To maintain energy homeostasis during cold exposure, the increased energy demands of thermogenesis must be counterbalanced by increased energy intake. To investigate the neurobiological mechanisms underlying this cold-induced hyperphagia, we asked whether agouti-related peptide (AgRP) neurons are activated when animals are placed in a cold environment and, if so, whether this response is required for the associated hyperphagia. We report that AgRP neuron activation occurs rapidly upon acute cold exposure, as do increases of both energy expenditure and energy intake, suggesting the mere perception of cold is sufficient to engage each of these responses. We further report that silencing of AgRP neurons selectively blocks the effect of cold exposure to increase food intake but has no effect on energy expenditure. Together, these findings establish a physiologically important role for AgRP neurons in the hyperphagic response to cold exposure.

Background: Sarcoidosis is a systemic inflammatory disorder and can often affect any other organs beyond the heart. Whole-body 18F-fluorodeoxyglucose positron emission tomography (FDG-PET) is used to detect not only cardiac but also extra-cardiac involvement of sarcoidosis. However, the features and clinical impact of extra-cardiac lesions have not yet been fully elucidated. Therefore, this study aimed to clarify these using FDG-PET. Methods and results: We enrolled 120 consecutive patients with abnormal findings clinically suggesting cardiac sarcoidosis who underwent whole-body FDG-PET. In this study, a patient with suspected cardiac sarcoidosis was defined as one having both clinically suspected findings and FDG-PET positive cardiac uptake. Subsequently, a total of 36 patients with suspected cardiac sarcoidosis were found and analyzed. Extra-cardiac involvement was detected in 35 lesions of 14 patients (39% per patient). In particular, the extra-cardiac lesions were widely distributed throughout the body, and mediastinal/hilar lymph node involvement was most commonly observed. In most of the patients (93% per patient, 13/14), the extra-cardiac lesions were localized in the regions that were considered more accessible with less risk of complication compared with endomyocardial biopsy (EMB). Based on the FDG-PET findings, 8 patients underwent extra-cardiac biopsy without complication, and its diagnostic sensitivity for histological sarcoidosis was high (75%, 6/8). Moreover, FDG-PET-guided extra-cardiac biopsy could confirm histological sarcoidosis in 4 lesions that EMB failed to prove. Conclusions: Extra-cardiac involvement in patients with suspected cardiac sarcoidosis was relatively high. FDG-PET-guided extra-cardiac biopsy may be safe and useful for the imaging based diagnosis of cardiac sarcoidosis.

Androgen deprivation in men leads to increased adiposity, but the mechanisms underlying androgen regulation of fat mass have not been fully defined. Androgen receptor (AR) is expressed in monocytes/macrophages, which are resident in key metabolic tissues and influence energy metabolism in surrounding cells. Male mice bearing a cell-specific knockout of the AR in monocytes/macrophages (M-ARKO) were generated to determine whether selective loss of androgen signaling in these cells would lead to altered body composition. Wild-type (WT) and M-ARKO mice (12–22 weeks of age, n = 12 per group) were maintained on a regular chow diet for 8 weeks and then switched to a high-fat diet for 8 additional weeks. At baseline and on both the regular chow and high-fat diets, no differences in lean mass or fat mass were observed between groups. Consistent with the absence of differential body weight or adiposity, no differences in food intake (3.0 ± 0.5 g per day for WT mice vs 2.8 ± 0.4 g per day for M-ARKO mice) or total energy expenditure (0.6 ± 0.1 Kcal h−1 for WT mice vs 0.5 ± 0.1 Kcal h−1 for M-ARKO mice) were evident between groups during high-fat feeding. Liver weight was greater in M-ARKO than that in WT mice (1.5 ± 0.1 g vs 1.3 ± 0.0 g, respectively, P = 0.02). Finally, M-ARKO mice did not exhibit impairments in glucose tolerance or insulin sensitivity relative to WT mice at any study time point. In aggregate, these findings suggest that AR signaling specifically in monocytes/macrophages does not contribute to the regulation of systemic energy balance, adiposity, or insulin sensitivity in male mice.

Regulating the frequencies of power grids by controlling electric vehicle charging and discharging, known as vehicle-to-grid (V2G) ancillary services, is a promising and profitable means of providing flexibility that integrates variable renewable energy (VRE) into traditional power systems. However, the ancillary services market is a niche, and the scale, saturation, and time-dependency are unclear when assuming future changes in the power system structure. We studied the marginal value of V2G ancillary services as a balancing capacity of the power system operation on the load-frequency control (LFC) timescale and evaluated the reasonable maximum capacity of the LFC provided by V2G. As a case study, we assumed that the Japanese power system would be used under various VRE penetration scenarios and considered the limited availability time of V2G, based on the daily commuter cycle. The power system operation was modeled by considering pumped storage, interconnection lines, and thermal power–partial load operations. The results show that the marginal value of V2G was greater during the daytime than overnight, and the maximum cost saving (USD 705.6/EV/year) occurred during the daytime under the high-VRE scenario. Improving the value and size of V2G ancillary services required coordination with energy storage and excess VRE generation.

Normal aging in humans and rodents is accompanied by a progressive increase in adiposity. To investigate the role of hypothalamic neuronal circuits in this process, we used a Cre-lox strategy to create mice with specific and progressive degeneration of hypothalamic neurons that express agouti-related protein (Agrp) or proopiomelanocortin (Pomc), neuropeptides that promote positive or negative energy balance, respectively, through their opposing effects on melanocortin receptor signaling. In previous studies, Pomc mutant mice became obese, but Agrp mutant mice were surprisingly normal, suggesting potential compensation by neuronal circuits or genetic redundancy. Here we find that Pomc-ablation mice develop obesity similar to that described for Pomc knockout mice, but also exhibit defects in compensatory hyperphagia similar to what occurs during normal aging. Agrp-ablation female mice exhibit reduced adiposity with normal compensatory hyperphagia, while animals ablated for both Pomc and Agrp neurons exhibit an additive interaction phenotype. These findings provide new insight into the roles of hypothalamic neurons in energy balance regulation, and provide a model for understanding defects in human energy balance associated with neurodegeneration and aging.

Wind power has been increasingly deployed in the last decade to decarbonize the electricity sector. Wind power output changes intermittently depending on weather conditions. In electrical power systems with high shares of variable renewable energy sources, such as wind power, system operators aim to respond flexibly to fluctuations in output. Here, we investigated very short-term fluctuations, short-term fluctuations (STFs), and long-term fluctuations (LTFs) in wind power output by analyzing historical output data for two northern and one southern balancing areas in Japan. We found a relationship between STFs and the average LTFs. The percentiles of the STFs in each month are approximated by linear functions of the monthly average LTFs. Furthermore, the absolute value of the slope of this function decreases with wind power capacity in the balancing area. The LTFs reflect the trend in wind power output. The results indicate that the flexibility required for power systems can be estimated based on wind power predictions. This finding could facilitate the design of the balancing market in Japan.

This case report describes sustained monomorphic ventricular tachycardia (VT) caused by a large epicardial scar, related to dilated-phase hypertrophic cardiomyopathy mimicking VT originating from the apical septum. VT resolved with epicardial catheter ablation. The exit of the VT circuit suggested that a 12-lead electrocardiogram can be remote with respect to the critical isthmus in this case. In patients with structural heart disease, it is difficult to identify the VT reentrant circuit by surface electrocardiography, which shows only the exit site. VT originating in the epicardium should be considered, even if the suspected origin is another ventricular site.

The increase in the number of electric vehicles (EVs) has led to increased global expectations that the application of this technology may result in the reduction of CO2 emissions through the replacement of conventional petrol vehicles and ensure the flexibility of power systems such as batteries. In this paper, we propose a residential demand response (DR) evaluation model that considers the degradation mechanism of the EV battery and examines the effective battery operation. We adopted the already-proposed NiMnCo battery degradation model to develop an EV DR evaluation model. In this model, the battery operation is optimized to minimize the electricity and degradation costs affected by ambient temperature, battery state of charge (SOC), and depth of discharge. In this study, we evaluated the impact of the relevant parameters on the economics of the DR of EV batteries for 10 all-electric detached houses with photovoltaic system assuming multiple EV driving patterns and battery (dis)charging constraints. The results indicated that the degradation costs are greatly affected by the SOC condition. If a low SOC can be managed with a DR strategy, the total cost can be reduced. This is because the sum of the reduction of purchased cost from the utility and calendar degradation costs are higher than the increase of the cycle degradation cost. In addition, an analysis was conducted considering different driving patterns. The results showed that the cost reduction was highest when a driving pattern was employed in which the mileage was low and the staying at home time was large. When degradation costs are included, the value of optimized charging and discharging operations is more apparent than when degradation costs are not considered.

Purpose. To compare the morphology of two types of vortex keratopathy: amiodarone-induced keratopathy and the Fabry disease-associated keratopathy. Patients and Methods. Eight patients who were receiving oral amiodarone therapy and 3 patients with Fabry disease, a mother and her 2 daughters, were examined by slit-lamp biomicroscopy and in vivo confocal microscopy (IVCM) regularly. Results. Amiodarone-induced keratopathy developed in 7 of the 8 patients, and it was detected as early as 7 days by IVCM and 14 days by slit-lamp biomicroscopy. The in vivo confocal microscopic images showed a clustering of corneal epithelial cells with a highly reflective cytoplasm in both types of keratopathy. In the amiodarone-induced keratopathy, the highly reflective epithelial cells were first found at the center of the cornea and then spread to the periphery with increasing time on amiodarone. In Fabry disease, the highly reflective epithelial cells were consistently observed extending from the limbus to the central cornea. Conclusion. These findings suggest that the corneal epithelial cells most likely endocytose amiodarone from the tear film in the amiodarone-induced keratopathy. In Fabry disease, globotriaosylceramide deposits are taken up by the lysosomes of the limbal epithelial stem cells, and they differentiate and migrate to the center of the cornea to form the whorl pattern.
