Objectives: Diabetes mellitus is one of the most frequent stroke-related comorbid states, and it is characterized by accumulation of reactive carbonyl compounds (RCOs), leading to "carbonyl stress". This pilot study was aimed to evaluate the effect of the consumption of the nutritional supplement ALAnerv® on some serum carbonyl stress markers, as well as on the activity of erythrocytes' glyoxalases in post-acute stroke patients undergoing rehabilitation. Material and Methods: We created a study population of 28 patients, organized into (-) ALA and (+) ALA groups. Patients from (+) ALA group received ALAnerv® for two weeks (2 pills/day). All the subjects followed the same rehabilitation program. In both groups, blood samples were taken at the hospitalization and at the discharge moments, respectively. On these samples we assessed lactic acid, fructosamine and RCOs concentrations, as well as the activities of glyoxalases 1 and 2 from erythrocytes' lysates. Outcomes: In (-) ALA group the concentrations of fructosamine and RCOs significantly increased (0.90 ± 0.04 vs. 1.02 ± 0.04, p = 0.020; 0.19 ± 0.03 vs. 0.28 ± 0.07, p = 0.027) during the study period. Also, glyoxalase 2 activity decreased in this group (27.04 ± 6.10 vs. 14.43 ± 3.02, p = 0.027). In (+) ALA group, the variation of these parameters did not reach statistical significance. Only, the activity of Glo1, which catalyzes the rate-limiting step in the glyoxalase pathway, had an increasing trend in (+) ALA group. The percentage of variation of fructosamine between (-) ALA and (+) ALA groups reached statistical significance (14.8 ± 5.2 vs. - 1.0 ± 13.3, p = 0.047). Regression analysis indicated that the activity of glyoxalase 2 was significantly influenced by the treatment with ALAnerv® (p < 0.001), while the concentration of RCOs was significantly influenced by diabetes mellitus (p = 0.030). Conclusions: Our preliminary results suggest that ALAnerv® could be useful for the correction of the carbonyl stress status in post-acute stroke patients with diabetes. Gleditsia triacanthos is an aggressive invasive species in Eastern Europe, producing a significant number of pods that could represent an inexhaustible resource of raw material for various applications. The aim of this study was to extract cellulose from the Gleditsia triacanthos pods, characterize it by spectrophotometric and UHPLC–DAD-ESI/MS analysis, and use it to fabricate a wound dressing that is multi-functionalized with phenolic compounds extracted from the leaves of the same species. The obtained cellulose microfibers (CM) were functionalized, lyophilized, and characterized by ATR-FTIR and SEM. The water absorption and retention capacity as well as the controlled release of phenolic compounds with antioxidant properties evaluated in temporal dynamics were also determined. The antimicrobial activity against reference and clinical multi-drug-resistant Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, Acinetobacter baumannii, Enterobacter cloacae, Candida albicans, and Candida parapsilosis strains occurred immediately after the contact with the tested materials and was maintained for 24 h for all tested microbial strains. In conclusion, the multi-functionalized cellulose microfibers (MFCM) obtained from the reproductive organs of an invasive species can represent a promising alternative for the development of functional wound dressings with antioxidant and antimicrobial activity, as well as being a scalable example for designing cost-effective, circular bio-economy approaches to combat the accelerated spread of invasive species. Cerebrovascular accidents are currently the second major cause of death and the third leading cause of disability in the world, according to the World Health Organization (WHO), which has provided protocols for stroke prevention. Although there is a multitude of studies on the health benefits associated with anthocyanin (ACN) consumption, there is no a rigorous systematization of the data linking dietary ACN with stroke prevention. This review is intended to present data from epidemiological, in vitro, in vivo, and clinical studies dealing with the stroke related to ACN-rich diets or ACN supplements, along with possible mechanisms of action revealed by pharmacokinetic studies, including ACN passage through the blood-brain barrier (BBB). Objectives: Stroke is a pathological condition associated with a redox imbalance. Both the acute and the post-acute phases after a stroke are characterized by a pro-oxidant state, which could be corrected through antioxidant supplementation. The aim of the present study was to evaluate the effect of the ALAnerv® upon the redox status of erythrocytes. Material and Methods: For this pilot study were enrolled 28 post-acute stroke patients. They were randomly divided into a control group [(-) ALA] and a study group [(+) ALA]. Patients were hospitalized for a period of two weeks. Blood samples were taken at the beginning and at the end of this period. Patients from (+) ALA group received the nutritional supplement ALAnerv®. Catalase, SOD, GPx, GRed and GT activities were assessed on erythrocytes' lysates. Also, the total antioxidant capacity as well as the concentration of total thiols were evaluated. Outcomes: During the study period only SOD (1393.48 ± 69.14 U/g Hb vs. 1514.06 ± 60.80) and GRed (4.47 ± 0.37 U/g Hb vs. 5.06 ± 0.24) significantly increased in (+) ALA group. Catalase (173.70 ± 14.62 k/g Hb vs. 123.41 ± 8.71) and GT (5.55 ± 0.26 U/g Hb vs. 4.95 ± 0.23) activities significantly decreased during the study period. Multiple regression indicated that only SOD, GT and GRed activities were influenced by the ALAnerv® consumption. There was no significant increase if the total thiols concentration. Conclusions: ALAnerv® consumption could be associated with a correction of the erythrocytes' redox status. There is a need to investigate if administration of ALAnerv® over a longer time period could have a more significant influence on the erythrocytes' antioxidant system. [ABSTRACT FROM AUTHOR]

The hydrodistilled essential oils and volatile compounds (by static headspaces technique) of Mentha piperita L. and M spicata L. were characterized by GC-MS. Headspace analysis of Mentha piperita revealed the existence of menthone (25.4%), 1,8-cineole (17.7%) and menthol (12.1%) as the main components, while the essential oil contained high amounts of menthol (46.8%) and menthone (25.6%). By contrast, headspace analysis of M. spicata showed a high content of limonene (37.0%) together with carvone (13.0%), β-pinene (10.4%) and α-pinene (9.8%), while the essential oil was reach in carvone (51.7%), dihydrocarveol (11.5%) and cis-dihydrocarvone (9.1%). Eleven samples of peppermint tea available on the Romanian market were analysed by headspace GC-MS. The volatile profile of the tea samples was compared with that of Mentha piperita L. and certain differences were emphasized and discussed.

In vitro antimicrobial and antioxidant activities of Amaranthus retroflexus leaves and inflorescence alcoholic (ethanol 70%) extracts of various concentrations ranging from 0.78 to 400 μL/ml were analyzed on different clinical and reference bacterial strains (Staphylococcus aureus, Bacillus subtills, Enterococcus faecalis, Klebsiella pneumoniae, Escherichia coli, Pseudomonas aeruginosa, Acinetobacter baumannii) and fungal strains (Candida albicans, C. famata, C. utilis, Saccharomyces cerevisiae) using agar disk diffusion method and broth dilution method (MIC determination) for antimicrobial activity and TEAC (Trolox capacity) assay for the evaluation of the antioxidant activity. The adapted diffusion method was used to test the antimicrobial effect of the extracts separately and in combination with a range of standard antibiotics, in order to evidence any synergic effects of A. retroflexus extracts on the antibiotics activity. The extracts showed the highest inhibitory effect against K. pneumoniae and B. subtilis with no activity against S. aureus among the bacterial strains, while in case of the fungal strains the most intensive effect was exhibited against C. famata by both extracts. The A. retroflexus leaves extract improved the ciprofloxacin and ticarcillin-clavulanic acid activity towards the P. aeruginosa clinical strain. The inflorescences extract significantly increased chloramphenicol activity on B. subtilis strain. The antioxidant activity assay showed that the studied extracts exhibited the ability to neutralize the free radicals leading to the conclusion that the tested extracts bear compounds with a broad spectrum of antimicrobial and antioxidant activity that could represent a potential alternative for treating various infectious diseases.

The aim of present study was to investigate the microbial colonization of worn contact lenses (CLs) and to evaluate the inhibitory effect of pomelo (Citrus maxima) peels essential oil on the biofilm development on unworn CLs. The essential oil was isolated by steam distillation and analyzed by gas chromatography coupled with mass spectrometry, twenty compounds being isolated. The antimicrobial activity of pomelo oil was tested against S. epidermidis and P. aeruginosa strains, known for their ability to develop biofilms on prosthetic devices, by qualitative screening methods and quantitative assay of the minimal inhibitory concentrations (MIC) in order to evaluate the antibiofilm activity. Our study revealed that all worn CLs where 100% colonized by staphylococci and Enterobacteriaceae strains. The pomelo essential oil inhibited the development of bacterial biofilms formed by Gram-positive and Gram-negative microorganisms on soft CLs, its antibiofilm activity being specific and dependent on different physical parameters (contact time and temperature). The architecture of bacterial biofilms developed on soft contact lenses was analyzed using confocal scanning laser microscopy (CSLM).

The vegetal extracts are used as an ecological alternative to classical anti-infectious treatments based on antibiotics, exhibiting the advantage of reduced secondary effects. Most of these compounds are secondary metabolites, especially aromatic substances synthesized by plants in a reduced concentration. The aim of this study was to investigate the influence of usnic acid against quorum sensing and response mechanisms involved in the initiation and development of the dental plaque biofilm and its tolerance to antimicrobials. Three samples of super-gingival dental plaque were treated for different time intervals with usnic acid at 200 ig/ml in dimethyl-sulfoxide, representing the MIC value. Each dental plaque sample was inoculated in Brain Heart Infusion medium to establish the microbial growing curve by viable cells counts using the tenfold microdilutions method. For strains identification there were used the microtest galleries: API 20Strep, API Staph, API 20NE, API 20E. MIC value for usnic acid was determined by twofold microdilution technique. Usnic acid selectively inhibited the biofilm development by Gram positive bacteria and the expression of haemolytic properties of strains isolated from the dental plaque. The growth curve of the isolated strains was affected by usnic acid, the changes consisting of the lag phase extension to 6-10 h (this time interval being considered as the persistence time of antimicrobial activity) and the significant decrease of the viable cell number and consecutively, the prolongation of the generation time. These effects are demonstrating the interference of the usnic acid with the intra- and interspecies signalling mechanisms based on quorum sensing and response and dependent on cell density, giving the possibility to use them as an active principle in some new pharmaceutical formula intended for the prevention and treatment of gingival and periodontal pathologies.

Regulatory constraints and environmental and human health concerns have promoted the search for alternative bio-control strategies of fire blight, a destructive disease of rosaceous plants which produces serious losses in apple and pear orchards all over the world. The aim of this study was to establish the antimicrobial activity of Citrus maxima essential oil against Erwinia amylovora. An agar diffusion method was used for the screening of the inhibitory effect of Citrus maxima essential oil on bacterial strains growth. The quantitative inhibitory effect of pomelo oil on in vitro biofilm development was established by a microtiter colorimetric assay. In order to investigate the ability of pomelo oil to interfere with bacterial adherence and subsequent biofilm development on leaves obtained from different pomaceous fruit trees species and cultivars: Pyrus (Napoca, Williams), Malus (Golden Delicious) and Cydonia (Aromate), leaves were immersed in pomelo oil for 1, 2, 3, 5 and 10 minutes before exposing them to bacterial colonization. The architecture of bacterial biofilms developed on leaf surface was analyzed using Confocal Scanning Laser Microscopy (CSLM). Our results showed that Citrus maxima essential oil inhibited the development of bacterial biofilms on leaves, pomelo oil being more active on Cydonia (Aromate) leaves when the leaves were treated for 5 minutes. The results obtained from this study may contribute to the development of new bio-control agents as alternative strategies to protect fruit trees from fire blight disease.

In order to survive in changing environments, bacteria possess enormous adaptive capabilities that allow them to modulate their behavior and reprogram gene expression in response to environmental cues. Vibrios are inhabitants of estuarine and fresh waters and some species are pathogenic to humans, and marine vertebrates and invertebrates. Surface attachment is believed to be essential for colonization of all of these natural environments. Vibrio (V.) parahaemolyticus, an ubiquitous marine bacterium and human pathogen, seems to be particularly adapted to growth on surfaces or in biofilms. In response to its physical environment, V. parahaemolyticus induces the expression of a large number of genes. The purpose of the present study was to investigate the influence of different physicochemical parameters (temperature and osmolarity) on the virulence factors expression in Vibrio strains using different conditions simulating environmental stress factors. Some of the tested strains displayed a decreased adherence capacity to the inert substrate under stressful conditions, and the adherence capacity on HeLa cell was generally reduced, while the soluble enzymatic factors showed only slight changes. However, it is to be noticed that the haemolysins and Kanagawa enterotoxin were better expressed at higher temperature and osmolarity, these factors probably contributing to the bacterial adaptation and survival in the extern medium of certain Vibrio species.
