61. Anti-Photoaging Effects of the Ethanol Extract from Eucommia ulmoidesOliv. Seed Meal on UVB-irradiated HaCaT Cells and Mice

Abstract

Background Eucommia ulmoidesOliv. is a traditional Chinese medicine, and its flowers, leaves, and seeds are widely used as food. Currently, E. ulmoides seed oil is capable of improving glucose and lipid metabolism in type 2 diabetic mice, but E. ulmoidesseed meal is not fully utilized, and the anti-photoaging effect of it is still unclear.Objectives We examined the anti-photoaging effect of the ethanol extracts of E. ulmoidesseed meal (EUSM) in UVB-stimulated HaCaT cells and mice.Materials and Methods EUSM was analyzed using Q-Orbitrap-based high-resolution LC-MS/MS. In vivoand in vitroexperiments were conducted using normal mice and HaCaT cells, respectively. Hematoxylin and eosin (H&E) staining and Masson’s staining were used to evaluate epidermal thickness and degree of skin fibrosis. The production of hyaluronic acid (HA) and total protein using ELISA kits. The expression of levels for collagen type I α 1 (COL1A1), hyaluronidase-1 (HYAL-1), MMP-3, and MMP-9 were assessed using semi-quantitative real-time polymerase chain reaction (qRT-PCR).Results EUSM contains all-trans-retinoic acid, medicagenic acid, and other chemical constituents. EUSM increased the level of total collagen by reducing matrix metalloproteinase expression, and it enhanced the content of HA. In addition, EUSM reduced the production of wrinkles in UVB-induced mouse skin and showed that EUSM decreased the level of malondialdehyde (MDA) and increased the contents of superoxide dismutase (SOD) and catalase (CAT) in the mouse skin tissue.Conclusion EUSM has therapeutic potential for UVB-induced skin photoaging and provided a rationale for the application of EUSM in the cosmetic field.

Abstract

Phase change material (PCM) selection for particular applications mainly depends on its phase change temperature and latent heat, which were commonly measured by the differential scanning calorimetry (DSC). However, the DSC instruments are not absolute measuring devices and calibration is extremely important to ensure the accuracy of determined data. Meanwhile, the PCM with melting temperature between 0 and 65 °C is the most common interest for energy saving in buildings. This study, therefore, calibrated the DSC produce (NanoTechnology Inc.) by the high-purity metals with various regression methods for low melting temperature PCM thermal properties analysis. In the end, the paraffin n-eicosane was introduced to evaluate the accuracy of the temperature and latent heat measurements of the DSC. The experimental results showed that the accuracy of calibration could be improved by higher-order and more reference materials in a broad temperature range. However, the measurement results of n-eicosane proved that the 0-order regression method could offer good accuracy with only one suitable calibrant in a particular temperature range. Therefore, this research suggests the selection of reference materials for DSC calibration should consider the target of measurement temperature range as well as the thermal stabilities of calibrants.

Abstract

Finding target persons in full scene images with a query of text description has important practical applications in intelligent video surveillance.However, different from the real-world scenarios where the bounding boxes are not available, existing text-based person retrieval methods mainly focus on the cross modal matching between the query text descriptions and the gallery of cropped pedestrian images. To close the gap, we study the problem of text-based person search in full images by proposing a new end-to-end learning framework which jointly optimize the pedestrian detection, identification and visual-semantic feature embedding tasks. To take full advantage of the query text, the semantic features are leveraged to instruct the Region Proposal Network to pay more attention to the text-described proposals. Besides, a cross-scale visual-semantic embedding mechanism is utilized to improve the performance. To validate the proposed method, we collect and annotate two large-scale benchmark datasets based on the widely adopted image-based person search datasets CUHK-SYSU and PRW. Comprehensive experiments are conducted on the two datasets and compared with the baseline methods, our method achieves the state-of-the-art performance.

Abstract

The mechanisms that induce the reduction of beta-myosin heavy chain (MHC) expression in response to non-weight bearing activity were analyzed in soleus muscles isolated from mice subjected to hindlimb suspension (HS). Northern blot analysis of HS mouse soleus indicated the presence of reduced levels of messenger RNA (mRNA) expression for beta-MHC, cardiac/slow troponin C and slow myosin light chain one and two. HS also increased the levels of muscle creatine kinase and skeletal troponin C mRNA expression.

Thee induction of beta-myosin heavy chain (beta-MHC) gene expression was investigated in transgenic mouse lines containing serial deletions of the human beta-MHC promoter to nucleotides -293, -201, and -141. Expression assays identified only the 293-base pair beta-MHC in both adult and fetal mice muscle. Expression of the 293 gene was site specific at the plantaris and soleus muscles.

Abstract

Mutations of the DNA regulatory sub-regions, muscle chloramphenicol acetyltransferase (MCAT), C-rich and beta-e3, fail to eliminate the induction of beta-myosin heavy chain (MHC) transgene in mechanically overloaded plantaris muscle of transgenic mice. Protein extracts from these wild-type muscles and those containing a mutant transgene show a marked increase in CAT specific activity. In transgenic mice with mutations in a 5,600-base pair of the transgene, the increased CAT activity is restricted to type I MHC fibres.

Abstract

Skeletal muscles respond to extended periods of mechanical overload by increasing production of beta-myosin heavy chains (beta-MHC) and slow myosin light chain (SMLC) proteins. Rat plantaris muscles subjected to overload conditions for seven weeks increase mRNA for beta-MHC and SMLC. Rats that possessed transgenes for these muscle proteins undergo synthesis at elevated levels in response to overload. In addition, genes for beta-MHC were identified in the human genome.

Abstract

Hypertrophy of the skeletal muscles caused by mechanical overload causes a decrease in the expression of the muscle creatine kinase (MCK) gene determined by a decrease in the amount of MCK mRNA and MM-CK isoprotein. The decrease in MCK gene expression in transgenic mice containing the 5'-flanking DNA present upstream from the enhancer region is more than in the mice containing the +enh206 enhancer region. Thus the expression of the MCK gene is controlled by both the enhancer region and the 5' region present upstream.

97. A simple and efficient gene functional analysis method for studying the growth and development of peach seedlings

Abstract

Stable genetic transformation of peach [Prunus persica(L.) Batsch] still faces many technical challenges, and existing transient expression methods are limited by tissue type or developmental stage, making it difficult to conduct functional analysis of genes regulating shoot growth. To overcome this dilemma, we developed a three-step method for efficient analysis of gene functions during peach seedling growth and development. This method resulted in transformation frequencies ranging from 48 to 87%, depending on the gene. From transformation of germinating seeds to phenotyping of young saplings took just 1.5 months and can be carried out any time of year. To test the applicability of this method, the function of three tree architecture-related genes, namely PpPDS, PpMAX4, and PpWEEP, and two lateral root-related genes, PpIAA14–1and −2, were confirmed. Since functional redundancy can challenge gene functional analyses, tests were undertaken with the growth-repressor DELLA, which has three homologous genes, PpDGYLA(DG), PpDELLA1(D1), and −2(D2), in peach that are functionally redundant. Silencing using a triple-target vector (TRV2-DG-D1-D2) resulted in transgenic plants taller than those carrying just TRV2-DGor TRV2. Simultaneously silencing the three DELLAgenes also attenuated the stature of two dwarf genotypes, ‘FHSXT’ and ‘HSX’, which normally accumulate DELLA proteins. Our study provides a method for the functional analysis of genes in peach and can be used for the study of root, stem, and leaf development. We believe this method can be replicated in other woody plants.

Abstract

The Air Cooling Garment (ACG) was employed to study skin temperature and Clothing Inner Microclimate (CIMC) Regulation, which provides theoretical basis for further optimization of aerodynamic layout of AGG towards human thermal comfort improving. 15 male postgraduates served as subjects in the experiment, each runs at 5 km/h (to simulate Moderate Work, MW) or 7.5 km/h (Heavy Work, HW) on the treadmill in an artificial climate chamber with or without the Air Cooling Garment (ACG) for 40 min respectively. The temperature of the artificial climate chamber changed from 24 to 34 °C. The skin temperature of upper body, clothing inner Wind Velocity (WV), Relative Humidity (RH) and temperature were measured. Results showed that the skin temperature decreased much more significantly with the ACG than without when the ambient temperature is over 26 °C, especially when engaged in HW. The WV and RH in clothes changed with the labor intensity, rather than the ambient temperature. The variation of clothing inner temperature field was different, with the front and back sides decreased with labor intensity, while the left and right sides were not affected. It proved that the ACG can improve CIMC and enhance human thermal comfort effectively.
