The calcium-transporting ATPase ATP2A2, also known as SERCA2a, is a critical ATPase responsible for [Ca.sup.2+] re-uptake during excitation-contraction coupling. Impaired [Ca.sup.2+] uptake resulting from decreased expression and reduced activity of SERCA2a is a hallmark of heart failure (1). Accordingly, restoration of SERCA2a expression by gene transfer has proved to be effective in improving cardiac function in heart-failure patients (2), as well as in animal models (3). The small ubiquitin-related modifier (SUMO) can be conjugated to lysine residues of target proteins (4), and is involved in many cellular processes (5). Here we show that SERCA2a is SUMOylated at lysines 480 and 585 and that this SUMOylation is essential for preserving SERCA2a ATPase activity and stability in mouse and human cells. The levels of SUMO1 and the SUMOylation of SERCA2a itself were greatly reduced in failing hearts. SUMO1 restitution by adeno-associated-virus-mediated gene delivery maintained the protein abundance of SERCA2a and markedly improved cardiac function in mice with heart failure. This effect was comparable to SERCA2Agene delivery. Moreover, SUMO1 overexpression in isolated cardiomyocytes augmented contractility and accelerated [Ca.sup.2+] decay. Transgene-mediated SUMO1 overexpression rescued cardiac dysfunction induced by pressure overload concomitantly with increased SERCA2a function. By contrast, downregulation of SUMO1 using small hairpin RNA (shRNA) accelerated pressure-overload-induced deterioration of cardiac function and was accompanied by decreased SERCA2a function. However, knockdown of SERCA2a resulted in severe contractile dysfunction both in vitroand in vivo, which was not rescued by overexpression of SUMO1. Oxidative stress, caused by the accumulation of reactive oxygen species (ROS) during acute myocardial infarction (AMI), is one of the main factors leading to myocardial cell damage and programmed cell death. Phosphatidylinositol-3-kinase-AKT (PI3K-AKT) signaling is essential for regulating cell proliferation, differentiation, and apoptosis. Phosphoinositide-3-kinase (PI3K)-interacting protein 1 (PIK3IP1) is an intrinsic inhibitor of PI3K in various tissues, but its functional role during AMI remains unknown. In this study, the anti-ischemic role of PIK3IP1 in an in vitro AMI setting was evaluated using H9c2 cells. The MTT assay demonstrated that cell viability decreased significantly via treatment with H
2 O2 (200–500 μM). The TUNEL assay results revealed substantial cellular apoptosis following treatment with 200 μM H2 O2 . Under the same conditions, the expression levels of hypoxia-inducible factor (HIF-1α), endothelin-1 (ET-1), bcl-2-like protein 4 (BAX), and cleaved caspase-3 were elevated, whereas those of PIK3IP1, LC3II, p53, and Bcl-2 decreased significantly. PIK3IP1 overexpression inhibited H2 O2 -induced and PI3K-mediated apoptosis; however, PIK3IP1 knockdown reversed this effect, suggesting that PIK3IP1 functions as an anti-apoptotic molecule. To identify both the upstream and downstream molecules associated with PIK3IP1, ET-1 receptor type-specific antagonists (BQ-123 and BQ-788) and PI3K subtype-specific antagonists (LY294002 and IPI-549) were used to determine the participating isoforms. Co-immunoprecipitation was performed to identify the binding partners of PIK3IP1. Our results demonstrated that ROS-induced cardiac cell death may occur through the ETA-PI3Kγ-AKT axis, and that PIK3IP1 inhibits binding with both ETA and PI3Kγ. Taken together, these findings reveal that PIK3IP1 plays an anti-ischemic role by reducing the likelihood of programmed cell death via interaction with the ETA-PI3Kr-AKT axis. The aim of this study was to examine how global cardiac gene transfer of sarcoplasmic reticulum [Ca.sup.2+]-ATPase (SERCA2a) can influence left ventricular (LV) mechanical and energetic function, especially in terms of [O.sub.2] cost of LV contractility, in normal rats. Normal rats were randomized to receive an adenovirus carrying the SERCA2a (SERCA) or [beta]-galactosidase ([beta]-Gal) gene or saline by a catheter-based technique. LV mechanical and energetic function was measured in cross-circulated heart preparations 2-3 days after the infection. The end-systolic pressure-volume relation was shifted upward, end-systolic pressure at 0.1 ml of intraballoon water volume was higher, and equivalent maximal elastance, i.e., enhanced LV contractility, was higher in the SERCA group than in the normal, [beta]-Gal, and saline groups. Moreover, the LV relaxation rate was faster in the SERCA group. There was no significant difference in myocardial [O.sub.2] consumption per beat-systolic pressure-volume area relation among the groups. Finally, [O.sub.2] cost of LV contractility was decreased to subnormal levels in the SERCA group but remained unchanged in the [beta]-Gal and saline groups. This lowered [O.sub.2] cost of LV contractility in SERCA hearts indicates energy saving in [Ca.sup.2+] handling during excitation-contraction coupling. In the approach, an NHS-ester linked methallylsilane is utilized as a bifunctional linker to couple the biomolecule to the silica surface. Weak organic acid such as acetic acid activates the silica surface enough to react with bifunctional linker without destroying activity of biomolecule.

First published June 10, 2004; 10.1152/ajpheart.01211.2003.--Impaired sarcoplasmic reticulum (SR) Ca release has been suggested to contribute to the depressed cardiac function in heart failure. The release of Ca from the SR may be regulated by the ryanodine receptor, triadin, junctin, calsequestrin, and a histidine-rich, Ca-binding protein (HRC). We observed that the levels of HRC were reduced in animal models and human heart failure. To gain insight into the physiological function of HRC, we infected adult rat cardiac myocytes with a recombinant adenovirus that contains the full-length mouse HRC cDNA. Overexpression (l.7-fold) of HRC in adult rat cardiomyocytes was associated with increased SR Ca load (28%) but decreased SR Ca-induced Ca release (37%). resulting in impaired Ca cycling and depressed fractional shortening (36%) as well as depressed rates of shortening (38%) and relengthening (33%). Furthermore, the depressed basal contractile and Ca kinetic parameters in the HRC-infected myocytes remained significantly depressed even after maximal isoproterenol stimulation. Interestingly, HRC overexpresssion was accompanied by increased protein levels of junctin (1.4-fold) and triadin (1.8-fold), whereas the protein levels of ryanodine receptor, calsequestrin, phospholamban, and sarco(endo)plasmic reticulum Ca-ATPase remained unaltered. The autoprocessing reaction is shown to proceed via an internal thioester intermediate and result in a covalent modification that increases the signaling domain's hydrophobic character and influences its subcellular and spatial distribution. Truncated unprocessed amino-terminal protein is shown to cause embryonic mispatterning even when expression is localized to cells that normally express Hh. This suggests the role of autoprocessing in the spatial regulation of hh signaling.

Prehair formation by inturned (in) gene of Drosophila is cold-sensitive, and the in-gene regulates the initiation of hair morphogenesis before wing hair development. The in-temperature sensitive period, which occurs before the first sign of prehair morphogenesis involves a cold-sensitive microtubule cytoskeleton structures. In-heat sensitive point starts at the end of cold-sensitive period, and induces multiple wing hair phenocopy.

Well‐defined recyclable Pd‐ and Rh‐bipyridyl group‐impregnated SBA‐15 catalysts were prepared for C−C bond coupling reaction and selective hydrogenation reactions, respectively. These SBA‐15 derived ligands for the catalysts were prepared by direct and indirect co‐condensation method using bipyridyl‐linked methallylsilane. This indirect method, involving methoxysilane generated from methallylsilane shows higher loading efficiency of transition metal catalysts on SBA‐15 than the direct use of methallylsilane. We present the first wide-range measurement of the charged-particle pseudorapidity density distribution, for different centralities (the 0-5%, 5-10%, 10-20%, and 20-30% most central events) in Pb-Pb collisions at $\sqrt{s_{\rm NN}} = 2.76$ TeV at the LHC. The measurement is performed using the full coverage of the ALICE detectors, $-5.0 < ��< 5.5$, and employing a special analysis technique based on collisions arising from LHC "satellite" bunches. We present the pseudorapidity density as a function of the number of participating nucleons as well as an extrapolation to the total number of produced charged particles ($N_{\rm ch} = 17165 \pm 772$ for the 0-5% most central collisions). From the measured ${\rm d}N_{\rm ch}/{\rm d}��$ distribution we derive the rapidity density distribution, ${\rm d}N_{\rm ch}/{\rm d}y$, under simple assumptions. The rapidity density distribution is found to be significantly wider than the predictions of the Landau model. We assess the validity of longitudinal scaling by comparing to lower energy results from RHIC.
