IntroductionCentral pancreatectomy (CP) is a standard surgical procedure for benign and low-grade malignant pancreatic neoplasms in the body and neck of the pancreas. Higher incidence of clinically relevant postoperative pancreatic fistula (CR-POPF) after CP than after pancreaticoduodenectomy (PD) or distal pancreatectomy (DP) has been reported, but no nomogram for prediction of CR-POPF after open CP has been previously established.MethodsPatients undergoing open CP for benign or low-grade malignant pancreatic neoplasms in the department of Hepatobiliary and Pancreatic (HBP) surgery of Shanghai Changhai Hospital affiliated to Naval Medical University between January 01, 2009 and December 31,2020 were enrolled. Pre-, intra- and post-operative parameters were analyzed retrospectively.ResultsA total of 194 patients, including 60 men and 134 women, were enrolled with median age of 52 years (21~85 years). 84 patients (43.3%) were overweight (BMI>23.0 Kg/m2) and 14 (7.2%) were obese (BMI>28.0 Kg/m2). Pathological diagnoses ranged from serous cystic neoplasm (32.5%), solid pseudopapillary neoplasm (22.2%), pancreatic neuroendocrine tumor (20.1%), intraductal papillary mucinous neoplasm (18.0%) to mucinous cystic neoplasm (5.2%). All patients had soft pancreatic texture. Main pancreatic duct diameters were ≤0.3cm for 158 patients (81.4%) and were ≥0.5cm in only 12 patients (6.2%). A stapler (57.7%) or hand-sewn closure (42.3%) were used to close the pancreatic remnant. The pancreatic anastomosis techniques used were duct to mucosa pancreaticojejunostomy (PJ)-interrupted suture (47.4%), duct to mucosa PJ-continuous suture (43.3%), duct to mucosa “HO” half-purse binding PJ (5.2%) and invaginating pancreaticogastrostomy (4.1%). Post-surgical incidences of CR-POPF of 45.9%, surgical site infection of 28.9%, postpancreatectomy hemorrhage of 7.7% and delayed gastric emptying of 2.1% were found. Obesity and pancreatic anastomosis technique were independent risk factors of CR-POPF, with a concordance index of 0.675 and an Area Under the Curve of 0.678.DiscussionThis novel nomogram constructed according to obesity and pancreatic anastomosis technique showed moderate predictive performance of CR-POPF after open CP.

Besides insulin deficiency, glucagon excess is also regarded as a critical factor of type 2 diabetes mellitus (T2DM) according to the bi-hormonal hypothesis. Triterpenic acid-enriched fraction (TAE) of Cyclocarya paliurus leaves have been proved to effectively inhibit glucagon secretion, but the active ingredient for the efficacy and mechanism of TAE on glucagon secretion remains unclear. The present study was designed to identify the therapeutic component of TAE to treat T2DM and explore the exact mechanism. Herein, arjunolic acid (AA) screened from TAE selectively inhibited glucagon secretion in palmitate (PA)-induced αTC1-6 cells and isolated islets. Additionally, AA ameliorated hyperglycemia and hyperglucagonemia and improve pancreatic islet morphology and function in streptozotocin (STZ)-induced T2DM mice. Noticeably, the selective inhibition effect of AA on glucagon secretion from α-cell was regulated by facilitating ephrin-A1 and EphA4 interaction, subsequently activating PI3K and Akt pathway. Therefore, AA may be a promising agent for T2DM treatment.

Two new genera and species of Ithonidae, i.e. Stictopolystoechotes sparsulus Gao, Xu et Wang gen. et sp. nov. and Pycnopolystoechotes striatus Gao, Xu et Wang gen. et sp. nov., are described from the Middle Jurassic Jiulongshan Formation, Inner Mongolia, China. The two new genera can be assigned to the Polystoechotes genus-group of Ithonidae by the combination of following characters: elongated forewing, Sc and RA fused distally, crossveins in radical sector sparse except for the gradate series, and simple CuP branches. The known wing markings among the Jurassic Polystoechotes genus-group were outlined, which suggested the diversification of wing markings was possibly positively associated with the species radiation of this lineage during this period. Moreover, a key to fossil genera of the Polystoechotes genus-group was provided. Grylloidea (crickets) and Gryllotalpoidea (mole crickets and ant crickets) are relatively ancient lineages within Orthoptera but their fossil records are not very rich. They are currently considered sister clades within the Gryllidea but with obvious differences in morphology and ecological adaptations. We report a new gryllidean family, †Pseudogryllotalpidae fam. nov. with three new genera and four new species, viz. †Pseudogryllotalpa scalprata gen. et sp. nov., †Unidigitus longialatus gen. et sp. nov., †Petilus zhengi gen. et sp. nov. and †Chunxiania fascia sp. nov. from the mid‐Cretaceous of northern Myanmar (ca. 99 million years ago). Their external and short ovipositors and their modified fossorial forelegs suggest a soil‐inhabiting preference. This new family exhibits a series of 'intermediate' character states between Grylloidea and Gryllotalpoidea and seems to be a 'transitional' fossil group linking these two superfamilies. This new family was resolved as a sister group of the extant Gryllotalpoidea and provides a new insight into the relationship and evolution of Grylloidea and Gryllotalpoidea. [ABSTRACT FROM AUTHOR]

Summary: We investigated the mining mode of insect feeding, involving larval consumption of a plant's internal tissues, from the Middle Jurassic (165 million years ago) Daohugou locality of Northeastern China. Documentation of mining from the Jurassic Period is virtually unknown, and results from this time interval would address mining evolution during the temporal gap of mine–seed plant diversifications from the previous Late Triassic to the subsequent Early Cretaceous.Plant fossils were examined with standard microscopic procedures for herbivory and used the standard functional feeding group–damage‐type system of categorizing damage. All fossil mines were photographed and databased.We examined 2014 plant specimens, of which 27 occurrences on 14 specimens resulted in eight, new, mine damage types (DTs) present on six genera of bennettitalean, ginkgoalean, and pinalean gymnosperms.Three conclusions emerge from this study. First, these mid‐Mesozoic mines are morphologically conservative and track plant host anatomical structure rather than plant phylogeny. Second, likely insect fabricators of these mines were three basal lineages of polyphagan beetles, four basal lineages of monotrysian moths, and a basal lineage tenthredinoid sawflies. Third, the nutrition hypothesis, indicating that miners had greater access to nutritious, inner tissues of new plant lineages, best explains mine evolution during the mid‐Mesozoic. Burmese amber preserves a diverse assemblage of Cretaceous arachnids, and among pseudoscorpions (Arachnida: Pseudoscorpiones), ten species in five families have already been named. Here, we describe a new fossil species from Burmese amber in the pseudoscorpion family Hyidae, providing detailed measurements, photographs and 3D-models from synchrotron scanning. Based on morphology, the new fossil, Hya fynni sp. nov. is placed in the genus Hya, and is nearly identical to extant species in the genus, except for the position of trichobothrium est on the pedipalpal chela, thereby indicating extreme morphological stasis in this invertebrate lineage over the last 99 million years. Hya fynni represents the first described fossil species in Hyidae, and the third described Burmese fossil in the superfamily Neobisioidea. It also joins the garypinid, Amblyolpium burmiticum, in representing the oldest fossil records for extant pseudoscorpion genera. Considering proposed divergence dates, the newly described fossil species bolsters a Gondwanan origin for Hyidae, and provides evidence for the "Late Jurassic Rifting" hypothesis for the Burma Terrane, in which this landmass rifted from Gondwana in the Late Jurassic and collided with Eurasia by the Cretaceous/Eocene. Like Hya species today, H. fynni likely inhabited humicolous microhabitats in tropical forests on the Burma Terrane, supporting ecological niche stasis for this family since the Mesozoic. BACKGROUND:Post-infarction cardiovascular remodeling and heart failure are the leading cause of myocardial infarction (MI)-driven death during the past decades. Experimental observations have involved intestinal microbiota in the susceptibility to MI in mice; however, in humans, identifying whether translocation of gut bacteria to systemic circulation contributes to cardiovascular events post-MI remains a major challenge. RESULTS:Here, we carried out a metagenomic analysis to characterize the systemic bacteria in a cohort of 49 healthy control individuals, 50 stable coronary heart disease (CHD) subjects, and 100 ST-segment elevation myocardial infarction (STEMI) patients. We report for the first time higher microbial richness and diversity in the systemic microbiome of STEMI patients. More than 12% of post-STEMI blood bacteria were dominated by intestinal microbiota (Lactobacillus, Bacteroides, and Streptococcus). The significantly increased product of gut bacterial translocation (LPS and D-lactate) was correlated with systemic inflammation and predicted adverse cardiovascular events. Following experimental MI, compromised left ventricle (LV) function and intestinal hypoperfusion drove gut permeability elevation through tight junction protein suppression and intestinal mucosal injury. Upon abrogation of gut bacterial translocation by antibiotic treatment, both systemic inflammation and cardiomyocyte injury in MI mice were alleviated. CONCLUSIONS:Our results provide the first evidence that cardiovascular outcomes post-MI are driven by intestinal microbiota translocation into systemic circulation. New therapeutic strategies targeting to protect the gut barrier and eliminate gut bacteria translocation may reduce or even prevent cardiovascular events post-MI.

Abstract Activation of adipose tissue macrophages (ATMs) contributes to chronic inflammation and insulin resistance in obesity. However, the transcriptional regulatory machinery involved in ATM activation during the development of obesity is not fully understood. Here, we profiled the chromatin accessibility of blood monocytes and ATMs from obese and lean mice using assay for transposase-accessible chromatin sequencing (ATAC-seq). We found that monocytes and ATMs from obese and lean mice exhibited distinct chromatin accessibility status. There are distinct regulatory elements that are specifically associated with monocyte or ATM activation in obesity. We also discovered several transcription factors that may regulate monocyte and ATM activation in obese mice, specifically a predicted transcription factor named ETS translocation variant 5 (ETV5). The expression of ETV5 was significantly decreased in ATMs from obese mice and its downregulation was mediated by palmitate stimulation. The decrease in ETV5 expression resulted in macrophage activation. Our results also indicate that ETV5 suppresses endoplasmic reticulum (ER) stress and Il6 expression in macrophages. Our work delineates the changes in chromatin accessibility in monocytes and ATMs during obesity, and identifies ETV5 as a critical transcription factor suppressing ATM activation, suggesting its potential use as a therapeutic target in obesity-related chronic inflammation.
