Astaxanthin, a carotenoid extensively found in marine organisms and increasingly used as a dietary supplement, has been reported to have neuroprotective effect. In this study, the neuroprotective effects and mechanisms of astaxanthin against glutamate-induced neurotoxicity were explored. The results showed that pretreatment with astaxanthin inhibited neuronal PC12 cell death induced by L-glutamate and reduced the production of reactive oxygen species (ROS) significantly. Astaxanthin prevented glutamate-induced apoptosis by depressing the elevation of Bax/Bcl-2 ratio, caspase-3 activation and Ca2+ influx. Moreover, the loss of mitochondrial membrane potential (MMP) as well as the activation of ROS associated NF-κB and MAPK pathways were also inhibited upon treatment with astaxanthin. Therefore, the neuroprotective effects reflected the ability of astaxanthin in the regulation of multiple signaling pathways that protect mitochondria as well as cellular damage induced by L-glutamate. Thus, astaxanthin has the potential to be used as a prophylactic or remediation agent against neuronal disorders.

Chitosan oligosaccharides (COSs), depolymerized products of chitosan composed of β-(1→4) d-glucosamine units, have broad range of biological activities such as antitumour, antifungal, and antioxidant activities. In this study, peracetylated chitosan oligosaccharides (PACOs) and N-acetylated chitosan oligosaccharides (NACOs) were prepared from the COSs by chemcal modification. The structures of these monomers were identified using NMR and ESI-MS spectra. Their antagonist effects against glutamate-induced PC12 cell death were investigated. The results showed that pretreatment of PC12 cells with the PACOs markedly inhibited glutamate-induced cell death in a concentration-dependent manner. The PACOs were better glutamate antagonists compared to the COSs and the NACOs, suggesting the peracetylation is essential for the neuroprotective effects of chitosan oligosaccharides. In addition, the PACOs pretreatment significantly reduced lactate dehydrogenase release and reactive oxygen species production. It also attenuated the loss of mitochondrial membrane potential. Further studies indicated that the PACOs inhibited glutamate-induced cell death by preventing apoptosis through depressing the elevation of Bax/Bcl-2 ratio and caspase-3 activation. These results suggest that PACOs might be promising antagonists against glutamate-induced neural cell death.

This paper focuses on the development of modern intelligent air combat decision-making technology, and analyzes the elements and characteristics of intelligent air combat scenarios. It introduces the research status and practical application of existing intelligent air combat decision-making methods, including decision-making methods based on game theory, prior data-driven decision-making method, and decision-making methods based on autonomous learning, and especially focuses on deep reinforcement learning intelligent decision-making methods based on value and strategy. Finally, facing to various challenges of future intelligent air combat and the limitations of traditional deep reinforcement learning, the paper gives the future development direction of deep reinforcement learning technology in the field of air combat, which are multi-agent intelligent decision-making technology for cluster warfare, efficient intelligent decision-making technology for wide area space-time, and generalized intelligent decision-making technology for complex scenarios.

A combined numerical-experimental methodology is presented to measure dynamic Mode-I fracture properties of fiber reinforced composites. A modified wedge-DCB test using a Split-Hopkinson Bar technique along with cohesive zone modelling is utilised for this purpose. Three different comparison metrics, namely, strain-displacement response, crack propagation history and crack opening history are employed in order to extract unique values for the cohesive fracture properties of the delaminating interface. More importantly, the complexity of dealing with the frictional effects between the wedge and the DCB specimen is effectively circumvented by utilising right acquisition techniques combined with an inverse numerical modelling procedure. The proposed methodology is applied to extract the high rate interlaminar fracture properties of carbon fiber reinforced epoxy composites and it is further shown that a high level of confidence in the calibrated data can be established by adopting the proposed methodology.

Diaphragma juglandis fructus contains various bioactive constituents. Fourteen compounds were isolated from Diaphragma juglandis fructus by preparative high performance liquid chromatography (pre-HPLC) and high-speed counter-current chromatography (HSCCC). Their structures were identified by nuclear magnetic resonance (NMR) and electrospray ionization mass spectrometry (ESI-MS). Compounds (+)-dehydrovomifoliol (12), (6R,9R)-9-hydroxymegastigman-4-en-3-one (13) and (6R,9S)-9-hydroxymegastigman-4-en-3-one (14) are found from Juglans regia L. for the first time. Compounds dihydrophaseic acid (2), blumenol B (3) and (4S)-4-hydroxy-1-tetralone (11) are isolated from Diaphragma juglandis fructus for the first time. The anti-inflammatory effects of isolated compounds were evaluated by an in vitro model of lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. Compounds gallic acid (1), ethyl gallate (9) and (+)-dehydrovomifoliol (12) exhibited inhibitory activity on the nitric oxide production of RAW 264.7 at a concentration of 25 μM. The result indicated that the combination HSCCC with pre-HPLC is an effective way for compound separation and purification. And Diaphragma juglandis fructus constituents have the potential for the treatment of inflammatory-related diseases.

In our previous studies, we prepared novel oligomannuronate-chromium(III) complexes (OM2, OM4) from marine alginate, and found that these compounds sensitize insulin action better than oligomannuronate(OM), chromium, and metformin in C2C12 skeletal muscle cells. In the present study, we studied their effects on mitochondrial biogenesis, lipid metabolism, and the underlying molecular mechanisms in differentiated 3T3-L1 adipocytes.We firstly used the pGL3-PGC1α and pGL3-ATGL promoter plasmids to compare their effects on PGC1α and ATGL transcription activities. Then mitochondrial biogenesis was quantified by transmission electron microscopy and MitoTracker staining. Mitochondrial oxygen consumption and fatty acid oxidation were measured by an oxygen biosensor system and ³H-labelled water scintillation. The mitochondrial DNA and mRNA involved in mitochondrial biogenesis and lipid oxidation were evaluated by real-time PCR. AMPK together with other protein expression levels were measured by western blotting. The inhibitor compound C and siRNA of PGC1α were used to inhibit the OM2-induced AMPK-PGC1α signaling pathway. And we found that OM2 stimulated AMPK-PGC1α pathway in the 3T3-L1 adipocytes, which were correlated with induced mitochondrial biogenesis, improved mitochondrial function, and reduced lipid accumulation by enhanced fatty acid β-oxidation and augmented ATGL protein expression.Our data indicated that the marine oligosaccharide-derived OM2 might represent a novel class of molecules that could be useful for type 2 diabetes prevention and treatment by up-regulating AMPK-PGC1α signaling pathway.

Maize is one of the major cultivated crops of China, having a central role in ensuring the food security of the country. There has been a significant increase in studies of maize under interactive effects of elevated CO2 concentration ([CO2]) and other factors, yet the interactive effects of elevated [CO2] and increasing precipitation on maize has remained unclear. In this study, a manipulative experiment in Jinzhou, Liaoning province, Northeast China was performed so as to obtain reliable results concerning the later effects. The Open Top Chambers (OTCs) experiment was designed to control contrasting [CO2] i.e., 390, 450 and 550 µmol·mol(-1), and the experiment with 15% increasing precipitation levels was also set based on the average monthly precipitation of 5-9 month from 1981 to 2010 and controlled by irrigation. Thus, six treatments, i.e. C550W+15%, C550W0, C450W+15%, C450W0, C390W+15% and C390W0 were included in this study. The results showed that the irrigation under elevated [CO2] levels increased the leaf net photosynthetic rate (Pn) and intercellular CO2 concentration (Ci) of maize. Similarly, the stomatal conductance (Gs) and transpiration rate (Tr) decreased with elevated [CO2], but irrigation have a positive effect on increased of them at each [CO2] level, resulting in the water use efficiency (WUE) higher in natural precipitation treatment than irrigation treatment at elevated [CO2] levels. Irradiance-response parameters, e.g., maximum net photosynthetic rate (Pnmax) and light saturation points (LSP) were increased under elevated [CO2] and irrigation, and dark respiration (Rd) was increased as well. The growth characteristics, e.g., plant height, leaf area and aboveground biomass were enhanced, resulting in an improved of yield and ear characteristics except axle diameter. The study concluded by reporting that, future elevated [CO2] may favor to maize when coupled with increasing amount of precipitation in Northeast China.
