We present results on the spin and quark content of the nucleon using $N_f=2$ twisted mass clover-improved fermion simulations with a pion mass close to its physical value. We use recently developed methods to obtain accurate results for both connected and disconnected contributions. We provide results for the axial charge, quark and gluon momentum fraction as well as the light, strange and charm $\sigma$-terms., Comment: 7pages, 7 figures. Physical review letters 119(14), 142002 (2017). doi:10.1103/PhysRevLett.119.142002, We determine within lattice QCD the nucleon spin carried by valence and sea quarks and gluons.The calculation is performed using an ensemble of gauge configurations with two degenerate light quarks with mass fixed to approximately reproduce the physical pion mass.We find that the total angular momentum carried by the quarks in the nucleon is $J_{u+d+s}$ = 0.408(61)$_{stat}$(48)$_{syst}$ and the gluon contribution is $J_g$ = 0.133(11)$_{stat}$(14)$_{syst}$, giving a total of $J_N$ = 0.54(6)$_{stat}$(5)$_{syst}$ that is consistent with the spin sum. For the quark intrinsic spin contribution, we obtain $\frac 1{2} \Delta \Sigma _{u+d+s}$ 0.201(17)$_{stat}$(5)$_{syst}$. All quantities are given in the modified minimal subtraction scheme at 2 GeV. The proposal for a new General Data Protection Regulation has been billed as a harbinger of increased harmonisation, better enforcement and modernised rules within the area of data protection law. Through an analysis of several central elements in the draft Regulation—and European data protection law in general—as well as an assessment of the practical implications the proposal is likely to have if adopted, this chapter challenges whether the proposal will be able to deliver the harmonised rules that have been promised. It focuses particularly on the proposed regulations scope of application, its legal architecture, the use of discretionary provisions and related issues.It is argued that the proposal not only fails to address the root causes of why the current data protection directive (Directive 95/46) failed to bring about harmonisation and effective rules, but also looks set to transplant them into the new regulation.

Compared to a previous calculation by us, the errors are reduced by a factor of about 2.5. Moreover, we present our first results for the polarized sector of the proton, which indicate an asymmetry in the proton sea in favor of the $u$ antiquarks for the case of helicity distributions, and an asymmetry in favor of the $d$ antiquarks for the case of transversity distributions., Published by SISSA, Trieste

In this talk we would like to report on recent non-perturbative results and ongoing efforts to explore the proton spin from lattice QCD. We present results for the relevant generalized form factors from gauge field ensembles that feature a physical value of the pion mass. These generalized form factors can be used to determine the total spin and angular momentum carried by the quarks. It was shown that racemic (±)-2 [1'-benzyl-3-(3-fluoropropyl)-3H-spiro[[2]benzofuran-1,4'-piperidine], WMS-1813] represents a promising positron emission tomography (PET) tracer for the investigation of centrally located σ(1) receptors. To study the pharmacological activity of the enantiomers of 2, a preparative HPLC separation of (R)-2 and (S)-2 was performed. The absolute configuration of the enantiomers was determined by CD-spectroscopy together with theoretical calculations of the CD-spectrum of a model compound. In receptor binding studies with the radioligand [(3)H]-(+)-pentazocine, (S)-2 was thrice more potent than its (R)-configured enantiomer (R)-2. The metabolic degradation of the more potent (S)-enantiomer was considerably slower than the metabolism of (R)-2. The structures of the main metabolites of both enantiomers were elucidated by determination of the exact mass using an Orbitrap-LC-MS system. These experiments showed a stereoselective biotransformation of the enantiomers of 2.

A series of various N-substituted 3-(3-fluoropropyl)-3H-spiro[[2]benzofuran-1,4'-piperidines] (7) has been synthesized. In receptor binding studies, the N-benzyl derivative 7a (WMS-1813) revealed extraordinarily high sigma(1) receptor affinity (K(i) = 1.4 nM) and excellent sigma(1)/sigma(2) selectivity (600 fold). In vitro biotransformation of 7a with rat liver microsomes led to three main metabolites. N-Debenzylation was inhibited by introduction of an N-phenylethyl residue (7 g). The PET tracer [(18)F]7a was synthesized by nucleophilic substitution of the tosylate 13 with K[(18)F]F-K222-carbonate complex. The decay corrected radiochemical yield of [(18)F]7a was 35-48% with a radiochemical purity of99.5% and a specific activity of 150-238 GBq/micromol. The radiotracer properties were evaluated in female CD-1 mice by organ distribution and ex vivo brain autoradiography. The radiotracer uptake in the brain was fast and sufficient, with values of approximately 4% injected dose per gram. Target specificity of [(18)F]7a was validated in blocking studies by preapplication of haloperidol, and significant reduction in the uptake of radioactivity was observed in the brain and peripheral organs expressing sigma(1) receptors.

Objectives The pharmacology and metabolism of the potent σ1 receptor ligand 1′-benzyl-3-methoxy-3H-spiro[[2]benzofuran-1,4′-piperidine] were evaluated. Methods The compound was tested against a wide range of receptors, ion channels and neurotransmitter transporters in radioligand binding assays. Analgesic activity was evaluated using the capsaicin pain model. Metabolism by rat and human liver microsomes was investigated, and the metabolites were identified by a variety of analytical techniques. Key findings 1′-Benzyl-3-methoxy-3H-spiro[[2]benzofuran-1,4′-piperidine] (compound 1) is a potent σ1 receptor ligand (Ki 1.14 nM) with extraordinarily high σ1/σ2 selectivity (>1100). It was selective for the σ1 receptor over more than 60 other receptors, ion channels and neurotransmitter transporters, and did not interact with the human ether-a-go-go-related gene (hERG) cardiac potassium channel. Compound 1 displayed analgesic activity against neuropathic pain in the capsaicin pain model (53% analgesia at 16 mg/kg), indicating that it is a σ1 receptor antagonist. It was rapidly metabolised by rat liver microsomes. Seven metabolites were unequivocally identified; an N-debenzylated metabolite and a hydroxylated metabolite were the major products. Pooled human liver microsomes formed the same metabolites. Studies with seven recombinant cytochrome P450 isoenzymes revealed that CYP3A4 produced all the metabolites identified. The isoenzyme CYP2D6 was inhibited by 1 (IC50 88 nM) but did not produce any metabolites. Conclusions 1′-Benzyl-3-methoxy-3H-spiro[[2]benzofuran-1,4′-piperidine] is a potent and selective σ1 receptor antagonist, which is rapidly metabolised. Metabolically more stable σ1 ligands could be achieved by stabilising the N-benzyl substructure.

Several 3H-spiro[[2]benzofuran-1,4'-piperidines] bearing a p-fluorobenzyl residue at the N-atom and various substituents in position 3 of the benzofuran system were synthesized. The crucial reaction steps are the addition of a lithiated benzaldehyde derivative to the p-fluorobenzylpiperidone 5 and the BF(3).OEt(2) catalyzed substitution of the methoxy group of 2a by various nucleophiles. Structure-affinity relationship studies revealed that compounds with two protons (2d), a methoxy group (2a), and a cyano group (2e) in position 3 possess subnanomolar sigma(1) affinity (K(i)=0.18 nM, 0.79 nM, 0.86 nM) and high selectivity against the sigma(2) subtype. The metabolites of 2a, 2d, and 2e, which were formed upon incubation with rat liver microsomes, were identified. Additionally, the rate of metabolic degradation of 2a, 2d, and 2e was determined and compared with the degradation rate of the non-fluorinated spirocyclic compound 1. For the synthesis of the potential PET tracers [(18)F]2a and [(18)F]2e two different radiosynthetic approaches were followed.

This essay presents a small case study that traces the history of American Jewish scholarship in search of its language and its place in the American world before 1945. Instead of dealing with the scholarship of languages, the author focuses on two languages of Jewish scholarship that emerged during the nineteenth century, that had and still have, albeit in different degrees, a deep impact on modern Jewish Studies. The author does not elaborate on theories of multilinguism, but rather explores the process of transformation that took place when Jewish emigration to the United States in the nineteenth and twentieth centuries led to the transplantation of Wissenshaft des Judentums into a totally different cultural and linguistic context.

Inorganic and organic salts, amino acids, sugars, and phosphate esters (concentrations usually 25 mM) were fed via the transpiration stream through the petiole into detached leaves of Lepidium sativum and Solanum tuberosum. While water was lost by transpiration, solutes did not accumulate in the apoplast. Uptake into leaf cells was indicated by stimulation of respiration and by changes of membrane potential and apoplastic pH. Apoplastic alkalinization (followed by transient acidification) and membrane depolarization (followed by repolarization) indicated energization of transport at the expense of the proton motive force (PMF) across the plasma membrane in all examined cases. Loss of ATP in the symplast during proton extrusion into the apoplast by the plasmalemma ATPase is thought to be responsible for stimulation of respiration. Even unphysiological solutes such as β-morpholinoethane sulfonate (Mes), or potentially toxic salts such as CdCl2 or AlCl3, and metabolites involved in energy conservation such as AMP and NAD, were readily transported into leaf cells at the expense of metabolic energy. At the maximum stimulation of CO2 release by D-serine (which is unlikely to be metabolized) respiration exceeded basal respiration by an average of 33%. Occasionally, and with other solutes, basal respiration was almost doubled. The ratio of transported solute to released extra CO2 was 6.9 ± 1.1 (n = 11) in the case of D-serine. From this, maximum energized transport of D-serine was calculated to be close to 500 nmol/(m2 leaf area s). Solute/CO2 ratios similar to those observed with D-serine were also obtained for sucrose. Lower ones were observed with organic solutes such as L-glycine, pyruvate, malate or citrate where secondary metabolic conversions may contribute to CO2 release.

Solutions of sucrose, glucose, raffinose, and stachyose were fed via the petiole to detached leaves of plant species known to transfer sugars during photosynthesis into the phloem using either the apoplastic or the symplastic pathway of phloem loading. Symplastic phloem loaders, which translocate raffinose-type oligosaccharides and sucrose in the phloem, and apoplastic plants, translocating exclusively sucrose, were selected for this study. As the sugars arrived with the transpiration stream in the leaf blade within little more than a minute, dark respiration increased. Almost simultaneously, fluorescence of a potential-indicating dye, which had been infiltrated into the leaves, indicated membrane depolarization. Another fluorescent dye used to record the apoplastic pH revealed apoplastic alkalinization that occurred with a slight lag phase after respiration and membrane depolarization responses. Occasionally, alkalinization was preceded by transient apoplastic acidification. Whereas membrane depolarization and apoplastic acidification are interpreted as initial responses of the proton motive force across the plasma membrane to the advent of sugars in the leaf apoplast, the following apoplastic alkalinization showed that sugars were taken up from the apoplast into the symplast in cotransport with protons. This was true not only for glucose and sucrose, but also for raffinose and stachyose. Similar observations were made for sugar uptake not only in leaves of plants known to export sugars by symplastic phloem loading but also of plants using the apoplastic pathway. Increased respiration during sugar uptake revealed tight coupling between respiratory ATP production and ATP consumption by proton-translocating ATPase of the plasma membrane, which exports protons into the apoplast, thereby compensating for the proton loss in the apoplast when protons are transported together with sugars into the symplast. The extent of stimulation of respiration by sugars indicated that sugar uptake was not limited to phloem tissue. Ratios of the extra CO2 released during sugar uptake to the amounts of sugars taken up were variable, but lowest values were lower than 0.2. When a ratio of 0.2 is taken as a basis to calculate rates of sugar uptake from observed maxima of sugar-dependent increases in respiration, rates of sugar uptake approached 350 nmol/(m2 leaf surface s). Sugar uptake rates were half-saturated at sugar concentrations in the feeding solutions of about 10–25 mM indicating a low in vivo affinity of sugar uptake systems for sugars.

Dissipation of light energy was studied in the moss Rhytidiadelphus squarrosus (Hedw.) Warnst., and in leaves of Spinacia oleracea L. and Arabidopsis thaliana (L.) Heynh., using chlorophyll fluorescence as an indicator reaction. Maximum chlorophyll fluorescence of 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU)-treated spinach leaves, as produced by saturating light and studied between and -20 degrees C, revealed an activation energy delta E of 0.11 eV. As this suggested recombination fluorescence produced by charge recombination between the oxidized primary donor of photosystem II and reduced pheophytin, a mathematical model explaining fluorescence, and based in part on known characteristics of primary electron-transport reactions, was developed. The model permitted analysis of different modes of fluorescence quenching, two localized in the reaction center of photosystem II and one in the light-harvesting system of the antenna complexes. It predicted differences in the relationship between quenching of variable fluorescence Fv and quenching of basal, so-called F0 fluorescence depending on whether quenching originated from antenna complexes or from reaction centers. Such differences were found experimentally, suggesting antenna quenching as the predominant mechanism of dissipation of light energy in the moss Rhytidiadelphus, whereas reaction-center quenching appeared to be important in spinach and Arabidopsis. Both reaction-center and antenna quenching required activation by thylakoid protonation but only antenna quenching depended on or was strongly enhanced by zeaxanthin. De-protonation permitted relaxation of this quenching with half-times below 1 min. More slowly reversible quenching, tentatively identified as so-called qI or photoinhibitory quenching, required protonation but persisted for prolonged times after de-protonation. It appeared to originate in reaction centers.

Kinetics of the dark relaxation of variable chlorophyll fluorescence, Fv, were studied after brief illumination of dark-adapted barley leaves in order to understand the rapid reversibility of pulse-induced fluorescence increases, which is observed even when fast linear electron transport to an external electron acceptor is not possible. Four kinetically distinct components were observed which reveal complexity in the oxidation of the reduced primary quinone acceptor of Photosystem II, QA −: the slowest component accounted for 4–5% of maximal Fv and had a life-time of several seconds. It is suggested to represent a minor population of inactive Photosystem II centers. The other three components displayed first-order kinetics with half-time of 6–8 ms (`fast' component), 60–80 ms (`middle' component) and 650–680 ms (`slow' component). The fast component dominated Fv when methyl viologen or far-red light accelerated oxidation of plastohydroquinone. It shows rapid oxidation of QA − during electron flow to plastoquinone commensurate with maximum linear electron flow through the electron transport chain. The other two components were observed under conditions of restricted electron flow and excessive reduction of electron carriers. Unexpectedly, the slow component, which is interpreted to reflect the recombination between QA − and an intermediate on the oxidizing side of Photosystem II, saturated already at low irradiances of actinic light when plastoquinone was not yet strongly reduced suggesting that dark-adaptation of leaves results not only in the loss of activity of light-regulated enzymes of the carbon cycle but affects also electron flow from QA − to plastoquinone. KCN poisoning or high temperature treatment of leaves produced a nonexponential pattern of slow Fv relaxation. This effect was largely (heat treatment) or even completely (KCN) abolished by far-red light.

Summary Leaves of several plant species were illuminated either in air or in a gas atmosphere with reduced oxygen content, and photosynthesis and transpiration were recorded after brief exposure to high concentrations of SO2. Inhibition of photosynthesis by SO2 was similar in air and at reduced oxygen concentrations or, in some experiments, even larger under low oxygen. Recovery from inhibition was always faster in high than in low oxygen. In leaves of Pelargonium zonale, stomata remained open or closed only slowly after fumigation, whereas in spinach and potato leaves stomata closed rapidly under the influence of high SO2. After cessation of fumigation, stomata always reopened. pH-indicating fluorescent dyes were employed to monitor SO2-dependent pH changes in the apoplast and the cytosol of leaf tissue. In the cytosol, acidification was weaker in the dark than in the light. In the light, it was stronger in 21 % oxygen than in 1% oxygen. Cytosolic acidification could not fully be attributed to the hydration and oxidation of SO2 but was also caused by a transient breakdown of transmembrane proton gradients that resulted in the influx of acid from acidic leaf compartments into the cytosol. This was concluded from opposite pH changes in the cytosol and the apoplast under the influence of SO2. Even chloroplasts were transiently acidified by SO2 as shown by the kinetics of 505 nm absorption, which reflect the pH-sensitive interconversion of violaxanthin to zeaxanthin. Differences in the kinetics of SO2-dependent inhibition of photosynthesis and of cytosolic acidification in high and low oxygen suggest combined nucleophilic attack of sulfite, and of radicals generated in the light, on sensitive cellular constituents. However, the reversibility of photosynthesis inhibition and of acidification, particularly in the presence of air levels of oxygen, demonstrates fast repair. The observations suggest that detrimental effects on plants caused by long-term exposure to ambient concentrations of SO2 which are several hundredfold lower than used in the present investigation, can neither be explained by radical damage nor by immediate effects of cellular acidification. Both radical detoxification and pH regulation are highly effective in healthy plants.

In illuminated intact spinach chloroplasts, warming to and beyond 40 °C increased the proton permeability of thylakoids before linear electron transport through Photosystem II was inhibited. Simultaneously, antimycin A-sensitive cyclic electron transport around Photosystem II was activated with oxygen or CO2, but not with nitrite as electron acceptors. Between 40 to 42 °C, activation of cyclic electron transport balanced the loss of protons so that a sizeable transthylakoid proton gradient was maintained. When the temperature of darkened spinach leaves was slowly increased to 40°C, reduction of the quinone acceptor of Photosystem II, QA, increased particularly when respiratory CO2 production and autoxidation of plastoquinones was inhibited by decreasing the oxygen content of the atmosphere from 21 to 1%. Simultaneously, Photosystem II activity was partially lost. The enhanced dark QA reduction disappeared after the leaf temperature was decreased to 20 °C. No membrane energization was detected by light-scattering measurements during heating the leaf in the dark. In illuminated spinach leaves, light scattering and nonphotochemical quenching of chlorophyll fluorescence increased during warming to about 40 °C while Photosystem II activity was lost, suggesting extra energization of thylakoid membranes that is unrelated to Photosystem II functioning. After P700 was oxidized by far-red light, its reduction in the dark was biphasic. It was accelerated by factors of up to 10 (fast component) or even 25 (slow component) after short heat exposure of the leaves. Similar acceleration was observed at 20 °C when anaerobiosis or KCN were used to inhibit respiratory oxidation of reductants. Methyl viologen, which accepts electrons from reducing side of Photosystem II, completely abolished heat-induced acceleration of P700+ reduction after far-red light. The data show that increasing the temperature of isolated chloroplasts or intact spinach leaves to about 40 °C not only inhibits linear electron flow through Photosystem II but also activates Photosystem I-driven cyclic electron transport pathways capable of contributing to the transthylakoid proton gradient. Heterogeneity of the kinetics of P700+ reduction after far-red oxidation is discussed in terms of Photosystem I-dependent cyclic electron transport in stroma lamellae and grana margins.
