Our objective was to find herd factors associated with pigs testing seropositive for Salmonella. Data were collected from 359 finishing-pig herds in Germany, Denmark, Greece, The Netherlands and Sweden, between 1996 and 1998. Pigs fed non-pelleted feed (dry or wet) had 2- and 2.5-times lower odds of seropositivity, compared to pigs fed pelleted feed. The protective effect of non-pelleted feed over pelleted feed may be ascribed to the structure and composition. Also, pigs that were given whey (to drink or as the liquid part of the diet) had 2.6-times lower odds to test seropositive than pigs not getting whey. Pigs produced in batches in herds with hygienic-lock facilities had >3-times lower odds for testing seropositive compared to pigs in herds where only one or neither factor was present. In herds where the caretaker(s) washed hands consistently before tending to the animals, pigs had 1.5-times lower odds of seropositivity than pigs in herds where the caretaker did not. Pigs which were able to have snout contact with pigs in neighbouring pens (because pen separations were either open or too low) had 1.7-times higher odds to test seropositive compared to pigs for which such contact was prevented. Pigs in herds recruiting from more than three supplier herds had three-times higher odds to test seropositive than pigs in herds which breed their own replacement stock or recruit from a maximum of three supplier herds.

Blood samples were taken from 50 finishing pigs at 90–105 kg in each of 59 randomly selected farrow-to-finish herds. The sera were tested for antibodies to Salmonella enterica by the Danish mix-ELISA. Samples with an optical density of >10% were considered to be positive. Associations between the odds of seropositivity of pigs and possible risk factors were evaluated in multivariable logistic regression models. The results of the analysis indicated that pigs fed non-pelleted dry or wet ration had 11 (P=0·0004) or 9 (P=0·02) times, respectively, lower odds of seropositivity than those fed pelleted ration. The risk of seropositivity was 4 (P=0·0006) times higher in pigs fed a combination of chlortetracycline, procaine penicillin and sulphamethazine during fattening than in those fed an approved growth promotor or a probiotic.

Ticks were collected from 249 dogs, admitted over a one-year period (1996–1997) to a Veterinary Teaching Hospital and four private veterinary practices located in the county of Thessaloniki, Greece. A total of 2812 ticks (Acari: Ixodidae) were collected, of which 2511 (89.3%) were Rhipicephalus sanguineus (Latreille) adults (1070 males, 1441 females), 156 (5.5%) R. turanicus Pomerantsev adults (35 males, 121 females), 100 (3.6%) Rhipicephalus spp. nymphs and 45 (1.6%) Rhipicephalus spp. larvae. Dogs living outdoors, in rural areas and in close proximity to farm animals, were infested with higher numbers of ticks than dogs living indoors. Tick attachment sites were recorded on 237 dogs, harbouring a total of 1711 ticks. The most heavily infested sites were, in decreasing frequency, the ear pinnae, neck, interdigital skin folds, trunk, head, ventrum, extremities and the tail. Cutaneous lesions at the attachment sites were noticed in 129 dogs and then mainly in the more severely infested animals.

The purpose of this study was the comparison of the diagnostic sensitivity between buffy coat (BC), peripheral blood (PB), lymph node (LN), bone marrow (BM) and short-term culture (P-D) cytology that has been based on the detection of Ehrlichia canis morulae, in the acute phase of canine monocytic ehrlichiosis (CME). Their cellular localization, total numbers and microscopic differentials were also investigated. The highest sensitivities were achieved after evaluating 1000 oil immersion fields (OIFs) in BC (66%) and an equal number in LN (60.9%) smears, separately or together (74%). The morulae were more often detected into lymphocytes than monocytes. The highest total number of morulae (n=143) were found in P-D smears. Finally, to avoid false positive diagnoses, platelets, lymphocytic azurophilic granules, lymphoglandular bodies and phagocytosed nuclear material should not be confused with the morulae.

Μeταξύ της ανθeκτικότητας του S. Evidence-based information of a cause-and-effect relationship between Ehrlichia canis infection and polyarthritis in naturally- or experimentally-infected dogs is currently lacking. The aim of this prospective study was to investigate whether synovial fluid cytological evidence of arthritis could be documented in dogs with acute monocytic ehrlichiosis. Direct synovial fluid cytology smears from eight Beagle dogs experimentally infected with E. canis were examined prior to, and on 21, 35 and 63 days post-inoculation. The cytological variables assessed included cellularity, percentages of mononuclear cells and neutrophils, macrophage reactivity and evidence of E. canis morulae. The median cellularity and percentages of mononuclear cells and neutrophils prior to inoculation did not differ when compared to post-inoculation cytological evaluation. Increased cellularity, E. canis morulae or cytological evidence of arthritis or macrophage reactivity were not observed throughout the course of the study. In the present study, no cytological evidence of arthritis was found in dogs with experimental acute canine monocytic ehrlichiosis, suggesting that E. canis infection should be considered a rather uncommon cause of arthritis in dogs.

We included 92 pairs of ewes with or without retention of fetal membranes in a cohort study of 25 flocks in Southern Greece. We obtained two uterine content samples under aseptic conditions, by introducing a swab into the uterus of these ewes, on the 2nd-4th and the 5th-9th day after lambing. We used conventional bacteriological techniques to isolate and identify bacteria and to carry out antimicrobial agents susceptibility testing. The prevalence of bacterial intrauterine contamination among ewes with retention was 24% on the first and 46% on the second sampling (P0.0001) and that among ewes without retention was 8 and 2% (P0.05), respectively. Clinical signs accompanying the retention of fetal membranes were more frequently observed among ewes with intrauterine contamination than among those without (P = 0.0007). The odds of an ewe having an intrauterine contamination increased multiplicatively by 1.06 when the median duration of retention in the flock increased by 6 h. The principal bacteria isolated from the ewes with retention were Arcanobacterium pyogenes and Escherichia coli; 21% of 73 isolates tested were found resistant to at least one antimicrobial agent.

One hundred and seventy-four dogs diagnosed with cutaneous neoplasms in the Animal Medical and Surgical Clinic, Faculty of Veterinary Medicine, Aristotle University of Thessaloniki, were studied. Thirty-one types of neoplasm were diagnosed, among which mast cell tumours (13.8%), hepatoid gland adenomas (9.8%), lipomas (5.7%) and histiocytomas (5.7%) were the most common. The prevalence of epithelial, mesenchymal, lymphohistiocytic and melanocytic tumours was 47.7, 40.8, 8.6 and 2.9%, respectively. Potentially malignant neoplasms were less frequently recorded than benign neoplasms. The tumours were single (80.5%) or multiple (19.5%) and located on the head and neck (18.4%), the body trunk (49.4%), the limbs (25.9%) or at multiple sites (6.3%). The factors evaluated in multivariable logistic regression models for possible association with the odds of a tumour's potential for malignancy included the age, the sex and the breed of the dog, as well as the histological type of the neoplasm. Dogs with mesenchymal tumours had two times higher odds of potential for malignancy than those with epithelial tumours. In contrast, dogs with either lymphohistiocytic or melanocytic tumours did not have increased risk of malignancy compared with dogs with epithelial tumours. The odds of tumour malignancy linearly increased with increasing age of the dog by a factor of 1.1 per year. Finally, the effect of the sex and the breed of the dog on the risk of developing cutaneous neoplasms was investigated in an age-matched case-control sample of 348 dogs by conditional logistic regression analysis. The odds of neoplasm presence were two times higher in pure bred dogs than in mongrels but did not differ between cross-breeds and mongrels.

In this study 50 clinically normal and 50 cats with cutaneous and/or systemic signs that lived in an endemic area and had been previously examined for infection by L. infantum using PCR in four different tissues were serologically tested for the presence of anti-Leishmania IgG (IFAT and ELISA) and IgM (IFAT). The aim was to compare the results of IFAT, ELISA and PCR and to investigate the possible associations between seropositivity to Leishmania spp and signalment, living conditions, season of sampling, health status of the cats, and seropositivity to other infectious agents. Low concentrations of anti-Leishmania IgG were detected by IFAT in 10% of the cats and by ELISA in 1%, whereas anti-Leishmania IgM were detected by IFAT in 1%. Otodectes cynotis is responsible for at least 50% of canker cases diagnosed in cats world-wide. The role of Demodex cati in the pathogenesis of otitis and acne is still obscure. The aims of this study were to estimate the prevalence of O. cynoyis and D. cati infestations in clinically normal cats in northern Greece, to determine the factors that are associated with the probability and severity of infestation in the cat, and to examine the importance of these mites in the pathogenesis of feline acne. Samples from 161 cats were examined by flushing the ear canals and by taking skin scrapings of the chin and lip area. The results were combined with various factors (sex, age, living style, hair coat type and presence of pruritus, of ear discharge, of acne-like lesions) in order to carry out a risk analysis. Two separate logistic regression analyses were performed. One, on the infestation/non-infestation potential with O. cynotis and the other, on the degree of such infestation as mild-to-moderate (≤5 mites/field) or severe (>5 mites/field). D. cati was not detected in any of the 161 cats. The prevalence of O. cynotis was estimated at 25.5% (95% confidence interval (CI) 19–32). The rate of mite infestation was higher with the presence of ear discharge (odds ratio 9, 95% CI 3.3–24.5), periaural pruritus (odds ratio 3.6, 95% CI 1.8–8) and acne-like lesions (odds ratio 3.3, 95% CI 1.2–9). Cats with mild-to-moderate degree of infestation had 18 times higher chance of exhibiting an ear discharge than those with a severe infestation. The log-odds of mild-to-moderate parasitism were linearly related to the age.

The objective of this field trial was to compare the odds of pregnancy at 25 days (P25) and at 60 days (P60), of embryonic death (ED) between 25 and 60 days, and of non-pregnant cows returning to heat 20-25 days after artificial insemination (AI). Three fixed-time insemination protocols involving combined administration of either double gonadotrophin-releasing hormone (GnRH) and prostaglandins (A) or GnRH, human chorionic gonadotrophin (hCG) and prostaglandins (B), or double treatment with prostaglandins (C) were used. Cows included in the trial were selected from five herds, comprising a total of 735 (herd size range 85-250, median 120) Holstein dairy cows. Animals used for the study were multiparous, 4-7 years of age, had expressed at least one overt postpartum heat, were between 60 and 120 days of lactation and had at least one palpable corpus luteum and no gross morphological anomalies of their reproductive tracts. They were randomly allocated to receive treatment A (n = 89), B (n = 93), or C (n = 65). Pregnancy of cows not observed in heat 18-24 days post AI was diagnosed by transrectal ultrasonography at 25-30 days and confirmed by rectal palpation at 60-75 days post AI. Those that were detected in heat in the normal period post AI were re-inseminated. No differences were detected among the three treatment groups concerning the odds of P25 (P = 0.5), P60 (P = 0.2) and ED (P = 0.13). In contrast, non-pregnant cows that were administered treatment A were less likely to exhibit estrous signs in the normal period post AI than those administered treatments B (P = 0.02) or C (P = 0.005).

We defined retention of fetal membranes (RFM) in dairy ewes as failure to expel the placenta within 6h after lambing the last lamb and designed a matched case-control study to identify factors that affect the risk of retention. For each ewe with RFM, the next ewe in the flock that lambed and expelled the placenta in6h after lambing the last lamb was selected as control. Data analyzed included 92 pairs of ewes from 25 flocks comprising a total of 7275 ewes (median flocksize 270 ewes). Factors investigated for associations with RFM were induction of lambing, obstetrical assistance because of dystocia, parity, the number of liveborn lambs, the occurrence of stillbirth(s), the mean weight of the litter on the third day post-lambing and the occurrence of neonatal death in the litter. Conditional logistic regression indicated (1) that the risk of RFM increased linearly with increasing number of liveborn lambs and (2) that the risk of retention was 4-fold higher in ewes that received assistance at lambing than those that lambed normally.

In a field investigation of 10 flocks in Southern Greece, 3367 dairy ewes were examined twice, in order to estimate the incidence risk and the aetiology of mammary abnormalities during the dry-period. Abnormal secretion, lumps, nodules, diffuse hardness, abscesses and cysts were the abnormalities detected. The cumulative incidence of mammary abnormalities during the dry-period was 5.1% (95% confidence interval: 4.4–5.8%); 47% of the cases detected developed during the first three weeks after cessation of lactation. Despite variation in the flock size, there was no between-flock variation in the risk of a ewe developing mammary abnormalities. Staphylococci ( Staphylococcus aureus and coagulase-negative isolates) were the most frequently isolated bacteria from mammary samples; Actinomyces pyogenes , Clostridium perfringens , streptococci and Escherichia coli were also isolated. Resistance was encountered among the staphylococcal isolates.

This study aimed to investigate the possible association between canine parvoviral enteritis and asymptomatic bacteriuria. Forty-three puppies that were admitted to the outpatient service of the Animal Medical Clinic with clinical signs compatible with parvoviral enteritis formed the exposed group. The clinical diagnosis was subsequently confirmed by a positive fecal ELISA test (CITE test: IDDEX Lab., Westbrook, ME). Twenty-three (53.5%) of these puppies were males and 20 (46.5%) were females. Their age ranged from 1.5 to 5.5 months. Forty-eight clinically normal and age-matched puppies, that had been admitted to the clinic for vaccinations and had a negative result in the aforementioned ELISA test, were randomly selected to form the unexposed group. Urine samples were collected by antebupic cystocentesis from all puppies and submitted for bacterial culture. In the parvovirus exposed group, 11 of 43 puppies had detectable bacteriuria. The isolates were Escherichia coli alone (8/11 – 72.7%) Staphylococcus aureus alone (1/11 – 9.1%) and mixed cultures of E. coli and S. epidermitis (2/11 – 18.2%). In the unexposed group there were three puppies with detectable bacteriuria, one isolate each of E. coli , Enterococcus durans and Corynebacterium spp. Puppies with parvoviral enteritis had five (95% CI: 1.3–19.8) times higher odds of developing asymptomatic bacteriuria than puppies without the disease. The observed increased risk of asymptomatic bacteriuria among puppies with parvoviral enteritis was probably due to the fecal contamination of the external genitalia and the neutropenia these puppies exhibited.

Ehrlichia canis infection causes multisystemic disease in dogs (canine monocytic ehrlichiosis, CME) which is associated with variable morbidity and mortality. Atypical clinical manifestations, including gastrointestinal signs, may occasionally occur in CME and approximately 10-15% of dogs are presented with historical or clinical evidence of vomiting, diarrhea, and/or abdominal discomfort. The objective of this study was to investigate if there are any alterations in serum canine pancreatic lipase immunoreactivity (cPLI) in dogs with experimentally induced or naturally occurring monocytic ehrlichiosis. Serum samples from 10 Beagle dogs experimentally infected with E. canis and two healthy uninfected Beagles were serially examined; samples from 20 naturally infected dogs (10 with non-myelosuppressive [NME] and 10 with myelosuppressive [ME] ehrlichiosis) were also examined at a given point in time (cross-sectional sampling). None of the experimentally infected Beagles showed gastrointestinal signs or increased cPLI concentrations prior to or following the artificial infection. Three naturally infected dogs with NME and one with ME demonstrated serum cPLI concentrations in the diagnostic range for pancreatitis (>400 μg/L) without showing gastrointestinal signs. The results of the present study indicated that 4/20 (20%) of dogs naturally infected with E. canis demonstrated increased serum cPLI concentrations consistent with mild and clinically inapparent pancreatitis.

We validated a commercial (Idexx Pourquier, Montpellier, France) serum and milk indirect ELISA that detects antibodies against Mycobacterium avium ssp. paratuberculosis (MAP) in Greek dairy goats. Each goat was sampled 4 times, starting from kidding and covering early, mid, and late lactation. A total of 1,268 paired milk (or colostrum) and serum samples were collected during the 7-mo lactation period. Bayesian latent class models, which allow for the continuous interpretation of test results, were used to derive the distribution of the serum and milk ELISA response for healthy and MAP-infected individuals at each lactation stage. Both serum and milk ELISA, in all lactation stages, had average and similar overall discriminatory ability as measured by the area under the curve (AUC). For each test, the smallest overlap between the distribution of the healthy and MAP-infected does was in late lactation. At this stage, the AUC was 0.89 (95% credible interval: 0.70; 0.98) and 0.92 (0.74; 0.99) for the milk and serum ELISA, respectively. Both tests had comparable sensitivities and specificities at the recommended cutoffs across lactation. Lowering the cutoffs led to an increase in sensitivity without serious loss in specificity. In conclusion, the milk ELISA was as accurate as the serum ELISA. Therefore, it could serve as the diagnostic tool of choice, especially during the implementation of MAP control programs that require frequent testing, because milk sampling is a noninvasive, rapid, and easy process.

Objectives To assess the efficacy of rifampicin in achieving clinical and haematological recovery and clearing infection in dogs with experimentally induced acute monocytic ehrlichiosis. Methods Five Ehrlichia canis-infected Beagle dogs were treated with rifampicin (10 mg/kg/24 h orally for 3 weeks), nine E. canis-infected dogs received no treatment (infected untreated dogs) and two dogs served as uninfected controls. Clinical score, platelet counts, immunofluorescent antibody titres and PCR detection of E. canis-specific DNA in blood, bone marrow and spleen aspirates were evaluated on post-inoculation days 21 (start of rifampicin), 42 (end of rifampicin) and 98 (end of the study). Results By day 21 post-inoculation, all infected dogs became clinically ill and thrombocytopenic, seroconverted and were PCR positive in at least one tissue. Clinical scores and antibody titres did not differ between the treated and infected untreated dogs throughout the study. The rifampicin-treated dogs experienced an earlier resolution of their thrombocytopenia (Kaplan-Meier survival plot, P=0.048), and the median platelet counts were significantly higher in the treated compared with the infected untreated dogs on post-inoculation days 42 (P=0.0233) and 98 (P=0.0195). At the end of the study, three treated and six untreated infected dogs remained PCR positive in one tissue each. Conclusions The rifampicin treatment regimen applied in this study hastened haematological recovery, but was inconsistent in eliminating the acute E. canis infection.

SUMMARYDisease cases are often clustered within herds or generally groups that share common characteristics. Sample size formulae must adjust for the within-cluster correlation of the primary sampling units. Traditionally, the intra-cluster correlation coefficient (ICC), which is an average measure of the data heterogeneity, has been used to modify formulae for individual sample size estimation. However, subgroups of animals sharing common characteristics, may exhibit excessively less or more heterogeneity. Hence, sample size estimates based on the ICC may not achieve the desired precision and power when applied to these groups. We propose the use of the variance partition coefficient (VPC), which measures the clustering of infection/disease for individuals with a common risk profile. Sample size estimates are obtained separately for those groups that exhibit markedly different heterogeneity, thus, optimizing resource allocation. A VPC-based predictive simulation method for sample size estimation to substantiate freedom from disease is presented. To illustrate the benefits of the proposed approach we give two examples with the analysis of data from a risk factor study on Mycobacterium avium subsp. paratuberculosis infection, in Danish dairy cattle and a study on critical control points for Salmonella cross-contamination of pork, in Greek slaughterhouses.

Bayesian mixture models can be used to discriminate between the distributions of continuous test responses for different infection stages. These models are particularly useful in case of chronic infections with a long latent period, like Mycobacterium avium subsp. paratuberculosis (MAP) infection, where a perfect reference test does not exist. However, their discriminatory ability diminishes with increasing overlap of the distributions and with increasing number of latent infection stages to be discriminated. We provide a method that uses partially verified data, with known infection status for some individuals, in order to minimize this loss in the discriminatory power. The distribution of the continuous antibody response against MAP has been obtained for healthy, MAP-infected and MAP-infectious cows of different age groups. The overall power of the milk-ELISA to discriminate between healthy and MAP-infected cows was extremely poor but was high between healthy and MAP-infectious. The discriminatory ability increased with increasing age. The great overlap between the distributions of the different infection stages would have hampered our ability to discriminate between the different infection stages. Thus, the proposed method, which uses partially verified data on the true status for some individuals, is an intuitive extension to the standard non-gold standard methods, especially in the case of infections with a long latent period.

The presence of important house dust and storage mite species in the microenvironment of atopic dogs has not been thoroughly investigated.To compare the presence and population of five dust mite species (Dermatophagoides farinae, Dermatophagoides pteronyssinus, Acarus siro, Tyrophagus putrescentiae and Lepidoglyphus destructor) among households with mite-sensitive atopic dogs (Group A), households with clinically healthy dogs (Group B) and households without pets (Group C, n=25) in Greece.Twenty mite-sensitive atopic dogs and 20 clinically healthy dogs.Dust samples were collected with a vacuum cleaner from owners' mattresses (all groups) and from dogs' sleeping areas (Groups A and B) or living room couch (Group C), once every season of the year. Following dust flotation, mites were counted and identified.Dermatophagoides farinae was the most prevalent (60, 40 and 64% in Groups A, B and C, respectively), followed by D. pteronyssinus (45, 35 and 48%, respectively), whereas the three storage mites were found in fewer households. No major differences could be found between Groups A and B or between households with (Groups A and B) and without dogs (Group C) regarding the presence or numbers of the five dust mite species.The presence and population of five common house dust and storage mite species does not differ among Greek households with mite-sensitive atopic dogs, households with healthy dogs and households without pets.

We conducted a longitudinal retrospective study in order to assess the effect of health disorders (HDs) on culling in the 1st or 2nd lactation cows, in Iranian dairy herds. In total, 7067 first- and second-parity Holstein cows, from 32 Iranian dairy herds, which calved from March 2007 to March 2008, were followed until the next calving or culling. Parametric survival models with time-dependent covariates were used to capture the effect of HDs with different duration and recurrence episodes on the risk of culling. Mastitis, locomotor disorders, ovarian cysts, abortion, diarrhea, rumen disorders, displaced abomasum and respiratory disorders were associated with an increased risk of culling. Hazard ratios and 95% confidence intervals were 2.78 (2.31; 3.35), 3.4 (2.79; 4.13) and 1.62 (1.15; 2.78) for mastitis, locomotor disorders and ovarian cysts as common HDs, respectively. Parity and milk-yield were identified as confounder and effect modifier, respectively. The risk of culling in the presence of uterine infection or traumatic reticulo-peritonitis increased with decreasing milk yield.

The aim of this study was to determine whether serum cardiac troponin I (cTnI) concentration measured on admission was associated with the clinical severity of canine monocytic ehrlichiosis and was predictive of clinical outcome (death or survival) in dogs naturally infected with Ehrlichia canis. Serum cTnI concentration was compared among 22 dogs with non-myelosuppressive ehrlichiosis (NME), 22 dogs with myelosuppressive ehrlichiosis (ME) and 10 healthy dogs. Unlike healthy dogs, 45.5% NME and 59.1% ME dogs had increased cTnI concentrations. There was no difference in the frequency of cTnI increase or mean cTnI concentrations between the NME and ME groups, whereas mean cTnI concentration was significantly lower in healthy dogs. No association was established between cTnI concentration on admission and clinical outcome.

Isospora suis is an important parasitic infection in intensive pig production worldwide, responsible for significant economic losses. In this study the efficacy of toltrazuril treatment against isosporosis was evaluated, under field conditions and throughout the nursing period, in reducing (i) the mean time to onset of diarrhoea and oocyst excretion, (ii) the odds of diarrhoea and, (iii) the odds and level of oocyst excretion, adjusting for the heterogeneity of I. suis infection among litters and across time. In a 300-sow farrow-to-finish commercial operation, twenty-five litters were randomly allocated to receive toltrazuril (thirteen litters) or no treatment (twelve litters). The course of infection was followed in all piglets by coprological examination from day 6 after farrowing until weaning. Parametric shared frailty models, generalised linear mixed models and a two-part random effects model were used in the analyses. Treated piglets had longer mean time to onset of oocyst excretion, lower odds of excreting oocysts and lower mean amount of excreted oocysts on any day during the nursing period. Diarrhoea was less likely to occur in treated piglets. Variance partition coefficients revealed that almost half of the variation in the odds of oocyst excretion and diarrhoea was ascribed to unknown or unmeasured factors that operate at higher than the piglet levels of aggregation. Thus, beyond toltrazuril treatment, control of isosporosis in commercial pig farms can be improved by identification and quantification of these factors.

The effect of transportation and confinement stress on Dicrocoelium dendriticum egg production was investigated. Sheep passing a minimum of 200 eggs g-1 of faeces were selected from a naturally infected flock. A group of six ewes (group A) was transferred to the laboratory premises and kept indoors for 28 days, while another group (B) of six ewes remained on pasture and was used as a control. Faecal examinations and egg counting were performed weekly, on all sheep, from one week before to 28 days after the transportation of the animals. Comparison of faecal egg counts between groups revealed higher (P < 0.01) counts in transported sheep sampled on days 7, 14 and 28 of the trial. Furthermore, egg counts obtained from sheep that were transferred remained consistently high while the ones from sheep that remained on pasture showed significant variation. Therefore, it is concluded that stress-inducing factors, such as transportation and confinement may enhance egg production of D. dendriticum.

The potential impact of preanalytical factors, such as type of anticoagulant, storage temperature, and time, on the formation of macroplatelets and platelet aggregates (platelet clumping) in dogs is largely elusive. The objective of the current study was to assess the effect of anticoagulant, temperature, and blood storage time in the light microscopy-generated macroplatelet percentages and the frequency of visually inspected platelet aggregates in clinically healthy dogs. Giemsa-stained blood smears from 70 healthy dogs were reviewed after exposure to different anticoagulants (ethylenediamine tetra-acetic acid [EDTA] vs. citrate), temperatures (25 degrees C vs. 4 degrees C), and storage times (up to 24 hr postsampling). The mean percentage of macroplatelets (platelets with diameter or lengthor =5 microm) was higher (P = 0.0006) when EDTA was used as the anticoagulant. For either anticoagulant, the mean percentage of macroplatelets was higher (P0.0001) at 25 degrees C than at 4 degrees C. Platelet clumping was 1.9 times (P0.0001) more likely to occur when citrate- rather than EDTA-anticoagulated blood was examined; regardless of the anticoagulant used, clumping occurred 3 times (P0.0001) more often when samples were preserved at 4 degrees C than when they were preserved at 25 degrees C. Storage time did not significantly influence the macroplatelet percentages or the frequency of platelet clumping. The results of this study indicate that macroplatelet percentages in the canine blood should be interpreted in relation to anticoagulant- and temperature-specific reference intervals and that future studies are warranted in order to investigate the clinical relevance of this calculation. In addition, the significant association of citrate with the formation of platelet aggregates may preclude its use for platelet enumeration in the dog.

Summary The requirements for the isolation of Mycobacterium avium subsp. paratuberculosis (Map) may be related to the strain-type [sheep (S)- or cattle (C)-type] and not to the host. The objective of this paper was to estimate and compare strain- and biological sample (faeces or pooled-tissue) – specific sensitivities (Ses) of two solid culture media, Herrold’s egg yolk medium (HEYM) and Lowenstein–Jensen (LJ) medium, for the isolation of Map from Greek dairy sheep and goats. From 400 faecal samples collected from sub-clinically infected sheep and goats of four flocks and from 214 pooled-tissue samples (142 from sheep and 72 from goats) collected, at the abattoir, from >1-year-old routinely slaughtered animals, with gross pathology suggestive of paratuberculosis, we isolated 34 Map strains. Of those, by the IS1311 PCR, 18 were categorized into the C-type and nine into the S-type; seven were not typed. We used a Bayesian approach to estimate the strain-specific Ses. SeHEYM-C-faecal = 17% (95% credible interval: 7, 40) was higher than SeHEYM-S-faecal = 2% (0.3, 11). Also, SeHEYM-C-faecal was higher than SeLJ-C-faecal = 4% (1, 12). In pooled-tissue samples, the strain-specific Ses did not differ between the two media.

Background: Canine leishmaniasis (CanL) is a common cause of epistaxis in dogs residing in endemic areas. The pathogenesis of CanL-associated epistaxis has not been fully explored because of the limited number of cases reported so far. Hypothesis: Epistaxis in CanL could be attributed to more than 1 pathomechanism such as hemostatic dysfunction, biochemical abnormalities, chronic rhinitis, and coinfections occurring in various combinations. Animals: Fifty-one dogs with natural CanL. Methods: The allocation of 51 dogs in this cross-sectional study was based on the presence (n = 24) or absence (n = 27) of epistaxis. The potential associations among epistaxis and concurrent infections (Ehrlichia canis, Bartonella spp., and Aspergillus spp.), biochemical and hemostatic abnormalities, and nasal histopathology were investigated. Results: Hypergammaglobulinemia (P= .044), increased serum viscosity (P= .038), decreased platelet aggregation response to collagen (P= .042), and nasal mucosa ulceration (P= .039) were more common in the dogs with epistaxis than in those without epistaxis. The other significant differences between the 2 groups involved total serum protein (P= .029) and γ-globulin (P= .013) concentrations, which were higher, and the percentage platelet aggregation to collagen, which was lower (P= .012) in the epistaxis dogs. Clinical Importance: CanL-associated epistaxis appears to be the result of multiple and variable pathogenetic factors such as thrombocytopathy, hyperglobulinemia-induced serum hyperviscosity, and nasal mucosa ulceration.

Objectives: To determine the prevalence and identify possible clinicopathologic indicators of the diseases associated with canine epistaxis. Methods: The medical records of 61 dogs with epistaxis were reviewed. Results: Systemic diseases, diagnosed in fifty-six dogs, included canine leishmaniasis in twenty-three dogs, canine monocytic ehrlichiosis in twenty-two, concurrent canine leishmaniasis and canine monocytic ehrlichiosis in six, rodenticide toxicity in two and primary immune-mediated thrombocytopenia, suspected oestrogen toxicity and systemic arterial hypertension in one dog each. Intranasal diseases were documented in the remaining five dogs, including transmissible venereal tumour in three dogs, and nasal adenocarcinoma and nasal aspergillosis in one dog each. Mucosal pallor and a generalised bleeding tendency were significantly more common among dogs with canine monocytic ehrlichiosis compared with those with canine leishmaniasis, whereas the opposite was true for peripheral lymphadenomegaly. Also, dogs with canine monocytic ehrlichiosis presented with pancytopenia more frequently compared with those with canine leishmaniasis; in the latter dogs, the median values of haematocrit, leucocyte and platelet counts and serum total protein concentrations were higher. Clinical Significance: Canine leishmaniasis and canine monocytic ehrlichiosis are the leading causes of canine epistaxis in Greece. Mucosal pallor, bleeding tendency and pancytopenia are more likely to be indicative of canine monocytic ehrlichiosis, as opposed to peripheral lymphadenomegaly and hyperproteinaemia in canine leishmaniasis.

SUMMARYThis study aimed to: (1) investigate whether non-ruminant wildlife interfacing with dairy sheep and goats of four Greek flocks endemically infected withMycobacterium aviumsubspeciesparatuberculosis(MAP) harboured MAP and (2) genetically compare the strains isolated from the wildlife to those isolated from the small ruminants of these flocks. We cultured and screened, by polymerase chain reaction (PCR), pooled-tissue samples from 327 wild animals of 11 species for the MAP-specific IS900insertion sequence. We also cultured faecal samples from 100 sheep or goats from each of the four flocks. MAP was detected in samples from 11 sheep, 12 goats, two mice, two rats, a hare and a fox. Only one rat had histopathological findings. Genetic typing categorized 21 isolates as cattle-type strains and two, from a house mouse and a goat respectively, as sheep-type strains; this is the first report of a rodent harbouring a sheep-type strain. The MAP types that were most frequently isolated amongst the sheep and goats of each flock were also the ones isolated from sympatric rodents; those isolated from the fox and hare also belonged to the predominant ruminant strains.

Experiments were conducted to determine the effects of lamb age, frequency of follicular aspirations, and hormone stimulation by fixed or variable FSH dose, on the number of collected oocytes and their maturational competence. In trial 1, the characteristics of follicular population (number and diameter of follicles) were studied in 40 lambs which were slaughtered at the age of 30 days (S1), 42 days (S2), 60 days (S3) and 5-6 months (S4), each n = 10. In trial 2, 27 lambs were divided into four groups. group MF lambs (n = 6) had follicular aspiration (OPU) in four monthly intervals commencing from the age of 8-9 weeks (sessions MF1, MF2, MF3 and MF4). In groups SF2, SF3 and SF4 (each n = 6), OPU was conducted once during the 12-13, 16-17 and 20-21 week of age, respectively. Ovarian stimulation was conducted with fixed FSH dose (3.52 mg/animal). In trial 3, 10 lambs (group MV) were treated as those of group MF apart from the FSH dose, which was administered according to the body weight in a dose of 0.27 mg/kg. The number and the size of follicles, the number and the quality of collected oocytes and the maturational competence of the oocytes were compared between and within groups. In trial 1, the total number and the number of small follicles were greater in groups S1 and S2 compared with those of S3 and S4 (p0.01). Similarly, the follicular population was greater in group MF1 than in group SF3 (p0.01). In sessions MF2, MF3, MV2, MV3 and MV4, more oocytes were collected in comparison with those from the respective once-aspirated age mates (groups SF2, SF3 and SF4). In total, more (p = 0.02) oocytes per donor were collected from group MV (15.2 +/- 5.5) than from group MF (9.0 +/- 3.2). An absolute maturational failure was observed in oocytes collected from groups SF2 and SF3. Maturational competence varied between 16.7% and 58.3% (p = 0.017) among sessions of group MF, but it was more uniform among sessions of group MV (range 12.5-42.9%, p0.05). Our results indicate that firstly, the number and the quality of harvested oocytes from juvenile lambs can be much improved if follicular stimulation regime is adjusted to the body weight. Secondly, in terms of follicular population and oocyte quality, 3 and 4-month-old lambs are naturally bad oocyte donors, but this characteristic can be reversed by a previous follicular ablation.

Objective To investigate the spectrum, prevalence and treatment response rate of ocular manifestations associated with natural canine monocytic ehrlichiosis (CME) due to Ehrlichia canis . Methods The medical records of 90 dogs that were admitted for primary ocular problems and reacted positive for E. canis serum antibodies were reviewed. All the dogs were treated with oral doxycycline. In 79 dogs, a combination of systemic corticosteroids and/or topical anti-inflammatory, mydriatics/cycloplegics and antibacterial agents were applied. Results The dogs ranged from 0.5 to 15 years in age; 55 were male and 35 female. A total of 19 breeds were represented. In the majority of dogs, ocular abnormalities were noticed in conjunction with other CME-induced clinical signs. In 30/90 dogs, ophthalmic signs were the sole presenting complaint. Unilateral (22/90, 24.5%) and bilateral (68/90, 75.5%) uveitis was the most common ophthalmic diagnosis and was classified as anterior, posterior or panuveitis in 58 (64.5%), 8 (8.9%) and 24 (26.6%) of the dogs, respectively. Bilateral uveitis was significantly more common than unilateral uveitis ( P < 0.0001), and anterior significantly more common than posterior uveitis ( P < 0.0001) or panuveitis ( P < 0.0001). In addition, corneal ulceration (12/90, 13.3%), necrotic scleritis (10/90, 11.1%), low tear production (8/90, 8.9%) and orbital cellulitis (3/90, 3.3%) were seen. Of the 45 (50%) dogs with a satisfactory follow-up, 25 (55.5%), 11 (24.5%), and 9 (20%) showed complete, partial and poor response to treatment, respectively. The number of dogs exhibiting complete resolution of the ocular manifestations was significantly higher than those with partial ( P < 0.0001) or poor ( P < 0.0001) response. Conclusions CME should be considered a major differential for a wide range of ocular manifestations exhibited by dogs residing in the endemic areas of the disease. Anterior bilateral uveitis appears to be the most prevalent ocular lesion and a favorable outcome to systemic and topical treatment may be expected in the majority of the affected dogs.

The aim of this study was to compare the accuracy of two methods used to clinically estimate testicular volume in the dog. Caliper and ultrasonographic measurements of testicular dimensions (length, width and height) of both testes were performed on 21 adult Beagles. Either measurement was taken in vivo over the scrotal skin and again in vitro after castration, excluding or including, when possible, the epididymis. In a sub-sample of 14 testes of 7 randomly selected dogs, the testicular volume was calculated by the formula of an ellipsoid: length (l) × width (w) × height (h) × 0.5236, and the empiric formula of Lambert: (l) × (w) × (h) × 0.71. The calculated volumes were then compared to the actual ones, which were estimated by water displacement. For each measurement method, the formula that estimated more accurately the true testicular volume was, subsequently, applied to the calculation of the testicular volume of the remaining testes. The formula of ellipsoid estimated testicular volume more accurately when in vivo and in vitro caliper measurements and in vitro ultrasound measurements were performed. The formula of Lambert estimated testicular volume more accurately when in vivo ultrasound measurements were performed. The volumes calculated from the measurements of calipers over the scrotal skin overestimated true volumes, on average, by 69 ± 27% (concordance correlation coefficient rccc = 0.49). By contrast, those calculated from the ultrasonography measurements were more accurate, overestimating the true volumes by 17 ± 24% (rccc = 0.81). Therefore, ultrasonography when the formula of Lambert is used is more accurate than caliper methods for the estimation of testicular volume in clinical practice and should be the method of choice.

Objective—To determine clinical signs and clinicopathologic abnormalities in dogs with naturally occurring clinical spirocercosis. Design—Retrospective case series. Animals—39 dogs with spirocercosis. Procedures—Medical records were reviewed, and information on signalment, residence (rural vs urban), owner complaints, physical examination findings, clinicopathologic abnormalities, radiographic and endoscopic findings, and concurrent systemic diseases was recorded. Results—Hellenic hounds and mixed-breed dogs were overrepresented, compared with a group of 117 control dogs without spirocercosis that were examined because of gastrointestinal tract disease, and mean body weight of dogs with spirocercosis was significantly higher than mean body weight of control dogs. Odynophagia (34 [87%]), regurgitation (24 [62%]), and excessive salivation (14 [36%]) were the most common clinical findings. The most common radiographic abnormalities were a mass in the caudodorsal aspect of the mediastinum (15/35 [43%]) and spondylitis of the caudal thoracic vertebrae (10 [29%]). Parasitic nodules were seen during esophagoscopy in all 39 dogs. Normocytic, normochromic, nonregenerative anemia; neutrophilic leukocytosis; hyperproteinemia; and high alkaline phosphatase activity were significantly more common in dogs with spirocercosis than in a control group of 56 healthy dogs. Concurrent systemic diseases, mainly leishmaniosis, dirofilariosis, and monocytic ehrlichiosis, were documented in 14 (36%) dogs. Conclusions and Clinical Relevance—Results suggest that clinical spirocercosis occurs more often in young-adult, large-breed dogs. Nonregenerative anemia, neutrophilic leukocytosis, hyperproteinemia, and high alkaline phosphatase activity may be useful clinicopathologic indicators of this disease.

Background: The expected number of thrombopoietic cells in normal canine bone marrow is poorly defined and there is no consensus on the most appropriate way to prepare cytologic smears to evaluate these cells nor on the optimum method for their quantification. Objective: The purpose of this study was to determine total and differential counts of thrombopoietic cells in the bone marrow of clinically healthy Beagle dogs by comparing 4 different smear types and bone marrow core biopsies. Methods: Twenty-two clinically healthy, male Beagle dogs, 10 to 12 months old, were used in the study. Following bone marrow aspiration and core biopsy from the iliac crest, Giemsa-stained smears were prepared by 4 techniques: drop-squash, particle-squash, buffy coat, and fat-layer smears. Thrombopoietic cells were counted in up to 100 low-power fields (LPF, ×10 objective) in the aspiration smears and in all possible high-power fields (HPF, ×40 objective) in H&E–stained biopsy sections. Results: Mean total thrombopoietic cell counts were 2.76 cells/LPF (drop-squash), 1.55 cells/LPF (particle-squash), 8.05 cells/LPF (buffy coat), and 3.08 cells/LPF (fat-layer). Core biopsies yielded 5.31 cells/HPF but frequently failed to provide interpretable specimens. There was a significant difference in cell counts among the 4 smear types (P < .001). Based on evaluation of buffy coat smears, thrombopoietic cells included 1.23% megakaryoblasts, 8.77% promegakaryocytes, and 90% megakaryocytes, with a mean maturation index of 0.11. Conclusion: Thrombopoietic cell counts in canine bone marrow are influenced by the smear technique. Buffy coat and fat-layer smears may be useful to obtain cellular smears in hemodiluted or small aspirate samples.

The sensitivities of the Knott's test (four 20-μl sediment aliquots), quantitative buffy coat capillary tube method (QBC tube, 111 μl of whole blood) and direct blood smear (DBS, 20 μl of whole blood) were evaluated for the detection of microfilaraemia in dogs. Undiluted whole blood samples taken from 70 Dirofilaria immitis antigen-positive dogs and 10 serially diluted microfilaraemic blood samples at concentrations of 400, 200, 100, 50, 25 and 12 microfilariae (mff) ml−1 were examined. For filarial speciation, the buffy coat of QBC tubes was mixed with one drop of methylene blue–formalin solution and examined as a direct smear. In 52/70 microfilaraemic blood samples, the number of mff ranged from 12 to 321987 ml−1 (median: 3199 ml−1). The diagnostic sensitivity of the Knott's test, QBC tube method and DBS in undiluted blood samples attained the 100%, 98% and 92.3% levels, respectively. Eighteen dogs tested amicrofilaraemic by all three methods. At concentrations of 400 mff ml−1, a 100% sensitivity was found by all three methods, while at 200 mff ml−1 the Knott's test, QBC tube and DBS were 100%, 100% and 90% sensitive, respectively. The relevant figures at 100 mff ml−1 were 100%, 100% and 80%, at 50 mff ml−1 100%, 100% and 50%, at 25 mff ml−1 100%, 100% and 10% and at 12 mff ml−1 80%, 50% and 10%. At 50 and 25 mff ml−1, the DBS was less sensitive compared to the other two methods, while at 12 mff ml−1, only to the Knott's test. A significant correlation was found between the QBC tube method and Knott's test regarding mff speciation. Therefore, the QBC method may be considered a reliable alternative to the Knott's test for both the detection and speciation of mff in the dog.

Nineteen dogs from Greece with chronic ehrlichiosis were studied. The dogs exhibited bicytopenia or pancytopenia, bone marrow hypoplasia, seroreactivity to Ehrlichia canis (E. canis) antigens, and had no history of drug or radiation exposure. Anorexia, depression, severe bleeding tendencies, hypoalbuminemia, and increased serum alanine aminotransferase activity were also hallmarks of the disease. All these animals eventually died, irrespective of the treatment applied. Some dogs were also serologically positive for Rickettsia conorii, Leishmania infantum (L. infantum), and Bartonella vinsonii subspp. berkhoffii. Polymerase chain reaction testing of bone marrow samples revealed E. canis, Anaplasma phagocytophilia, Anaplasma platys, and L. infantum in some dogs. Concurrent infections did not appear to substantially influence the clinical course and final outcome of the chronic canine ehrlichiosis.

A total of 216 local crossbred sheep from 16 scrapie-affected Greek flocks and 210 purebred sheep of the milk breeds Chios and Karagouniko from healthy flocks were analysed for scrapie-linked polymorphisms in the prion protein (PrP) gene. Of the 216 sheep in this case–control study, 96 sheep were clinical cases, 25 subclinical cases (asymptomatic at the moment of euthanasia but positive by histopathology and/or ELISA detecting proteinase-resistant PrP) and 95 healthy controls (negative by all evaluations). Polymorphisms at codons 136, 154 and 171 were determined by denaturing gradient gel electrophoresis, followed by RFLP and sequencing. Scrapie, both clinical and subclinical, was associated with the genotypes ARQ/ARQ (88 of 110 sheep of that genotype), ARQ/TRQ (9 of 13), ARQ/AHQ (15 of 38) and VRQ/VRQ (9 of 17). Histopathological lesions were more severe in the clinical cases. Genotypes ARQ/ARR (26 sheep), ARQ/ARK (seven sheep), AHQ/ARR (one sheep), ARH/ARH (one sheep) and ARR/ARH (three sheep) were detected exclusively in healthy control sheep. In the purebred survey, four genotypes were present in the Chios sheep (ARQ/ARQ, ARQ/TRQ, ARQ/AHQ and ARQ/ARR) and four in the Karagouniko sheep (ARQ/ARQ, ARQ/AHQ, ARQ/ARR and ARQ/ARH).

The aim of this study was to assess the probability of detecting Salmonella from pen faecal samples in seropositive classified finishing pig herds. The study involved 77 herds from Denmark (20), The Netherlands (20), Greece (17) and Germany (20). The serological herd status was determined by the blood-sampling of 50 finishing pigs. Bacteriological sampling was performed by 20 pen faecal samples per herd. Over-all, 47% of the blood samples had an OD% larger than 10 and 23% larger than 40. Salmonella was isolated from 135 (9.3%) pen faecal samples in 32 herds (42%). Twenty-eight of these herds (87.5%) had a within-herd seroprevalence larger than 50% at sample cut-off OD%>10. In our study, there was an increasing probability of recovering Salmonella with increasing within-herd seroprevalence. However, this was only a moderate correlation. A correlation coefficient of 0.62 was found between the proportion of culture positive- and seropositive samples in a herd at cut-off OD%>10 and of 0.58 at cut-off OD%>40. Serology is a measure of historical exposure, which may or may not correlate closely to the microbiological burden at the time of sampling. Due to the low sensitivity of culture methods, apparent ‘false-positive’ serological results may well represent real infections not detected by bacteriological testing. For screening purposes, serological testing provides an indication of exposure to Salmonella, which forms the basis for targeted sampling, intervention and logistic slaughter procedures.

Two experiments were performed to compare the severity of encephalomyocarditis virus (EMCV) infection in pigs. The pigs were challenged with the Greek myocardial strain, at different ages and with different doses. In the first experiment, nineteen susceptible pigs, 40 days old, were divided into three groups and were experimentally infected with 10(6) TCID(50), 10(4) TCID(50) or 10(2) TCID(50) of the Greek EMCV strain. In the second experiment, 10 susceptible pigs, of either 20 or 105 days, were divided into two groups according to age and were experimentally infected with 10(6) TCID(50) of the Greek EMCV strain. In addition, five piglets, each one the same age as its experimental group, were used as uninfected controls. No clinical signs were observed after infection, except a transient temperature rise in some pigs. Another important observation was the difference in mortality between groups. The survival rate of the 40-day-old pigs was inversely related to the viral dose. In these pigs, a positive association between the viral dose and the severity of macroscopical and histopathological lesions of the heart was also evident. Viral isolations from various organs of the challenged 40-day-old pigs increased with the increasing dose level. When challenged with 10(6) TCID(50) of EMCV, there was no difference in the fatality rate of the 20- and 40-day-old pigs, but none of the 105-day-old pigs died. The severity of the macroscopical and the histopathological heart lesions was inversely related to the age of the pigs. Furthermore, viral isolations from the various organs were higher in 20- and 40-day-old pigs than in the older ones. In 40-day-old pigs, neutralizing antibodies linearly increased as the dose increased. These antibodies were consistently lower in 20-day-old pigs. Viraemia, and nasal and faecal excretions were detected in all groups and lasted 1-3 days, except for the 105-day-old pigs whose symptoms lasted for an additional day.

A total of 73 clinically healthy hunting dogs, experiencing an outdoor lifestyle and originating from an area where canine leishmaniasis is endemic, were included in the study. Polymerase chain reaction (PCR) and indirect immunofluorescent antibody test (IFAT) for Leishmania spp. were done on bone marrow and serum samples, respectively, obtained from all 73 dogs, just before the beginning of the sandfly season. PCR was found positive in 46/73 (63%) whereas, IFAT only in 9/73 (12.3%) of the dogs. The prevalence and the incidence of Leishmania infection by PCR were 61.9 and 47.1%, respectively. No association was found between the breed, age, sex, length of hair coat of the dog, urban or rural life and the presence of ample vegetation and water collections in the proximity of their living quarters, and the result of PCR. These findings clearly demonstrate that most of the dogs residing areas where leishmaniasis is endemic become infected but usually remain seronegative. Serological screening of the general canine population in these areas may result in an underestimation of the true prevalence of the infection rate.

A total of 45 non-uremic dogs, with clinical signs indicating leishmaniosis, entered the study. Diagnosis was confirmed by indirect immunofluorescence assay (IFA) on serum and polymerase chain reaction (PCR) on bone marrow samples. The dogs were randomly allocated into Group A (n=37) that received allopurinol (10mg/kg B.W., per os, twice daily) for 4 consecutive months, and Group B (n=8) that were placebo-treated. Clinical signs were scored just before and at monthly intervals throughout the study period, in a blinded and independent fashion. Complete blood count, serum biochemistry profile, urinalysis, lymph node and bone marrow parasitology, IFA and enzyme-linked immunosorbent assay (ELISA) serology and bone marrow PCR were carried out at the beginning and at the end of the trial. A total of three Group A and one Group B dogs died of end stage kidney disease that developed during the trial. In Group A animals that endured the trial there was a significant improvement in the general body condition, conjunctivitis, peripheral lymphadenopathy, splenomegaly, masticatory muscle atrophy, ulcerative stomatitis, epistaxis, exfoliative dermatitis, cutaneous ulcerations, blepharitis and nasodigital hyperkeratosis. The same observation was made for anemia, lymphopenia, hyperproteinemia, hyperglobulinemia, hyperphosphatemia, increased alkaline phosphatase activity and the low albumin/globulin ratio. By contrast, no improvement of any kind was seen in Group B dogs. Lymph node and bone marrow parasite numbers were significantly decreased in Group A animals. In Group B, that occurred only in the lymph nodes. Apart from remission of clinical signs and restoration to normal of clinicopathological abnormalities, allopurinol did not eliminate Leishmania organisms, as the PCR result on bone marrow was still positive in all the dogs that finished the trial.

Blood samples were taken from 50 pigs in each of 59 farrow-to-finish production herds and from 40 pigs in each of four of five registered multiplying herds. Samples of feed and faeces were also collected from 17 of the production herds and from the four multiplying herds. The sera were tested for antibodies to Salmonella enterica by the Danish mix-ELISA, and the organisms were isolated, serotyped and sensitivity tested by standard techniques. The average within-herd seroprevalence was 3.4 per cent and at least one pig tested seropositive in 21 of the 59 herds. In the multiplying herds, only a single seroreactor was detected. Salmonellae were isolated from only five of 95 feed samples, from two of the 17 herds sampled, Salmonella tennessee in four of five samples from one herd and an untypable strain in one of five samples from another. Four infected faecal samples were detected in four herds; they harboured Salmonella typhimurium, Salmonella bredeney or Salmonella london. No salmonellae were isolated from the samples of feed and faeces taken from the multiplying herds. The S london and S typhimurium had a low sensitivity to streptomycin, kanamycin and neomycin, and the S typhimurium also had low sensitivity to amoxycillin, ticarcillin, piperacillin, amoxycillin + clavulanic acid, cefalotin and cefoperazone. The other isolates were sensitive to all the antimicrobial agents tested.

In a field investigation of 28 flocks in southern Greece, 7660 lambings were monitored. Retention of foetal membranes (defined as failure to expel the foetal membranes within 6 h of lambing the last lamb) was recorded in 92 ewes. The median within-flock incidence risk was 1.25% (range: 0-1.9%). The heterogeneity of the risk among flocks was not significant (p = 0.99); no correlation was found between the incidence risk and the flock size (r(sp) = 0.33, p = 0.09). Overall, the median duration of retention was 72 h (range: 9-288 h); it did not differ among flocks (p = 0.89) and was not correlated with flock size (r(sp) = 0.24, p = 0.27). During the initial stage of retention the membranes were fleshy and humid; the cotyledons were thick and congested. Progressively, the membranes became thin, dry and stringy; the cotyledons shrunk and were pale. Finally the membranes dropped as a mass. In ewes with retention for12 h, a variety of accompanying signs was recorded: straining (in 22% of the ewes with retention), vulval oedema and reddening (in 16%), anorexia (in 13%), recumbency (in 13%) and increased temperature (in 12%) were the most frequent ones. Overall, the median clinical score of the disorder was '2' (range: '1'-'5'); it did not differ among flocks (p = 0.98) and was not correlated with flock size (r(sp) = 0.26, p = 0.25). In 4% of the ewes with retention for4 days accompanying signs were recorded (median clinical score: '2'), whilst in 80% of the ewes with retention foror =4 days accompanying signs were recorded (median clinical score: '3'). This difference in the prevalence of clinical signs was significant (p0.0001).

Forty-eight dairy ewes and 45 lambs naturally infested with sarcoptic mange, were divided into three equal groups and treated with moxidectin 1% inj. sol. at a dose rate of 0.2 mg moxidectin kg(-1) bodyweight once or twice ten days apart. During the study, the skin lesions were scored and the number of mites recovered in facial skin scrapings from the animals were recorded. Moxidectin was found to be effective against ovine sarcoptic mange, when given twice; the percentage reduction of mites was 100% (p < 0.0001) and the skin lesions disappeared completely (p = 0.0001). In sheep treated once only, the percentage reduction of mites varied from 75% to 92%. Treated ewes produced up to 22.4% more milk than the infested ones (p = 0.0001). Treated lambs had up to 15% greater final weight and up to 23.7% more growth than infested controls (p = 0.0001). It is concluded that two injections of moxidectin 1% inj. sol. at a dose rate of 0.2 mg kg(-1) bodyweight, given ten days apart, were effective against ovine sarcoptic mange. Furthermore, it is considered that sarcoptic mange adversely affects the production of the infested animals.

Background – The presence of important house dust and storage mite species in the microenvironment of atopic dogs has not been thoroughly investigated. Objectives – To compare the presence and population of five dust mite species (Dermatophagoides farinae, Dermatophagoides pteronyssinus, Acarus siro, Tyrophagus putrescentiae and Lepidoglyphus destructor) among households with mite-sensitive atopic dogs (Group A), households with clinically healthy dogs (Group B) and households without pets (Group C, n = 25) in Greece. Animals – Twenty mite-sensitive atopic dogs and 20 clinically healthy dogs. Methods – Dust samples were collected with a vacuum cleaner from owners’ mattresses (all groups) and from dogs’ sleeping areas (Groups A and B) or living room couch (Group C), once every season of the year. Following dust flotation, mites were counted and identified. Results – Dermatophagoides farinae was the most prevalent (60, 40 and 64% in Groups A, B and C, respectively), followed by D. pteronyssinus (45, 35 and 48%, respectively), whereas the three storage mites were found in fewer households. No major differences could be found between Groups A and B or between households with (Groups A and B) and without dogs (Group C) regarding the presence or numbers of the five dust mite species. Conclusions and clinical importance – The presence and population of five common house dust and storage mite species does not differ among Greek households with mite-sensitive atopic dogs, households with healthy dogs and households without pets. Hypotheses/Objectifs – Comparer la presence et la population de cinq especes d’acariens de poussiere (Dermatophagoides farinae, D. pteronyssinus, Acarus siro, Tyrophagus putrescentiae, Lepidoglyphus destructor) parmi les habitations de chiens atopiques (groupe A), de chiens cliniquement sains (groupe B) et les habitations sans animal domestique (groupe C, n = 25) en Grece. Animaux – 20 chiens atopiques sensibilises aux acariens et 20 chiens cliniquement sains. Methodes – Des echantillons de poussiere ont ete preleves a l’aide d’un aspirateur sur les matelas des proprietaires (tous les groupes) et sur les lieux de couchage des chiens (groupes A et B) ou sur le canape du salon (groupe C), une fois a chaque saison de l’annee. Apres mise en flottaison de la poussiere, les acariens ont ete comptes et identifies. Resultats – Dermatophagoides farinaeetait le plus prevalent (60%, 40% et 64% dans les groupes A, B et C, respectivement), suivi par D. pteronyssinus (45%, 35% et 48%, respectivement), alors que les trois acariens de stockage ont ete trouves dans peu d’habitations. Aucune difference significative n’a pu etre trouvee entre les groupes A et B ou entre les habitations avec (groupes A et B) ou sans chiens (groupe C) pour la presence ou le nombre des cinq especes d’acariens de poussiere. Conclusions et importance clinique – la presence et la population de cinq acariens de poussieres communs et d’especes d’acariens de stockage ne differe pas entre les habitations grecques de chiens atopiques sensibilises aux acariens, de chiens sains ou d’habitations sans animaux de compagnie. Resumen Introduccion – la presencia importante de acaros del polvo del hogar y de almacenamiento en el microambiente de perros atopicos no ha sido investigada a fondo. Hipotesis/objetivos – comparar la presencia y el tipo de poblacion de cinco especies del acaro del polvo (Dermatophagoides farinae, D. pteronyssinus, Acarus siro, Tyrophagus putrescentiae, Lepidoglyphus destructor) entre las casas con perros atopicos sensibles a acaros (grupo A), las casas con perros clinicamente sanos (grupo B) y las casas sin animales domesticos (grupo C, n = 25), en Grecia. Animales – 20 perros atopicos sensibles a acaros y 20 clinicamente perros sanos. Metodos – las muestras del polvo fueron recogidas con un aspirador de los colchones de los propietarios (todos los grupos) y de las areas donde dormian habitualmente los perros (grupos A y B) o del sofa de la sala de estar (grupo C), una vez en cada estacion del ano. Despues de realizar la flotacion del polvo, los acaros fueron contados e identificados. Resultados – Dermatophagoides farinae fueron los acaros mas frecuentes (un 60%, un 40% y un 64% en los grupos A, B y C, respectivamente), seguido por D. pteronyssinus (un 45%, un 35% y un 48%, respectivamente), mientras que los tres acaros de almacenaje fueron encontrados en pocas casas. No se pudo encontrar ninguna diferencias importante entre los grupos A y B o entre las casas con (grupos A y B) y sin perros (grupo C) con respecto a la presencia o a los numeros de las cinco especies del acaro del polvo. Conclusiones e importancia clinica – la presencia y la poblacion de cinco acaros del polvos y de almacenaje no presento diferencias entre los hogares de perros atopicos sensibles a acaros, de perros sanos y de casas sin animales domesticos en Grecia. Hypothese /Ziele – Ein Vergleich des Vorkommens und des Bestandes von funf Hausstaubmilbenspezies (Dermatophagoides farinae, D. pteronyssinus, Acarus siro, Tyrophagus putrescentiae, Lepidoglyphus destructor) in Haushalten mit milben-empfindlichen atopischen Hunden (Gruppe A), in Haushalten mit klinisch gesunden Hunden (Gruppe B) und in Haushalten ohne Haustiere (Gruppe C, n = 25) in Griechenland war das Ziel. Tiere – 20 milben-empfindliche atopische Hunde und 20 klinisch gesunde Hunde Methoden – Staubproben wurden einmal in jeder Jahreszeit mit einem Staubsauger von den Matratzen der BesitzerInnen gesammelt (alle Gruppen) und von den Schlafplatzen der Hunde (Gruppen A und B) oder der Couch im Wohnraum (Gruppe C). Nach Staubflotation wurden die Milben gezahlt und identifiziert. Ergebnisse – Dermatophagoides farinae war die pravalenteste Milbe (60%, 40% bzw. 64% in den Gruppen A, B bzw. C), gefolgt von D. pteronyssinus (45%, 35% bzw. 48%), wahrend die drei Vorratsmilben in weniger Haushalten gefunden wurden. Es wurden keine grosen Unterschiede zwischen den Gruppen A und B oder zwischen den Haushalten mit (Gruppe A und B) und ohne Hunde (Gruppe C) im Bezug auf das Vorkommen oder die Anzahl der funf Staubmilbenspezies gefunden. Schlussfolgerung und klinische Bedeutung – das Vorkommen und der Bestand der funf haufigen Hausstaub- und Vorratsmilbenspezies unterscheiden sich nicht in griechischen Haushalten mit milben-empfindlichen atopischen Hunden, gesunden Hunden und in Haushalten ohne Haustiere.
