Background: HIV protease inhibitor (PI)-induced inflammatory response in macrophages is a major risk factor for cardiovascular diseases. We have previously reported that berberine (BBR), a traditional herbal medicine, prevents HIV PIinduced inflammatory response through inhibiting endoplasmic reticulum (ER) stress in macrophages. We also found that HIV PIs significantly increased the intracellular concentrations of BBR in macrophages. However, the underlying mechanisms of HIV PI-induced BBR accumulation are unknown. This study examined the role of P-glycoprotein (P-gp) in HIV PI-mediated accumulation of BBR in macrophages. Methodology and Principal Findings: Cultured mouse RAW264.7 macrophages, human THP-1-derived macrophages, Wild type MDCK (MDCK/WT) and human P-gp transfected (MDCK/P-gp) cells were used in this study. The intracellular concentration of BBR was determined by HPLC. The activity of P-gp was assessed by measuring digoxin and rhodamine 123 (Rh123) efflux. The interaction between P-gp and BBR or HIV PIs was predicated by Glide docking using Schrodinger program. The results indicate that P-gp contributed to the efflux of BBR in macrophages. HIV PIs significantly increased BBR concentrations in macrophages; however, BBR did not alter cellular HIV PI concentrations. Although HIV PIs did not affect Pgp expression, P-gp transport activities were significantly inhibited in HIV PI-treated macrophages. Furthermore, the molecular docking study suggests that both HIV PIs and BBR fit the binding pocket of P-gp, and HIV PIs may compete with BBR to bind P-gp. Conclusion and Significance: HIV PIs increase the concentration of BBR by modulating the transport activity of P-gp in macrophages. The role of endoplasmic reticulum (ER) stress is a rapidly emerging field of interest in the pathogenesis of metabolic diseases. Recent studies have shown that chronic activation of ER stress is closely linked to dysregulation of lipid metabolism in several metabolically important cells including hepatocytes, macrophages, β-cells, and adipocytes. Adipocytes are one of the major cell types involved in the pathogenesis of the metabolic syndrome. Recent advances in dissecting the cellular and molecular mechanisms involved in the regulation of adipogenesis and lipid metabolism indicate that activation of ER stress plays a central role in regulating adipocyte function. In this paper, we discuss the current understanding of the potential role of ER stress in lipid metabolism in adipocytes. In addition, we touch upon the interaction of ER stress and autophagy as well as inflammation. Inhibition of ER stress has the potential of decreasing the pathology in adipose tissue that is seen with energy overbalance. [ABSTRACT FROM AUTHOR]

We report experimental results on relativistic electron beam (REB) transport in a set of cold and shock-heated carbon samples using the high-intensity kilojoule-class OMEGA EP laser. The REB energy distribution and transport were diagnosed using an electron spectrometer and x-ray fluorescence measurements from a Cu tracer buried at the rear side of the samples. The measured rear REB density shows brighter and narrower signals when the targets were shock-heated. Hybrid PIC simulations using advanced resistivity models in the target warm-dense-matter (WDM) conditions confirm this observation. We show that the resistivity response of the media, which governs the self-generated resistive fields, is of paramount importance to understand and correctly predict the REB transport.

Heavy metal pollution has become an increasingly recognized threat to the aquatic environment. In response to heavy metal stresses, various terrestrial plants produce the hormone nitric oxide (NO), but minimal information is available about the role of NO in aquatic macrophytes under high zinc (Zn) stress. In this study, we measured physiochemical parameters in the leaves of Hydrilla verticillata exposed to Zn2+ (10 mg l-1) alone or combined with NO (supplied as sodium nitroprusside, SNP) for 1 d and 4 d. Excess Zn disturbed nutrient uptake and altered the redox status of the aquatic plants. NO application partially reversed negative impacts of Zn stress on the contents of photosynthetic pigments, malondialdehyde, nutrient concentration, ascorbic acid, and the activity of antioxidant enzymes. However, the metabolites of SNP did not have similar effects. After treatments for 4 d, plants were further cultured in a solution containing Zn2+ (10 mg l-1) for 10 d. NO application partially reversed Zn-induced negative effects in parameters such as nutrient uptake, antioxidant enzymatic activities, and biomass, as compared to treatments with Zn alone. These results indicate that NO supply could mitigate Zn stress in H. verticillata, as a defense mechanism of the plant against Zn toxicity.

Abstract Background and Objective: The emergence of a novel H1N1influenza A virus of animal origin with transmissibility from human to human poses pandemic concern. Current subtypes of Seasonal influenza A viruses spread in human are influenza A H1N1 influenza A H3N2 and influenza type B viruses. The aim of this study was to determine current strains of the H3N2 and new H1N1 subtypes of influenza A virus from patients suspected influenza infection in 2009 flu pandemic in Golestan province, Iran. Material and Methods: In this descriptive study, respiratory samples (n = 153) from patients with acute respiratory symptoms were collected in 2009 flu pandemic applied during 2009 pandemic influenza in Golestan province. After reverse transcription of extracted viral RNA, PCR was developed for both H1N1and H3N2subtypes using CDC specific primers. Results: The mean age of patients was 16.59. Of them 45.1% were male. Thirteen (8.49%) were infected with seasonal influenza H1N1 and 25(16.33%) with seasonal H3N2influenza. Conclusion: The rate of infection with seasonal H1N1and H3N2is similar to other studies reported from Iran, but lower than the rate reported from other parts of the world. Division of tectonic units in the eastern part of the Central Asian Orogenic Belt in northeast China has been a major concern and resulted in much fieldwork, but the division of these tectonic units in NE China is still controversial. Although detection of tectonic units in seismic sections is not straightforward, for this meeting, we shall try to relate tectonic units with the crustal and upper mantle structure and deformation derived from a ~2500 km long reflection seismic profile (Figure, red lines) in this area, recently acquired or reprocessed with support of China Geological Survey and the Chinese SinoProbe Project.

The effluent of an RF argon atmospheric pressure plasma jet, the so-called kinpen, is investigated with focus on the nitric-oxide (NO) distribution for laminar and turbulent flow regimes. An additional dry air gas curtain is applied around the plasma effluent to prevent interaction with the ambient humid air. By means of laser-induced fluorescence (LIF) the absolute spatially resolved NO density is measured as well as the rotational temperature and the air concentration. While in the laminar case, the transport of NO is attributed to thermal diffusion; in the turbulent case, turbulent mixing is responsible for air diffusion. Additionally, measurements with a molecular beam mass-spectrometer (MBMS) absolutely calibrated for NO are performed and compared with the LIF measurements. Discrepancies are explained by the contribution of the ${\rm N}{{{\rm O}}_{2}}$ and ${{{\rm N}}_{2}}{\rm O}$ to the MBMS NO signal. Finally, the effect of a conductive substrate in front of the plasma jet on the spatial distribution of NO and air diffusion is also investigated.

ABSTRACT The Beijing oil (BJO) chicken is an autochthona Chinese breed which shows outstanding meat quality characteristics compared with fast-growing imported chickens such as Arbor Acres (AA) chickens. Gut microbial diversity has been shown to influence host factors such as energy and nutrient metabolism, immune response and fertility. Consequently, it is hoped that analyses into host microbial populations will ultimately help to improve the quality of associated meat products. Two distinct chicken breeds, Arbor Acres (AA) and Beijing oil (BJO), were selected to analyze the composition of the 16S rRNA gene V3-V5 hypervariable regions using high-throughput sequencing technology. Upon elucidation of gut-colonizing bacteria, the Firmicutes were observed to encompass the dominant phylum. The abundance of Firmicutes in the BJO chickens (84.9%) was lower than in the AA chickens (89.9%). Conversely, the prevalence of Proteobacteria was 4.8% in BJO chickens, with a significantly lower abundance observed in AA chickens (1.3%). In the cecum, the Firmicutes were once more the dominant phylum in BJO chickens (60.2%) and AA chickens (63.7%). The abundance of bacteroidetes was 37.7% in BJO chickens and 34.9% in AA chickens, respectively. These discoveries provide a perception into the composition of the gut microbiotain both breeds. The study also provides a foundation for future research relating to gut bacterial factors that may influence the development and progression of gastrointestinal disease in chickens and other animals.

T-cell prolymphocytic leukemia (T-PLL) is a rare malignancy with a poor prognosis. Here, the authors investigate the genomic landscape, gene expression profiles and functional mechanisms in 111 patients, highlighting TCL1 overexpression and ATM aberrations as core lesions which co-operate to impair DNA damage processing.

Soil respiration (Rs) and its biophysical controls were measured over a fixed sand dune in a desert–shrub ecosystem in northwest China in 2012 to explore the mechanisms controlling the spatial heterogeneity in Rs and to understand the plant effects on the spatial variation in Rs in different phenophases. The measurements were carried out on four slope orientations (i.e., windward, leeward, north- and south-facing) and three height positions on each slope (i.e., lower, upper, and top) across the phenophases of the dominant shrub species (Artemisia ordosica). Coefficient of variation (i.e., standard deviation/mean) of Rs across the 11 microsites over our measurement period was 23.5 %. Soil respiration was highest on the leeward slope, and lowest on the windward slope. Over the measurement period, plant-related factors, rather than microhydrometeorological factors, affected the microtopographic variation in Rs. During the flower-bearing phase, root biomass affected Rs most, explaining 72 % of the total variation. During the leaf coloration–defoliation phase, soil nitrogen content affected Rs the most, explaining 56 % of the total variation. Our findings highlight that spatial pattern in Rs was dependent on plant distribution over a desert sand dune, and plant-related factors largely regulated topographic variation in Rs, and such regulations varied with plant phenology.

The effects of foliar application of 24-epibrassinolide (EBR) on chlorophyll content, oxidative damage, and antioxidant capacity in eggplant (Solanum melongena L.) seedlings during a short-term chilling stress and subsequent recovery were investigated. The eggplant seedlings pretreated with 0.1 μM EBR were exposed to 5 °C for 12 h in the dark and then recovered under a normal temperature for another 12 h. The results show that EBR alleviated a chilling-induced oxidative stress reflected by a decrease in malondialdehyde and H2O2 content, O2 .- production rate, and an increase in chlorophyll content and activities of superoxide dismutase, guaicol peroxidase, and catalase. The EBR pretreatment also promoted activities of ascorbate peroxidase, glutathione reductase, dehydroascorbate reductase, as well as the accumulation of ascorbic acid (AsA) and glutathione (GSH) involved in the AsA-GSH cycle. Moreover, after the12 h recovery, EBR plus chilling treated leaves showed no significant changes in content of MDA and chlorophyll a, lower reactive oxygen species (ROS) content, and greater activities of AsA-GSH cycle enzymes than in seedlings only chilled. The results suggest that EBR alleviated an oxidative damage caused by the dark chilling stress and accelerated a recovery rate mainly through increasing the ROS scavenging system including the AsA-GSH cycle.

This study was carried out to understand the mechanism of protection of plants under cold stress by exogenous 24-epibrassinolide (EBR). The eggplant (Solanum melongena L.) seedlings were pretreated with five concentrations of EBR (0, 0.05, 0.1, 0.2 and 0.4 °M) and then exposed to day/night temperatures of 10/5 °C for 8 d. The results show that EBR, especially 0.1 °M EBR, dramatically alleviated growth suppression and a decrease in chlorophyll content and photosynthetic rate caused by the cold stress. In addition, EBR also decreased malondialdehyde content and O2 .- production rate induced by the cold stress, and increased the activities of superoxide dismutase, guaiacol peroxidase, catalase, and ascorbate peroxidase, and proline content. The results of the present study suggest that exogenous EBR could improve cold tolerance of eggplant by regulating photosynthesis and antioxidative systems.

The current understanding of the responses of soil respiration (Rs) to soil temperature (Ts) and soil moisture is limited for desert ecosystems. Soil CO2 efflux from a desert shrub ecosystem was measured continuously with automated chambers in Ningxia, northwest China, from June to October 2012. The diurnal responses of Rs to Ts were affected by soil moisture. The diel variation in Rs was strongly related to Ts at 10 cm depth under moderate and high volumetric soil water content (VWC), unlike under low VWC. Ts typically lagged Rs by 3–4 h. However, the lag time varied in relation to VWC, showing increased lag times under low VWC. Over the seasonal cycle, daily mean Rs was correlated positively with Ts, if VWC was higher than 0.08 m3 m−3. Under lower VWC, it became decoupled from Ts. The annual temperature sensitivity of Rs (Q10) was 1.5. The short-term sensitivity of Rs to Ts varied significantly over the seasonal cycle, and correlated negatively with Ts and positively with VWC. Our results highlight the biological causes of diel hysteresis between Rs and Ts, and that the response of Rs to soil moisture may result in negative feedback to climate warming in desert ecosystems. Thus, global carbon cycle models should account the interactive effects of Ts and VWC on Rs in desert ecosystems.

The effects of exogenous 24-epibrassinolide (EBR) on the growth, oxidative damage, antioxidant system and ion contents in eggplant (Solanum melongena L.) seedlings under salt stress were investigated. Eggplant seedlings were exposed to 90 mM NaCl with 0, 0.025, 0.05, 0.10 and 0.20 mg dm-3 EBR for 10 d. EBR, especially at concentration 0.05 mg dm-3, alleviated growth suppression caused by NaCl stress, decreased electrolyte leakage, superoxide production and content of malondialdehyde and H2O2 in NaCl-treated plants. EBR also increased activities of superoxide dismutase, guaiacol peroxidase, catalase and ascorbate peroxidase and the contents of ascorbic acid and reduced glutathione. Furthermore, we also found that Na+, Cl- contents were decreased, K+, Ca2+ contents and K+/Na+, Ca2+/Na+ ratios were increased in the presence of EBR under salt stress.

BackgroundHIV protease inhibitors (PI) are core components of Highly Active Antiretroviral Therapy (HAART), the most effective treatment for HIV infection currently available. However, HIV PIs have now been linked to lipodystrophy and dyslipidemia, which are major risk factors for cardiovascular disease and metabolic syndrome. Our previous studies have shown that HIV PIs activate endoplasmic reticulum (ER) stress and disrupt lipid metabolism in hepatocytes and macrophages. Yet, little is known on how HIV PIs disrupt lipid metabolism in adipocytes, a major cell type involved in the pathogenesis of metabolic syndrome.Methodology and principal findingsCultured and primary mouse adipocytes and human adipocytes were used to examine the effect of frequently used HIV PIs in the clinic, lopinavir/ritonavir, on adipocyte differentiation and further identify the underlying molecular mechanism of HIV PI-induced dysregulation of lipid metabolism in adipocytes. The results indicated that lopinavir alone or in combination with ritonavir, significantly activated the ER stress response, inhibited cell differentiation, and induced cell apoptosis in adipocytes. In addition, HIV PI-induced ER stress was closely linked to inhibition of autophagy activity. We also identified through the use of primary adipocytes of CHOP(-/-) mice that CHOP, the major transcriptional factor of the ER stress signaling pathway, is involved in lopinavir/ritonavir-induced inhibition of cell differentiation in adipocytes. In addition, lopinavir/ritonavir-induced ER stress appears to be associated with inhibition of autophagy activity in adipocytes.Conclusion and significanceActivation of ER stress and impairment of autophagy activity are involved in HIV PI-induced dysregulation of lipid metabolism in adipocytes. The key components of ER stress and autophagy signaling pathways are potential therapeutic targets for HIV PI-induced metabolic side effects in HIV patients.

A double-channel methane (CH 4 ) sensor was developed using a dual-pass multipass cell (DP-MPC) and a novel method that combines averaging dual-channel concentration signals with optimized detector gain configuration. This DP-MPC features two input/output coupling holes, resulting in absorption path lengths of approximately 95.8 m and 35.8 m, respectively. By optimizing the photodetector gain configuration and averaging the dual-channel concentration signals, the detection performance of the sensor was further enhanced. Allan deviation analysis indicated that after optimizing the detector gain, the measurement precision after dual-channel averaging reaches 21 ppb with an integration time of 1 s at a concentration of 2 ppm CH 4 , which is approximately 1.4 times higher than the measurement precision of the long-path channel (31 ppb) and short-path channel (30 ppb). The time required to achieve a measurement precision of 21 ppb is 2.4 s for the long-path channel and 2.1 s for the short-path channel. The response speed of the dual-channel averaging is approximately 2 times that of any single channel. Meanwhile, the sensor demonstrated its stability and reliability through continuous outdoor atmospheric CH 4 measurements.

Background: In certain situations, masks are worn during sleep to prevent respiratory infections. However, the effects of mask wearing on cardiopulmonary function during sleep are unknown. This study aimed to determine whether wearing masks during sleep has an impact on cardiopulmonary function, including in patients with obstructive sleep apnea., Methods: This was a prospective, randomized crossover-controlled trial. The effects of wearing surgical masks or N95 respirators on cardiopulmonary function were measured in healthy subjects and patients with mild-moderate obstructive sleep apnea. Sleep breathing parameters were monitored during nocturnal sleep using a sleep monitor, and subjective feelings about mask wearing were assessed using a questionnaire., Results: Wearing masks during sleep at night did not significantly impact sleep breathing parameters. Furthermore, there were no significant differences in heart rate, blood oxygenation, and blood pressure before and after wearing masks. However, wearing masks, especially the N95 mask, had an adverse impact on sleep quality, and masks were found to be subjectively uncomfortable., Conclusion: Wearing masks during sleep at night does not adversely affect cardiopulmonary function but they can be uncomfortable, especially the N95 mask. Thus, in circumstances where wearing N95 masks during nocturnal sleep proves intolerable, we recommend the use of surgical masks as a more comfortable alternative., (Copyright © 2024 Elsevier Inc. Intermittent hypoxia (IH) precipitates pulmonary vasoconstriction, culminating in the onset of pulmonary hypertension (PH) among individuals afflicted with sleep apnea. While Nuclear receptor subfamily 1 group D member 1 (Nr1d1) is progressively recognized as pivotal regulator of cellular physiology, the role in the pathogenesis of IH-induced PH remains largely uncharted. The expression of Nr1d1 was examined in IH-induced rodent PH and in IH-treated PASMCs. To elucidate the contribution of Nr1d1 to the development of IH-induced PH, we employed siRNA to modulate Nr1d1 expression in vitro and employed serotype 1 adeno-associated virus (AAV1) in vivo. Nr1d1 levels were elevated in IH-induced rodents PH lung tissues and IH-treated PASMCs. Knocking down Nr1d1 by AAV1 effectively inhibited PH progression in chronic IH-induced PH models. Mechanistic investigations identified dual specificity phosphatase 1 (Dusp1), as a direct target that Nr1d1 trans-repressed, mediating Nr1d1's regulatory influence on Erk1/2/Drp1 signaling. Nr1d1 deficiency ameliorates mitochondrial dysfunction and fission by restoring Dusp1 dysregulation and Drp1 phosphorylation. Activation of Erk1/2 with PMA reversed the Dusp1-mediated regulation of Drp1 phosphorylation, indicating the involvement of the Erk1/2 pathway in Drp1 phosphorylation controlled by Dusp1. Meanwhile, intermittent hypoxia induced more severe PH in Dusp1 knockout mice compared with wild-type mice. Background: The clinical and prognostic characteristics of mild-to-moderate COPD with and without emphysema remain inadequately investigated., Research Question: Do the clinical and prognostic characteristics differ between mild- to-moderate COPD with and without emphysema?, Study Design and Methods: We obtained clinical data of 989 participants with mild-to-moderate COPD from the Subpopulations and Intermediate Outcome Measures in COPD Study (SPIROMICS). They were categorized into 2 groups based on their baseline low-attention lung voxels with a density < -950 Hounsfield units of < 5% on CT scans: mild-to-moderate COPD with emphysema (EC) group and mild-to-moderate COPD without emphysema (NEC) group. Linear mixed-effects models were used to assess the differences in the decline of lung function, health-related quality of life, and quantitative CT indexes between these 2 groups. Zero-inflated negative binomial regressions were used to evaluate the rates of acute respiratory exacerbations between the groups., Results: Among participants with mild-to-moderate COPD, 428 (43.3%) exhibited emphysema on CT scans. The annual decline in FEV 1 was -56.1 mL/y for the EC group and -46.9 mL/y for the NEC group, with a nonsignificant between-group difference of 9.1 mL/ (95% CI, -24.0 to 5.7 mL/y). The rate of emphysema progression in the EC group was significantly lower than in the NEC group (-0.173%; 95% CI, -0.252 to -0.094). The EC group also showed a more pronounced annual increase in the St. George's Respiratory questionnaire score (0.9 points) compared with the NEC group. The EC group had a higher rate of acute respiratory exacerbations (0.36 per person-year) than the NEC group (0.25 per person-year), with a rate ratio of 1.42 (95% CI, 1.27-1.54)., Interpretation: The EC group did not have accelerated rates of decline in FEV 1 , but they experienced significantly worse health-related quality of life and a higher rate of acute respiratory exacerbations. Poly (ADP-ribose) polymerase (PARP) 1 and 2 enzymatic inhibitors (PARPi) are promising cancer treatments. But recently, their use has been hindered by unexplained severe anemia and treatment-related leukemia. In addition to enzymatic inhibition, PARPi also trap PARP1 and 2 at DNA lesions. Here we report that, unlike Parp2 -/- mice, which develop normally, mice expressing catalytically inactive Parp2 (E534A and Parp2 EA/EA ) succumb to Tp53- and Chk2-dependent erythropoietic failure in utero, mirroring Lig1 -/- mice. While DNA damage mainly activates PARP1, we demonstrate that DNA replication activates PARP2 robustly. PARP2 is selectively recruited and activated by 5'-phosphorylated nicks (5'p-nicks), including those between Okazaki fragments, resolved by ligase 1 (Lig1) and Lig3. Inactive PARP2, but not its active form or absence, impedes Lig1- and Lig3-mediated ligation, causing dose-dependent replication fork collapse, which is detrimental to erythroblasts with ultra-fast forks. Background: Post-acute sequelae of COVID-19 (PASC) is incurring a huge health and economic burden worldwide. There is currently no effective treatment or recommended drug for PASC., Methods: This prospective randomized controlled study was conducted in a hospital in China. The effect of intermittent hypoxia exposure (IHE; 5-min hypoxia alternating with 5-min normal air, repeated five times) on dyspnea and fatigue was investigated in patients meeting the NICE definition of PASC. Patients were computationally randomized to receive normoxia exposure (NE) and routine therapy or IHE and routine therapy. Six-minute walk distance (6MWD) and spirometry were tested before and after the interventions; the Borg Dyspnea Scale (Borg) and the modified Medical Research Council Dyspnea Scale (mMRC) were used to assess dyspnea; and the Fatigue Assessment Scale (FAS) and the Chalder Fatigue Scale-11 (CFQ-11) were used to assess fatigue. The study was registered in the Chinese Clinical Trial Registry (ChiCTR2300070565)., Findings: Ninety-five participants (33 males and 62 females) were recruited between March 1, 2023 and December 30, 2023. Forty-seven patients in the IHE group received 10.0 (9.0, 15.0) days of IHE, and 48 patients in NE group received 10.0 (8.0, 12.0) days of NE. 6MWD, forced vital capacity (FVC), FVC %pred, forced expiratory volume in 1 s (FEV 1 ), FEV 1 %pred, tidal volume (V T ), and dyspnea and fatigue scales markedly improved after IHE (p < 0.05), and improvements were greater than in the NE group (all p < 0.05). Furthermore, participants in IHE group had better subjective improvements in dyspnea and fatigue than those in the NE group (p < 0.05). Compared with <10 days of IHE, ≥10 days of IHE had a greater impact on 6MWD, FVC, FEV 1 , FEV 1 %pred, V T , FAS, and CFQ-11. This prospective, randomized, controlled clinical trial assessed the therapeutic effects of major ozone autohemotherapy (O 3 -MAH) in patients with post-acute sequelae of COVID-19 (PASC). Seventy-three eligible participants were randomly assigned to an O 3 -MAH plus conventional therapy group (n = 35) or a conventional therapy alone group (n = 38). Symptom score, pulmonary function, 6-minute walk distance (6MWD), and hematological, biochemical, and immunological parameters were evaluated before and after the interventions. Both groups demonstrated improvements in various parameters post-intervention, but efficacy was greater in the O 3 -MAH group than the conventional treatment group; with intervention effectiveness defined as a ≥ 50 % reduction in symptom score, 25 of 35 patients (71 %) responded to O 3 -MAH, while 17/38 patients (45 %) responded to conventional treatment alone (P = 0.0325). Significant improvements in symptom scores (P = 0.0478), tidal volume (P = 0.0374), predicted 6MWD (P = 0.0032), and coagulation and inflammatory indicators were noted in the O 3 -MAH group compared with the conventional treatment group. O 3 -MAH was more likely to be effective in patients with elevated CRP levels. Furthermore, O 3 -MAH markedly improved cellular immunity, and this improvement became more pronounced with extended treatment duration. In summary, combining O 3 -MAH with conventional treatment was more effective than conventional therapy alone in improving symptoms, pulmonary function, inflammation, coagulation, and cellular immunity in patients with PASC. Zinc-sulfur (Zn-S) batteries exhibit a high theoretical energy density, nontoxicity, and cost-effectiveness, demonstrating significant potential for integration into large-scale energy storage systems. However, the phenomenon of polysulfide (including dissolved S 8 and S x ) shuttling is a major issue that results in rapid capacity decay and a short lifespan, limiting the practical performance of sulfur-based batteries. Herein, we fabricated an ionic covalent organic framework (iCOF) membrane as an active separator for the Zn-S battery. Sulfonic acid groups were introduced to the COF membrane, providing abundant negative charge sites in its pore wall. By combining size sieving and charge interaction between the polysulfide and pore wall, the iCOF membrane inhibited the crossover of polysulfides to the Zn metal anode without affecting the transport of metal ions. The Zn-S battery with the iCOF membrane as the separator shows a high-performance and low attenuation rate of 0.05% per cycle over 300 cycles at 2.5 A g 2- . Capturing carbon dioxide (CO 2 ) from flue gases is a crucial step towards reducing CO 2 emissions. Among the various carbon capture methods, facilitated transport membranes (FTMs) have emerged as a promising technology for CO 2 capture owing to their high efficiency and low energy consumption in separating CO 2 . However, FTMs still face the challenge of losing mobile carriers due to weak interaction between the carriers and membrane matrix. Herein, we report a sulfonated chitosan (SCS) gel membrane with confined amine carriers for effective CO 2 capture. In this structure, diethylenetriamine (DETA) as a CO 2 -mobile carrier is confined within the SCS gel membrane via electrostatic forces, which can react reversibly with CO 2 and thus greatly facilitate its transport. The SCS ion gel membrane allows for the fast diffusion of amine carriers within it while blocking the diffusion of nonreactive gases, like N 2 . Thus, the prepared membrane exhibits exceptional CO 2 separation capabilities when tested under simulated flue gas conditions with CO 2 permeance of 1155 GPU and an ultra-high CO 2 /N 2 selectivity of above 550. Moreover, the membrane retains a stable separation performance during the 170 h continuous test. A novel method is introduced to improve the detection performance of photoacoustic spectroscopy for trace gas detection. For effectively suppressing various types of noise, this method integrates photoacoustic spectroscopy with residual networks model which encompasses a total of 40 weighted layers. Firstly, this approach was employed to accurately retrieve methane concentrations at various levels. Secondly, the analysis of the signal-to-noise ratio (SNR) of multiple sets of photoacoustic spectroscopy signals revealed significant enhancement. The SNR was improved from 21 to 805, 52-962, 98-944, 188-933, 310-941, and 587-936 across the different concentrations, respectively, as a result of the application of the residual networks. Finally, further exploration for the measurement precision and stability of photoacoustic spectroscopy system utilizing residual networks was carried out. The measurement precision of 0.0626 ppm was obtained and the minimum detectable limit was found to be 1.47 ppb. Compared to traditional photoacoustic spectroscopy method, an approximately 46-fold improvement in detection limit and 69-fold enhancement in measurement precision were achieved, respectively. Ataxia telangiectasia (A-T) is a rare genetic disorder characterized by neurological defects, immunodeficiency, cancer predisposition, radiosensitivity, decreased blood vessel integrity, and diabetes. ATM, the protein mutated in A-T, responds to DNA damage and oxidative stress, but its functional relationship to the progressive clinical manifestation of A-T is not understood. CD98HC chaperones cystine/glutamate (x c - ) and cationic/neutral amino acid (y + L) antiporters to the cell membrane, and CD98HC phosphorylation by ATM accelerates membrane localization to acutely increase amino acid transport. Loss of ATM impacts tissues reliant on SLC family antiporters relevant to A-T phenotypes, such as endothelial cells (telangiectasia) and pancreatic α-cells (fatty liver and diabetes) with toxic glutamate accumulation. Bypassing the antiporters restores intracellular metabolic balance both in ATM-deficient cells and mouse models. Obstructive sleep apnea (OSA) incurs a huge individual, societal, and economic burden. Specific and selective targeting of hypoglossal motor neurons could be an effective means to treat OSA. Bioluminescent-optogenetics (BL-OG) is a novel genetic regulatory approach in which luminopsins, fusion proteins of light-generating luciferase and light-sensing ion channels, increase neuronal excitability when exposed to a suitable substrate. Here we develop and validate the feasibility of BL-OG for sleep-disordered breathing (SDB). Upon confirming that diet-induced obese mice represent an excellent SDB model, we employed a method of targeting the hypoglossal nucleus (12 N) by peripherally injecting retrogradely transported rAAV2/Retro. With AAV transduction, the eLMO3 protein is expressed in hypoglossal motor neurons (HMN); administration of CTZ results in production of bioluminescence that in turn activates the tethered channelrhodopsin, leading to an increase in the firing of HMN and a 2.7 ± 0.8-fold increase in phasic activity of the genioglossus muscle, a 7.6 ± 1.8-fold increase in tonic activity, and improvements in hypoventilation and apnea index without impacting sleep structure. This is therefore the first study to leverage the rAAV2/Retro vector to execute the BL-OG approach in SDB, which amplified genioglossus muscle discharge activity and increased airflow in mice after activation. Efficient recovery of valuable metals from copper smelting slag (CSS) can not only alleviate the pressure from resource scarcity, but also has important practical significance for the realization of green and sustainable production in the copper smelting industry. In this paper, a composite medium of FeS-O 2 is used as a synergistic modifier to transform the solid-state valuable metals in CSS into leachable state of sulphates, and achieves efficient and comprehensive recovery of zinc and copper through neutral leaching. XRD, FTIR, XPS, etc and comparative analysis methods are used to deeply analyze the characteristics of occurrence phase and transformation rules of valuable metal in CSS, roasted slag and leached slag. The results show under the optimal roasting conditions of T Roasting  = 650 °C, M (copper slag) : M (FeS)  = 1:1, V (O2) : V (Ar)  = 1:6 and t Holding  = 90 min, the recovery rate for zinc is approximately 95.1 %, and that for copper is 99.3 %, almost all of which is recovered. Intermittent hypoxia (IH) and intermittent transcutaneous electrical stimulation (ITES) might benefit patients with obstructive sleep apnea (OSA). However, the therapeutic value of combined IH and ITES in OSA is unknown. In this prospective, randomized, controlled crossover study, normoxia (air exposure for 50 min before sleep and sham stimulation for 6 h during sleep), IH (5 repeats of 5 min 10-12 % O 2 alternating with 5 min air for 50 min, and sham stimulation for 6 h), ITES (air exposure for 50 min and 6 repeats of 30 min transcutaneous electrical stimulation alternating with 30 min of sham stimulation for 6 h), and IH&ITES (10-12 % O 2 alternating with air for 50 min and transcutaneous electrical stimulation alternating with sham stimulation for 6 h) were administered to patients with OSA over four single-night sessions. The primary endpoint was difference in OSA severity between the interventions according to apnea-hypopnea index (AHI) and oxygen desaturation index (ODI). The efficacy was response to IH, ITES, IH&ITES defined as a ≥50 % reduction in AHI compared with normoxia. Twenty participants (17 male, 3 female) completed the trial. The median (IQR) AHI decreased from 14.5 (10.8, 17.5) events/h with normoxia to 6.9 (3.9, 14.8) events/h with IH (p=0.020), 5.7 (3.4, 9.1) events/h with ITES (p=0.001), and 3.5 (1.8, 6.4) events/h with IH&ITES (p=0.001). AHI was significantly different between IH and IH&ITES (p=0.042) but not between ITES and IH&ITES (p=0.850). For mild-moderate OSA (n=17), IH, ITES, and IH&ITES had a significant effect on AHI (p=0.013, p=0.001, p=0.001, respectively) compared with normoxia, but there were no differences in post hoc pairwise comparisons between intervention groups. No serious adverse events were observed. In conclusion, IH, ITES, and IH&ITES significantly reduced OSA severity. IH&ITES showed better efficacy in mild-moderate OSA than IH and was comparable to ITES. Background: The mpox pandemic has caused widespread public concern around the world. The spread of misinformation through the internet and social media could lead to an infodemic that poses challenges to mpox control., Objective: This review aims to summarize mpox-related infodemiology studies to determine the characteristics, influence, prevention, and control measures of the mpox infodemic and propose prospects for future research., Methods: The scoping review was conducted based on a structured 5-step methodological framework. A comprehensive search for mpox-related infodemiology studies was performed using PubMed, Web of Science, Embase, and Scopus, with searches completed by April 30, 2024. After study selection and data extraction, the main topics of the mpox infodemic were categorized and summarized in 4 aspects, including a trend analysis of online information search volume, content topics of mpox-related online posts and comments, emotional and sentiment characteristics of online content, and prevention and control measures for the mpox infodemic., Results: A total of 1607 articles were retrieved from the databases according to the keywords, and 61 studies were included in the final analysis. After the World Health Organization's declaration of an mpox public health emergency of international concern in July 2022, the number of related studies began growing rapidly. Google was the most widely used search engine platform (9/61, 15%), and Twitter was the most used social media app (32/61, 52%) for researchers. Researchers from 33 countries were concerned about mpox infodemic-related topics. Among them, the top 3 countries for article publication were the United States (27 studies), India (9 studies), and the United Kingdom (7 studies). Studies of online information search trends showed that mpox-related online search volume skyrocketed at the beginning of the mpox outbreak, especially when the World Health Organization provided important declarations. There was a large amount of misinformation with negative sentiment and discriminatory and hostile content against gay, bisexual, and other men who have sex with men. Given the characteristics of the mpox infodemic, the studies provided several positive prevention and control measures, including the timely and active publishing of professional, high-quality, and easy-to-understand information online; strengthening surveillance and early warning for the infodemic based on internet data; and taking measures to protect key populations from the harm of the mpox infodemic., Conclusions: This comprehensive summary of evidence from previous mpox infodemiology studies is valuable for understanding the characteristics of the mpox infodemic and for formulating prevention and control measures. The non-homologous end-joining (NHEJ) pathway is critical for DNA double-strand break repair and is essential for lymphocyte development and maturation. The Ku70/Ku80 heterodimer (KU) binds to DNA ends, initiating NHEJ and recruiting additional factors, including DNA-dependent protein kinase catalytic subunit (DNA-PKcs) that caps the ends and pushes KU inward. The C-terminus of Ku70 in higher eukaryotes includes a flexible linker and a SAP domain, whose physiological role remains poorly understood. To investigate this, we generated a mouse model with knock-in deletion of the SAP domain ( Ku70 ΔSAP/ΔSAP ). Ku70 ΔSAP supports KU stability and its recruitment to DNA damage sites in vivo . In contrast to the growth retardation and immunodeficiency seen in Ku70 -/- mice, Ku70 ΔSAP/ΔSAP mice show no defects in lymphocyte development and maturation. Structural modeling of KU on long dsDNA, but not dsRNA suggests that the SAP domain can bind to an adjacent major groove, where it can limit KU's rotation and lateral movement along the dsDNA. Accordingly, in the absence of DNA-PKcs that caps the ends, Ku70 ΔSAP fails to support stable DNA damage-induced KU foci. In DNA-PKcs -/- mice, Ku70 ΔSAP abrogates the leaky T cell development and reduces both the qualitative and quantitative aspects of residual V(D)J recombination. In the absence of DNA-PKcs, purified Ku70 ΔSAP has reduced affinity for DNA ends and dissociates more readily at lower concentration and accumulated as multimers at high concentration. These findings revealed a physiological role of the SAP domain in NHEJ by restricting KU rotation and lateral movement on DNA that is largely masked by DNA-PKcs., Highlight: Ku70 is a conserved non-homologous end-joining (NHEJ) factor. Using genetically engineered mouse models and biochemical analyses, our study uncovered a previously unappreciated role of the C-terminal SAP domain of Ku70 in limiting the lateral movement of KU on DNA ends and ensuring end protection. The presence of DNA-PKcs partially masks this role of the SAP domain.

DNA strand breaks activate Poly(ADP-ribose) polymerase (PARP) 1 and 2, which use NAD+ as the substrate to covalently conjugate ADP-ribose on themselves and other proteins (e.g., Histone) to promote chromatin relaxation and recruit additional DNA repair factors. Enzymatic inhibitors of PARP1 and PARP2 (PARPi) are promising cancer therapy agents that selectively target BRCA1- or BRCA2- deficient cancers. As immediate early responders to DNA strand breaks with robust activities, PARP1 and PARP2 normally form transient foci (<10 minutes) at the micro-irradiation-induced DNA lesions. In addition to enzymatic inhibition, PARPi also extend the presence of PARP1 and PARP2 at DNA lesions, including at replication forks, where they may post a physical block for subsequent repair and DNA replication. The dynamic nature of PARP1 and PARP2 foci made live cell imaging a unique platform to detect subtle changes and the functional interaction among PARP1, PARP2, and their regulators. Background: Yunnan province borders Myanmar, Laos, and Vietnam, giving it one of the longest borders in China. We aimed to determine the trends in prevalence and impact of COVID-19 on depressive symptoms among adolescents (12-18 years) from 2018 to 2022 in Yunnan, southwest China., Objective: We evaluated the impact of the COVID-19 epidemic on adolescents' mental health, with the aim of reducing the effect of psychological emergency syndrome and promoting healthy, happy adolescent growth., Methods: This longitudinal, observational study used Students' Health Survey data on adolescents' depressive symptoms from 2018 to 2022 (before and during COVID-19) in Yunnan. We used multistage, stratified sampling in 3 prefectures in 2018 and 16 prefectures from 2019 to 2022. In each prefecture, the study population was classified by gender and residence (urban or rural), and each group was of equal size. Depressive symptoms were diagnosed based on Center for Epidemiological Studies Depression Scale (CES-D) scores. We used ANOVA to assess the differences in mean CES-D scores stratified by gender, age, residence, grade, and ethnicity. Chi-square tests were used to compare depressive symptoms by different variables. For comparability, the age-standard and gender-standard population prevalences were calculated using the 2010 China Census as the standard population. The association between COVID-19 and the risk of a standardized prevalence of depressive symptoms was identified using unconditional logistic regression analysis., Results: The standardized prevalence of depressive symptoms for all participants was 32.98%: 28.26% in 2018, 30.89% in 2019, 29.81% in 2020, 28.77% in 2021, 36.33% in 2022. The prevalences were 30.49% before COVID-19,29.29% in early COVID-19, and 36.33% during the COVID-19 pandemic. Compared with before COVID-19, the risks of depressive symptoms were 0.793 (95% CI 0.772-0.814) times higher in early COVID-19 and 1.071 (95% CI 1.042-1.100) times higher than during COVID-19. The average annual increase in depressive symptoms was 1.61%. During the epidemic, the prevalence of depressive symptoms in girls (36.87%) was higher than that in boys (28.64%), and the acceleration rate of girls was faster than that of boys. The prevalences of depressive symptoms and acceleration rates by age group were as follows: 27.14% and 1.09% (12-13 years), 33.99% and 1.8% (14-15 years), 36.59% and 1.65% (16-18 years). Prevalences did not differ between Han (32.89%) and minority (33.10%) populations. However, the acceleration rate was faster for the former than for the latter. The rate for senior high school students was the highest (34.94%). However, the acceleration rate for vocational high school students was the fastest (2.88%), followed by that for junior high school students (2.32%). Rural residents (35.10%) had a higher prevalence and faster acceleration than urban residents (30.16%)., Conclusions: From 2018 to 2022, there was a significant, continuous increase in the prevalence of depressive symptoms among adolescents in Yunnan, China, especially during the COVID-19 pandemic. This represents an emergency public health problem that should be given more attention. Purpose: SARS-CoV-2 can invade the thyroid gland. This study was to delineate the risk of thyroid dysfunction amidst the prevalence of the Omicron variant, and to investigate the correlation between thyroid function and Coronavirus disease 2019 (COVID-19) outcomes. The study also aimed to ascertain whether thyroid dysfunction persisted during COVID-19 recovery phase., Methods: This was a retrospective cohort study. COVID-19 patients from the Renmin Hospital of Wuhan University, China during the epidemic of Omicron variants were included, and their thyroid function were analyzed in groups., Results: A history of thyroid disease was not associated with COVID-19 outcomes. COVID-19 can lead to a bimodal distribution of thyroid dysfunction. The severity of COVID-19 was inversely proportional to the levels of thyroid- stimulating hormone (TSH), free triiodothyronine (FT3) and free thyroxine (FT4), leading to a higher prevalence of thyroid dysfunction. Severe COVID-19 was a risk factor for euthyroid sick syndrome (ESS) (OR=22.5, 95% CI, 12.1 - 45.6). Neutrophil to lymphocyte ratio mediated the association between severe COVID-19 and ESS (mediation effect ratio = 41.3%, p < 0.001). ESS and decreased indicators of thyroid function were associated with COVID-19 mortality, while high levels of FT3 and FT4 exhibited a protective effect against death. This effect was more significant in women (p < 0.05). During the recovery period, hyperthyroidism was quite uncommon, while a small percentage of individuals (7.7%) continued to exhibit hypothyroidism., Conclusion: COVID-19 severity was linked to thyroid dysfunction. Severe COVID-19 increased the risk of ESS, which was associated with COVID-19 mortality. In the past, research on the cognitive neural mechanism of second language (L2) learners' processing time information has focused on Indo-European languages. It has also focused on the temporal category expressed by morphological changes. However, there has been a lack of research on L2 learners' various time coding means, especially for Mandarin, which lacks morphological changes. Using event-related potentials (ERPs), we investigated the cognitive neural mechanism of L2 learners with native Indonesian background in processing two time coding means (time adverbs and aspect markers) in Chinese. Indonesian has time adverb encoding time information similar to that of Chinese, but there are no aspect markers similar to Chinese in Indonesian. We measured ERPs time locked to the time adverb " (cengjing)" and the aspect marker "verb + (verb + guo)" in two different conditions, i.e., a control condition (the correct sentence) and a temporal information violation. The experimental results showed that the native speaker group induced the biphasic N400-P600 effect under the condition of time adverb violation, and induced P600 under the condition of the aspect marker "verb + (verb + guo)" violation. Indonesian L2 learners of Chinese only elicited P600 for the violation of time adverbs, and there was no statistically significant N400 similar to that of Chinese native speakers. In the case of aspect marker violation, we observed no significant ERPs component for the Indonesian L2 learners of Chinese. Both groups of subjects induced elicited a widely distributed and sustained negativity on the post-critical words after "verb + (verb + guo)" and "(cengjing)". Iron-nitrogen-carbon single-atom catalysts derived from zeolitic-imidazolate-framework-8 (ZIF-8) have presented its great potential for the oxygen reduction reaction (ORR) in Zn-air batteries (ZABs). However, due to insufficient active Fe-N sites, its ORR activity is inferior to Pt-based catalysts. Herein, a carboxylate (OAc) linker strategy is proposed to design a ZIF-8-derived FeNCOAc catalyst with abundant accessible Fe-N 4 single-atom sites. Except that imidazole groups can coordinate with Fe ions, the OAc linker on the unsaturated coordination Zn nodes can anchor and coordinate with more Fe ions, resulting in a significant increase in Fe-N 4 site density. Meanwhile, the corrosion of carbon skeleton by OAc oxidation during heat-treatment leads to improved porosity of catalyst. Benefitting from the highly dense Fe-N 4 sites and hierarchical pores, the FeNCOAc endows superior performance in alkaline medium (E 1/2  = 0.906 V), which is confirmed by density functional theory calculation results. Meanwhile, the assembled liquid ZAB delivers a favorable peak power density of 173.9 mW cm -2 , and a high specific capacity of 770.9 mAh g -1 as well as outstanding durability. In disordered glass structures, the structural modelling and analyses based on local experimental data are not yet established. Here we investigate the icosahedral short-range order (SRO) in a Zr-Pt metallic glass using local structural modelling, which is a reverse Monte Carlo simulation dedicated to two-dimensional angstrom-beam electron diffraction (ABED) patterns, and local pair distribution function (PDF) analysis. The local structural modelling invariably leads to the icosahedral SRO atomic configurations that are similarly distorted by starting from some different initial configurations. Furthermore, the SRO configurations with 11-13 coordination numbers reproduce almost identical ABED patterns, indicating that these SRO structures are similar to each other. Further local PDF analysis explicitly indicates the presence of the wide distribution of atomic bond distances, which is comparable to the global PDF profile, even at the SRO level. The SRO models based on the conventional MD simulation can be strengthened by comparison with those obtained by the present local structural modelling and local PDF analysis based on the ABED data., (© 2024.
