ObjectiveHypothalamic-pituitary axis dysfunction is a common complication in post-operative craniopharyngioma(CP) patients, and it greatly impacts the long-term quality of life of such patients. To better understand the effects of postoperative hypothalamic-pituitary dysfunction and long-term hormone replacement therapy in patients with childhood CP, we assessed approximately 200 patients with childhood-onset CP postoperatively.MethodsClinical details of patients with childhood-onset CP who underwent sellar tumor resection in Beijing Children’s Hospital and Beijing Tiantan Hospital from 2018 to 2019 were retrieved retrospectively. The participants were followed up to assess the effects of post-operative long-term hormone replacement therapy and assess the tumor recurrence rate.ResultsThe median age of admission was 8.1 (1.8, 14.3) years. Headache (45.5%), visual impairment (39.5%), and nausea (33.0%) were the most common clinical manifestations. ACP accounted for 95% of all CP cases. The incidence of central adrenal insufficiency and central hypothyroidism within the first week after surgery was 56.2% and 70.3%, respectively. At the same time 85.5% of the patients required at least one dose of desmopressin to control urine output. Total survival and tumor recurrence rates were 98.6% and 26.1%, respectively, with a median follow-up time of 29.7 (19.0, 40.3) months. During the follow-up period, 28.1% patients met the diagnostic criteria for short stature, while 54.4% fit the criteria for obesity. In addition, 94.4% of the patients were taking at least one kind of hormone substitution, and 74.7% were taking three or more. The prevalence of levothyroxine, glucocorticoid, desmopressin, and growth hormone replacement therapy was 87.3%, 77.5%, 78.9% and 31.0%, respectively. The proportion of patients treated with the substitutive combination of levothyroxine, hydrocortisone, and desmopressin was 54.9%.ConclusionThis study is a large-sample systematic postoperative endocrine function evaluation of patients with childhood-onset CP. Due to the high prevalence of post-operative hypothalamic-pituitary dysfunction, patients with CP usually require long-term multiple hormone substitution therapy. Individualized management and accurate hormone replacement dosage for postoperative childhood-onset CP patients are of great importance.

Objective: To evaluate the relationship between periodontitis and postmenopausal osteoporosis. Methods: This research was carried out according to the principles laid down by the PRISMA (Preferred Reporting Items for Systematic Reviews and Meta-Analyses) guideline statement. We searched the Web of Science, Embase, PubMed, The Cochrane Library, CNKI, VIP, and WanFang databases from inception to July 1, 2023 to collect all relevant publications, with no restrictions on publication date or Languages. Cochrane's tool for assessing RoB was used to evaluate the RoB for RCTs. The Newcastle-Ottawa Scale was used to assess the RoB for cohort studies and case-control studies. Mean differences (MD) with 95 % confidence intervals (CI) were used for analysis of continuous data. Heterogeneity was measured using the I2 statistic. Revman 5.4 software was used for the meta-analysis. Results: 28 observational studies with 19611 patients, including 5813 cases in the postmenopausal osteoporosis group and 13798 cases in the non-osteoporosis group. The habitats of oyster reefs had played various ecological services functions, as an important means of marine ecological restoration and fishery resources conservation, which had been widely used worldwide. And the characteristics of microbial communities would be inevitably affected by habitat of oyster reefs. The aim of this study was to reveal the relationship between oyster reefs and the intestinal microbiota of fish (Takifugu ocellatus, Acanthopagrus latus, Clupanodon thrissa, and Mugil cephalus). Here, based on 16S rRNA sequencing, this research analyzed the microbial communities of fishes gut in the oyster reefs from the most crucial estuary economic zone in Southern China. It was significantly lower that the bacterial alpha-diversity in the fish intestines than in the sediments of oyster reefs. The non-metric multidimensional scaling (NMDS) and analysis of similarities (ANOSIM) results showed that intestines microbiota of T. ocellatus and A. latus were more homologous to sediments microbiota, but intestines microbiota of C. thrissa and M. cephalus were more similar to the water microbiota. There were obvious differences in the contribution of oyster reefs habitat to intestinal microorganisms of four fishes. Source tracking analysis as well reached the similar conclusion. For instance, contribution of water to T. ocellatus and A. latus was lesser than sediments, which was related to their benthic habits. Based on the differences in microbial community compositions, the functional prediction analysis had demonstrated that special functional pathways and independent core indicators had been formed separately from oyster reefs and fish intestines. Overall, these findings highlighted the characteristics of microbial diversity and functions from the omnivorous fish living in oyster reefs habitats. This research proposed new insights into the function of oyster reefs habitats, which have enriched the ecological service value of oyster reefs from microbial ecology and provided a reference for the application of oyster reefs habitats in other estuaries.

The efficient synthesis of myofibrillar protein(MRN)-gallic acid (GAD) complex in ultrasound (UID)-assisted processing is a challenging problem in food manufacturing. In this investigation, the effect of viscosity characteristics on the efficiency of UID processing in MRN-based beverages was analyzed. Both viscosity and surface tension can increase sono-physico-chemical effects on the degradation of terephthalic acid and crystal violet, with surface tension having a more significant effect (negative correlation, R2 = 0.99) than viscosity (positive correlation, R2 = 0.79). The structural indicators and microstructure demonstrated that the reaggregation and refolding of the MRN structure during the modification procedure occurred with relatively small three-dimensional dimensions. Compared to the MRN/GAD4 group, the water contact angle of the MRN/GAD7 system enhanced by 129.44%, leading to greater system stability. The ABTS-scavenging capacity of the system increased by approximately 19.45% due to the increase in viscosity of these two categories.

There was no statistically significant difference in postpartum hemorrhage (PPH) incidence between the aspirin and control groups. This study aimed to evaluate the potential bleeding risk of 100 mg aspirin in high-risk pregnant women and the difference in the incidence of PPH according to maternal characteristics. Methods:. SiC fibers have important application in aerospace and other high-tech equipment due to their excellent mechanical properties and thermal resistance. In this work, SiC fibers with diameters of 9, 14, 60 μm were prepared in order to investigate the influence of different diameters on the precursor-derived SiC fibers. The results showed that the polycrystalline structure and near stoichiometric composition of the SiC-9 and SiC-14 fibers were achieved by the “one-step” approach. However, the sintering for SiC-60 fiber was insufficient due to the largest diameter. Moreover, the tensile strength of SiC fibers decreased from 1.88 GPa to only 0.28 GPa with increment of the fiber diameter. The fibers exhibited excellent high temperature resistance with tensile strength retention reaching over 94% after being kept at 1900 °C in argon for 1 h. Furthermore, the SiC fibers with sintered and densified microstructure showed good oxidation resistance. This work could help to further optimize the structure of the polycrystalline near stoichiometric SiC fibers for higher mechanical and thermal performance.

The power transformer is an essential equipment in the UHV transmission system. High energy discharge resulting from arc faults in the transformer turret can lead to an explosion, posing a serious threat to the safe and stable operation of the power system. At present, there is a lack of experimental research on arc discharge faults in the transformer turret. The ignition and explosion process remains unclear, which limits the improvement of transformer explosion-proof performance. To address this issue, a test platform of arc discharge fault in the transformer turret was constructed. High-current and high-deflagration capacity simulation short-circuit tests were carried out, and the voltage and current waveforms in the arc discharge process were obtained. This facilitated the quantitative characterization of discharge energy and analysis of the arc energy flow conversion. The test results highlighted that arc energy and arc current are the important factors affecting the voltage boost in the riser. The arc process of casing rupture under high current (1400 A) can be divided into two stages: “smooth” and “sudden”. The insulating oil after high energy discharge and the gas escaping from the oil were analyzed by chromatographic methods. The results revealed that the oil cracking occurs after high energy discharge, significantly increasing various fault characteristic gases in the oil. Among these gases, H2, CH4, C2H2, and C2H4 exhibited the highest total content in the oil, while H2, CH4, and C2H4 presented the greatest range in the escaped gas, accounting for more than 75% of the total volume. This work elucidates the physical mechanism of the internal short-circuit fault of the transformer turret, and holds crucial guiding significance for the transformer explosion-proof design.

There is an accumulating body of evidence implicating the muscarinic acetylcholine receptor 4 (M4) in schizophrenia and dementia with Lewy bodies, however, a clinically validated M4 positron emission tomography (PET) radioligand is currently lacking. As such, the aim of this study was to develop a suitable M4 PET ligand that allows the non-invasive visualization of M4 in the brain. Structure–activity relationship studies of pyrazol-4-yl-pyridine derivates led to the discovery of target compound 12 ― a subtype-selective positive allosteric modulator (PAM). The radiofluorinated analogue, [18F]12, was synthesized in 28 ± 10% radiochemical yield, >37 GBq/μmol and an excellent radiochemical purity >99%. Initial in vitro autoradiograms on rodent brain sections were performed in the absence of carbachol and showed moderate specificity as well as a low selectivity of [18F]12 for the M4-rich striatum. However, in the presence of carbachol, a significant increase in tracer binding was observed in the rat striatum, which was reduced by >60% under blocking conditions, thus indicating that orthosteric ligand interaction is required for efficient binding of [18F]12 to the allosteric site. Remarkably, however, the presence of carbachol was not required for high specific binding in the non-human primate (NHP) and human striatum, and did not further improve the specificity and selectivity of [18F]12 in higher species. These results pointed towards significant species-differences and paved the way for a preliminary PET study in NHP, where peak brain uptake of [18F]12 was found in the putamen and temporal cortex. In conclusion, we report on the identification and preclinical development of the first radiofluorinated M4 PET radioligand with promising attributes. The availability of a clinically validated M4 PET radioligand harbors potential to facilitate drug development and provide a useful diagnostic tool for non-invasive imaging.

To aid cocrystal screening, a deep forest-based cocrystal prediction model was developed in this study using data from the Cambridge Structural Database (CSD). The positive samples in the experiment came from the CSD. The negative samples were partly from the failure records in other papers, and some were randomly generated according to specific rules, resulting in a total of 8576 pairs. Compared with the models of traditional machine learning methods and simple deep neural networks models, the deep forest model has better performance and faster training speed. The accuracy is about 95% on the test set. Febuxostat cocrystal screening was also tested to verify the validity of the model. Our model correctly predicted the formation of cocrystal. It shows that our model is practically useful in practice.

Non-alcoholic fatty liver disease (NAFLD) is a common chronic hepatic condition whose impact on human health is increasingly significant. The imbalance of the gut microbiome, linked to insulin resistance, heightened intestinal permeability, and pro-inflammatory reactions, may be the linchpin in the development of NAFLD. In our research, the impact of Lactiplantibacillus plantarum ZDY2013 administration for 12 weeks on gut microbiota dysbiosis induced by a high-fat, high-fructose, high-cholesterol (FHHC) diet in male C57BL/6n mice was investigated. Research results presented that the intervention of L. plantarum ZDY2013 in mice fed with the FHHC diet could restore their liver function and regulate oxidative stress. Compared to mice in the model group, the intervention of L. plantarum ZDY2013 significantly regulated the gut microbiota, inhibited the LPS/NF-κB pathway, and led to a lower level of colonic inflammation in the mice administered with L. plantarum ZDY2013. It also improved insulin resistance to regulate the PI3K/Akt pathway and lipid metabolism, thereby resulting in reduced fat accumulation in the liver. The above results suggest that the intervention of L. plantarum ZDY2013 can hinder the progression of diet-induced NAFLD by reducing inflammation to regulate the PI3K/Akt pathway and regulating gut microbiota disturbance.

Tomato is the vegetable with the largest greenhouse area in China, and low temperature is one of the main factors affecting tomato growth, yield, and quality. Hydrogen sulfide (H2S) plays an important role in regulating plant chilling tolerance, but its downstream cascade reaction and mechanism remain unclear. Mitogen-activated protein kinases (MAPK/MPKs) are closely related to a variety of signaling substances in stress signal transmission. However, whether H2S is related to the MPK cascade pathway in response to low-temperature stress is rarely reported. In this study, NaHS treatment significantly decreased the electrolyte leakage (EL), superoxide anion (O2−) production rate, and hydrogen peroxide (H2O2) content of seedlings at low temperatures. In addition, the activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) were obviously increased; and the photochemical efficiency of PSII (Fv/Fm) was enhanced with treatment with NaHS, indicating that NaHS improved the seedlings’ cold tolerance by alleviating the degree of membrane lipid peroxidation and oxidative damage. However, H2S scavenger hypotaurine (HT) treatment showed the opposite effect. We found that H2S content, L-cysteine desulfhydrase (LCD) activity, and mRNA expression were increased by chilling stress but reduced by MPK inhibitor PD98059; PD98059 reversed the alleviating effect of H2S via increasing the EL and H2O2 contents. The expression levels of MPK1–MPK7 at low temperatures showed that SlMPK4 was significantly induced by exogenous NaHS and showed a trend of first increasing and then decreasing, while the expression level of SlMPK4 in HT-treated seedlings was lower than that of the control. After SlMPK4 was silenced by virus-induced gene silencing, the H2S-induced upregulation of C-repeat-Binding Factor (CBF1), inducer of CBF expression 1 (ICE1), respiratory burst oxidase homologs (RBOH1, RBOH2) at low temperatures disappeared, and tomato cold tolerance decreased. In conclusion, H2S improves the cold tolerance of tomato plants by increasing the activity of antioxidant enzymes and reducing reactive oxygen species (ROS) accumulation and membrane lipid peroxidation. MPK4 may act as a downstream signaling molecule in this process.

This study focuses on exploring the effects of niobium (Nb)-microalloying on the properties of steel for ultra-high-strength bridge cables during hot-rolling processes. We employed a combination of dual-pass compression tests, stress–strain curve analysis, and Electron Backscatter Diffraction (EBSD) techniques to investigate the influence of Nb-microalloying on the static recrystallization behavior and grain size of the steel. The key findings reveal that Nb-microalloying effectively inhibits static recrystallization, particularly at higher temperatures, significantly reducing the volume fraction of recrystallized grains, resulting in a finer grain size and enhanced deformation resistance. Secondly, at a deformation temperature of 975 °C, Nb-containing steel exhibited finer grain sizes compared to Nb-free steel when held for 10 to 50 s; however, the grain size growth accelerated when the hold time exceeded 50 s, likely linked to the increased deformation resistance induced by Nb. Lastly, this research proposes optimal hot-rolling process parameters for new bridge cable steel, recommending specific finishing rolling temperatures and inter-pass times for both Nb-containing and Nb-free steels during the roughing and finishing stages. This study suggests optimal hot-rolling parameters for both Nb-containing and Nb-free steels, providing essential insights for improving hot-rolling and microalloying processes in high-carbon steels for bridge cables.

CircRNA, a recently characterized non-coding RNA (ncRNA) variant, functions as a molecular sponge, exerting regulatory control by binding to microRNA (miRNA) and modulating the expression of downstream proteins, either promoting or inhibiting their expression. Among poultry species, geese hold significant importance, prized by consumers for their delectable taste and rich nutritional content. Despite the prominence of geese, research on the growth and development of goose muscle, particularly the regulatory role of circRNAs in goose muscle formation, remains insufficiently explored. In this study, we constructed comprehensive expression profiles of circRNAs and messenger RNAs (mRNAs) within the myoblasts and myotubes of Shitou geese. We identified a total of 96 differentially expressed circRNAs (DEcircRNAs) and 880 differentially expressed mRNAs (DEmRNAs). Notably, the parental genes of DEcircRNAs and DEmRNAs exhibited enrichment in the Wnt signaling pathway, highlighting its potential impact on the proliferation and differentiation of goose myoblasts. Employing RNAhybrid and miRDB, we identified circRNA-miRNA pairs and mRNA-miRNA pairs that may play a role in regulating myogenic differentiation or muscle growth. Subsequently, utilizing Cytoscape, we constructed a circRNA–miRNA–mRNA interaction network aimed at unraveling the intricate regulatory mechanisms involved in goose muscle growth and development, which comprises 93 circRNAs, 351 miRNAs, and 305 mRNAs. Moreover, the identification of 10 hub genes (ACTB, ACTN1, BDNF, PDGFRA, MYL1, EFNA5, MYSM1, THBS1, ITGA8, and ELN) potentially linked to myogenesis, along with the exploration of their circRNA–miRNA–hub gene regulatory axis, was also conducted. These competitive endogenous RNA (ceRNA) regulatory networks elucidate the molecular regulatory mechanisms associated with muscle growth in Shitou geese, providing deeper insights into the reciprocal regulation of circRNA, miRNA, and mRNA in the context of goose muscle formation.

China’s recent collective forest tenure reform (CFTR) aims to improve forest conditions by devolution of forest management rights from community collectives to individual households. Studies reveal substantial local variation in CFTR implementation, but the ecological effects and underlying mechanism have received little attention. Our study focused on community-managed forests and household-managed forests in four communities located in the mountains of Southwest China. The objective was to investigate the relationship between CFTR and forest conditions at the local scale. We employed a combination of remote sensing technology, field forest observations and socioeconomic surveys to gather data for analysis. The results showed that the forest cover change trends of collective forests and household forests in each community were consistent from 1994 to 2014. There was no significant relationship between CFTR implementation and local-level differences in species diversity and soil fertility in forests. The aboveground biomass of collective forests was significantly higher than that of household forests. Differentiated access and use, low compensation amounts, and loose payment terms jointly affected CFTR’s effectiveness to improve local-scale forest conditions in the study area. Attempts to formalize forest tenure rights must be flexible enough and adapted to the local environment so that changes induced by the CFTR can improve local access to forest resources and its benefits.

Summary: Overly strong fear memories can cause pathological conditions. Histamine H3 receptor (H3R) has been viewed as an optimal drug target for CNS disorders, but its role in fear memory remains elusive. We find that a selective deficit of H3R in cholinergic neurons, but not in glutamatergic neurons, enhances freezing level during contextual fear memory retrieval without affecting cued memory. Consistently, genetically knocking down H3R or chemogenetically activating cholinergic neurons in the ventral basal forebrain (vBF) mimics this enhanced fear memory, whereas the freezing augmentation is rescued by re-expressing H3R or chemogenetic inhibition of vBF cholinergic neurons. Spatiotemporal regulation of H3R by a light-sensitive rhodopsin-H3R fusion protein suggests that postsynaptic H3Rs in vBF cholinergic neurons, but not presynaptic H3Rs of cholinergic projections in the dorsal hippocampus, are responsible for modulating contextual fear memory. Therefore, precise modulation of H3R in a cell-type- and subcellular-location-specific manner should be explored for pathological fear memory.

Genipin is a natural crosslinker that improves the functional properties of proteins by modifying its structures. This study aimed to investigate the effects of sonication on the emulsifying properties of different genipin concentration-induced myofibrillar protein (MP) cross-linking. The structural characteristics, solubility, emulsifying properties, and rheological properties of genipin-induced MP crosslinking without sonication (Native), sonication before crosslinking (UMP), and sonication after crosslinking (MPU) treatments were determined, and the interaction between genipin and MP were estimated by molecular docking. The results demonstrated that hydrogen bond might be the main forces for genipin binding to the MP, and 0.5 μM/mg genipin was a desirable concentration for protein cross-linking to improve MP emulsion stability. Ultrasound treatment before and after crosslinking were better than Native treatment to improve the emulsifying stability index (ESI) of MP. Among the three treatment groups at the 0.5 μM/mg genipin treatment, the MPU treatment group showed the smallest size, most uniform protein particle distribution, and the highest ESI (59.89%). Additionally, the highest α-helix (41.96%) in the MPU + G5 group may be conducive to the formation of a stable and multilayer oil–water interface. Furthermore, the free groups, solubility, and protein exposure extent of the MPU groups were higher than those of UMP and Native groups. Therefore, this work suggests that the treatment of cross-linking followed by ultrasound (MPU) could be a desirable approach for improving the emulsifying stability of MP.

Tetracycline (TC) is widely used in the livestock industry, but undigested TC is excreted with livestock waste and accumulates in the environment. In this study, swine manure-derived biochar (SBC) was modified with KMnO4 (MnOx-SBC), and used to remove TC. SEM-EDS, FTIR, XPS and elemental analysis all indicated that ultrafine MnOx particles were attached to the biochar surface. The surface properties and composition of the oxygen-containing functional groups were enhanced by KMnO4 modification. Batch sorption experiments showed that MnOx-SBC's TC-adsorption capacity was 105.9 mg·g−1, 46.4% higher than SBC's. The TC-adsorption onto MnOx-SBC agreed well with the pseudo-second-order model and Freundlich isotherm. A new platform is proposed for reusing swine manure while solving the livestock industry's antibiotic pollution risk by ‘treating waste with waste’. HIGHLIGHTS TC adsorption capacity of swine manure derived biochar enhanced after KMnO4 modification.; Initial solution pH, temperature and initial solution concentration affected TC removal.; Pseudo-second-order and Freundlich models can well fit the adsorption behavior.; Adsorption mechanism included pore-filling, surface complexation, π-π interactions and H-bonding.;

Abstract COX7A1, a subunit of cytochrome c oxidase, holds an important position in the super-assembly which integrates into multi-unit heteromeric complexes peripherally in the mitochondrial electron transport chain (ETC). Recently, some studies indicated the significant potential of COX7A1 in cancer metabolism and therapy. However, the underlying metabolic process and therapy mechanism remain unclear. In this study, COX7A1-overexpressed cell line was established via lentivirus transduction. The relationship between COX7A1 and ferroptosis, a novel form of cell death driven by iron-dependent lipid peroxidation, was further analyzed in different human non-small-cell lung carcinoma (NSCLC) cells respectively. Our results showed that COX7A1 increased the sensitivity of NSCLC cells to the ferroptosis induced by cysteine deprivation via enhancing the tricarboxylic acid (TCA) cycle and the activity of complex IV in mitochondrial ETC. Meanwhile, COX7A1 suppressed mitochondrial dynamics as well as mitochondrial biogenesis and mitophagy through blocking autophagic flux. The autophagy activator, rapamycin, relieved the autophagic blockage and further strengthened the sensitivity to cysteine deprivation-induced ferroptosis of NSCLC cells in vitro and in vivo. Taken together, our data indicate the close association of COX7A1 with cysteine deprivation-induced ferroptosis, and provide a novel insight into the therapy mode against human NSCLC.

Abstract This study mainly studied sensory and metabolite migration from the skin to the soup in the boiling process of tilapia skin using content analysis, electronic nose technique, electronic tongue technique, and metabolomics technique based on ultra-high performance liquid chromatography-mass spectrometry/mass spectrometry and gas chromatography-time-of-flight-mass spectrometry. The content changes, flavor changes, taste changes, metabolite numbers and differential metabolite numbers for both tilapia skin and soup mainly occurred in the initial 30 min. Moreover, the initial 10 min was the key period for the metabolite changes in the boiling process. Further, the differential metabolites in these three periods (0–10, 10–30, and 30–60 min) were identified to show the metabolites migration process. Six (adenine, gingerol, terephthalic acid, vanillin, pentanenitrile, and 2-pyrrolidinonede) and seven (butyramide, lysope(0:0/20:4(5z,8z,11z,14z)), lysope(22:6(4z,7z,10z,13z,16z,19z)/0:0), linoleic acid, N-acetylneuraminic acid, L-threose, and benzoin) chemicals were screened out in the differential metabolites of tilapia skin and soup, respectively, with Variable Importance in the Projection of >1 and p value of

nARs are ligand-activated transcription factors associated with gonadal development and reproductive regulation. In this study, two nuclear androgen receptor genes (ara and arb) were cloned from an ovoviviparous teleost, black rockfish (Sebastes schlegelii). The phylogenetic analysis of nARs showed that nARα and nARβ clustered into teleost nARα and nARβ, respectively, but differed from the nARs of tetrapods. Four module domains of the nuclear receptor superfamily are present in the black rockfish nARs, an N-terminal domain (NTD), a DNA-binding domain (DBD), a hinge region (HR) and a ligand-binding domain (LBD). Among the four domains, only the DBD and LBD of nARα and nARβ are relatively conserved. Tissue distribution analysis revealed that ara was mainly expressed in the gonad, muscle, intestine and kidney in males and in female, while arb was mainly detected in the gonad, followed by the intestine, kidney and head kidney. Abstract Doxorubicin (DOX)-based chemotherapy is widely used to treat malignant tumors; however, the cardiotoxicity induced by DOX restricts its clinical usage. A therapeutic dose of DOX can activate ubiquitin-proteasome system. However, whether and how ubiquitin-proteasome system brings out DOX-induced cardiotoxicity remains to be investigated. Here we conducted a proteomics analysis of a DOX-induced cardiotoxicity model to screen the potentially ubiquitination-related molecules. Dysregulated TRIM25 was found to contribute to the cardiotoxicity. In vivo and in vitro cardiotoxicity experiments revealed that TRIM25 ameliorated DOX-induced cardiotoxicity. Electron microscopy and endoplasmic reticulum stress markers revealed that TRIM25 mitigated endoplasmic reticulum stress and apoptosis in DOX-induced cardiomyocytes. Mechanistically, the Co-immunoprecipitation assays and CHX pulse-chase experiment determined that TRIM25 affected p85α stability and promoted its ubiquitination and degradation. This leads to increase of nuclear translocation of XBP-1s, which mitigates endoplasmic reticulum stress. These findings reveal that TRIM25 may have a therapeutic role for DOX-induced cardiotoxicity.

This work studied the physicochemical properties and odor profiles of tilapia muscles after exposure to four types of thermal processing methods: microwaving, roasting, boiling, or steaming. The effect of thermal processing on textural properties followed a pH–water state–water content–tissue microstructure–mass loss–textural properties route, expressed in the following manner: microwaving > roasting > steaming ≈ boiling. After processing, muscle pH increased from 6.59 ± 0.10 to 6.73 ± 0.04–7.01 ± 0.06, and hardness changed from 1468.49 ± 180.77 g to 452.76 ± 46.94–10723.66 ± 2898.46 g. Gas chromatography-based E-nose analysis confirmed that these methods had significant odor fingerprint effects on the tilapia muscles. Finally, the combined analysis of headspace solid-phase microextraction–gas chromatography–mass spectrometry, statistical MetaboAnalyst, and odor activity value showed that the microwaved, roasted, steamed, and boiled tilapia muscles had, respectively, three (hexanal, nonanal, and decanal), four (2-methyl-butanal, 3-methyl-butanal, decanal, and trimethylamine), one (2-methyl-butanal), and one (decanal) relatively important volatile compounds.

This study aimed to examine the sono-physico-chemical effects of ultrasound (UND) and its impact on the conjugate rates of morin (MOI) following the addition of polysaccharides in various conditions. In comparison to the control group, the incorporation of quaternary ammonium chitosan decreased the rate of MOI conjugation by 17.38%, but the addition of locust bean gum enhanced the grafting rate by 29.89%. Notably, the highest degree of myofibrillar protein (MRN) unfolding (fluorescence intensity: 114435.50), the most stable state (−44.98 mV), and the greatest specific surface area (393.06 cm2/cm3) were observed in the UMP/LBG group. The outcomes of atomic force microscopy and scanning electron microscopy revealed that the inclusion of locust bean gum led to a different microscopic morphology than the other two polysaccharides, which may be the primary cause of the strongest sono-physico-chemical effects of the system. This work demonstrated that acoustic settings can be tuned based on the characteristics of polysaccharides to maximize the advantages of sono-physico-chemical impacts in UND-assisted MOI processing.

Optimizing ultrasound (ULD)-assisted flavonoid modification is an important component of enhancing its application potential. In this work, diverse flavonoids, such as quercetin (Que), apigenin (Api), and morin (Mor), were used to modify protein in myofibrillar protein (MP)/cellulose nanocrystal (CN) complexes using ULD-assisted method. Compared with the MP/CNs group, the triiodide contents of MP-Que/CNs, MP-Api/CNs, and MP-Mor/CNs increased by 1175.84%, 479.05%, and 2281.50% respectively. The findings revealed that the actual intensity of ULD was drastically reduced by the molecular weight decrease of these flavonoids. For olive oil emulsions prepared with mixed emulsifiers, the low interfacial diffusion rates (0.03 mN·m·s−1/2) and weak emulsifying activity (8.33 m2/g) of the MP/CN complexes were significantly improved by the flavonoids after ULD-assisted treatment. Notably, the emulsions prepared using MP-Api/CNs contained smaller oil droplets and exhibited better emulsifying properties, compared to emulsions prepared with MP-Mor/CNs or MP-Que/CNs. This study is essential for ULD-assisted treatment since the processing impact may be increased by choosing the most suitable flavonoid.

Located on the southern margin of the Qinghai-Tibet Plateau, the Northwest Yunnan-Southeast Tibet (NYST) is a biodiversity hotspot. However, plant diversity in the region faces enormous threats, including forest fragmentation and overexploitation of natural resources. Here, we updated the catalogue of higher plants in NYST by integrating various data sources and referring to the newest results of molecular systematics and taxonomy. Combining phylogenetic approaches and a spatial analysis of forest landscape fragmentation, we assessed spatial variations in taxonomic richness and phylogenetic diversity, and identified potential geographic units for conservation gaps. NYST harbours nearly one-third of China's higher plant species, of which ∼60% of the species listed as threatened are endemic. Among the genera, the highest number of endemic and threatened species belong to Rhododendron. The largest increase in the number of threatened species was observed in the Orchidaceae over the last 15 years. Seed plant taxonomic richness and phylogenetic diversity were significantly positively correlated at both the species and genus levels. Counties with large mountains are hotspots of phylogenetic diversity as well as total, endemic, and threatened species richness. The pattern of diversity of higher plants in the study area is likely related to the Himalayan-Tibetan Plateau uplift. The western part of Yulong County and the central to southern parts of Shangri-La County are conservation gaps in this area. In-depth local field surveys of plant diversity are needed, especially in those counties where there is large disparity between the number of species present and collected specimens (e.g., Yulong County). This study provides data and a theoretical basis for the study and conservation of NYST plant diversity.

Polypropylene melt-blown nonwoven fabric (PP MNF) masks can effectively block pathogens in the environment from entering the human body. However, the adhesion of surviving pathogens to masks poses a risk of human infection. Thus, embedding safe and efficient antibacterial materials is the key to solving pathogen infection. In this study, stable chlorinated poly(methacrylamide-N,N′-methylenebisacrylamide) polypropylene melt-blown nonwoven fabrics (PP-P(MAA-MBAA)-Cl MNFs) have been fabricated by a simple UV cross-link and chlorination process, and the active chlorine content can reach 3500 ppm. The PP-P(MAA-MBAA)-Cl MNFs show excellent hydrophilic and antibacterial properties. The PP-P(MAA-MBAA)-Cl MNFs could kill all bacteria (both Escherichia coli and Staphylococcus aureus) with only 5 min of contact. Therefore, incorporating PP-P(MAA-MBAA)-Cl MNF as a hydrophilic antimicrobial layer into a four-layer PP-based mask holds great potential for enhancing protection and comfort.

Objectives: Translocator protein 18 kDa (TSPO) positron emission tomography (PET) can be harnessed for the non-invasive detection of macrophage-driven inflammation. [18F]LW223, a newly reported TSPO PET tracer which was insensitive to rs6971 polymorphism, showed favorable performance characteristics in a recent imaging study involving a rat myocardial infarction model. To enable quantitative neuroimaging with [18F]LW223, we conducted kinetic analysis in the non-human primate (NHP) brain. Further, we sought to assess the utility of [18F]LW223-based TSPO imaging in a first-in-human study. Methods: Radiosynthesis of [18F]LW223 was accomplished on an automated module, whereas molar activities, stability in formulation, lipophilicity and unbound free fraction (fu) of the probe were measured. Brain penetration and target specificity of [18F]LW223 in NHPs were corroborated by PET-MR imaging under baseline and pre-blocking conditions using the validated TSPO inhibitor, (R)-PK11195, at doses ranging from 5 to 10 mg/kg. Kinetic modeling was performed using one-tissue compartment model (1TCM), two-tissue compartment model (2TCM) and Logan graphical analyses, using dynamic PET data acquisition, arterial blood collection and metabolic stability testing. Clinical PET scans were performed in two healthy volunteers (HVs). Regional brain standard uptake value ratio (SUVr) was assessed for different time intervals. Results: [18F]LW223 was synthesized in non-decay corrected radiochemical yields (n.d.c. RCYs) of 33.3 ± 6.5% with molar activities ranging from 1.8 ± 0.7 Ci/µmol (n = 11). [18F]LW223 was stable in formulation for up to 4 h and LogD7.4 of 2.31 ± 0.13 (n = 6) and fu of 5.80 ± 1.42% (n = 6) were determined. [18F]LW223 exhibited good brain penetration in NHPs, with a peak SUV value of ca. 1.79 in the whole brain. Pre-treatment with (R)-PK11195 substantially accelerated the washout and attenuated the area under the time-activity curve, indicating in vivo specificity of [18F]LW223 towards TSPO. Kinetic modeling demonstrated that 2TCM was the most suitable model for [18F]LW223-based neuroimaging. Global transfer rate constants (K1) and total volumes of distribution (VT) were found to be 0.10 ± 0.01 mL/cm3/min and 2.30 ± 0.17 mL/cm3, respectively. Dynamic PET data analyses across distinct time windows revealed that the VT values were relatively stable after 60 min post-injection. In a preliminary clinical study with two healthy volunteers, [18F]LW223 exhibited good brain uptake and considerable tracer retention across all analyzed brain regions. Greenhouse gas emissions from human activities have become the leading cause of climate problems. Reducing greenhouse gas emissions from human economic activities and realizing carbon neutralization are the main means of sustainable economic development. Among them, carbon emission reduction of large-scale activities including auctions bears the brunt. Through the emission factor method, this paper estimates the carbon footprint of a typical artwork auction and divides the auction market into different sizes, according to the average round-trip distance of the number of participants. The results show that a typical 3-day medium-sized artwork auction with 500 people’s carbon footprint is about 270 tons of carbon dioxide. The traffic carbon emission of participants accounts for a large proportion of the total carbon footprint, particularly composed of the traffic carbon emissions of a small proportion of long-distance participants. Therefore, the transition from offline to virtual artwork auctions can significantly reduce the carbon footprint by 90%–95%. We put forward suggestions on improving the auction carbon footprint accounting process and industry carbon neutralization system, and promoting the development of relevant technologies for the virtual artwork auction market.

Using computer simulation tools such as finite element analysis (FEA) to perform material stress analysis is a common design method in engineering practice. In order to model more realistic real‐world systems, simulation models have become more complex, and calculation becomes more expensive as a result. The rise of artificial intelligence technologies has made it possible to integrate deep learning methods and material stress analysis. Herein, FEA software is employed to obtain a large number of analysis cases as training samples and uses a fully connected neural network and long‐short‐term memory neural network as surrogate models, which can predict the stress distribution and stress sequence in the process of the bullet impacting target plates with different materials. These models can give results similar to FEA with 92.19% and 92.41% accuracy, respectively. The experimental results show that the deep learning surrogate models have great potential in material stress analysis.

The blockchain technology system has gradually come to be employed in the food supply chain system, and it has emerged that the system offers the unique function of effectively curbing counterfeiting by food manufacturers. Unlike previous research on adoption by enterprises of new technology, this paper probes into the specific evolutionary routes of game subjects from the perspective of the precondition for enterprises' willingness to employ blockchain technology on the basis of China's social co-governance framework and by establishing a tripartite evolutionary game model of food manufacturer, government and consumer. The study then tests and verifies the stability conditions of equilibrium points and the relationship between these equilibrium points and the social co-governance level through numerical simulation analysis. On the above basis, the expected market proceeds of food producers employing blockchain technology and the influence of government and consumer behavior on enterprises' selection of a behavior strategy and the level of social co-governance are analyzed. The results show that every subject selects their own behavior strategy on the basis of the balance of their respective interests, and the final stability condition of the system is independent of their initial intentions. Rather, the expected sales volume of foods employing blockchain technology, governmental behavior (e.g., supervision, casual inspection, economic punishment, and fiscal subsidies), and complaints made by consumers constitute the main factors that influence food enterprises' selection of a behavior strategy. The level of social co-governance and the behavior of both government and consumers will ultimately be accomplished by influencing enterprises' expected economic returns, and the selection of an enterprise behavior strategy internally depends on the expected economic returns from producing foods employing blockchain technology. Therefore, this paper makes relevant proposals in an attempt to assist the Chinese government to better promote and popularize the blockchain technology system among food manufacturing enterprises.

In recent years, sustainable development has become a focal issue in the globalization process due to environmental uncertainty in the global context, which has attracted the extensive attention of scholars. This study explores the dynamic transmission mechanism and time-varying impacts among green finance, renewable energy and sustainable development in China on the basis of a stochastic volatile time-varying vector autoregressive (TVP-VAR-SV) model using time-series data from January 4, 2013, to October 14, 2022. The results revealed that there is an interaction among green finance, renewable energy and sustainable development in China, and the degree of influence and direction of action at different points in time show time-varying and heterogeneous characteristics. Further, this study subdivides renewable energy into hydro, wind, geothermal and solar energy. The dynamic impact of these types of energy on sustainable development is heterogeneous, and this relationship is affected by relevant economic events.

Lipid metabolism disorders contribute to hyperlipidemia and hepatic steatosis. It is ideal to develop drugs simultaneous improving both hyperlipidemia and hepatic steatosis. Nitazoxanide is an FDA-approved oral antiprotozoal drug with excellent pharmacokinetic and safety profile. We found that nitazoxanide and its metabolite tizoxanide induced mild mitochondrial uncoupling and subsequently activated AMPK in HepG2 cells. Gavage administration of nitazoxanide inhibited high-fat diet (HFD)-induced increases of liver weight, blood and liver lipids, and ameliorated HFD-induced renal lipid accumulation in hamsters. Nitazoxanide significantly improved HFD-induced histopathologic changes of hamster livers. In the hamsters with pre-existing hyperlipidemia and hepatic steatosis, nitazoxanide also showed therapeutic effect. Gavage administration of nitazoxanide improved HFD-induced hepatic steatosis in C57BL/6J mice and western diet (WD)-induced hepatic steatosis in Apoe–/– mice. The present study suggests that repurposing nitazoxanide as a drug for hyperlipidemia and hepatic steatosis treatment is promising.

Monoacylglycerol lipase (MAGL) is a pivotal enzyme in the endocannabinoid system, which metabolizes 2-arachidonoylglycerol (2-AG) into the proinflammatory eicosanoid precursor arachidonic acid (AA). MAGL and other endogenous cannabinoid (EC) degrading enzymes are involved in the fibrogenic signaling pathways that induce hepatic stellate cell (HSC) activation and ECM accumulation during chronic liver disease. Our group recently developed an 18F-labeled MAGL inhibitor ([18F]MAGL-4-11) for PET imaging and demonstrated highly specific binding in vitro and in vivo. In this study, we determined [18F]MAGL-4-11 PET enabled imaging MAGL levels in the bile duct ligation (BDL) and carbon tetrachloride (CCl4) models of liver cirrhosis; we also assessed the hepatic gene expression of the enzymes involved with EC system including MAGL, NAPE-PLD, FAAH and DAGL that as a function of disease severity in these models; [18F]MAGL-4-11 autoradiography was performed to assess tracer binding in frozen liver sections both in animal and human. [18F]MAGL-4-11 demonstrated reduced PET signals in early stages of fibrosis and further significantly decreased with disease progression compared with control mice. We confirmed MAGL and FAAH expression decreases with fibrosis severity, while its levels in normal liver tissue are high; in contrast, the EC synthetic enzymes NAPE-PLD and DAGL are enhanced in these different fibrosis models. In vitro autoradiography further supported that [18F]MAGL-4-11 bound specifically to MAGL in both animal and human fibrotic liver tissues. Our PET ligand [18F]MAGL-4-11 shows excellent sensitivity and specificity for MAGL visualization in vivo and accurately reflects the histological stages of liver fibrosis in preclinical models and human liver tissues.

The RSA is one of the current default cryptosystems that provides security with applications such as encryptions and digital signatures. It is important to further study the weak characteristics of the RSA to ensure correct utilisation in order not to be susceptible to adversaries. In this paper, we give detailed analysis on security of the Murru-Saettone variant of the RSA cryptosystem that utilised a cubic Pell $ed-k\left ({p^{2}+p+1}\right )\left ({q^{2}+q+1}\right )=1$ as key equation and $N=pq$ as RSA modulus. We propose some attacks on this variant when the prime difference $|p-q|$ is small. Our first approach is to utilise the continued fractions algorithm to determine the parameter $d$ which enables us to determine the secret $p$ and $q$ . Our second approach considers the Coppersmith's method and lattice basis reduction to factor the modulus $N$ . Our attacks improve recent cryptanalyses on the cubic Pell equation variant of RSA. Furthermore, our attacks prove that under small prime difference scenario, the number of susceptible private exponents for the cubic Pell equation variant of RSA is much larger than the standard RSA.

Covalent organic frameworks (COFs) have unique features, including intrinsic porosity, crystallinity, and tunability, making them desirable materials for diverse applications ranging from environmental remediation to energy harvesting. Among these applications, COFs are extensively studied for their photocatalytic hydrogen evolution by converting solar energy into clean and renewable fuel via water splitting. COFs have several advantages over conventional inorganic catalysts, such as tunable band structures, high surface areas, and low cost. However, the research in this field is still in the early stages, and COFs still face some challenges, such as low charge carrier mobility, high exciton binding energy, and poor stability. To overcome these challenges, various design strategies relying on a mechanistic approach have been developed to design and modify COFs for enhanced photocatalytic performance. These include extending the π-conjugation, incorporating heteroatoms or metal complexes, and donor–acceptor (D–A) configuration, which ultimately improves the light absorption charge separation of COFs. Additionally, blending COFs with other functional materials, such as inorganic-organic semiconductors, can create synergistic effects to boost photocatalytic activity. In this review, the design aspects of the fabrication of COFs as effective photocatalysts have been reported.

The unique odors of yeast proteins (YPs) are decisive for their application in meat substitutes. Sensory evaluation, electronic nose, and gas chromatography–mass spectrometry/olfactory (GC-MS/O) were combined to characterize the aroma profiles and aroma-active compounds of YPs. The sensory evaluation results indicate that the sweaty aroma had the strongest intensity in YP #10, followed by rice bran, sour, and plastic. The electronic nose could effectively distinguish the aroma differences among five YPs. A total of 27 aroma-active compounds in the five YPs were identified by GC-MS/O. The concentration of 2-methyl-propanoic acid (6.37 μg/kg), butanoic acid (47.46 μg/kg), 3-methyl-butanoic acid (22.50 μg/kg), and indole (943.40 μg/kg) in YP #10’s aroma was higher than that of the other YPs. The partial least squares regression method results show that o-cresol, (3S)-3,7-dimethyloct-7-en-1-ol, benzyl alcohol, octanal, 2-methyl-propanoic acid, butanoic acid, 3-methyl-butanoic acid, hexanal, heptanal, and indole were predicted as the potential aroma-active compounds significantly contributing to the aroma profiles of the five YPs. Addition experiments confirmed that the overall aroma profile intensities of the five YP samples were extended with the addition of these ten compounds, verifying their significant contributions.

FKBP10, a member of the FK506-binding protein (FKBP) family, has been implicated in cancer development, although its prognostic function remains controversial. In this study, we analyzed the expression of FKBP10 in tumor tissues using online databases (TCGA) as well as our CRC cohort, and investigated the relationship between its subcellular expression pattern and patient outcomes. Cox regression analysis was used to determine the associations between different subcellular expression patterns of FKBP10 and clinical features of patients. We also discussed the expression level of FKBP10 based on different subcellular expression patterns. Our results showed that FKBP10 was significantly elevated in CRC tissues and exhibited three different subcellular expression patterns which were defined as ‘FKBP10-C’ (concentrated), ‘FKBP10-T’ (transitional) and ‘FKBP10-D’ (dispersive). The FKBP10-D expression pattern was only found in tumor tissues and was associated with unfavorable disease-free survival in CRC patients. High expression levels of FKBP10-C predicted an unfavorable prognosis of recurrence of CRC, while FKBP10-D did not. Our findings suggest that the subcellular expression patterns and expression level of FKBP10 play crucial prognostic roles in CRC, which revealed that FKBP10 may be a viable prognostic and therapeutic target for the diagnosis and treatment of CRC.

The direction of human gaze is an important indicator of human behavior, reflecting the level of attention and cognitive state towards various visual stimuli in the environment. Convolutional neural networks have achieved good performance in gaze estimation tasks, but their global modeling capability is limited, making it difficult to further improve prediction performance. In recent years, transformer models have been introduced for gaze estimation and have achieved state-of-the-art performance. However, their slicing-and-mapping mechanism for processing local image patches can compromise local spatial information. Moreover, the single down-sampling rate and fixed-size tokens are not suitable for multiscale feature learning in gaze estimation tasks. To overcome these limitations, this study introduces a Swin Transformer for gaze estimation and designs two network architectures: a pure Swin Transformer gaze estimation model (SwinT-GE) and a hybrid gaze estimation model that combines convolutional structures with SwinT-GE (Res-Swin-GE). SwinT-GE uses the tiny version of the Swin Transformer for gaze estimation. Res-Swin-GE replaces the slicing-and-mapping mechanism of SwinT-GE with convolutional structures. Experimental results demonstrate that Res-Swin-GE significantly outperforms SwinT-GE, exhibiting strong competitiveness on the MpiiFaceGaze dataset and achieving a 7.5% performance improvement over existing state-of-the-art methods on the Eyediap dataset.

Abstract Background Transmembrane proteins are vital for intercellular signalling and play important roles in the control of cell fate. However, their physiological functions and mechanisms of action in myogenesis and muscle disorders remain largely unexplored. It has been found that transmembrane protein 182 (TMEM182) is dramatically up‐regulated during myogenesis, but its detailed functions remain unclear. This study aimed to analyse the function of TMEM182 during myogenesis and muscle regeneration. Methods RNA sequencing, quantitative real‐time polymerase chain reaction, and immunofluorescence approaches were used to analyse TMEM182 expression during myoblast differentiation. A dual‐luciferase reporter assay was used to identify the promoter region of the TMEM182 gene, and a chromatin immunoprecipitation assay was used to investigate the regulation TMEM182 transcription by MyoD. We used chickens and TMEM182‐knockout mice as in vivo models to examine the function of TMEM182 in muscle growth and muscle regeneration. Chickens and mouse primary myoblasts were used to extend the findings to in vitro effects on myoblast differentiation and fusion. Co‐immunoprecipitation and mass spectrometry were used to identify the interaction between TMEM182 and integrin beta 1 (ITGB1). The molecular mechanism by which TMEM182 regulates myogenesis and muscle regeneration was examined by Transwell migration, cell wound healing, adhesion, glutathione‐S‐transferse pull down, protein purification, and RNA immunoprecipitation assays. Results TMEM182 was specifically expressed in skeletal muscle and adipose tissue and was regulated at the transcriptional level by the myogenic regulatory factor MyoD1. Functionally, TMEM182 inhibited myoblast differentiation and fusion. The in vivo studies indicated that TMEM182 induced muscle fibre atrophy and delayed muscle regeneration. TMEM182 knockout in mice led to significant increases in body weight, muscle mass, muscle fibre number, and muscle fibre diameter. Skeletal muscle regeneration was accelerated in TMEM182‐knockout mice. Furthermore, we revealed that the inhibitory roles of TMEM182 in skeletal muscle depend on ITGB1, an essential membrane receptor involved in cell adhesion and muscle formation. TMEM182 directly interacted with ITGB1, and this interaction required an extracellular hybrid domain of ITGB1 (aa 387–470) and a conserved region (aa 52–62) within the large extracellular loop of TMEM182. Mechanistically, TMEM182 modulated ITGB1 activation by coordinating the association between ITGB1 and laminin and regulating the intracellular signalling of ITGB1. Myogenic deletion of TMEM182 increased the binding activity of ITGB1 to laminin and induced the activation of the FAK‐ERK and FAK‐Akt signalling axes during myogenesis. Conclusions Our data reveal that TMEM182 is a novel negative regulator of myogenic differentiation and muscle regeneration.

Abstract In this work, lipid profile migration from muscle to juice during the tilapia muscle steaming process was revealed by a transactional analysis of data from ultra-high-performance liquid chromatography coupled with Q Exactive (UHPLC-QE) Orbitrap mass spectrometry (MS) and lipidomics. Firstly, the lipids in tilapia muscles and juices at different steaming time points were extracted and examined by UHPLC-QE Orbitrap mass spectrometry. Secondly, a transactional analysis procedure was developed to analyze the data from UHPLC-QE Orbitrap MS and lipidomics. Finally, the corrected lipidomics data and the normalized MS data were used for lipid migration analysis. The results suggested that the transactional analysis procedure was efficient to significantly decrease UHPLC-QE Orbitrap MS workloads and delete the false-positive data (22.4–36.7%) in lipidomics data, which compensated the disadvantages of the current lipidomics method. The lipid changes could be disappearance, full migration into juice, appearance in juice, appearance in muscle, appearance in both muscle and juice, and retention in the muscle. Moreover, the results showed 9 (compared with 52), 5 (compared with 116), and 10 (compared with 178) of lipid class (compared with individual lipid) variables showed significant differences among the different steaming times (0, 10, 30, and 60 min) in all the muscles, juices, and muscle-juice systems, respectively. These results showed significant lipid profile migration from muscle to juice during the tilapia steaming process.

Abstract Background Ovarian cancer, a highly metastatic malignancy, has benefited tremendously from advances in modern human genomics. However, the genomic variations related to the metastasis remains unclear. Methods We filtered various significant genes (n = 6722) associated with metastasis within a large-scale functional genomic CRISPR/Cas9 knock-out library including 122,756 single guide RNAs, and identified ITK (IL2 Inducible T Cell Kinase) as a potential cancer suppressor gene for ovarian cancer metastasis. Downstream bioinformatic analysis was performed for ITK using public databases. Results We found that patients in low-ITK group had poor prognosis and more distant metastasis than those in high-ITK group in TCGA and GEO databases. We also demonstrated that ITK combined with the clinical factors could accurately predict prognosis through multiple Cox regression analysis and ROC analysis. Moreover, alterations correlated with distant metastasis emereged with significantly increased expression in SAMRCD1 in low-ITK group, but CD244 and SOCS1 in high-ITK group. Integrated analysis revealed dysregulated molecular processes including predominantly oncogenic signaling pathways in low-ITK group but immune related pathways in high-ITK group, which suggested ITK might inhibit distant metastasis in ovarian cancer. Furtherly, deconvolution of the cellular composition of all samples validated the close correlation between ITK and immune related function especially for cytotoxic lymphocytes. Conclusions Together, these data provide insights into the potential role of ITK, with implications for the future development of tansformative ovarian cancer therapeutics.

The exploration of deodorization is important for the application of gelatin in food industry. In this work, the effect of β-cyclodextrin (β-CD) deodorization on the volatile chemicals and functional properties of three types of gelatins (commercial porcine skin gelatin, cold water fish skin gelatin, and Chinese longsnout catfish skin gelatin) were studied. The results suggested the odors of commercial gelatins were significantly less than home-extracted gelatins. The β-CD deodorization efficiency was dependent on both β-CD concentration and volatile chemical. (E)-2-Octenal (C8H14O), 1-octen-3-ol (C8H16O), 2-pentyl-furan (C9H14O), and hentriacontane (C17H36) could be deodorized at low β-CD concentration (even at 2 mg/mL). The best β-CD deodorization concentration for 66.7 mg/mL of Chinese longsnout catfish skin gelatin was 30 mg/mL. β-CD addition could not change the gel forming ability and emulsion activity of gelatins, whereas it had different and concentration-dependent effects on the emulsion stability of gelatins. β-CD addition had no obvious effects on the droplet sizes, droplet coalescence and liquid-gel transition behaviors, but had different effects on the creaming of the emulsions stabilized by three types of gelatins. The encapsulation of β-carotene did not significantly change the droplet trimodal size distribution and liquid-gel transition of fish oil-loaded emulsions. However, β-carotene might delay the droplet coalescence. The creaming stability of β-carotene/fish oil-loaded gelatin/β-CD-stabilized emulsions was dependent on the gelatins, β-CD, and β-carotene. Finally, the β-carotene retention in the emulsions was dependent not on β-CD addition but on the nature of the gelatins. These results provided useful information to understand the molecular deodorization behaviors and explore the deodorization of emulsifiers for food emulsions.

Machine learning has shown great potential in bridging materials structures and properties. However, efficient and general descriptors for materials structures are still missing. This work dedicates to establishing an efficient and self-adaptable materials structure descriptor based on the graph attention network (GAT). In the GAT descriptor generation, the concrete structure is represented by graphic atomic clusters (atoms as nodes, edges as bonds) consisting of one central atom and the surroundings. A multi-head-GAT is applied to gather the short- and long-distance topological information from the graphical atomic clusters to generate dimension-flexible descriptors regardless of atom number; the GAT descriptors are contracted and help avoiding overfitting during the training process theoretically. Further verification of the GAT descriptors is carried out by performing the phase classification, energy prediction, and potential energy surface (PES) prediction task for Al-Cu alloys with complex precipitates and defects. The GAT descriptors adapt themselves for cross tasks without manual intervention and produce accurate predictions that agree with the density functional theory (DFT) results. The GAT descriptors are also testified as differentiable, universal, and precise on structural characterization containing point-, line-, and planar- defects. Further applications will be found for the materials design field demanding accurate structure descriptions.

Estuaries are one of the most important ecosystems with high biological productivity. They connect freshwater and marine ecosystems, and the fish community assemblages face intense environmental changes, so estuarine fish communities have unique ecological characteristics. With the increase of human activities and the discharge of pollutants, the pressure on the ecosystem of the Pearl River Estuary (PRE) is intensifying, and its ecosystem functions and services are inevitably affected. To better understand the functional structure of the fish community in the PRE and its response to environmental changes, we analyzed the trait composition of the community and its relationships to environmental factors based on 10 functional traits of 130 fish species collected in the PRE during the summer and winter of 2020–2021. In addition, we built a multi-dimensional functional space to visualize the overall functional structure of the PRE and explore the seasonal variation of the selected traits in the functional space. Our findings suggest that the functional traits of the fish communities in the PRE were mainly affected by temperature, dissolved oxygen, chlorophyll-a, and depth. Among them, the effects of dissolved oxygen, temperature, and chlorophyll-a on CWM traits were reflected in seasonal differences, while the effects of depth were reflected in spatial differences. We also found the main trait categories that occupied the most functional space and contributed the most to the seasonal differences in functional space. The results of this study are significant for understanding the seasonal variation of the functional structure in the PRE and how environmental gradients drive the functional traits of fish communities, and also provide insights for effective ecosystem-based management in the PRE.

We conducted a phase I bioequivalence trial in healthy Chinese subjects in the fasting and postprandial states. The goal of this trial was to compare the pharmacokinetics and safety of the test preparation Cefaclor granule (Disha Pharmaceutical Group Co., Ltd.) and the reference preparation Cefaclor suspension (Ceclor®, Eli Lilly and Company). In this trial, 24 subjects were selected in the fasting and postprandial states, respectively. Enrolled subjects randomly accepted a single dose of 0.125 g Cefaclor granule or Cefaclor suspension. The washout period was set as 2 days. Blood samples were collected within 8 h after administration in the fasting state and within 10 h after administration in the postprandial state. Plasma concentrations were determined by Liquid chromatography-tandem mass spectrometry (LC-MS/MS). Pharmacokinetic parameters (AUC, Cmax) were used to evaluate bioequivalence of the two drugs. In the fasting trial, the geometric mean ratios (90% confidence intervals CIs) for Cmax, AUC0-t, and AUC0-∞ were 93.01% (85.96%–100.63%), 97.92% (96.49%–99.38%) and 97.95% (96.52%–99.41%), respectively. The GMR (90% CIs) for Cmax, AUC0-t, and AUC0-∞ in postprandial state were 89.27% (81.97%–97.22%), 97.31% (95.98%–98.65%) and 97.31% (95.93%–98.71%), respectively. The 90% CIs of AUC and Cmax in the fasting and postprandial states were within the 80–125% bioequivalence range. Therefore, Cefaclor granule and Cefaclor suspension were bioequivalent and displayed similar safety profiles. Furthermore, food intake affected the pharmacokinetic parameters of both drugs.

Abstract Background Foot-and-mouth disease (FMD) is a highly contagious disease of livestock worldwide. Russia is a big agricultural country with a wide geographical area where FMD outbreaks have become an obstacle for the development of the animal and animal products trade. In this study, we aimed to assess the export risk of FMD from Russia. Results After simulation by Monte Carlo, the results showed that the probability of cattle infected with FMD in the surveillance zone (Surrounding the areas where no epidemic disease has occurred within the prescribed time limit, the construction of buffer areas is called surveillance zone.) of Russia was 1.29 × 10− 6. The probability that at least one FMD positive case was exported from Russia per year in the surveillance zone was 6 %. The predicted number of positive cattle of the 39,530 - 50,576 exported from Russia per year was 0.06. A key node in the impact model was the probability of occurrence of FMD outbreaks in the Russian surveillance zone. By semi-quantitative model calculation, the risk probability of FMD defense system defects was 1.84 × 10− 5, indicating that there was a potential risk in the prevention and control measures of FMD in Russia. The spatial time scan model found that the most likely FMD cluster (P
