62. The coronavirus transmissible gastroenteritis virus causes infection after receptor-mediated endocytosis and acid-dependent fusion with an intracellular compartment

Abstract

Udgivelsesdato: 1998-Jan, Aminopeptidase N is a species-specific receptor for transmissible gastroenteritis virus (TGEV), which infects piglets, and for the 229E virus, which infects humans. It is not known whether these coronaviruses are endocytosed before fusion with a membrane of the target cell, causing a productive infection, or whether they fuse directly with the plasma membrane. We have studied the interaction between TGEV and a cell line (MDCK) stably expressing recombinant pig aminopeptidase N (pAPN). By electron microscopy and flow cytometry, TGEV was found to be associated with the plasma membrane after adsorption to the pAPN-MDCK cells. TGEV was also observed in endocytic pits and apical vesicles after 3 to 10 min of incubation at 38 degrees C. The number of pits and apical vesicles was increased by the TGEV incubation, indicating an increase in endocytosis. After 10 min of incubation, a distinct TGEV-pAPN-containing population of large intracellular vesicles, morphologically compatible with endosomes, was found. A higher density of pAPN receptors was observed in the pits beneath the virus particles than in the surrounding plasma membrane, indicating that TGEV recruits pAPN receptors before endocytosis. Ammonium chloride and bafilomycin A1 markedly inhibited the TGEV infection as judged from virus production and protein biosynthesis analyses but did so only when added early in the course of the infection, i.e., about 1 h after the start of endocytosis. Together our results point to an acid intracellular compartment as the site of fusion for TGEV.

Abstract

To assess risk for cattle-to-human transmission of prions that cause uncommon forms of bovine spongiform encephalopathy (BSE), we inoculated mice expressing human PrP Met129 with field isolates. Abstract

The summer-dormancy period among collections of Phalaris tuberosa L. from the eastern Mediterranean region when grown at Davis, California, ranged from 30 to 127 days. Plants from the more arid sites of origin tended to exhibit the longer dormancy. There were exceptions, however, and these, together with the diversity of behaviour in this material, suggest that genotypes with a potential for increased summer growth may be selected for use on rangelands. Abstract

Because variant Creutzfeldt-Jakob disease (vCJD) in humans probably results from consumption of products contaminated with tissue from animals with bovine spongiform encephalopathy, whether infectious prion protein is present in ruminant muscles is a crucial question. Here we show that experimentally and naturally scrapie-affected sheep accumulate the prion protein PrPSc in a myocyte subset. In naturally infected sheep, PrPSc is detectable in muscle several months before clinical disease onset. The relative amounts of PrPSc suggest a 5,000-fold lower infectivity for muscle as compared to brain. [ABSTRACT FROM AUTHOR]

Abstract

Non-infectious UV-inactivated transmissible gastroenteritis virus (TGEV) was previously shown to induce interferon alpha (IFIMα) secretion following in vitroincubation with blood mononuclear cells. In this study, pig foetuses at different stages of gestation were injected in uterowith (a) partially UV-inactivated wild TGEV or (b) fully UV-inactivated wild or dm49-4 mutant TGEV Coronavirus. Nucleated cells from foetal liver, bone marrow, spleen and blood were isolated 10 or 20 h after injection and assayed ex vivo for IFNα secretion by ELISPOT and ELISA techniques. The administration of TGEV induced IFNα-secreting cells in foetal lymphohaematopoietic organs at mid-gestation. In contrast, IFNα was not detected in control sham-operated foetuses. A specific point mutation in the amino acid sequence of the viral membrane glycoprotein M of TGEV mutant dm49-4 was associated with lower or absent IFNα in uteroinducibility by mutant virus as compared with wild virus. Row cytometry analysis did not show differences in leukocyte surface marker expression between control and TGEV- or between dm49-4 and wild virus-treated foetus cells, with the exception of a reduction in percentages of polymorphonuclear cells in TGEV-treated lymphohaematopoietic tissues, which is probably due to IFNα secretion. The present data provided in vivoevidence of IFNα secretion at the cell level in foetal lymphohaematopoietic organs. Such IFNα-secreting cells in lymphohaematopoietic tissues may be the source of IFNα detected during foetal infections.

Abstract

We looked for the presence of interferon in the digestive tract of newborn piglets infected with a virulent strain of transmissible gastroenteritis virus (Coronaviridae). High levels of type 1 interferon activity were found early in the disease in jejunal and ileal parts of the intestine as well as in the serum. Enterocytes appeared to be involved in the interferon synthesis. These findings raise the question of the role of interferon in the pathogenesis of viral enteritis.

86. Induction of alpha interferon by transmissible gastroenteritis coronavirus: role of transmembrane glycoprotein E1

Abstract

Epithelial cells infected with the coronavirus transmissible gastroenteritis virus (TGEV) and fixed by glutaraldehyde induced a high alpha interferon (IFN-alpha) production in nonimmune porcine as well as human or bovine peripheral blood mononuclear cells (PBMC). IFN-alpha was detected as early as 3 h after exposure of PBMC to infected cells and at producer/inducer cell ratios as low as 1/1. Two of four monoclonal antibodies directed against the viral transmembrane glycoprotein E1 could block the IFN-inducing capacity of both TGEV-infected cells and viral particles. On the other hand, IFN-alpha induction was not markedly affected by monoclonal antibodies directed against other E1 epitopes, against peplomer glycoprotein E2, or against nucleocapsid protein. Thus, these findings strongly imply that IFN induction by TGEV results from interactions between an outer membrane domain of E1 and the PBMC membrane.

87. De novo methylation of tumour suppressor genes CDKN2A and CDKN2B is a rare finding in B-cell chronic lymphocytic leukaemia

Abstract

De novo methylation of the 5′CpG island has been recently reported as an alternative mechanism of inactivation for the tumour suppressor genes CDKN2A and CDKN2B. We examined CDKN2A methylation status at diagnosis in 42 B-cell chronic lymphocytic leukaemia (CLL) patients, in 19 cases the CDKN2B methylation status was also analysed. No homozygous CDKN2A/2B deletion was detected, but four patients (9%) displayed an aberrant CDKN2A methylation status and only one had hypermethylated CDKN2B. De novo methylation was associated with silencing of gene expression. These results confirm that CDKN2A/2B inactivation by deletion is a rare event in CLL and suggest that aberrant methylation could be an alternative way of inactivation very rarely involved in the development of some CLL.

92. Influence of Lodging on Yield and Other Characters in Winter Wheat1

Abstract

Yields were reduced about one‐third due to lodging 1 to 2 weeks before heading and 1 to 2 weeks after heading. Lodging between those periods caused less reduction averaging about 15% just before heading. Early lodging mainly reduced the number of kernels per head and later lodging reduced the size of the kernel. Protein content of the grain was higher in lodged than standing wheat. The total amount of protein per acre, however, was less in lodged areas.
