Abstract

National audience; Une génisse Holstein âgée de dix-neuf mois est référée pour une fracture complète transversale du corps de la mandibule gauche. Elle présente des signes d’atteinte du nerf facial (VII). Un ptyalisme provoque une acidose métabolique. Le pH sanguin et la volémie sont rétablis. La fracture est réduite, puis fixée à l’aide d’un cerclage. Une antibiothérapie et un traitement anti-inflammatoire sont administrés. La plaie chirurgicale s’infecte. La couverture antibiotique est modifiée. L’abcès est drainé. Le ptyalisme et le déficit neurologique persistent pendant cinq semaines, mais la vache retourne en production. Les fractures mandibulaires surviennent fréquemment au cornadis. Souvent ouvertes et infectées, elles sont toutefois de bon pronostic après réduction chirurgicale. Elles peuvent provoquer des déséquilibres hydro-électrolytiques et acidobasiques, ainsi qu’une lésion du nerf vague.

Abstract

Hors-série "Parasitologie des ruminants laitiers"; National audience; Les principales affections parasitaires chez la vache laitière sont les affections digestives et respiratoires. Le principal symptôme digestif rencontré dans cette parasitose est la diarrhée, sous forme aiguë ou chronique. L’enquête préliminaire a pour objectif de cerner les modalités d’apparition de la maladie dans l’élevage. L’atteinte digestive est objectivée par un examen attentif de la cavité buccale, du rumen et des bouses. Les principaux prélèvements consistent en un prélèvement des fèces dans l’ampoule rectale, et un prélèvement sanguin. De plus en plus souvent, la recherche directe de parasites à la clinique est possible. Le laboratoire permet la recherche dans les bouses d’agents de diarrhée : bactéries, virus, parasites. Lorsque l’affection se traduit par la mort d’un animal, l’étude des lésions à l’autopsie peut fournir des éléments utiles au diagnostic. Enfin, des saisies ou des découvertes d’abattoir connues de l’éleveur peuvent aider le praticien dans la mise en place du diagnostic différentiel et éviter des examens complémentaires superflus.

Abstract

National audience; Spécifique à son statut de pré-ruminant et à son âge, les maladies nerveuses non congénitales du veau se répartissent en trois groupes : les troubles en hypo, les troubles en hyper et les affections à expression clinique localisée. Les trois quarts des maladies à l’origine de troubles nerveux en hypo chez le veau sont dues au syndrome myopathie-dyspnée et aux gastro-entérites néonatales. Les affections du système nerveux proprement dit qui provoquent une dépression sont : l’anorexie cérébrale, la gastro-entérite paralysante, les méningo-encéphalites, le botulisme et la carence en vitamine A. Seules les atteintes spécifiques du système nerveux peuvent entraîner des symptômes d’excitation chez le veau. Parmi ces affections, celles qui sont à l’origine d’une amaurose (nécrose du cortex cérébral, intoxication par le plomb et par le sélénium) sont plus rares que celles qui n’en provoquent pas. (méningo-encéphalites, entéroxémie, tétanos, hypomagnésémie). Enfin, certaines affections nerveuses, le plus souvent en début d’évolution, peuvent n’avoir qu’une expression clinique localisée : listériose, abcès cérébraux, otite moyenne, otite interne ou sinusite.

Abstract

National audience; La neuropathologie du veau dans sa première semaine de vie, en dehors des conséquences de l’hypoxie éventuellement induite par les conditions du part, regroupe les conséquences et les séquelles de diverses affections virales et parasitaires du fœtus, ainsi que les défauts de l’ontogénèse d’origine héréditaire, infectieuse ou inconnue. Le praticien est le plus souvent appelé pour un syndrome cérébelleux, plus rarement pour une encéphalite ou une méningo-encéphalite. Après un examen clinique le plus rigoureux possible en fonction du format de l’animal, le praticien entreprend une recherche étiologique. L’examen nécroscopique permet la détection de lésions macroscopiquement visibles et la préparation d’échantillons destinées au laboratoire d’analyses. L’anomalie tératologique la plus fréquente est l’hydrocéphalie. Le tableau clinique du syndrome cérébelleux ou les lésions d’hydrocéphalie doivent entraîner en première intention la recherche directe et/ou indirecte, à l’échelon individuel et à l’échelon du troupeau, du pestivirus de la diarrhée virale bovine maladie des muqueuses (B.V.D./M.D.V.).

National audience; Les diarrhées néonatales peuvent être dues à des facteurs nutritionnels ou infectieux. Parmi ces derniers, une importance particulière doit être accordée aux colibacilles, aux clostridies, aux salmonelles, aux rotavirus et aux cryptosporidies. Les symptômes et les tableaux néocropsiques sont très peu spécifiques. Le recours aux examens de laboratoire est indispensable pour préciser les causes des épisodes d’entérites néonatales, souvent multifactorielles. Le traitement et surtout la prévention doivent être envisagés à l’échelle du troupeau. Bien que des moyens de prophylaxie médicale existent pour plusieurs maladies, les mesures hygiéniques et la maîtrise des facteurs de risque sont des piliers de la prévention.

National audience; Les infections bactériennes du système nerveux central des bovins sont relativement rares. Parmi celles que l’ont peut rencontrer chez les adultes figurent la listériose (cf. Dossier Spécial Listérioses et Listéria, Bulletin des GTV, n°11, mai-août 2001) et la méningo-encéphalite thrombosante à Haemophilus somnus. Chez les veaux, la clinique est dominée par les méningites et méningo-encéphalites. Le diagnostic différentiel est difficile à mettre en œuvre. Il doit prendre en compte d’autres affections neurologiques, telles que la polio-encéphalomalacie ou nécrose du cortex cérébral ; les abcès intracrâniennes : le prélèvement de liquide céphalorachidien peut constituer une aide précieuse.

National audience; Le botulisme des ruminants est le plus souvent d’origine alimentaire. Associé à la présence de cadavres, il peut aussi apparaître en présence d’un aliment mal conservé : ensilage de mauvaise qualité, foin. L’abreuvement des animaux avec des eaux stagnantes, constitue un autre facteur de risque important, en particulier en zone de marais. Cette affection évolue sous forme de cas sporadiques ou de foyers d’étable ou de pâturage. Il se manifeste par une paralysie flasque. Il convient de la différencier de l’ESB. Le traitement, peu efficace, reste symptomatique. Les sérums spécifiques, comme les vaccins, ne sont pas disponibles en France.

Abstract

An epidemiological investigation was carried out on one hundred Salmonella isolates from broiler farms, slaughterhouses, and human patients in the Constantine region of Algeria, in order to explore the contribution of avian strains to human salmonellosis cases in this region over the same period of time. The isolates were characterized by phenotypic as well as genotypic methods. A large variety of antimicrobial resistance profiles was found among human isolates, while only seven profiles were found among avian isolates. Enterobacterial Repetitive Intergenic Consensus-PCR (ERIC-PCR), Insertion Sequence 200-PCR (IS200-PCR), and Pulsed Field Gel Electrophoresis (PFGE) resulted in the allocation of the isolates to 16, 20, and 34 different profiles, respectively. The 3 genotyping methods led to complementary results by underlining the clonality of some serovars with the diffusion and persistence of a single clone in the Constantine area as well as stressing the polymorphism present in isolates belonging to other serovars, indicating the diversity of potential reservoirs of nontyphoidal Salmonella. Altogether, our results seem to indicate that nontyphoidal avian Salmonella may play an important role in human salmonellosis in the Constantine region. [ABSTRACT FROM AUTHOR]

Abstract

ABSTRACTThe whole sequence of plasmid pENVA carrying the extended-spectrum β-lactamase gene blaCTX-M-15was determined. It was identified from a series of clonally related Klebsiella pneumoniaesequence type 274 strains recovered from companion animals. This plasmid was 253,984 bp in size and harbored, in addition to blaCTX-M-15, a large array of genes encoding resistance to many antibiotic molecules, including β-lactams (blaTEM-1, blaDHA-1), aminoglycosides (aacA2, aadA1), tetracycline (tetA), quinolones (qnrB4), trimethoprim (dfrA15), and sulfonamides (two copies of sul1). In addition, genes encoding resistance to mercury, tellurium, nickel, and quaternary compounds were identified. It also carried genes encoding DNA damage protection and mutagenesis repair and a locus for a CRISPR system, which corresponds to an immune system involved in protection against bacteriophages and plasmids. Comparative analysis of the plasmid scaffold showed that it possessed a structure similar to that of only a single plasmid, which was pNDM-MAR encoding the carbapenemase NDM-1 and identified from human K. pneumoniaeisolates. Both plasmids possessed two replicons, namely, those of IncFIB-like and IncHIB-like plasmids, which were significantly different from those previously characterized. The blaCTX-M-15gene, together with the other antibiotic resistance genes, was part of a large module likely acquired through a transposition process. We characterized here a new plasmid type carrying the blaCTX-M-15gene identified in a K. pneumoniaeisolate of animal origin. The extent to which this plasmid type may spread efficiently and possibly further enhance the dissemination of blaCTX-M-15among animal and human isolates remains to be determined.

Abstract

ABSTRACTScreening of extended-spectrum β-lactamase (ESBL)-producing Gram-negative bacteria in companion animals living in the Paris area in France identified a high rate of CTX-M-15-producing Klebsiella pneumoniae. Those isolates were recovered during the 2010-2011 period from both infections and asymptomatic colonizations. Sequence typing revealed that most of these isolates belonged to sequence type ST274. Interestingly, the blaCTX-M-15gene was located on a specific and novel plasmid scaffold. These findings highlight that companion animals may be reservoirs for CTX-M-15-producing K. pneumoniaeevolving separately from the human reservoir of CTX-M-15 producers.

Abstract

ABSTRACTA procedure that uses an original molecular marker (IS200-PCR) and that is based on the amplification of DNA with outward-facing primers complementary to each end of IS200has been evaluated with a collection of 85Salmonella entericasubsp. entericaserotype Typhimurium isolates. These strains were isolated from a group of 10 cows at different stages: during transportation between the farm and the slaughterhouse, on the slaughter line, from the environment, and from the final product (ground beef). The 85 isolates were characterized by their antibiotic resistance patterns and were compared by IS200-PCR and by use of four other genotypic markers. Those markers included restriction profiles for 16S and 23S rRNA (ribotypes) and amplification profiles obtained by different approaches: random amplified polymorphic DNA analysis, enterobacterial repetitive intergenic consensus PCR, and PCR ribotyping. The results of the IS200-PCR were in accordance with those of other molecular typing methods for this collection of isolates. Five different genotypes were found, which made it possible to refine the hypotheses on transmission obtained from phenotypic results. The genotyping results indicated the massive contamination of the whole group of animals and of the environment by one clonal strain originally recovered from one cow that excreted the strain. On the other hand, a few animals and their environment appeared to be simultaneously contaminated with genetically different strains.

86. Functional characterization of a miniature inverted transposable element at the origin of mcr-5 gene acquisition in Escherichia coli

Abstract

Plasmid-mediated colistin resistance of the mobile colistin resistance (MCR) type is a growing concern in Enterobacteriaceae since it has been described worldwide in humans and animals. Here, we identified a series of MCR-producing Escherichia coli isolates corresponding to two different clones (represented by isolates PS1 and PS8b) producing MCR-1 and MCR-5, respectively, obtained from pig fecal samples in France. Plasmid analysis showed that the plasmid carrying the mcr-1 gene (pPS1) possesses an IncHI2 backbone, whereas the mcr-5 gene was carried onto a 6,268-bp nontypeable non-self-conjugative plasmid (pPS8b). Detailed analysis of plasmid pPS8b revealed a 3,803-bp-long cassette containing the mcr-5 gene that was bracketed by two inverted-repeat (IR) sequences with 5-bp-long direct repeats at each extremity, similarly to an insertion sequence, but with the exception that no transposase gene was identified within this cassette. By performing in vitro transposition experiments, we showed that the mcr-5 cassette could be mobilized by the TnAs1 transposase provided in trans, displaying a mobilization mechanism similar to that of miniature inverted-repeat transposable elements (MITEs).

Abstract

Abstract: A study was made of Salmonella contamination in beef sampled from a slaughterhouse and from retailers in Dakar, Senegal. The serotypes as well as antibiotic-resistance patterns of the Salmonella isolates were determined. A total of 435 meat samples (236 from the slaughterhouse, 199 from retailers) were tested. Among them, 275 (63%) were positive for Salmonella, 43% (101/236) from the slaughterhouse and 87% (174/199) from the retailers. Furthermore, 97% of the investigated retailers had at least one beef sample contaminated by Salmonella. The 286 Salmonella isolates were divided into 51 serotypes. The most prevalent serotypes were Salmonella bredeney (25%), S. muenster (8%), S. waycross (7%), S. corvallis (4%) and S. kentucky (4%). About 62% of the isolates were resistant to nitrofurans. Resistance rates were lower to streptomycin (22%), sulfamethoxazole (15%), spectinomycin (1%), chloramphenicol (1%), and tetracycline (0,4%) while low-level resistance to quinolones was detected. About 16% of the Salmonella strains were multiresistant to two or more antibiotic families. Finally, ten resistance profiles have been identified. This study shows the huge spread of Salmonella in the beef production chain in Dakar, Senegal. Finally, this study provides the very first data about Salmonella prevalence in sub-saharian Africa. 89. Surveillance of Extended-Spectrum β-Lactamase-, Cephalosporinase- and Carbapenemase-Producing Gram-Negative Bacteria in Raw Milk Filters and Healthy Dairy Cattle in Three Farms in Île-de-France , France.

Abstract

The aim of this work was to test a surveillance protocol able to detect extended-spectrum β-lactamase (ESBL)-, cephalosporinase (AmpC)- and carbapenemase (CP)-producing gram-negative bacteria in three conveniently chosen dairy farms with known prior occurrences of ESBL- and CP-producing strains. The protocol was applied monthly for a year. At each visit, 10 healthy lactating dairy cows were rectally swabbed, and raw milk filters (RMFs) were sampled in two of the three farms. Bacterial isolation was based on a first screening step with MacConkey agar supplemented with 1 mg/L cefotaxime and commercial carbapenem-supplemented media. We failed to detect CP-producing strains but showed that ESBL- Escherichia strains, found in one farm only (13 strains), were closely associated with multi-drug resistance (12 out of 13). The limited number of conveniently selected farms and the fact that RMFs could not be retrieved from one of them limit the validity of our findings. Still, our results illustrate that ESBL-status changes monthly based on fecal swabs and negative herds should be qualified as "unsuspected" as proposed by previous authors. Although surveillance of farm statuses based on RMF analysis could theoretically allow for a better sensitivity than individual swabs, we failed to illustrate it as both farms where RMFs could be retrieved were constantly negative. Determination of CP herd-level status based on RMFs and our surveillance protocol was hindered by the presence of intrinsically resistant bacteria or strains cumulating multiple non-CP resistance mechanisms which means our protocol is not specific enough for routine monitoring of CP in dairy farms., Competing Interests: The authors declare that this study received funding from Ceva Santé Animale.
