Abstract

AQUA 2018, #WeRAquaculture we are the producers and investors, the scientists and technical advisors, the legislators and educators, the students, civil society organizations and consumers of farmed aquatic products, 25-29 August 2018, Montpellier, France, The European sea bass, Dicentrarchus labrax, a major aquaculture species in Europe, has a peculiar polygenic system of sex determination, with several QTLs determining the genetic component of sex. Temperature during the early phases of larval rearing (before 60 days post hatch, dph) also has large effects on sex-ratio: high temperatures (>17°C, HT) masculinize fish that would have developed as females in low temperature (neomales> in fish with chromosomal sex determination). We produced two families by mating two males with the same female. The progenies were reared in common garden, half of them in a sex-neutral LT temperature protocol (16°C until 60 dph), and the other half in a masculinizing HT treatment (21°C until 60 dph). The fish were sexed at 319 dph, and assigned to their parents with 12 microsatellite markers. Among the assigned fish, 92 HT fish and 84 LT fish from sire A, as well as 78 LT fish from sire B, were selected for genotyping on a 3K SNP Illumina ISelect array. In total, 966 SNP markers were polymorphic within or between the two families, with a call rate >0.90, a sufficient minor allele frequency (MAF>0.05) and no mendelian transmission errors. Sex was predicted by the genomic estimated breeding value of sex tendency using a genomic best linear unbiased prediction (GBLUP) model, applied to each family at each temperature and various combinations thereof. When data from two temperature treatments were applied, a fixed effect of temperature was included in the model. To the prediction of sex, a receiver operating characteristic (ROC) curve was built using all possible sex tendency thresholds to separate males from females. The quality of the prediction was assessed by the area under the curve (AUC). The AUC was close to its maximum value of 1 in family A, HT (AUC=0.999). In family A, LT, the AUC was 0.947, and it was 0.994 in family B, LT. Thus, prediction of sex was very good in the three groups. When the data from both LT and HT were combined in family A, the AUC was 0.962, showing coherence between the models at two temperatures. However, when data from family A and family B were combined, the AUC dropped to 0.900, showing that the optimal model was not the same in both families. We show that despite its polygenic nature, sex in The European sea bass can be predicted with very high accuracy within family using a genome-wide approach (GBLUP). Cysts containing spermatozoa were observed in ovaries with pre-vitellogenic oocytes. This feature is very uncommon, especially owing to the fact that male cells do not normally reach this stage in captivity, and an environmentally controlled transdifferentation process may not be excluded. Besides, the expression of the gonadal aromatase gene (cyp19a1a) was found to be higher in these 2 intersexual fish compared to normal females, although these results must be considered with caution since only 2 intersexual fish were available. A possible feminizing effect of this ‘abnormal’ upregulation of aromatase is discussed.

Abstract

The aim of this review is to present an overview of the sex differentiation and sex determination processes in eels in relation to the urgent need to provide scientific knowledge to better protect and manage the Anguilla genus. Indeed, the global decline of the three main temperate eel stocks, Anguilla anguilla, Anguillidae (Fisheries Management and Ecology, 2003, 10, 365); Anguilla japonica, Anguillidae (Casselman, Eel Biology, Springer Japan, 2003, 293) and Anguilla rostrata, Anguillidae (Tatsukawa, Eel Biology, Springer, Japan, 2003, 255), raises concerns about the necessity to better understand all stages of the life cycle of eels (Righton and Walker, Journal of Fish Biology, 2013, 83, 754). Little is known about the mechanisms involved in the production of males and females in this species with environmental sex determination. Previous reviews identifying the density of individuals as the major factor influencing sex determination were undertaken (Krueger and Oliveira, Environmental Biology of Fishes, 1999, 55, 381; Reviews in Fish Biology and Fisheries, 2005, 15, 37). Here, we review the current advances on the subject, focusing on the roles of early growth rate and interindividual relationships, which are mechanisms underpinned by density, as well as the sex differentiation process, and we question how this knowledge might influence global conservation measures. Abstract

To address the impact of cadmium sulphide nanoparticles (CdSNPs) of two different sizes (8 and 50 nm), Danio rerio zebrafish were dietary exposed to very low doses: 100 or 40 ng CdSNPs/day/g body weight for 36 or 60 days, respectively. The results obtained using RAPD-PCR genotoxicity test showed genomic alteration since the number of hybridisation sites of the RAPD probes was significantly modified after CdSNPs exposure. In addition, selected stress response genes were either repressed or upregulated in tissues of CdSNPs-exposed fish. Mitochondrial dysfunction was also caused by the presence of CdSNPs in food. Cadmium accumulation in fish tissues (brain and muscles) could only be observed after 60 days of exposure. CdSNPs toxicity was dependent on their size and concentration. [ABSTRACT FROM AUTHOR]

Abstract

The impact of a daily ration of food containing gold nanoparticles (AuNPs) of two sizes (12 and 50 nm) was investigated in the zebrafish Danio rerio at very low doses (from 36-106 ng gold/fish/day). AuNP exposure resulted in various dysfunctions at the sub cellular scale, and AuNP concentration in food, AuNP size and exposure duration modulated the observed adverse effects. Indeed, we showed alteration of genome composition using a RAPD-PCR genotoxicity test as the number of hybridization sites of the RAPD probes was significantly modified after AuNP exposure. Moreover, the expression of genes involved in DNA repair, detoxification processes, apoptosis, mitochondrial metabolism and oxidative stress was also modulated in response to AuNP contamination. Mitochondrial dysfunctions appeared in brain and muscle for both tested doses (40 and 100 ng gold/fish/day), but gold accumulation in fish tissues could only be observed in the case of the highest exposure dose. Abstract

Exposure to metal-contaminated water has been shown to result in a number of reproductive abnormalities in adult and larvae fish, such as failure of oocyte maturation and teratogenic effects. Recently, dietary uptake of metals by fish has been recognized as a critical route of exposure, however, the mechanisms of metal uptake and toxicity are poorly understood and in need of further investigation. The objectives of the present study are to quantify uranium (U dietary transfers from spiked artificial diets) in Danio rerio tissues and embryos, as well as establish its effect on reproduction and embryonic development. Uranium's environmental prominence is currently increasing because of new mining and milling activities. Uranium concentrations range from 0.02 µg/L in natural waters to 2 mg/L. The focus of this study was to examine the trophic transfer and effects of U following exposure modalities (dose, exposure duration 1 to 20 d). Two different isotopes were used to distinguish between chemical and radioactivity toxicity of U. Results showed that U trophic transfer was low (0.52%). Uranium tissue distributions showed that accumulation occurred in digestive organs (liver, digestive tract) following dietary exposure. High levels of U were measured in the gonads (female in particular, >20% of relative burden). High U accumulation levels in eggs indicated maternal transfer of the contaminant. Moreover, U trophic exposure led to a reduction in reproduction success as a function of U accumulated levels.
