This study reports the purification and identification of a novel 28 kDa phosphoprotein from rat pancreatic acini, previously described as being highly regulated by calcium mobilizing secretagogues, which we have designated calcium-regulated heat-stable protein 28 (CRHSP-28). Internal amino acid sequences of purified CRHSP-28 were obtained following trypsin digestion and found to match with >95% identity the predicted amino acid sequence of a novel cDNA recently identified as being highly expressed in human breast carcinomas. Verification that this cDNA codes for human CRHSP-28 was demonstrated by the ability of antiserum raised against purified rat CRHSP-28 to recognize the recombinant human protein when expressed in bacteria. Furthermore, this antibody was found to specifically react with CRHSP-28 in rat acini following one- and two-dimensional electrophoresis and underwent a marked acidic shift in mobility after cholecystokinin stimulation, a phenomenon indicative of an increase in its phosphorylation. CRHSP-28 is predicted to be extremely hydrophilic, is phosphorylated entirely on serine residues, and bears little homology to any known proteins. Finally, the distribution of the CRHSP-28 protein in various rat tissues revealed that although it was present at low levels in almost all tissues, it was most highly expressed in pancreas, followed by the gastric, intestinal, and colonic mucosa. In view of its relative abundance throughout the digestive system and its apparent regulation by calcium-mobilizing agents, this protein may provide valuable insight into the mechanism(s) of calcium signaling in these tissues.

Stimulation of rat pancreatic acinar cells with cholecystokinin (CCK) is known to result in a significant inhibition of CTP:phosphocholine cytidylyltransferase (CT), a rate-limiting enzyme in phosphatidylcholine biosynthesis. Immunoprecipitation of CT from 32P-labeled acinar cells revealed that CCK treatment also caused a marked reduction in CT phosphate levels. The effects of CCK were maximal over 60 min and dependent on concentration, exhibiting an EC50 of 800 pM. Other calcium mobilizing secretagogues such as carbamylcholine (100 microM) and bombesin (10 nM) also reduced CT phosphate levels to 20 and 39% of control, respectively. Treatment of cells with thapsigargin and/or 12-O-tetradecanoyl-phorbol-13-acetate established that a combination of increased intracellular Ca2+ and protein kinase C activation was necessary to decrease phosphorylated CT content. Conversely, secretin (10 nM) or 8-(4-chlorophenylthio)-cAMP (100 microM) added alone had no effects. Use of the compound JMV-180 indicated CCK was acting through the low affinity state of the CCKA receptor to reduce CT phosphate levels. Further, the decrease in phosphorylated CT caused by CCK was blocked by the phosphatase inhibitors okadaic acid (3 microM) and calyculin A (100 nM). Finally, immunoblotting from whole cell lysates revealed CT was partially degraded in response to CCK, providing a novel mechanism by which the inhibition of CT enzyme activity occurs in response to the hormone. Moreover, this degradation was also blocked by a phosphatase inhibitor. These data suggest that the dephosphorylation of either CT itself or some other regulatory molecule(s) which mediates the CCK-induced protease activation may play a central role in reducing CT enzyme levels in acinar cells.

The immunosuppressant cyclosporin A (CsA) was utilized as a highly specific inhibitor of the Ca2+/calmodulin-dependent protein phosphatase, PP2B in rat pancreatic acinar cells. Treatment of cells with CsA for 20 min resulted in a concentration-dependent inhibition of PP2B that was maximal (> 90%) at 1 microM and exhibited an IC50 of 65 nM. CsA also inhibited cholecystokinin-, 100 pM, or carbamylcholine-, 10 microM, induced amylase release in a concentration-dependent manner. A maximal inhibition to 55% of stimulated control cells occurred at 1 microM CsA with half-maximal inhibition occurring at approximately 200 nM. Secretion in response to 1 microM 12-O-tetradecanoylphorbol-13-acetate (TPA) was uneffected by CsA treatment. Conversely, amylase release stimulated by the Ca2+ mobilizing agent, thapsigargin, when added alone at 2 microM or in combination with TPA, was inhibited by CsA to 66 and 61% of control cells, respectively. These data indicate that CsA-mediated inhibition occurs only when stimulation involves an increase in intracellular Ca2+. In addition, analogues of CsA, 6-methyl-alanine-CsA, and 11-methyl-leucine-CsA had no effect on PP2B activity or amylase secretion. The chemically distinct immunosuppressant, FK506, produced only partial inhibition of PP2B activity and did not significantly inhibit amylase secretion at concentrations up to 1 microM. Two-dimensional gel electrophoresis of proteins from 32P-labeled acinar cells revealed that CsA specifically blocked the cholecystokinin-stimulated dephosphorylation of a 24-kDa protein in a concentration range similar to that seen for inhibition of secretion. Using 32P-labeled cytosol and purified calcineurin, a Ca(2+)- and calmodulin-dependent dephosphorylation of the 24-kDa protein was also demonstrated in vitro. Collectively, these data indicate that the Ca2+/calmodulin-dependent protein phosphatase, PP2B, plays a significant role in stimulus-secretion coupling in pancreatic acinar cells.

Experiments were performed to immunologically identify protein kinase C (PKC) in cultured IEC-6 cells. Polyclonal antibodies specific to PKC revealed an immunoreactive band of approximately 84 kDa in both cytosolic and solubilized particulate fractions. Treatment with phorbol 12-myristate 13-acetate (PMA; 10 nM x 60 min) increased the intensity of the 84-kDa band by 25% in the solubilized particulate fraction while decreasing it by 36% in the cytosolic fraction. Prolonged 24-h treatment with 300 nM PMA completely abolished the 84-kDa band in both fractions. Isoform-specific antisera demonstrated that alpha- and epsilon-isoforms of PKC were expressed in IEC-6 cells. Treatment of quiescent cultures with PMA induced a maximal 400% increase in ornithine decarboxylase (ODC) activity. Similarly, addition of exogenous phospholipase C (PLC) to quiescent cells stimulated ODC activity. Downregulation of PKC with 300 nM PMA x 24 h inhibited basal, serum, and PLC-stimulated ODC activity by 70%. Northern analysis revealed that PKC downregulation was correlated with a marked reduction in ODC mRNA levels, suggesting regulation of ODC enzyme at this level. Despite their ability to modulate ODC activity in quiescent cultures, neither PMA nor PLC induced [3H]thymidine incorporation at 24 h. Furthermore, downregulation of PKC did not attenuate thymidine incorporation. However, chronic PMA treatment caused the cells to contact-inhibit at a 30% lower cell density, 3.16 x 10(6) vs. 2.1 x 10(6) cells/35-mm plate, respectively.(ABSTRACT TRUNCATED AT 250 WORDS)

Regulation of putrescine uptake in a small intestinal crypt cell line, IEC-6 cells, was examined. Uptake of [14C]putrescine was measured throughout a normal growth curve and was found to be inversely related to growth. Kinetic analysis at low and high cell density revealed the inhibition of uptake in confluent cells was due to a five-fold reduction in Vmax of uptake, 199.5 vs. 43.1 pmol.10(5) cells-1.h-1, respectively. Three gastrointestinal hormones, gastrin, secretin, and cholecystokinin, produced partial inhibition of [14C]putrescine uptake. Conversely, treatment of quiescent cells with 5% fetal bovine serum to stimulate growth did not affect uptake. Influence of putrescine uptake on free ionized intracellular Ca2+ ([Ca2+]i) was measured by microspectrofluorometry using the Ca(2+)-sensitive fluoroprobe fura-2. Basal [Ca2+]i was calculated to be 112 nM and increased rapidly to 313 nM upon addition of 10 microM putrescine. Preventing the rise in [Ca2+]i using an intracellular Ca2+ buffer, 1,2-bis(2-aminophenoxy)ethane-N,N,N',N'-tetraacetic acid acetoxymethyl ester, decreased [14C]putrescine uptake to 29.5 +/- 5.3% of control values. 45Ca2+ flux experiments and measurement of transport in 0 Ca2+ and 0.5 mM EDTA suggested an intracellular source of calcium was mobilized during putrescine uptake. Finally, use of the putative calmodulin antagonist N-(6-aminohexyl)-5-chloro-l-naphthalenesulfonamide caused a dose-dependent inhibition of [14C]putrescine uptake with 50% inhibitory concentration of approximately 7 microM. These data suggest that putrescine uptake in IEC-6 cells may be regulated by a Ca2+/calmodulin-dependent mechanism.
