51. Production and preliminary characterization of monoclonal antibodies highly specific to pork fat protein

Abstract: Meat and food products adulterated with pork fat concern Islamic consumers whose dietary laws prohibit consumption of pork meat. The objectives of this study are to investigate the existence of thermal stable-soluble protein (TSSP) in pork fat tissues and to develop and characterize monoclonal antibodies (MAbs) highly specific to the TSSP in pork fat tissues. The TSSP extracted was used as an immunogen. After cell fusion and cloning, seven monoclonal antibodies (MAbs; PF 2B8-3, 2B8-9, 2B8-20, 2B8-28, 2B8-31, 2B8-32, and 2B8-33) were developed and confirmed to be specific to pork fat protein without cross-reaction to other animal meats and fats in an indirect ELISA and Western blot. The ELISA assay based on MAb PF 2B8-31 can sensitively detect 1% pork fat protein mixed in other animal meats. These results support that the developed MAbs especially PF 2B8-31 can be sufficiently used as a bio-receptor in the development of immunoassay for the easy and rapid detection of pork fat adulteration. In addition, the ELISA based on the MAbs developed could be a useful tool to rapidly detect low levels of pork fat fraudfully adulterated in meat and food products.

Abstract: This study analyzed contents of 12 kinds of middle school technology·Home Economics① textbooks, 9 high school Technology·Home Economics textbooks and 2 high school home science textbooks based on the 5 areas and 9 subject areas of `Understanding of market and consumer`, `Reasonable consumption`, `Resolution and prevention of consumer problems`, `Formation of desirable consumption culture`, `Career and occupation related to consumption life` for the purpose of analyzing connectivity of contents in the area of `consumption life` in the Technology·Home Economics curriculum revised in 2009, and analyzed connectivity of contents on the basis of `developed`, `repeated`, `different` and `reduced` based on the results of analysis on the contents of the textbooks. Analysis results show that middle school Technology·Home Economics① textbooks are mainly dealing with problems related to consumption life and ways to practice healthy consumption life, high school Technology·Home Economics textbooks emphasized understanding of consumption culture in modern life and practice and method of sustainable consumption life, and high school home science textbooks contain overall contents of household financial management and consumption life. In general, contents were distributed as `developed` 11 times (32.4%), `repeated` 6 times (17.6%), `reduced` 13 times (38.2%), `different` 4 times (11.8%). Contents of the majority of textbooks are composed suitable for the curriculum achievement standard, but connectivity of `developed` cannot be considered a desirable type of connectivity. various formation plans as suggested shall be devised not to let learning elements of `consumption life` area discontinued considering school levels and achievement standard of curriculum.

Abstract: Estrogen affects endometrial cellular proliferation by regulating the expression of the c-myc gene. B-cell translocation gene 1 (BTG1), a translocation partner of the c-myc, is a tumor suppressor gene that promotes apoptosis and negatively regulates cellular proliferation and cell-to-cell adhesion. The aim of this study was to determine the role of BTG1 in the pathogenesis of endometriosis. BTG1 mRNA and protein expression was evaluated in eutopic and ectopic endometrium of 30 patients with endometriosis (endometriosis group), and in eutopic endometrium of 22 patients without endometriosis (control group). The effect of BTG1 downregulation on cellular migration, proliferation, and apoptosis was evaluated using transfection of primarily cultured human endometrial stromal cells (HESCs) with BTG1 siRNA. BTG1 mRNA expression level of eutopic and ectopic endometrium of endometriosis group were significantly lower than that of the eutopic endometrium of the control group. Migration and wound healing assays revealed that BTG1 downregulation resulted in a significant increase in migration potential of HESCs, characterized by increased expression of matrix metalloproteinase 2 (MMP2) and MMP9. Downregulation of BTG1 in HESCs significantly reduced Caspase 3 expression, indicating a decrease in apoptotic potential. In conclusion, our data suggest that downregulation of BTG1 plays an important role in the pathogenesis of endometriosis.

Abstract: The flat spectrum radio quasar 4C 38.41 showed a significant increase of its radio flux density during the period 2012 March - 2015 August which correlates with gamma-ray flaring activity. Multi-frequency simultaneous VLBI observations were conducted as part of the interferometric monitoring of gamma-ray bright active galactic nuclei (iMOGABA) program and supplemented with additional monitoring observations at various bands across the electromagnetic spectrum. The epochs of the maxima for the two largest gamma-ray flares coincide with the ejection of two respective new VLBI components and the evolution of the physical properties seem to be in agreement with the shock-in-jet model. Derived synchrotron self absorption magnetic fields, of the order of 0.1 mG, do not seem to dramatically change during the flares, and are much smaller, by a factor 10,000, than the estimated equipartition magnetic fields, indicating that the source of the flare may be associated with a particle dominated emitting region.

Abstract: The JEM-EUSO (Joint Experiment Missions for the Extreme Universe Space Observatory) program aims at developing Ultra-Violet (UV) fluorescence telescopes for efficient detections of Extensive Air Showers (EASs) induced by Ultra-High Energy Cosmic Rays (UHECRs) from satellite orbit. In order to demonstrate key technologies for JEM-EUSO, we constructed the EUSO-Balloon instrument that consists of a ∼1 m 2 refractive telescope with two Fresnel lenses and an array of multi-anode photo-multiplier tubes at the focus. Distinguishing it from the former balloon-borne experiments, EUSO-Balloon has the capabilities of single photon counting with a gate time of 2.3 µs and of imaging with a total of 2304 pixels. As a pathfinder mission, the instrument was launched for an 8 h stratospheric flight on a moonless night in August 2014 over Timmins, Canada. In this work, we analyze the count rates over ∼2.5 h intervals. The measurements are of diffuse light, e.g. of airglow emission, back-scattered from the Earth's atmosphere as well as artificial light sources. Count rates from such diffuse light are a background for EAS detections in future missions and relevant factor for the analysis of EAS events. We also obtain the geographical distribution of the count rates over a ∼780 km 2 area along the balloon trajectory. In developed areas, light sources such as the airport, mines, and factories are clearly identified. This demonstrates the correct location of signals that will be required for the EAS analysis in future missions. Although a precise determination of count rates is relevant for the existing instruments, the absolute intensity of diffuse light is deduced for the limited conditions by assuming spectra models and considering simulations of the instrument response. Abstract: We present the intraday flaring activity of Cyg X-3 in the rise phase of a major flare observed in November-December 2008 at 22 GHz with the VLBI Exploration of Radio Astrometry (VERA). The two consecutive intraday flares with a time scale of ~ 3 - 4 hours are similar in their light curve morphologies. The intraday flares observed in 2008 are compared to other interferometric observations of intraday flares in Cyg X-3, as well as to those observed in GRS 1915+105. The intraday flares observed in Cyg X-3 can be classified into three types: flares with a fast rise and exponential decay (type A), flares with an almost equal time scale in the rise and decay (type B), and asymmetric flares with a slow rise and rapid decay (type C). Such types of intraday flares have also been observed in another representative microquasar GRS 1915+105, and are, therefore, a common characteristic in both microquasars. The long-term multiwavelengh light curves in 2007 and 2008 are similar except during a few-day-long transition from an unltrasoft to harder states. In the transition state, the intraday flares of type A were observed in the 2008 VERA observation whereas, in the 2007 VERA observation, a type C flare was observed, opposite to the flares observed in 2008. The 2007 observation is the only observation of a type C flare in Cyg X-3. A possible cause of the different types of intraday flares is briefly discussed in the context of the jet propagation process.

Abstract: EUSO-Balloon is a pathfinder mission for the Extreme Universe Space Observatory onboard the Japanese Experiment Module (JEM-EUSO). It was launched on the moonless night of the 25th of August 2014 from Timmins, Canada. The flight ended successfully after maintaining the target altitude of 38 km for five hours. One part of the mission was a 2.5 hour underflight using a helicopter equipped with three UV light sources (LED, xenon flasher and laser) to perform an inflight calibration and examine the detectors capability to measure tracks moving at the speed of light. We describe the helicopter laser system and details of the underflight as well as how the laser tracks were recorded and found in the data. These are the first recorded laser tracks measured from a fluorescence detector looking down on the atmosphere. Abstract: The current investigation was performed to evaluate the chemical analytical techniques for PAHs (organic, carcinogenic and mutagenic compounds) in food. It also determines the content of eight polycyclic aromatic hydrocarbons (PAHs) in frequently consumed fruit, vegetables, meats and their products. The methodology included saponification or ultrasonication, liquid-liquid extraction with solvents like n-hexane, clean-up using a silica solid phase extraction cartridge and a gas chromatography-mass spectrometry technique. A good linearity (R2 > 0.99) was achieved for the PAHs at different concentrations. Recovery results for PAHs extended from 88.75 to ∼100.00%. The limit of detection was 0.08–0.25 µg/kg and the limit of quantification was 0.24–0.75 µg/kg. The mean concentration (n = 3) of benzo[a]pyrene was not identified in fruits, was 0.05 µg/kg in vegetables, and 0.64 µg/kg in meat products. Overall concentration of the eight PAHs was 0.67 µg/kg in fruits, 0.82 µg/kg in vegetables, and 3.37 µg/kg in meat products.

Abstract: The flat spectrum radio quasar 1633+382 (4C~38.41) showed a significant increase of its radio flux density during the period 2012 March - 2015 August which correlates with gamma-ray flaring activity. Multi-frequency simultaneous VLBI observations were conducted as part of the interferometric monitoring of gamma-ray bright active galactic nuclei (iMOGABA) program and supplemented with additional radio monitoring observations with the OVRO 40m telescope, the Boston University VLBI program, and the Submillimeter Array. The epochs of the maxima for the two largest gamma-ray flares coincide with the ejection of two respective new VLBI components. Analysis of the spectral energy distribution indicates a higher turnover frequency after the flaring events. The evolution of the flare in the turnover frequency - turnover flux density plane probes the adiabatic losses in agreement with the shock-in-jet model. Derived synchrotron self absorption magnetic fields, of the order of 0.1mG, do not seem to dramatically change during the flares, and are much smaller, by a factor $10^4$, than the estimated equipartition magnetic fields, indicating that the source of the flare may be associated with a particle dominated emitting region., 12 pages, 7 figures, 4 tables. Abstract: EUSO-TA is a ground-based telescope, installed at the Telescope Array (TA) site in Black Rock Mesa, Utah, USA. This is the first detector to successfully use a Fresnel lens based optical system and multi-anode photomultipliers (64 channels per tube, 2304 channels encompassing a 10.6° × 10.6° field of view) for detection of Ultra High Energy Cosmic Rays (UHECR). The telescope is located in front of one of the fluorescence detectors of the TA experiment. Since its installation in 2013, the detector has observed several ultra-high energy cosmic ray events and, in addition, meteors. The limiting magnitude of 5.5 on summed frames (∼ 3 ms) has been established. Measurements of the UV night sky emission in different conditions and moon phases and positions have been completed. Abstract: We present multi-frequency simultaneous VLBI radio observations of the flat spectrum radio quasar 1633+382 (4C~38.41) as part of the interferometric monitoring of gamma-ray bright active galactic nuclei (iMOGABA) program combined with additional observations in radio, optical, X-rays and $\gamma-$rays carried out between the period 2012 March - 2015 August. The monitoring of this source reveals a significant long-lived increase in its activity since approximately two years in the radio bands, which correlates with a similar increase on all other bands from sub-millimeter to $\gamma-$rays. A significant correlation is also found between radio fluxes and simultaneous spectral indices during this period. The study of the discrete correlation function (DCF) indicates time lags smaller than the $\sim40$ days uncertainties among both radio bands and also high-energy bands, and a time lag of $\sim$70 days, with $\gamma-$rays leading radio. We interpret that the high-energy and radio fluxes are arising from different emitting regions, located at $1\pm12$ and $40\pm13$ pc from the central engine respectively., Comment: 16 pages, 15 figures, and 5 tables. Abstract: The aim of this study was to investigate microbiological contamination levels in the manufacturingcompany of ginseng products (white and red ginseng). Firstly, the contamination level for ginseng and each stage gin-seng were 1.8~4.9 log CFU/g (total bacteria), 1.2~3.0 log CFU/g (coliform), 0.8~4.1 log CFU/g (fungi). However,only Bacillus cereus among pathogenic bacteria was detected from a few sample. The contamination of total bacteriatended to decrease as ginseng was being processing. Therefore, that of finished products (white and red ginseng)showed the lowest contamination level among each stage ginseng sample. That of fungi decreased steadily, althoughthe contamination of fungi has tended to increase right after ginseng was steamed. Secondly, the contamination levelfor working tools and facilities were 1.7~4.7 log CFU/cm 2 (total bacteria), 0.4~4.0 log CFU/cm 2 (coliform), 0.9~4.2log CFU/cm 2 (fungi). Especially, washing and peeling machines were higher contamination level. Finally, the contam-ination level of worker who washed and steamed ginseng was higher than worker who shaped, sorted and stored gin-seng. Also, Staphylococus aureus was detected at 0.2~0.7 log CFU/hand on some wokers’ hands. These resultsshowed proper heating condition (temperature and time) and tidy manufacturing facility are the most important toavoid developing any microbiological problem of Ginseng Products.Key words : ginseng products, red ginseng, white ginseng, manufacturing process, microbial contamination

Abstract: This study is to investigate microbiological hazards which can be used as fundamental data to ade-quately control leeks hazards and develop leeks GAP model for those who want to get GAP system. The microbiolog-ical investigations on cultivation environments (soil and water), crops (leeks), personal hygiene (workers’ hands,clothes and gloves) and working tools (boxes) have been conducted for one year, so the period was classified undernon-cultivation, cultivation, and post harvest. Total bacteria was detected from soil (4.0~6.7 log CFU/g), leeks(4.6~5.1 log CFU/g), hands (ND~3.3 log CFU/hand) and gloves (ND~5.4 log CFU/cm 2 ) while nothing was detectedfrom the other samples. The coliform contamination of leeks (4.8~5.0 log CFU/g) was more high than that of soil(3.9~4.2 log CFU/g). In case of foodborne pathogens, only B. cereus was detected at the level of 0.5~4.6 log CFU/g(or hand, 100 cm 2 ). Fungi was observed at the level of 2.1~3.8 log CFU/g (or hand, 100 cm 2 ) excepting water andsome working tools. These results demonstrate that the contamination of leeks is comparatively higher than that of soilsample. The reason may be the cross-contamination by biological hazards presenting on soil. Abstract: The study aims to review airline services like the domestic line performance, the general flight fares and website discount fairs, reservation cancellation and cancellation charges, reroutings and personal discount system of the low cost carriers(LCC), and suggest what needs to be done to improve the airline service. The improvements to be made by the LCC should include increasing regular domestic lines, providing information compared for consumers, relieving the toll to call the service center, revising the conditions on reservation cancellation penalty, rerouting charges, revising the consumer dispute resolution standards, and promoting frequent fliers program. Development strategies should be established that can boost consumer benefits through the improvement of the airline services. Abstract: Aims: Our objective was to predict natural pregnancy after endometriosis surgery using the endometriosis fertility index (EFI). Methods: A retrospective medical records review was conducted, examining patients surgically treated for endometriosis at a single center in Korea between January 2009 and February 2015. In total, 68 women attempting natural conception were analyzed by assessing age, preoperative serum CA-125, body mass index, revised American Fertility Society (rAFS) stage, EFI, and pregnancy outcome. Kaplan-Meier estimates and log-rank tests were used to generate cumulative natural pregnancy rate curves based on an EFI cut-point. A receiver-operating characteristic (ROC) curve was plotted for EFI. Results: Seventy-seven patients attempted conceptions, resulting in 33 natural and 9 assisted conceptions. Excluding assisted conceptions, the mean EFI scores of 68 women who were not pregnant and pregnant were 5.43 ± 0.36 and 6.88 ± 0.28 respectively. The relation between EFI and natural pregnancy was significant (cumulative overall pregnancy rate, p = 0.006), whereas rAFS stage was not (univariate logistics, p = 0.853). The cut-point for maximum natural pregnancy outcomes was 6 (area under ROC curve = 0.710, 95% CI 0.586–0.835). Conclusion: The EFI is a reliable staging system for predicting natural pregnancy after endometriosis surgery. EFI scores can be used to guide postoperative treatment of women with endometriosis.

Abstract: Ochratoxin A (OTA), a mycotoxin classified as a potentially carcinogenic to humans, is slightly soluble in water. In this study, the transfer rate of OTA in herbal medicines to decoctions was investigated. Herbal medicine decoctions were prepared by boiling and autoclaving herbal medicines contaminated with OTA after soaking and not soaking in water. OTA amounts in decoctions and residues were analyzed using HPLC. Transfer of OTA (12.72–61.33%) from herbal medicines to decoctions was observed. Soaking and autoclaving steps enhanced the transfer rate of OTA to decoctions. The results meant that there is a potential risk for consumption of herbal medicine decoctions. Therefore, regular monitoring for mycotoxins, including OTA, in herbal medicines and proper management to ensure the safety of herbal medicines is necessary to protect consumers.

Abstract: UNLABELLED The objective of this study was to develop a 1-step simultaneous lateral flow strip test for the rapid and simple detection of deoxynivalenol (DON) and zearalenone (ZEA) in grains. Two monoclonal antibodies (MAbs) against DON and ZEA were respectively conjugated with gold nanoparticles and used to develop a lateral flow strip test for a single toxin and multiple toxins. First, individual lateral flow strips for a single toxin were optimized, and their conditions were used to develop a simultaneous lateral flow strip for multiple toxins. Limits of detection of both lateral flow strip tests for DON and ZEA were the same (DON: 50 ng/mL, ZEA: 1 ng/mL). Both methods showed cross-reactivity for α-zearalenol and β-zearalenol, but no cross-reaction to other mycotoxins. The results can be completed obtained within 15 min. The cut-off values of the simultaneous lateral flow strip for the spiked rice and corn were 500 and 10 ng/g for DON and ZEA, respectively. The results demonstrated that the developed simultaneous lateral flow strip test offers a rapid, easy-to-use, and portable analytical system and can be used as a convenient qualitative tool for the on-site detection of DON and ZEA in food and agricultural commodities. PRACTICAL APPLICATION Simultaneous lateral strip test is useful for a rapid detection of DON and ZEA at a time in food and grain samples.

Abstract: This research is focused on coagulation and sand filtration process as a pretreatment of RO seawater desalination. RO systems require sufficient and reliable pretreatment process to produce superior quality of RO feedwater that can mitigate RO membrane fouling. This experiment was conducted to investigate the effectiveness of coagulation and filtration process under various experimental conditions including different coagulant dose, flocculation mixing intensity and time, turbidity, and filtration rate. The experimental results showed that the optimum pretreatment conditions resulting in lower SDI value suitable for RO feedwater were coagulation pH 6.5, raw water turbidity greater than 4 NTU, and media bed depth greater than 550 mm. However, flocculation mixing intensity, coagulant dose, and filtration rate relatively affected little on the filtration efficiency.

Abstract: Aims We elucidated the therapeutic potential of human umbilical vein endothelial cells (HUVECs) for ameliorating progressive heart failure in a myocardial infarction (MI) rat model. Main methods MI was induced by ligation of left anterior descending artery, and HUVEC was transplanted 1 week after MI. Cardiac function was evaluated by echocardiography, and histological analyses were performed. Key findings Phosphate-buffered saline (MI-V, n = 5) or HUVEC (MI-HV, n = 5) were injected into the border zone and infarcted area 7 days after ligation of the left coronary artery in rats. The MI-HV group showed attenuation of left ventricular (LV) remodeling compared with the MI-V group. In the infarcted myocardium, a few of injected HUVEC was retained up to 28 days. The ratios of matrix metalloproteinase (MMP)-2 or MMP-9 to tissue inhibitor of metalloproteinase (TIMP)-1 or TIMP-3 were decreased in the MI-HV group compared with the MI-V group. In vivo zymography analysis showed that HUVEC transplantation decreased the activities of MMP-2 and MMP-9. In immunohistochemistry, decreased MMP-2 and increased TIMP-1 and TIMP-3 expression were observed at 48 h after HUVEC transplantation. These effects on MMP/TIMP balance were inhibited by L-NAME administration (an eNOS inhibitor, 10 mg/kg). NOS inhibition decreased the protein expressions of TIMP-1 and TIMP-3 but did not change the protein expressions of MMP-2 and MMP-9. Significance Our data suggest that altered balance between MMP and TIMP by HUVEC transplantation contributed to attenuation of ventricular remodeling after MI via eNOS.

Abstract: This study examined the contents of 'Adolescent’s Consumption Life' unit in 12 Technology·Home Economics textbooks used in the 2009 revised curriculum to analyze the contents of school consumer education. The main summary of this study is as follows: First, the contents of consumer education in the unit of 'Adolescent’s Consumption Life' consisted of five criteria including the understanding of adolescent consumer, consumer's right and responsibility, reasonable purchase, resolution to consumer problems, and the formation of new consumption culture. Second, significant differences were found in the contents of consumer education among textbooks because basic concepts have been not been clearly defined yet. Third, essential content was either omitted or hard to understand. Thus, some contents elaborations were inappropriate for students' level. Fourth, there were some limitations in drawing interest and attention from learners and developing dispute resolution competence, since the textbooks mainly comprised the theory-oriented contents of consumer education. Fifth, cautious consideration needs to be taken with regards to clear understanding of basic concepts and systematic and consistent contents composition for coherent and organized school consumer education. These research findings are anticipated to provide basic information of Technology·Home Economics textbooks to teachers, and will be utilized as basic data in organizing textbook contents or developing efficient teaching-learning programs for consumer education. Abstract: To analyze the hazards associated with cucumber and hot pepper cultivation areas, a total of 72 samples were obtained and tested to detect the presence of biological (sanitary indicative, pathogenic bacteria and fungi) and chemical hazards (heavy metals and pesticide residues). The levels of sanitary indicative bacteria (aerobic plate counts and coliforms) and fungi were ND-7.2 and ND-4.8 log CFU/(g, mL, hand, or 100 cm 2 ) in cucumber cultivation areas, and ND- 6.8 and 0.4-5.3 log CFU/(g, mL, hand, or 100 cm 2 ) in hot pepper cultivation areas. More specifically, the soil of hot pepper cultivation areas was contaminated with coliforms at a maximum level of 5.6 log CFU/g. Staphylococcus aureus was detected only in glove samples at a level of 1.4 log CFU/100 cm 2 and Bacillus cereus was detected in the majority of samples at a level of ND-4.8 log CFU/(g, mL, hand, or 100 cm 2 ). Listeria monocytogenes, Escherichia coli O157:H7, and Salmonella spp. were not detected. Heavy metal (Zn, Cu, Ni, Pb, and Hg) chemical hazards were detected at levels lower than the regulation limit. Residual insecticides were not detected in cucumbers; however, hexaconazole was detected at a level of 0.016 mg/kg (maximum residue limit: 0.3 mg/kg) in hot peppers.

Abstract: Thyroid hormone (TH) signaling plays an important role in the regulation of long-wavelength vision in vertebrates. In the retina, thyroid hormone receptor β (thrb) is required for expression of long-wavelength-sensitive opsin (lws) in red cone photoreceptors, while in retinal pigment epithelium (RPE), TH regulates expression of a cytochrome P450 enzyme, cyp27c1, that converts vitamin A1 into vitamin A2 to produce a red-shifted chromophore. To better understand how TH controls these processes, we analyzed the phenotype of zebrafish with mutations in the three known TH nuclear receptor transcription factors (thraa, thrab, and thrb). We found that no single TH nuclear receptor is required for THmediated induction of cyp27c1 but that deletion of all three (thraa-/-;thrab-/-;thrb-/-) completely abrogates its induction and the resulting conversion of A1- to A2-based retinoids. In the retina, loss of thrb resulted in an absence of red cones at both larval and adult stages without disruption of the underlying cone mosaic. RNA-sequencing analysis revealed significant downregulation of only five genes in adult thrb-/- retina, of which three (lws1, lws2, and miR-726) occur in a single syntenic cluster. In the thrb-/- retina, retinal progenitors destined to become red cones were transfated into ultraviolet (UV) cones and horizontal cells. Taken together, our findings demonstrate cooperative regulation of cyp27c1 by TH receptors and a requirement for thrb in red cone fate determination. 87. Microbial Contamination Levels of Ginseng and Ginseng Products Distributed in Korean Markets

Abstract: The objective of this study was to investigate the microbial contamination levels of ginseng and ginseng products distributed in markets. A total of 81 ginseng and ginseng products samples (fresh ginseng 37, white ginseng 15, red ginseng 15, red ginseng beverage 14) were tested to analyze sanitary indicator bacteria (total aerobic bacteria, coliforms and Escherichia coli), major foodborne pathogens, and fungi. The contamination levels of total aerobic bacteria and coliforms were in the range of 3.19 to 7.02 log CFU/g for fresh ginseng, 0.25 to 7.31 log CFU/g for white ginseng, 0 to 2.89 log CFU/g for red ginseng and 0 to 1.70 log CFU/mL for red ginseng beverage. In case of major foodborne pathogens, B. cereus was detected at levels of 0.50 to 2.41 log CFU/g in samples except red ginseng beverage. Fungi was detected at levels of 2.41 log CFU/g in fresh ginseng, the contamination levels of the other gin- seng products samples were lower than 1 log CFU/g or mL. These results indicate that the ginseng and ginseng prod- ucts were comparatively safe with respect to microbiological hazard.

A total of 32 samples were collected from onion farms and tested for biological (sanitary indicators, and pathogenic bacteria and fungi) and chemical (heavy metals and pesticide residues) hazards. Aerobic bacteria and coliforms were detected at a level of 0.27.1 log CFU/g (or mL) in the soil and agricultural water, 1.6-3.6 log CFU/g on surface of the onion, 0.0-6.0 log CFU/ hand (or cm) on the workers’ hands, clothes, and gloves, and 4.7 log CFU/cm on the onion bags. Fungi were detected at a level of 0.0-5.0 log CFU/g (or mL, hand, or 100 cm) in all the samples. Staphylococcus aureus was detected at a level of 1.2 log CFU/hand on the workers’ hands, the detection level of Bacillus cereus was up to 4.8 log CFU/g in the soil. However, Escherichia coli (and in particular strain O157:H7), Listeria monocytogenes, and Salmonella spp. were not detected. Although heavy metals were detected in the environment (in soil and agricultural water) and pesticide residues were detected in onion, the levels were lower than the regulation limits.

Abstract: This study validated microbiological hazards of ginseng farms at the cultivation stage and sug- gested recommendations to develop a good agricultural practices (GAP) model. A total of 96 samples were collected from cultivation environments (soil, irrigation water, and atmosphere), plants (ginseng and its leaf), personnel hygiene (glove, cloth, and hand) of 3 ginseng farms (A, B, and C) and were tested to analyze sanitary indicator bacteria (aero- bic plate count, coliforms and Escherichia coli), major foodborne pathogens (E. coli O157:H7, Listeria monocytoge- nes, Salmonella spp., Staphylococcus aureus, and Bacillus cereus), and fungi. Total bacteria, coliform, and fungi in the 3 ginseng farms were detected at the level of 1.3~6.0, 0.1~5.0, and 0.4~4.9 v/g (or mL, hand, and 100 cm 2 ), respec- tively. Only irrigation water collected from one ginseng farm was confirmed to be E. coli positive. In case of patho- genic bacteria, B. cereus was detected at levels of 0.1~5.0 log CFU/g (or mL, hand, and 100 cm 2 ) in all samples, but other pathogen bacterias were not detected in any samples from all farms. Although E. coli were detected in irrigation water, the level of microbial for the three farms was lower than the regulation limit. According to the results, the gin- sengs produced from the 3 farms were comparatively safe with respect to microbiological hazard. However, cross-con- tamination of bacteria from environments and workers to ginseng has been considered as potential risks. Therefore, to minimize microbial contamination in ginseng, GAP model should be applied for ensuring the safety of ginsengs.

Abstract: The application of ultrafiltration (UF) and microfiltration (MF) membranes has been increased for drinking water purification. The advantages of UF/MF membrane process compared to conventional treatment processes are stable operation under varying feed water quality, smaller construction area, and automatic operation. Most membrane treatment plants are designed with polymeric membranes. Recently, some studies suggested that the process of treating surface water with ceramic membranes is competitive to the application of polymeric membranes. Higher water flux, less frequent cleaning, and much longer lifetime are the advantages of ceramic membrane comparing to polymeric membrane. Therefore, this research focused on the application of ceramic MF membrane pilot plant at the slow sand filtration plant. The ceramic membrane pilot plant has three trains that used raw water and sand filtered water as a feed water, respectively. For optimizing the pilot plant process, the coagulation with PACl coagulant was used as a pretreatment of ceramic membrane process. In addition, CEB (Chemical Enhanced Backwash) process using H2SO4 and NaOCl was used for 1.5 days, respectively. The experimental results showed that applying the optimum coagulant dose before membrane filtration showed enhancing membrane fluxes for both raw water and sand filtered water. Also, when using raw water as a feed of membrane, minimum fouling rate was 2.173 kPa/cycle with 25 mg/L of PACl and when using sand filtered water, the minimum fouling rate was 0.301 kPa/cycle with 5 mg/L of PACl.

91. Research on Bacterial Contamination Levels in Apartment Tower Elevators

Abstract: Objectives: The aim of this study was to investigate the microbial contamination levels in elevators in apartment buildings and to provide information on such microbial contamination. Methods: A total of 144 samples, including from the exterior buttons, interior buttons, elevator handrails, walls, ventilators and airborne bacteria were collected in the morning and afternoon from July to August 2013 for six different elevators. The samples were used to detect sanitary indicator bacteria (total bacteria, coliform, and Escherichia coli), pathogenic bacteria (E. coli O157, L. monocytogenes, Salmonella spp., B. cereus, S. aureus) and fungi. Results: Contamination levels of total bacteria were 0.3-3.8 and 0.0-2.4 log CFU/100 in the morning and afternoon, respectively. In the case of coliform bacteria, the levels were 0.0-3.7 log CFU/100 in the morning and 0.0-0.3 log CFU/100 in the afternoon. However, E. coli was not detected among all samples. Bacillus cereus, pathogenic bacteria, was only detected in 13 (11%) among 144 samples. E. coli O157, L. monocytogenes, Salmonella spp. and S. aureus were not detected among all samples. Comparing the samples collected in the morning and afternoon, we could confirm that the samples in the afternoon were cleaner. Conclusions: This study indicates that the samples in the afternoon were cleaner because these samples were collected following routine cleaning. Also, the levels of contamination in the elevators were low and the sanitary conditions were comparatively well-managed. Therefore it is deemed necessary for elevators be cleaned regularly to provide good conditions for people using elevators.

Abstract: Objectives: This study was undertaken to analyze Staphylococcus aureus from cultivation environments for agricultural products and to confirm antibiotic resistance and enterotoxin genes for the isolated S. aureus. Methods: A total of 648 samples were collected from apple, peach, ginseng and balloon flower farms. S. aureus was isolated from soil, agricultural water, personal hygiene elements (hands, gloves and clothes) and work utensils (boxes). Results: S. aureus was detected in a total of 25 samples and 72 strains were isolated. The resistance rate of the isolated S. aureus strains was confirmed at 33.3%, with 24 resistant strains among the total of 72. Fourteen different patterns types were found, and three pattern types (NV, OX, VA) were confirmed most frequently. As result of the detection of enterotoxin gene type, four gene types (sea: 1, sed: 4, seg: all isolated S. aureus, sei: all isolated S. aureus) were analyzed among a total of nine types. Conclusions: This study demonstrates that personal hygiene techniques should be properly managed, such as washing and sterilization before or after work, because agricultural contamination by S. aureus frequently developed through improper management.

93. Rapid and Sensitive Immunochromatographic Strip for On-site Detection of Sulfamethazine in Meats and Eggs

Abstract: A rapid immunochromatographic (ICG) strip based on a conjugate of colloidal gold and monoclonal antibody (mAb) was developed for the rapid, sensitive, and on-site detection of sulfamethazine in meat and egg samples. The detection limit of the ICG strip is 2 ng/mL, and the assay can be completed in 10 min. A cross-reactivity test indicated that the ICG strip was highly specific to sulfamethazine with no cross-reaction with sulfonamide compounds and other antibiotics. The results of the recovery test from meat and egg samples spiked with sulfamethazine were in good agreement with those obtained by the indirect competitive enzyme-linked immunosorbent assay. These results demonstrated that the ICG strip can be used as a rapid and qualitative tool for on-site screening of sulfamethazine in meat and egg samples. Practical Application The present immunochromatographic strip could be used as a rapid and on-site screening tool for sulfamethazine in livestock products.

Abstract: This study was carried out to find the optimal conditions for red cabbage seed sprouts in terms of their physicochemical and sensory qualities by electron-beam irradiation, cultivation and storage using the response surface methodology (RSM). Moisture content (R 2 =0.9638) was affected by irradiation dose and cultivation time. Total phenolics content (R 2 =0.9117) was mainly affected by irradiation dose, but carotenoid content (R 2 =0.8338) was affected in the order of irradiation dose, cultivation time and storage time. Sensory properties were also affected by irradiation dose, and thus scores decreased as irradiation dose increased. The optimum conditions estimated by superimposing total phenolics content and overall acceptance were 2.2-3.8 kGy of the irradiation dose, 3.0-4.0 days of cultivation and 2.0-3.0 days of storage.

Abstract: Reference materials for quantitative determination of Cd, Cr, Hg and Pb in polycarbonate were developed. Reference materials with two concentration level of elements were prepared by adding appropriate amounts of chemicals to a blank polycarbonate base material. It was shown that ten bottles with triplicate analysis are enough to demonstrate the homogeneity of these candidate reference materials. The statistical results also showed no significant trends in both short-term stability test for four weeks and long-term stability test for twelve months. The certification of the four elements was carried out by isotope-dilution-inductively coupled plasma mass spectrometry (ID-ICP-MS) with microwave-assisted digestion. Certification of candidate reference materials in a single laboratory was confirmed with interlaboratory comparison participated by a certain number of well-recognized testing laboratories in Korea. The certified values and expanded uncertainties (k = 2) for the candidate reference material with low level and the one with high level were (51.7 ± 2.1) mg kg−1 Cd, (103.8 ± 2.9) mg kg−1 Cd, (98.8 ± 4.5) mg kg−1 Cr, (1004 ± 49.8) mg kg−1 Cr, (107.4 ± 4.6) mg kg−1 Hg, (1133 ± 50.7) mg kg−1 Hg, (94.8 ± 3.7) mg kg−1 Pb and (988.4 ± 53.6) mg kg−1 Pb, respectively. The reference materials developed in this study demonstrated their suitability for the quality assurance in Cd, Cr, Hg and Pb analysis for the implementation of RoHS Directive.

Abstract: We present results of single-epoch very long baseline interferometry (VLBI) observations of gamma-ray bright active galactic nuclei (AGNs) using the Korean VLBI Network (KVN) at 22, 43, 86, and 129~GHz bands, which are part of a KVN key science program, Interferometric Monitoring of Gamma-ray Bright AGNs (iMOGABA). We selected a total of 34 radio-loud AGNs of which 30 sources are gamma-ray bright AGNs with flux densities of $>6\times10^{-10}$~ph~cm$^{-2}$~s$^{-1}$. Single-epoch multi-frequency VLBI observations of the target sources were conducted during a 24-hr session on 2013 November 19 and 20. All observed sources were detected and imaged at all frequency bands with or without a frequency phase transfer technique which enabled the imaging of 12 faint sources at 129~GHz, except for one source. Many of the target sources are resolved on milliarcsecond scales, yielding a core-jet structure with the VLBI core dominating the synchrotron emission on the milliarcsecond scale. CLEAN flux densities of the target sources are 0.43-28~Jy, 0.32-21~Jy, 0.18-11~Jy, and 0.35-8.0~Jy in the 22, 43, 86, and 129~GHz bands, respectively., 42 pages, 8 figures, 3 tables, to appear in ApJS

Abstract: This study was carried out to develop pilot plant Net3FM(Net Fit Fiber Filter Module) system and to suggest optimum operating condition for municipal wastewater reuse. SS concentration of biologically treated sewage effluent was reduced from 1.5~5.4mg/L to 0.4~1.0mg/L without coagulant injection in Net3FM system, and the SS removal efficiency was average 84.7%. And also, the removal efficiencies of COD and T-P were decreased slightly due to the SS removal by filtration. Coagulation-Filtration test was conducted to enhance the removal efficiencies of SS and T-P. The optimum dosage of coagulant was injected automatically by auto-controlling system, which is controlled by detecting value of turbidity of secondary sewage effluent. SS, COD and T-P concentrations in filtrated effluent were 0.21~0.57, 1.6~6.2 and 0.137~0.392mg/L with coagulant injection by in-line mixer in Net3FM system, respectively. The removal efficiencies of SS and T-P were highly increased to 92.8% and 89.8%, respectively. It was due to the combined the processes of coagulation and filtration. Net3FM system was evaluated that the removal efficiency of pollutants in secondary sewage effluent and the utilization potential as reclaimed water technology were very high.

Data were collected using structured questionnaires which included Kolb`s Learning Style Inventory, Academic Achievement in Fundamental Nursing and Health Assessment, Self Directed Learning Readiness Scale, and California Critical Thinking Disposition Inventory. Data were analyzed using test, ANOVA, Pearson` correlation coefficients, and Spearman rank correlation coefficient. Methods: The subjects of this study were dental technicians, dental technology students, a professor of dental technology in Daegu and Gyeongbuk area and a self-administered structured questionnaire survey was conducted for 350 randomly selected people. Collected data were analyzed using SPSS Win 17.0 statistics program and analysis techniques include the frequency, percentage and cross-analysis. Results: The results of the survey are as follows. The usefulness of Major subjects was Full Denture laboratory (4.72 points), Dental Morphology (4.71 points), Crown and Bridge laboratory (4.69 points) in the instructor`s group. The usefulness of Major subjects was Dental Morphology (4.58 points), Dental Morphology laboratory (4.34 points), Partial Denture laboratory (4.30 points) in the dental technician`s group.
