This study investigates the creation of multifunctional scaffolds based on alginate dialdehyde (ADA) and gelatin (GEL) incorporating ferulic acid (FA), a phytotherapeutic agent with diverse pharmacological properties, and human platelet lysate (HPL). The multimaterial hydrogels were characterized in terms of degradation/swelling behavior showing that HPL decelerates the degradation process of ADA‐GEL hydrogels in absence of FA. A FA release study confirmed that the presence of HPL does not affect the FA release ability of ADA‐GEL hydrogel. A trinitrobenzenesulfonic acid (TNBS) assay confirmed no influence of HPL on the Schiff's base formation. The addition of HPL was shown to lead to a decrease in stiffness, mechanical stability, and viscosity of ADA‐GEL‐HPL inks. This effect was also observed for ADA‐GEL‐HPL inks when investigating their three‐dimensional (3D) printing ability. Cytocompatibility tests and a 3D bioprinting study confirmed the beneficial impact of the addition of HPL on MC3T3‐E1 cells. Moreover, an antibacterial effect provided by the addition of FA was confirmed while the incorporation of HPL led to higher VEGF‐A expression by MC3T3‐E1 cells. Overall, the addition of HPL and FA to ADA‐GEL hydrogel results in a promising ink, which has potential for future biofabrication applications. [ABSTRACT FROM AUTHOR]

Despite advances in treatment modalities, bone tumour therapies still face significant challenges. Severe side effects of conventional approaches, such as chemo- and radiation therapy, result in poor survival rates and high tumour recurrence rates, which are the most common issues that need to be improved upon. The aim of this study was to evaluate the therapeutic properties of 45S5 bioactive glass (BG) for targeting bone tumours. The viability of the cells derived from osteosarcoma, chondrosarcoma, and giant cell tumours was significantly reduced in the presence of 45S5-BG. In contrast, the viability of non-malignant osteoblast-like cells, chondrocytes, and bone marrow-derived stromal cells was not or only slightly affected. While alterations to the particle surface induced by heat treatment, acid etching, or incubation in a simulated body fluid had only minor effects on cytotoxicity, reducing the particle size or sintering the material significantly improved the cytotoxic effect of 45S5-BG. Further, using a chicken chorioallantoic membrane assay, the co-transplantation of 45S5-BG resulted in a significant reduction in tumour formation in vivo. Given the known positive effects of BGs on bone regeneration, our findings suggest that 45S5-BG holds great potential for the development of new and effective bone tumour therapies, with minimal side effects on non-malignant cells and simultaneous contribution to bone healing. The 0106‐B1‐bioactive glass (BG) composition (in wt %: 37.5 SiO2, 22.6 CaO, 5.9 Na2O, 4.0 P2O5, 12.0 K2O, 5.5 MgO, and 12.5 B2O3) has demonstrated favorable processing properties and promising bone regeneration potential. The present study aimed to evaluate the biological effects of the incorporation of highly pro‐angiogenic copper (Cu) in 0106‐B1‐BG in vitro using human bone marrow‐derived mesenchymal stromal cells (BMSCs) as well as its in vivo potential for bone regeneration. CuO was added to 0106‐B1‐BG in exchange for CaO, resulting in Cu‐doped BG compositions containing 1.0, 2.5 and 5.0 wt % CuO (composition in wt %: 37.5 SiO2, 21.6/ 20.1/17.6 CaO, 5.9 Na2O, 4.0 P2O5, 12.0 K2O, 5.5 MgO, 12.5 B2O3, and 1.0/ 2.5/ 5.0 CuO). In vitro, the BGs' impact on the viability, proliferation, and growth patterns of BMSCs was evaluated. Analyses of protein secretion, matrix formation, and gene expression were used for the assessment of the BGs' influence on BMSCs regarding osteogenic differentiation and angiogenic stimulation. The presence of Cu improved cytocompatibility, osteogenic differentiation, and angiogenic response when compared with unmodified 0106‐B1‐BG in vitro. In vivo, a critical‐size femoral defect in rats was filled with scaffolds made from BGs. Bone regeneration was evaluated by micro‐computed tomography. Histological analysis was performed to assess bone maturation and angiogenesis. In vivo effects regarding defect closure, presence of osteoclastic cells or vascular structures in the defect were not significantly changed by the addition of Cu compared with undoped 0106‐B1‐BG scaffolds. Hence, while the in vitro properties of the 0106‐B1‐BG were significantly improved by the incorporation of Cu, further evaluation of the BG composition is necessary to transfer these effects to an in vivo setting. Next to the choice of a material, the crosslinking characteristics play an important role in the development of hydrogels for applications in the biomedical field. By decreasing crosslinking time or concentration one can reduce the density of the network and hence the stiffness of the hydrogel. However, at the same time in many cases the stability of the matrix is significantly reduced, leading to a rapidly degrading hydrogel structure. In this study, we evaluate and compare two different crosslinking methods while maintaining the material chemistry. The hydrogel system evaluated consisted of oxidized hyaluronic acid and gelatin, and it was crosslinked with microbial transglutaminase. We found that by in situ crosslinking (CL) instead of post CL we can significantly increase the stability of the hydrogel while maintaining the matrix stiffness. Encapsulated embryonic mouse fibroblasts showed an increased viability in in situ crosslinked samples. These findings indicate that it is possible to change hydrogel parameters independently, even if they are otherwise interrelated. [ABSTRACT FROM AUTHOR]

Doping of brushite cements with metal ions can entail many positive effects on biological and physicochemical properties. Cu2+ ions are known to exhibit antibacterial properties and can additionally have different positive effects on cells as trace elements, whereas high Cu2+ concentrations are cytotoxic. For therapeutical applications of bone cement, a combination of good biocompatibility and sufficient mechanical properties is required. Therefore, the aim of this study was to investigate different physicochemical and biological aspects, relevant for application, of a brushite cement with Cu2+‐doped β‐tricalcium phosphate, monocalcium phosphate monohydrate and phytic acid as setting retarder. Additionally, the ion release was compared with a cement with citric acid as setting retarder. The investigated cements showed good injectability coefficients, as well as compressive strength values sufficient for application. Furthermore, no antibacterial effects were detected irrespective of the Cu2+ concentration or the bacterial strain. The cell experiments with eluate samples showed that the viability of MC3T3‐E1 cells tended to decrease with increasing Cu2+ concentration in the cement. It is suggested that these biological responses are caused by the difference in the Cu2+ release from the hardened cement depending on the solvent medium. Furthermore, the cements showed a steady release of Cu2+ ions to a lesser extent in comparison with a cement with citric acid as setting retarder, where a burst release of Cu2+ was observed. In conclusion, despite the anticipated antibacterial effect of Cu2+‐doped cements was lacking and mammalian cell viability was slightly affected, Cu2+‐concentrations maintained the physicochemical properties as well as the compressive strength of cements and the slow ion release from cements produced with phytic acid is considered advantageous compared to citric acid‐based formulations. Biofabrication techniques enable the performance of bioinspired three‐dimensional (3D) matrices resembling primary tumors. To validate their reliability, embedded cells may express complex biophysical responses. Among others, the emergence of tumor heterogeneity and the generation of Polyploid Giant Cancer Cells (PGCC), as a result of the mechanical stress, are two of the most challenging hallmarks to resemble in vitro. Here, these phenomena are studied in cells cultured on two‐dimensional (2D) flasks, in 3D spheroids, or immobilized within 3D polymer‐based tumor‐like microcapsules. These results show that cells cultured in 3D microcapsules exhibited an enhanced biomechanical heterogeneity, a higher number of PGCC, and an increased exertion of cell‐matrix attachment forces with respect to the other two experimental conditions. Additionally, cells isolated from tumor‐like microcapsules redistribute and align the cytoplasmatic protein Caveolin‐1, and upregulate markers involved in cell proliferation (i.e., Ki67), metastasis (i.e., TGF‐β1, TGF‐β‐R2), and epithelial to mesenchymal transition, to name a few. These hallmarks are barely described in the past as a result of the confinement and mechanical stress. Thus, in this work it is demonstrated that both the mechanical stress and confinement are required to stimulate cell polyploidy and biomechanical heterogeneity, which can be easily addressed by immobilizing breast cancer cells in tumor‐like microcapsules. This perspective article delves into the transformative potential of alkali‐activated materials, acid‐activated materials, and geopolymers in mitigating climate change and market challenges. To harness the benefits of these materials, a comprehensive strategy is proposed. This strategy aims to integrate these materials into existing construction regulations, facilitate certification, and promote market access. Emphasizing research and innovation, the article advocates for, increased funding to refine the chemistry and production of these materials, prioritizing low‐cost alternatives and local waste materials. Collaboration between academia and industry is encouraged to expedite technological advances and broaden applications. This article also underscores the need to develop economic and business models emphasizing the long‐term benefits of these materials, including lower life‐cycle costs and reduced environmental impact. Incentivizing adoption through financial mechanisms like tax credits and subsidies is suggested. The strategy also includes scaling up production technology, fostering industrial collaboration for commercial viability, and developing global supply chains. Educational programs for professionals and regulators are recommended to enhance awareness and adoption. Additionally, comprehensive life‐cycle assessments are proposed to demonstrate environmental benefits. The strategy culminates in expanding the applications of these materials beyond construction, fostering international collaboration for knowledge sharing, and thus positioning these materials as essential for sustainable construction and climate change mitigation.
