Abstract

International audience; Neutron-rich light nuclei and their reactions play an important role for the creation of chemical elements. Here, data from a Coulomb dissociation experiment on $^{20,21}$N are reported. Relativistic $^{20,21}$N ions impinged on a lead target and the Coulomb dissociation cross section was determined in a kinematically complete experiment. Using the detailed balance theorem, the $^{19}\mathrm{N}(\mathrm{n},\gamma)^{20}\mathrm{N}$ and $^{20}\mathrm{N}(\mathrm{n},\gamma)^{21}\mathrm{N}$ excitation functions and thermonuclear reaction rates have been determined. Reifarth et al: ; 12 págs.; 9 figs.; Open Access funded by Creative Commons Atribution Licence 3.0 ; Nuclear Physics in Astrophysics VI (NPA6), The nucleosynthesis of elements beyond iron is dominated by neutron captures in the s and r processes. However, 32 stable, proton-rich isotopes cannot be formed during those processes, because they are shielded from the s-process ow and r-process -decay chains. These nuclei are attributed to the p and rp process. For all those processes, current research in nuclear astrophysics addresses the need for more precise reaction data involving radioactive isotopes. Depending on the particular reaction, direct or inverse kinematics, forward or time-reversed direction are investigated to determine or at least to constrain the desired reaction cross sections. The Facility for Antiproton and Ion Research (FAIR) will oer unique, unprecedented opportunities to investigate many of the important reactions. The high yield of radioactive isotopes, even far away from the valley of stability, allows the investigation of isotopes involved in processes as exotic as the r or rp processes., This project was supported by the HGF Young Investigators Project VH-NG-327, EMMI, H4F, HGS-HIRe, JINA, NAVI, DFG and ATHENA.

Abstract

We present experimental results on the electromagnetic excitation of neutron-rich nickel isotopes, making use of the (RB)-B-3-LAND setup at GSI. Exotic beams were produced at approximately 500 MeV/u and their reactions were studied in inverse kinematics. Integral cross sections for Ni-58 are discussed and compared to previous data, providing a validation of our experimental method. The El excitation-energy distribution of the unstable Ni-68 is presented as well, showing an excess in cross section in the 1n decay channel when compared only with a typical Giant Dipole Resonance.

Abstract

The advantages and disadvantages of different types of shaping operations (extrusion, forging, combination variants) for sintered semifinished products of powder molybdenum alloys are examined. The article presents the mechanical properties and microstructure of bars of cast and powder molybdenum alloys obtained from semifinished products deformed by different methods.

Abstract

The properties of oxide powders from different suppliers are considered. The production of molybdenum powder by the reduction of oxides in hydrogen is discussed. The influence of the oxide properties on the properties of the molybdenum powder is demonstrated. Recommendations are made regarding means of assessing powder quality.

Abstract

Atacicept, a recombinant fusion protein containing the extracellular, ligand-binding portion of the transmembrane activator and calcium modulator and cyclophilin-ligand interactor receptor, and the Fc portion of human immunoglobulin (Ig) G, is designed to block the activity of B-lymphocyte stimulator and a proliferation-inducing ligand, and may have utility as a treatment for B-cell-mediated diseases, such as systemic lupus erythematosus (SLE). This Phase Ib study investigated the safety, tolerability, pharmacokinetics (PK) and pharmacodynamics of intravenous (i.v.) atacicept in patients with mild-to-moderate SLE. Patients (n = 24) were randomised (5:1) to receive atacicept (single dose: 3, 9 or 18 mg/kg; or multiple dose: 2 x 9 mg/kg) or matching placebo. Patients were followed for 6 weeks after dosing (9 weeks in the 2 x 9 mg/kg cohort). Local tolerability of atacicept was comparable with that of placebo, with only mild injection-site reactions reported with atacicept. Atacicept i.v. was generally well tolerated, both systemically and locally, in patients with mild-to-moderate SLE. Atacicept displayed non-linear PK, which was predictable across doses and between single and repeat doses. The biological activity of atacicept was demonstrated by its marked effect in reducing B-cells and Ig levels in patients with SLE. This supports the utility of this therapeutic approach in the treatment of autoimmune diseases, such as SLE.

Abstract

This work presents a direct measurement of the $^{96}$Ru($p, \gamma$)$^{97}$Rh cross section via a novel technique using a storage ring, which opens opportunities for reaction measurements on unstable nuclei. A proof-of-principle experiment was performed at the storage ring ESR at GSI in Darmstadt, where circulating $^{96}$Ru ions interacted repeatedly with a hydrogen target. The $^{96}$Ru($p, \gamma$)$^{97}$Rh cross section between 9 and 11 MeV has been determined using two independent normalization methods. As key ingredients in Hauser-Feshbach calculations, the $\gamma$-ray strength function as well as the level density model can be pinned down with the measured ($p, \gamma$) cross section. Furthermore, the proton optical potential can be optimized after the uncertainties from the $\gamma$-ray strength function and the level density have been removed. As a result, a constrained $^{96}$Ru($p, \gamma$)$^{97}$Rh reaction rate over a wide temperature range is recommended for $p$-process network calculations.

Two-proton decay of the unbound Tz = -2 nucleus 16Ne , produced in one-neutron knockout from a 500 MeV/u 17Ne beam, has been studied at GSI. The ground state, at a resonance energy 1.388(15) MeV, (Γ = 0.082(15) MeV) above the 14O +p+p threshold, and two narrow resonances at Er = 3.220(46) MeV and 7.57(6) MeV have been investigated. A comparison of the energy difference between the first excited 2+ state and the 0+ ground state in 16Ne with its mirror nucleus 16C reveals a small Thomas-Ehrman shift (TES) of +70(46) keV. A trend of the TES for the T = 2 quintet is obtained by completing the known data with a prediction for 16F obtained from an IMME analysis. The decay mechanisms of the observed three resonances were revealed from an analysis of the energy and angular correlations of the 14O +p+p decay products. The ground state decay can be considered as a genuine three-body (democratic) mode and the excited states decay sequentially via states in the intermediate nucleus 15F , the 3.22 MeV state predominantly via the 15F ground-state resonance, while the 7.57 MeV state decays via the 5/2+ resonance in 15F at 2.8 MeV above the 14O +p+p threshold. Abstract

The effect of the sintering medium, i.e., hydrogen at different degrees of drying and evacuation, on the mechanical properties of specimens of Mo – TiN and Mo – ZrN powder compositions with 2% nitrides, which are sintered with relative density of 93 – 96%, is investigated. It is shown that vacuum sintering of the Mo – ZrN composition is much more advantageous with respect to the efficiency of hardening of molybdenum than sintering in hydrogen. The Mo – ZrN composition has the best combination of mechanical properties (strength, ductility) as a result of formation of a fine-grain structure and fine (finer than 1 μm) oxide (ZrO2) and nitride (ZrN) phases.

Abstract

The effect of additives of titanium, zirconium, hafnium, and niobium in an amount of 2% on the technological and mechanical properties of sintered molybdenum and tungsten is considered. The changes in the chemical and phase compositions of pressed specimens after sintering in a flow of hydrogen are described. The temperature ranges of the effective hardening of molybdenum and tungsten by nitride additives are established. The composition with the best parameters of ductility and strength in tensile tests is determined.

Abstract

The radionuclide Fe-60 has been of great interest to the nuclear astrophysics community for over a decade. An initial discrepancy between the observed and modeled Galactic Fe-60/Al-26 ratio motivated numerous studies focused on the nucleosynthesis of these two isotopes, though the cross section of the primary astrophysical production reaction, Fe-59(n,gamma)Fe-60, has remained purely theoretical. The present work offers a first experimental constraint on the Fe-59(n,gamma)Fe-60 cross section at astrophysical energies, obtained indirectly via Coulomb dissociation, and demonstrates that the theoretical reaction rates used in present stellar models are not highly erroneous.

Abstract

We report on the first observation of the unbound proton-rich nucleus 15Ne. Its ground state and first excited state were populated in two-neutron knockout reactions from a beam of 500 MeV/u 17Ne. The 15Ne ground state is found to be unbound by 2.522(66) MeV. The decay proceeds directly to 13O with simultaneous two-proton emission. No evidence for sequential decay via the energetically allowed 2- and 1- states in 14F is observed. The 15Ne ground state is shown to have a strong configuration with two protons in the (sd) shell around 13O with a 63(5)% (1s1/2)2 component.

Abstract

Coulomb breakup at high energy in inverse kinematics of proton-rich Cl-31 was used to constrain the thermonuclear S-30(p,gamma)Cl-31 capture reaction rate under typical Type I x-ray burst conditions. This reaction is a bottleneck during rapid proton-capture nucleosynthesis (rp process), where its rate depends predominantly on the nuclear structure of Cl-31. Two low-lying states just above the proton-separation threshold of S-p = 296( 50) keV in Cl-31 have been identified experimentally using the (RB)-B-3-LAND setup at the GSI Helmholtzzentrum fur Schwerionenforschung GmbH. Both states are considered to play a key role in the thermonuclear S-30( p,gamma)Cl-31 capture reaction. Excitation energies of the first J(pi) = 1/2(+), 5/2(+) states have been extracted and the reaction rate for proton capture on S-30 under typical rp-process temperatures has been investigated.

Radioactive beams of 14,15B produced by fragmentation of a primary 40Ar beam were directed onto a Pb target to investigate the neutron breakup within the Coulomb field. The experiment was performed at the LAND/R3B setup. Abstract

The E1 strength distribution in Ni68 has been investigated using Coulomb excitation in inverse kinematics at the RB3-LAND setup and by measuring the invariant mass in the one- and two-neutron decay channels. The giant dipole resonance and a low-lying peak (pygmy dipole resonance) have been observed at 17.1(2) and 9.55(17) MeV, respectively. The measured dipole polarizability is compared to relativistic random phase approximation calculations yielding a neutron-skin thickness of 0.17(2) fm. A method and analysis applicable to neutron-rich nuclei has been developed, allowing for a precise determination of neutron skins in nuclei as a function of neutron excess.

Abstract

The effect of extrusion and heat treatment regimes on mechanical properties of rods 40 – 45 mm in diameter fabricated from powder molybdenum alloys is investigated. The alloys are fabricated from a mixture of Mo and 0.1 – 2.0% ZrB2. A deformed alloy of molybdenum with 2% ZrB2 is shown to be hardened effectively. An optimum composition is chosen for a high-temperature molybdenum alloy possessing good ductility at 20°C in deformed and recrystallized states.

Abstract

Mechanical properties of deformed rods fabricated from a cast niobium alloy of grade 5VMTs-2 and powder molybdenum and tungsten alloys of grades M-MP, MV-2-MP, and V-5-MP are studied in the temperature range of 120–2000°C. It is shown that the behavior of the temperature dependences of the mechanical properties of the rods in the lengthwise and widthwise directions is similar. The ductility characterstics differ substantially in the range of the brittleness threshold. By increasing the degree of deformation the anisotropy of the mechanical properties is diminished and the level of the ductility is increased.

Abstract

The technology of powder metallurgy is widely used in the industry for the production of deformed semiproducts (rods, sheets, strips, forgings) from refractory metals, molybdenum (of grade M-PM) in particular. Molybdenum in the form of deformed rods is used in structures operating at a temperature of up to 2000°C. The requirements on the stability of the mechanical properties of structural materials are very strict (especially at room temperature). The stability of the mechanical properties of molybdenum rods is affected substantially by the types and conditions of the deformation treatment of the sintered preforms. 73. Enhancement of Oil Recovery During Improved Thermal-Mining Development of Yarega Field

Abstract

This work is devoted to the actual problem – increase in completeness of extraction of oil on Yaregskaya area of Yarega oil field. In present time the most effective method of exploitation of Yaregskaya area is thermo-mine technology using underground-surface system with radial scheme of formation exposing 300 m wells. Improve ment of thermo-mine method of exploitation allows to increase recovery rates and oil recovery factor. In present time to recover the remaining reserves it is proposed to construct mini-mines using underground and surface system with panel system of formation exposing 800 m wells. Using of this system will allow to reduce penetration of excavation in 2 times. Besides, it will allow save temperature of the miner atmosphere in admissible limits till the end of oil field exploration. Construction of mini-mines will allow to reduce charges of excavation. Yarega oil field of high-viscosity oil is situated in 18 km to the south-west from Ukhta. The oil field is dated for the Yaregskay, Lyayelskay and Vezhavozhskay structures having the general extent of 36 km (drawing 1). Yarega`s oil viscosity reaches 16000 mPa*s, initial gas to oil ratio (GOR) is 10–13 m3/t, initial reservoir pressure is 1,3 MPa.

Abstract

The very neutron-rich oxygen isotopes O-25 and O-26 are investigated experimentally and theoretically. The unbound states are populated in an experiment performed at the R3B-LAND setup at GSI via proton-knockout reactions from F-26 and F-27 at relativistic energies around 442 and 414 MeV/nucleon, respectively. From the kinematically complete measurement of the decay into O-24 plus one or two neutrons, the O-25 ground-state energy and width are determined, and upper limits for the O-26 ground-state energy and lifetime are extracted. In addition, the results provide indications for an excited state in O-26 at around 4 MeV. The experimental findings are compared to theoretical shell-model calculations based on chiral two- and three-nucleon (3N) forces, including for the first time residual 3N forces, which are shown to be amplified as valence neutrons are added.

Abstract

Exotic Ni isotopes have been measured at the R3B-LAND setup at GSI in Darmstadt, using Coulomb excitation in inverse kinematics at beam energies around 500 MeV/u. As the experimental setup allows kinematically complete measurements, the excitation energy was reconstructed using the invariant mass method. The GDR and additional low-lying strength have been observed in 68Ni, the latter exhausting 4.1(1.9)% of the E1 energy-weighted sum rule. Also, the branching ratio for the non-statistical decay of the excited 68Ni nuclei was measured and amounts to 24(4)%., Comment: 11 pages, 7 figures. Abstract

5 pags., 3 figs., 1 tab. -- 11th Symposium on Nuclei in the Cosmos, 19-23 July 2010, Heidelberg, Germany, A Coulomb dissociation experiment on the proton-rich 32Ar and 34Ar isotopes was performed at the ALADIN-LAND setup at GSI in Darmstadt. Recent RQRPA calculations show a low-lying E1 soft-vibrational mode at an excitation energy Ex ≈ 9 MeV for proton-rich argon isotopes at the dripline. In a macroscopic picture, this can be understood as an out-of-phase oscillation of a thin proton skin against the isospin-saturated core, similar to the neutron pygmy resonance at the neutron dripline. On the other hand, the measured (γ, p) reactions are interesting for the calculation of reaction cross-sections and radiative proton capture rates for the rp-process. In this hydrogen burning process a lot of nuclear structure inputs are still missing. Especially in the argon region a bottleneck for the reaction flow is assumed at 30S and 34Ar. The impact of the predicted proton pygmy resonance on the reaction flow is not yet clear. The experimental motivation and the experiment itself are described. Identification plots for incoming and outgoing particles are shown and a tracking algorithm is applied and shows to work succesfully., This project is supported by the HGF Young Investigators Project VH-NG-327.

Abstract

Photo-dissociation reactions play an important role in p-process nucleosynthesis, which takes place in supernova explosions. Theoretical calculations of isotopic abundances of the p-nuclei require a vast reaction network linking thousands of isotopes, where most of the reaction rates must be derived from the Hauser-Feshbach statistical model. However, as many rates as possible need to be determined experimentally, in order to provide a reliable reference for the calculations. Measuring reaction rates on Mo isotopes is important to explain the problem of the significant underproduction of Mo and Ru in all existing models of p-process nucleosynthesis. Another aspect of the project is to verify the accuracy of the Coulomb dissociation method by comparing our data with experiments performed with real photons at S-DALINAC (TU Darmstadt) and ELBE (FZD) [1].

Abstract

A concentration of dipole strength at energies below the giant dipole resonance was observed in neutron-rich nuclei around Sn-132 in an experiment using the FRS-LAND setup. This so-called "pygmy" dipole strength can be related to the parameters of the symmetry energy and to the neutron skin thickness on the grounds of a relativistic quasiparticle random-phase approximation. Using this ansatz and the experimental findings for Sn-130 and 132 Sri, we derive a value of the symmetry energy pressure of (p) over bar (0) = 2.2 +/- 0.5 MeV/fm(3). Neutron skin thicknesses of R-n-R-p = 0.23 +/- 0.03 fm and 0.24 +/- 0.03 fm for Sn-130 and Sn-132, respectively, have been determined. Preliminary results on Ni-68 from a similar experiment using an improved setup indicate an enhanced cross section at low energies, while the results for Ni-58 are in accordance with results from photoabsorption measurements.

Abstract

The presence of a vertebral deformity increases the risk of subsequent spinal deformities. The aim of this analysis was to determine whether the presence of vertebral deformity predicts incident hip and other limb fractures. Six thousand three hundred and forty-four men and 6788 women aged 50 years and over were recruited from population registers in 31 European centers and followed prospectively for a median of 3 years. All subjects had radiographs performed at baseline and the presence of vertebral deformity was assessed using established morphometric methods. Incident limb fractures which occurred during the follow- up period were ascertained by annual postal questionnaire and confirmed by radiographs, review of medical records and personal interview. During a total of 40348 person-years of follow-up, 138 men and 391 women sustained a limb fracture. Amongst the women, after adjustment for age, prevalent vertebral deformity was a strong predictor of incident hip fracture, (rate ratio (RR) = 4.5; 95% CI 2.1-9.4) and a weak predictor of 'other' limb fractures (RR = 1.6; 95% CI 1.1-2.4), though not distal forearm fracture (RR = 1.0; 95% CI 0.6-1.6). The predictive risk increased with increasing number of prevalent deformities, particularly for subsequent hip fracture: for two or more deformities, RR = 7.2 (95% CI 3.0-17.3). Amongst men, vertebral deformity was not associated with an increased risk of incident limb fracture though there was a nonsignificant trend toward an increased risk of hip fracture with increasing number of deformities. Abstract

Almost all of the heavy elements are produced via neutron-induced processes in a multitude of stellar production sites. The remaining minor part is produced via photon- and proton-induced reactions. The predictive power of the underlying stellar models is currently limited because they contain poorly constrained physics components such as convection, rotation or magnetic fields. An important tool to determine such components is the comparison of observed with modeled abundance distributions based on improved nuclear physics input. The FRANZ facility at the Goethe University Frankfurt, which is currently under construction will provide unprecedented neutron fluxes and proton currents available for nuclear astrophysics. It will be possible to investigate important branchpoint nuclei of the s-process nucleosynthesis path and proton-induced reactions important for p-process modeling. At the GSI close to Darmstadt radioactive isotopes can be investigated in inverse kinematics. This allows experiments such as proton-induced cross section measurements using a heavy-ion storage ring or measurements of gamma-induced reactions using the Coulomb dissociation method. The future FAIR facility will allow similar experiments on very exotic nuclei, since orders of magnitude higher radioactive ions beams will be possible.

Abstract

The appearance of the pygmy-dipole-resonance is a recently observed phenomenon that can be related to neutron-matter properties. Its study can be a tool to determine the nuclear symmetry-energy parameters and thus can contribute constraining neutron star models. We present the ( γ,n ) cross sections for different Ni isotopes obtained from a measurement in inverse kinematics at about 500 MeV/u in the LAND reaction setup at GSI. The question of the disentanglement of the Coulomb and nuclear contributions is addressed.

Abstract

A pioneering experiment was recently performed at the Experimental Storage Ring (ESR) at GSI. Fully stripped ions of 96Ru were injected into the storage ring and slowed down to a few MeV per nucleon. The 97Rh ions from the 96Ru(p,γ) reaction at a newly developed hydrogen jet target were detected with Double Sided Silicon Strip Detectors (DSSSD) mounted inside a pocket. The experiment and the status of the analysis at a beam energy of 11 MeV per nucleon will be presented. © 2010 IOP Publishing Ltd.

Abstract

1. It is shown that in the whole pressure range investigated (9–180 kg/mm2) coarse-grained spherical tungsten powders surpass standard fine-grained powders in compressibility. 2. In a wide pressure range (from 9 to 100–150 kg/mm2) the compressibility of the powders investigated is satisfactorily described by M. Yu. Bal'shin's second equation. The limiting pressure below which this equation is applicable falls with increase in the relative density of powders. 3. For spherical powders the character of density, microstress, and electrical resistivity vs compaction pressure curves changes at pressures above 100 kg/mm2, which is apparently indicative of a change in the mechanism of their densification.

Abstract

1. A study was made of the effect of 0.25 wt.% lanthanum and hafnium additions on the initial recrystallization temperature of tungsten. It was established that hafnium raises the initial recrystallization temperature of tungsten from 1200 to 1300°C; alloying with lanthanum fails to raise the initial recrystallization temperature of tungsten. 2. A 0.25 wt.% addition of hafnium lowers the brittle transition temperature of tungsten from 200 to 100°C, while alloying with lanthanum lowers the brittle transition temperature of tungsten in the recrystallized condition by 100–150°C. 3. Compared with unalloyed tungsten, hafnium-containing tungsten alloys retain good mechanical properties to higher temperatures.

Abstract

First results are reported on the ground state configurations of the neutron-rich 29,30Na isotopes, obtained via Coulomb dissociation (CD) measurements. The invariant mass spectra of these nuclei have been obtained through measurement of the four-momenta of all decay products after Coulomb excitation of those nuclei on a 208Pb target at energies of 400–430 MeV/nucleon using the FRS-ALADIN-LAND setup at GSI, Darmstadt. Integrated inclusive CD cross-sections (CD) of 89 (7) mb and 167 (13) mb for one neutron removal from 29Na and 30Na, respectively, have been extracted up to an excitation energy of 10 MeV. The major part of one neutron removal, CD cross-sections of those nuclei populate the core, in its ground state. A comparison with the direct breakup model, suggests the predominant occupation of the valence neutron in the ground state of 29Na and 30Na is the d-orbital with a small contribution from the s-orbital, which are coupled with the ground state of the core. One of the major components of the ground state configurations of these nuclei are 28Na and 29Na, respectively. The ground state spin and parity of these nuclei obtained from this experiment are in agreement with earlier reported values. The spectroscopic factors for the valence neutron occupying the s and d orbitals for these nuclei in the ground state have been extracted and reported for the first time. A comparison of the experimental findings with shell model calculation using the MCSM suggests a lower limit of around 4.3 MeV of the sd–pf shell gap in 30Na. [ABSTRACT FROM AUTHOR]

Abstract

This paper presents the results of studies on the pervaporation properties (for benzene/hexane mixtures) and gas permeability (for He, H2, N2, O2, CO2, CH4, C2H6, and C4H10) of ladder-like polyphenylsesquioxanes (L-PPSQ) with improved physical and chemical properties. These polymers were obtained by condensation of cis-tetraphenylcyclotetrasiloxanetetraol in ammonia medium. The structure of L-PPSQ was fully confirmed by a combination of physicochemical analysis methods: 1H, 29Si NMR, IR spectroscopy, HPLC, powder XRD, and viscometry in solution. For the first time, a high molecular weight of the polymer (Mn = 238 kDa, Mw = 540 kDa) was achieved, which determines its improved mechanical properties and high potential for use in membrane separation. Using TGA and mechanical analysis methods, it was found that this polymer has high thermal (Td5% = 537 °C) and thermal-oxidative stability (Td5% = 587 °C) and good mechanical properties (Young’s module (E) = 1700 MPa, ultimate tensile stress (σ) = 44 MPa, elongation at break (ε) = 6%), which is important for making membranes workable under various conditions. The polymer showed a high separation factor for a mixture of 10% wt. benzene in n-hexane (126) at a benzene flow of 33 g/(m2h).

Abstract

In order to expand the range of materials that can be used in outer space and in development of small spacecraft, ladder polyphenylsilsesquioxanes with different molar weights and the Nb-siloxane composites based on them were studied. The properties of the polymer films were studied, including tests in an oxygen plasma flow. Both initial and filled ladder polymers feature extremely low erosion coefficients in the region of 10–26 cm3/atom O at a high fluence of atomic oxygen of 1.0 × 1021 atom O/cm2. Ladder polyphenylsilsesquioxane films irradiated with atomic oxygen (AO) retain their integrity, do not crack, and exhibit good optical properties, in particular, a high transmittance. The latter slightly decreases during AO exposure. The Nb-siloxane filling retains the AO resistance and slight decrease in optical transmission due to diffuse scattering on the formed Nb-[(SiO)x] nanoparticles. Ladder polyphenylsilsesquioxanes demonstrate their suitability for creating protective, optically transparent coatings for small spacecraft that are resistant to the erosive effects of incoming oxygen plasma.

Abstract

Glycerol has found its applications as a heat-transfer fluid in heat exchangers, and as a component of functional liquids in biosensor analysis. Flowing non-aqueous fluids are known to be able to induce electromagnetic fields due to the triboelectric effect. These triboelectrically generated electromagnetic fields can affect biological macromolecules. Horseradish peroxidase (HRP) is widely employed as a convenient model object for studying how external electric, magnetic, and electromagnetic fields affect enzymes. Herein, we have studied whether the flow of glycerol in a ground-shielded cylindrical coil affects the HRP enzyme incubated at a 2 cm distance near the coil’s side. Atomic force microscopy (AFM) has been employed in order to study the effect of glycerol flow on HRP at the nanoscale. An increased aggregation of HRP on mica has been observed after the incubation of the enzyme near the coil. Moreover, the enzymatic activity of HRP has also been affected. The results reported that their application can be found in biotechnology, food technology and life sciences applications, considering the development of triboelectric generators, enzyme-based biosensors and bioreactors with surface-immobilized enzymes. Our work can also be of interest for scientists studying triboelectric phenomena, representing one more step toward understanding the mechanism of the indirect action of the flow of a dielectric liquid on biological macromolecules.

Abstract

Abstract In our present paper, the influence of a pyramidal structure on physicochemical properties of a protein in buffer solution has been studied. The pyramidal structure employed herein was similar to those produced industrially for anechoic chambers. Pyramidal structures are also used as elements of biosensors. Herein, horseradish peroxidase (HRP) enzyme was used as a model protein. HRP macromolecules were adsorbed from their solution onto an atomically smooth mica substrate, and then visualized by atomic force microscopy (AFM). In parallel, the enzymatic activity of HRP was estimated by conventional spectrophotometry. Additionally, attenuated total reflection Fourier-transform infrared spectroscopy (ATR-FTIR) has been employed in order to find out whether or not the protein secondary structure changes after the incubation of its solution either near the apex of a pyramid or in the center of its base. Using AFM, we have demonstrated that the incubation of the protein solution either in the vicinity of the pyramid’s apex or in the center of its base influences the physicochemical properties of the protein macromolecules. Namely, the incubation of the HRP solution in the vicinity of the top of the pyramidal structure has been shown to lead to an increase in the efficiency of the HRP adsorption onto mica. Moreover, after the incubation of the HRP solution either near the top of the pyramid or in the center of its base, the HRP macromolecules adsorb onto the mica surface predominantly in monomeric form. At that, the enzymatic activity of HRP does not change. The results of our present study are useful to be taken into account in the development of novel biosensor devices (including those for the diagnosis of cancer in humans), in which pyramidal structures are employed as sensor, noise suppression or construction elements.

Abstract

MicroRNAs, which circulate in blood, are characterized by high diagnostic value; in biomedical research, they can be considered as candidate markers of various diseases. Mature microRNAs of glial cells and neurons can cross the blood–brain barrier and can be detected in the serum of patients with autism spectrum disorders (ASD) as components of macrovesicles, macromolecular protein and low-density lipoprotein particles. In our present study, we have proposed an approach, in which microRNAs in protein complexes can be concentrated on the surface of AFM chips with oligonucleotide molecular probes, specific against the target microRNAs. MicroRNAs, associated with the development of ASD in children, were selected as targets. The chips with immobilized molecular probes were incubated in serum samples of ASD patients and healthy volunteers. By atomic force microscopy (AFM), objects on the AFM chip surface have been revealed after incubation in the serum samples. The height of these objects amounted to 10 nm and 6 nm in the case of samples of ASD patients and healthy volunteers, respectively. MALDI-TOF-MS analysis of protein components on the chip surface allowed us to identify several cell proteins. These proteins are involved in the binding of nucleic acids (GBG10, RT24, RALYL), in the organization of proteasomes and nucleosomes (PSA4, NP1L4), and participate in the functioning of the channel of active potassium transport (KCNE5, KCNV2).

Abstract

External electromagnetic fields are known to be able to concentrate inside the construction elements of biosensors and bioreactors owing to reflection from their surface. This can lead to changes in the structure of biopolymers (such as proteins), incubated inside these elements, thus influencing their functional properties. Our present study concerned the revelation of the effect of spherical elements, commonly employed in biosensors and bioreactors, on the physicochemical properties of proteins with the example of the horseradish peroxidase (HRP) enzyme. In our experiments, a solution of HRP was incubated within a 30 cm-diameter titanium half-sphere, which was used as a model construction element. Atomic force microscopy (AFM) was employed for the single-molecule visualization of the HRP macromolecules, adsorbed from the test solution onto mica substrates in order to find out whether the incubation of the test HRP solution within the half-sphere influenced the HRP aggregation state. Attenuated total reflection Fourier transform infrared spectroscopy (ATR-FTIR) was employed in order to reveal whether the incubation of HRP solution within the half-sphere led to any changes in its secondary structure. In parallel, spectrophotometry-based estimation of the HRP enzymatic activity was performed in order to find out if the HRP active site was affected by the electromagnetic field under the conditions of our experiments. We revealed an increased aggregation of HRP after the incubation of its solution within the half-sphere in comparison with the control sample incubated far outside the half-sphere. ATR-FTIR allowed us to reveal alterations in HRP’s secondary structure. Such changes in the protein structure did not affect its active site, as was confirmed by spectrophotometry. The effect of spherical elements on a protein solution should be taken into account in the development of the optimized design of biosensors and bioreactors, intended for performing processes involving proteins in biomedicine and biotechnology, including highly sensitive biosensors intended for the diagnosis of socially significant diseases in humans (including oncology, cardiovascular diseases, etc.) at early stages.

Flow-based coiled systems, through which a heat transfer fluid (such as glycerol) is pumped, are widely used for thermal stabilization of bioreactors and biosensor cuvettes and cells. Previously, using horseradish peroxidase (HRP) as a model protein, we have demonstrated that the incubation of a protein solution in a flow-based system over coiled pipe with flowing glycerol leads to a change in the adsorption properties of the protein macromolecules. Herein, we have studied the effect of the glycerol flow on the properties of HRP, the solution of which was placed differently: i.e., near either the inflow or the outflow linear sections of the pipe, while the coiled section of the pipe was shielded with a grounded metallic cover. Atomic force microscopy (AFM) has been employed in order to visualize the HRP protein macromolecules adsorbed from its solution onto the mica substrate surface. The quantity of adsorbed protein was estimated based on the AFM data. The enzymatic activity of HRP was estimated by spectrophotometry. We demonstrate that a change in the properties of HRP enzyme was observed after the incubation of its solution near the inflow/outflow linear sections of the pipe with flowing glycerol. Namely, after the incubation of HRP solution near the inflow section, a decrease in the protein adsorption onto mica was observed, but its enzymatic activity remained unchanged in comparison to the control sample. In another case, when the HRP solution was incubated near the outflow section, an increased protein adsorption was observed, while the enzyme exhibited considerably lower activity.
