Item does not contain fulltext In the WAG/Rij rat, a model for human absence epilepsy, spike-wave discharges and absence epileptic behaviour are fully symptomatic at an age of 6 months. The hypothesis is tested that the brain disorder underlying absence epilepsy is related to a changed distribution of neuronal parvalbumin (PV). Quantitative immunocytochemistry of PV shows that, in comparison to non-epileptic ACI control rats, the number of PV-containing neurons in WAG/Rij rats is considerably lower in the somatosensory cortex (parietal cortex area 1 and the forelimb area) and in the olfactory tubercle, but similar in the rostral reticular thalamic nucleus. The possible significance of the low amount of PV in the cortical areas in relation to absence epilepsy is discussed.

Abstract

The adult WAG/Rij rat is a well-established animal model for human absence epilepsy characterized by the presence of spike-wave discharges (SWD) in the EEG concomitant with behavioural symptoms such as vibrissae twitching. The rostral part of the reticular thalamic nucleus (rRTN) is generally assumed to contain the pacemaker for the occurrence of SWD. We investigated if SWD maintenance is related to the synaptic organization of the rRTN, by studying the ultrastructure of the rRTN of absence epileptic WAG/Rij rats in comparison with non-epileptic, age-matched ACI control rats. In WAG/Rij rats, we show D-, L- and F-type terminals on somata, dendrites, dendritic spines and axons, in single as well as in convergent and divergent configurations. Semi-quantitative observations indicate that D-type synapses, especially axo-dendritic ones, dominate. L- and F-type terminals are common, but less frequent than D-type terminals. Occasionally, F-type terminals were observed to innervate the axon hillock. As to multiple inputs, all three terminal types are involved in multiple synaptic configurations, with somata, dendrites and axons as postsynaptic structures. Convergent synaptic configurations outnumber divergent ones. In the non-epileptic ACI rat, the synaptology of the rRTN is similar to that of the WAG/Rij rat. This indicates that the role of the rRTN in WAG/Rij rats in the maintenance absence epilepsy does not depend on its synaptic organization.

Contains fulltext : 57824.pdf (Publisher’s version ) (Closed access) Dopamine inhibits pituitary melanotrope cells of the amphibian Xenopus laevis through activation of a dopamine (D2) receptor that couples to a Gi protein. Activated Gi protein subunits are known to affect voltage-operated Ca2+ currents (ICa). In the present study we investigated which Ca2+ currents are regulated by D2-receptor activation and which Gi protein subunits are involved. Whole-cell voltage-clamp patch-clamp experiments from holding potentials (HPs) of -80 and -30 mV show that 28.6 and 36.9%, respectively, of the total ICa was inhibited by apomorphin, a D2-receptor agonist. The inhibited current had fast activation and inactivation kinetics. From an HP of -80 mV, inhibition of N-type Ca2+ currents with omega-conotoxin GVIA and R-type current by SNX-482 reduced the efficacy of the apomorphin-induced inhibition. From an HP of -30 mV this reduction for omega-conotoxin GVIA was still observed. Blocking L-type current by nifedipine or P/Q-type current by omega-agatoxin IVA did not affect apomorphin-induced inhibition at either HP. Our results imply that D2-receptor activation inhibits both N- and R-type Ca2+ currents. Using a strong depolarizing pre-pulse partially reversed the inhibition of the total current by apomorphin. About 50% of this inhibition was achieved through interaction of Gbeta/gamma proteins, and this part of the inhibited ICa had fast activating and inactivating kinetics. However, the other part of the current inhibited by D2-receptor activation may proceed through Galpha-PKA phosphorylation.

Item does not contain fulltext ddThe distribution of cocaine- and amphetamine-regulated transcript peptide (CARTp)-like immunoreactivity was studied only in the rat central nervous system (CNS). In mammals, CART peptides occur among others in brain areas that control feeding behavior. We mapped CARTp-immunoreactive structures in the CNS of the frog Rana esculenta and assumed that differences may exist in the CARTp-containing neuronal populations between the frog, which does not feed in winter, and the rat. In the forebrain, immunoreactive cells and fibers were found in the olfactory bulb, nucleus accumbens, amygdala, medial pallium, septum, striatum, the preoptic nuclei, ventromedial nucleus, central thalamic nucleus, and the hypothalamus. The optic pathway was free of immunoreactivity. The neurohypophysis showed intense immunostaining. In the mesencephalon, many cells were stained in the Edinger-Westphal nucleus, and a few in the optic tectum, where fibers were stained in all plexiform layers. In the retina, some cells in the inner nuclear layer contained CARTp. In the rhombencephalon, cells were stained in the raphe nuclei, central gray, nucleus of the solitary tract, and the vicinity of motor nuclei. Neurons of the motor cranial nerves were densely innervated by CARTp-positive fibers originating from the spinal cord. In the spinal cord, preganglionic cells were stained, and motoneurons were surrounded by immunoreactive varicose axon terminals. Major differences were found between the frog and the rat brains in the distribution of CARTp in the visual system, olfactory bulb, preoptic area, and the motor nuclei. The leptin receptor is present in hypothalamic centers controlling energy metabolism as well as in the centrally projecting Edinger-Westphal nucleus (EWcp), a region implicated in the stress response and in various aspects of stress-related behaviors. We hypothesized that the stress response by cocaine- and amphetamine-regulated transcript (CART)-producing EWcp-neurons would depend on the animal's energy state. To test this hypothesis, we investigated the effects of changes in energy state (mimicked by low, normal and high leptin levels, which were achieved by 24 h fasting, normal chow and leptin injection, respectively) on the response of CART neurons in the EWcp of rats subjected or not to acute restraint stress. Our data show that leptin treatment alone significantly increases CART mRNA expression in the rat EWcp and that in leptin receptor deficient (db/db) mice, the number of CART producing neurons in this nucleus is reduced. This suggests that leptin has a stimulatory effect on the production of CART in the EWcp under non-stressed condition. Under stressed condition, however, leptin blunts stress-induced activation of EWcp neurons and decreases their CART mRNA expression. Interestingly, fasting, does not influence the stress-induced activation of EWcp-neurons, and specifically EWcp-CART neurons are not activated. These results suggest that the stress response by the EWcp depends to some degree on the animal's energy state, a mechanism that may contribute to a better understanding of the complex interplay between obesity and stress.

Item does not contain fulltext Ca2+ oscillations regulate secretion of the hormone alpha-melanphore-stimulating hormone (alpha-MSH) by the neuroendocrine pituitary melanotrope cells of the amphibian Xenopus laevis. These Ca2+ oscillations are built up by discrete increments in the intracellular Ca2+ concentration, the Ca2+ steps, which are generated by electrical membrane bursting firing activity. It has been demonstrated that the patterns of Ca2+ oscillations and kinetics of the Ca2+ steps can be modulated by changing the degree of intracellular Ca2+ buffering. We hypothesized that neurotransmitters known to regulate alpha-MSH secretion also modulate the pattern of Ca2+ oscillations and related electrical membrane activity. In this study, we tested this hypothesis for the secretagogue sauvagine. Using high temporal-resolution Ca2+ imaging, we show that sauvagine modulated the pattern of Ca2+ signalling by increasing the frequency of Ca2+ oscillations and inducing a broadening of the oscillations through its effect on various Ca2+ step parameters. Second, we demonstrate that sauvagine caused a small but significant decrease in K+ currents measured in the whole-cell voltage-clamp, whereas Ca2+ currents remained unchanged. Third, in the cell-attached patch-clamp mode, a stimulatory effect of sauvagine on action current firing was observed. Moreover, sauvagine changed the shape of individual action currents. These results support the hypothesis that the secretagogue sauvagine stimulates the frequency of Ca2+ oscillations in Xenopus melanotropes by altering Ca2+ step parameters, an action that likely is evoked by an inhibition of K+ currents.

Contains fulltext : 103841.pdf (publisher's version ) (Closed access), Brain-derived neurotrophic factor (BDNF) is, despite its name, also found outside the central nervous system (CNS), but the functional significance of this observation is largely unknown. This review concerns the expression of BDNF in the pituitary gland. While the presence of the neurotrophin in the mammalian pituitary gland is well documented its functional significance remains obscure. Studies on the pars intermedia of the pituitary of the amphibian Xenopus laevis have shown that BDNF is produced by the neuroendocrine melanotrope cells, its expression is physiologically regulated, and the melanotrope cells themselves express receptors for the neurotrophin. The neurotrophin has been shown to act as an autocrine factor on the melanotrope to promote cell growth and regulate gene expression. In doing so BDNF supports the physiological function of the cell to produce and release alpha-melanophore-stimulating hormone for the purpose of adjusting the animal's skin color to that of its background.

Item does not contain fulltext, Classical studies in amphibians have concluded that the endocrine pituitary and pars intermedia are derived from epithelial buccal epidermis and do not require the infundibulum for their induction. These studies also assumed that the pituitary is not subsequently determined by infundibular induction. Our extirpation, auto-transplantation and immunohistochemical studies with Xenopus laevis were initiated to investigate early presumptive pituitary development. These studies were conducted especially with reference to the pars intermedia melanotrope cell's induction, and its production and release of alpha-melanophore stimulating hormone (alpha-MSH) from the precursor protein proopiomelanocortin (POMC). Auto-transplantation studies demonstrated that the pituitary POMC-producing cells are determined at a stage prior to pituitary-infundibular contact. The results of experiments involving the extirpation of the presumptive infundibulum also indicated that the infundibulum is not essential for the differentiation of POMC-producing cells. We also demonstrated that early pituitary development involves adherence to the prechiasmatic area of the diencephalon with the pituitary placode growing in a posterior direction toward the infundibulum where contact occurs at Xenopus stage 39/40. Overall, our studies provide a model for early tissue relations among presumptive pituitary, suprachiasmatic nucleus, pars tuberalis and infundibulum during neurulation and later neural tube stages of development. It is hypothesized that the overlying chiasmatic area suppresses pituitary differentiation.

In rodents, cocaine- and amphetamine-regulated transcript (CART) and nesfatin-1/NUCB2 have not only been implicated in metabolism, but in the pathobiology of anxiety and depressive-like behaviour, however they have not previously been investigated in depressed subjects. Both peptides are highly expressed in centrally projecting neurons in the Edinger-Westphal nucleus (EWcp) in the midbrain. The EWcp has been implicated in stress adaptation and stress-related mood disorders like major depressive disorder in a sex-specific manner. This is intriguing, given the fact that females have higher prevalence of mood disorders. Here, we hypothesized that the expression of CART and nesfatin-1 in EWcp would exhibit a sex-specific difference between depressed suicide victims vs. controls. We found that CART and nesfatin/NUCB2 colocalized in the human EWcp, and that CART mRNA content was much higher in both male (x3.8) and female (x5.9) drug-free suicide victims than in controls (persons who died without any diagnosed neurodegenerative or psychiatric disorder). Similarly, NUCB2 mRNA content was also higher (x1.8) in male suicides, whereas in female suicide victims, these contents were x2.7 lower compared to controls. These observations are the first to show changes in the dynamics of CART and nesfatin/NUCB2 expressions in the midbrain of drug-free depressed suicide victims vs. controls. Co-promotor : Kozicz, L.T., Contains fulltext : 91338.pdf (publisher's version ) (Open Access), The aim of this thesis research was to test our hypothesis that the non-preganglionic Edinger-Westphal nucleus (npEW) serves as an integrating center linking energy metabolism with stress adaptation. We have shown indeed that the npEW receives information about stress stimuli and the peripheral metabolic state. Moreover, it appears that not only the npEW as a whole responds to this information by changing its activity, but also that this reaction is a result of the differential reactions of the various neuropeptides expressed in this nucleus. Our results extend knowledge of the energy-dependent regulation of the stress adaptation response and, hopefully, will contribute to broadening insight into the etiology of stress-induced anxiety, depression and eating disorders, as well as may stimulate research to identify novel treatments to combat these disorders. In this respect, future studies on the mechanisms by which energy balance and leptin participate in the regulation of the npEW-Ucn1/CART system will be of particular interest.
