Abstract

Precipitating antibodies prepared in rabbits with human seminal plasma (HSP) have been used in a preliminary study to define the antigenicity of HSP by use of gel precipitin methods. Also, a method for evaluating and preserving liquid precipitates of acid phosphatase in polyacrylamide is described. Some ten or eleven antigens were demonstrated by Ouchterlony agar immunodiffusion (ID). Eight of these appeared to be HSP-specific and two or three were also found in normal human serum (NILS). Five of the HSP-specific antigens were shown by agar immunoelectrophoresis (IEP) and by ID to be acid phosphatase. Four of these constituted the basic isoenzyme pattern and were shown by IEP and ID to be shared by prostatic fluid (HPF). Titration of HPF by ID yielded antigen titres of 46, 33, 17 and 3 for the four isoenzymes. IEP patterns obtained with lISP and HPF showed two isoenzymes migrating to the a-globulin zone. The third isoenzyme migrated to the a-2-fast β area, while a fourth was seen as a slow beta precipitin arc formed consistently by HPF, and variably by HSP. HSP contained a fifth isoenzyme which also migrated to the slow β area. Human prostate extract (HPE) showed only two, sometimes three, of these components, one of the slower components being frequently faint in appearance or absent. In HSP and HPF isoenzyme migrating to the a-2-fast β zone exhibited two distinct characteristics: biphasic migration and limited diffusion, suggesting a heterogeneous high molecular weight isoenzyme or an isoenzyme complex. However, observed differences between HSP, HPF and HPE in migration rate, relative concentrations of subfractions, lateral diffusion, etc. leave open the question of the nature of the heterogeneity. 75. Isolation of a detergent-solubilized maltase/glucoamylase from rat intestine and its comparison with a maltase/glucoamylase solubilized by papain

Abstract

Maltase/glucoamylase from the rat intestinal brush-border membrane was solubilized by homogenization of the intestinal mucosa in buffer containing 0.5% Triton X-100. After removal of the detergent with butan-1-ol, the enzyme was purified by chromatography on Sepharose 4B and DEAE-cellulose. The final specific activity was 70.3 units/mg of protein in six preparations, comparing favourably with the specific activity of 65.0 units/mg of protein of a pure papain-solubilized maltase/glucoamylase previously isolated and characterized by us [Flanagan & Forstner (1978) Biochem. J.173, 553–563]. The two enzymes were compared. Both migrated as single bands with the same mobility on sodium dodecyl sulphate/polyacrylamide-gel electrophoresis, were eluted at the same volume from Sepharose 4B, and had the same sedimentation pattern in mannitol gradients. The amino acid composition was similar; content of total apolar residues differed by 1.0mol%. Antibodies prepared against either enzyme gave identical precipitin lines with each. Neither enzyme bound tritiated Triton X-100. The only difference noted was the tendency of the detergent-solubilized enzyme to aggregate on storage, whereas the papain-solubilized enzyme remained unchanged. Both enzymes had two N-termini, glycine and arginine. When the two enzymes were dissociated by boiling in sodium dodecyl sulphate, each exhibited the same five species on sodium dodecyl sulphate/polyacrylamide-gel electrophoresis. Single N-termini were found in the two smaller species, 1 (glycine) and 2 (arginine), whereas larger species (3–5) had both N-terminal amino acids. Both the Triton- and papain-solubilized enzymes appear to be oligomers of species 1 and 2, indicating that the native enzyme contains two subunit types. Aggregation in aqueous solutions does not depend on a proteolytically susceptible peptide fragment at the N-terminus of either subunit.

Abstract

Medulloblastoma (MB) is a neoplasm linked to dysregulated cerebellar development. Previously, we demonstrated that the Sonic Hedgehog (SHH) subgroup grows hierarchically, with Sox2+cells at the apex of tumor progression and relapse. To test whether this mechanism is rooted in a normal developmental process, we studied the role of Sox2 in cerebellar development. We find that the external germinal layer (EGL) is derived from embryonic Sox2+precursors and that the EGL maintains a rare fraction of Sox2+cells during the first postnatal week. Through lineage tracing and single-cell analysis, we demonstrate that these Sox2+cells are within the Atoh1+lineage, contribute extensively to adult granule neurons, and resemble Sox2+tumor cells. Critically, constitutive activation of the SHH pathway leads to their aberrant persistence in the EGL and rapid tumor onset. We propose that failure to eliminate this rare but potent developmental population is the tumor initiation mechanism in SHH-subgroup MB.

77. Biochemical studies of chloroleukemia in female Sprague-Dawley rats

Abstract

The chloroleukemia (chloroma) in the Cancer Research Centre, U.B.C. could be transplanted into adult Sprague-Dawley rats with tumour homogenates, whole blood and ascitic fluid containing intact cells. Cell-free filtrates of the ascitic fluid did not give rise to tumours. It was considered probable that intact cells were required for the transmission of this leukemia. The present study showed that there was a 'latent period' of 10 to 12 days after which the chloroma grew rapidly until the death of the animal (ca 27 days). The tumour contained substantial amounts of free protoporphyrin and had high myeloperoxidase activity. The protoporphyrin concentration increased markedly and continuously with the age of the tumour but its rate of synthesis (from glycine-2-C¹⁴) declined after the 19th day, i.e..protoporphyrin accumulated in the tumour. The myeloperoxidase activity increased between the 13th and 15th day after transplantation and thereafter fell sharply. The effect of two drugs - Vinblastine (VLB) and 3,5-dicarbethoxy-1,4-dihydrocollidine (DDC) on certain aspects of the biosynthetic activity of this particular adult chloroma was investigated. VLB inhibited the incorporation of glycine-2-C¹⁴ into both RNA and protein earlier and to a greater extent than into the DNA and protoporphyrin. This tumour, although of hematopoietic origin, did not therefore show the high sensitivity of normal bone marrow to VLB. DDC markedly increased the Δ -aminolevulinic acid synthetase activity and protoporphyrin concentration in the liver of rats but had no effect on the tumour protoporphyrin synthesis although there was more than a 20-fold increase in Δ -aminolevulinic acid synthetase activity. It was suggested therefore that the enzyme Δ-aminolevulinic acid synthetase may be rate-limiting in the liver but not in the tumour. In the course of the present work, an improved method for the separation of porphyrin esters based on partition chromatography on alumina was developed.

87. Disputed ethnic identity and the role of public education: the case of Moldova

Abstract

This dissertation examines the case of Moldova, where two ethnic nationalisms (Moldovan and Romanian) have battled over the content of national identity over the last two decades. Historically, the land on which Moldova lies was caught in a tug-of-war between Russia (later Soviet Union) and Romania. Sharing the same ethnic traits with Romania, Romanian nationalism emerged early in Moldova, only to be later deconstructed by the Soviets through deportations and executions of Romanian nationalists, and eventually reconstructed as a "Moldavian" identity. This dissertation has two goals. First, through archival and historical research it traces the process of formation of ethnic identity and the emergence of two conflicting nationalisms in Moldova. Second, it investigates the role of public education in ethno-national identity formation through interviews and a survey of Moldovan students. I hypothesize that because self-identified Romanians control the school curricula, the younger generation is more likely to identify as Romanian than the rest of the population - whose connection with school is more distant. To test this thesis, I conducted an original survey of students from seven schools. In contrast to the primordialist theory of nationalism, these findings indicate a relatively fluid national identity. However, the case of Moldovan nationalism also contradicts the instrumentalist school of thought, which over-emphasizes the socio-economic interests of nationalist agents and fails to take into account the cultural motivations of nationalism. Moldovan story indicates that at the fore-front of Romanian nationalist movement were the relatively well-off intellectuals and not the rural and urban working people as the accounts of Cash and Crowther indicate. Lastly, the structuralist (materialist) school fails to acknowledge the power of ideas and the effect they have on historical events. While material means like print media, capital markets, and urbanization facilitated the diffusion of these ideas, they did not create them. As the case of Moldova illustrates, the emergence of nationalism cannot be explained without an understanding of the motivations of the agents involved.
