Clinicians often have negative attitudes toward clients diagnosed with antisocial personality disorder (ASPD), which can sabotage treatment and lead to clinician burnout and job dissatisfaction. Researchers recommend clinicians receive regular clinical supervision; however, clinical supervision strategies and models related to working with ASPD are lacking. We identify supervisors' primary task as exploring and improving clinicians' attitudes toward clients having ASPD and examine this task within the discrimination model of clinical supervision. Newcastle University offers undergraduate students an opportunity to interact with older people by undertaking an intergenerational course about aging. This study systematically evaluated the impacts of involvement in the course on the older adults who participate using a framework analysis method. Three key themes relating to impact emerged: 'Learning,' 'Barriers,' and 'Contributing.' Participation largely led to beneficial impacts on older people through increased knowledge, identity work and generativity, although some examples of potentially negative outcomes were also suggested. In Wales, the barrel jellyfish Rhizostoma octopus is commercially harvested to produce high-value medical grade collagen. Although the fishery is presently not regulated, there are concerns how it may affect the leatherback turtle (Dermochelys coriacea), which preys on R. octopus in local waters. We combined monitoring data and morphometric and weight measurements in models to estimate the potential impact of R. octopus fishery on foraging turtles. We found a significant quadratic relationship between bell diameter and wet weight of R. octopus, with bell diameter explaining 88% of the variability in wet weight. R. octopus biomass in the Carmarthen Bay varied inter-annually between 38.9 and 594.2 tonnes y-1. The amount of R. octopus needed to satisfy a leatherback turtle's daily energetic requirements was estimated at 85.1-319.1 kg. Using leatherback turtle sighting data, our models show that during a jellyfish 'low year", the R. octopus population could be completely depleted by an average of two foraging turtles along with the current level of commercial harvesting (4.3 tonnes). During a jellyfish "high year", the current level of commercial harvesting is predicted to have relatively little impact on food supply for even the maximum number of foraging leatherback turtle reported in the area. This article relates the experiences of several European translators of Haruki Murakami's novel, 1Q84, discussing difficulties they encountered in the text and different solutions they chose. These are in turn compared to the solutions chosen in the English translation, which was published later than many European versions. How, for example, should the translator handle such issues as the use of narrative present or fluctuations between the first and third person of the narrator? How to express in alphabetic languages the author's creative manipulation of the Japanese writing system? The article also discusses the issue of mistakes, including both those appearing in the original text and those made inadvertently by translators, as well as editorial cuts in the English translation and the reasons behind them. GEN1 was recently identified as a key Holliday junction resolvase involved in homologous recombination. Somatic truncating GEN1 mutations have been reported in two breast cancers. Together these data led to the proposition that GEN1 is a breast cancer predisposition gene. In this article we have formally investigated this hypothesis. We performed full-gene mutational analysis of GEN1 in 176 BRCA1/ 2-negative familial breast cancer samples and 159 controls. We genotyped six SNPs tagging the 30 common variants in the transcribed region of GEN1 in 3,750 breast cancer cases and 4,907 controls. Mutation analysis revealed one truncating variant, c.2515_2519delAAGTT, which was present in 4% of cases and 4% of controls. We identified control individuals homozygous for the deletion, demonstrating that the last 69 amino acids of GEN1 are dispensable for its function. We identified 17 other variants, but their frequency did not significantly differ between cases and controls. Analysis of 3,750 breast cancer cases and 4,907 controls demonstrated no evidence of significant association with breast cancer for six SNPs tagging the 30 common GEN1 variants. These data indicate that although it also plays a key role in double-strand DNA break repair, GEN1 does not make an appreciable contribution to breast cancer susceptibility by acting as a high- or intermediate-penetrance breast cancer predisposition gene like BRCA1, BRCA2, CHEK2, ATM, BRIP1 and PALB2 and that common GEN1 variants do not act as low-penetrance susceptibility alleles analogous to SNPs in FGFR2. Furthermore, our analyses demonstrate the importance of undertaking appropriate genetic investigations, typically full gene screening in cases and controls together with large-scale case-control association analyses, to evaluate the contribution of genes to cancer susceptibility. PALB2 interacts with BRCA2, and biallelic mutations in PALB2 (also known as FANCN), similar to biallelic BRCA2 mutations, cause Fanconi anemia. We identified monoallelic truncating PALB2 mutations in 10/923 individuals with familial breast cancer compared with 0/1,084 controls (P = 0.0004) and show that such mutations confer a 2.3-fold higher risk of breast cancer (95% confidence interval (c.i.) = 1.4–3.9, P = 0.0025). The results show that PALB2 is a breast cancer susceptibility gene and further demonstrate the close relationship of the Fanconi anemia–DNA repair pathway and breast cancer predisposition. We identified constitutional truncating mutations of the BRCA1-interacting helicase BRIP1 in 9/1,212 individuals with breast cancer from BRCA1/BRCA2 mutation–negative families but in only 2/2,081 controls (P = 0.0030), and we estimate that BRIP1 mutations confer a relative risk of breast cancer of 2.0 (95% confidence interval = 1.2–3.2, P = 0.012). Biallelic BRIP1 mutations were recently shown to cause Fanconi anemia complementation group J. Thus, inactivating truncating mutations of BRIP1, similar to those in BRCA2, cause Fanconi anemia in biallelic carriers and confer susceptibility to breast cancer in monoallelic carriers.
