AbstractBoron (B) deficiency is detrimental to Glycine max (L.) Merr., which is one of the leading oil crops. In this study, physiological analysis of soybean seedlings under B deficiency and control after 12 h, 24 h, 72 h and 8 days was carried out, and the roots were subjected to transcriptome sequencing analysis. The results showed that under B deficiency, the plant height, SPAD and chlorophyll fluorescence value of soybean seedlings decreased significantly, indicating that B stress significantly inhibited plant growth and photosynthesis. RNA-seq revealed a total of 5126 DEGs (Differentially Expressed Genes), and nine DEGs co-existed at the four-time points, among which GLYMA_03G130600 regulates transcription factor ORG3, which is related to plant growth. GO analysis revealed a total of annotated 4018 DEGs, among which the terms plasma membrane, extracellular, enzyme activity, ion transport metabolic process and oxidoreductase activity were significantly enriched at the four time points, which might hinder the growth of soybean and accelerate senescence. KEGG analysis showed that at level 1, the DEGs were mainly enriched in photosynthesis-related, phenylpropanoid biosynthesis, nitrogen metabolism and plant hormone signal transduction. At level 2 of secondary metabolism, the DEGs were mainly enriched in energy metabolism and amino acid metabolism pathways, which indicates that B stresses mainly affect photosynthesis, hormone regulation and amino acid metabolism of soybean, thus affecting plant growth. The above results give deeper insight into the soybean response to B deficiency and lay the foundation for further studies of the molecular mechanism of soybean response to boron deficiency.

Abstract Mortality from non cancer causes in patients with gallbladder cancer (GBC) still unclear. This study evaluated the causes and risk factors of non cancer death during different follow-up periods after GBC diagnosis. Non cancer causes of death for GBC patients diagnosed between 2000 and 2017 in Surveillance, Epidemiology and End Results database were analyzed and standardized mortality rates (SMR) for each non cancer death were calculated. Predictors for non cancer death were identified through multivariate competing risk analysis. A total 11,927 GBC patients were identified for further analysis, 9393 died during follow up. The largest proportion of non cancer deaths occurred > 3 years after diagnosis (39.4%). Most common non cancer cause were cardiovascular disease (43.3%), followed by other cause of death (34.4%) and infectious diseases (8.6%). Compared with US general population, GBC patients has higher risk of death from disease of heart (SMR, 1.58; 95%CI, 1.41–1.75), septicemia (SMR,3.21; 95%CI, 2.27–4.40), diabetes mellitus (SMR,1.97; 95%CI, 1.43–2.63), alone with other causes. Non cancer causes accounted for a significant proportion of deaths during the follow-up period after GBC diagnosis. The risk of non cancer death is higher in GBC patients than in the general population. Our study provides comprehensive assessment of death from non cancer cause in GBC patients, which has important implications for health management in GBC patients.

Abstract Background Although radiotherapy has improved local control in esophageal squamous cell carcinoma (ESCC), a considerable number of patients still experience relapse due to resistance. In this study, we aimed to evaluate the effects of cetuximab on radiosensitivity in two ESCC cell lines (ECA109 and TE‐13) and to investigate their underlying mechanisms. Methods Cells were pretreated with or without cetuximab before irradiation. The MTT assay and clonogenic survival assay were performed to evaluate cell viability and radiosensitivity. Flow cytometry was performed to analyze cell cycle distribution and apoptosis. The γH2AX foci were counted to determine cellular DNA‐repairing capacity via immunofluorescence assay. The phosphorylation of key molecules involved in the epidermal growth factor receptor (EGFR) signaling pathway and DNA double‐strand break (DSB) repair were measured by western blot. Results Cetuximab alone was not sufficient to suppress cell viability, but significantly enhanced radiation‐induced inhibition of clonogenic survival in ECA109 and TE‐13. The radiation sensitivity enhancement ratio (SER) was 1.341 and 1.237 for ECA109 and TE‐13, respectively. ESCC cells treated with cetuximab were arrested at the G2/M phase in response to radiation. No significant increase in apoptotic rate was observed in irradiated cells that were treated with cetuximab. The average number of γH2AX foci increased in the combination group (cetuximab and radiation). Cetuximab suppressed phosphorylation of EGFR and downstream ERK, but had no significant effect on AKT. Conclusions These results indicate the potential for use of cetuximab as an effective radiosensitizer in ESCC. Cetuximab promotes G2/M cycle arrest and reduces DSB repair, as well as inhibiting EGFR and downstream ERK pathways in ESCC.

This paper summarized the postoperative nursing of a patient undergoing surgery for lumbar disc herniation. Based on the comprehensive nursing assessment and analysis of key nursing issues, targeted nursing interventions including pain control, dietary guidance, catheter maintenance, postoperative functional exercise and prevention of complications were carried out to improve the recovery of the patient. Abstract Major depressive disorder (MDD) is a highly heterogeneous psychiatric disorder. The pathogenesis of MDD remained unclear, and it may be associated with exposure to different stressors. Most previous studies have focused on molecular changes in a single stress-induced depression model, which limited the identification of the pathogenesis of MDD. The depressive-like behaviors were induced by four well-validated stress models in rats, including chronic unpredictable mild stress, learned helplessness stress, chronic restraint stress and social defeat stress. We applied proteomic and metabolomic to investigate molecular changes in the hippocampus of those four models and revealed 529 proteins and 98 metabolites. Ingenuity Pathways Analysis (IPA) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis identified differentially regulated canonical pathways, and then we presented a schematic model that simulates AKT and MAPK signaling pathways network and their interactions and revealed the cascade reactions. Further, the western blot confirmed that p-AKT, p-ERK12, GluA1, p-MEK1, p-MEK2, p-P38, Syn1, and TrkB, which were changed in at least one depression model. Importantly, p-AKT, p-ERK12, p-MEK1 and p-P38 were identified as common alterations in four depression models. The molecular level changes caused by different stressors may be dramatically different, and even opposite, between four depression models. However, the different molecular alterations converge on a common AKT and MAPK molecular pathway. Further studies of these pathways could contribute to a better understanding of the pathogenesis of depression, with the ultimate goal of helping to develop or select more effective treatment strategies for MDD.

Two new species from China, Hymenogaster pseudoniveus and H. zunhuaensis, are described and illustrated based on morphological and molecular evidence. Hymenogaster pseudoniveus is distinguished from other species of the genus based on the color of peridium, which is snow white to white when unexcavated but then turns yellowish to earth yellow, and the basidiospores, which are ornamented with nearly longitudinally arranged ridges up to 2 μm high. H. zunhuaensis is diagnosed by its dirty white to pale yellow peridium, yellow brown to brown gleba, and the smaller (Lm × Wm = 11.7 μm × 9.8 μm) broadly ellipsoidal to subglobose basidiospores (Q = 1.1–1.3). ITS/LSU-based phylogenetic analysis supports the erection of the two new species, each placed in distinct clades with strong statistical support, suggesting that they represent two distinct species novel to science. Based on the morphological and molecular evidence, we have published two new species of Hymenogaster. A key for Hymenogaster species from China is provided.

In this study, five new species from China, Hymenogaster latisporus, H. minisporus, H. papilliformis, H. perisporius, and H. variabilis, are described and illustrated based on morphological and molecular evidence. Hymenogaster latisporus was distinguished from other species of the genus by the subglobose, broad ellipsoidal, ovoid basidiospores (average = 13.7 μm × 11.6 μm) with sparse verrucose and ridge-like ornamentation (1–1.2 μm high); H. minisporus by the ellipsoidal to broadly ellipsoidal and small basidiospores (average = 11.7 μm × 9.5 μm); H. papilliformis was characterized by the whitish to cream-colored basidiomes, and broadly fusiform to citriform basidiospores with a pronounced apex (2–3 μm, occasionally up to 4 μm high), papillary, distinct warts and ridges, and pronounced appendix (2–3 μm long); H. perisporius by the dirty white to pale yellow basidiomes, broad ellipsoidal to ellipsoidal, and yellow-brown to dark-brown basidiospores with warts and gelatinous perisporium; H. variabilis by the peridium with significant changes in thickness (167–351 μm), and broad ellipsoidal to subglobose basidiospores ornamented with sparse warts and ridges. An ITS/LSU-based phylogenetic analysis supported the erection of the five new species. A key for Hymenogaster species from northern China is provided.

The UAV industry has witnessed an unprecedented boom in recent years. Among various kinds of UAV platforms, the vertical take-off and landing (VTOL) aircraft with fixed-wing configurations has received more and more attention due to its flexibility and long-distance flying abilities. However, due to the fact that the advance ratio of regular propeller systems during the cruise phase is significantly higher than that during the VTOL phase, a variable-pitch propeller system is proposed and designed which can be applied without additional propulsion mechanisms during both flying stages. Thus, a VTOL aircraft platform is proposed based on the propulsion system constructed of variable-pitch propellers, and appropriate control manners are precisely analyzed, especially during its VTOL phase. As a basic propulsion system, a nonlinear model for variable-pitch propellers is constructed, and an optimization-based control allocation module is developed because of its multi-solution and high-order characteristics. Finally, the objective function is designed according to the stability and energy consumption requirements. Simulation experiments demonstrate that the proposed controller is able to lower energy consumption and maintain the stability of the aircraft while tracking aggressive trajectories for large-scale VTOLs with noises at the same time.

Tilt-rotor unmanned aerial vehicles combine the advantages of multirotor and fixed-wing aircraft, offering features like rapid takeoff and landing, extended endurance, and wide flight conditions. This article provides a summary of the design, modeling, and control of a composite tilt-rotor. During modeling process, aerodynamic modeling was performed on the tilting and non-tilting parts based on the subcomponent modeling method, and CFD simulation analysis was conducted on the entire unmanned aerial vehicle to obtain its accurate aerodynamic characteristics. In the process of modeling the motor propeller, the reduction of motor thrust and torque due to forward flow and tilt angle velocity is thoroughly examined, which is usually ignored in most tilt UAV propeller models. In the controller design, this paper proposes a fusion ADRC control strategy suitable for vertical takeoff and landing of this type of tiltrotor. The control system framework is built using Simulink, and the control algorithm’s efficiency has been verified through simulation testing. Through the proposed control scheme, it is possible for the composite tiltrotor unmanned aerial vehicle to smoothly transition between multirotor and fixed-wing flight modes.

Wine grape quality is influenced by the variety and growing environment, and the quality of the grapes has a significant impact on the quality of the wine. Tannins are a crucial indicator of wine grape quality, and, therefore, rapid and non-destructive methods for detecting tannin content are necessary. This study collected spectral data of Pinot Noir and Chardonnay using a geophysical spectrometer, with a focus on the 500–1800 nm spectrum. The spectra were preprocessed using Savitzky–Golay (SG), first-order differential (1D), standard normal transform (SNV), and their respective combinations. Characteristic bands were extracted through correlation analysis (PCC). Models such as partial least squares (PLS), support vector machine (SVM), random forest (RF), and one-dimensional neural network (1DCNN) were used to model tannin content. The study found that preprocessing the raw spectra improved the models’ predictive capacity. The SVM–RF model was the most effective in predicting grape tannin content, with a test set R2 of 0.78, an RMSE of 0.31, and an RE of 10.71%. These results provide a theoretical basis for non-destructive testing of wine grape tannin content.

This paper summarized the postoperative nursing measures for a patient with cervical spondylotic myelopathy. Based on the postoperative nursing assessment, interventions such as pain control, gastrointestinal function care, airway management were carried out. Traditional Chinese Medicine nursing techniques including intradermal needling, auricular acupoint pressing, Chinese herbal medicine hot-compress, external application and fumigation were conducted under the guidance of syndrome differentiation. The nursing of integrated traditional Chinese and western medicine is effective to relieve the pain, improve the gastrointestinal function and prevent the postoperative complications. Abstract Background The sex‐specific associations between birth weight and the risk of chronic kidney disease (CKD) later in life remain controversial. This study aimed to examine the shape of the relationship between birth weight and the risk of CKD in men and women. Methods A total of 277,252 participants free of CKD at baseline from the United Kingdom Biobank (UKB) prospective cohort were included in the analysis. Cox proportional hazard regression model was used to estimate the association between birth weight and the risk of incident CKD with adjustment for potential confounders. Results During a median follow‐up of 11.9 years, we identified 13,030 (4.7%) CKD cases. Compared with normal birth weight (2.5–4.0 kg), low birth weight (LBW, 4.0 kg) was associated with an 11% lower risk of CKD (HR: 0.89 [0.83–0.95]) in men but not in women (HR: 0.96 [0.89–1.04]) (p for interaction = 0.13). Furthermore, the LBW‐CKD association was stronger in obese (HR: 1.28 [1.18–1.39]) than in nonobese participants (HR: 1.16 [1.09–1.24], p for interaction = 0.03). Conclusions We found an L‐shaped relationship between birth weight and the risk of CKD in women but a linear shape in men. The LBW‐CKD association was mitigated to some extent by maintaining healthy body weight in adulthood. Our findings support the notion that the CKD risk should be managed from a lifecycle perspective.

PURPOSEPreoperative prediction of visceral pleural invasion (VPI) is important because it enables thoracic surgeons to choose appropriate surgical plans. This study aimed to develop and validate a multivariate logistic regression model incorporating the maximum standardized uptake value (SUVmax) and valuable computed tomography (CT) signs for the non-invasive prediction of VPI status in subpleural clinical stage IA lung adenocarcinoma patients before surgery.METHODSA total of 140 patients with subpleural clinical stage IA peripheral lung adenocarcinoma were recruited and divided into a training set (n = 98) and a validation set (n = 42), according to the positron emission tomography/CT examination temporal sequence, with a 7:3 ratio. Next, VPI-positive and VPI-negative groups were formed based on the pathological results. In the training set, the clinical information, the SUVmax, the relationship between the tumor and the pleura, and the CT features were analyzed using univariate analysis. The variables with significant differences were included in the multivariate analysis to construct a prediction model. A nomogram based on multivariate analysis was developed, and its predictive performance was verified in the validation set.RESULTSThe size of the solid component, the consolidation-to-tumor ratio, the solid component pleural contact length, the SUVmax, the density type, the pleural indentation, the spiculation, and the vascular convergence sign demonstrated significant differences between VPI-positive (n = 40) and VPI-negative (n = 58) cases on univariate analysis in the training set. A multivariate logistic regression model incorporated the SUVmax [odds ratio (OR): 1.753, P = 0.002], the solid component pleural contact length (OR: 1.101, P = 0.034), the pleural indentation (OR: 5.075, P = 0.041), and the vascular convergence sign (OR: 13.324, P = 0.025) as the best combination of predictors, which were all independent risk factors for VPI in the training group. The nomogram indicated promising discrimination, with an area under the curve value of 0.892 [95% confidence interval (CI), 0.813–0.946] in the training set and 0.885 (95% CI, 0.748–0.962) in the validation set. The calibration curve demonstrated that its predicted probabilities were in acceptable agreement with the actual probability. The decision curve analysis illustrated that the current nomogram would add more net benefit.CONCLUSIONThe nomogram integrating the SUVmax and the CT features could non-invasively predict VPI status before surgery in subpleural clinical stage IA lung adenocarcinoma patients.

Abstract Background Since renal cell carcinoma(RCC) is insensitive to conventional chemoradiotherapy, molecularly targeted drugs are commonly used treatments for unresectable advanced RCC. The aim of this study was to explore the efficacy and safety of TACE + sunitinib vs. sunitinib in the treatment of unresectable advanced RCC. Methods This study included 98 patients with unresectable advanced RCC who were treated in Union Hospital from January 2015 to December 2018, and they met the criteria. They were divided into two groups: TACE + Sunitinib group (N = 47) and Sunitinib group (N = 51). We conducted a retrospective study to analyze the efficacy and safety of the two groups of patients. Results (1)TACE + Sunitinib group: 4 patients (8.5%) achieved CR, 27 patients (57.5%) achieved PR, 9 patients (19.1%) achieved SD, and 7 patients (14.9%) achieved PD. Objective: Although radiation workers are exposed to much lower doses of neutron-γ rays than those suffered in nuclear explosions and accidents, it does not mean that their health is not affected by radiation. Lower doses of radiation do not always cause morphological aberrations in chromosomes, so more sophisticated tests must be sought to specific alterations in the exposed cells. Our goal was to characterize the specific gene expression in lymphocytes from logging workers who were continuously exposed to low doses of neutron-γ radiation. We hypothesized that the combination of cell type-specific transcriptomes and open chromatin profiles would identify lymphocyte-specific gene alterations induced by long-term radiation with low-dose neutron-γ-rays and discover new regulatory pathways and transcriptional regulatory elements.Methods: Lymphocytes were extracted from workers who have been occupationally exposed to neutron-γ and workers unexposed to radiation in the same company. mRNA-seq and ATAC-seq (Assay for Transposase-Accessible Chromatin with high-throughput sequencing) were performed, followed integrative analysis to identify specific gene regulatory regions induced by neutron-γ radiation. A qPCR assay was then performed to verify the downregulation of RNA coding for ribosomal proteins and flow cytometry was used to detect ribosomal protein expression and cell cycle alterations.Results: We identified transcripts that were specifically induced by neutron-γ radiation and discovered differential open chromatin regions that correlated with these gene activation patterns. Notably, we observed a downward trend in the expression of both differentially expressed genes and open chromatin peaks. Our most significant finding was that the differential peak upregulated in ATAC-seq, while the differential gene was downregulated in the ribosome pathway. We confirmed that neutron-γ radiation leads to transcriptional inhibition by analyzing the most enriched promoters, examining RPS18 and RPS27A expression by qPCR, and analyzing protein-protein interactions of the differential genes. Ribosomal protein expression and cell cycle were also affected by neutron-γ as detected by flow cytometry.Conclusion: We have comprehensively analyzed the genetic landscape of human lymphocytes based on chromatin accessibility and transcript levels, enabling the identification of novel neutron-γ induced signature genes not previously known. By comparing fine-mapping of open chromatin and RNA reads, we have determined that neutron-γ specifically leads to downregulation of genes in the ribosome pathway, with pseudogenes potentially playing a crucial role.

Conventional water purification technologies struggle to simultaneously address purification efficiency and energy consumption. Molecular orbital level surface micro-electric field (MEF)-driven water purification is an original and innovative concept conceived and developed by our group in recent years. The core idea involves creating electron-rich and electron-poor micro-areas on the nanomaterial surface, which drive pollutants or H2O molecules to provide electrons in the electron-poor micro-areas while other environmental factors (such as H2O2 and O2) obtain electrons in the electron-rich micro-areas. This process effectively utilizes the internal energy contained within wastewater and emerging contaminants (ECs). Centered on this core, this review systematically examines the discovery, construction, and characteristics of MEF and MEF-like systems and summarizes their application directions. The challenges, bottlenecks, and future development directions of MEF technology are also analyzed and discussed. Reviews of MEFs can facilitate the development of low-consumption, high-efficiency water purification technologies.

Background and purposeThe pathogenesis of idiopathic sudden sensorineural hearing loss (ISSNHL) is still unclear, and there is no targeted treatment. This research aimed to verify the role of oxidative stress in ISSNHL and explore whether melatonin has a protective effect on hearing.Materials and methodsA total of 43 patients with ISSNHL and 15 healthy controls were recruited to detect the level of melatonin, reactive oxygen species (ROS), and total antioxidant capacity (TAC) in the blood and compared before and after treatment. Multivariate logistic regression models were performed to assess the factors relevant to the occurrence and improvement of ISSNHL.ResultsThe patients with ISSNHL showed significantly higher ROS levels than controls (4.42 ± 4.40 vs. 2.30 ± 0.59; p = 0.031). The levels of basal melatonin were higher (1400.83 ± 784.89 vs. 1095.97 ± 689.08; p = 0.046) and ROS levels were lower (3.05 ± 1.81 vs. 5.62 ± 5.56; p = 0.042) in the effective group as compared with the ineffective group. Logistic regression analysis showed that melatonin (OR = 0.999, 95% CI 0.997–1.000, p = 0.049), ROS (OR = 1.154, 95% CI 1.025–2.236, p = 0.037), and vertigo (OR = 3.011, 95% CI 1.339–26.983, p = 0.019) were independent factors associated with hearing improvement. Besides, the level of melatonin (OR = 0.999, 95% CI 0.998–1.000, p = 0.023) and ROS (OR = 3.248, 95% CI 1.109–9.516, p = 0.032) were associated with the occurrence of ISSNHL.ConclusionOur findings may suggest oxidative stress involvement in ISSNHL etiopathogenesis. The level of melatonin and ROS, and vertigo appear to be predictive of the effectiveness of hearing improvement following ISSNHL treatment.

Aiming at the existence of multiple complex vibration transmission paths in the multistage transmission gearbox, a mathematical model of the multistage transmission vibration transmission path of the gearbox is established, and the frequency spectrum structure of the vibration model is deduced by the Fourier series analysis method. Vibration source, time-varying transmission path and multistage transmission gear vibration frequency characteristics are considered comprehensively, an equivalent time-varying transmission path function from the meshing point to the accelerometer mounted on the ring tooth is proposed and the time-varying transmission path is modeled as having different periodic function of decay amplitude. Taking the multistage transmission gearbox as an example, the validity of the proposed mathematical model is verified by simulation and experiment. The model can provide reference for the condition monitoring and fault diagnosis of the multistage gearbox.

Abstract Background Acute coronary syndrome (ACS) and diabetes mellitus (DM) are the leading health risks for the elderly. Triglyceride-glucose (TyG) index is a novel and reliable indicator of insulin resistance (IR). This study aims to explore the relationship between the TyG index and all-cause mortality in oldest-old patients with ACS and DM. Methods Seven hundred twenty hospitalized patients with ACS aged ≥ 80 years were enrolled, and 699 patients signed informed consent for the study. During the follow-up period, 37 were lost to follow-up, and the follow-up rate was 94.7%. 231 ACS patients with DM were selected for the study’s analyses. Kaplan–Meier curve, Cox regression model and receiver operating characteristic (ROC) curve were used to analyze the association between the TyG index and all-cause mortality. Results The mean age of participants was 81.58 ± 1.93 years, and 32.47% were women. Compared to TyG tertile 1, the Hazard Ratio (HR) [95% confidence interval (CI)] of all-cause mortality was 2.04 (1.09, 3.81) for TyG tertile 3 in the fully adjusted model. For the TyG index per standard deviation (SD) increment, the HR (95% CI) of all-cause mortality was 1.44 (1.13, 1.83). Further, the association between the TyG index and all-cause mortality was dose–response (P for trend = 0.026). ROC curve analyses indicated that the TyG index outperformed FBG and TG in the prediction of mortality risk and improved the prognostic value of the Gensini score combined with LVEF. Conclusion The TyG index predicts the risk of all-cause mortality in the oldest-old ACS patients with DM.

Lithocholic acid (LCA) is a classical secondary bile acid formed by the metabolism of gut microbiota. The TGR5 receptor (also known as G protein-coupled receptor 1, GPBAR1) is an important bile acid membrane receptor that mediates a variety of metabolic processes in vivo. In recent years, most studies have focused on the role of bile acid receptors in the intestine and liver. However, there are few reports on its effect on skeletal muscle regeneration, and the specific mechanism remains unclear. Therefore, it is necessary to investigate the mechanism of the TGR5 receptor in the regulation of skeletal muscle regeneration. The results demonstrate that muscle injection with LCA significantly reduces the necrosis rate of injured muscle and improves muscle injury. Moreover, treatment of C2C12 cells with LCA significantly increases AKT/mTOR/FoxO3 phosphorylation through the TGR5 receptor, enhances MyoG transcription and reduces FBXO32 transcription. These findings indicate that LCA can activate the TGR5/AKT signaling pathway, inhibit protein degradation and promote protein synthesis to enhance the myogenic process and promote skeletal muscle regeneration.

When realizing rolling bearing fault identification through deep learning, there is a low recognition rate and convergence rate due to ambient noise. Aiming at the above problem, a fault identification model based on enhanced empirical wavelet transform (EEWT) and enhanced dictionary learning (EDL) is proposed. Firstly, the vibration signals of rolling bearing are transformed by envelope spectrum, and envelope spectrum adaptive segmentation is implemented through the relationship between the envelope point and the adaptive threshold, and signals are decomposed into several amplitude modulation-frequency modulation (AM-FM) components. Secondly, a new component screening index is proposed, and then the appropriate AM-FM components are reconstructed to effectively reduce the noise of signals. Finally, the sparsity constraint is used to learn the typical structural characteristics in the bearing fault sample layer by layer, and the deep fault dictionary (DFD) is constructed. Then the fault samples are fed into the DFD to determine the fault category according to the reconstruction error of the samples. The test results show that the proposed method is robust to noise and has better fault recognition ability than other models. And the proposed method utilizes the sparse constraint driving dictionary to automatically extract the fault features in the vibration signal samples, while the EDL structure makes the extracted fault features have better hierarchical and physical meaning, which is in line with people's intuitive understanding of the fault and can be used in the rolling bearing fault identification engineering.

The German city of Bamberg offers lessons in how continuity and change interact within the context of the inner-urban land use of commercial horticulture, thereby informing sustainable urban transformations in historic cities. The case of Bamberg shows that urban food production is not just well-established, but a consistent and centuries-old cultural structure that influences the fabric of today’s city. In this article, we discuss what forms of urban horticulture (and thus also food production) are evident from Bamberg’s past and which may prevail in the future. Two questions structure our analysis. First, how are historical sites and spatial structures of horticulture shaped in the tension between continuity and change? Second, which practices/forms of urban horticulture are taken up and how are they updated by which actors? Both the heritage and contemporary practices of urban horticulture, it is argued, can be conceived of as a resource to create sustainable places and ways of life for citizens. Two new contributions result from this work. First, the article highlights the ongoing cultural heritage dimensions of urban horticulture in a field still dominated by eco-technical contributions associated with post-industrial innovation in urban planning; in this respect, heritage should be recognised as a dynamic that shapes urban change. In addition, secondly, the application of Luhmannian concepts of evolution in social systems reinforces the interdependence of continuity and change in urban settings.

Abstract Objective Atherosclerosis (AS) is the major cause of cardiovascular disease, and dyslipidemia is a principal determinant of the initiation and progression of AS. Numerous works have analyzed the lipid signature of blood, but scarce information on the lipidome of vascular tissue is available. This study investigated the lipid profile in the aorta of ApoE −/− mice. Method ApoE −/− mice were randomly divided into two groups: (1) the normal diet (ND) group and (2) the high-fat diet (HFD) group. After feeding for 8 weeks, the plasma low-density lipoprotein (LDL), total cholesterol (TC), and triglyceride (TGs) levels were measured. UHPLC-Q Exactive plus MS was used to assess the lipid profile using both positive and negative ionization modes. Results LDL and TC levels were significantly increased in HFD mice, and lipid deposition, plaque area and collagen fiber levels were increased in HFD group. In addition, a total of 131 differential lipids were characterized, including 57 lipids with levels that were increased in the HFD group and 74 with levels that were decreased. Further analysis revealed that the levels of several differentially expressed phosphocholines (PCs) and lysophosphocholines (LPCs) were significantly increased. These PCs included PC (38:3), PC (36:4), PC (36:3), PC (36:2), PC (36:1), PC (34:1e), PC (34:1), PC (32:1), PC (18:0/18:1), and PC (38:5), and the LPCs included LPC (18:1), LPC (18:0) and LPC (16:0). Conclusion Our findings indicate the presence of a comprehensive lipid profile in the vascular tissue of atherosclerotic mice, particularly involving PC and LPC, which exhibited significantly increased levels in AS.

Abstract The TBC1 domain family member 10B (EPI64B/TBC1D10B), a member of the RabGAP EPI64 subfamily, contains a TBC domain that confers GTPase-activating protein activity. Even though overexpression of TBC1D10B has been reported to promote tumor invasion and metastasis in gastric adenocarcinoma, the prognostic value of TBC1D10B and its correlation with DNA methylation and immune infiltration in hepatocellular carcinoma are still not known. Transcriptional expression profiles of TBC1D10B between hepatocellular carcinoma tissues and normal tissues were downloaded from The Cancer Genome Atlas and Gene Expression Omnibus. The Clinical Proteomic Tumor Analysis Consortium and the Human Protein Atlas were used to assess the TBC1D10B protein expression. The biological functions of TBC1D10B were evaluated by the Metascape database and by Gene Set Enrichment Analysis (GSEA). Receiver operating characteristic (ROC) curve analysis was used to distinguish hepatocellular carcinoma from adjacent normal tissues. The effect of TBC1D10B on survival was estimated using the Kaplan–Meier method. DNA methylation in the TBC1D10B gene was assessed using the online MEXPRESS and MethSurv tools. The association between TBC1D10B mRNA expression and immune cell infiltration was investigated by the TIMER2 web server, tumor immune estimation resource and single-sample GSEA. This study found that TBC1D10B is highly expressed in hepatocellular carcinoma and that increased TBC1D10B mRNA expression is associated with female sex, lower Body Mass Index, high level of alpha fetal protein, and worse clinical stages. The mRNA and protein levels of TBC1D10B were verified in cells. Functional annotation indicated enrichment with negative regulation of the cell cycle, extracellular matrix, and corresponding pathways in the high-TBC1D10B phenotype. The ROC curve analysis showed that, with a cutoff level of 2.912, the accuracy, sensitive, and specificity in differentiate TBC1D10B hepatocellular carcinoma from adjacent controls were 0.931, 0.920, and 0.802, respectively. Kaplan–Meier survival analysis showed that hepatocellular carcinoma patients with high TBC1D10B had a worse prognosis than those with low TBC1D10B, especially in patients with a weight below 70 kg, height above 170 cm, and histological G2 and G3. We also found that the methylation of TBC1D10B was associated with the prognosis in patients with hepatocellular carcinoma. Moreover, correlation analysis indicated that TBC1D10B mRNA expression was positively correlated with infiltration levels of most immune cells, but negatively correlated with Th17 and cytotoxic cells infiltration. Our study indicates that increased TBC1D10B expression in hepatocellular carcinoma may play a role in tumorigenesis by regulating the cell cycle and extracellular matrix. TBC1D10B may be a novel prognostic and predictive marker and immune therapeutic target in hepatocellular carcinoma patients.

Abstract Background HL-085 is a selective, orally administered MEK1/2 inhibitor. We aimed to evaluate the safety and efficacy of HL-085 in patients with advanced melanoma harboring NRAS mutations. Methods This was a multicenter phase 1 study. HL-085 was administered twice daily in a standard 3 + 3 dose-escalation design (10 dose cohorts; 0.5–18 mg twice daily), followed by dose expansion at the recommended phase II dose (RP2D). The primary endpoints included tolerability, dose-limiting toxicity (DLT), maximum tolerated dose (MTD) and RP2D. Results Between September 13, 2017, and January 18, 2021, 42 patients were enrolled (dose escalation phase: n = 30; dose expansion phase: n = 12). No DLT was reported during dose escalation and MTD was not reached with HL-085 doses up to 18 mg twice daily. The RP2D was 12 mg twice daily. The most common all-grade drug-related adverse events (AEs) across all dose levels were rash (61.9%), increased creatine phosphokinase (CK, 59.5%), face edema (50.0%), increased aspartate aminotransferase (47.6%), peripheral edema (40.5%), diarrhea (33.3%), alanine aminotransferase (33.3%), and paronychia (19.0%), most of which were grade 1 and 2. Most frequency of grade ≥ 3 AEs were CK (14.2%), asthenia (7.1%), peripheral edema (4.8%), and acneiform dermatitis (4.8%). In the cohort of 12 mg twice daily dose (15 patients), confirmed objective response rate was 26.7%; disease control rate was 86.7%; median duration of response was 2.9 months; median progression-free survival was 3.6 months. Conclusions The HL-085 showed acceptable tolerability and substantial clinical activity in patients with advanced melanoma harboring NRAS mutations. As an environmentally friendly, green, and low-carbon construction material, cemented sand and gravel (CSG) dam is widely used in civil engineering and water conservation projects in China. This study describes the progress of research on CSG dams (materials) in recent years, including the progress of engineering related to cemented gravel dams, the static, dynamic, and thermodynamic properties and intrinsic structure models of dam building materials, the static, dynamic, and temperature field analysis of dam structures, and the design of dam profiles; and on this basis, it proposes the direction of further research and development of cemented gravel structures, which will provide research for new cemented gravel structures in China. Structure design, old and diseased engineering structure removal and reinforcement, and safety assessment to provide research status and new ideas.

Oil-based drilling cuttings (OBDCs) contain petroleum hydrocarbons with complex compositions and high concentrations, which have highly carcinogenic, teratogenic, and mutagenic properties. In this study, three highly efficient petroleum hydrocarbon-degrading bacteria were screened from OBDCs of different shale gas wells in Chongqing, China, and identified as Rhodococcus sp. and Dietzia sp. Because of their ability to degrade hydrocarbons of various chain lengths, a new method was proposed for degrading petroleum hydrocarbons in shale gas OBDCs by combining different bacterial species. Results showed that the bacterial consortium, consisting of the three strains, exhibited the highest degradation rate for petroleum hydrocarbons, capable of degrading 74.38% of long-chain alkanes and 93.57% of short-chain alkanes, respectively. Moreover, the petroleum hydrocarbon degradation performance of the bacterial consortium in actual OBDCs could reach 90.60% in the optimal conditions, and the degradation kinetic process followed a first-order kinetic model. This study provides a certain technical reserve for the bioremediation of shale gas OBDCs.

Vaccination-route-dependent adjuvanticity was identified as being associated with the specific features of antigen-carrying nanoparticles (NPs) in the present work. Here, we demonstrated that the mechanical properties and the decomposability of NP adjuvants play key roles in determining the antigen accessibility and thus the overall vaccine efficacy in the immune system when different vaccination routes were employed. We showed that soft nano-vaccines were associated with more efficient antigen uptake when administering subcutaneous (S.C.) vaccination, while the slow decomposition of hard nano-vaccines promoted antigen uptake when intravenous (I.V.) vaccination was employed. In comparison to the clinically used aluminum (Alum) adjuvant, the NP adjuvants were found to stimulate both humoral and cellular immune responses efficiently, irrespective of the vaccination route. For vaccination via S.C. and I.V. alike, the NP-based vaccines show excellent protection for mice from Staphylococcus aureus (S. aureus) infection, and their survival rates are 100% after lethal challenge, being much superior to the clinically used Alum adjuvant.

Nanoparticles have been identified in numerous studies as effective antigen delivery systems that enhance immune responses. However, it remains unclear whether this enhancement is a result of increased antigen uptake when carried by nanoparticles or the adjuvanticity of the nanoparticle carriers. Consequently, it is important to quantify antigen uptake by dendritic cells in a manner that is free from artifacts in order to analyze the immune response when antigens are carried by nanoparticles. In this study, we demonstrated several scenarios (antigens on nanoparticles or inside cells) that are likely to contribute to the generation of artifacts in conventional fluorescence-based quantification. Furthermore, we developed the necessary assay for accurate uptake quantification. PLGA NPs were selected as the model carrier system to deliver EsxB protein (a Staphylococcus aureus antigen) in order to testify to the feasibility of the established method. The results showed that for the same antigen uptake amount, the antigen delivered by PLGA nanoparticles could elicit 3.6 times IL-2 secretion (representative of cellular immune response activation) and 1.5 times IL-12 secretion (representative of DC maturation level) compared with pure antigen feeding. The findings above give direct evidence of the extra adjuvanticity of PLGA nanoparticles, except for their delivery functions. The developed methodology allows for the evaluation of immune cell responses on an antigen uptake basis, thus providing a better understanding of the origin of the adjuvanticity of nanoparticle carriers. Ultimately, this research provides general guidelines for the formulation of nano-vaccines.

ObjectiveTo develop and validate the model for predicting benign and malignant ground-glass nodules (GGNs) based on the whole-lung baseline CT features deriving from deep learning and radiomics.MethodsThis retrospective study included 385 GGNs from 3 hospitals, confirmed by pathology. We used 239 GGNs from Hospital 1 as the training and internal validation set; 115 and 31 GGNs from Hospital 2 and Hospital 3 as the external test sets 1 and 2, respectively. An additional 32 stable GGNs from Hospital 3 with more than five years of follow-up were used as the external test set 3. We evaluated clinical and morphological features of GGNs at baseline chest CT and extracted the whole-lung radiomics features simultaneously. Besides, baseline whole-lung CT image features are further assisted and extracted using the convolutional neural network. We used the back-propagation neural network to construct five prediction models based on different collocations of the features used for training. The area under the receiver operator characteristic curve (AUC) was used to compare the prediction performance among the five models. The Delong test was used to compare the differences in AUC between models pairwise.ResultsThe model integrated clinical-morphological features, whole-lung radiomic features, and whole-lung image features (CMRI) performed best among the five models, and achieved the highest AUC in the internal validation set, external test set 1, and external test set 2, which were 0.886 (95% CI: 0.841-0.921), 0.830 (95%CI: 0.749-0.893) and 0.879 (95%CI: 0.712-0.968), respectively. In the above three sets, the differences in AUC between the CMRI model and other models were significant (all P < 0.05). Moreover, the accuracy of the CMRI model in the external test set 3 was 96.88%.ConclusionThe baseline whole-lung CT features were feasible to predict the benign and malignant of GGNs, which is helpful for more refined management of GGNs.

ABSTRACT Species of Discinaceae are common macrofungi with a worldwide distribution. Some of them are commercially consumed, while a few others are reported as poisonous. Two genera were accepted in the family: the epigeous Gyromitra with discoid, cerebriform to saddle-shaped ascomata and the hypogeous Hydnotrya with globose or tuberous ascomata. However, due to discrepancies in their ecological behaviors, a comprehensive investigation of their relationship was not thoroughly explored. In this study, phylogenies of Discinaceae were reconstructed using sequence analyses of combined and separate three gene partitions (internal transcribed spacer [ITS], large subunit ribosomal DNA [LSU], and translation elongation factor [TEF]) with a matrix containing 116 samples. As a result, the taxonomy of the family was renewed. Eight genera were recognized: two of them (Gyromitra and Hydnotrya) were retained, three (Discina, Paradiscina, and Pseudorhizina) were revived, and three (Paragyromitra, Pseudodiscina, and Pseudoverpa) were newly established. Nine new combinations were made in four genera. Two new species in Paragyromitra and Pseudodiscina and an un-named taxon of Discina were described and illustrated in detail based on the materials collected from China. Furthermore, a key to the genera of the family was also provided. IMPORTANCE Taxonomy of the fungal family Discinaceae (Pezizales, Ascomycota) was significantly renewed on the basis of sequence analyses of internal transcribed spacer (ITS), large subunit ribosomal DNA (LSU), and translation elongation factor (TEF). Eight genera were accepted, including three new genera; two new species were described; and nine new combinations were made. A key to the accepted genera of the family is provided. The aim of this study is to deepen the understanding of the phylogenetic relationships among genera of the group, as well as the associated generic concepts.

ObjectiveThis study aims to investigate the correlation between sleep duration and all-cause and cardiovascular mortality in the general population.MethodsA total of 26,977 participants aged ≥18 years were included in the analysis from the National Health and Nutrition Examination Survey (NHANES) database covering the period from 2005 to 2014. Data on cardiovascular and all-cause deaths were collected until December 2019. Sleep duration was assessed using a structured questionnaire, and participants were categorized into five groups based on their reported sleep duration (≤5, 6, 7, 8, or ≥9 h). Kaplan-Meier survival curves were employed to examine the mortality rates across different sleep duration groups. Multivariate Cox regression models were utilized to explore the association between sleep duration and mortality. Additionally, a restricted cubic spline regression model was employed to identify the non-linear relationship between sleep duration and all-cause and cardiovascular mortality.ResultsThe average age of participants was 46.23 ± 18.48 years, with 49.9% of the subjects being male. Over a median follow-up period of 9.42 years, 3,153 (11.7%) participants died from all-cause mortality, among which 819 (3.0%) were attributed to cardiovascular causes. The groups with sleep durations of ≥9 and ≤5 h exhibited the lowest cumulative survival rates for all-cause mortality and cardiovascular mortality, respectively. When using a sleep duration of 7 h as the reference, the hazard ratios (with 95% confidence intervals) for all-cause mortality were 1.28 (1.14–1.44) for ≤5 h, 1.10 (0.98–1.23) for 6 h, 1.21 (1.10–1.34) for 8 h, and 1.53 (1.35–1.73) for ≥9 h. The hazard ratios (with 95% confidence intervals) for cardiovascular mortality were 1.32 (1.04–1.67) for ≤5 h, 1.22 (0.97–1.53) for 6 h, 1.29 (1.05–1.59) for 8 h, and 1.74 (1.37–2.21) for ≥9 h. A U-shaped non-linear relationship between sleep duration and all-cause and cardiovascular mortality was observed, with inflection point thresholds at 7.32 and 7.04 h, respectively.ConclusionThe findings suggest that the risk of all-cause and cardiovascular mortality is minimized when sleep duration is approximately 7 h.

Objectives: Long-term use of evidence-based antiplatelet therapy is recommended for management of stable coronary artery disease (SCAD). However, non-adherence to antiplatelet drugs is common in older patients. This study aimed to evaluate the incidence and impact of antiplatelet therapy cessation on clinical outcomes of older patients with SCAD.Methods: A total of 351 consecutive eligible very older patients (≥80 years) with SCAD from the PLA General Hospital were included. Baseline demographics, clinical characteristics, and clinical outcomes were collected during follow-up. Patients were divided into cessation group and standard group based on whether discontinuing of antiplatelet drugs. The primary outcome was major adverse cardiovascular events (MACE) and secondary outcomes were minor bleeding and all-cause mortality.Results: A total of 351 participants, with a mean age of 91.76 ± 5.01 years old (range 80–106 years) were included in statistical analysis. The antiplatelet drug cessation rate was 60.1%. There were 211 patients in cessation group and 140 patients in standard group. During a median follow-up of 98.6 months, the primary outcome of MACE occurred in 155 patients (73.5%) in the cessation group and 84 patients (60.0%) in the standard group (HR = 1.476, 95% CI:1.124-1.938, p = 0.005). Cessation of antiplatelet drugs increased the rates of angina (HR = 1.724, 95% CI:1.211-2.453, p = 0.002) and non-fatal MI (HR = 1.569, 95% CI:1.093-2.251, p = 0.014). The secondary outcomes of minor bleeding and all-cause mortality were similar between the two groups.Conclusion: Among very older patients with SCAD, antiplatelet therapy cessation significantly increased the risk of MACE, and continuous antiplatelet drug therapy didn’t increase the risk of minor bleeding.

Purpose: It is vital to develop noninvasive approaches with high accuracy to discriminate the preserved ratio impaired spirometry (PRISm) group from the chronic obstructive pulmonary disease (COPD) groups. Radiomics has emerged as an image analysis technique. This study aims to develop and confirm the new radiomics-based noninvasive approach to discriminate these two groups. Methods: Totally 1066 subjects from 4 centers were included in this retrospective research, and classified into training, internal validation or external validation sets. The chest computed tomography (CT) images were segmented by the fully automated deep learning segmentation algorithm (Unet231) for radiomics feature extraction. We established the radiomics signature (Rad-score) using the least absolute shrinkage and selection operator algorithm, then conducted ten-fold cross-validation using the training set. Last, we constructed a radiomics signature by incorporating independent risk factors using the multivariate logistic regression model. Model performance was evaluated by receiver operating characteristic (ROC) curve, calibration curve, and decision curve analyses (DCA). Results: The Rad-score, including 15 radiomic features in whole-lung region, which was suitable for diffuse lung diseases, was demonstrated to be effective for discriminating between PRISm and COPD. Its diagnostic accuracy was improved through integrating Rad-score with a clinical model, and the area under the ROC (AUC) were 0.82(95 %CI 0.79–0.86), 0.77(95 %CI 0.72–0.83) and 0.841(95 %CI 0.78–0.91) for training, internal validation and external validation sets, respectively. As revealed by analysis, radiomics nomogram showed good fit and superior clinical utility. Conclusions: The present work constructed the new radiomics-based nomogram and verified its reliability for discriminating between PRISm and COPD.

In recent years, with the development of artificial intelligence, especially deep learning technology, researches on automatic detection of cerebrovascular diseases on medical images have made tremendous progress and these models are gradually entering into clinical practice. However, because of the complexity and flexibility of the deep learning algorithms, these researches have great variability on model building, validation process, performance description and results interpretation. The lack of a reliable, consistent, standardized design protocol has, to a certain extent, affected the progress of clinical translation and technology development of computer aided detection systems. After reviewing a large number of literatures and extensive discussion with domestic experts, this position paper put forward recommendations of standardized design on the key steps of deep learning-based automatic image detection models for cerebrovascular diseases. With further research and application expansion, this position paper would continue to be updated and gradually extended to evaluate the generalizability and clinical application efficacy of such tools.

The aim of this study is to investigate an approach to the electricity conductivity of nano-graphite in concrete by experiments, and different doses of nano-graphite have a different influence on the electrical conductivity of the whole structure. In this research project, the compression test was carried out on a 50 mm × 100 mm cylinder using a Baldwin machine and a digital multimeter to measure the conductivity. To find out the patterns of how different nano-graphites influence the properties of concrete structures, multifarious percentages of nano-graphite’s content are employed for testing both the electrical resistivity and compressive strength. These results are important to predict and prevent structural failure and fill the gap in nano-graphene concrete research, thereby saving human lives and significantly reducing the structures’ cost of maintenance and repair.

Abstract It is challenging to insulate sound transmission in low frequency-bands without blocking the air flow in a pipe. In this work, a small and light membrane-based cubic sound insulator is created to block acoustic waves in multiple low frequency-bands from 200 to 800 Hz in pipes. Due to distinct vibration modes of the membrane-type faces of the insulator and co-action of acoustic waves transmitting along different paths, large sound attenuation is achieved in multiple frequency-bands, and the maximum transmission loss reaches 25 dB. Furthermore, because the sound insulator with a deep subwavelength size is smaller than the cross-sectional area of the pipe, it does not block ventilation along the pipe.

AIM: To demonstrate the feasibility of mesenchymal stem cell (MSC)-mediated nano drug delivery, which was characterized by the “Trojan horse”-like transport of hypoxia-inducible factor-1α small interfering RNA (HIF-1α siRNA) between MSCs and retinal pigment epithelial cells (RPE) under hypoxia environment. METHODS: Plasmid and lentivirus targeting the human HIF-1α gene were designed and constructed. HIF-1α siRNA was encapsulated into poly(lactic-co-glycolic acid) nanoparticles (PLGA-NPs) through the water-in-oil-in-water (w/o/w) multiple emulsion technique. The effect of PLGA-NPs uptake on the expression of HIF-1α mRNA was tested in RPE cells by real-time quantitative polymerase chain reaction (qPCR) and additional transfected conditions were used as control, including lentivirus group, nude plasmid group and blank PLGA group. MSCs were transfected with the NPs and the transfection efficacy was evaluated by flow cytometry. Transwell co-culture system of transfected MSCs and RPE cells was constructed under hypoxia environment. The effects of MSC-loaded HIF-1α siRNA PLGA-NPs on proliferation, apoptosis, and migration of RPE cells were then evaluated. The durability of concrete structures is often reduced owing to the corrosion of reinforcement in an aggressive environment. Ordinary reinforcement methods, such as wrapping section steel or steel plate, are also vulnerable to corrosion. Using 6061-T6 aluminium alloy as near-surface reinforcement of the concrete structure is a feasible method. In this study, the corrosion resistance of 6061-T6 aluminium alloy bars was studied by simulating the coastal environment, atmospheric environment, and concrete internal environment with chloride solution, simulated acid rain solution, and saturated Ca(OH)2 solution. The corrosion rate of the 6061-T6 aluminium alloy in the above environments was tested using a weight loss method, and its corrosion resistance was evaluated using the metal corrosion resistance classification standard. Based on the electrochemical reaction mechanism, the polarisation properties and AC impedance spectra of steel and 6061-T6 aluminium alloy were compared, and the corrosion resistance mechanisms of steel and the 6061-T6 aluminium alloy in the above corrosive environments were obtained. The results show that the 6061-T6 aluminium alloy has better corrosion resistance than steel bars in chloride and atmospheric environments, with corrosion currents of 0.012 and 0.037 µA·cm−2, and 8-day corrosion rates of 0.051 and 0.031 mm·a−1, respectively. However, owing to the activity of the aluminium alloy, its corrosion resistance in an alkaline environment inside concrete is poor; the corrosion current is 0.22 µA·cm−2 and the 8-day corrosion rate is 16.166 mm·a−1. The research results can provide a reference for applying aluminium alloy bars as external prestressed concrete bars and near-surface steel bars.

Atherosclerosis is characterized by inflammation in the arterial wall, which is known to be exacerbated by diabetes. Therapeutic repression of inflammation is a promising strategy for treating atherosclerosis. In this study, we showed that diabetes aggravated atherosclerosis in apolipoproteinE knockout (ApoE−/-) mice, in which increased expression of long-chain acyl-CoA synthetase 1 (Acsl1) in macrophages played an important role. Knockdown of Acsl1 in macrophages (MφshAcsl1) reprogrammed macrophages to an anti-inflammatory phenotype, especially under hyperglycemic conditions. Injection of MφshAcsl1 reprogrammed macrophages into streptozotocin (STZ)-induced diabetic ApoE−/- mice (ApoE−/-+ STZ) alleviated inflammation locally in the plaque, liver and spleen. Consistent with the reduction in inflammation, plaques became smaller and more stable after the adoptive transfer of reprogrammed macrophages. Taken together, our findings indicate that increased Acsl1 expression in macrophages play a key role in aggravated atherosclerosis of diabetic mice, possibly by promoting inflammation. Adoptive transfer of Acsl1 silenced macrophages may serve as a potential therapeutic strategy for atherosclerosis.

Abstract. Background:. Changes in thyroid hormone levels are commonly recognized characters among the elderly, which were reported to potentially influence incident frailty. Therefore, we examined the cross-sectional associations of thyroid hormones (THs) with frailty as well as the five components characterizing frailty (fatigue, resistance, ambulation, number of illnesses, and loss of weight) among the oldest-old. Methods:. Four hundred and eighty-seven community-dwelling oldest-old from a local community in Haidian District, Beijing, participated in our recruitment campaign between April 2019 and May 2020. The primary outcomes were a definitive diagnosis of frailty according to the FRAIL scale (Fatigue, Resistance, Ambulation, Illnesses, Loss of weight) and a positive score for each frailty subdomain. Demographic information (age, sex, marital status, and educational status), comorbidities, and details on the participants’ lifestyles were recorded. Serum THs including free triiodothyronin (fT3), triiodothyronine (T3), free thyroxine (fT4), and thyroxine (T4) and thyroid stimulating hormone (TSH) levels were also measured at the beginning of our study. Logistic regressions were conducted to screen for potential risk factors for frailty and its subdomains. Results:. Among the total 487 subjects at enrollment, 60 (12.23%) of them were diagnosed with subclinical hypothyroidism and 110 (22.59%) of the total population scored positive for frailty. Logistic regression analyses adjusted for all potential confounders, showed that frailty was significantly associated with the serum TSH concentration (odds ratio [OR]: 1.06), fT3 concentration (OR: 0.54), and subclinical hypothyroidism score (OR: 2.18). The association between fT4 and frailty was absent in our observational study. The fT3/fT4 ratio characterizing peripheral hormone conversion was also tested to be correlated with frailty. Conclusion:. Subclinical hypothyroidism, higher TSH level, lower fT3 level, and decreased fT3/fT4 ratio were all associated with frailty assessed by the FRAIL scale among the community-dwelling oldest-old, suggesting a relevant role of thyroid function in aging. Future longitudinal studies are warranted to determine the casual relationship between thyroid dysfunction and frailty in the oldest-old.

Objective To observe the clinical efficacy of 3D printed microporous titanium artificial vertebral body in the treatment of ossification of posterior longitudinal ligament (OPLL). Methods The clinical data of 64 patients with OPLL who underwent anterior subtotal vertebral resection and decompression followed by 3D printed microporous titanium artificial vertebral body or titanium mesh cage implantation and titanium plate internal fixation in our department from January of 2018 to January of 2020 were collected and retrospectively analyzed. Finally, only 57 cases meeting the inclusion and exclusion criteria were recruited. According to different implants, they were divided into observation group (n=28, of 3D printed microporous titanium artificial vertebral body) and control group (n=29, titanium mesh cage). The operation time, intraoperative blood loss, Japanese Orthopaedic Association (JOA) score and improvement rate, as well as incidence of prosthesis settlement and bone graft fusion were compared between the 2 groups. Results The operation time and intraoperative blood loss in the treatment group were 59.32±12.09 min and 102.75±11.25 mL respectively, while those in the control group was 67.55±4.71 min and 102.06±13.83 mL respectively. The operation time was significantly shorter in the observation group than the control group (P=0.001), but there was no statistical difference in intraoperative blood loss between them. During the follow-up time from 12 to 15 (14.3±2.7) months, 1 case (control group) complicated with wound infection, and 7 cases (3 cases in the treatment group and 4 cases in the control group) complicated with neck swallowing discomfort, which disappeared in 1~2 weeks later. All patients were bone healing at the last follow-up, and no fracture or malposition of implants were seen in all patients. At the last follow-up, the treatment group had a JOA score increased from 11.61±0.99 to 16.10±0.68, and an improvement rate of (83.03±12.68)%, and that in the control group was elevated from 11.37±1.08 to 16.03±0.56, and an improvement rate was (81.58±12.67)%. However, there was no significant difference in the improvement rate between the 2 groups. Significant difference was seen in prosthesis settlement between the treatment group (1 case, 3.57%) and the control group (12 cases, 41.38%, P=0.001). Conclusion 3D printed microporous titanium artificial vertebral body can reconstruct the stability of cervical spine, with the advantages of better bone material interface, faster installation, and less prosthesis sinking when compared with titanium mesh cage.

In the present work, four groups of spherical chromium carbide reinforced NiCrBSi hardmetal coatings were prepared on AISI 4145 steel by plasma transferred arc (PTA) technique. The corrosion behavior of the four as-received hardmetal coatings in 0.5 mol/L H2SO4 solution was investigated by polarization curve and electrochemical impedance spectroscopy (EIS). The results revealed that more Cr-rich carbides (Cr3C2, Cr7C3 and M23(C, B)6) are formed in the chromium carbide reinforced coatings, while for the NiCrBSi hardmetal coating only Cr7C3 carbide was detected by XRD. The polarization results show that the chromium carbide reinforced NiCrBSi hardmetal coatings have positive corrosion potential and lower corrosion current, providing a better protective effect to the substrate metal. The combined effects of Cr-rich carbide ceramic phases and a more stable passive film of Cr2O3 greatly improved the corrosion resistances of the chromium carbide reinforced NiCrBSi hardmetal coatings. The coating with the highest spherical chromium carbide addition has more pores because of the thermal stress due to the difference of thermal expansion coefficient between the NiCrBSi bonding phase and chromium carbide reinforced phase. The negative effects of the pores weaken the corrosion resistance, and the coating with the 30% chromium carbide content shows the best corrosion resistance. For NiCrBSi hardmetal coatings with higher reinforced chromium carbide content, the repeatability of the corrosion current obtained by polarization fitting is not as good as that of coatings with lower chromium carbide content. The repeatability of polarization results becomes worse when the specimens keep in a more stable passive state.

Abstract Sepsis is one of the most common causes of death, which is closely related to the uncontrolled systemic inflammation. Dysregulation of M1 macrophage polarization is the primary contributor to serious inflammation. In this study, it is revealed that the murine homologue of circRNA SCAR (steatohepatitis‐associated circRNA ATP5B regulator), denoted as circRNA mSCAR hereafter, decreases in the macrophages of septic mice, which correlates with the excessive M1 polarization. To restore circRNA mSCAR in mitochondria, exosomes encapsulated with circRNA mSCAR are further electroporated with poly‐D‐lysine‐graft‐triphenylphosphine (TPP‐PDL), and thus TPP‐PDL facilitates the bound circRNA delivered into mitochondria when the exosomes engulf by the recipient cells. In in vivo septic mouse model and in vitro cell model, it is shown that the exosome‐based mitochondria delivery system delivers circRNA mSCAR into mitochondria preferentially in the macrophages, favoring macrophage polarization toward M2 subtype. Accordingly, the systemic inflammation is attenuated by exosome‐based mitochondrial delivery of circRNA mSCAR, together with alleviated mortality. Collectively, the results uncover the critical role of circRNA mSCAR in sepsis, and provide a promising approach to attenuate sepsis via exosome‐based mitochondrial delivery of circRNA mSCAR.

ObjectivesThe objectives of this study were to assess the changes in the left myocardial function after chemotherapy for childhood lymphoma and observe the predictive or monitor value for cancer treatment-related cardiac dysfunction (CTRCD) by speckle-tracking echocardiography.MethodsA total of 23 children with histopathological diagnoses of lymphoma were included, with age-matched normal controls. Comparative analysis of clinical serological tests and left heart strain parameters in children with lymphoma, including left ventricular global longitudinal strain (LVGLS); global myocardial work (GMW) indices, which include global work index (GWI), global constructive work (GCW), global wasted work, and global work efficiency; and the LS of subendocardial, middle, and subepicardial layer myocardium during left ventricular systole were measured: left atrial strain of reservoir phase (LASr), left atrial strain of conduit phase (LAScd), and left atrial strain of contraction phase (LASct).ResultsOne-way ANOVA showed that GLS, GWI, GCW, LASr, and LAScd were closely associated with CTRCD and multivariate logistic regression analysis showed that GLS was the most sensitive predictor for detecting patients at lofty risk of anthracycline-related cardiotoxicity. Both before and after chemotherapy, GLS in the left ventricle showed a pattern of basal segment 0.05). After chemotherapy, maximum flow rate in early mitral relaxation/left atrial systolic maximum flow rate (E/A) and left atrial volume index of each group were in the normal range, and the values of LASr, LAScd, and LASct slightly increased in the second cycle and decreased significantly in the fourth cycle after chemotherapy, reaching the lowest level; LASr and LAScd were positively correlated with GLS.ConclusionLVGLS is a more sensitive and earlier indicator to predict CTRCD compared with conventional echocardiography-related parameters and serological markers, and GLS of each myocardial layer showed a certain regularity. Left atrial strain can be used for early monitoring of cardiotoxicity in children with lymphoma after chemotherapy.
