52. A Novel SNP in the Promoter Region of IGF1 Associated With Yunshang Black Goat Kidding Number via Promoting Transcription Activity by SP1

Abstract

IGF1, a member of the insulin-like growth factor (IGF) superfamily, is also known as the growth-promoting factor (somatomedin C). IGF1 is involved in vertebrate growth and development, immunity, cell metabolism, reproduction, and breeding. However, there are relatively few studies on the relationship between IGF1 and goat reproduction. In this study, a new transcription factor SP1 bound to the IGF1 g. 64943050T>C promoted granulosa cell (GC) proliferation. A mutation g.64943050T>C located in the promoter region of IGF1 was identified. Association analysis revealed that the kidding number in the first and second litters and the average number of first three litters of the CC genotype (2.206 ± 0.044, 2.254 ± 0.056, and 2.251 ± 0.031) were significantly higher than those in the TC genotype (1.832 ± 0.049, 1.982 ± 0.06, and 1.921 ± 0.034) and TT genotype (1.860 ± 0.090, 1.968 ± 0.117, and 1.924 ± 0.062) (p < 0.05). The kidding number in the third litter of the CC genotype (2.355 ± 0.057) was significantly higher than that in the TT genotype (2.000 ± 0.107) (p < 0.05). Then, the function of this mutation was validated by the dual-luciferase reporter assay and EMSA. The results showed that the luciferase activity of IGF1-mutant-C was significantly higher than that of IGF1-Wild-T (p < 0.05). The EMSA also showed that the binding ability of IGF1-mutant-C was higher than that of IGF1-Wild-T (p < 0.05). Subsequently, the transcription factor SP1 was predicted to bind to the mutation of IGF1 (g.64943050T>C). Overexpression of SP1 promotes the expression of IGF1 in the primary granulosa cells (GCs). The results of the CCK-8 assay and the expression of GC proliferation factors (CDK4, cyclin D1, and cyclin D2) demonstrated that SP1 promoted GC proliferation by regulating IGF1 expression. Our results suggested that the IGF1 g.64943050T>C was significantly associated with the kidding number of Yunshang black goats, and SP1 as a transcription factor of IGF1 binding to the mutation T>C regulated the expression of IGF1. Furthermore, SP1 promoted goat GC proliferation by regulating the expression of IGF1, which provides a new insight for the goat fertility trait.

66. An Immune-suppressive Triterpenoid from the Stem of Kadsura coccinea

Abstract

Kadsura coccinea has been used as a folk medicine in China, treating various diseases, including chronic gastritis and rheumatoid arthritis (RA). Chemical studies have revealed that lignans and triterpenes are the main constituents of its roots and stems. In this paper, a chemical investigation on the stem of K. coccinea was performed and a previously undescribed 3,4-secolanostane (1) was afforded. The structure of 1 was determined through comprehensive NMR and HRESI-MS data analysis. The stereo-configuration of 1 was established based on a comparison of experimental and calculated ECD spectra. Bioactivity assay carried out in immune cells indicated that 1 could show moderate effects on ConA (5 μg/mL)-induced T-cell proliferation with an IC50 value of 13.28 μM (cyclosporine A as the positive drug, IC50 = 0.13 μM) while the IC50 value was 23.12 μM in LPS (10 μg/mL)-induced B cells (mycophenolate mofetil as the positive drug, IC50 = 16.71 μM).

87. The formation of Inorganic Fouling Scales on RO Membrane in A Landfill Leachate Treatment system.

Abstract

Landfill leachate treatment with reverse osmosis (RO) process can inevitably generate membrane fouling, which increasing operation costs and declining system performance. Thus it is important to reveal the mechanism of fouling formation and propose the control approaches. In this paper, some pieces of waste RO membranes, which were collected from a Disk-Tube Reverse Osmosis (DTRO) system in a landfill site were determined. The surface aggregation state, chemical constitutions and ions species of surface inorganic contaminants on RO membrane fouling layer were characterized by Atomic Force Microscope(AFM), Scanning Electron Mictoscopy-X-ray Energy Dispersive Analysis(SEM-EDX), and Ion chromatography(IC), respectively. The results demonstrated that the fouling mass on the edge of the membrane was more larger than that on the centre, but there were no significant differences of components of the fouling substance. The main inorganic constitutions were barium and calcium carbonate and sulfate, SiO2, Al (OH)3, and Fe (OH)3 colloids. The initial ions from landfill leachate might form the precipitation of BaSO4, CaCO3 and CaSO4, MgCO3, Fe (OH)3 and FeCO3. Therefore, the precipitation from cations and anions in the landfill leachate is responsible for the inorganic fouling formation on the RO membrane. [ABSTRACT FROM AUTHOR]

88. Characteristics of organic scales on RO membrane from MSW leachate treatment systems.

Abstract

Reverse osmosis (RO) is a dominant treatment technology for leachate produced in municipal solid waste (MSW) incineration plants and landfills. However, the RO application is restricted by membrane fouling. In this study, the foulants of disc tube reverse osmosis (DTRO) membranes from a MSW incineration plant and a landfill were characterized with different analytical techniques, including Atomic Force Microscope (AFM), Scanning Electron Microscopy with Energy Dispersive X-ray Spectroscopy (SEM-EDX), Fourier Transform Infrared Spectrometer (FTIR) and Three-dimensional Fluorescence Exceitation-Emission Spectrophotometer (3D-EEM). Results indicate that the floulant mass on the edge of the fouling layer was greater than that on the center of the layer. However, there was not any significant difference of elemental composition between the two locations. The main elements of fouling substance were C and O, while the minor elements include Mg, Al, Si, Fe, Ba and Ca. The functional groups of these foulants were mainly characterized with aromatic and polysaccharide structures. The primary organic pollutants on the RO membrane were fulvic acid and proteins. Abstract

Reported is a highly chemoselective intermolecular annulation of indole‐based biaryls with bromoalkyl alkynes by using palladium/norbornene (Pd/NBE) cooperative catalysis. This reaction is realized through a sequence of Catellani‐type C−H alkylation, alkyne insertion, and indole dearomatization, by forming two C(sp2)−C(sp3) and one C(sp2)−C(sp2) bonds in a single chemical operation, thus providing a diverse range of pentacyclic molecules, containing a spiroindolenine fragment, in good yields with excellent functional‐group tolerance. Preliminary mechanistic studies reveal that C−H bond cleavage is likely involved in the rate‐determining step, and the indole dearomatization might take place through an olefin coordination/insertion and β‐hydride elimination Heck‐type pathway. Abstract

The rapid pace of technological change in the internet age has fundamentally transformed information technology (IT) outsourcing. Thus, organisations should be able to re-imagine their IT outsourcing strategies to meet the needs of increasingly turbulent business environments. Bimodal IT requires the shaping of IT services to be either agile oriented or efficient oriented, thereby bringing new demands to an organisations' adoption strategies. On the basis of the review of traditional IT outsourcing service research, this paper presents the traditional and nonlinear modes of IT outsourcing service, discusses risk-based IT outsourcing service adoption strategy, relationship and contract management strategies from the static and dynamic perspectives and provides several directions for future research.

Abstract

Based on the strong robust performance of filter for the uncertainty of the system model and noises, a new method based on filter and Kalman inverse filter for gravity anomaly distortion correction is proposed to ameliorate the performance of single inverse Kalman filter. This method deduces the system state equation according to the transfer function of actual gravimeter system firstly, then filter is used to inhibit the influence of the measurement noises, finally inverse Kalman filter is adapted and the corrected gravity anomaly signal is obtained. Simulations using actual measurement data proved that the decay introduced by measurement noises of gravity anomaly correction procedure had been effectively inhibit and the precision of system had been improved apparently.

Abstract

In this work, carbon/SiO 2 composites, using amylose and tetraethyl orthosilicate (TEOS) as raw materials, were successfully prepared by a facial hydrothermal carbonization process. The carbon/SiO 2 composites were characterized by X-ray diffraction (XRD), scanning electron microscope (SEM), Energy Dispersive Spectroscopy (EDS), transmission electron microscope (TEM), N 2 adsorption and Thermogravimetric (TG) analysis. The composites, which were made up of amorphous SiO 2 and amorphous carbon, were found to have hierarchical porous structures. The mass ratios of amylose and SiO 2 and the hydrothermal carbonization time had significant effects on the morphology of the composites, which had three shapes including monodispersed spheres, porous pieces and the nano-fibers combined with nano-spheres structures. The adsorption performance of the composites was studied using Pb 2+ as simulated contaminants from water. When the mass ratio of amylose and SiO 2 was 9/1, the hydrothermal time was 30 h and the hydrothermal temperature was 180 °C, the adsorption capacity of the composites achieved to 52 mg/g. Experimental data show that adsorption kinetics of the carbon/SiO 2 composites can be fitted well by the Elovich model, while the isothermal data can be perfectly described by the Langmuir adsorption model and Freundlich adsorption model. The maximum adsorption capacity of the carbon/SiO 2 composites is 56.18 mg g −1 . [ABSTRACT FROM AUTHOR]
