Abstract

Recent studies have suggested that 5'AMP-activated protein kinase (AMPK) is activated in response to metabolic stresses, such as contraction, hypoxia, and the inhibition of oxidative phosphorylation, which leads to insulin-independent glucose transport in skeletal muscle. In the present study, we hypothesized that acute oxidative stress increases the rate of glucose transport via an AMPK-mediated mechanism. When rat epitrochlearis muscles were isolated and incubated in vitro in Krebs buffer containing the oxidative agent [H.sub.2][O.sub.2], AMPK[alpha]1 activity increased in a time- and dose-dependent manner, whereas AMPK[alpha]2 activity remained unchanged. The activation of AMPK[alpha]1 was associated with phosphorylation of AMPK [Thr.sup.172], suggesting that an upstream kinase is involved in the activation process. [H.sub.2][O.sub.2]-induced AMPK[alpha]1 activation was blocked in the presence of the antioxidant N-acetyl-L-cysteine (NAC), and [H.sub.2][O.sub.2] significantly increased the ratio of oxidized glutathione to glutathione (GSSG/GSH) concentrations, a sensitive marker of oxidative stress. [H.sub.2][O.sub.2] did not cause an increase in the conventional parameters of AMPK activation, such as AMP and AMP/ATP. [H.sub.2][O.sub.2] increased 3-O-methyl-D-glucose transport, and this increase was partially, but significantly, blocked in the presence of NAC. Results were similar when the muscles were incubated in a superoxide-generating system using hypoxanthine and xanthine oxidase. Taken together, our data suggest that acute oxidative stress activates AMPK[alpha]1 in skeletal muscle via an AMP-independent mechanism and leads to an increase in the rate of glucose transport, at least in part, via an AMPK[alpha]1-mediated mechanism. contraction; epitrochlearis muscle; hydrogen peroxide; hypoxanthine: xanthine oxidase

Abstract

The expression of the obese (ob) gene in the adipose tissue of KK mice and congenic lethal yellow obese KKA(super y) mice is dependent on the nutritional status of the mice. The ob mRNA levels in epididymal, mesenteric and subcutaneous white adipose tissue (WAT) are equal in four wks old KK and KKA(super y) mice. However, it increases in the mesenteric and subcutaneous WAT of KKA(super y) mice at 12 wks and the mice develop obesity. Fasting decreases ob gene expression in the epididymal and mesenteric WAT but fails to affect ob gene expression in subcutaneous WAT.

Abstract

The endothelin-B receptor gene (EDNRB) exon 4 in Hirschsprung's disease (HSCR)-affected patients contains a missense G to T mutation that replaces the highly conserved Trp-276 residue by a Cys residue in the fifth membrane helix of the G protein-coupled receptor. A slight damage in the ligand-induced calcium transient levels is seen in the mutant W276C receptor. The risk of HSCR development in W276C heterozygotes and homozygotes is 21% and 74%, respectively. Genotype studies reveal that the HSCR is a multigenic disorder.

Abstract

Natural and targeted piebald-lethal mutations in the endothelin-B receptor (EDNRB) gene in mouse cause aganglionic megacolon with coat color spotting in the mice. The recessive phenotype of the natural piebald-lethal (s') mutations is similar to that of targeted mutations, indicating that the EDNRB gene deletion localizes to the s' chromosome. The effects of the s' mutations indicate their involvement in the growth of two neural-crest-derived cell lineages, the epidermal melanocytes and the myenteric ganglion. Abnormalities in the human EDNRB gene initiate Hirschsprung's disease that is genetically mapped to chromosome 13 that consists of the EDNRB.

Abstract

Targeted endothelin-3 ligand (EDN3) gene disruption in mice produces the same recessive phenotype of coat color spotting and megacolon as does the disruption of endothelin-B receptor gene. A natural recessive EDN3 gene mutation in lethal spotting (ls) mice does not complement the targeted EDN3 allele, indicating that ls is allelic to the EDN3 gene in the mice. A point gene mutation in the ls mice blocks the endothelin-converting enzyme induced proteolytic stimulations between endothelin-B receptor. Thus, interactions between endothelin-B and EDN3 are necessary for growth of neural crest-derived lineages.
