This study highlights hyperspectral infrared observations from the Marine-Atmospheric Emitted Radiance Interferometer (M-AERI) collected as part of the Department of Energy (DOE) Atmospheric Radiation Measurement (ARM) Mobile Facility (AMF) deployment on the icebreaker RV Polarstern during the Multidisciplinary drifting Observatory for the Study of Arctic Climate (MOSAiC) expedition from October 2019 to September 2020. The ARM M-AERI directly measures the infrared radiance emission spectrum between 520 cm−1 and 3000 cm−1 (19.2–3.3 μm) at 0.5 cm−1 spectral resolution. These ship-based observations provide a valuable set of radiance data for the modeling of snow/ice infrared emission as well as validation data for the assessment of satellite soundings. Remote sensing using hyperspectral infrared observations provides valuable information on sea surface properties (skin temperature and infrared emissivity), near-surface air temperature, and temperature lapse rate in the lowest kilometer. Comparison of the M-AERI observations with those from the DOE ARM meteorological tower and downlooking infrared thermometer are generally in good agreement with some notable differences. Operational satellite soundings from the NOAA-20 satellite were also assessed using ARM radiosondes launched from the RV Polarstern and measurements of the infrared snow surface emission from the M-AERI showing reasonable agreement.

Although phloem-feeding insects such as aphids can cause significant damage to plants, relatively little is known about early plant defenses against these insects. As a first line of defense, legumes can stop the phloem mass flow through a conformational change in phloem proteins known as forisomes in response to Ca2+ influx. However, specialized phloem-feeding insects might be able to suppress the conformational change of forisomes and thereby prevent sieve element occlusion. To investigate this possibility, we triggered forisome dispersion through application of a local heat stimulus to the leaf tips of pea (Pisum sativum), clover (Trifolium pratense) and broad bean (Vicia faba) plants infested with different pea aphid (Acyrthosiphon pisum) host races and monitored forisome responses. Pea aphids were able to suppress forisome dispersion, but this depended on the infesting aphid host race, the plant species, and the age of the plant. Differences in the ability of aphids to suppress forisome dispersion may be explained by differences in the composition and quantity of the aphid saliva injected into the plant. Various mechanisms of how pea aphids might suppress forisome dispersion are discussed.

Two major diseases that affect grapevine leaves and berries are controlled by the oomycete Pythium oligandrum. As the efficacy of biocontrol agents strongly depends on factors such as the trophic behaviors of pathogens and cultivar susceptibility, a two-disease approach was implemented to evaluate the activity of P. oligandrum against Botrytis cinerea (the necrotrophic fungus of gray mold) and Plasmopara viticola (the biotrophic oomycete of downy mildew) on two grapevine cultivars with different susceptibilities to these two pathogens. The results show that grapevine root inoculation with P. oligandrum significantly reduced P. viticola and B. cinerea infection on the leaves of the two cultivars, but with differences. This was observed when the relative expression of 10 genes was measured in response to each pathogen, and could be attributed to their lifestyles, i.e., biotrophic or necrotrophic, which are related to the activation of specific metabolic pathways of the plant. In response to P. viticola infection, genes from the jasmonate and ethylene pathways were mainly induced, whereas for B. cinerea, the genes induced were those of the ethylene–jasmonate pathway. The different levels of defense against B. cinerea and P. viticola could also explain the difference in cultivar susceptibility to these pathogens.

Abstract Objective The objective of this study was to investigate and compare the effectiveness of several toothpastes containing nanohydroxyapatite (nano-HAP) to relieve dentin hypersensitivity (DHS) with that of a commercial desensitizing dentifrice containing calcium sodium phosphosilicate (CSPS). Materials and methods In this double-blind, randomized, parallel-group clinical trial, patients diagnosed with DHS and qualified to participate were randomized into four groups: toothpaste containing 10% nano-HAP (10%nano-HAP), 15% nano-HAP (15%nano-HAP), 10% nano-HAP supplemented with potassium nitrate (KNO3) (10%nano-HAPKN), or CSPS. Subjects’ baseline and post-treatment sensitivities were assessed using visual analog scale (VAS) after the application of ice-cold and air stimuli. Abstract Two-component plant defenses such as cyanogenic glucosides are produced by many plant species, but phloem-feeding herbivores have long been thought not to activate these defenses due to their mode of feeding, which causes only minimal tissue damage. Here, however, we report that cyanogenic glycoside defenses from cassava (Manihot esculenta), a major staple crop in Africa, are activated during feeding by a pest insect, the whitefly Bemisia tabaci, and the resulting hydrogen cyanide is detoxified by conversion to beta-cyanoalanine. Additionally, B. tabaci was found to utilize two metabolic mechanisms to detoxify cyanogenic glucosides by conversion to non-activatable derivatives. First, the cyanogenic glycoside linamarin was glucosylated 1–4 times in succession in a reaction catalyzed by two B. tabaci glycoside hydrolase family 13 enzymes in vitro utilizing sucrose as a co-substrate. Second, both linamarin and the glucosylated linamarin derivatives were phosphorylated. Both phosphorylation and glucosidation of linamarin render this plant pro-toxin inert to the activating plant enzyme linamarase, and thus these metabolic transformations can be considered pre-emptive detoxification strategies to avoid cyanogenesis.

The Choloepus Hoffmani is a mammal belonging to the Xenarthra superorder; xenarthrans are distributed from North to South America. It is common for these animals to require medical attention at wildlife rescue centers after being attacked by domestic animals or run over by cars. A proper understanding of this species’ anatomy is vital in order to be able to offer them a proper level of clinical attention. This publication aims to describe the spine’s anatomical and radiographic characteristics of the Choloepus Hoffmani. Four individuals were used in this research; the spine bones were cleaned by boiling and maceration. In the results, it was possible to observe how the postcranial axial skeleton in the sloths is made up by five of distinctive vertebra types. In the spine were found: six cervical vertebrae, a variable number of thoracic vertebrae, xenarthrous lumbar vertebrae, and a fusion between the sacrum and coxal bone. Finally, four underdeveloped caudal vertebrae were also identified in a small stump-like tail. Radiographically, no pathologies were observed in the alignment or structure of the spine. In conclusion, the present study described both the osteology alongside the anatomical radiography of the vertebral column of the Choloepus hoffmani, highlighting the particularities that are not found in domestic mammals and other members of the Xenarthra superorder. Information of this kind is relevant for forensic wildlife analysis, alongside aiding the treatment of animals in this species who suffered lesions in their spine.

Abstract Background Protease inhibitors are defense proteins widely distributed in the plant kingdom. By reducing the activity of digestive enzymes in insect guts, they reduce the availability of nutrients and thus impair the growth and development of the attacking herbivore. One well-characterized class of protease inhibitors are Kunitz-type trypsin inhibitors (KTIs), which have been described in various plant species, including Populus spp. Long-lived woody perennials like poplar trees encounter a huge diversity of herbivores, but the specificity of tree defenses towards different herbivore species is hardly studied. We therefore aimed to investigate the induction of KTIs in black poplar (P. nigra) leaves upon herbivory by three different chewing herbivores, Lymantria dispar and Amata mogadorensis caterpillars, and Phratora vulgatissima beetles. Results We identified and generated full-length cDNA sequences of 17 KTIs that are upregulated upon herbivory in black poplar leaves, and analyzed the expression patterns of the eight most up-regulated KTIs via qRT-PCR. We found that beetles elicited higher transcriptional induction of KTIs than caterpillars, and that both caterpillar species induced similar KTI expression levels. Furthermore, KTI expression strongly correlated with the trypsin-inhibiting activity in the herbivore-damaged leaves, but was not dependent on damage severity, i.e. leaf area loss, for most of the genes. Conclusions We conclude that the induction of KTIs in black poplar is controlled at the transcriptional level in a threshold-based manner and is strongly influenced by the species identity of the herbivore. However, the underlying molecular mechanisms and ecological consequences of these patterns remain to be investigated.

Terpenes and phenolics are important constitutive and inducible conifer defenses against bark beetles and their associated fungi. In this study, the inducible defenses of mature Norway spruce (Picea abies) trees with different histories of attack by the spruce bark beetle, Ips typographus were tested by inoculation with the I. typographus-associated fungus Endoconidiophora polonica. We compared trees that had been under previous attack with those under current attack and those that had no record of attack. After fungal inoculation, the concentrations of mono-, sesqui-, and diterpenes in bark increased 3- to 9-fold. For the phenolics, the flavan-3-ols, catechin, and gallocatechin, increased significantly by 2- and 5-fold, respectively, while other flavonoids and stilbenes did not. The magnitudes of these inductions were not influenced by prior bark beetle attack history for all the major compounds and compound classes measured. Before fungal inoculation, the total amounts of monoterpenes, diterpenes, and phenolics (constitutive defenses) were greater in trees that had been previously attacked compared to those under current attack, possibly a result of previous induction. The transcript levels of many genes involved in terpene formation (isoprenyl diphosphate synthases and terpene synthases) and phenolic formation (chalcone synthases) were significantly enhanced by fungal inoculation suggesting de novo biosynthesis. Similar inductions were found for the enzymatic activity of isoprenyl diphosphate synthases and the concentration of their prenyl diphosphate products after fungal inoculation. Quantification of defense hormones revealed a significant induction of the jasmonate pathway, but not the salicylic acid pathway after fungal inoculation. Our data highlight the coordinated induction of terpenes and phenolics in spruce upon infection by E. polonica, a fungal associate of the bark beetle I. typographus, but provide no evidence for the priming of these defense responses by prior beetle attack.

Objective: The objective of the study was to evaluate clinical characteristics and analytical abnormalities at admission in hospitalized patients with COVID-19 and to identify which are associated with severe disease. Methods: A retrospective cohort study was performed. All adult patients admitted with COVID-19 from March 1, 2020, to May 31, 2020, were included consecutively. A descriptive analysis of clinical characteristics and analytical abnormalities at admission was made. We evaluated what comorbidities and biomarkers are associated with severe COVID-19 using a binary logistic regression model. Results: A total of 336 patients were included, 83 patients (24.7%) with severe disease. In patients with severe COVID-19, 76% were male, mean age was 71 years, and the most prevalent comorbidities were hypertension (57.8%), obesity (55.4%), dyslipidemia (50.6%), and diabetes (42.2%). In multivariate analysis, age (OR: 1.03; 95% CI 1.01-1.05; p = 0.004), male sex (OR: 2.92; 95% CI 1.62-5.27; p < 0.001), obesity (OR: 1.84; 95% CI 1.06-3.20; p = 0.030), and obstructive sleep apnea (OSA) (OR: 5.41; 95% CI 1.63-17.94; p = 0.006) were identified as comorbidities associated with severity. Patients with severe disease presented a lower arterial partial pressure of oxygen fraction and a greater inflammatory response at admission. Biomarkers associated with severe COVID-19 were lactate dehydrogenase (LDH) > 600 U/L (OR: 2.35; 95% CI 1.10-5.04; p = 0.027), serum ferritin >600 mcg/L (OR: 2.66; 95% CI 1.24-5.70; p = 0.012), and interleukin-6 (IL-6) > 40 pg/mL (OR: 4.30; 95% CI 2.04-9.04;p < 0.001). Conclusions: Patients with severe COVID-19 disease present more comorbidities and inflammatory response atadmission. Age, male sex, obesity, and OSA are associated with severity. Biomarkers at admission associated with severeCOVID-19 are LDH > 600 U/L, serum ferritin > 500 mcg/L, and IL-6 > 40 pg/mL.

Introduction: Although the development of COVID-19 vaccines represents a triumph of modern medicine, studies suggest vaccine hesitancy exists among key populations, including healthcare professionals. In December 2020, a large academic medical center offered COVID-19 vaccination to 3439 students in medicine, nursing, dentistry, and other health professions. With limited vaccine hesitancy research in this population, this study evaluates the prevalence of COVID-19 vaccine hesitancy among healthcare students, including predictors of hesitancy and top concerns with vaccination. Methods: The authors distributed a cross-sectional survey to all healthcare students (n = 3,439) from 12/17/2020 to 12/23/2020. The survey collected age, sex, perceived risk of contracting SARS-CoV-2 without vaccination, perceived impact on health if infected with SARS-CoV-2, vaccine hesitancy, and vaccine concerns. In 2021, logistic regressions identified risk factors associated with hesitancy. Results: The response rate was 30.0% (n = 1030) with median age of 25.0. Of respondents, 19.4% were hesitant to accept COVID-19 vaccination, while 66.6% reported at least one concern with the vaccine. ObjectiveAngiosarcoma is a rare malignant vascular tumor, and the management and outcome of this disease are not well-described. The aim of this study was to report the incidence, patient demographics, and outcomes of angiosarcoma based on national data.MethodsData on patients with angiosarcoma were obtained from the Surveillance, Epidemiology, and End Results (SEER) database. Inverse probability treatment weights (IPTW) were used to assess the survival benefit of operation with additional chemo or radiation therapy compared to operation alone. These variables were further compared against patients who did not receive an operation despite being initially offered one. Cox regression was used to assess survival. Statistical analyses were performed on RStudio.ResultsFor this study, 5,135 patients (46% men; median age 69, range 0–102) with angiosarcoma were identified in the SEER database between 1975 and 2016. The age-adjusted incidence rate was 1–4%. Patients were mostly non-Hispanic Caucasian (75.4%). The average tumor size was 4.7 cm, range (.1–98.9). Tumor grades were high at presentation (Grade III 17.2, Grade IV 19, and unknown 50.6%), but half were considered localized tumors. Most patients underwent an operation (66.1%). In 5.6% of patients, the operation was recommended but not performed. The overall 5-year survival was 26.7% (95% CI 25.4–28.1%). IPTW with adjusted Cox proportional hazard model demonstrated worse survival, showing that operation compared to no operation and operation with chemo/radiation compared to operation alone had worse survival between months 0 and 25 but had improved survival after month 25.ConclusionsThe incidence of angiosarcoma is low and long-term survival is poor. Multimodal therapy in the form of neoadjuvant or adjuvant chemo/radiation therapy offers significant long-term survival benefits over operation alone.

Telomerase activity correlates with distinct cell states, but can be challenging to quantify. Here the authors quantify telomerase activity across a range of biological samples using the expression of 13 genes, and show it correlates with cancer cell proliferation and stemness.

As satellite imagery collections continue to grow at an astonishing rate, so is the demand for automated and scalable object detection and segmentation. Scaling computational activities demand models that generalize well across various challenges that can hamper progress, including diverse imaging and geographic conditions, sampling bias in training data, manual ground truth collection, tooling for model reuse and accountability assessment, and poor model training strategies. A great deal of progress has been made on these challenges. We contribute to the improvement through further development of ReSFlow, a workflow that breaks the problem of model generalization into a collection of specialized exploitations. ReSFlow partitions imagery collections into homogeneous buckets equipped with exploitation models trained to perform well under each bucket.s specific context. Essentially, ReSFlow aims for generalization through stratification. Therefore, within a bucket, exploitation is a homogeneous process that mitigates heterogeneity challenges, including the number of training data and data biases that can occur over varied conditions. Furthermore, custom model architectures and rich training strategies effective for within-bucket conditions can be developed. Meanwhile, across buckets, performance metrics support systematic views of the workflow leading to optimal data labeling allocations and indications that further specialization is warranted. Herein, we discuss the formation of models during the framework's “Offline Initialization” stage. Lastly, we exploit the inherent parallelism due to bucketing to introduce model reuse and demonstrate efficacy by reducing an 89-day manual data labeling cost to zero-days in a new area of interest.

Mammography is considered the gold standard for breast cancer screening and diagnostic imaging; however, there is an unmet clinical need for complementary methods to detect lesions not characterized by mammography. Far-infrared ‘thermogram’ breast imaging can map the skin temperature, and signal inversion with components analysis can be used to identify the mechanisms of thermal image generation of the vasculature using dynamic thermal data. This work focuses on using dynamic infrared breast imaging to identify the thermal response of the stationary vascular system and the physiologic vascular response to a temperature stimulus affected by vasomodulation. The recorded data are analyzed by converting the diffusive heat propagation into a virtual wave and identifying the reflection using component analysis. Clear images of passive thermal reflection and thermal response to vasomodulation were obtained. In our limited data, the magnitude of vasoconstriction appears to depend on the presence of cancer. The authors propose future studies with supporting diagnostic and clinical data that may provide validation of the proposed paradigm.

The AGN STORM 2 campaign is a large, multiwavelength reverberation mapping project designed to trace out the structure of Mrk 817 from the inner accretion disk to the broad emission line region and out to the dusty torus. As part of this campaign, Swift performed daily monitoring of Mrk 817 for approximately 15 months, obtaining observations in X-rays and six UV/optical filters. The X-ray monitoring shows that Mrk 817 was in a significantly fainter state than in previous observations, with only a brief flare where it reached prior flux levels. The X-ray spectrum is heavily obscured. The UV/optical light curves show significant variability throughout the campaign and are well correlated with one another, but uncorrelated with the X-rays. Combining the Swift UV/optical light curves with Hubble Space Telescope UV continuum light curves, we measure interband continuum lags, τ ( λ ), that increase with increasing wavelength roughly following τ ( λ ) ∝ λ ^4/3 , the dependence expected for a geometrically thin, optically thick, centrally illuminated disk. Modeling of the light curves reveals a period at the beginning of the campaign where the response of the continuum is suppressed compared to later in the light curve—the light curves are not simple shifted and scaled versions of each other. The interval of suppressed response corresponds to a period of high UV line and X-ray absorption, and reduced emission line variability amplitudes. We suggest that this indicates a significant contribution to the continuum from the broad-line region gas that sees an absorbed ionizing continuum.

UV and optical continuum reverberation mapping is a powerful tool for probing the accretion disk and inner broad-line region. However, recent reverberation mapping campaigns in the X-ray, UV, and optical have found lags consistently longer than those expected from the standard disk reprocessing picture. The largest discrepancy to date was recently reported in Mrk 335, where UV/optical lags are up to 12 times longer than expected. Here, we perform a frequency-resolved time lag analysis of Mrk 335, using Gaussian processes to account for irregular sampling. For the first time, we compare the Fourier frequency-resolved lags directly to those computed using the popular interpolated cross-correlation function method applied to both the original and detrended light curves. We show that the anticipated disk reverberation lags are recovered by the Fourier lags when zeroing in on the short-timescale variability. This suggests that a separate variability component is present on long timescales. If this separate component is modeled as reverberation from another region beyond the accretion disk, we constrain a size scale of roughly 15 lt-days from the central black hole. This is consistent with the size of the broad-line region inferred from H β reverberation lags. We also find tentative evidence for a soft X-ray lag, which we propose may be due to light travel time delays between the hard X-ray corona and distant photoionized gas that dominates the soft X-ray spectrum below 2 keV.

Continuum reverberation mapping probes the size scale of the optical continuum-emitting region in active galactic nuclei (AGN). Through 3 yr of multiwavelength photometric monitoring in the optical with robotic observatories, we perform continuum reverberation mapping on Mrk 876. All wave bands show large-amplitude variability and are well correlated. Slow variations in the light curves broaden the cross-correlation function (CCF) significantly, requiring detrending in order to robustly recover interband lags. We measure consistent interband lags using three techniques (CCF, JAVELIN, and PyROA), with a lag of around 13 days from u to z . These lags are longer than the expected radius of 12 days for the self-gravitating radius of the disk. The lags increase with wavelength roughly following λ ^4/3 , as would be expected from thin disk theory, but the lag normalization is approximately a factor of 3 longer than expected, as has also been observed in other AGN. The lag in the i band shows an excess that we attribute to variable H α broad-line emission. A flux–flux analysis shows a variable spectrum that follows f _ν ∝ λ ^−1/3 , as expected for a disk, and an excess in the i band that also points to strong variable H α emission in that band.

The BlueFlux field campaign, supported by NASA’s Carbon Monitoring System, will develop prototype blue carbon products to inform coastal carbon management. While blue carbon has been suggested as a nature-based climate solution (NBS) to remove carbon dioxide (CO _2 ) from the atmosphere, these ecosystems also release additional greenhouse gases (GHGs) such as methane (CH _4 ) and are sensitive to disturbances including hurricanes and sea-level rise. To understand blue carbon as an NBS, BlueFlux is conducting multi-scale measurements of CO _2 and CH _4 fluxes across coastal landscapes, combined with long-term carbon burial, in Southern Florida using chambers, flux towers, and aircraft combined with remote-sensing observations for regional upscaling. During the first deployment in April 2022, CO _2 uptake and CH _4 emissions across the Everglades National Park averaged −4.9 ± 4.7 μ mol CO _2 m ^−2 s ^−1 and 19.8 ± 41.1 nmol CH _4 m ^−2 s ^−1 , respectively. When scaled to the region, mangrove CH _4 emissions offset the mangrove CO _2 uptake by about 5% (assuming a 100 year CH _4 global warming potential of 28), leading to total net uptake of 31.8 Tg CO _2 -eq y ^−1 . Subsequent field campaigns will measure diurnal and seasonal changes in emissions and integrate measurements of long-term carbon burial to develop comprehensive annual and long-term GHG budgets to inform blue carbon as a climate solution.

We present reverberation mapping measurements for the prominent ultraviolet broad emission lines of the active galactic nucleus Mrk 817 using 165 spectra obtained with the Cosmic Origins Spectrograph on the Hubble Space Telescope. Our ultraviolet observations are accompanied by X-ray, optical, and near-infrared observations as part of the AGN Space Telescope and Optical Reverberation Mapping Program 2 (AGN STORM 2). Using the cross-correlation lag analysis method, we find significant correlated variations in the continuum and emission-line light curves. We measure rest-frame delayed responses between the far-ultraviolet continuum at 1180 Å and Ly α λ 1215 Å ( ${10.4}_{-1.4}^{+1.6}$ days), N v λ 1240 Å ( ${15.5}_{-4.8}^{+1.0}$ days), Si iv + ]O iv λ 1397 Å ( ${8.2}_{-1.4}^{+1.4}$ days), C iv λ 1549 Å ( ${11.8}_{-2.8}^{+3.0}$ days), and He ii λ 1640 Å ( ${9.0}_{-1.9}^{+4.5}$ days) using segments of the emission-line profile that are unaffected by absorption and blending, which results in sampling different velocity ranges for each line. However, we find that the emission-line responses to continuum variations are more complex than a simple smoothed, shifted, and scaled version of the continuum light curve. We also measure velocity-resolved lags for the Ly α and C iv emission lines. The lag profile in the blue wing of Ly α is consistent with virial motion, with longer lags dominating at lower velocities, and shorter lags at higher velocities. The C iv lag profile shows the signature of a thick rotating disk, with the shortest lags in the wings, local peaks at ±1500 km s ^−1 , and a local minimum at the line center. The other emission lines are dominated by broad absorption lines and blending with adjacent emission lines. These require detailed models, and will be presented in future work.

We present the first results from a 100-day Swift, NICER, and ground-based X-ray–UV–optical reverberation mapping campaign of the Narrow-line Seyfert 1 Mrk 335, when it was in an unprecedented low X-ray flux state. Despite dramatic suppression of the X-ray variability, we still observe UV–optical lags as expected from disk reverberation. Moreover, the UV–optical lags are consistent with archival observations when the X-ray luminosity was >10 times higher. Interestingly, both low- and high-flux states reveal UV–optical lags that are 6–11 times longer than expected from a thin disk. These long lags are often interpreted as due to contamination from the broad line region; however the u- band excess lag (containing the Balmer jump from the diffuse continuum) is less prevalent than in other active galactic nuclei. The Swift campaign showed a low X-ray-to-optical correlation (similar to previous campaigns), but NICER and ground-based monitoring continued for another 2 weeks, during which the optical rose to the highest level of the campaign, followed ∼10 days later by a sharp rise in X-rays. While the low X-ray countrate and relatively large systematic uncertainties in the NICER background make this measurement challenging, if the optical does lead X-rays in this flare, this indicates a departure from the zeroth-order reprocessing picture. If the optical flare is due to an increase in mass accretion rate, this occurs on much shorter than the viscous timescale. Alternatively, the optical could be responding to an intrinsic rise in X-rays that is initially hidden from our line of sight.

The AGN STORM 2 Collaboration targeted the Seyfert 1 galaxy Mrk 817 for a year-long multiwavelength, coordinated reverberation mapping campaign including Hubble Space Telescope, Swift, XMM-Newton, NICER, and ground-based observatories. Early observations with NICER and XMM revealed an X-ray state 10 times fainter than historical observations, consistent with the presence of a new dust-free, ionized obscurer. The following analysis of NICER spectra attributes variability in the observed X-ray flux to changes in both the column density of the obscurer by at least one order of magnitude ( N _H ranges from ${2.85}_{-0.33}^{+0.48}\times {10}^{22}\,{\mathrm{cm}}^{-2}$ to ${25.6}_{-3.5}^{+3.0}\times {10}^{22}\,{\mathrm{cm}}^{-2}$ ) and the intrinsic continuum brightness (the unobscured flux ranges from 10 ^−11.8 to 10 ^−10.5 erg s ^−1 cm ^−2 ). While the X-ray flux generally remains in a faint state, there is one large flare during which Mrk 817 returns to its historical mean flux. The obscuring gas is still present at lower column density during the flare, but it also becomes highly ionized, increasing its transparency. Correlation between the column density of the X-ray obscurer and the strength of UV broad absorption lines suggests that the X-ray and UV continua are both affected by the same obscuration, consistent with a clumpy disk wind launched from the inner broad-line region.

La Inteligencia Artificial ha ayudado a lidiar diferentes problemas relacionados con los datos masivos y a su vez con su tratamiento, diagnóstico y detección de enfermedades como la que actualmente nos preocupa, la Covid-19. El objetivo de esta investigación ha sido analizar y desarrollar la clasificación de imágenes de neumonía a causa de covid-19 para un diagnostico efectivo y óptimo. Se ha usado Transfer-Learning aplicando ResNet, DenseNet, Poling y Dense layer para la elaboración de los modelos de red propios Covid-UPeU y Covid-UPeU-TL, utilizando las plataformas Kaggle y Google colab, donde se realizaron 4 experimentos. El resultado con una mejor clasificación de imágenes se obtuvo en el experimento 4 prueba N°2 con el modelo Covid-UPeU-TL donde Acc.Train: 0.9664 y Acc.Test: 0.9851. Los modelos implementados han sido desarrollados con el propósito de tener una visión holística de los factores para la optimización en la clasificación de imágenes de neumonía a causa de COVID-19. Abstract Artificial Intelligence has helped to deal with different problems related to massive data in turn to the treatment, diagnosis and detection of diseases such as the one that currently has us in concern, Covid-19. The objective of this research has been to analyze and develop the classification of images of pneumonia due to covid-19 for an effective and optimal diagnosis. Transfer-Learning has been used applying ResNet, DenseNet, Poling and Dense layer for the elaboration of the own network models Covid-Upeu and Covid-UpeU-TL, using Kaggle and Google colab platforms, where 4 experiments have been carried out. The result with a better classification of images was obtained in experiment 4 test N ° 2 with the Covid-UPeU-TL model where Acc.Train: 0.9664 and Acc.Test: 0.9851. The implemented models have been developed with the purpose of having a holistic view of the factors for optimization in the classification of pneumonia COVID-19 images. Some plants produce toxic isothiocyanates that protect them against pathogens. Here, Chen et al. show that the plant pathogenic fungus Sclerotinia sclerotiorum converts isothiocyanates into non-toxic compounds via glutathione conjugation and, more effectively, via hydrolysis to amines using an isothiocyanate hydrolase.

Abstract The purpose of the EANM Dosimetry Committee Series on “Standard Operational Procedures for Dosimetry” (SOP) is to provide advice to scientists and clinicians on how to perform patient-specific absorbed dose assessments. This SOP describes image and data acquisition parameters and dosimetry calculations to determine the absorbed doses delivered to whole-body, tumour and normal organs following a therapeutic administration of 131I mIBG for the treatment of neuroblastoma or adult neuroendocrine tumours. Recommendations are based on evidence in recent literature where available and on expert opinion within the community. This SOP is intended to promote standardisation of practice within the community and as such is based on the facilities and expertise that should be available to any centre able to perform specialised treatments with radiopharmaceuticals and patient-specific dosimetry. A clinical example is given to demonstrate the application of the absorbed dose calculations.

The shear volumes of data generated from earth observation and remote sensing technologies continue to make major impact; leaping key geospatial applications into the dual data and compute-intensive era. As a consequence, this rapid advancement poses new computational and data processing challenges. We implement a novel remote sensing data flow (RESFlow) for advancing machine learning to compute with massive amounts of remotely sensed imagery. The core contribution is partitioning massive amounts of data into homogeneous distributions for fitting simple models. RESFlow takes advantage of Apache Spark and the availability of modern computing hardware to harness the acceleration of deep learning inference on expansive remote sensing imagery. The framework incorporates a strategy to optimize resource utilization across multiple executors assigned to a single worker. We showcase its deployment in both computationally and data-intensive workloads for pixel-level labeling tasks. The pipeline invokes deep learning inference at three stages; during deep feature extraction, deep metric mapping, and deep semantic segmentation. The tasks impose compute-intensive and GPU resource sharing challenges motivating for a parallelized pipeline for all execution steps. To address the problem of hardware resource contention, our containerized workflow further incorporates a novel GPU checkout routine and the ticketing system across multiple workers. The workflow is demonstrated with NVIDIA DGX accelerated platforms and offers appreciable compute speed-ups for deep learning inference on pixel labeling workloads; processing 21 028 TB of imagery data and delivering output maps at area rate of 5.245 sq.km/s, amounting to 453 168 sq.km/day-reducing a 28 day workload to 21 h.

Kagami-Ogata syndrome (KOS) is a rare genomic imprinting disorder of chromosome 14 with characteristic facial features, a small, bell-shaped thorax, muscular hypotonia, and abdominal wall defects. We present a case of a kidney incarcerated in a posterolateral Bochdalek type congenital diaphragmatic hernia (CDH), intestinal malrotation and stage 2 rectocele in KOS. Successful repair of CDH and malrotation was achieved with Ladd's procedure, CDH repair, Nissen fundoplication and gastrostomy tube placement. She had no post-operative complications and is currently tolerating 100% enteral feeds. CDH, intestinal malrotation and rectocele have not been previously reported in KOS. Despite overall poor prognosis, KOS patients with CDH and malrotation can be repaired successfully with improvement in quality of life.

Gut enzymes can metabolize plant defense compounds and thereby affect the growth and fitness of insect herbivores. Whether these enzymes also influence feeding preference is largely unknown. We studied the metabolization of taraxinic acid β-D-glucopyranosyl ester (TA-G), a sesquiterpene lactone of the common dandelion (Taraxacum officinale) that deters its major root herbivore, the common cockchafer larva (Melolontha melolontha). We have demonstrated that TA-G is rapidly deglucosylated and conjugated to glutathione in the insect gut. A broad-spectrum M. melolontha β-glucosidase, Mm_bGlc17, is sufficient and necessary for TA-G deglucosylation. Using cross-species RNA interference, we have shown that Mm_bGlc17 reduces TA-G toxicity. Furthermore, Mm_bGlc17 is required for the preference of M. melolontha larvae for TA-G-deficient plants. Thus, herbivore metabolism modulates both the toxicity and deterrence of a plant defense compound. Our work illustrates the multifaceted roles of insect digestive enzymes as mediators of plant-herbivore interactions.

Cruciferous plants in the order Brassicales defend themselves from herbivory using glucosinolates: sulfur-containing pro-toxic metabolites that are activated by hydrolysis to form compounds, such as isothiocyanates, which are toxic to insects and other organisms. Some herbivores are known to circumvent glucosinolate activation with glucosinolate sulfatases (GSSs), enzymes that convert glucosinolates into inactive desulfoglucosinolates. This strategy is a major glucosinolate detoxification pathway in a phloem-feeding insect, the silverleaf whitefly Bemisia tabaci, a serious agricultural pest of cruciferous vegetables. In this study, we identified and characterized an enzyme responsible for glucosinolate desulfation in the globally distributed B. tabaci species MEAM1. In in vitro assays, this sulfatase showed a clear preference for indolic glucosinolates compared with aliphatic glucosinolates, consistent with the greater representation of desulfated indolic glucosinolates in honeydew. B. tabaci might use this detoxification strategy specifically against indolic glucosinolates since plants may preferentially deploy indolic glucosinolates against phloem-feeding insects. In vivo silencing of the expression of the B. tabaci GSS gene via RNA interference led to lower levels of desulfoglucosinolates in honeydew. Our findings expand the knowledge on the biochemistry of glucosinolate detoxification in phloem-feeding insects and suggest how detoxification pathways might facilitate plant colonization in a generalist herbivore.

Abstract Induced responses in plants, initiated by herbivory, create potential for trait‐mediated indirect interactions among herbivores. Responses to an initial herbivore may change a number of plant traits that subsequently alter ecological processes with additional herbivores. Although common, indirect interactions between taxonomically distant herbivores, such as mammals and insects, are less studied than between taxonomically related species (i.e., insect–insect). In terms of mammal–insect interactions, effects on insect numbers (e.g., density) are relatively well studied, whereas effects on performance (e.g., fecundity) are rarely explored. Moreover, few studies have explored mammal–insect interactions on coniferous plants. The aim of this study was to investigate the effect of mammalian induced responses on insect performance. We specifically investigated the effect of moose (Alces alces) browsing on Scots pine (Pinus sylvestris) and subsequent effects on sawfly (Neodiprion sertifer) performance. Sawfly larvae were reared on browsed, clipped, and unbrowsed control pine trees in a controlled field experiment. Afterward, cocoon weight was measured. Needle C:N ratio and di‐terpene content were measured in response to browsing. Sawfly performance was enhanced on trees browsed by moose. Cocoon weight (proxy for fecundity) was 9 and 13% higher on browsed and clipped trees compared to unbrowsed trees. Cocoon weight was weakly related to needle C:N ratio, and browsed trees had lower a C:N ratio compared to unbrowsed trees. Needle di‐terpene content, known to affect sawfly performance, was neither affected by the browsing treatments nor did it correlate with sawfly weight. We conclude that mammalian herbivory can affect insect herbivore performance, with potential consequences for ecological communities and with particular importance for insect population dynamics. The measured plant variables could not fully explain the effect on sawfly performance providing a starting point for the consideration of additional plant responses induced by mammalian browsing affecting insect performance.

Abstract Background The central aim was to examine the accuracy of the full range of daily activities recorded in self-report time-use diaries against data from two objective passive data collection devices (wearable camera and accelerometer) serving as criterion reference instruments. This enabled systematic checks and comparisons on the timing, sequence and duration of activities recorded from the three data sources. Methods Participants (n = 148) were asked to complete a single-day self-report paper time-use diary designed for use in the Harmonised European Time Use Study (HETUS), while simultaneously wearing a camera that continuously recorded images of their activities, and an accelerometer tracking physical movement. In a reconstruction interview shortly after the data collection period, participants viewed the camera images to help researchers interpret the image sequences. Of the initial 148 recruits (multi-seed snowball sample, 59% women, aged 18–91, 43% > 40) 131 returned usable diary and camera records (of whom 124 also provided a usable whole-day accelerometer record. We compare time allocation estimates from the diary and camera records, and also match the diary and camera records to the simultaneously recorded accelerometer vector magnitudes. Results The data were examined at three analytic levels: aggregate, individual diarist and timeslot. Abstract Background This section defined time use (TU) research, illustrating its relevance for public health. TUR in the health context is the study of health-enhancing and health-compromising behaviours that are assessed across a 24 h day. The central measurement is the use of Time Use Diaries, which capture 24–48 h, typically asking about behaviour in each 15-min period. TUR is used for understanding correlates of health behaviours, and as a form of population surveillance, assessing behavioural trends over time. Main body This paper is a narrative review examining the history of time use research, and the potential uses of TU data for public health research. The history of TUR started in studies of the labour force and patterns of work in the late 19th and early twentieth century, but has more recently been applied to examining health issues. Initial studies had a more economic purpose but over recent decades, TU data have been used to describe the distribution and correlates of health-enhancing patterns of human time use. These studies require large multi-country population data sets, such as the harmonised Multinational Time Use Study hosted at the University of Oxford. TU data are used in physical activity research, as they provide information across the 24-h day, that can be examined as time spent sleeping, sitting/standing/light activity, and time spent in moderate-vigorous activities. TU data are also used for sleep research, examining eating and dietary patterns, exploring geographic distributions in time use behaviours, examining mental health and subjective wellbeing, and examining these data over time. The key methodological challenge has been the development of harmonised methods, so population TU data sets can be compared within and between-countries and over time. Conclusions TUR provides new methods for examining public health research questions where a temporal dimension is important. These time use surveys have provided unique data over decades and in many countries that can be compared. They can be used for examining the effects of some large public health interventions or policies within and between countries.

Abstract Background Behaviour has diverse economic, social and health consequences. Linking time spent in different daily activities to energy expenditure (EE) is one way of investigating the health and physiological consequences of behaviour and identifying targets to improve population health and well-being. Methods We estimated behaviour-related EE for respondents to time use surveys (TUS) from three countries: UK 2001, Poland 2012 and US 2003–13. The Harmonised Multinational Time Use Survey (MTUS) activity categories were matched to MET estimates from the 2011 Compendium of Physical Activities. We attach METs values to each successive activity in the TUS, together with both the original UK, Polish and US activity classifications and the 68-category MTUS activity classification. We used TUS estimates of activity durations across 24-h to estimate the Physical Activity Level (PAL) for respondents from the three countries and the average time spent and MET values for different activity categories. Results PAL values ranged from 1.59 in the US to 1.74 in Poland. The main sources of daily EE from PA were paid and unpaid work activities. Discretionary PA accounted for only a very small part (~ 3%) of adult daily energy expenditures. Using the harmonised MTUS 68-activity classification reduced the variability of the aggregate PAEE measure by ~ 20%, but the patterns of association between key demographics (age, sex, educational attainment) were unaffected. TUS data were further used to (1) identify sources of daily PA, and (2) assess adherence to physical activity guidelines (PAG) on a single-day basis. Estimated adherence levels were similar to those reported from other TUS as well as frequency based estimates. Conclusions Comparative studies of energy expenditure based on harmonised time use activity categories could provide insight into the relative importance of different activities for energy expenditure across different countries and demographic groups. However, new observational studies combining TUS data with accelerometer, direct observation and other measures of activity intensity are required for more accurate MET assignments to activity categories in TUS.

Abstract Background Over the last 150 years, advanced economies have seen the burden of disease shift to non-communicable diseases. The risk factors for these diseases are often co-morbidities associated with unhealthy weight. The prevalence of overweight/obesity among adults in the advanced countries of the English-speaking world is currently more than two-thirds of the adult population. However, while much attention has concentrated on changes in diet that might have provoked this rapid increase in unhealthy weight, changes in patterns of eating have received little attention. Methods This article examines a sequence of large-scale, time use surveys in urban Australia stretching from 1974 to 2006. The earliest survey in 1974 (conducted by the Cities Commission) was limited to respondents aged between 18 and 69 years, while the later surveys (by the Australian Bureau of Statistics) included all adult (15 years of age or over) living private dwellings. Since time use surveys capture every activity in a day, they contain much information about mealtimes and the patterns of eating. This includes duration of eating, number of eating occasions and the timing of eating. Inferential statistics were used to test the statistical significance of these changes and the size of the effects. Results The eating patterns of urban Australian adults have changed significantly over a 32-year period and the magnitude of this change is non-trivial. Total average eating time as main activity has diminished by about a third, as have eating occasions, affecting particularly luncheon and evening meals. However, there is evidence that eating as secondary activity that accompanies another activity is now almost as frequent as eating at mealtimes. Moreover, participants seem not to report it. Conclusions Contemporary urban Australians are spending less time in organized shared meals. These changes have occurred the over same period during which there has been a public health concern about the prevalence of unhealthy weight. Preliminary indications are that societies that emphasize eating as a commensal, shared activity through maintaining definite, generous lunch breaks and prioritizing eating at mealtimes, achieve better public health outcomes. This has implications for a strategy of health promotion, but to be sure of this we need to study countries with these more socially organized eating patterns.

Abstract Because of today's advancements in surgical techniques and perioperative management skills, surgeons are beginning to explore the usefulness of the laparoscopic approach in managing periampullary tumors. However, as a result of its innate complexity and associated high surgery‐related complications, its applicability to the general surgical community remains controversial. To date, only retrospective data from high‐volume centers support the safety and feasibility of laparoscopic pancreaticoduodenectomy (Lap PD) for the treatment of benign conditions and malignant periampullary tumors. In addition, various surgical techniques in terms of port placement, dissection, and reconstruction have evolved in different centers depending on the preferred method commonly used by the surgeon through accumulated experience. In our center, we used a stepwise approach and standardized our surgical technique to overcome this technically demanding procedure. A collaborative implementation of video review and analysis, practice training and simulation, operating room didactics, and strict adherence to our stepwise approach in Lap PD, might potentially improve the surgical skills of young hepatobiliary surgeons and possibly overcome the volume‐based learning curve of Lap PD.

While the role of effective population size (N e) in explaining variation in genetic diversity has received much attention, the role of spontaneous mutation rate is largely ignored. Here, Xu et al. show that giant duckweed has a high N e yet low genetic diversity, likely due to its low mutation rate.

Abstract Background Plants grow in multi-species communities rather than monocultures. Yet most studies on the emission of volatile organic compounds (VOCs) from plants in response to insect herbivore feeding focus on one plant species. Whether the presence and identity of neighboring plants or plant community attributes, such as plant species richness and plant species composition, affect the herbivore-induced VOC emission of a focal plant is poorly understood. Methods We established experimental plant communities in pots in the greenhouse where the focal plant species, red clover (Trifolium pratense), was grown in monoculture, in a two species mixture together with Geranium pratense or Dactylis glomerata, or in a mixture of all three species. We measured VOC emission of the focal plant and the entire plant community, with and without herbivory of Spodoptera littoralis caterpillars caged on one red clover individual within the communities. Results Herbivory increased VOC emission from red clover, and increasing plant species richness changed emissions of red clover and also from the entire plant community. Neighbor identity strongly affected red clover emission, with highest emission rates for plants growing together with D. glomerata. Conclusion The results from this study indicate that the blend of VOCs perceived by host searching insects can be affected by plant-plant interactions.

Abstract Background Nitrilases are nitrile-converting enzymes commonly found within the plant kingdom that play diverse roles in nitrile detoxification, nitrogen recycling, and phytohormone biosynthesis. Although nitrilases are present in all higher plants, little is known about their function in trees. Upon herbivory, poplars produce considerable amounts of toxic nitriles such as benzyl cyanide, 2-methylbutyronitrile, and 3-methylbutyronitrile. In addition, as byproduct of the ethylene biosynthetic pathway upregulated in many plant species after herbivory, toxic β-cyanoalanine may accumulate in damaged poplar leaves. In this work, we studied the nitrilase gene family in Populus trichocarpa and investigated the potential role of the nitrilase PtNIT1 in the catabolism of herbivore-induced nitriles. Results A BLAST analysis revealed three putative nitrilase genes (PtNIT1, PtNIT2, PtNIT3) in the genome of P. trichocarpa. While PtNIT1 was expressed in poplar leaves and showed increased transcript accumulation after leaf herbivory, PtNIT2 and PtNIT3 appeared not to be expressed in undamaged or herbivore-damaged leaves. Recombinant PtNIT1 produced in Escherichia coli accepted biogenic nitriles such as β-cyanoalanine, benzyl cyanide, and indole-3-acetonitrile as substrates in vitro and converted them into the corresponding acids. In addition to this nitrilase activity, PtNIT1 showed nitrile hydratase activity towards β-cyanoalanine, resulting in the formation of the amino acid asparagine. The kinetic parameters of PtNIT1 suggest that the enzyme utilizes β-cyanoalanine and benzyl cyanide as substrates in vivo. Indeed, β-cyanoalanine and benzyl cyanide were found to accumulate in herbivore-damaged poplar leaves. The upregulation of ethylene biosynthesis genes after leaf herbivory indicates that herbivore-induced β-cyanoalanine accumulation is likely caused by ethylene formation. Conclusions Our data suggest a role for PtNIT1 in the catabolism of herbivore-induced β-cyanoalanine and benzyl cyanide in poplar leaves.

Background Although intravesical BCG is the standard treatment of high-grade non-muscle invasive bladder cancer (NMIBC), response rates remain unsatisfactory. In preclinical models, rapamycin enhances BCG vaccine efficacy against tuberculosis and the killing capacity of γδ T cells, which are critical for BCG’s antitumor effects. Here, we monitored immunity, safety, and tolerability of rapamycin combined with BCG in patients with NMIBC.Methods A randomized double-blind trial of oral rapamycin (0.5 or 2.0 mg daily) versus placebo for 1 month was conducted in patients with NMIBC concurrently receiving 3 weekly BCG instillations (NCT02753309). The primary outcome was induction of BCG-specific γδ T cells, measured as a percentage change from baseline. Post-BCG urinary cytokines and immune cells were examined as surrogates for local immune response in the bladder. Secondary outcomes measured were adverse events (AEs) and tolerability using validated patient-reported questionnaires.Results Thirty-one patients were randomized (11 placebo, 8 rapamycin 2.0 mg, and 12 rapamycin 0.5 mg). AEs were similar across groups and most were grade 1–2. One (12.5%) patient randomized to 2.0 mg rapamycin was taken off treatment due to stomatitis. No significant differences in urinary symptoms, bowel function, or bother were observed between groups. The median (IQR) percentage change in BCG-specific γδ T cells from baseline per group was as follows: −26% (−51% to 24%) for placebo, 9.6% (−59% to 117%) for rapamycin 0.5 mg (versus placebo, p=0.18), and 78.8% (−31% to 115%) for rapamycin 2.0 mg (versus placebo, p=0.03). BCG-induced cytokines showed a progressive increase in IL-8 (p=0.02) and TNF-α (p=0.04) over time for patients on rapamycin 2.0 mg, whereas patients receiving placebo had no significant change in urinary cytokines. Compared with placebo, patients receiving 2.0 mg rapamycin had increased urinary γδ T cells at the first week of BCG (p=0.02).Conclusions Four weeks of 0.5 and 2.0 mg oral rapamycin daily is safe and tolerable in combination with BCG for patients with NMIBC. Rapamycin enhances BCG-specific γδ T cell immunity and boosts urinary cytokines during BCG treatment. Further study is needed to determine long-term rapamycin safety, tolerability and effects on BCG efficacy.

The emphasis on team science in clinical and translational research increases the importance of collaborative biostatisticians (CBs) in healthcare. Adequate training and development of CBs ensure appropriate conduct of robust and meaningful research and, therefore, should be considered as a high-priority focus for biostatistics groups. Comprehensive training enhances clinical and translational research by facilitating more productive and efficient collaborations. While many graduate programs in Biostatistics and Epidemiology include training in research collaboration, it is often limited in scope and duration. Therefore, additional training is often required once a CB is hired into a full-time position. This article presents a comprehensive CB training strategy that can be adapted to any collaborative biostatistics group. This strategy follows a roadmap of the biostatistics collaboration process, which is also presented. A TIE approach (Teach the necessary skills, monitor the Implementation of these skills, and Evaluate the proficiency of these skills) was developed to support the adoption of key principles. The training strategy also incorporates a “train the trainer” approach to enable CBs who have successfully completed training to train new staff or faculty.

We present findings from three waves of a population-representative, UK time-use diary survey conducted both pre- and in real time during full 'lockdown', and again following the easing of social restrictions. We used an innovative online diary instrument that has proved both reliable and quick-to-field. Combining diary information on activity, location, and co-presence to estimate infection risks associated with daily behavior, we show clear changes in risk-associated behavior between the pre, full-lockdown and post full-lockdown periods. We document a shift from more to less risky daily behavior patterns (combinations of activity/location/co-presence categories) between the pre-pandemic pattern and full lockdown in May/June 2020, followed by a reversion (although not a complete reversal) of those patterns in August 2020 following the end of the first lockdown. Because, in general, a populations' time use changes relatively slowly, the behavioral changes revealed may be interpreted as a consequence of the UK COVID-19 lockdown social restrictions and their subsequent relaxation.

Time-use data can often be perceived as inaccessible by non-specialists due to their unique format. This article introduces the ATUS-X diary visualization tool that aims to address the accessibility issue and expand the user base of time-use data by providing users with opportunity to quickly visualize their own subsamples of the American Time Use Survey Data Extractor (ATUS-X). Complementing the ATUS-X, the online tool provides an easy point-and-click interface, making data exploration readily accessible in a visual form. The tool can benefit a wider academic audience, policy-makers, non-academic researchers, and journalists by removing accessibility barriers to time use diaries.

The relationship between plants and insects is continuously evolving, and many insects rely on biochemical strategies to mitigate the effects of toxic chemicals in their food plants, allowing them to feed on well-defended plants. Spodoptera frugiperda, the fall armyworm (FAW), accepts a number of plants as hosts, and has particular success on plants of the Poaceae family such as maize, despite their benzoxazinoid (BXD) defenses. BXDs stored as inert glucosides are converted into toxic aglucones by plant glucosidases upon herbivory. DIMBOA, the main BXD aglucone released by maize leaves, can be stereoselectively re-glucosylated by UDP-glycosyltransferases (UGTs) in the insect gut, rendering it non-toxic. Here, we identify UGTs involved in BXD detoxification by FAW larvae and examine how RNAi-mediated manipulation of the larval glucosylation capacity toward the major maize BXD, DIMBOA, affects larval growth. Our findings highlight the involvement of members of two major UGT families, UGT33 and UGT40, in the glycosylation of BXDs. Most of the BXD excretion in the frass occurs in the form of glucosylated products. Furthermore, the DIMBOA-associated activity was enriched in the gut tissue, with a single conserved UGT33 enzyme (SfUGT33F28) being dedicated to DIMBOA re-glucosylation in the FAW gut. The knock-down of its encoding gene reduces larval performance in a strain-specific manner. This study thus reveals that a single UGT enzyme is responsible for detoxification of the major maize-defensive BXD in this pest insect.

The C5 hemiterpenes isoprene and 2-methyl-3-buten-2-ol (MBO) are important biogenic volatiles emitted from terrestrial vegetation. Isoprene is emitted from many plant groups, especially trees such as Populus, while emission of MBO is restricted to certain North American conifers, including species of Pinus. MBO is also a pheromone emitted by several conifer bark beetles. Both isoprene and MBO have typically been measured by proton-transfer reaction mass spectrometry (PTR-MS), but this method cannot accurately distinguish between them because of their signal overlap. Our study developed a method for using selective ion flow tube mass spectrometry (SIFT-MS) that allows simultaneous on-line measurement of isoprene and MBO by employing different reagent ions. The use of m/z(NO+) = 68 u for isoprene and m/z(O2+) = 71 u for MBO gave minimal interference between the compounds. We tested the suitability of the method by measuring the emission of young trees of Populus, Picea, and Pinus. Our results largely confirm previous findings that Populus nigra, Picea glauca, and Picea abies emit isoprene and Pinus ponderosa emits MBO, but we also found MBO to be emitted by Picea abies. Thus SIFT-MS provides a reliable, easy to use, on-line measuring tool to distinguish between isoprene and MBO. The method should be of use to atmospheric chemists, tree physiologists and forest entomologists, among others.

Grapevine trunk diseases (GTDs) are increasing in vineyards in many grape production regions. Among the pathogens causing these diseases, Neofusicoccum parvum, is one of the most frequent and virulent. To control GTDs, biocontrol is being developed using plant beneficial microorganisms. Strains of the oomycete Pythium oligandrum have been shown to naturally colonize grapevine roots in vineyards in several countries in Europe. This study examined the ability of the root-coloniser P. oligandrum to induce grapevine resistance against N. parvum, by deciphering the gene expression changes in a set of 62 genes involved in different grapevine defence pathways. Two greenhouse assays showed that the wood necrosis of vine cuttings caused by N. parvum was reduced by 65% when P. oligandrum colonized root systems of the plants. The relative expression levels of selected genes in the host trunks were studied by real-time PCR. Plant responses were assessed after inoculation by P. oligandrum and/or N. parvum, at three different sampling time points (0, 14 and 150 d after N. parvum inoculation). Sampling time influenced gene expressions for the different inoculation treatments. At each sampling time, specific host responses to the different treatments were also detected, for controls, and for inoculations with P. oligandrum, N. parvum or P. oligandrum + N. parvum. When P. oligandrum colonized grapevine root systems, inoculation with the pathogen was associated with increased up-regulation and over-expression of particular genes, including those regulating Pathogen-Related proteins, cell wall reinforcement proteins and hormone signalling pathways. A priming effect of the grapevine defence system was induced in roots colonized by P. oligandrum.

The methylerythritol 4-phosphate (MEP) pathway of isoprenoid biosynthesis produces chlorophyll side chains and compounds that function in resistance to abiotic stresses, including carotenoids, and isoprene. Thus we investigated the effects of moderate and severe drought on MEP pathway function in the conifer Picea glauca, a boreal species at risk under global warming trends. Although moderate drought treatment reduced the photosynthetic rate by over 70%, metabolic flux through the MEP pathway was reduced by only 37%. The activity of the putative rate-limiting step, 1-deoxy-D-xylulose-5-phosphate synthase (DXS), was also reduced by about 50%, supporting the key role of this enzyme in regulating pathway metabolic flux. However, under severe drought, as flux declined below detectable levels, DXS activity showed no significant decrease, indicating a much-reduced role in controlling flux under these conditions. Both MEP pathway intermediates and the MEP pathway product isoprene incorporate administered 13CO2 to high levels (75–85%) under well-watered control conditions indicating a close connection to photosynthesis. However, this incorporation declined precipitously under drought, demonstrating exploitation of alternative carbon sources. Despite the reductions in MEP pathway flux and intermediate pools, there was no detectable decline in most major MEP pathway products under drought (except for violaxanthin under moderate and severe stress and isoprene under severe stress) suggesting that the pathway is somehow buffered against this stress. The resilience of the MEP pathway under drought may be a consequence of the importance of the metabolites formed under these conditions.

White spruce (Picea glauca) emits monoterpenes that function as defensive signals and weapons after herbivore attack. We assessed the effects of drought and methyl jasmonate (MeJA) treatment, used as a proxy for herbivory, on monoterpenes and other isoprenoids in P. glauca. The emission of monoterpenes was significantly increased after MeJA treatment compared to the control, but drought suppressed the MeJA-induced increase. The composition of the emitted blend was altered strongly by stress, with drought increasing the proportion of oxygenated compounds and MeJA increasing the proportion of induced compounds such as linalool and (E)-β-ocimene. In contrast, no treatment had any significant effect on the levels of stored monoterpenes and diterpenes. Among other MEP pathway-derived isoprenoids, MeJA treatment decreased chlorophyll levels by 40%, but had no effect on carotenoids, while drought stress had no impact on either of these pigment classes. Of the three described spruce genes encoding 1-deoxy-D-xylulose-5-phosphate synthase (DXS) catalyzing the first step of the MEP pathway, the expression of only one, DXS2B, was affected by our treatments, being increased by MeJA and decreased by drought. These findings show the sensitivity of monoterpene emission to biotic and abiotic stress regimes, and the mediation of the response by DXS genes.

O-Methylated benzoxazinoids (BXs) and flavonoids are widespread defenses against herbivores and pathogens in the grasses (Poaceae). Recently, two flavonoid O-methyltransferases (FOMTs), ZmFOMT2 and ZmFOMT3, have been reported to produce phytoalexins in maize (Zea mays). ZmFOMT2 and ZmFOMT3 are closely related to the BX O-methyltransferases (OMTs) ZmBX10-12 and ZmBX14, suggesting a common evolutionary origin in the Poaceae. Here, we studied the evolution and enzymatic requirements of flavonoid and BX O-methylation activities in more detail. Using BLAST searches and phylogenetic analyses, we identified enzymes homologous to ZmFOMT2 and ZmFOMT3, ZmBX10-12, and ZmBX14 in several grasses, with the most closely related candidates found almost exclusively in species of the Panicoideae subfamily. Biochemical characterization of candidate enzymes from sorghum (Sorghum bicolor), sugar cane (Saccharum spp.), and teosinte (Zea nicaraguensis) revealed either flavonoid 5-O-methylation activity or DIMBOA-Glc 4-O-methylation activity. However, DIMBOA-Glc 4-OMTs from maize and teosinte also accepted flavonols as substrates and converted them to 3-O-methylated derivatives, suggesting an evolutionary relationship between these two activities. Homology modeling, sequence comparisons, and site-directed mutagenesis led to the identification of active site residues crucial for FOMT and BX OMT activity. However, the full conversion of ZmFOMT2 activity into BX OMT activity by switching these residues was not successful. Only trace O-methylation of BXs was observed, indicating that amino acids outside the active site cavity are also involved in determining the different substrate specificities. Altogether, the results of our study suggest that BX OMTs have evolved from the ubiquitous FOMTs in the PACMAD clade of the grasses through a complex series of amino acid changes.

Within the southern California Current ecosystem there are two well-documented breaks in marine community structure at Point Conception and Punta Eugenia. We explored the presence of similar breaks in a diverse zooplankton community through metabarcoding of mixed net tow tissue samples collected during an expedition from Monterey to Baja California in February of 2012. We recovered a high diversity of species as well as patterns of species presence that align with their previously documented ranges in this region. We found a clear break at Punta Eugenia in overall zooplankton community structure, while Point Conception was weakly linked to changes in community structure. We analyzed this dataset through two parallel bioinformatic pipelines to examine the robustness of these results. Our overall conclusions were consistent across both pipelines, however there were differences in species detection. This study illustrates the utility of metabarcoding analysis on mixed tissue samples for recovering known patterns of diversity, as well as allowing elucidation of broad patterns of community differentiation across many groups of organisms.

Background: In developed countries, Human Immunodeficiency Virus type-1 (HIV-1) infection has become a chronic disease despite the positive effects of anti-retroviral therapies (ART), but still at least half of the HIV infected population shown signs of cognitive impairment. Therefore, biomarkers of HIV cognitive decline are urgently needed. Methods: We analyze the opening of one of the larger channels expressed by humans, pannexin-1 (Panx-1) channels, in the uninfected and HIV infected population (n = 175). We determined channel opening and secretion of intracellular second messengers released through the channel such as PGE2 and ATP. Also, we correlated the opening of Panx-1 channels with the circulating levels of PGE2 and ATP as well as cogntive status of the individuals analyzed. Findings: Here, we demonstrate that Panx-1 channels on fresh PBMCs obtained from uninfected individuals are closed and no significant amounts of PGE2 and ATP are detected in the circulation. In contrast, in all HIV-infected individuals analyzed, even the ones under effective ART, a spontaneous opening of Panx-1 channels and increased circulating levels of PGE2 and ATP were detected. Circulating levels of ATP were correlated with cognitive decline in the HIV-infected population supporting that ATP is a biomarker of cognitive disease in the HIV-infected population. Interpretation: We propose that circulating levels of ATP could predict CNS compromise and lead to the breakthroughs necessary to detect and prevent brain compromise in the HIV-infected population.
