53. Demonstration of resistance to satyrization behavior in Aedes aegypti from La Réunion island

Abstract

Aedes aegypti and Aedes albopictus are competent vectors of arboviruses such as dengue and chikungunya viruses which co-exist in some areas, including La Réunion island, Indian Ocean. A type of reproductive interference called satyrization has been described in sympatric species where dominant species mating fails to produce hybrids and thus reduces the fitness and tends to control the spread of the other species. Here, we investigated satyrization in laboratory experiments to provide insights on the potential impact on Ae. aegypti of a control campaign including a sterile insect technique component against Ae. albopictus. Different mating crosses were used to test sympatric, conspecific-interspecific and allopatric effects of irradiated and non-irradiated male Ae. albopictus on female Ae. aegypti, including in a situation of skewed male ratio. Our results suggest that there was only a low level of satyrization between sympatric populations of Ae. aegypti and Ae. albopictus colonized from La Réunion island. A male Ae. albopictus to female Ae. aegypti ratio of 3:1 did not increase the level of satyrization. Female Ae. aegypti previously mated to male Ae. albopictus were not prevented from being inseminated by conspecific males. A satyrization effect was not seen between allopatric Ae. albopictus and Ae. aegypti strains from La Réunion Island either. The tested Ae. aegypti strain from La Réunion island has therefore developed full resistance to satyrization and so releasing sterile male Ae. albopictus may not suppress Ae. aegypti populations if an overflooding of irradiated male Ae. albopictus leads to similar results. The management strategy of two competent species in a sympatric area is discussed.

Abstract

The production of a large number of mosquitoes of high biological qualities and reliable sex sorting before release are key challenges when applying the sterile insect technique as part of an area-wide integrated pest management approach. There is a need to fully evaluate the production capacity of the equipment developed in order to plan and maintain a daily production level for large-scale operational release activities. This study aimed to evaluate the potential use of the FAO/IAEA larval rearing unit for Aedes aegypti and the subsequent female contamination rate after sex sorting with a Fay–Morlan glass separator. Trays from each rack were tilted and their contents sorted either for each individual tray or after mixing the content of all trays from the rack. The pupal production and the female contamination rate were estimated with respect to day of collection, position of the tray, type of pupae collection, and sorting operator. Results showed significant daily variability of pupal production and female contamination rate, with a high male pupal production level achieved on the second day of collection and estimated female contamination of male pupae reached around 1%. Neither tray position nor type of pupae collection affected the pupal production and female contamination rate. However, the operator had a significant effect on the female contamination rate. These results highlight the need to optimize pupal production at early days of collection and to develop a more effective and automated method of sex separation.

Abstract

The mass production of mosquitoes is becoming more wide-spread due to the increased application of the sterile insect technique (SIT) and other genetic control programmes. Due to the variable availability and high cost of the bovine liver powder (BLP) constituent of many current larval diets, there is an urgent demand for new ingredients in order to support sustainable and efficient mosquito production while reducing rearing cost, without affecting the quality of the insects produced. Two black soldier fly (BSF) powder-based diet formulations (50% tuna meal, 35% BSF powder, 15% brewer’s yeast and 50% tuna meal + 50% BSF powder) were tested for their suitability to support the development of Aedes aegypti and Ae. albopictus mosquitoes in mass-rearing conditions. Overall, the results indicate that the use of the BSF powder did not negatively impact the development and quality of the produced insects in terms of time to pupation, adult production and male flight ability. Furthermore, depending on the species and diet formulations, there were improvements in some parameters such as female body size, egg production, egg hatch rate and male longevity. BSF powder is a valuable ingredient that can effectively replace costly BLP for the mass production of high quality Ae. aegypti and Ae. albopictus mosquitoes. Both diet formulations can be used for Ae. aegypti showing high plasticity to nutrition sources. However, for Ae. albopictus we recommend the combination including brewer’s yeast.

Abstract

Balancing process efficiency and adult sterile male biological quality is one of the challenges in the success of the sterile insect technique (SIT) against insect pest populations. For the SIT against mosquitoes, many stress factors need to be taken into consideration when producing sterile males that require high biological quality to remain competitive once released in the field. Pressures of mass rearing, sex sorting, irradiation treatments, packing, transport and release including handling procedures for each step, add to the overall stress budget of the sterile male post-release. Optimizing the irradiation step to achieve maximum sterility while keeping off-target somatic damage to a minimum can significantly improve male mating competitiveness. It is therefore worth examining various protocols that have been found to be effective in other insect species, such as dose fractionation. A fully sterilizing dose of 70 Gy was administered to Aedes aegypti males as one acute dose or fractionated into either two equal doses of 35 Gy, or one low dose of 10 Gy followed by a second dose of 60 Gy. The two doses were separated by either 1- or 2-day intervals. Longevity, flight ability, and mating competitiveness tests were performed to identify beneficial effects of the various treatments. Positive effects of fractionating dose were seen in terms of male longevity and mating competitiveness. Although applying split doses generally improved male quality parameters, the benefits may not outweigh the added labor in SIT programmes for the management of mosquito vectors.

Abstract

International audience; Spatio-temporally heterogeneous environments may lead to unexpected population dynamics. Knowledge is needed on local properties favouring population resilience at large scale. For pathogen vectors, such as tsetse flies transmitting human and animal African trypanosomosis, this is crucial to target management strategies. We developed a mechanistic spatio-temporal model of the age-structured population dynamics of tsetse flies, parametrized with field and laboratory data. It accounts for density- and temperature-dependence. The studied environment is heterogeneous, fragmented and dispersal is suitability-driven. We confirmed that temperature and adult mortality have a strong impact on tsetse populations. When homogeneously increasing adult mortality, control was less effective and induced faster population recovery in the coldest and temperature-stable locations, creating refuges. To optimally select locations to control, we assessed the potential impact of treating them and their contribution to the whole population. This heterogeneous control induced a similar population decrease, with more dispersed individuals. Control efficacy was no longer related to temperature. Dispersal was responsible for refuges at the interface between controlled and uncontrolled zones, where resurgence after control was very high. The early identification of refuges, which could jeopardize control efforts, is crucial. Abstract

International audience; Aedes albopictus and Aedes aegypti are invasive mosquito species that impose a substantial risk to human health. To control the abundance and spread of these arboviral pathogen vectors, the sterile insect technique (SIT) is emerging as a powerful complement to most commonly-used approaches, in part, because this technique is ecologically benign, specific, and non-persistent in the environment if releases are stopped. Because SIT and other similar vector control strategies are becoming of increasing interest to many countries, we offer here a pragmatic and accessible ‘roadmap’ for the pre-pilot and pilot phases to guide any interested party. This will support stakeholders, non-specialist scientists, implementers, and decision-makers. Applying these concepts will ensure, given adequate resources, a sound basis for local field trialing and for developing experience with the technique in readiness for potential operational deployment. This synthesis is based on the available literature, in addition to the experience and current knowledge of the expert contributing authors in this field. We describe a typical path to successful pilot testing, with the four concurrent development streams of Laboratory, Field, Stakeholder Relations, and the Business and Compliance Case. We provide a graphic framework with criteria that must be met in order to proceed.

Abstract

Geographic information systems (GIS) and spatial modelling are crucial for designing, implementing, and optimizing area-wide programmes of insect and/or disease control. This chapter provides a basic introduction to the science of GIS, Global Positioning System (GPS), satellite remote sensing (RS), and spatial modelling, and reviews the principal ways in which these technologies can be used to assist various stages of development of the sterile insect technique (SIT) as part of area-wide integrated pest management (AW-IPM) programmes — from the selection of project sites, and feasibility assessments and planning of pre-intervention surveys, to the monitoring and analysis of insect suppression programmes, and the release of sterile insects. Potential barriers to the successful deployment of GIS tools are also discussed.

In addition to the destruction of breeding sites (prevention) and mass trapping, methods based on the sterile insect technique (SIT), the autodissemination of pyriproxyfen (ADT), and a fusion of elements from both of these known as boosted SIT (BSIT), are being developed to meet the urgent need for effective vector control. However, the comparative potential of these methods has yet to be explored in different environments. This is needed to propose and integrate informed guidelines into sustainable mosquito management plans. We extended a weather-dependent model of Aedes albopictus population dynamics to assess the effectiveness of these different vector control methods, alone or in combination, in a tropical (Reunion island, southwest Indian Ocean) and a temperate (Montpellier area, southern France) climate. Our results confirm the potential efficiency of SIT in temperate climates when performed early in the year (mid-March for northern hemisphere). In such a climate, the timing of the vector control action was the key factor in its success. In tropical climates, the potential of the combination of methods becomes more relevant. BSIT and the combination of ADT with SIT were twice as effective compared to the use of SIT alone.

Abstract

International audience; Tsetse flies (Diptera: Glossinidae) are the main vectors of animal and human trypanosomoses in Africa. The Sterile Insect Technique (SIT) has proven effective in controlling tsetse flies when applied to isolated populations but necessitates the production of large numbers of sterile males. A new approach, called boosted SIT, combining SIT with the contamination of wild females by sterile males coated with biocides has been proposed for large-scale control of vector populations. The aim of the study was to evaluate this new approach using pyriproxyfen on the riverine species Glossina palpalis gambiensis (Vanderplank, 1949) in the laboratory. The contamination dose and persistence of pyriproxyfen on sterile males, the impact of pyriproxyfen on male survival, and the dynamics of pyriproxyfen transfer from a sterile male to a female during mating, as well as the impact of pyriproxyfen on pupal production and adult emergence, were evaluated in the laboratory. For this purpose, a method of treatment by impregnating sterile males with a powder containing 40% pyriproxyfen has been developed. The results showed that the pyriproxyfen has no impact on the survival of sterile males. pyriproxyfen persisted on sterile males for up to 10 days at a dose of 100 ng per fly. In addition, the horizontal transfer of pyriproxyfen from a treated sterile male to a female during mating could be measured with an average of 50 ng of pyriproxyfen transferred. After contacts without mating, the average quantity transferred was more than 10 ng. Finally, the pyriproxyfen powder was very effective on G. p. gambiensis leading to 0% emergence of the pupae produced by contaminated females. These promising results must be confirmed in the field. A large-scale assessment of this boosted pyriproxyfen-based SIT approach will be carried out against tsetse flies in Senegal (West Africa). On the African continent, tsetse flies (Diptera: Glossinidae) are the main vectors of the parasites responsible for Human African Trypanosomosis (HAT), or sleeping sickness, and Animal African Trypanosomosis (AAT), also called Nagana 1. These diseases caused by Trypanosomatidae belong to the group of neglected tropical diseases (NTDs), occurring in developing areas 2. Sub-Saharan African countries suffer from a significant impact of NTDs on public health and economic development 3. Glossina palpalis gambiensis (Vanderplank, 1949) is one of the main vectors of the HAT parasite in West Africa and is responsible for the persistence of many residual outbreaks of this disease in forest and savannah areas 4. There is still no vaccine against HAT and curative treatments are difficult to access for the most exposed and vulnerable populations 5. Glossina palpalis gambiensis is also involved in the transmission of AAT, which affects livestock and reduces animal production thus limiting the availability of food resources 6,7. Abstract

International audience; The Sterile Insect Technique (SIT), used to control insect populations, consists of releasing males sterilized by ionizing radiations. Wild females that mate with these males can no longer produce viable offspring, which may drives the population decline. Although this method has proved its efficiency, its effect may be more limited for fast-reproducing large-population species, such Aedes albopictus. A novel approach, named ”boosted SIT” has been designed to strengthen the SIT technique: It consists of coating sterile males with a biocide that will be transferred to the mated females, which will then contaminate the oviposition sites. This study is aimed at exploring demographic effects of both techniques (SIT and boosted SIT) through their inclusion in a weather-driven abundance model of the Aedes albopictus population dynamics in the geographical context of La Reunion Island. Sensitivity analysis showed that the date to start the release, as well as the quantity of sterile males released and their competitiveness, are of key importance for both control methods. According to our results, boosted SIT allows 1) Increasing the effectiveness of the SIT when the sterile males released are of medium quality in terms of competitiveness, and 2) extending the optimal window to start the control period.

Abstract

Le projet REVOLINC (ERC Consolidator grant no. 682387) vise à développer des stratégies innovantes et respectueuses de l'environnement pour lutter contre trois espèces d'insectes nuisibles (mouche tsé-tsé, moustique tigre et mouche méditerranéenne des fruits), dont les deux dernières ont envahi !'Europe. Ce projet repose sur le concept de renforcer la technique de l'insecte stérile (TIS) par le transfert spécifique de biocides des males stériles a leurs homologues femelles sauvages pendant l'accouplement Nous voulons améliorer la TIS qui consiste à élever en masse des insectes mâles, à les stériliser par irradiation et à les libérer dans l'environnement, où ils sont en compétition avec leurs homologues sauvages non stériles pour se reproduire. La TIS " boostée " consiste à s'assurer que les mâles stériles traités par un biocide et relâchés dans l'environnement transmettent également ce biocide aux femelles afin de les tuer ou de tuer leur progéniture. Dans Revolinc, nous étudions ce concept à l'aide de deux biocides : un analogue de l'hormone juvénile (pyriproxifène) et un densovirus spécifique (AalDV2) contre le moustique tigre. Le pyriproxifène sera également teste contre la mouche tsé-tsé et la mouche des fruits méditerranéenne. Les évaluations sont réalisées au laboratoire à Montpellier, France au sein du Vectopole de l'IRD et en conditions contrôlées sur le terrain à Valence, Espagne et a La Réunion. Par modélisation, nous étudions les impacts relatifs de la TIS et de la TIS renforcée sur la dynamique des populations ciblées. Enfin, nous comparerons la réponse évolutive de la population d'insecte cible a ces différentes pressions de contrôle et pour différents biocides et stratégies de reproduction. Ce projet s'inscrit dans la nécessité de développer des méthodes alternatives aux insecticides employés dans la lutte contre les insectes vecteurs ou ravageurs et qui soulèvent des inquiétudes légitimes quant à leur toxicité pour la santé et l'environnement, tout en améliorant la rentabilité de la TIS.

Abstract

The sterile insect technique (SIT) is an environmentally safe and proven technology to suppress wild populations. To further advance its utility, a novel CRISPR-based technology termed precision guided SIT (pgSIT) is described. PgSIT mechanistically relies on a dominant genetic technology that enables simultaneous sexing and sterilization, facilitating the release of eggs into the environment ensuring only sterile adult males emerge. Importantly, for field applications, the release of eggs will eliminate burdens of manually sexing and sterilizing males, thereby reducing overall effort and increasing scalability. Here, to demonstrate efficacy, we systematically engineer multiple pgSIT systems in Drosophila which consistently give rise to 100% sterile males. Importantly, we demonstrate that pgSIT-generated sterile males are fit and competitive. Using mathematical models, we predict pgSIT will induce substantially greater population suppression than can be achieved by currently-available self-limiting suppression technologies. Taken together, pgSIT offers to potentially transform our ability to control insect agricultural pests and disease vectors., Sterile Insect Technique (SIT) is used to suppress wild populations. Here the authors integrate CRISPR-based technology and SIT to develop a precision guided SIT (pgSIT), and demonstrate its proof-of-principle by generating 100% sterile males.
