65. Complex population evolutionary history of four cold-tolerant Notopterygium herb species in the Qinghai-Tibetan Plateau and adjacent areas

Abstract

Historical geological and climatic events are the most important drivers of population expansions/contractions, range shifts, and interspecific divergence in plants. However, the species divergence and spatiotemporal population dynamics of alpine cold-tolerant herbal plants in the high-altitude Qinghai-Tibetan Plateau (QTP) and adjacent areas remain poorly understood. In this study, we investigated population evolutionary history of four endangered Notopterygium herb species in the QTP and adjacent regions. We sequenced 10 nuclear loci, 2 mitochondrial DNA regions, and 4 chloroplast DNA regions in a total of 72 natural populations from the 4 species, and tested the hypothesis that the population history of these alpine herbs was markedly affected by the Miocene–Pliocene QTP uplifts and Quaternary climatic oscillations. We found that the four Notopterygium species had generally low levels of nucleotide variability within populations. Molecular dating and isolation-with-migration analyses suggested that Notopterygium species diverged ~1.74–7.82 million years ago and their differentiation was significantly associated with recent uplifts of the eastern margin of the QTP. In addition, ecological niche modeling and population history analysis showed that N. incisum and N. franchetii underwent considerable demographic expansions during the last glacial period of the Pleistocene, whereas a demographic contraction and a expansion occurred for N. forrestii and N. oviforme during the antepenultimate interglacial period and penultimate glacial period, respectively. These findings highlight the importance of geological and climatic changes during the Miocene–Pliocene and Pleistocene as causes of species divergence and changes in population structure within cold-tolerant herbs in the QTP biodiversity hotspot.

74. Preparation of Gel Bacterial Testing Plate.

Abstract

Brain heart infusion (BHI) broth as nutrient components and two cold water soluble gel, xanthan gum and carrageenan as complex gel were used to develop a gel bacterial testing plate for the detection of aerobic bacterial count in food. The optimal combination of the key additional components was determined by orthogonal test, and the effects of the ratio of the two gels on physical and chemical and microbiological results were examined. The optimal results were obtained when the components of medium were BHI 38.5 g/L, TTC 0.008 g/L, SAP 0.05 g/L, sodium pyruvate 1.5 g/L, and the proportion of complex gel was xanthan gum: carrageenan=8:2. Compared with national standard method, the gel testing plate method of this study had a good correlation (R²>0.99), achieving the same limit (2 CFU/mL) detection and sensitivity (100%); Compared with 3M Petrifilm, the quantitative results showed no significant difference, the two methods linear relationship was good (R²=0.99). The detection limit, sensitivity, and accuracy of the colony count testing plate in this study were no significant differences in comparison with the national method, it can be applied to the detection of colony count in food. 75. Efficacy and safety of imiquimod 5% cream for basal cell carcinoma: a meta-analysis of randomized controlled trial.

Abstract

Background: We conducted this meta-analysis to compare the efficacy and safety of imiquimod with other treatments in patients with basal cell carcinoma (BCC). Methods: A comprehensive literature search was performed in the database of PubMed, Embase, and Web of Science, focusing on studies that evaluated the efficacy and safety profile of BCC patients who were treated with imiquimod. The main outcome measures included histological/composite clearance rate, success rate, complete response rate, tumor free survival, and adverse events. Results were expressed as risk ratio (RR) with 95% confidence intervals (CIs). Pooled estimates were calculated using a fixed-effects or random-effects model according to the heterogeneity among studies. Results: A total of 13 studies involving 4256 patients were included in this meta-analysis. Imiquimod was associated with significantly higher histological clearance rate and composite clearance rate. Moreover, imiquimod also significantly increased complete response rate but had no effect in the success rate and probability of tumor free survival, as compared with other treatments. There were more patients with imiquimod developed adverse events than those with other treatments. Conclusion: The present meta-analysis indicated the effects of imiquimod in improving the histological/composite clearance rates as compared with other treatments. [ABSTRACT FROM AUTHOR]

Abstract

Pyoderma gangrenosum is an uncommon ulcerative cutaneous lesion manifesting as rapidly progressing single or multiple skin ulcers. Permanent stoma in inflammatory bowel disease patients remains an independent risk of pyoderma gangrenosum. In the current report, we describe a case of pyoderma gangrenosum in a post‐colostomy ulcerative colitis patient with chronic hepatitis B. Pyoderma gangrenosum began seemingly as peristomal dermatitis that rapidly developed into painful ulcerations with subsequent appearance of sterile pustules and ulcerations in the left lower leg. The patient significantly improved after active management with prednisolone, antiviral therapy with entecavir, and wound dressings. Our case suggests that physicians and surgeons should have a high index of suspicion of pyoderma gangrenosum in post‐colostomy ulcerative colitis patients who develop peristomal dermatitis. Abstract

Background: This study examined the use of low doses of antipsychotic medications (300mg/day CPZeq or less) in older Asian patients with schizophrenia and its demographic and clinical correlates. Methods: Information on hospitalized patients with schizophrenia, aged 55 years or older, was extracted from the database of the Research on Asian Psychotropic Prescription Patterns (REAP) study (2001-2009). Data on 1,452 patients in eight Asian countries and territories including China, Hong Kong, Japan, Korea, Singapore, Taiwan, India, and Malaysia were analyzed. Sociodemographic and clinical characteristics and antipsychotic prescriptions were recorded using a standardized protocol and data collection procedure. Results: The prescription frequency for low doses of antipsychotic medications was 40.9% in the pooled sample. Multiple logistic regression analysis of the whole sample showed that patients on low doses of antipsychotic medications were more likely to be female, have an older age, a shorter length of illness, and less positive symptoms. Of patients in the six countries and territories that participated in all the surveys between 2001 and 2009, those in Japan were less likely to receive low doses of antipsychotics. Conclusion: Low doses of antipsychotic medications were only applied in less than half of older Asian patients with schizophrenia

Abstract

The purpose of the present study was to review the prescription patterns of antidepressants in different countries in East Asia. The survey was conducted in China, Japan, Korea, Singapore and Taiwan from October 2003 to March 2004 using the unified research protocol and questionnaire. Twenty teaching hospitals and major psychiatric hospitals participated and a total of 1898 patients receiving antidepressants were analyzed. The survey provided a number of interesting characteristics on the prescription patterns of antidepressant in East Asia. Out of 56 antidepressants listed in the Anatomical Therapeutic Chemical Classification (ATC) index by the World Health Organization (WHO) Collaborating Center for Drug Statistics Methodology (Oslo), only 26 antidepressants were prescribed in participating countries in East Asia. On average 38.4% of prescriptions of antidepressants were for patients with diagnoses other than depressive disorders. The availability and commonly prescribed antidepressants varied greatly by country. The selective serotonin re-uptake inhibitors (SSRI) and other newer antidepressants were prescribed in approximately 77.0% of all cases. At the time of the survey, only two SSRI medications were available in Japan. However, five types of SSRI were available and were often prescribed in Korea.

Abstract

Alkaline pulping liquors are problematic for anaerobic treatment due to their toxicity to methanogens and their relatively large fraction of inert lignin. In previous research, toxicity due to wood extractives was shown to be highly eliminated during aerobic wastewater treatment, but not during anaerobic treatment. These observations have led to the proposal of a detoxification strategy denominated upfront dilution, based on the sequenced anaerobic-aerobic treatment of the pulping liquor, recirculating the aerobic effluent to dilute the incoming influent to sub-toxic concentrations. In this study, the treatment of highly toxic hemp stem wood black liquor (HSWBL) in lab-scale UASB reactors with upfront dilution was compared with direct anaerobic treatment and with direct aerobic treatment. Direct anaerobic treatment of 12 g COD/ l HSWBL led to almost complete inhibition of the methanogenic activity within 14 d. However, recirculation of 75% of the aerobic post-treatment effluent for upfront dilution of the toxic HSWBL, enabled anaerobic treatment at loading rates up to 21.5 g COD/ l UASB ·d without significant inhibition of the methanogenic activity. Extensive detoxification was confirmed during anaerobic-aerobic treatment of 20 g COD/ l HSWBL recirculating 86% of the aerobic effluent. COD and BOD removal was 72% and 97%, respectively, after anaerobic-aerobic treatment at an overall loading rate of 3.6 g COD/ l ·d, while 30–35% of the incoming COD was recovered as methane. Batch-assays demonstrated significant detoxification after anaerobic-aerobic treatment of HSWBL. Treatment efficiencies and detoxification during anaerobic-aerobic and aerobic treatment were similar. However, the anaerobic-aerobic treatment system provided 50% lower surplus sludge production, production of 0.16 m 3 methane/kg COD removed as an energy source, less nutrient dosage and substantial reductions in aeration costs. During anaerobic-aerobic treatment as well as aerobic treatment significant lignin removal was obtained, ranging from 28–58%. Lignin removal could be attributed to biodegradation of low molecular weight lignin (MW 34 kD.

Abstract

To assess the pharmacokinetics of vildagliptin at different doses and build a mechanism-based population model that simultaneously describes vildagliptin pharmacokinetics and its effects on DPP-4 activity based on underlying physiology and biology.Vildagliptin concentrations and DPP-4 activity vs. time from 13 type 2 diabetic patients after oral vildagliptin 10, 25 or 100 mg and placebo twice daily for 28 days were co-modelled. NONMEM VI and S-ADAPT were utilized for population modelling.A target-mediated drug disposition (TMDD) model accounting for capacity-limited high affinity binding of vildagliptin to DPP-4 in plasma and tissues had good predictive performance. Modelling the full time course of the vildagliptin-DPP-4 interaction suggested parallel vildagliptin dissociation from DPP-4 by a slow first-order process and hydrolysis by DPP-4 to an inactive metabolite as a disposition mechanism. Due to limited amounts of DPP-4, vildagliptin concentrations increased slightly more than dose proportionally. This newly proposed model and the parameter estimates are supported by published in vitro studies. Mean parameter estimates (inter-individual coefficient of variation) were: non-saturable clearance 36 l h−1 (25%), central volume of distribution 22 l (37%), half-life of dissociation from DPP-4 1.1 h (94%) and half-life of hydrolysis 6.3 h (81%).Vildagliptin is both an inhibitor and substrate for DPP-4. By utilizing the TMDD approach, slow dissociation of vildagliptin from DPP-4 was found in patients and the half-life of hydrolysis by DPP-4 estimated. This model can be used to predict DPP-4 inhibition effects of other dosage regimens and be modified for other DPP-4 inhibitors to differentiate their properties.

Abstract

This article is introducing the latest progress and important results of bio-trickling filter for the purification of biogas. The paper summarized the mechanism of bio-trickling filter, the selection of bacterial species, and the automatical control system of the reactor. The survey also indicated that the main parameters such as pH, dissolved oxygen (DO), residence time and fillers are the critical factors in the bio-desulfurization process. However, this technology is not so immature that more intensive research activities are needed to improve the process of bio-desulfurization. This paper may provide reference for further research in the purification of biogas.
