Abstract

Background: Selenium is an essential element; however, at higher doses, it can be toxic. Therefore, alternative nanotechnological solutions are required to overcome toxicological issues, rather than conventional alternatives. Nanoparticles show new and promising properties that may be able to suppress toxicity while maintaining the positive effects of selenium on an organism. The aim of the experiment was to determine the influence of sodium selenite and selenium nanoparticles (SeNPs) on the antioxidant status of rats. Methods: The males of the outbreed rat strain Wistar albino were selected as a model organism. Animals were fed different forms of selenium. The control group was given a mixture without selenium addition, whereas other groups were fed a mixture containing sodium selenite, Se-49, and Se-100 SeNPs respectively. The duration of the trial was 30 days. Results: Analysis of blood and liver was performed where the concentration of reduced (GSH) and oxidised (GSSG) glutathione, and total selenium content were measured. In the liver, a significant reduction in GSSG was found for all experiment groups. Blood samples showed a significant reduction in GSH and an increase in GSSG. Discussion: These results show that SeNPs may be an alternative to dietary selenium for animal organisms. [ABSTRACT FROM AUTHOR]

Abstract

The aim of the experiment was to determine the effect of selenium nanoparticles (SeN) and selenium nanoparticles bound with glucose (SeN-GLU) on the antioxidant status of rats. The rats were fed with two defined forms of selenium nanoparticles for ten days. The first experimental group (n=6) was dosed with SeN solution (0.06 mg of Se per kg of body weight / day). The second experimental group (n=6) was dosed with SeN-GLU (0.06 mg of Se and 0.3 mg of glucose per kg of body weight / day). In both experimental groups, the antioxidant activity was decreased in rat plasma and increased in liver, when measured using the both free radicals (FR) and Ferric Reducing Antioxidant Power (FRAP) method. Electrochemical technique (differential pulse voltammetry-DPV) was applied to analyze the content of metallothionein (MT) and heavy metals. The reduced and oxidized forms of glutathione were analyzed by high performance liquid chromatography with electrochemical detection (HPLC-ED). In the analysis of whole blood, a significant increase of reduced glutathione (GSH) by 41% (P < 0.05) was observed in the SeN-GLU group compared with the control group. Higher levels of GSH were also observed in the SeN group by 12%, but without any statistical significance. On the other hand, a smaller increase in oxidized glutathione (GSSG) by 22% (P < 0.05) was observed for the SeN-GLU group and an insignificant decrease by 13% was recorded for the SeN group which implies increased GSH/GSSG ratio and higher antioxidant activity for both SeN-GLU and SeN groups in the whole blood. In the assessing of GSH in erythrocytes, a linear increase was observed for each group., Cílem experimentu bylo stanovit vliv selenových nanočástic (SeN) a selenových nanočástic vázáných s glukózou (SeN-GLU) na antioxidační status potkanů. Potkani byli krmeni dvěma definovanými formami selenových nanočástic po dobu deseti dnů. První experimentální skupině (n = 6) byl podáván roztok SeN (Se 0,06 mg/kg tělesné hmotnosti/den). Druhé experimentální skupině (n = 6) byl podáván SeN-GLU (0,06 mg Se a 0,3 mg glukózy na kg tělesné hmotnosti/den). V obou experimentálních skupinách byla snížena antioxidační aktivita v plazmě potkanů a zvýšena v játrech, měřeno metodou volných radikálů (FR) a metodou FRAP. Elektrochemická metoda (diferenční pulsní voltametrie – DPV) byla použita pro analyzování obsahu metalothioneinu (MT) a těžkých kovů. Redukované a oxidované formy glutathionu byly analyzovány pomocí vysokoúčinné kapalinové chromatografie s elektrochemickou detekcí (HPLC-ED). Při analýze krve bylo pozorováno výrazné zvýšení redukovaného glutathionu (GSH) o 41 % (P<0,05) ve skupině SeN-GLU ve srovnání s kontrolní skupinou. Vyšší hladiny GSH o 12 % byly pozorovány také ve skupině SeN, ale bez jakékoliv statistické významnosti. Na druhé straně, menší nárůst oxidovaného glutathionu (GSSG) o 22 % (P<0,05) byl pozorován u skupiny SeN-GLU a nevýznamný pokles o 13% byl zaznamenán u skupiny SeN, což znamená zvýšení poměru GSH/GSSG a vyšší antioxidační aktivitu pro SeN-GLU a SeN skupiny v krvi. Při posuzování GSH v erytrocytech byl pozorován lineární nárůst pro každou skupinu.

Abstract

The aim of the dissertation thesis was to analyze reproductive performance of sows recovered by the means of repopulation, in operating conditions of a production farm. Another objective was to analyze productive parameters of piglets and losses of piglets from birth to weaning. Based on achieved results it can be stated, that repopulated group of sows showed significantly higher reproductive performance compared to sows before repopulation. An anylsis of reproductive performance of gilts coming from two commercial programs was also conducted. The differences in reproductive performance were not found significant, however the differences in the losses of piglets were significant in favor of the program II. From the analysis of the productive parameters of piglets it is evident that piglets in the commercial program I had better growth intensity compared to piglets in the commercial program II, demonstrated by significantly higher weaning weight and daily gain of piglets. Also reproductive performance of repopulated sows from the 1st to the 4th litter was analyzed. The results indicate that there is a significant difference between parities. Reproductive performance had increasing tendency up to the 3rd litter and in the 4th litter it decreased. The most frequent causes of discarding of sows were problems with musculoskeletal system and abortions.
