No. of bitstreams: 0 Previous issue date: 2009-07-01 Ten estrous cycles of seven Machador Brasileiro jennies located at Estacao Experimental de Zootecnia - Colina - Sao Paulo were monitored and the plasmatic progesterone concentration determined by the radioimmunoassay method in solid phase, employing commercial kits (DPC). For progesterone concentration determination and CL diameter there were excluded the animals which presented diestrus ovulations. The corpora lutea formed from single ovulations or double ovulations presented a mean diameter of 26.2 +/- 4.4 and 22.1 +/- 2.7 mm, respectively, which represented 66.1% and 64.1% of the preovulatory follicle diameter. The presence of two CLs from double ovulations was not reflected by a significant increase on progesterone concentrations. No. of bitstreams: 0 Previous issue date: 2009-07-01 The ovarian activity was accessed by ultrasound throughout 10 estrous cycles from Jennies (Marchador Brasileira). Daily ultrasound examinations were performed and the dada was retrospectively studied based on daily identification of each follicle detected. Blood samples were collected every 24 hours from ovulation (D0) until the next identified ovulation. The follicles measuring 11mm were detected and their mean diameter was registered daily using an ovarian map and permitting a retrospective evaluation of the dada, which represented the follicular growth dynamics. One and two major follicular waves were detected in six (60%) and four (40%) cycles respectively from ten estrous cycles in the present study. The primary wave emergency and follicle deviation from the cycles observed during the present study with one major follicular wave occurred at day 10.2 +/- 0.75 and at day14.1 +/- 0.81 and for cycles with two major waves, those events occurred at 9.0 +/- 2.0 and 14.0 +/- 1.15 days after ovulation. The maximum diameter of the dominant follicle at the cycles with one and two follicular waves were 37.2 +/- 3.35 mm and 37.3 +/- 1.1, respectively. The mean intervals from two ovulations were 23.0 +/- 1.79 and 22.3 +/- 1.26 days when observed in cycles with one and two major follicular waves. The maximum diameter of the dominant follicle was slightly smaller when double ovulations were observed if compared with the single ovulations (P > 0.05). It can be concluded that wave emergency period is observed around 9.6 days of the estrous cycle and the occurrence of deviation can be observed on day 14 of the estrous cycle. Abstract

The effect of prostaglandin (PGF2α) vs CIDR and eCG (equine corionic gonadotrophin) on dynamic of foUicular population and its relationship with P4plasma concentrations were investigated in cyclic ewes. Fourteen Bergamascia ewes were used, Group 1 was synchronized using two i.m. injections of PGF2α, and Group 2 treated for 14 d with CIDR and 500IU of eCG at dispositive withdrawal on day 14. Ovarian foUicular dynamics were ultrasonically monitored. All ovarian follicles ≥ 2 mm were measured, and their relative locations were recorded on an ovarian map in order to follow the sequential development of each individual follicle. Blood samples for P4determination were collected daily starting the day before the second injection of PGF2α, (Gl) and from administration of eCG (G2) until day 10 after ovulation of the cycle. Abstract

Summary: The use of frozen semen for artificial insemination is the main approach utilised for the genetic improvement of most domesticated species. The advantages include lower transportation costs, continuous availability of semen, fewer occurrences of sexually transmitted diseases and the incorporation of desirable genes in a relatively short amount of time. Nevertheless, the use of frozen semen in buffalo herds remains limited due to the loss of sperm quality when buffalo semen is frozen. So, the goal of this study was to evaluate the pre‐ and post‐cryopreservation quality of buffalo semen diluted in three distinct freezing media: Tris‐egg yolk, Botu‐bov® (BB) and ACP‐111®. Thirty‐two ejaculates from four bulls were analysed in terms of kinetics, morphology and sperm viability by epifluorescence microscope. Thawed samples were also evaluated for capacitation‐like damage, DNA fragmentation and plasma and acrosomal membrane integrity using flow cytometry. The Tris‐egg yolk and BB® extenders yielded better results than the ACP‐111® extender for kinetics parameter (total motility, progressive motility and percentage of rapid cells). However, semen samples were similar for parameters evaluated by flow cytometry. Taken together, the data indicate that in comparison with Tris‐egg yolk and BB extender, ACP‐111® can also be used as an extender for buffalo semen cryopreservation. [ABSTRACT FROM AUTHOR]

63. Effects of selenium supplementation on serum cortisol in repeatedly handled cattle

Abstract

This study evaluated the relationship between Se supplementation and serum cortisol in repeatedly handled cattle. Sixty Nellore calves were randomly distributed into four experimental groups, which were fed a mineral-protein mixture added with Se to achieve individual daily supplementation of 0 (Gc), 3.6 (G 3.6 ), 5.4 (G 5.4 ) and 6.4 (G 6.4 ) mg Se. Stressful handling procedures and blood sampling were performed on days 0, 15, 30, 60, 90 and 120. Serum cortisol increased until day 90 and decreased on day 120, irrespective of Se supplementation. This finding shows that cattle reached a stress state but adapted. Cortisol and Se levels were not markedly correlated. Serum Se increased over the course of the experiment in the supplemented groups. However, because serum Se decreased in Gc, it may have mobilized to form the selenoproteins needed to remove metabolic wastes from handling-related oxidative stress., Este estudo avaliou a relação entre suple- mentação com Se e cortisol sérico em gado mane- jado repetidamente. Sessenta bezerros Nelore foram distribuídos em quatro grupos, os quais se alimentaram de uma mistura mineral protéica acrescida de Se de modo a alcançar suple- mentação individual diária de 0 (Gc), 3.6 (G 3.6 ), 5.4 (G 5.4 ) or 6.4 (G 6.4 ) mg Se. O estresse de manejo e as colheitas das amostras de sangue foram nos dias 0, 15, 30, 60, 90 e 120. O cortisol sérico aumentou até o dia 90 e reduziu no dia 120, independente da suplementação com Se. Isso indica que os bovinos ficaram estressados, mas se adaptaram ao longo do tempo. O cortisol e Se sérico não foram marcantemente correlacionados. A concentração de Se sérico aumentou ao longo do experimento nos grupos suplementados. En- tretanto, como o Se sérico foi reduzido em Gc, deve-se considerar que ele pode ter sido mobilizado para formação de selenoproteínas necessárias à remoção dos restos metabólicos resultantes do estresse oxidativo desencadeado pelo estresse de manejo.

65. Correlações entre morfometria ovariana e hormônios em vacas Zebus não prenhes

Abstract

The study of ovarian morphometry is directly related to its practical applications to interpret the examination of the reproductive tract of the cow. The objective of this work was to study the ovarian morphometry collected in slaughterhouse and correlations between corpus luteum area, progesterone and cortisol in no pregnant zebu cows. One hundred fourteen pairs of ovaries were collected from slaughterhouse, measured in thickness, length, width and volume, diameter and volume of the follicle, diameter and area of the corpus luteum. Significative difference (p<0.05) were observed for widths of (1.95 cm and 1.83 cm) and volume (p<0.05) of 7.26 ml and 6.23 ml of left and right ovaries respectively. For size and volume of follicles, and diameter and area of the corpus luteum, there was no significative difference between the sides. There was correlation of 0.46 (p<0.01) between left ovary volume x luteum corpus area, in the presence of follicles with diameters equal or greater than 9 mm, the corpus luteum of the massive and protuberant type were present in 23 (43.39%) of 53 ovaries, predominating in relation to type of massive and incluse 16 (30.18%). There were no significative correlation between corpus luteum area x progesterone, corpus luteum area x cortisol and progesterone x cortisol. In conclusion, the presence of the included corpus luteum in zebu cows could result in error during the rectal palpation exam to estimate the ovarian activity., O estudo da morfometria ovariana está diretamente ligado as suas aplicações práticas para realizar e interpretar os achados dos exames ginecológicos em vacas. O objetivo desse trabalho foi estudar a morfometria de ovários coletados em abatedouro e as correlações entre área do corpo lúteo, progesterona e cortisol em vacas zebu não prenhes. Foram coletados 114 pares de ovários em abatedouro, mensurados quanto a sua espessura, comprimento, largura e volume; diâmetro e volume do folículo, diâmetro e área do corpo lúteo, e concentrações de progesterona e cortisol. Foi observada diferença significativa (p<0,05) para largura 1,95 cm e 1,83 cm; e volume (p<0,05) 7,26 ml e 6,23 ml do ovário esquerdo e direito, respectivamente. Houve correlação de 0,46 (p<0,01) entre o volume do ovário esquerdo x área do corpo lúteo. Na presença de folículos com diâmetro igual ou superior a 9 mm, o corpo lúteo do tipo maciço e protruso presente em 23 (43,39%) dos 53 ovários, predominou em relação ao tipo maciço e incluso 16 (30,18%) dos ovários. Dos 84 ovários com corpos lúteos 26,20% eram do tipo incluso. Não houve correlação significativa entre área do corpo lúteo x progesterona, área do corpo lúteo x cortisol e progesterona x cortisol. Conclui-se que a presença de corpos lúteos inclusos, nas vacas zebu, pode resultar em falha durante o exame de palpação retal para estimar a atividade ovariana.

Abstract

Contents The present work aimed to estimate heritability and genetic correlations of reproductive features of Nellore bulls, offspring of mothers classified as superprecocious (M1), precocious (M2) and normal (M3). Twenty one thousand hundred and eighty-six animals with average age of 21.29 months were used, evaluated through the breeding soundness evaluation from 1999 to 2008. The breeding soundness features included physical semen evaluation (progressive sperm motility and sperm vigour), semen morphology (major, minor and total sperm defects), scrotal circumference (SC), testicular volume (TV) and SC at 18 months of age (SC18). The components of variance, heritability and genetic correlations for and between the features were estimated simultaneously by restricted maximum likelihood, with the use of the vce software system vs 6. The heritability estimates were high for SC18, SC and TV (0.43, 0.63 and 0.54; 0.45, 0.45 and 0.44; 0.42, 0.45 and 0.41, respectively for the categories of mothers M1, M2 and M3) and low for physical and morphological semen aspects. The genetic correlations between SC18 and SC were high, as well as between these variables with TV. High and positive genetic correlations were recorded among SC18, SC and TV with the physical aspects of the semen, although no favourable association was verified with the morphological aspects, for the three categories of mothers. It can be concluded that the mother's sexual precocity did not affect the heritability of their offspring reproduction features. [ABSTRACT FROM AUTHOR]

Abstract

Ovaries of 86 buffaloes and 95 cows were collected from slaughterhouses and transported to the laboratory in saline solution at 36°C. The Cumulus-oocyte complexes (COCs) were recovered by follicular aspiration and only grades I and II COCs were selected and matured in TCM 199 supplemented with 10% fetal calf serum, sodium pyruvate, LH, FSH, estradiol, gentamicin and cysteamin, for 22-24 hours. A total of 714 and 1983 COCs were recovered from buffaloes and cows, respectively. In the buffaloes, the recovery rates of each COCs categories (grade I: 25.9%; grade II: 30.7%; grade III: 10.2%; denuded 18.6% and expanded: 14.6%) were lower than cows (31.8; 30.6; 15.4; 4.5; 17.7%, respectively) according to Mann-Whitney Test (p< 0.05). The percentage of bubaline oocytes that reached metaphase II (63.4% - 242/ 396) was lower than bovine oocytes (67.8% - 696/ 1234) under the same laboratory conditions. These differences observed in all analysis realized indicate that each species has peculiar phisiological characteristics. [ABSTRACT FROM AUTHOR]

100. Review of the literature on corpus luteum insufficiency in women (2015–2020) and in domestic animals (1980–2020)

Abstract

The corpus luteum (CL) is a transitory endocrine structure found in female mammals which secretes progesterone (P4), rendering the endometrium secretory and thus facilitating embryo implantation in the uterus. CL insufficiency (CLI) is defined as a condition in which CL P4 cannot maintain the endometrium and gestation, causing infertility and pregnancy loss. The present study aimed to compare specific aspects related to CLI in humans and domestic animals to identify its main aetiological differences and those related to treatment of the condition in different species.
