Abstract

The FY-4B satellite is one of the second generation of China's geosynchronous meteorological satellites aiming at numerical weather forecasts. The space environment monitoring package (SEMP) onboard the FY-4B is a comprehensive instrument package for plasma, high-energy particle, and energetic neutral particle measurements. The low-energy particle analyzer (LEPA) is one of the instruments of the SEMP and consists of two top hat electrostatic analyzers designed for plasma detection. The electron and ion sensors are back-to-back assembled and are integrated to a shared electronic box. It measures the three-dimensional velocity distribution of low-energy electrons and ions on the geosynchronous orbit. In this paper, we present the ground calibration and in-flight performance of the instrument. With the electrostatic deflectors and the cylindrically symmetric structure, the instrument provides high-cadence measurements of electron and ion velocity distributions with a wide field of view (FOV) of 180° by 100°, an angular resolution of 16.7° × 20°, and a broad energy range for both the electrons and ions from tens of eV to above 30 keV, with a 1 s time resolution. The geometric factors of the electron and ion analyzers are 1.1 × 10−3 cm2·sr·eV/eV and 1.4 × 10−3 cm2·sr·eV/eV, respectively, which fulfills the requirements of the low-energy plasma measurement. The LEPA monitored typical space environment disturbance such as geomagnetic storms and successfully recorded the responses of plasma energy fluxes. Abstract

Malicious code threatens the safety of computer systems. Researching malicious code design techniques and mastering code behavior patterns are the basic work of network security prevention. With the game of network offense and defense, malicious code shows the characteristics of invisibility, polymorphism, and multi-dismutation. How to correctly and effectively understand malicious code and extract the key malicious features is the main goal of malicious code detection technology. As an important method of program understanding, program slicing is used to analyze the program code by using the idea of “decomposition”, and then extract the code fragments that the analyst is interested in. In recent years, data mining and machine learning techniques have been applied to the field of malicious code detection. The reason why it has become the focus of research is that it can use data mining to dig out meaningful patterns from a large amount of existing code data. Machine learning can It helps to summarize the identification knowledge of known malicious code, so as to conduct similarity search and help find unknown malicious code. The machine learning heuristic malicious code detection method firstly needs to automatically or manually extract the structure, function and behavior characteristics of the malicious code, so we can first slice the malicious code and then perform the detection. Through the improvement of the classic program slicing algorithm, this paper effectively improves the slicing problem between binary code processes. At the same time, it implements a malicious code detection system. Abstract

The Cancer/Testis Antigen (CTA) genes comprise a group of genes whose expression under physiological conditions is restricted to the testis but is activated in many human cancers. Depending on the particular expression pattern, the CTA genes are speculated to play a role in spermatogenesis, but evidence is limited thus far. Here, we reported patients with a hemizygous nonsense mutation in cancer-testis antigen 55 (CT55) suffering from male infertility with extreme disruption in sperm production, morphology, and locomotion. Specifically, the insufficiency of sperm individualization, excessive residue of unnecessary cytoplasm, and defects in acrosome development were evident in the spermatozoa of the patients. Furthermore, mouse models with depletion of Ct55showed accelerated infertility with age, mimicking the defects in sperm individualization, unnecessary cytoplasm removal, and meanwhile exhibiting the disrupted cumulus-oocyte complex penetration. Mechanistically, our functional experiments uncovered CT55 as a new autophagic manipulator to regulate spermatogenesis via selectively interacting with LAMP2 and GABARAP (which are key regulators in the autophagy process) and further fine-tuning their expression. Therefore, our findings revealed CT55as a novel CTA gene involved in spermatogenesis due to its unprecedented autophagy activity.

Abstract

The testis-specific protein, Y-linked 1 (TSPY1), a newly recognized cancer/testis antigen, has been suggested to accelerate tumor progression. However, the mechanisms underlying TSPY1 cancer-related function remain limited. By mining the RNA sequencing data of lung and liver tumors from The Cancer Genome Atlas, we found frequent ectopic expression of TSPY1 in lung adenocarcinoma (LUAD) and liver hepatocellular carcinoma (LIHC), and the male-specific protein was associated with higher mortality rate and worse overall survival in patients with LUAD and LIHC. Overexpression of TSPY1 promotes cell proliferation, invasiveness, and cycle transition and inhibits apoptosis, whereas TSPY1 knockdown has the opposite effects on these cancer cell phenotypes. Transcriptomic analysis revealed the involvement of TSPY1 in PI3K/AKT and RAS signaling pathways in both LUAD and LIHC cells, which was further confirmed by the increase in the levels of phosphorylated proteins in the PI3K-AKT and RAS signaling pathways in TSPY1-overexpressing cancer cells, and by the suppression on the activity of these two pathways in TSPY1-knockdown cells. Further investigation identified that TSPY1 could directly bind to the promoter of insulin growth factor binding protein 3 (IGFBP3) to inhibit IGFBP3 expression and that downregulation of IGFBP3 increased the activity of PI3K/AKT/mTOR/BCL2 and RAS/RAF/MEK/ERK/JUN signaling in LUAD and LIHC cells. Taken together, the observations reveal a novel mechanism by which TSPY1 could contribute to the progression of LUAD and LIHC. Our finding is of importance for evaluating the potential of TSPY1 in immunotherapy of male tumor patients with TSPY1 expression.
