (27-35 μm et d=0.99 ; 27-32 μm et d=1.20 ; 63-75 μm et d=1.20) ainsi qu’à un cocktail d'échantillons environnementaux prélevés dans l’estuaire de la Seine (100 μm) afin de déterminer la cinétique d'ingestion et d'égestion. Pour mettre en évidence d'éventuelles différences en fonction du stade de développement, cette exposition a été réalisée avant (7 jours post éclosion, jpe), pendant (21-22 jpe) et après la métamorphose (41-42 jpe). Dans une seconde expérience, des juvéniles de sole ont été nourris avec des vers estuariens (Hediste diversicolor) préalablement exposés au MP via le sédiment. Comme précédemment les MP utilisés sont d’origines industrielles ou d'estuaire de Seine. Plusieurs marqueurs d’état de santé tels que la survie, la croissance, le comportement, le métabolisme énergétique, le système immunitaire, le système digestif ont été étudiés. Résultats Quelque soit le stade de développement, les larves ont ingérés tous les plastiques indépendamment de la taille et/ou du type et aucune différence de cinétique d'ingestion ou d'égestion n'a été mise en évidence. Pendant la métamorphose le comportement natatoire des larves de sole a été modifié : la distance parcourue est plus faible chez les larves ayant consommé des MP et pour tous les MP testés. Chez les juvéniles de soles nourris avec des vers exposés à des MP environnementaux (400 μm) ou industriels (100-250 μm) nous avons mis en évidence une modification de comportement dans le choix du sol proposé (noir ou blanc) et de couleur des individus exposés pour la concentration la plus haute et les MP enrobés de Benzo(a)Pyrène ou d’Oxybenzone. Ces observations sont indicatrices d'un stress physiologiques. Ces premiers résultats mettent en évidence les effets délétères des microplastiques lors d'expositions directes ou indirectes à différents stades de vie. Ce travail sera complété par une exposition de larves de soles pendant 1 mois à partir de l'éclosion à des copépodes (Acartia tonsa) ayant eux-mêmes été exposés à des MP.

67. Measuring individual feed efficiency and its correlations with performances and agonistic behaviours in juvenile Nile tilapia Oreochromis niloticus reared in groups

Abstract

Estimating individual feed intake of fish held in groups has long been a challenge precluding precise knowledge of the individual variation in feed efficiency (FE). In this study, we measured individual feed conversion ratio (FCR) of 100 juvenile Nile tilapia Oreochromis niloticus held in groups and estimated the correlations between FCR, performance and agonistic behavioural traits. For this purpose, fish were video-recorded to estimate the occurrence of agonistic behaviour during a fasting and a refeeding period, occurring just before the individual feed intake (FI) measurement. To measure FI, we counted the number of feed pellets eaten by each individual tilapia using video recordings made over a period of 10 days where fish were fed twice daily. Accumulated measures of FI over 11 meals were found to achieve 95% repeatability and a highly accurate FI estimate. Thus, with this approach, we can accurately investigate FI and FCR in fish. During the FI measurement period, average fish growth was 12.0 ± 3.6 g and FCR was 0.86 ± 0.20. FCR was negatively correlated with body weight gain (BWG, r = -0.62 ± 0.06) but not with FI (r = -0.02 ± 0.10). Conversely, residual feed intake (RFI) was positively correlated with FI (r = 0.60 ± 0.06) but not with BWG (r = -0.03 ± 0.10). These findings highlight the complex relationships between the main feed efficiency traits, FCR and RFI. Fish were 58% less aggressive during the fasting period compared to the refeeding period, but generally, an aggressive fish during the fasting period was also aggressive during the refeeding period. The correlations between agonistic behaviour and growth and feed conversion ratio were low, and in most cases not significant. We conclude that the hierarchical rank and agonistic behaviour should not affect Nile tilapia growth and feed conversion ratio. Consequently, a dominant fish should not necessarily be more efficient than a subordinate fish.

Abstract

International audience; Awareness of the presence of microplastics, i.e. plastic particles ranging in size from 1 μm to 5 mm, in marine and freshwaters has recently risen but detection and quantification is challenging. Furthermore, whether they pose a risk to aquatic organisms is not yet clear. Fish, for example, have been demonstrated to ingest microplastics particles but the link between quantification of uptake and impact assessment has not yet been made. In this context, we are exploring methods for particles quantification upon feeding juvenile fish with regular food and microplastics and assess whether exposure impacts behavior and growth. For quantification of uptake, we hypothesized that it is possible to analyze the fish tissue by flow cytometry in combination with viSNE, which allows the 2D clustering of particles with different features according to the fluorescence measured. Exposure experiments were carried out for up to three weeks, using different types of microplastic particles and a wide concentration range. In the flow, when particles were mixed with fish tissue, flow cytometry/viSNE was able to differentiate particles natures, numbers and sizes. About 10% of added particles were internalized by the fish from all particles that floated or settled on the bottom. Particles ingestion resulted in a slight impact on behavior. Yet, floating particles were massively incorporated by the fish and significant numbers remained even after 24h of depuration. Based on this, we are currently exploring if continuous feeding with microparticles contaminated food has consequences on juvenile fish growth. Taken together, our study demonstrates the power and limits of flow cytometry/viSNE for microplastics quantification in a complex biological matrix like fish.The setup could be extended to other types and forms of microplastics in different environmental matrices. Moreover, our study sheds light on ecological consequences that microplastics exposure might have on fish.

International audience; Microplastic (MP) occurrence is ubiquitous in fresh and marine waters and species living inEstuary are particularly exposed to this kind of pollution. In this context, the Seine River isinteresting for investigating the impacts of MP on fish, because of intense and numerous humanactivities through its watershed. The goal of this study is to evaluate the fate and impact of MPingestion using experimental exposure of the common sole, Solea solea, a representative species ofthe Seine Estuary. Larvae of fish (from 7 days post hatch (dph) to 42 dph) were exposed during 2hours to MP particles before (7 dph), during (21 and 22 dph) and after metamorphosis (41 and 42dph). Three types of industrial MP were chosen with different properties (middle size floating (27-35μg mL-1, DC: 50 ng mL-1). Post-exposurem, d=0.99), middle size sinking (27-32μg mL-1, DC: 50 ng mL-1). Post-exposurem, d=1.20) and large size sinking (63- 75μg mL-1, DC: 50 ng mL-1). Post-exposurem, d=1.20).In addition, at the last stage, fish were also exposed to natural plastic particles collected in the SeineRiver and ground into a fraction with a size less than 100 μg mL-1, DC: 50 ng mL-1). Post-exposurem (PVC, PE, PET, PS and PP testedseparately). After exposure, fish which had ingested MP were counted (Figure 1) and isolated tomonitor egestion. Solea solea ingested all types of plastics in variable (1 to more than 100)quantities and egested them on average less than 24 hours after exposure at all stages. A behaviouralchallenge, was performed on MP contaminated larvae. Behavioral responses of fish did not seem tobe affected by MP exposures before metamorphosis compared to controls but distance moveddecreased during metamorphosis for fish that had ingested MP particles. This could be explained bythe fact that before metamorphosis fish can still use yolk energy reserve and are thus less affected byMP than later on when external feeding becomes mandatory.

Abstract

We review knowledge on applications of sustained aerobic swimming as a tool to promote productivity and welfare of farmed fish species. There has been extensive interest in whether providing active species with a current to swim against can promote growth. The results are not conclusive but the studies have varied in species, life stage, swimming speed applied, feeding regime, stocking density and other factors. Therefore, much remains to be understood about mechanisms underlying findings of 'swimming‐enhanced growth', in particular to demonstrate that swimming can improve feed conversion ratio and dietary protein retention under true aquaculture conditions. There has also been research into whether swimming can alleviate chronic stress, once again on a range of species and life stages. The evidence is mixed but swimming does improve recovery from acute stresses such as handling or confinement. Research into issues such as whether swimming can improve immune function and promote cognitive function is still at an early stage and should be encouraged. There is promising evidence that swimming can inhibit precocious sexual maturation in some species, so studies should be broadened to other species where precocious maturation is a problem. Swimming performance is a heritable trait and may prove a useful selection tool, especially if it is related to overall robustness. Abstract

Adaptive phenotypic plasticity is a key component of the ability of organisms to cope with changing environmental conditions. Fish have been shown to exhibit a substantial level of phenotypic plasticity in response to abiotic and biotic factors. In the present study, we investigate the link between environmental sensitivity assessed globally (revealed by phenotypic variation in body weight) and more targeted physiological and behavioral indicators that are generally used to assess the sensitivity of a fish to environmental stressors. We took advantage of original biological material, the rainbow trout isogenic lines, which allowed the disentangling of the genetic and environmental parts of the phenotypic variance. Ten lines were characterized for the changes of body weight variability (weight measurements taken every month during 18 months), the plasma cortisol response to confinement stress (3 challenges) and a set of selected behavioral indicators. This study unambiguously demonstrated the existence of genetic determinism of environmental sensitivity, with some lines being particularly sensitive to environmental fluctuations and others rather insensitive. Correlations between coefficient of variation (CV) for body weight and behavioral and physiological traits were observed. This confirmed that CV for body weight could be used as an indicator of environmental sensitivity. As the relationship between indicators (CV weight, risk-taking, exploration and cortisol) was shown to be likely depending on the nature and intensity of the stressor, the joint use of several indicators should help to investigate the biological complexity of environmental sensitivity.

Abstract

Cette étude caractérise pour la première fois les productions sonores chez S. melanotheron et montre les différences avec O. niloticus dans un contexte d'hybridation. Les comportements de dix couples par mode d'appariement intra et interspécifique entre les deux espèces (individus de même taille 21.0 +- 1.8 cm) ont été étudiés. Pendant 15 min (à 10h, 12h et 14h) les couples ont été filmés et les sons produits enregistrés; deux stéro'ides sexuels (11- KT-Ketotestostérone et E2 (Estradiol) ont été mesurés pour chaque individu. Le premier type de son enregistré ressemble à des sons de tambour chez les deux espèces et le second ressemble à des grognements chez O. niloticus et à des 'br-r-r' chez S. melanotheron. La signature des sons agressifs et non agressifs se distingue chez S. melanotheron. Les pulses du son varient de 1 à 7 chez O. niloticus et jusqu'à 12 chez S. melanotheron. La fréquence du son chez O. niloticus est supérieure à celle de S. melanotheron dans les comportements agressifs. Pour les deux espèces, les individus d'un couple ont les mêmes périodes, durées et intensités du son. Les concentrations en 11-KT sont supérieures chez O. niloticus mâle et le nombre de sons non agressifs est positivement corrélé à la concentration en 11-KT chez cette espèce. Il n'existe pas de corrélation entre le nombre de sons et le niveau de E2 chez les femelles ni de corrélation avec la 11-KT chez S. melanotheron. L'absence de grognements et la signature des sons associés aux comportements non agressifs spécifiques à S. melanotheron pourraient constituer une barrière reproductive entre les deux espèces. Abstract

International audience; Although the use of polychlorinated biphenyls (PCBs) has been banned for several decades, they are still present in the environment. Field analyses have established correlations between exposure to PCBs and alterations in fish physiology including reproductive function and behaviour. In the present study, we performed a life-cycle exposure using zebrafish and mixtures representative of some environmental situations in terms of doses, composition and containing mainly non dioxin-like congeners. Exposure was performed through diet which is the main contamination route in the wild. We demonstrated a bioaccumulation of PCBs in males and females as well as a maternal transfer to the eggs.Several reproductive traits were altered after exposure to a PCB-contaminated diet, including a reduction in the number of fertilized eggs per spawn as well as an increase in the number of poorly fertilized spawns. This was related to modifications of ovary histology revealing a decrease of maturing follicles and an increase of atretic follicles in the ovaries of females exposed to PCBs.In addition, several behavioural traits were monitored. Fish exposed to the highest dose (equivalent to that found in the Seine Estuary) displayed an increased swimming activity during the period at the end of the night. Fish exposed to an intermediate dose (equivalent to that found in the Loire estuary) displayed behavioural disruption analogous to hyperactivity that showed some similarities to a human behavioural deficit syndrome known as attention deficit / hyperactivity disorder (ADHD), which can be observed in humans after exposure to PCBs.These results indicated that exposure to PCBs mixtures mimicking some environmental situations, can lead to a dramatic reduction in the number of offspring produced by a female over a lifetime and disrupt behaviour. Potential ecological consequences are discussed with regard to reproduction, fitness and survival.
