Abstract

Acute myeloid leukemia (AML) is a high remission, high relapse fatal blood cancer. Although mTORC1 is a master regulator of cell proliferation and survival, its inhibitors have not performed well as AML treatments. To uncover the dynamics of mTORC1 activity in vivo, fluorescent probes are developed to track single cell proliferation, apoptosis and mTORC1 activity of AML cells in the bone marrow of live animals and to quantify these activities in the context of microanatomical localization and intra-tumoral heterogeneity. When chemotherapy drugs commonly used clinically are given to mice with AML, apoptosis is rapid, diffuse and not preferentially restricted to anatomic sites. Dynamic measurement of mTORC1 activity indicated a decline in mTORC1 activity with AML progression. However, at the time of maximal chemotherapy response, mTORC1 signaling is high and positively correlated with a leukemia stemness transcriptional profile. Cell barcoding reveals the induction of mTORC1 activity rather than selection of mTORC1 high cells and timed inhibition of mTORC1 improved the killing of AML cells. These data define the real-time dynamics of AML and the mTORC1 pathway in association with AML growth, response to and relapse after chemotherapy. They provide guidance for timed intervention with pathway-specific inhibitors.

Abstract

We prove a purely local form of a result of Saito and Yatagawa. They proved that the characteristic cycle of a constructible \'etale sheaf is determined by wild ramification of the sheaf along the boundary of a compactification. But they had to consider ramification at all the points of the compactification. We give a pointwise result, that is, we prove that the characteristic cycle of a constructible \'etale sheaf around a point is determined by wild ramification at the point. Abstract

In this paper, we consider linear prediction models in the form of a sparse linear combination of rules, where a rule is an indicator function defined over a hyperrectangle in the input space. Since the number of all possible rules generated from the training dataset becomes extremely large, it has been difficult to consider all of them when fitting a sparse model. In this paper, we propose Safe Optimal Rule Fit (SORF) as an approach to resolve this problem, which is formulated as a convex optimization problem with sparse regularization. The proposed SORF method utilizes the fact that the set of all possible rules can be represented as a tree. By extending a recently popularized convex optimization technique called safe screening, we develop a novel method for pruning the tree such that pruned nodes are guaranteed to be irrelevant to the prediction model. This approach allows us to efficiently learn a prediction model constructed from an exponentially large number of all possible rules. We demonstrate the usefulness of the proposed method by numerical experiments using several benchmark datasets.

76. Imaging Findings of Human Papillomavirus-Positive and Human Papillomavirus-Negative Oropharyngeal Squamous Cell Carcinoma Associated with Recurrence.

Abstract

Objectives: This study aimed to compare the imaging findings associated with the recurrence of HPV-positive and HPV-negative oropharyngeal squamous cell carcinoma (OPSCC). Methods: In total, 68 patients (51 men; mean age, 64.4 years; age range, 41–86 years; 48 HPV-positive patients and 20 HPV-negative patients) with histopathologically proven OPSCC who underwent CT, MRI, and 18F-FDG-PET/CT before treatment between October 2014 and July 2022 were enrolled in this study. The imaging findings were retrospectively evaluated and statistically compared. Results: HPV-positive OPSCC had a significantly lower recurrence rate compared with that of HPV-negative OPSCC (p < 0.01). Among HPV-positive OPSCCs, patients with recurrence were considerably older than those without recurrence (p < 0.05); however, the T and N categories did not differ between the two groups. Meanwhile, among HPV-negative OPSCCs, the T and N categories were associated with recurrence (p < 0.05). Furthermore, the attenuation on contrast-enhanced CT (p < 0.05) and signal intensity on contrast-enhanced T1-weighted images (p < 0.05) of nodal metastases were significantly lower in recurrence patients compared to those in nonrecurrence patients. Cystic change in nodal metastases in HPV-positive and HPV-negative OPSCCs were similar in patients with and without recurrence. Conclusions: The T and N categories were associated with recurrence in HPV-negative OPSCC but not in HPV-positive OPSCC. We prove that wild ramification of a constructible sheaf on a surface is determined by that of the restrictions to all curves. We deduce from this result that the Euler-Poincar\'e characteristic of a constructible sheaf on a variety of arbitrary dimension over an algebraically closed field is determined by wild ramification of the restrictions to all curves.
