Abstract

Drought stress is one of the primary stresses that reduce crops’ yield in some parts of the world. In this regard, to mitigate drought stress effects on plants, farmers usually apply chemical fertilizers that lead to environmental issues. This study aimed to investigate the effect of biofertilizer application containing potassium-solubilizing bacteria (Pseudomonas koreensisand Pseudomonas vancouverensis), phosphorus-solubilizing bacteria (Pseudomonas putida), and nitrogen-fixing bacteria (Pantoea agglomerans) as an environmentally friendly biofertilizer for the growth and uptake of some essential nutrients in maize and its effect on drought tolerance. To achieve this, a Completely Randomized Design with ten treatments experiment was conducted in greenhouse conditions. The findings revealed that biofertilizers have the potential to improve maize yield and physiological characteristics, particularly under drought stress. The discussion section explores the mechanisms through which biofertilizers exert their effect and discusses practical implications for agricultural practices and environmental sustainability. Overall, this study contributes valuable insights into sustainable agricultural practices and has the potential to inform decision-making processes for farmers and policymakers.

60. Oxygen in the Earth's core: a first principles study

Abstract

First principles electronic structure calculations based on density functional theory have been used to study the thermodynamic, structural and transport properties of solid solutions and liquid alloys of iron and oxygen at Earth's core conditions. Aims of the work are to determine the oxygen concentration needed to account for the inferred density in the outer core, to probe the stability of the liquid against phase separation, to interpret the bonding in the liquid, and to find out whether the viscosity differs significantly from that of pure liquid iron at the same conditions. It is shown that the required concentration of oxygen is in the region 25-30 mol percent, and evidence is presented for phase stability at these conditions. The Fe-O bonding is partly ionic, but with a strong covalent component. The viscosity is lower than that of pure liquid iron at Earth's core conditions. It is shown that earlier first-principles calculations indicating very large enthalpies of formation of solid solutions may need reinterpretation, since the assumed crystal structures are not the most stable at the oxygen concentration of interest., Comment: 21 pages, 12 figures

Abstract

Lung cancer is the leading cause of cancer-associated mortality worldwide. Here we analysed 1,644 tumour regions sampled at surgery or during follow-up from the first 421 patients with non-small cell lung cancer prospectively enrolled into the TRACERx study. This project aims to decipher lung cancer evolution and address the primary study endpoint: determining the relationship between intratumour heterogeneity and clinical outcome. In lung adenocarcinoma, mutations in 22 out of 40 common cancer genes were under significant subclonal selection, including classical tumour initiators such as TP53 and KRAS. We defined evolutionary dependencies between drivers, mutational processes and whole genome doubling (WGD) events. Despite patients having a history of smoking, 8% of lung adenocarcinomas lacked evidence of tobacco-induced mutagenesis. These tumours also had similar detection rates for EGFR mutations and for RET, ROS1, ALK and MET oncogenic isoforms compared with tumours in never-smokers, which suggests that they have a similar aetiology and pathogenesis. Large subclonal expansions were associated with positive subclonal selection. Patients with tumours harbouring recent subclonal expansions, on the terminus of a phylogenetic branch, had significantly shorter disease-free survival. Subclonal WGD was detected in 19% of tumours, and 10% of tumours harboured multiple subclonal WGDs in parallel. Subclonal, but not truncal, WGD was associated with shorter disease-free survival. Copy number heterogeneity was associated with extrathoracic relapse within 1 year after surgery. These data demonstrate the importance of clonal expansion, WGD and copy number instability in determining the timing and patterns of relapse in non-small cell lung cancer and provide a comprehensive clinical cancer evolutionary data resource.

Abstract

Introduction: Recent studies suggest that immune-related cells can be recruited for anti-tumor functions as well as tumor progression and the interplay between systemic inflammation and local immune response may play a major role in the development and progression of various cancers including lung cancer. Inflammatory markers, such as neutrophil-to-lymphocyte ratio (NLR), platelet-to-lymphocyte ratio (PLR), and systemic immune-inflammation index (SII) can be used as surrogate biomarkers of host immune status. In this work, associations between neutrophils, lymphocytes, platelets, NLR, PLR, SII and overall survival (OS) are investigated in two cohorts of non-small cell lung cancer (NSCLC) patients treated with fractionated radiotherapy (RT) and stereotactic body radiation therapy (SBRT) and a cohort of small cell lung cancer (SCLC) patients treated with fractionated RT. Material and Methods: Data from 2513 lung cancer patients were retrospectively analyzed. Baseline NLR, PLR, and SII (NLR × platelet count) were calculated from full blood test prior to RT initiation. Cox proportional hazards regression analyses were used to evaluate the association between systemic inflammation markers and known clinical factors with OS. Results: The two-year OS was 42%, 63%, and 62% in the NSCLC fractionated RT, SBRT, and SCLC cohort. NLR (per 1 unit: hazard ratio [HR]: 1.04, p < 0.05) and SII (per 100 × 109/L: HR: 1.01, p < 0.05) remained the strongest independent factors of OS in multivariable Cox analyses, correcting for clinical factors in early-stage and locally advanced NSCLC and SCLC patients treated with RT. Discussion: This single-center large-cohort study suggests that baseline NLR and SII are independent prognostic biomarkers associated with OS in locally advanced and early-stage NSCLC patients treated with either curative-intent fractionated RT or SBRT and SCLC patients treated with curative-intent fractionated RT. External validation is warranted to evaluate the utility of these biomarkers for patients’ stratification and adapting new treatment approaches.

Abstract

LCIA (low CO2-inducible protein A) is a chloroplast envelope protein associated with the CO2-concentrating mechanism of the green alga Chlamydomonas reinhardtii. LCIA is postulated to be a HCO3– channel, but previous studies were unable to show that LCIA was actively transporting bicarbonate in planta. Therefore, LCIA activity was investigated more directly in two heterologous systems: an Escherichia coli mutant (DCAKO) lacking both native carbonic anhydrases and an Arabidopsis mutant (βca5) missing the plastid carbonic anhydrase βCA5. Neither DCAKO nor βca5 can grow in ambient CO2 conditions, as they lack carbonic anhydrase-catalyzed production of the necessary HCO3– concentration for lipid and nucleic acid biosynthesis. Expression of LCIA restored growth in both systems in ambient CO2 conditions, which strongly suggests that LCIA is facilitating HCO3– uptake in each system. To our knowledge, this is the first direct evidence that LCIA moves HCO3– across membranes in bacteria and plants. Furthermore, the βca5 plant bioassay used in this study is the first system for testing HCO3– transport activity in planta , an experimental breakthrough that will be valuable for future studies aimed at improving the photosynthetic efficiency of crop plants using components from algal CO2-concentrating mechanisms. Abstract

Salinity and drought are two major abiotic stresses challenging global crop production and food security. In this study, the effects of individual and combined effects of drought (at different phenological stages) and salt stresses on growth, morphology, and physiology of triticale were evaluated. For this purpose, a 3 x 4 factorial design in three blocks experiment was conducted. The stress treatments included three levels of salinity (0, 50, and 100 mM NaCl) and four levels of drought (regular irrigation as well as irrigation disruption at heading, flowering, and kernel extension stages). The stresses, individual as well as combined, caused a significant decrease in chlorophyll contents, total dry matter, leaf area index, relative water content, and grain yield of triticale. In this regard, the highest reduction was recorded under combined stresses of 100 mM NaCl and drought stress at flowering. However, an increase in soluble sugars, leaf free proline, carotenoid contents, and electrolyte leakage was noted under stress conditions compared to the control. In this regard, the highest increase in leaf free proline, soluble sugars, and carotenoid contents were noted under the combination of severe salinity and drought stress imposed at the flowering stage. Investigating the growth indices in severe salinity and water deficit stress in different phenological stages shows the predominance of ionic stress over osmotic stress under severe salinity. The highest grain yield was observed under non-saline well-watered conditions whereas the lowest grain yield was recorded under severe salinity and drought stress imposed at the flowering stage. In conclusion, the flowering stage was more sensitive than the heading and kernel extension stages in terms of water deficit. The impact of salinity and water deficit was more pronounced on soluble sugars and leaf free proline; so, these criteria can be used as physiological indicators for drought and salinity tolerance in triticale.
