Abstract

Tooth root development occurs through the interaction of multiple growth factors and transcription factors expressed in Hertwig's epithelial root sheath (HERS) and dental mesenchyme. Previously, we demonstrated that bobby sox homolog (Bbx) regulates odontoblast differentiation of human dental pulp stem cells. Here, we generated Bbx knockout (Bbx−/−) mice to address the functional role of Bbx in tooth formation. During tooth development, Bbx was expressed in both dental epithelium and mesenchyme. However, molar and incisor morphology in Bbx−/− mice at postnatal Day 0 (P0) exhibited no prominent abnormalities compared with their wild‐type (Bbx+/+) littermates. Until P28, the crown morphology in Bbx−/− mice was not distinctively different from Bbx+/+ littermates. Meanwhile, the length of the mandibular base in Bbx−/− mice was notably less at P28. Compared with Bbx+/+ mice, the mesial and distal root lengths of the first molar were reduced by 21.33% and 16.28% at P14 and 16.28% and 16.24% at P28, respectively, in Bbx−/− mice. The second molar of Bbx−/− mice also showed 10.16% and 6.4% reductions at P28 in the mesial and distal lengths, compared with Bbx+/+ mice, respectively. The gene expression analysis during early tooth root formation (P13) showed that the expression of dentin sialophosphoprotein (Dspp) was significantly decreased in Bbx−/− mice. 78. Polydopamine Chemistry: Adhesion Mechanism, Copolymerization, and Application

Abstract

Polydopamine (PDA), inspired by mussel adhesive proteins, has garnered significant interest for its ability to coat surfaces independently of their nature, using a simple and environmentally friendly process in basic aqueous solutions. However, understanding its binding and polymerization mechanisms remains challenging due to the numerous reactive sites of the dopamine monomer and the insoluble nature of the resulting polymer film. Additionally, PDA's short, rigid backbone differs from the long, flexible sequences found in mussel-binding proteins, which limits its effectiveness in achieving the conformal contact necessary for efficient adhesion. In this dissertation, I systematically investigate the roles of PDA's building blocks—poly(catechol), poly(catecholamine), and PDA—to enhance our understanding of their binding mechanisms. Contrary to initial expectations, we find that PDA's adhesion primarily arises from the solubility limits of catecholamine oligomers in aqueous environments, complemented by catechol's multiple binding modes. Notably, in the absence of amines, poly(catechol) remains in solution or forms minor suspensions without surface coating, emphasizing the crucial role of amines in facilitating insoluble aggregate formation for adhesion. We validate our findings by inducing poly(catechol) aggregation with quaternized poly(4-vinylpyridine) (qPVP), demonstrating adhesion upon agglomerate formation. Based on these insights, we develop a sequential self-polymerization strategy of phenolic compounds combined with alkanedithiol (ADT) crosslinkers for versatile surface-independent coating and functionalization. The resulting copolymer leverages phenol's diverse binding modes and ADT's flexible aliphatic chain for conformal substrate contact, with Michael addition reactions yielding solvent-resistant crosslinked polymer films. This approach successfully functionalizes various surfaces with phenolic monomers, overcoming stability issues encountered with conventional polydopamine derivatives. Furthermore, we achieve 5,6-dihydroxyindole (DHI)-free poly(catecholamine) (PCA) coating using 3,4-dihydroxybenzylamine hydrobromide, yielding smooth, uniform surfaces. The suppressed indole ring formation preserves primary amine groups, offering an alternative surface functionalization strategy, particularly for hydroxyl-containing surfaces. DHI-free PCA modification enhances the thermal conductivity of graphene-polymer composites, achieving a significant increase compared to untreated graphene controls. Overall, this work advances understanding of PDA's binding mechanisms and offers novel strategies for surface coating and functionalization with broad implications across material science and engineering. By elucidating the intricate processes involved in PDA adhesion and developing innovative coating techniques, this research opens avenues for the design of advanced materials with tailored properties and enhanced performance in various applications.

79. Nationwide trends in the incidence of orbital lymphoma from 1999 to 2016 in South Korea

Abstract

Background/AimsLymphomas are the most frequent neoplasm of the orbit. However, the epidemiology of orbital lymphomas is not well reported. This study aimed to provide a population-based report on the epidemiology of orbital lymphomas and measure the trends in the incidence of orbital lymphoma cancer in South Korea.MethodsNationwide cancer incidence data from 1999 to 2016 were obtained from the Korea Central Cancer Registry. Age-standardised incidence rates and annual percent changes were calculated according to sex and histological types. The analysis according to the Surveillance, Epidemiology, and End Results summary stage classifications was performed from 2006 to 2016. Survival rates were estimated for cases diagnosed from 1999 to 2016.ResultsA total of 630 patients (median age: 54 years) with orbital lymphoma in the orbital soft tissue were included in this study. The age-standardised incidence rates increased from 0.03 to 0.08 per 100 000 individuals between 1999 and 2016, with an annual percent change of 6.61%. The most common histopathological type of orbital lymphoma was extra marginal zone B cell lymphoma, accounting for 82.2% of all orbital lymphomas during 1999–2016, followed by diffuse large B cell lymphoma (9.2%). Five-year, 10-year and 15-year overall survival (OS) of orbital lymphoma was 90.8%, 83.8% and 75.8%, respectively. OS showed a significant decrease as age increased and no significant differences between men and women.ConclusionThe incidence rate of orbital lymphoma is very low in South Korea. However, the incidence rate has increased over the past years. Orbital lymphomas have a worse prognosis as age increases.

82. Secretome Analysis of Human BMSCs and Identification of SMOC1 as an Important ECM Protein in Osteoblast Differentiation

Abstract

Extracellular matrix proteins have been implicated in the regulation of osteoblast differentiation of bone marrow derived mesenchymal stem cells (BMSCs) through paracrine or autocrine mechanisms. In the current study, we analyzed the secretory protein profiles of BMSCs grown in osteogenic medium (OSM) and identified SPARC-related modular calcium-binding protein 1 (SMOC1), a member of the SPARC family, as a regulator of osteoblast differentiation of BMSCs. BMSCs with high and low osteogenic potential were grouped and stimulated with OSM, after which conditioned medium was collected and analyzed by LC-MS/MS. We identified 410 proteins, 64 of which were selectively secreted by high osteogenic potential BMSCs. Of these 64 secreted proteins, we selected extracellular matrix proteins for validation in BMSCs undergoing osteoblast differentiation and found that SMOC1 is highly expressed and secreted in BMSCs stimulated with OSM. To examine the role of SMOC1 in osteoblast differentiation, we analyzed the effect of SMOC1 knockdown and overexpression using shRNAs and wild-type cDNA, respectively. Knockdown of SMOC1 significantly inhibited mineralization and the expression of osteoblast differentiation markers, while overexpression of SMOC1 substantially increased the expression of osteoblast differentiation-related genes. Thus, validation of secretome profiling data identified SMOC1 as a putative regulator of osteoblast differentiation of BMSCs.

Abstract

Health-related quality of life (HRQOL) is an important outcome factor in chronic diseases such as inflammatory bowel disease (IBD). This study used the Korean translation of the disease-specific, self-administered Inflammatory Bowel Disease Questionnaire (IBDQ) to compare HRQOL in ulcerative colitis (UC; n = 98), Crohn's disease (CD; n = 49), and intestinal Behcet's disease (BD; n = 34). In addition to the current status, patients were asked retrospectively to recall their symptoms at the beginning and during the worst period of their disease. Disease activity was measured by St. Mark's Activity Index, Crohn's disease Activity Index (CDAI), and the Harvey-Bradshaw Index (HBI). In all IBD patients, including those with BD, the IBDQ total score during the worst period was significantly lower than that at present and that at the beginning of the disease. However, there were no significant differences between groups regarding the total IBDQ score or its various dimensions. In UC a strong correlation between IBDQ scores and St. Mark's Activity Index was observed (r = -0.708, P < 0.001). IBDQ scores were also highly correlated with CDAI and HBI in both CD (r = -0.506, P < 0.001 for CDAI; r = -0.600, P < 0.001 for HBI) and BD (r = -0.687, P < 0.001 for CDAI; r = -0.531, P < 0.001 for HBI). However, the current IBDQ score was not related to demographic parameters such as gender, age, educational status, economic status, and marital status as well as disease factors such as duration of disease, history of operation or hospital admission, extent of disease in UC, involved region in CD, and clinical type in BD. We conclude that the Korean IBDQ is a responsive and promising instrument for measuring HRQOL of IBD patients in clinical trials. In addition, the IBDQ can be helpful in developing a disease-specific activity index in BD.

93. Assessment of macular function by multifocal electroretinography following epiretinal membrane surgery with internal limiting membrane peeling

Eighteen eyes of 18 patients who underwent vitrectomy with ILM removal were included. Best corrected visual acuity, optical coherence tomography (OCT), and multifocal electroretinography were assessed prior to surgery, and 3 and 12 months after surgery. All surgical samples were obtained and confirmed inclusion of an ILM by electron microscopy. Visual acuity and the central foveal thickness by OCT improved significantly 3 months postoperatively, with gradual recovery by 12 months. Preoperatively, only the P1 amplitude in rings 1 and 2 were decreased compared with the normal fellow eyes (P < 0.01). Three and 12 months after surgery, the P1 amplitude in rings 1 and 2 were decreased compared with the preoperative P1 amplitudes, but without significance. The photoreceptor status by OCT was related to the N1 amplitude before and after surgery. Although visual acuity and macular edema were improved after surgery, macular function, as indicated by multifocal ERG, had limited recovery at 12 months.Keywords: epiretinal membrane, macular function, multifocal electroretinography
