01 april 2023, Item does not contain fulltext, Genome-wide association studies (GWAS) have now correlated hundreds of genetic variants with complex genetic diseases and drug efficacy. Functional characterization of these factors remains challenging, particularly because of the lack of human model systems. Molecular and nanotechnological advances, in particular the ability to generate patient-specific PSC lines, differentiate them into diverse cell types, and seed and combine them on microfluidic chips, have led to the establishment of organ-on-a-chip (OoC) platforms that recapitulate organ biology. OoC technology thus provides unique personalized platforms for studying the effects of host genetics and environmental factors on organ physiology. In this review we describe the technology and provide examples of how OoCs may be used for disease modeling and pharmacogenetic research.

Item does not contain fulltext, BACKGROUND: Anti-TNF drugs, such as infliximab, are associated with attenuated antibody responses after SARS-CoV-2 vaccination. We aimed to determine how the anti-TNF drug infliximab and the anti-integrin drug vedolizumab affect vaccine-induced neutralising antibodies against highly transmissible omicron (B.1.1.529) BA.1, and BA.4 and BA.5 (hereafter BA.4/5) SARS-CoV-2 variants, which possess the ability to evade host immunity and, together with emerging sublineages, are now the dominating variants causing current waves of infection. METHODS: CLARITY IBD is a prospective, multicentre, observational cohort study investigating the effect of infliximab and vedolizumab on SARS-CoV-2 infection and vaccination in patients with inflammatory bowel disease (IBD). Patients aged 5 years and older with a diagnosis of IBD and being treated with infliximab or vedolizumab for 6 weeks or longer were recruited from infusion units at 92 hospitals in the UK. In this analysis, we included participants who had received uninterrupted biological therapy since recruitment and without a previous SARS-CoV-2 infection. The primary outcome was neutralising antibody responses against SARS-CoV-2 wild-type and omicron subvariants BA.1 and BA.4/5 after three doses of SARS-CoV-2 vaccine. We constructed Cox proportional hazards models to investigate the risk of breakthrough infection in relation to neutralising antibody titres. The study is registered with the ISRCTN registry, ISRCTN45176516, and is closed to accrual. - Abstract
Background Patients with Inflammatory bowel disease (IBD) receiving anti-TNF or JAK-inhibitor therapy have attenuated responses to COVID-19 vaccination. We aimed to determine how IBD treatments affect neutralising antibody responses against the currently dominant Omicron BA.4/5 variants. Methods We prospectively recruited 329 adults (68 healthy controls (HC) and 261 IBD) who had received three doses of COVID-19 vaccine at nine UK centres. The IBD population was established (>12 weeks therapy) on either thiopurine (n=60), infliximab (IFX) (n=43), thiopurine & IFX combination (n=46), ustekinumab (n=43), vedolizumab (n=46) or tofacitinib (n=23). Pseudoneutralisation assays were performed and the half maximal inhibitory concentration (NT50) of participant sera was calculated. The primary outcome was anti-SARS-CoV-2 neutralising response against wild-type (WT) virus and the BA.4/5 variant after the second and third doses of anti-SARS-CoV-2 vaccine, stratified by immunosuppressive therapy, adjusting for prior infection, ethnicity, vaccine type and age. Results Heterologous (two doses adenovirus vaccine, third dose mRNA vaccine) and homologous (three doses mRNA vaccine) vaccination strategies significantly increased neutralising titres against both WT SARS-CoV-2 virus and the BA.4/5 variants in HCs and IBD (fig 1). It is unknown how infliximab impacts vaccine-induced serological responses against highly transmissible Omicron variants, which possess the ability to evade host immunity and are now the dominating variants causing current waves of infection. Methods: CLARITY IBD is a prospective, multicentre, observational cohort study investigating the impact of infliximab and vedolizumab on SARS-CoV-2 infection and vaccination in patients with inflammatory bowel disease (IBD). In the current work, the primary outcome was neutralising antibody responses against SARS-CoV-2 wild-type and Omicron BA.1 and BA.4/5 variants after three doses of COVID-19 vaccination in 1288 patients with IBD without prior COVID-19 infection, who were established on either infliximab (n=871) or vedolizumab (n=417). Background: The coconut black-headed caterpillar, Opisina arenosella Walker (Lepidoptera: Xyloryctidae) has been a serious defoliator of coconut plants in many coconut producing countries including Vietnam. Trichospilus pupivorus Ferrière (Hymenoptera: Eulophidae) is pupal endo-parasitoid of O. arenosella with high percent parasitism and abundance in the coconut fields and appears to be a good biological control agent against the coconut black-headed caterpillar in Vietnam. The objective of this study was to investigate the developmental biology of T. pupivorus on O. arenosella. Methods: The experiment was carried out at the Faculty of Agronomy, Nong Lam University, Viet Nam during 2020 and 2021. The developmental biology of T. pupivorus on O. arenosella was studied in the laboratory at a constant temperature of 28°C and a photoperiod of 12L: 8D. Result: Total developmental time from egg to adult emergence was 14.0 days. The females laid a mean of 124.2 eggs during an average lifespan of 9.4 days. The number of progenies emergence from a single pupa of O. arenosella was 103.8. The offspring sex ratio was female-biased (8.0%). The results would contribute to the knowledge of the biology of this parasitoid to optimize a mass rearing system for a biological control program against O. arenosella. [ABSTRACT FROM AUTHOR]

Kim Le, Hady Saheb Department of Ophthalmology, McGill University, Montreal, QC, Canada Abstract: Trabecular micro-bypass stents, commonly known as iStents, are micro-invasive glaucoma surgery (MIGS) devices used to treat open-angle glaucoma. Like other MIGS procedures that enhance trabecular outflow, the iStent lowers intraocular pressure (IOP) by creating a direct channel between the anterior chamber and Schlemm’s canal. iStents are typically implanted at the time of phacoemulsification for patients with open-angle glaucoma and visually significant cataracts. This review summarizes the published data regarding the efficacy, safety, and cost considerations of trabecular micro-bypass stents. Most studies found statistically significant reductions in mean IOP and ocular medication use after combined phacoemulsification with single or double iStent implantation. The devices were found to be very safe, with a safety profile similar to that of cataract surgery. Complications were infrequent, with the most common complications being temporary stent obstruction or malposition, which resolved with observation or secondary procedures. Future studies are needed to evaluate long-term outcomes, patient satisfaction, cost effectiveness, and expanded indications. Several attempts have been made to explore the mechanism and new therapies for these patients. We intend to clarify the expression change of SRC and SIRT1 in tamoxifen-resistant breast cancer cells and explore their functions on tamoxifen resistance.Methods: SRC and SIRT1 expressions were analyzed by RNA sequencing, qPCR and Western blotting. Loss and gain of function of SRC and SIRT1 were utilized to indicate their oncogenic roles in tamoxifen resistance in vitro and in vivo. Kaplan–Meier analysis and receiver operating characteristic curve were used to evaluate the survival and the predicted effects of SRC and SIRT1 on patients’ prognosis.Results: High expressions of SRC and/or SIRT1 were found in tamoxifen-resistant cells and related to poor overall survival (p< 0.05 for SRC, p< 0.001 for SIRT1, p< 0.001 for SRC and SIRT1) and cancer-specific survival (p< 0.05 for SRC, p< 0.01 for SIRT1, p< 0.01 for SRC and SIRT1) of tamoxifen-treated breast cancer patients. Down-regulation of SRC (p< 0.01) or SIRT1 (p< 0.05) separately reversed the resistance to tamoxifen and the minimal concentration of SRC inhibitor KX-01 (p< 0.05) or SIRT1 inhibitor EX527 (p< 0.001) could also suppress cell proliferation. The expression level of SIRT1 was positively correlated with that of SRC. Overexpression of SRC significantly promotes the cell resistance to tamoxifen inhibited by SIRT1 (p< 0.01). In vivo experiments confirmed the effects of SRC on tumor growth by over- or down-regulating SRC expression (p< 0.001 and p< 0.001, respectively).Conclusion: SRC and SIRT1 are both up-regulated in tamoxifen-resistant breast cancer cells and related to a poor prognosis in tamoxifen-treated breast cancer. Moreover, SRC could promote tamoxifen resistance by up-regulating SIRT1. Current trauma recommendations do not advise whole-body computed tomography (CT) in hemodynamically stable patients.Objective: To measure the prevalence of abnormal CT findings in areas other than the prime area of clinical interest in multitrauma patients.Method: The records of 462 consecutive adult patients who underwent whole-body CT scans between 2004 and 2005 at Westmead Hospital, a Level 1 trauma center, were assessed. Clinical parameters, including suspected clinical injury, regional tenderness, bruising, loss of consciousness, scalp laceration, and unequal chest air entry, were examined. Correlation was made with CTs performed from the brain to symphysis pubis, on a Toshiba 16 slice machine, with evaluation of clinically significant findings.Results: The prevalence of abnormal CT findings distant to the prime area of concern varied between anatomical areas: brain (10.3%–88.7%), skull (6.7%–39.7%), facial bones (4.4%–54.3%), cervical spine (5.6%–13.7%), thoracolumbar spine (5.6%–26.7%), chest (30.8%–54.4%), and abdomen/pelvis (20%–27.2%).Conclusion: There is a high prevalence of injuries remote from the prime area of clinical concern in multitrauma patients. The improving access to increasing amounts of biomedical data provides completely new chances for advanced patient stratification and disease subtyping strategies. This requires computational tools that produce uniformly robust results across highly heterogeneous molecular data. Unsupervised machine learning methodologies are able to discover de-novo patterns in such data. Biclustering is especially suited by simultaneously identifying sample groups and corresponding feature sets across heterogeneous omics data. The performance of available biclustering algorithms heavily depends on individual parameterization and varies with their application. Here, we developed MoSBi (Molecular Signature identification using Biclustering), an automated multi-algorithm ensemble approach that integrates results utilizing an error model-supported similarity network. We evaluated the performance of MoSBi on transcriptomics, proteomics and metabolomics data, as well as synthetic datasets covering various data properties. Profiting from multi-algorithm integration, MoSBi identified robust group and disease specific signatures across all scenarios overcoming single algorithm specificities. Furthermore, we developed a scalable network-based visualization of bicluster communities that support biological hypothesis generation. MoSBi is available as an R package and web-service to make automated biclustering analysis accessible for application in molecular sample stratification.

Hydrothermally synthesized carbon nanodots (CDs) were impregnated on TiO2. The product (CDs/TiO2) was mechanically mixed with zeolite Y for application in toluene photocatalytic oxidation reaction under UV radiation. Material properties of the samples were investigated by different methods. Toluene vapor was chosen as a typical volatile organic compound to investigate the performance of CDs/TiO2 - zeolite Y photocatalyst when these technological parameters were changed: toluene concentration, gas flow rate, humidity and UV light intensity. In each reaction, only one parameter was changed and the remaining conditions were fixed. The toluene concentrations at the beginning and the end of each reaction were analyzed with the use of gas chromatography (GC). The results of different reaction conditions show the trends for toluene treatment of the CDs/TiO2 - zeolite Y catalyst, thereby providing specific explanations for these trends. The experiments also show that toluene removal is highest when the toluene concentration in the inlet gas is 314 ppmv, the flow rate is 3 L/h, the humidity is 60%, and the catalyst (CDs/TiO2 - zeolite Y composite with 70% zeolite in weight) is illuminated by 4 UV lamps. Total developmental time from egg to adult emergence was 13.8 days. The females laid a mean of 98.1 eggs during an average lifespan of 30.7 days. Fecundity peaked at a large range of age 7-29 days. Digitale transformatie staat centraal in de groeiambities van organisaties. Digitale transformatie heeft impact op de organisatieverandering en dwingt tot aanpassingen aan structuren, processen en organisatiecultuur. Onderzoek toont aan dat de essentiële factor, organisatiecultuur vaak wordt vergeten. Dit blijkt een cruciaal aspect te zijn voor het succes van digitale transformatie. Dit onderzoek richt zich op de vraag: Hoe kan een gevestigde financiële dienstverleningsorganisatie een digitale organisatiecultuur ontwikkelen om digitaal transformatiesucces mogelijk te maken? De digitale volwassenheid van de organisatie wordt gebruikt als maatstaf. Om deze vraag te beantwoorden wordt een gevalideerd instrument gebruikt om de digitale volwassenheid van de organisatie in kaart te brengen. Uit academisch onderzoek zijn twaalf culturele waarden geïdentificeerd van een geschikte organisatiecultuur ter ondersteuning van succesvolle digitale transformatie. Met een kwalitatieve onderzoeksstrategie wordt data verzameld om enerzijds een verdieping op de bestaande theorie omtrent relevante culturele waarden voor digitale transformatie en anderzijds prescriptieve inzichten bieden voor elk van de relevante culturele waarden hoe deze in de praktijk kunnen worden behaald.

Joubert syndrome is a rare neurodevelopmental disorder with malformations in cerebellum and brainstem. Many ocular abnormalities have been noted in Joubert syndrome, but the association of this syndrome with adult-onset ptosis has not been described to date. This 24-year-old Joubert patient developed a cerebrospinal fluid cyst in her midbrain. She had signs of bilateral third nerve palsy and abducens palsy in the left eye. The bilateral central third nerve palsy causing functional blindness secondary to severe bilateral levator palsy was treated successfully with silicone sling frontalis suspension, as the seventh nerve nucleus was not involved.Keywords: Joubert syndrome, ptosis, diplopia, molar tooth formation, silicone sling implants
