Background: Osteosarcoma (OS) is a highly metastatic primary bone malignancy, and the treatment options remain inadequate. Hence, exploring innovative natural medications is required. Prunetin (PRU) is an isoflavone that has been a proven anticancer agent in numerous cancer cell lines. However, the activity of PRU against OS remains uncertain. Materials and Methods: Here, we studied the anticancer activity of PRU (20 and 25 µM) on human OS cells MG-63 and investigated its latent mechanism. The PRU activity of MG-63 cells cytotoxicity, intracellular ROS, metastasis, apoptosis, anti-apoptotic proteins, MAPK/STAT-3, and AKT signaling pathways was assessed by MTT assay, DCFH-DA, DAPI, PI, AO/EB, cell adhesion, and RT-PCR analysis. Findings unveiled that PRU could constrain MG-63 cell viability and adhesion through elevated intracellular ROS and elicited apoptosis. Results: Likewise, PRU (20 and 25 µM) avert the MG-63 cell proliferation, which stimulates apoptosis by the enhancement of Bax and caspases, while it diminishes Bcl-2 in a dose-dependent way. Furthermore, PRU could reduce Pin-1, and anti-apoptotic elements, as well as trigger apoptotic signaling pathways. Our data established that PRU alleviates MG-63 cell proliferation and metastasis via ROS-mediated apoptosis, which triggers MAPKs/STAT3 and AKT pathways, suggesting that PRU is a promising natural remedy for OS. In order to comprehend the therapeutic target for cancer, we assessed the effect of PRU on the expression of Pin1, which is thought to be over-expressed in many human malignancies. According to our findings, PRU specifically suppressed Pin1 expression to reduce the expression of Akt, STAT3, P38, JNK, P65, and IL-6. We evaluated the impact of PRU on the expression of Pin1, which is allegedly over-expressed in many human malignancies, to better understand the therapeutic target for cancer. Researchers state that PRU inhibited the expression of Akt, STAT3, P38, JNK, P65, and IL-6 in particular, by suppressing Pin1 expression. Physical and climatic variables such as Tree Canopy coverage, Normalized Difference Vegetation Index (NDVI), Distance to Roads, Distance to the Coast, Impervious Surface, and Precipitation can affect land surface temperature (LST). This paper examines the relationships using linear regression models and explores LST trends in the Miami Statistical Area (MSA) between 2002 and 2021. This study evaluates the effect of dry and wet seasons as well as day and night data on LST. A multiscale investigation is used to examine LST trends at the MSA scale, the individual county level, and at the pixel level to provide a detailed local perspective. The multiscale results are needed to understand spatiotemporal LST distributions to plan mitigation measures such as planting trees or greenery to regulate temperature and reduce the impacts of surface urban heat islands. The results indicate that LST values are rising in the MSA with a positive trend throughout the 20-year study period. The rate of change (RoC) for the wet season is smaller than for the dry season. The pixel-level analysis suggests that the RoC is primarily in rural areas and less apparent in urban areas. New development in rural areas may trigger increased RoC. This RoC relates to LST in the MSA and is different from global or regional RoC using air temperature. Results also suggest that climatic explanatory variables have different impacts during the night than they do in the daytime. For instance, the Tree Canopy variable has a positive coefficient, while during the day, the Tree Canopy variable has a negative relationship with LST. The Distance to the Coast variable changes from day to night as well. The increased granularity achieved with the multiscale analysis provides critical information needed to improve the effectiveness of potential mitigation efforts. [ABSTRACT FROM AUTHOR]

Steam-assisted gravity oil drainage (SAGD flooding) is a cutting-edge technology for the development of oils which is gradually replacing steam huff and puff and is being used more and more widely. Low-permeability interlayers are generally developed in oil reservoirs in China, which may shield the migration of steam, oil and gas. Targeted acidizing fracturing was proposed to break through the low-permeability interlayers, and hence, the problem that the hindrance to the expansion of the steam chamber led to heat loss and seriously affected the development effect could be solved. A typical kind of well with SAGD flooding actually applied in China, Shuyi District of Liaohe Oilfield, was taken as the example for studying the optimization of crack parameters. Based on the study of reservoir sensitivity characteristics in this well, the formulations of working fluids for targeted acidizing fracturing were developed by optimizing the weight percentages of main acid solution and additives. The formula of '4% hydrochloric acid + 2% polyphosphoric acid + 5% fluoroboric acid + 4% acetic acid' could be used as the acidizing fracturing working fluid for typical blocks of the Shuyi District of Liaohe Oilfield, which can increase the permeability of the natural core by 40.19–57.06%. The permutation flowshop scheduling problem is a vital production management concern of industries such as electronic product manufacturing and integrated circuit fabrication. In many studies, jobs are prioritized independently according to their features or properties. This article develops a new perspective by introducing the self-attention mechanism into scheduling for the first time. The job similarities are characterized by the dot-product of processing time matrices and taken as job priorities. Based on this idea, a new priority rule denoted by PRSAT is proposed, and a heuristic named NEHLJP2 is presented with makespan minimization. The computational results with the Taillard and VRF benchmarks and 640 random instances demonstrate that the new priority rule and heuristic dominate the existing ones at a nominal cost of computation time.
