- Abstract
Background: The hippocampus is one of the first cortical regions to exhibit Alzheimer's Disease (AD) pathology. The spatially‐related firing of hippocampal place cells provides the cellular basis for spatial memory and this is impaired relatively early in AD, yet few studies examine place cell activity in AD mouse models. We undertook an in‐depth characterisation of hippocampal pyramidal cell activity in an APP knock‐in model of AD. Method: Microdrives carrying 8 tetrodes were implanted into the left CA1 hippocampal subfield of 14‐month female mice (four APPNL‐G‐F, four wild‐type littermates). Mice undertook daily foraging sessions in an open field and ran on a linear track as CA1 activity was recorded. On completion of experiments, 40µm coronal sections of fixed brain tissue were stained using Cresyl Violet to verify tetrode placement and Thioflavin‐S to quantify amyloid β plaque burden. Result: 270 cells were recorded (119 wild‐type, 151 APPNL‐G‐F). CA1 pyramidal cells were classified as "place cells" or "non‐place cells" based on their spatial information content (bits/spike). Pyramidal cells recorded in wild‐type mice had significantly greater spatial information content than APPNL‐G‐F pyramidal cells (Figure 1), and a higher proportion were classified as place cells. Wild‐type place cells had significantly greater spatial information content than APPNL‐G‐F place cells. Corresponding differences were identified in place field size (Figures 2, 3). A positive correlation was identified between Aβ plaque burden and pyramidal cell spatial information content in the four APPNL‐G‐F mice (Figure 4). Coordination of place cell firing by the hippocampal theta rhythm was disrupted in APPNL‐G‐F mice; phase‐locking of pyramidal cells and place cells was significantly reduced (Figure 5), there was less coherence in the preferred theta phase across the pyramidal cell population, and a reduction in theta phase precession was observed. Conclusion: APPNL‐G‐F CA1 place cells exhibited deficits in both rate coding and temporal coding of spatial information indicating that amyloid β pathology, in the absence tau tangles, was associated with a disruption of hippocampal place cell function. These results provide preliminary support for the hypothesis that disruption of hippocampal function by AD pathology manifests as altered place cell activity and spatial behaviour. - Abstract
In 2021, both social and economic activities began to return towards the levels preceding the COVID-19 pandemic for some parts of the globe, with others still experiencing restrictions. Meanwhile, the climate has continued to respond to the ongoing increase in greenhouse gases and resulting warming. La Niña, a phenomenon which tends to depress global temperatures while changing rainfall patterns in many regions, prevailed for all but two months of the year. Despite this, 2021 was one of the six-warmest years on record as measured by global mean surface temperature with an anomaly of between +0.21° and +0.28°C above the 1991–2020 climatology.

- Abstract
In 2021, both social and economic activities began to return towards the levels preceding the COVID-19 pandemic for some parts of the globe, with others still experiencing restrictions. Meanwhile, the climate has continued to respond to the ongoing increase in greenhouse gases and resulting warming. La Niña, a phenomenon which tends to depress global temperatures while changing rainfall patterns in many regions, prevailed for all but two months of the year. Despite this, 2021 was one of the six-warmest years on record as measured by global mean surface temperature with an anomaly of between +0.21° and +0.28°C above the 1991–2020 climatology. Lake surface temperatures were their highest on record during 2021. The number of warm days over land also reached a new record high. Exceptional heat waves struck the Pacific Coast of North America, leading to a new Canadian maximum temperature of 49.6°C at Lytton, British Columbia, on 29 June, breaking the previous national record by over 4°C. In Death Valley, California, the peak temperature reached 54.4°C on 9 July, equaling the temperature measured in 2020, and the highest temperature recorded anywhere on the globe since at least the 1930s. Over the Mediterranean, a provisional new European record of 48.8°C was set in Sicily on 11 August. In the atmosphere, the annual mean tropospheric temperature was among the 10 highest on record, while the stratosphere continued to cool. While La Niña was present except for June and July, likely influencing Australia’s coolest year since 2012 and wettest since 2016, other modes of variability played important roles. A negative Indian Ocean dipole event became established during July, associated with a warmer east and cooler west Indian Ocean. Northern Hemisphere winters were affected by a negative phase of the North Atlantic Oscillation at both the beginning and end of 2021. Neuronal grid cells fire in a spatial grid pattern laid out across the surface of a familiar environment, however the role of environmental boundaries in the construction of this pattern is not well understood; this study shows that the grid pattern orients to the walls of polarized environments such as squares but not circles and that the hexagonal grid symmetry is permanently broken in highly polarized environments such as trapezoids. Grid cell alignment reflects the environment The neuronal grid cells of the entorhinal cortex fire in a spatial grid pattern laid out across the surface of a familiar environment to provide the brain with an internal map of an animal's surroundings. The role of environmental boundaries in the construction of this pattern is not well understood. Early studies had suggested that properties such as symmetry, orientation and scale of grid cells' firing patterns were independent of an environment's shape. But now two separate papers in this issue of Nature -- one from Edvard Moser and colleagues and the other from John O'Keefe and colleagues -- demonstrate that grid orientation, scale, symmetry and homogeneity can be strongly affected by environmental geometry, with grid cells aligned with the borders of the environment at an offset of a few degrees such that it minimizes symmetry with boundaries. These findings suggest a mechanism by which the geometry of an environment causes local rotation and deformation of the hexagonal firing patterns of grid cells. Grid cells represent an animal's location by firing in multiple fields arranged in a striking hexagonal array.sup.1. Such an impressive and constant regularity prompted suggestions that grid cells represent a universal and environmental-invariant metric for navigation.sup.1,2. Originally the properties of grid patterns were believed to be independent of the shape of the environment and this notion has dominated almost all theoretical grid cell models.sup.3,4,5,6. However, several studies indicate that environmental boundaries influence grid firing.sup.7,8,9,10, though the strength, nature and longevity of this effect is unclear. Here we show that grid orientation, scale, symmetry and homogeneity are strongly and permanently affected by environmental geometry. We found that grid patterns orient to the walls of polarized enclosures such as squares, but not circles. Furthermore, the hexagonal grid symmetry is permanently broken in highly polarized environments such as trapezoids, the pattern being more elliptical and less homogeneous. Our results provide compelling evidence for the idea that environmental boundaries compete with the internal organization of the grid cell system to drive grid firing. There are currently a number of theories of rodent hippocampal function. They fall into two major groups that differ in the role they impute to space in hippocampal information processing. On one hand, the cognitive map theory sees space as crucial and central, with other types of nonspatial information embedded in a primary spatial framework. On the other hand, most other theories see the function of the hippocampal formation as broader, treating all types of information as equivalent and concentrating on the processes carried out irrespective of the specific material being represented, stored, and manipulated. One crucial difference, therefore, is the extent to which theories see hippocampal pyramidal cells as representing nonspatial information independently of a spatial framework. Studies have reported the existence of single hippocampal unit responses to nonspatial stimuli, both to simple sensory inputs as well as to more complex stimuli such as objects, conspecifics, rewards, and time, and these findings been interpreted as evidence in favor of a broader hippocampal function. Alternatively, these nonspatial responses might actually be feature-in-place signals where the spatial nature of the response has been masked by the fact that the objects or features were only presented in one location or one spatial context. In this article, we argue that when tested in multiple locations, the hippocampal response to nonspatial stimuli is almost invariably dependent on the animal's location. Looked at collectively, the data provide strong support for the cognitive map theory.

Traditional models of future alcohol use in adolescents have used variable-centered approaches, predicting alcohol use from a set of variables across entire samples or populations. Following the proposition that predictive factors may vary in adolescents as a function of family history, we used a two-pronged approach by first defining clusters of familial risk, followed by prediction analyses within each cluster. Thus, for the first time in adolescents, we tested whether adolescents with a family history of drug abuse exhibit a set of predictors different from adolescents without a family history. We apply this approach to a genetic risk score and individual differences in personality, cognition, behavior (risk-taking and discounting) substance use behavior at age 14, life events, and functional brain imaging, to predict scores on the alcohol use disorders identification test (AUDIT) at age 14 and 16 in a sample of adolescents (N = 1659 at baseline, N = 1327 at follow-up) from the IMAGEN cohort, a longitudinal community-based cohort of adolescents. In the absence of familial risk (n = 616), individual differences in baseline drinking, personality measures (extraversion, negative thinking), discounting behaviors, life events, and ventral striatal activation during reward anticipation were significantly associated with future AUDIT scores, while the overall model explained 22% of the variance in future AUDIT. In the presence of familial risk (n = 711), drinking behavior at age 14, personality measures (extraversion, impulsivity), behavioral risk-taking, and life events were significantly associated with future AUDIT scores, explaining 20.1% of the overall variance. Results suggest that individual differences in personality, cognition, life events, brain function, and drinking behavior contribute differentially to the prediction of future alcohol misuse. This approach may inform more individualized preventive interventions.
