Abstract

Blood pressure (BP) is a major risk factor for cardiovascular disease and more than 200 genetic loci associated with BP are known. Here, we perform a multi-stage genome-wide association study for BP (max N = 289,038) principally in East Asians and meta-analysis in East Asians and Europeans. We report 19 new genetic loci and ancestry-specific BP variants, conforming to a common ancestry-specific variant association model. At 10 unique loci, distinct non-rare ancestry-specific variants colocalize within the same linkage disequilibrium block despite the significantly discordant effects for the proxy shared variants between the ethnic groups. The genome-wide transethnic correlation of causal-variant effect-sizes is 0.898 and 0.851 for systolic and diastolic BP, respectively. Some of the ancestry-specific association signals are also influenced by a selective sweep. Our results provide new evidence for the role of common ancestry-specific variants and natural selection in ethnic differences in complex traits such as BP.

Abstract

Adenoid cystic carcinoma of the breast is a rare histological type of triple-negative breast cancer with an indolent clinical behavior, often driven by the MYB-NFIB fusion gene. Here we sought to define the repertoire of somatic genetic alterations in two adenoid cystic carcinomas associated with high-grade triple-negative breast cancer. The different components of each case were subjected to copy number profiling and massively parallel sequencing targeting all exons and selected regulatory and intronic regions of 488 genes. Reverse transcription PCR and fluorescence in situ hybridization were employed to investigate the presence of the MYB-NFIB translocation. The MYB-NFIB fusion gene was detected in both adenoid cystic carcinomas and their associated high-grade triple-negative breast cancer components. Although the distinct components of both cases displayed similar patterns of gene copy number alterations, massively parallel sequencing analysis revealed intratumor genetic heterogeneity. In case 1, progression from the trabecular adenoid cystic carcinoma to the high-grade triple-negative breast cancer was found to involve clonal shifts with enrichment of mutations affecting EP300, NOTCH1, ERBB2 and FGFR1 in the high-grade triple-negative breast cancer. In case 2, a clonal KMT2C mutation was present in the cribriform adenoid cystic carcinoma, solid adenoid cystic carcinoma and high-grade triple-negative breast cancer components, whereas a mutation affecting MYB was present only in the solid and high-grade triple-negative breast cancer areas and additional three mutations targeting STAG2, KDM6A and CDK12 were restricted to the high-grade triple-negative breast cancer. In conclusion, adenoid cystic carcinomas of the breast with high-grade transformation are underpinned by the MYB-NFIB fusion gene and, akin to other forms of cancer, may be constituted by a mosaic of cancer cell clones at diagnosis. Abstract

Magnetization vs temperature behavior of RuSr[sub 2]GdCu[sub 2]O[sub 8-δ] (Ru-1212) measured in an field of 5 Oe, shows a clear branching of zero-field-cooled (ZFC) and field-cooled (FC) curves around 140 K, a cusp at 135 K, and a diamagnetic transition around 20 K (in the ZFC branch). The cusp at 135 K is due to the antiferromagnetic ordering of the Ru moments. The magnetization-field isotherms, below 50 K, show a nonlinear contribution from a ferromagnetic component. The resistance vs temperature behavior of the compound, in applied fields of 0, 3, and 7 T, confirms that the sample is superconducting at around 20 K. The superconducting transition exhibits field broadening of a type different than that known for conventional high T[sub c] superconductors. The magnetoresistance (MR) is negative above the Ru magnetic ordering temperature of 135 K, while below this temperature, MR displays a positive peak in low fields and becomes negative in higher fields. A maximum of 2% is observed for the negative MR value at the Ru magnetic ordering temperature. An electron diffraction pattern obtained for this Ru-1212 sample shows two types of superstructure; one with a weak spot at the center of the a-b rectangle and the other only along the b direction. It is possible that either Ru/Cu or Ru[sup 4+] / Ru[sup 5+] ordering of 2b periodicity takes place along the b direction. [ABSTRACT FROM AUTHOR]

Abstract

We carried out a trans-ancestry genome-wide association and replication study of blood pressure phenotypes among up to 320,251 individuals of East Asian, European and South Asian ancestry. We find genetic variants at 12 new loci to be associated with blood pressure (P = 3.9 × 10 -11 to 5.0 × 10 -21). The sentinel blood pressure SNPs are enriched for association with DNA methylation at multiple nearby CpG sites, suggesting that, at some of the loci identified, DNA methylation may lie on the regulatory pathway linking sequence variation to blood pressure. The sentinel SNPs at the 12 new loci point to genes involved in vascular smooth muscle (IGFBP3, KCNK3, PDE3A and PRDM6) and renal (ARHGAP24, OSR1, SLC22A7 and TBX2) function. The new and known genetic variants predict increased left ventricular mass, circulating levels of NT-proBNP, and cardiovascular and all-cause mortality (P = 0.04 to 8.6 × 10 -6). Our results provide new evidence for the role of DNA methylation in blood pressure regulation.

Abstract

This goal of this paper is to measure and to make an analysis of the level of the familiar agricultural agribusiness in the state of Rio Grande do Sul, and comparing it with the one for the Brazilian economy as a whole, for the period from 1995 to 2003. Using input-output models it was possible to estimate the importance of the Gross Domestic Product of the familiar agribusiness in the national economy. The results show that around 1/3 of the Brazilian agribusiness comes from the agricultural production done by the familiar agriculture, the numbers are even more stricken for the state of Rio Grande do Sul, where 1/2 of the agribusiness GDP comes from the familiar agriculture. It was also noticed that the recent development of the familiar agriculture and of its links has been very positive, suppressing, the growth rates found in the non-familiar segment.

Lymphoepithelioma‐like carcinoma is fairly uncommon in the breast and cytopathological differential diagnosis of mammary cancers with medullary features requires recognising lymphoepithelial clusters and a fine nuclear chromatin pattern. Tumour‐infiltrating lymphocytes (TILs) should be taken into account not only in histological but also cytological specimens because intratumoural and stromal TILs have value both prognostically and as predictive indicators for responses to chemotherapy. Abstract

This paper aims to present the level of agribusiness family activity in Rio Grande do Sul, for the period 1995 to 2003. Through the input-output models has been possible to estimate the importance of gross domestic product generated by the agribusiness family. The results show that agribusiness family production in Rio Grande do Sul is of high importance in GDP not only in agribusiness, but in the overall state economy.

Abstract

Drosophila laminin alphabetagamma trimer assembly in Kc167 cells was perturbed by chain-specific RNA interference (RNAi). The intracellular pool of alpha and gamma chains remained unchanged under beta-chain RNAi by lipofection of double-stranded RNA encoding a beta-chain partial sequence. This was also the case for the intracellular pool of alpha and beta chains under gamma-chain-specific RNAi. Nonetheless, the intracellular pool of beta and gamma chains increased markedly under alpha-chain-specific RNAi. Non-reducing SDS/PAGE revealed that some of the increased beta and gamma chains migrated as disulphide-linked betagamma dimers but that the rest migrated as monomers. Since the monomeric beta and gamma bands detected under alpha-chain RNAi were denser than the beta band under gamma-chain RNAi and the gamma band under beta-chain RNAi, respectively, beta and gamma also appeared to accumulate by forming betagamma dimers without the disulphide linkage. We suggest that interconversion of these betagamma dimers is crucial for the replaceable and selective assembly of the alpha chain for alphabetagamma trimer formation.

Abstract

Breast cancer is a heterogeneous disease with varied morphological appearances, molecular features, behavior, and response to therapy. Current routine clinical management of breast cancer relies on the availability of robust clinical and pathological prognostic and predictive factors to support clinical and patient decision making in which potentially suitable treatment options are increasingly available. One of the best-established prognostic factors in breast cancer is histological grade, which represents the morphological assessment of tumor biological characteristics and has been shown to be able to generate important information related to the clinical behavior of breast cancers. Genome-wide microarray-based expression profiling studies have unraveled several characteristics of breast cancer biology and have provided further evidence that the biological features captured by histological grade are important in determining tumor behavior. Also, expression profiling studies have generated clinically useful data that have significantly improved our understanding of the biology of breast cancer, and these studies are undergoing evaluation as improved prognostic and predictive tools in clinical practice. Clinical acceptance of these molecular assays will require them to be more than expensive surrogates of established traditional factors such as histological grade. It is essential that they provide additional prognostic or predictive information above and beyond that offered by current parameters. Here, we present an analysis of the validity of histological grade as a prognostic factor and a consensus view on the significance of histological grade and its role in breast cancer classification and staging systems in this era of emerging clinical use of molecular classifiers.

91. Establishing a reproducible protocol for measuring index active extension strength.

Abstract

The goal of this study was to establish a reproducible protocol to measure active extension strength in the index finger. The secondary objectives consisted in correlating the independent or associated index extension strength to the other fingers force of contraction of the extensor indicis propius with hand dominance. The population studied consisted of 24 healthy volunteers, including 19 women and 20 right-handed individuals. The independent and dependent index extension strength in each hand was measured three times with a dynamometer by three examiners at Day 0 and again at Day 7. Intra and inter-examiner reproducibility were, respectively, >0.90 and >0.75 in all cases. The independent extension strength was lower than the dependent one. There was no difference between the independent index extension strength on the dominant and non-dominant sides. The same was true for the dependent strength. Our results show that our protocol is reproducible in measuring independent and dependent index extension strength. Dominance did not come into account.Level of evidence: II [ABSTRACT FROM AUTHOR]
