Abstract Background Emerging evidence suggests the critical roles of N6-methyladenosine (m6A) RNA modification in tumorigenesis and tumor progression. However, the role of m6A in non-small cell lung cancer (NSCLC) is still unclear. This study aimed to explore the role of the m6A demethylase fat mass and obesity-associated protein (FTO) in the tumor metastasis of NSCLC. Methods A human m6A epitranscriptomic microarray analysis was used to identify downstream targets of FTO. Quantitative real-time PCR (qRT‒PCR) and western blotting were employed to evaluate the expression levels of FTO and FAP in NSCLC cell lines and tissues. Gain-of-function and loss-of-function assays were conducted in vivo and in vitro to assess the effects of FTO and FAP on NSCLC metastasis. M6A-RNA immunoprecipitation (MeRIP), RNA immunoprecipitation (RIP), luciferase reporter assays, and RNA stability assays were used to explore the mechanism of FTO action. Co-immunoprecipitation (co-IP) assays were used to determine the mechanism of FAP in NSCLC metastasis. Results FTO was upregulated and predicted poor prognosis in patients with NSCLC. FTO promoted cell migration and invasion in NSCLC, and the FAK inhibitor defactinib (VS6063) suppressed NSCLC metastasis induced by overexpression of FTO. Mechanistically, FTO facilitated NSCLC metastasis by modifying the m6A level of FAP in a YTHDF2-dependent manner. Moreover, FTO-mediated metastasis formation depended on the interactions between FAP and integrin family members, which further activated the FAK signaling. Conclusion Our current findings provided valuable insights into the role of FTO-mediated m6A demethylation modification in NSCLC metastasis. FTO was identified as a contributor to NSCLC metastasis through the activation of the FAP/integrin/FAK signaling, which may be a potential therapeutic target for NSCLC. Video Abstract

Abstract Background Emerging metabolomics-based studies suggested links between amino acid metabolism and metabolic dysfunction-associated fatty liver disease (MAFLD) risk; however, whether there exists an aetiological role of amino acid metabolism in MAFLD development remains unknown. The aim of the present study was to assess the causal relationship between circulating levels of amino acids and MAFLD risk. Methods We conducted a two-sample Mendelian randomization (MR) analysis using summary-level data from genome-wide association studies (GWAS) to evaluate the causal relationship between genetically predicted circulating levels of amino acids and the risk of MAFLD. In the discovery MR analysis, we used data from the largest MAFLD GWAS (8434 cases and 770,180 controls), while in the replication MR analysis, we used data from a GWAS on MAFLD (1483 cases and 17,781 controls) where MAFLD cases were diagnosed using liver biopsy. We used Wald ratios or inverse variance-weighted (IVW) methods in the MR main analysis and weighted median and MR-Egger regression analyses in sensitivity analyses. Furthermore, we performed a conservative MR analysis by restricting genetic instruments to those directly involved in amino acid metabolism pathways. Results We found that genetically predicted higher alanine (OR = 1.43, 95% CI 1.13–1.81) and lower glutamine (OR = 0.83, 95% CI 0.73–0.96) levels were associated with a higher risk of developing MAFLD based on the results from the MR main and conservative analysis. The results from MR sensitivity analyses and complementary analysis using liver proton density fat fraction as a continuous outcome proxying for MAFLD supported the main findings. Conclusions Novel causal metabolites related to MAFLD development were uncovered through MR analysis, suggesting future potential for evaluating these metabolites as targets for MAFLD prevention or treatment.

Abstract Angelica sinensis roots (Angelica roots) are rich in many bioactive compounds, including phthalides, coumarins, lignans, and terpenoids. However, the molecular bases for their biosynthesis are still poorly understood. Here, an improved chromosome-scale genome for A. sinensis var. Qinggui1 is reported, with a size of 2.16 Gb, contig N50 of 4.96 Mb and scaffold N50 of 198.27 Mb, covering 99.8% of the estimated genome. Additionally, by integrating genome sequencing, metabolomic profiling, and transcriptome analysis of normally growing and early-flowering Angelica roots that exhibit dramatically different metabolite profiles, the pathways and critical metabolic genes for the biosynthesis of these major bioactive components in Angelica roots have been deciphered. Multiomic analyses have also revealed the evolution and regulation of key metabolic genes for the biosynthesis of pharmaceutically bioactive components; in particular, TPSs for terpenoid volatiles, ACCs for malonyl CoA, PKSs for phthalide, and PTs for coumarin biosynthesis were expanded in the A. sinensis genome. These findings provide new insights into the biosynthesis of pharmaceutically important compounds in Angelica roots for exploration of synthetic biology and genetic improvement of herbal quality.

The unconfined compressive strength (UCS) of alkali-activated slag (AAS)-based cemented paste backfill (CPB) is influenced by multiple design parameters. However, the experimental methods are limited to understanding the relationships between a single design parameter and the UCS, independently of each other. Although machine learning (ML) methods have proven efficient in understanding relationships between multiple parameters and the UCS of ordinary Portland cement (OPC)-based CPB, there is a lack of ML research on AAS-based CPB. In this study, two ensemble ML methods, comprising gradient boosting regression (GBR) and random forest (RF), were built on a dataset collected from literature alongside two other single ML methods, support vector regression (SVR) and artificial neural network (ANN). The results revealed that the ensemble learning methods outperformed the single learning methods in predicting the UCS of AAS-based CPB. Relative importance analysis based on the best-performing model (GBR) indicated that curing time and water-to-binder ratio were the most critical input parameters in the model. Finally, the GBR model with the highest accuracy was proposed for the UCS predictions of AAS-based CPB.

Abstract Esophageal carcinoma (EC), one of the most lethal human malignancies, lacks effective targeted therapies. Indoleamine 2,3-dioxygenase 1 (IDO1) plays a key role in a variety of cancers, but its role and mechanism in EC are still unclear. Immunohistochemistry and qRT-PCR were used to analyze the expression of IDO1 in EC, and the prognostic value of IDO1 in EC was evaluated by Kaplan-Meier test. The in vitro and in vivo function loss/acquisition tests were performed to evaluate the biological effects of IDO1 in EC. The mechanism of action of IDO1-regulation EC was explored through Firefly luciferase & Renilla luciferase activity reporter, chromatin immunoprecipitation (ChIP) and immunofluorescence (IF) assays. Clinically, IDO1 expression was abnormally elevated in EC and positively correlated with overall survival. Functionally, IDO1 was contributed to the proliferation and migration of EC cells. Mechanically, IDO1 regulated the expression of chemokine C-X-C ligand 10 (CXCL10) by promoting the entry of NF-κB into the nucleus to combine with the promoter of CXCL10. Consistently, IDO1 facilitated EC progression may dependent on the presence of CXCL10. Moreover, NF-κB alleviated the inhibitory effect of IDO1 knockdown on EC. IDO1 drove the progression of EC by directly binding NF-κB and CXCL10, the finding that may provide an effective theoretical basis for precise therapies for EC.

The purpose of this research was to compare the therapeutic efficacy of multiple-rod constructs vis-a-vis 2-rod constructs in the treatment of adult spinal deformity. A systematic review and meta-analysis were performed to determine whether the multiple-rod construct outperformed the 2-rod construct. Abstract Cytokine therapy, involving interleukin-15 (IL-15), is a promising strategy for cancer immunotherapy. However, clinical application has been limited due to severe toxicity and the relatively low immune response rate, caused by wide distribution of cytokine receptors, systemic immune activation and short half-life of IL-15. Here we show that a biomimetic nanovaccine, developed to co-deliver IL-15 and an antigen/major histocompatibility complex (MHC) selectively targets IL-15 to antigen-specific cytotoxic T lymphocytes (CTL), thereby reducing off-target toxicity. The biomimetic nanovaccine is composed of cytomembrane vesicles, derived from genetically engineered dendritic cells (DC), onto which IL-15/IL-15 receptor α (IL-15Rα), tumor-associated antigenic (TAA) peptide/MHC-I, and relevant costimulatory molecules are simultaneously anchored. We demonstrate that, in contrast to conventional IL-15 therapy, the biomimetic nanovaccine with multivalent IL-15 self-transpresentation (biNV-IL-15) prolonged blood circulation of the cytokine with an 8.2-fold longer half-life than free IL-15 and improved the therapeutic window. This dual targeting strategy allows for spatiotemporal manipulation of therapeutic T cells, elicits broad spectrum antigen-specific T cell responses, and promotes cures in multiple syngeneic tumor models with minimal systemic side effects.

Abstract The “Integrated Wastewater Discharge Standard” was implemented for water pollutant discharge in China’s pesticide industry, which has no control requirements for particular wastewater pollutants in the industry. In the standard, certain pollutants discharge limits are limited strictly or loosely, resulting in practical management implementation difficulties. In view of the highly selective targeting characteristics of organic pesticide active ingredients in fungicides, insecticides, and herbicides, a method for deriving discharge limits based on the water quality criteria for pesticides for the protection of nonsensitive species is established based on the idea of fully protecting aquatic organisms beyond sensitive objects. Through the use of malathion as an example, by screening its toxicity data in different species of aquatic organisms, the sequence point with the most significant change in the acute toxicity sensitivity of the species is taken as the variation point in the cumulative frequency of the sensitive and nonsensitive species to derive the water quality criteria, using the species sensitivity distribution method as the scientific basis for determining the discharge limits. After a comparative analysis of different simulation models, the sigmoid model, with the best fit, is selected to determine that the sensitive species hazard concentration (HCs) of malathion to aquatic organisms in China is 46.4 µg/L, and the discharge limit derived from the HCs based on the relationship between the environmental capacity and emissions is rounded to 250 µg/L. Studies showed that the relationship between the emissions limit derived from the water quality criteria for protecting nonsensitive species and malathion limit stipulated in the “Environmental Quality Standards for Surface Water” conforms to the corresponding relationship of the quality standard and discharge standard, which can be achieved by current pollution control technology, combined with water quality improvement. The discharge limit offers the advantages of technical accessibility and economic rationality.

Objective: To study the effect of β-nicotinamide mononucleotide (NMN) on the growth of Caenorhabditis elegans (C. elegans) and its mechanism. Methods: Nematodes were randomly divided into blank control group and experimental groups with different concentrations of NMN (0.1, 0.5, 1, 5, 10 mg/mL). The impact of NMN on the growth of C. elegans was evaluated by measuring the body length changes of C. elegans in 13 days after adulthood. To explore the inhibitory effect of total glycosides of Cistanche deserticola (TG) on HepG2 cells and its mechanism. In this paper, different concentrations (0, 3.5, 10.5, 21, 31.5, 42 μg/mL) TG were treated 24 h on HepG2 liver cancer cells, and the viability of HepG2 cells was detected using CCK8 assay. Hoechst 33342/PI double staining method and Annexin V-FITC/PI were used to detect HepG2 cells apoptosis. The phenomenon of cell migration was detected by cell migration assay. Meanwhile, cell cycle progression changes were detected by flow cytometry. And the expression of α-fetoprotein (AFP), β-catenin, Dishevelled (Dsh), GSK-3β were detected by Western blot. The results showed that TG could reduce the proliferation of HepG2 cells in a concentration dependent manner, with only 31.04% cell viability when TG was 42 μg/mL. In addition, TG could damage the cell structure and induce cell apoptosis, and the apoptosis rate could be as high as 32.44% by AV/PI detection. In this study, a brief measure of four internal attributes found to protect against the impact of adverse life events on mental health was developed and validated. In addition, profiles of internal strengths that significantly predict mental health outcomes in young Chinese adults were identified. The results of exploratory and confirmatory factor analyses on data from 831 Chinese university students supported the proposed four-factor model of the ISALES. Participants in the current study fell into two clusters, with one cluster being higher than the other in all four internal strengths, with the former cluster demonstrating better overall mental health than those in the latter cluster. The ISALES is a promising instrument for use in clinical settings and may be used to identify individuals who are more ‘at risk’ of developing poor mental health in the aftermath of a negative life event. The use of the ISALES may permit tailored interventions and timely support to be provided to individuals within clinical settings.

Excessive UV exposure can lead to skin roughness, wrinkles, pigmentation, and reduced elasticity, with severe cases potentially causing skin cancer. Nowadays, various anti-photoaging strategies have been developed to maintain skin health. Among them, dietary supplements with anti-photoaging properties are gaining increasing attention. Yak whey protein (YWP) possesses multiple benefits, including anti-inflammatory, antioxidant, and immune-boosting properties, effectively protecting the skin. This study used a mixed UVA and UVB light source to irradiate a nude mouse model, exploring the advantages of YWP in anti-photoaging and regulating gut microbiota. The results indicated that YWP alleviated UV-induced skin damage, wrinkles, dryness, and reduced elasticity by inhibiting oxidative stress, inflammatory factors (IL-1α, IL-6, and TNF-α), and matrix metalloproteinases (MMP-1, MMP-3, and MMP-12), thereby increasing the levels of elastin, type I collagen, and type III collagen in the extracellular matrix (ECM). Additionally, YWP significantly improved the abundance of Firmicutes and Bacteroidota in the gut microbiota of mice, promoting the growth of beneficial bacteria such as Lachnospiraceae_NK4A136_group, Ruminococcus_torques_group, and Clostridia_UCG_014, mitigating the dysbiosis caused by photoaging. These findings underscore the potential of YWP in anti-photoaging and gut microbiota improvement, highlighting it as a promising functional food for enhancing skin and gut health.

As a member of the pattern recognition receptor (PRR) class, scavenger receptor class B type 1 (SRB1) plays a key role in innate immunity. Grass carp (Ctenopharyngodon idellus) ranks among the most extensively cultivated freshwater aquaculture species in China. However, little is known about the function of SRB1 in C. idellus. In this research study, a SRB1 gene was identified in C. idellus, named CiSRB1. The full-length cDNA of CiSRB1 is 2486 bp long, with an open reading frame (ORF) of 2486 bp encoding a 497 amino acid (aa) protein containing a conserved CD36 domain. The identified genomic DNA length of CiSRB1 is 20,042 bp, including 12 exons and 11 introns. The predictive analysis of protein interactions revealed that CiSRB1 could interact with the outer capsid proteins of typical GCRV strains. The tissue distribution of CiSRB1 exhibited age-dependent characteristics. CiSRB1 displayed the highest expression in the intestines and moderate levels in muscle, spleen, liver, and brain of one-year-old grass carp while maintaining relatively low levels in three-year-old grass carp. Following grass carp reovirus (GCRV) infection, notable upregulation of CiSRB1 transcripts was observed in major immune tissues (gills, intestines, spleen, and liver). Furthermore, significant differences were found between one-year-old and three-year-old grass carp, with lower CiSRB1 expression levels being detected in the older group. Additionally, a distinct response to GCRV infection was observed in one-year-old and three-year-old grass carp. It was found that one-year-old individuals had a mortality rate of up to 84% 6 days post-infection (dpi), whereas all three-year-old counterparts survived after GCRV infection. The analysis of GCRV copy numbers across tissues revealed substantially higher levels in one-year-old grass carp compared with their older counterparts, confirming the existence of age-dependent susceptibility to GCRV infection in grass carp. Combined with these results, it was speculated that the decline in cell-surface CiSRB1 expression with age may impede reovirus binding to host cells, potentially explaining why older grass carp demonstrated enhanced resistance to GCRV infection. This observation accentuates the importance of CiSRB1 in the context of GCRV infection and provides insights into age-dependent susceptibility to reovirus.

Yellow wine fermented from highland barley is an alcoholic beverage with high nutritional value. However, the industrialization of barley yellow wine has been constrained to a certain extent due to the lack of a systematic starter culture. Therefore, the present study aims to simulate barley yellow wine fermentation using a starter culture consisting of Rhizopus arrhizus, Saccharomyces cerevisiae, Pichia kudriavzevii, and Lacticaseibacillus rhamnosus. In this study, changes in enzyme activity, fermentation characteristics, volatile substance production, and amino acid content during the fermentation of highland barley yellow wine brewed with different starter cultures were evaluated. The results of this study show that regulating the proportion of mixed starter bacteria can effectively control the various stages of the fermentation process and improve the organoleptic characteristics and quality of yellow wine to varying degrees. Additionally, we found that the addition of probiotics could effectively improve the palatability of yellow wine. To the best of our knowledge, we have validated for the first time the use of the above multispecies starter culture, consisting of R. arrhizus, S. cerevisiae, P. kudriavzevii, and L. rhamnosus, in the production of highland barley yellow wine. The obtained findings provided reference data for optimizing highland barley yellow wine fermentation.

Male reproductive dysfunction is a clinical disease, with a large number of cases being idiopathic. Reproductive disorders have been found in obese (diet-induced obesity and diet-induced obesity-resistant) mice, but the mechanism behind the male reproductive dysfunction between them may be different. The purpose of this study was to explore the possible role and mechanism of miR-34c on sperm production in high-fat-diet-induced obesity-resistant (DIO-R) mice and GC-1 spg cells, which may differ from those in high-fat-diet-induced obesity (DIO) mice. In vivo and in vitro experiments were performed. C57BL/6J mice were fed a high-fat diet for 10 weeks to establish the DIO and DIO-R mouse model. GC-1 spg cells were used to verify the mechanism of miR-34c on sperm production. During in vivo experiments, sperm production damage was found in both DIO and DIO-R male mice. Compared to the control mice, significantly decreased levels of testosterone, LH, activities of acrosome enzyme (ACE), HAse, and activating transcription factor 1 (ATF1) were found in both DIO and DIO-R male mice (p < 0.05). Compared with the control group, the ratio of B-cell lymphoma-2 (Bcl-2)/bcl-2-associated X protein (Bax) in the DIO group was significantly decreased, and the expression level of cleaved caspase-3 was significantly increased (p < 0.05). Compared with the control group, the Bcl-2 protein expression level in the testes of the DIO-R group significantly decreased (p < 0.05). However, the Bax expression level increased. Thus, the Bcl-2/Bax ratio significantly decreased (p < 0.01); however, the factor-related apoptosis (Fas), Fas ligand (FasLG), cleaved caspase-8, caspase-8, cleaved caspase-3, and caspase-3 protein expression levels significantly increased (p < 0.05). Compared with the DIO group, in DIO-R mice, the activities of ACE, ATF1, Bcl-2, and Bcl-2/Bax’s spermatogenesis protein expression decreased, while the apoptosis-promoting protein expression significantly increased (p < 0.05). During the in vitro experiment, the late and early apoptotic ratio in the miR-34c over-expression group increased. MiR-34c over-expression enhanced the expression of apoptosis-related proteins Fas/FasLG and Bax/Bcl-2 while inhibiting the expression of ATF1 and the sperm-associated protein in GC-1 spg cells. DIO and DIO-R could harm sperm production. DIO-R could impair sperm production by inducing the miR-34c-activated apoptosis and spermatogenesis pathway, which may be different from that of DIO.

Codonopsis convolvulacea is a highly valued Chinese medicinal plant containing diverse bioactive compounds. While roots/tubers have been the main medicinal parts used in practice, leaves and stems may also harbor valuable phytochemicals. However, research comparing volatiles across tissues is lacking. This study performed metabolomic profiling of leaves, stems, and tubers of C. convolvulacea to elucidate tissue-specific accumulation patterns of volatile metabolites. Ultra-high performance liquid chromatography–tandem mass spectrometry identified 302 compounds, belonging to 14 classes. Multivariate analysis clearly differentiated the metabolic profiles of the three tissues. Numerous differentially accumulated metabolites (DAMs) were detected, especially terpenoids and esters. The leaves contained more terpenoids, ester, and alcohol. The stems accumulated higher levels of terpenoids, heterocyclics, and alkaloids with pharmaceutical potential. The tubers were enriched with carbohydrates like sugars and starch, befitting their storage role, but still retained reasonable amounts of valuable volatiles. The characterization of tissue-specific metabolic signatures provides a foundation for the selective utilization of C. convolvulacea parts. Key metabolites identified include niacinamide, p-cymene, tridecanal, benzeneacetic acid, benzene, and carveol. Leaves, stems, and tubers could be targeted for antioxidants, drug development, and tonics/nutraceuticals, respectively. The metabolomic insights can also guide breeding strategies to enhance the bioactive compound content in specific tissues. This study demonstrates the value of tissue-specific metabolite profiling for informing the phytochemical exploitation and genetic improvement of medicinal plants.

RNA editing plays an extensive role in the initiation and progression of cancer. However, the overall profile and molecular functions of RNA editing in different ovarian cancer subtypes have not been fully characterized and elucidated. Here, we conducted a study on RNA editing in four cohorts of ovarian cancer subtypes through large-scale parallel reporting and bioinformatics analysis. Our findings revealed that RNA editing patterns exhibit subtype-specific characteristics within cancer subtypes. The expression pattern of ADAR and the number of differential editing sites varied under different conditions. CCOC and EOC exhibited significant editing deficiency, whereas HGSC and MOC displayed significant editing excess. The sites within the turquoise module of the coedited network also revealed their correlation with ovarian cancer. In addition, we identified an average of over 40,000 cis-edQTLs in the four subtypes. Finally, we explored the association between RNA editing and drug response, uncovering several potentially effective editing-drug pairs (EDP) and suggesting the conceivable utility of RNA editing sites as therapeutic targets for cancer treatment. Overall, our comprehensive study has identified and characterized RNA editing events in various subtypes of ovarian cancer, providing a new perspective for ovarian cancer research and facilitating the development of medical interventions and treatments.

The effect of underwater noise environment generated by equipment in industrial recirculating aquaculture systems (RAS) on fish is evident. However, different equipment generate noise in various frequency ranges. Understanding the effects of different frequency ranges noise on cultured species is important for optimizing the underwater acoustic environment in RAS. Given this, the effects of underwater noise across various frequency bands in RAS on the growth, physiology, and collective behavior of juvenile largemouth bass (Micropterus salmoides) were comprehensively evaluated here. In this study, three control groups were established: low-frequency noise group (80–1000 Hz, 117 dB re 1μPa RMS), high-frequency noise group (1–19 kHz, 117 dB re 1μPa RMS), and ambient group. During a 30-day experiment, it was found that: 1) industrial RAS noise with different frequency bands all had a certain inhibitory effect on the growth of fish, which the weight gain rate and product of length and depth of caudal peduncle in the ambient group were significantly higher than those of the two noise groups, with the low-frequency noise group showing significantly lower values than the high-frequency noise group; 2) industrial RAS noise had a certain degree of adverse effect on the digestive ability of fish, with the low-frequency noise group being more affected; 3) industrial RAS noise affected the collective feeding behavior of fish, with the collective feeding signal propagation efficiency and feeding intensity of the noise groups being significantly lower than those of the ambient group, and the high-frequency noise group performing better than the low-frequency noise group as a whole therein. From the above, the underwater noise across different frequency bands generated by equipment operation in industrial RAS both had an impact on juvenile largemouth bass, with the low-frequency noise group being more severely affected.

Background: Influenced by socio-cultural and world events, Chinese society has significant intergenerational differences. With rapid economic and cultural development, the unique characteristics of Generation Z nursing students in China may influence the clinical education environment. However, the research on Generation Z in China is still in its infancy. Objectives: This study aimed to explore the experiences and perceptions of Generation Z nursing students during their practicum in an intensive care unit (ICU) in the context of China's unique cultural and historical background. Methods: A phenomenological approach was used in this qualitative study. Semi-structured, face-to-face interviews were conducted with fifteen Generation Z nursing students doing practicum in the ICU in a third-level hospital in Shanghai, China. The data were analyzed using Colaizzi's seven-step method. Result: Three themes emerged: intelligent medical services empowering critical care, perception of multiple challenges, and affirmation of the teaching work in the ICU. Conclusions and implications: The clinical instructors should use a combination of online and offline pedagogy, give positive guidance through role modeling, and develop the self-learning skills of Generation Z nursing students. This might help Generation Z nursing students relieve the stress of practicum in the ICU, more quickly adapt to the clinical environment, and enter nursing positions. The result of this research provided valuable information to help clinical practicum programs in China effectively educate Generation Z nursing students. Good education ensures that health care is safe and effective, making it easier for patients to get better.

Abstract Background With the increasing incidence of diabetes worldwide, patients diagnosed with diabetes has been getting younger. Previous studies have shown that high remnant cholesterol (RC) level leads to an increased risk of cardiovascular disease events. However, the relationship between RC levels and newly diagnosed early‐onset type 2 diabetes mellitus (T2DM) is unknown. This study aimed to explore the association between RC and newly diagnosed early‐onset T2DM. Methods A total of 606 patients newly diagnosed with early‐onset T2DM and 619 gender‐matched subjects with normal blood glucose levels were retrospectively enrolled in this study. All T2DM patients showed onset age of 18–40 years. Binary logistic regression analysis was performed to analyze independent risk factors and receiver operating characteristic (ROC) analysis was used to explore the predictive value of RC and other unconventional lipids. Moreover, the correlation between RC and insulin resistance in patients with newly diagnosed early‐onset T2DM was also examined with binary logistic regression analysis and Spearman correlation analysis. Results Increased RC level was an independent risk factor for early‐onset T2DM (p .05). RC was still correlated with insulin resistance after adjusting the conventional lipid parameters (TG, TC, HDL‐C, and low‐density lipoprotein cholesterol) using partial correlation analysis. Conclusion RC level was higher in patients with early‐onset T2DM and was correlated to the degree of insulin resistance as well. Patients aged 18–40 years with RC >0.32 mmol/L showed an increased risk of developing T2DM.

This paper proposed a lightweight design method for orthotropic steel bridge deck U-ribs. Firstly, the U-rib is analyzed in static structural properties. An analytic hierarchy model was constructed to maximize the stiffness for multi-objective topology optimization. Secondly, a parametric reconfiguration model of the topologically optimized U-rib was established. Finally, static tests are carried out and the performance of the 3D printing scale model was tested. The test results show that the method is effective. Weight of the original model with U-rib is 99.8 kg, and the weight of its optimized model is 76.8 kg. The method achieved a weight reduction of 23% for the U-ribs, and the stiffness still met the permitted conditions. The orthotropic steel bridge deck composed of the optimized U-ribs effectively reduced the weight by 150 kg on the basis of basically unchanged stiffness. The method provides an important reference for the lightweight transformation of assembled bridge structures.

To evaluate the effects of dietary protein on reproductive performance, egg quality and larval health of the blotched snakehead Channa maculata, three groups were designed: a high protein group (HP) with 48% protein content, a low protein group (LP) with 44% protein content, and a control group (C) fed with commercial feed containing 40% protein. The results demonstrated that the HP diet significantly enhanced maternal absolute fecundity, egg size and offspring health; however, it had no impact on fertilization rate and hatching rate. The maternal plasma levels of 11-ketotestosterone, 17β-estradiol, and vitellogenin in the HP group were found to be significantly higher compared to those in the C group. Plasma SOD activity and T-AOC capacity in the HP group were significantly higher than those in the C group, while MDA content showed an opposite trend. The trend of T-AOC in 7 days post-hatching larvae exhibited a consistent pattern with that observed in the maternal. Metabolomics and transcriptomics results indirectly indicated that the differences in reproductive and egg quality of the broodstock were associated with the regulation of arachidonic acid metabolism, steroid hormone biosynthesis, cholesterol metabolism, glycerophospholipid metabolism, and glycerolipid metabolism. The expression levels of star, zp3, foxo3, tgf-β, ghr, igf, cyp17a1, and cyp19a1 genes associated with reproduction and were significantly upregulated in the HP group. By depleting the abundance of metabolites in key pathway such as the pentose phosphate pathway, glycolysis/gluconeogenesis, nicotinate and nicotinamide metabolism, vitamin B6 metabolism within the ovary, there is an elevation in arachidonic acid metabolite abundance, thereby facilitating ovarian maturation and regulating egg quality. In conclusion, the high protein diet may affect the reproductive process and egg quality by inducing an upregulation of TGF-beta signaling pathway and ovarian steroidogenesis and arachidonic acid metabolism, leading to increased plasma cholesterol levels and enhanced steroid hormone synthesis.

This paper introduced a new nonexplosive roadway excavation method, combining the reserved free space technology and the static expansion mechanical fracturing technology, where the former is implemented by the gasbag, while the piston splitter is for the latter. The numerical model of roadway excavation was set up via PFC3D to investigate the mechanical fracturing performance, including the single-hole fracturing and the hole network fracturing. The results show that the reasonable hole margin is about 1.0–1.5 m, and the optimal column spacing of the hole network pattern is 1.0 m, after comprehensively analyzing the fracturing performance and the splitting force evolution. Moreover, the mechanical fracturing excavation method was applied to construct a parking chamber in the Kalatongke Mine, to preliminarily verify the feasibility of the static expansion mechanical fracturing technology. The in situ investigation results indicate that the excavation footage is about 0.8 m with the piston splitter when adopting a hole margin of 1.0–1.5 m. To sum up, the preliminary field application and the numerical simulation result both support the feasibility of mechanical fracturing, and the reasonable fracturing hole margin is about 1.0 m.

In recent decades, climate change has significantly affected water quality and algal blooms in lakes worldwide. Studying the temporal dynamics patterns of algal production efficiency and its drivers in individual lakes is useful for analyzing the evolutionary trajectory of Chla-TP changes, and identifying the main controlling factors of eutrophication to guide lake management. In this study, we used a dataset for the Lake Gehu (1986–2020) to analyze the drivers of the ETP variability on interannual and seasonal time scales and to quantify the relative impacts and contributions of nutrient and climatic factors on the ETP. The results showed a negative correlation of the Chla-TP relationship appeared in years with very high ETP. The effects of nutritional and climatic factors on the ETP at different time scales were quantified using a generalized additive model. The results showed that the total phosphorus (TP) concentration had a significant effect on the interannual ETP variation, explaining up to 28.9 %. At the intra-annual scale, climatic variables had a much greater effect on the ETP than the nutrient concentration, and the explanatory effect was ranked as temperature > wind speed > rainfall > total nitrogen (TN). Based on the characteristics of seasonal ETP variations, the TPa of Lake Gehu were 50 μg/L, 33 μg/L, 20 μg/L with ETP levels of 0.2, 0.3, 0.5, respectively. This research highlights the significance of nutrient and climatic factors in interannual and seasonal scale, respectively. In addition, ETP influenced by the coupling of nutrient and climate provides a criterion for establishing the seasonal nutrient threshold of lakes.

Abstract Subspace outlier detection has emerged as a practical approach for outlier detection. Classical full space outlier detection methods become ineffective in high dimensional data due to the “curse of dimensionality”. Subspace outlier detection methods have great potential to overcome the problem. However, the challenge becomes how to determine which subspaces to be used for outlier detection among a huge number of all subspaces. In this paper, firstly, we propose an intuitive definition of outliers in subspaces. We study the desirable properties of subspaces for outlier detection and investigate the metrics for those properties. Then, a novel subspace outlier detection algorithm with a statistical foundation is proposed. Our method selectively leverages a limited set of the most interesting subspaces for outlier detection. Through experimental validation, we demonstrate that identifying outliers within this reduced set of highly interesting subspaces yields significantly higher accuracy compared to analyzing the entire feature space. We show by experiments that the proposed method outperforms competing subspace outlier detection approaches on real world data sets.

Abstract Background Human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs) can be used to treat heart diseases; however, the optimal maturity of hiPSC-CMs for effective regenerative medicine remains unclear. We aimed to investigate the benefits of long-term cultured mature hiPSC-CMs in injured rat hearts. Methods Cardiomyocytes were differentiated from hiPSCs via monolayer culturing, and the cells were harvested on day 28 or 56 (D28-CMs or D56-CMs, respectively) after differentiation. We transplanted D28-CMs or D56-CMs into the hearts of rat myocardial infarction models and examined cell retention and engraftment via in vivo bioluminescence imaging and histological analysis. We performed transcriptomic sequencing analysis to elucidate the genetic profiles before and after hiPSC-CM transplantation. Results Upregulated expression of mature sarcomere genes in vitro was observed in D56-CMs compared with D28-CMs. In vivo bioluminescence imaging studies revealed increased bioluminescence intensity of D56-CMs at 8 and 12 weeks post-transplantation. Histological and immunohistochemical analyses showed that D56-CMs promoted engraftment and maturation in the graft area at 12 weeks post-transplantation. Notably, D56-CMs consistently promoted microvessel formation in the graft area from 1 to 12 weeks post-transplantation. Transcriptomic sequencing analysis revealed that compared with the engrafted D28-CMs, the engrafted D56-CMs enriched genes related to blood vessel regulation at 12 weeks post-transplantation. As shown by transcriptomic and western blot analyses, the expression of a small heat shock protein, alpha-B crystallin (CRYAB), was significantly upregulated in D56-CMs compared with D28-CMs. Endothelial cell migration was inhibited by small interfering RNA-mediated knockdown of CRYAB when co-cultured with D56-CMs in vitro. Furthermore, CRYAB overexpression enhanced angiogenesis in the D28-CM grafts at 4 weeks post-transplantation. Conclusions Long-term cultured mature hiPSC-CMs promoted engraftment, maturation and angiogenesis post-transplantation in infarcted rat hearts. CRYAB, which was highly expressed in D56-CMs, was identified as an angiogenic factor from mature hiPSC-CMs. This study revealed the benefits of long-term culture, which may enhance the therapeutic potential of hiPSC-CMs.

Abstract Background To examine the relationships between polyunsaturated fatty acids (PUFAs) dietary intake and asthma in children. Methods In this cross-sectional study, a total of 14,727 participants from the United States National Health and Nutrition Examination Survey (NHANES) database in 1999–2018 were included, and the baseline characteristics of all participants were gathered. The description analysis was used to explore the possible covariates. Weighted multivariate logistic regression models were adopted to assessed the association between PUFAs dietary intake and asthma in children. In addition, we also performed subgroup analysis based on gender, age, and maternal smoking during pregnancy to investigate this relationship. Results The prevalence of asthma approximately was 15.38% in the present study. The result of weighted multivariate logistic regression indicated that, docosahexaenoic [weighted odds ratio (OR) = 0.37, 95% confidence interval (CI) 0.19–0.74], total n − 3 PUFAs (weighted OR = 0.63, 95%CI 0.43–0.91), and eicosapentaenoic (weighted OR = 0.35, 95%CI 0.13–0.95) dietary intake were negatively associated with asthma in children. The subgroup analysis described that when children were male (weighted OR = 0.28, 95%CI 0.10–0.84), or were 5–7 years (weighted OR = 0.04, 95%CI 0.01–0.37), were 7–12 years (weighted OR = 0.46, 95%CI 0.24–0.90), or their maternal smoking during pregnancy (weighted OR = 0.16, 95%CI 0.03–0.90), docosahexaenoic dietary intake was negatively related to childhood asthma. Conclusion Docosahexaenoic dietary intake was negatively associated with the asthma in children, especially if children were male, or were 5–12 years, or their maternal smoking during pregnancy.

Carallia brachiata (Lour.) Merr. (1919) is an important medical resource distributed across subtropical Asia. In this study, the complete chloroplast genome of C. brachiata was sequenced, revealing a total length of 162,460 bp, including four regions – a large single copy (89,814 bp), a small single copy (18,804 bp), and a pair of inverted repeats (26,921 bp each). The overall guanine + cytosine content was 35.76%. In total, 130 genes were annotated within the chloroplast genome, comprising 85 protein-coding, 37 tRNA, and 8 rRNA genes. Subsequent phylogenetic analyses revealed that C. brachiata is closely related to Carallia diplopetala.

Abstract Background Vascular smooth muscle cells (VSMCs) are the predominant cell type of the aortic middle layer, the abnormal number or function of which has been demonstrated to have a role in thoracic aortic aneurysm (TAA). Here, this study aimed to identify the function of circ_0008285 in VSMC apoptosis. Methods Human VSMCs were treated with angiotensin II (Ang II) for functional experiments. Cell counting kit‐8, 5‐ethynyl‐2′‐deoxyuridine (EdU), and flow cytometry were applied for function analysis. The interaction between miR‐150‐5p and circ_0008285 or brain acid‐soluble protein 1 (BASP1) was also evaluated by dual‐luciferase reporter assay and RNA immunoprecipitation assay. Exosomes were isolated by the commercial kit. Results A highly expressed circ_0008285 was observed in the aortic tissues of TAA patients and Ang‐II‐induced VSMCs. Circ_0008285 deficiency dramatically reversed Ang‐II‐induced proliferation arrest and apoptosis promotion in VSMCs. Circ_0008285 functionally targeted miR‐150‐5p. MiR‐150‐5p inhibition attenuated the inhibitory effects of circ_0008285 silencing on Ang‐II‐evoked apoptosis in VSMCs. BASP1 was verified to be a target of miR‐150‐5p, and was proved to attenuate miR‐150‐5p‐triggered apoptosis arrest in Ang‐II‐stimulated VSMCs. Additionally, extracellular circ_0008285 was packaged into exosomes, which could be transferred into the recipient cells. Conclusion Circ_0008285 silencing could suppress Ang‐II‐induced VSMCs apoptosis via miR‐150‐5p/BASP1 axis, adding further understanding of the pathogenesis of TAA.

Abstract There is increasing attention to the production of cellulose nanocrystals (CNCs) from lignocellulosic biomass by enzymatic hydrolysis with cellulase. In this study, the feasibility of the application of a cellulase system from engineered strain Penicillium oxalicum cEES in the production of CNCs was assessed. Using commercial eucalyptus dissolving pulp (EDP) as substrate, the CNCs were successfully obtained by enzymatic hydrolysis with the cellulase cEES, and the total yields of CNCs reached 15.7% through three-step enzymatic hydrolysis of total 72 h (24 h for each step). The prepared CNCs were characterized and found that their crystallinity and thermal stability were higher than that of EDP. In the later stage of enzymatic hydrolysis, the process efficiency of enzymatic preparation of CNCs greatly decreased because of the high crystallinity of cellulosic substrate, and a simple homogenization treatment can promote the enzymatic hydrolysis, as well as produce fusiform CNCs with more uniform size and more fermentable sugar that could be further converted into fuels and bulk chemicals through fermentation. This study provides a feasible enzymatic preparation process for CNCs with engineered cellulase and commercial cellulosic materials. Graphical Abstract

Salvia guaranitica is considered one of the most significant medicinal and aromatic herbs in terms of nutritional and medical benefits due to its wealth of important active components. Among these compounds, terpenoids are the most prominent and abundant, particularly monoterpenes (C10), sesquiterpenes (C15), and diterpenes (C20). They are biologically advantageous to plants and perform a multitude of functions. The current study aimed to clone the S. guaranitica gene that encodes for geranyllinalool synthases (SgGES, EC: 4.2.3.144), with consideration for these features. The open reading frame of the 867-amino-acid protein encoded by SgGES consists of 2.721 base pairs. In addition, the SgGES protein has five domains that belong to the terpene synthase family, which are related to the terpene and terpenoid synthase domains. We manipulated and overexpressed the SgGES gene in Nicotiana tabacum to explore its function. When compared to the GUS control, the transgenic N. tabacum plants displayed an increase in leaf production and diameter when compared with the wild-type plants. Finally, analysis of transgenic plants using gas chromatography/mass spectrometry (GC-MS) showed that SgGES is responsible for producing various terpene species, especially diterpenes.

The purpose of this study was to examine the dietary views and practices and to identify associated barriers to and facilitators of healthy eating of rural residents emerging from poverty in the context of rapid socio-economic development. A qualitative design with semi-structured interviews was utilized to collect detailed insights into the dietary behaviors of 22 rural residents from 10 villages in Fengshan town, Dafang county, Guizhou province, China. Thematic analysis was applied to interpret the interview data, guided by the socio-ecological model. Four themes were identified: traditional eating patterns, factors influencing food choice, limited knowledge of healthy diet, and lack of nutritional guidance. Barriers to healthy eating included traditional but unhealthy foods, limited nutritional knowledge, inadequate understanding of nutritional requirements, overwhelming nutritional information, and limited professional guidance. Facilitators encompassed healthy traditional eating patterns, household composition, home gardening, preference for traditional bean and soy products, improved food supply and availability owing to poverty alleviation efforts, and being open to receiving professional dietary guidance. As a preliminary investigation into the dietary views and practices of this population, the study highlights a significant gap in the nutritional knowledge and guidance available to rural residents in China, emphasizing the need for comprehensive strategies that address the complex socio-ecological factors influencing dietary behaviors.

Skin is the largest organ in the human body and requires proper dressing to facilitate healing after an injury. Wounds on movable parts, such as the elbow, knee, wrist, and neck, usually undergo delayed and inefficient healing due to frequent movements. To better accommodate movable wounds, a variety of functional hydrogels have been successfully developed and used as flexible wound dressings. On the one hand, the mechanical properties, such as adhesion, stretchability, and self-healing, make these hydrogels suitable for mobile wounds and promote the healing process; on the other hand, the bioactivities, such as antibacterial and antioxidant performance, could further accelerate the wound healing process. In this review, we focus on the recent advances in hydrogel-based movable wound dressings and propose the challenges and perspectives of such dressings.

Optimizing soil carbon content is essential for mitigating climate change. Understanding the soil organic carbon (SOC) contents and their relationship with plantation age is important for enhancing SOC in tea plantations. However, there is still a lack of studies in quantifying the SOC–age curve of the whole life cycle in tea plantations. Thus, in this study, we collected 140 soil samples aged 3 to 60 years in the four representative regions (Anxi, Datian, Qingliu, and Fuzhou) in Fujian Province to quantify the SOC contents and their relationship with plantation age. We found that the average SOC was 14.6 ± 6.1 g/kg in the four sampling regions. Nitrogen (N) emerged as having a highly significant positive correlation with SOC (R2 = 0.9). We also found a significant negative correlation between SOC and mean annual temperature (MAT) (R2 = 0.6), and a significant positive correlation with mean annual precipitation (MAP) (R2 = 0.6). The SOC increased with plantation age at 3–20 years old and peaked at 16–20 years old. After 35 years, the SOC decreased gradually with the aging of the plantation. The results indicated that tea plantations could be renewed after the age of 35. These results showed that optimizing age structure is important in enhancing SOC in tea plantations and is meaningful in achieving carbon neutrality.

Vehicle detection is a research direction in the field of target detection and is widely used in intelligent transportation, automatic driving, urban planning, and other fields. To balance the high-speed advantage of lightweight networks and the high-precision advantage of multiscale networks, a vehicle detection algorithm based on a lightweight backbone network and a multiscale neck network is proposed. The mobile NetV3 lightweight network based on deep separable convolution is used as the backbone network to improve the speed of vehicle detection. The icbam attention mechanism module is used to strengthen the processing of the vehicle feature information detected by the backbone network to enrich the input information of the neck network. The bifpn and icbam attention mechanism modules are integrated into the neck network to improve the detection accuracy of vehicles of different sizes and categories. A vehicle detection experiment on the Ua-Detrac dataset verifies that the proposed algorithm can effectively balance vehicle detection accuracy and speed. The detection accuracy is 71.19%, the number of parameters is 3.8 MB, and the detection speed is 120.02 fps, which meets the actual requirements of the parameter quantity, detection speed, and accuracy of the vehicle detection algorithm embedded in the mobile device.

Diarrhea, often caused by viruses like rotavirus (RV) and norovirus (NV), is a global health concern. This study focuses on RV and NV in Jining City from 2021 to 2022. Between 2021 and 2022, a total of 1052 diarrhea samples were collected. Real-Time Quantitative Fluorescent Reverse Transcriptase-PCR was used to detect RV-A, NV GI, and NV GII. For RV-A-positive samples, VP7 and VP4 genes were sequenced for genotype analysis, followed by the construction of evolutionary trees. Likewise, for NV-GII-positive samples, VP1 and RdRp genes were sequenced for genotypic analysis, and evolutionary trees were subsequently constructed. Between 2021 and 2022, Jining City showed varying detection ratios: RV-A alone (excluding co-infection of RV-A and NV GII) at 7.03%, NV GI at 0.10%, NV GII alone (excluding co-infection of RV-A and NV GII) at 5.42%, and co-infection of RV-A and NV GII at 1.14%. The highest RV-A ratios were shown in children ≤1 year and 2–5 years. Jining, Jinxiang County, and Liangshan County had notably high RV-A ratios at 24.37% (excluding co-infection of RV-A and NV GII) and 18.33% (excluding co-infection of RV-A and NV GII), respectively. Jining, Qufu, and Weishan had no RV-A positives. Weishan showed the highest NV GII ratios at 35.48% (excluding co-infection of RV-A and NV GII). Genotype analysis showed that, in 2021, G9P[8] and G2P[4] were dominant at 94.44% and 5.56%, respectively. In 2022, G8P[8], G9P[8], and G1P[8] were prominent at 75.86%, 13.79%, and 10.35%, respectively. In 2021, GII.3[P12], GII.4[P16], and GII.4[P31] constituted 71.42%, 14.29%, and 14.29%, respectively. In 2022, GII.3[P12] and GII.4[P16] accounted for 55.00% and 45.00%, respectively. RV-A and NV showed varying patterns for different time frames, age groups, and regions within Jining. Genotypic shifts were also observed in prevalent RV-A and NV GII strains in Jining City from 2021 to 2022. Ongoing monitoring of RV-A and NV is recommended for effective prevention and control.

Storing CO2 in oil and gas reservoirs offers a dual benefit: it reduces atmospheric CO2 concentration while simultaneously enhancing oil displacement efficiency and increasing crude oil production. This is achieved by injecting CO2 into producing oil and gas wells. Employing particle jet technology at the bottom of CO2 injection wells significantly expands the bottom hole diameter, thereby improving CO2 injection efficiency and storage safety. To further investigate the rock-breaking characteristics and efficiency, a finite element model for particle jet rock breaking is established by utilizing the smoothed particle hydrodynamics (SPH) method. Specifically, this new model considers the high temperature and confining pressure conditions present at the bottom hole. The dynamic response and fracturing effects of rock subjected to a particle jet are also revealed. The results indicate that particle jet impact rebound significantly influences the size of the impact crater, with the maximum first principal stress primarily concentrated on the crater’s surface. The impact creates a “v”-shaped crater on the rock surface, with both depth and volume increasing proportionally to jet inlet velocity and particle diameter. However, beyond a key particle concentration of 3%, the increase in depth and volume becomes less pronounced. Confining pressure is found to hinder particle impact rock-breaking efficiency, while high temperatures contribute to larger impact depths and breaking volumes. This research can provide theoretical support and parameter guidance for the practical application of particle impact technology in enhancing CO2 injection efficiency at the bottom hole.

Most multi-target movements are nonlinear in the process of movement. The common multi-target tracking filtering methods directly act on the multi-target tracking system of nonlinear targets, and the fusion effect is worse under the influence of different perspectives. Aiming to determine the influence of different perspectives on the fusion accuracy of multi-sensor tracking in the process of target tracking, this paper studies the multi-target tracking fusion strategy of a nonlinear system with different perspectives. A GM-JMNS-CPHD fusion technique is introduced for random outlier selection in multi-target tracking, leveraging sensors with limited views. By employing boundary segmentation from distinct perspectives, the posterior intensity function undergoes decomposition into multiple sub-intensities through SOS clustering. The distribution of target numbers within the respective regions is then characterized by the multi-Bernoulli reconstruction cardinal distribution. Simulation outcomes demonstrate the robustness and efficacy of this approach. In comparison to other algorithms, this method exhibits enhanced robustness even amidst a decreased detection probability and heightened clutter rates.

Diabetes is caused by insulin deficiency or impaired biological action, and long-term hyperglycemia leads to a variety of tissue damage and dysfunction. Therefore, the early prediction of diabetes and timely intervention and treatment are crucial. This paper proposes a robust framework for the prediction and diagnosis of type 2 diabetes (T2DM) to aid in diabetes applications in clinical diagnosis. The data-preprocessing stage includes steps such as outlier removal, missing value filling, data standardization, and assigning class weights to ensure the quality and consistency of the data, thereby improving the performance and stability of the model. This experiment used the National Health and Nutrition Examination Survey (NHANES) dataset and the publicly available PIMA Indian dataset (PID). For T2DM classification, we designed a convolutional neural network (CNN) and proposed a novel attention-oriented convolutional neural network (SECNN) through the channel attention mechanism. To optimize the hyperparameters of the model, we used grid search and K-fold cross-validation methods. In addition, we also comparatively analyzed various machine learning (ML) models such as support vector machine (SVM), logistic regression (LR), decision tree (DT), random forest (RF), and artificial neural network (ANN). Finally, we evaluated the performance of the model using performance evaluation metrics such as precision, recall, F1-Score, accuracy, and AUC. Experimental results show that the SECNN model has an accuracy of 94.12% on the NHANES dataset and an accuracy of 89.47% on the PIMA Indian dataset. SECNN models and CNN models show significant improvements in diabetes prediction performance compared to traditional ML models. The comparative analysis of the SECNN model and the CNN model has significantly improved performance, further verifying the advantages of introducing the channel attention mechanism. The robust diabetes prediction framework proposed in this article establishes an effective foundation for diabetes diagnosis and prediction, and has a positive impact on the development of health management and medical industries.

The diversity of alcohol beverage microorganisms is of great significance for improving the brewing process and the quality of alcohol beverage products. During the process of making alcoholic beverages, a group of microorganisms, represented by yeast and lactic acid bacteria, conducts fermentation. These microorganisms have complex synergistic or competitive relationships, and the participation of different microorganisms has a major impact on the fermentation process and the flavor and aroma of the product. Strain selection is one of the key steps. Utilizing scientific breeding technology, the relationship between strains can be managed, the composition of the alcoholic beverage microbial community can be improved, and the quality and flavor of the alcoholic beverage products can be increased. Currently, research on the microbial diversity of alcohol beverages has received extensive attention. However, the selection technology for dominant bacteria in alcohol beverages has not yet been systematically summarized. To breed better-quality alcohol beverage strains and improve the quality and characteristics of wine, this paper introduces the microbial diversity characteristics of the world’s three major brewing alcohols: beer, wine, and yellow wine, as well as the breeding technologies of related strains. The application of culture selection technology in the study of microbial diversity of brewed wine was reviewed and analyzed. The strain selection technology and alcohol beverage process should be combined to explore the potential application of a diverse array of alcohol beverage strains, thereby boosting the quality and flavor of the alcohol beverage and driving the sustainable development of the alcoholic beverage industry.

Abstract Optical fiber communication networks play an important role in the global telecommunication network. However, nonlinear effects in the optical fiber and transceiver noise greatly limit the performance of fiber communication systems. In this paper, the product of mutual information (MI) and communication bandwidth is used as the metric of the achievable information rate (AIR). The MI loss caused by the transceiver is also considered in this work, and the bit-wise MI, generalized mutual information (GMI), is used to calculate the AIR. This loss is more significant in the use of higher-order modulation formats. The AIR analysis is carried out in the QPSK, 16QAM, 64QAM and 256QAM modulation formats for the communication systems with different communication bandwidths and transmission distances based on the enhanced Gaussian noise (EGN) model. The paper provides suggestions for the selection of the optimal modulation format in different transmission scenarios. Graphical Abstract

Abstract Amyloid proteins are associated with a broad spectrum of neurodegenerative diseases. However, it remains a grand challenge to extract molecular structure information from intracellular amyloid proteins in their native cellular environment. To address this challenge, we developed a computational chemical microscope integrating 3D mid-infrared photothermal imaging with fluorescence imaging, termed Fluorescence-guided Bond-Selective Intensity Diffraction Tomography (FBS-IDT). Based on a low-cost and simple optical design, FBS-IDT enables chemical-specific volumetric imaging and 3D site-specific mid-IR fingerprint spectroscopic analysis of tau fibrils, an important type of amyloid protein aggregates, in their intracellular environment. Label-free volumetric chemical imaging of human cells with/without seeded tau fibrils is demonstrated to show the potential correlation between lipid accumulation and tau aggregate formation. Depth-resolved mid-infrared fingerprint spectroscopy is performed to reveal the protein secondary structure of the intracellular tau fibrils. 3D visualization of the β-sheet for tau fibril structure is achieved.

Abstract Tartary buckwheat flavonoids have a variety of effects on anti‐inflammatory, anti‐oxidation, as well as anti‐tumor and are valuable for academic research and industrial application. Helicobacter pylori (H. pylori) infection is associated with various gastrointestinal diseases in humans, and an increase in its resistance has led to the failure of many drugs. In this study, we quantified the main monomers of tartary buckwheat (Fagopyrum Tataricum (L.) Gaertn.) bran flavonoids extract through HPLC analysis. Then, we investigated the anti‐H. pylori activity and the effect on cell inflammation of tartary buckwheat flavonoids extract and its four main flavonoid monomers (rutin, quercetin, kaempferol, and nicotiflorin). The results showed that tartary buckwheat flavonoids extract and its four flavonoid monomers could inhibit the growth of H. pylori and down‐regulate the expression of proinflammatory factors IL‐6, IL‐8, and CXCL‐1 in H. pylori‐induced GES‐1 cells. Moreover, we also confirmed that tartary buckwheat flavonoids extract could reduce the expression of virulence factor gene of H. pylori. In summary, tartary buckwheat can alleviate the cell inflammation induced by H. pylori, which provides a theoretical basis for the development of tartary buckwheat healthcare products.

Abstract While exploring the process of CO/CO2 electroreduction (COxRR) is of great significance to achieve carbon recycling, deciphering reaction mechanisms so as to further design catalytic systems able to overcome sluggish kinetics remains challenging. In this work, a model single-Co-atom catalyst with well-defined coordination structure is developed and employed as a platform to unravel the underlying reaction mechanism of COxRR. The as-prepared single-Co-atom catalyst exhibits a maximum methanol Faradaic efficiency as high as 65% at 30 mA/cm2 in a membrane electrode assembly electrolyzer, while on the contrary, the reduction pathway of CO2 to methanol is strongly decreased in CO2RR. In-situ X-ray absorption and Fourier-transform infrared spectroscopies point to a different adsorption configuration of *CO intermediate in CORR as compared to that in CO2RR, with a weaker stretching vibration of the C–O bond in the former case. Theoretical calculations further evidence the low energy barrier for the formation of a H-CoPc-CO– species, which is a critical factor in promoting the electrochemical reduction of CO to methanol.

Abstract Background With the difficulties in choosing colistin sulfate and polymyxin B sulfate (PBS) for carbapenem-resistant gram-negative bacteria (CR-GNB), we compared the efficacy and safety of these two old polymyxins in treatment of critically ill patients infected with CR-GNB infection. Methods One hundred four patients infected with CR-GNB in ICU were retrospectively grouped by PBS (68 patients) or colistin sulfate (36 patients). Clinical efficacy including symptoms, inflammatory parameters, defervescence, prognosis and microbial efficacy were analyzed. Hepatotoxicity, nephrotoxicity, and hematotoxicity were evaluated by TBiL, ALT, AST, creatinine, and thrombocytes. Results Demographic characteristics between colistin sulfate and PBS were not significantly different. Most of the CR-GNB were cultured in respiratory tract (91.7% vs 86.8%), and almost all were polymyxin-sensitive (98.2% vs 100%, MIC ≤ 2 μg/ml). The microbial efficacy in colistin sulfate (57.1%) was significantly higher than PBS (30.8%) (p = 0.022), however, no significant difference in clinical success was seen in both groups (33.8% vs 41.7%), as well as mortality, defervescence, imaging remission, days in the hospital, microbial reinfections, and prognosis, and almost all patients defervesce within 7 days (95.6% vs 89.5%). Conclusions Both polymyxins can be administrated in critically ill patients infected with CR-GNB and colistin sulfate is superior to PBS in microbial clearance. These results highlight the necessity of identifying CR-GNB patients who may benefit from polymyxin and who are at higher risk of mortality.
