BAPTA-AM is the acetoxymethylester of the calcium chelator BAPTA and has demonstrated efficacy in several animal models of cerebral ischemia. This paper describes the development of a method for the determination of BAPTA-AM in rat plasma by liquid chromatography/tandem mass spectrometry. Owing to multiple ester groups in the structure of BAPTA-AM, [M + Na](+) was chosen as the analytical ion for quantification of BAPTA-AM. During the analytical method development, a high percentage of organic solvent and the addition of an amount of sodium acetate and formic acid in the mobile phase were found to favor the sensitivity and reproducibility of [M + Na](+). Poor fragmentation was usually observed in the MS/MS spectra of sodium adduct ions. However, abundant and reproducible fragment ions were observed for the BAPTA-AM sodium adduct ion, and therefore the traditional selective reaction-monitoring mode was used to further improve the sensitivity of MS detection. Because of the lability of the ester bond, a combination of fluoride and hydrochloric acid was applied to minimize the enzymatic hydrolysis, and acetonitrile was chosen to avoid the chemical hydrolysis or solvolysis during the sample collection and preparation procedure. On the basis of these studies, a rapid, sensitive and reproducible method for the determination of BAPTA-AM in rat plasma, using LC/ESI-MS/MS and a simple protein precipitation procedure, was developed and validated. Also, the present method was successfully applied to the determination of BAPTA-AM plasma concentrations for pharmacokinetic studies in rats.

To identify the major metabolites of resveratrol in rat, rat urine samples were pretreated by using solid-phase extraction technique (SPE) with polyamide cartridges. And a LC-MS/MS method with electrospray ionisation (ESI), negative ion mode and collision induced dissociation (CID), was used to elucidate the structures of the major metabolites of resveratrol. According to the results of our experiment, we found that the main metabolites of resveratrol were resveratrol monoglucuronide (M1), dihydroresveratrol monosulfate (M2), resveratrol monosulfate (M3) and dihydroresveratrol (M4).

Drying studies on a protective coating formed by spontaneous polymerization on aluminum are described. The polymer coating studied here was formed from styrene, n-phenyl maleimide (NPMI), bismaleimide (BMI), and 2-(methacryloyloxy) ethyl acetoacetate (MEA). The coating successfully changed into an adherent film through drying in the presence of a coalescing solvent. Drying at a high temperature (> 170°C) enhanced the performance of the coating in terms of corrosion resistance and adhesion. It was found that the drying process involved removal of surface water on the substrate followed by formation of a dense protective layer associated with thermal crosslinking of g -diketone functional groups in the coating. The reaction mechanism, glass transition temperature, adhesion strength to aluminum, and corrosion resistance of these coatings are reported. The resultant coatings show excellent adhesion strength in a torsional test and very good resistance under the ASTM B-117 accelerated salt fog test.

The corrosion resistance of a novel protective coating formed by spontaneous polymerization on aluminum was studied. The polymer coating was formed from styrene, n-phenyl maleimide, bismaleimide, 2-(methacryloyloxy) ethyl acetoacetate, and N-isobutoxymethyl acrylamide. A corrosion inhibitor, Nitrolotris (methylene) triphosphonic acid, was also used to protect the aluminum surface under the coating after polymerization. The resultant coating showed very good corrosion resistance in water immersion tests, salt fog and cyclic tests, and excellent adhesive bond strength, especially wet, in a torsional test. Incorporation of an internal plasticizer monomer, ethylene glycol methyl ether acrylate further enhanced coating durability.

YiGan San (YGS) has long been used in traditional Japanese and Chinese folk medicine and serves as a potent and novel therapeutic agent to treat Alzheimer's disease. In the present study, a rapid and sensitive method based on HPLC coupled with diode-array detection and quadrupole TOF MS (Q-TOF-MS) was designed to reveal the chemical constituents of YGS. Thirty-six compounds were identified and assigned in YGS, including 14 alkaloids, nine γ-lactones, six flavonoids, three triterpenoid saponinares, two small molecular organic acids, and two other types of compounds. In addition, the accurate fragment weight and MS/MS fragmentation reactions of a subtype indole alkaloid in Uncariae ramulus cum uncis were summarized for the first time to realize rapid identification without reference substances. For the first time, 11 major constituents were comprehensively quantified with a HPLC coupled with triple-quadrupole MS method. A three-section switch was used to realize such multicomponent identification. The contents of saikosaponin B2 and isoliquiritin, which produce anti-inflammatory and antidepressant-like effects, were extremely different, up to 700 times, in two sources of YGS. The developed qualitative and quantitative method was proved to be precise, accurate, and reproducible.

Three Callicarpa species, namely Callicarpa nudiflora Hook. et Arn., Callicarpa macrophylla Vahl. and Callicarpa kwangtungensis Chun. are astringency and hemostasis herbs in the traditional Chinese medical systems. Despite their wide use in Chinese medicine, no report on system comparison on their chemical constituents is available so far. High-performance liquid chromatography coupled with diode array detector and electrospray ionization trap mass spectrometry (HPLC–DAD–ESI-Trap MS) technique was used for qualitative and quantitative analyses of the three Callicarpa herbs. Phenylpropanoid glycosides, flavonoids and organic acids were identified by comparing with reference standards or according to their MS/MS fragmentation behaviors. A total of 33 compounds were identified identified or tentatively identified, and 23 of them were reported from these herbs for the first time. Phenylpropanoid glycosides were featured in the three species with their types and contents presenting significant differences. Furthermore, quantitative analysis was conducted by determining four marker phenylpropanoid glycosides (forsythoside B (14), acteoside (15), poliumoside (19), isoacteoside (21)) and two flavonoids (luteolin (30), apigenin (32)). Three flavonoid glucuronides (luteolin-diglucuronide-glucuronide (5), luteolin-diglucuronide (12), apigenin-7-O-β-glucuronide (24)) were semi-quantified according to their corresponding aglycones. The total contents of the nine major compounds in the three species varied significantly from 8.92 to 40.89 mg/g.

BACKGROUND AND PURPOSE An ATP-binding cassette (ABC) transporter, breast cancer resistance protein (BCRP)/ABCG2, limits oral bioavailability of sulphasalazine. Here we examined the effect of curcumin, the principal curcuminoid of turmeric, on oral bioavailability of microdoses and therapeutic doses of sulphasalazine in humans. EXPERIMENTAL APPROACH Effects of curcumin were measured on the ATP-dependent sulphasalazine uptake by hBCRP-expressing membrane vesicles and on oral bioavailability of sulphasalazine in wild-type and Bcrp(-/-) mice. Eight healthy Japanese subjects received an oral dose of sulphasalazine suspension (100 µg) or tablets (2 g) alone or after curcumin tablets (2 g). Uptake of sulphasalazine was studied in HEK293 cells transfected with the influx transporter (OATP)2B1. KEY RESULTS Curcumin was a potent hBCRP inhibitor in vitro (K(i) 0.70 ± 0.41 µM). Curcumin increased the area under the curve (AUC)(0-8) of plasma sulphasalazine eightfold in wild-type mice at 300 and 400 mg·kg(-1), but not in Bcrp(-/-) mice. Curcumin increased AUC(0-24) of plasma sulphasalazine 2.0-fold at microdoses and 3.2-fold at therapeutic doses in humans. Non-linearity of the dose-exposure relationship was observed between microdoses and therapeutic doses of sulphasalazine. Sulphasalazine was a substrate for OATP2B1 (K(m) 1.7 ± 0.3 µM). Its linear index (dose/K(m)) at the therapeutic dose was high and may saturate OATP2B1. CONCLUSIONS AND IMPLICATIONS Curcumin can be used to investigate effects of BCRP on oral bioavailability of drugs in humans. Besides the limited dissolution, OATP2B1 saturation is a possible mechanism underlying non-linearity in the dose-exposure relationship of sulphasalazine.

In the title compound, [Mn(C(20)H(14)N(2)O(2))(N(3))(H(2)O)]·0.5H(2)O, the Mn(III) ion is chelated by the N,N',O,O'-tetra-dentate Schiff base ligand and further coordinated by one azide ion and one water mol-ecule in trans positions, resulting in a distorted fac-MnN(3)O(3) octa-hedral arrangement. The O atom of the uncoordinated water mol-ecule lies on a crystallographic twofold axis. In the crystal, O-H⋯O and O-H⋯N hydrogen bonds help to establish the packing.

The selection of thermal and photo stabilizers for poly (vinyl chloride) (PVC) using conventional methods is a time-consuming process. The high-throughput screening method developed in this research demonstrates rapid and efficient ways to quantify the effectiveness of PVC stabilizers with respect to raw plastic materials, stabilizers, levels of use, and testing conditions. An experimental protocol using liquid sampling and fluorescence measurement was developed to determine the effectiveness of formulations. This was used to evaluate the performance of stabilizers based on the change of fluorescence emission at 440nm after thermal aging or ultraviolet (UV) radiation. The performance of PVC formulations using six different types of stabilizers was successfully mapped for both PVC resin and flexible PVC.
