Abstract

As promising photonic material, phototheranostics can be activated in the laser irradiation range of tumor with sensitivity and spatiotemporal precision. However, it is difficult to completely eradicate solid tumors due to their irregularity and limited laser irradiation area. Herein, multi‐stimulus responsive HA−Ce6@SWNHs were constructed with single‐walled carbon nanohorns (SWNHs) and chlorine e6 (Ce6) modified hyaluronic acid (HA) via non‐covalent binding. This SWNHs‐based phototheranostics not only exhibited water dispersion but also could target tumor and be activated by near‐infrared light for photodynamic therapy (PDT) and photothermal therapy (PTT). Additionally, HA−Ce6@SWNHs could be degraded by hyaluronidase in residual tumor cells, causing HA−Ce6 to fall off the SWNHs surfaces to restore autofluorescence, thus precisely guiding the programmed photodynamic treatments for residual tumor cells after the initial phototherapy. Thus, this work provides a rationally designed multiple‐stimulus‐response strategy to develop smart SWNHs‐based phototheranostics for precise PDT/PTT and post‐treatment imaging‐guided PDT of residual tumor cells. Abstract

Natural biopolymers can be controllably in situsynthesized in organisms and play important roles in biological activities. Inspired by this, the manipulation of in situbiosynthesis of functional polymers in vivowill be an important way to obtain materials for meeting biological requirements. Herein, in situbiosynthesis of functional conjugated polymer at the tumor site was achieved via the utilization of specific tumor microenvironment (TME) characteristics for the first time. Specially, a water-soluble aniline dimer derivative (N-(3-sulfopropyl) p-aminodiphenylamine, SPA) was artfully in situpolymerized into polySPA (PSPA) nanoparticles at the tumor site, which was activated via the catalysis of hydrogen peroxide (H2O2) overexpressed in TME to produce hydroxyl radical (•OH) by coinjected horseradish peroxidase (HRP). Benefiting from outstanding near-infrared (NIR)-II absorption of PSPA, the in situpolymerization process can be validly monitored by photoacoustic (PA) signal at the NIR-II region. Meanwhile, in situpolymerization would induce the size of polymeric materials from small to large, improving the distribution and retention of PSPA at the tumor site. On the combination of NIR-II absorption of PSPA and the size variation induced by polymerization, such polymerization can be applied for tumor-specific NIR-II light mediated PA image and photothermal inhibition of tumors, enhancing the precision and efficacy of tumor phototheranostics. Therefore, the present work opens the way to manipulate TME-activated in situbiosynthesis of functional conjugated polymer at the tumor site for overcoming formidable challenges in tumor theranostics.

74. NIR-II-responsive AuNRs@SiO2–RB@MnO2 nanotheranostic for multimodal imaging-guided CDT/PTT synergistic cancer therapy.

Abstract

Specific tumor-responsive capabilities and efficient synergistic therapeutic performance are the keys to effective tumor treatment. Herein, AuNRs@SiO2–RB@MnO2 was developed as a new type of tumor-responsive nanotheranostic for multimodal imaging and synergistic chemodynamic/photothermal therapy. In AuNRs@SiO2–RB@MnO2, the SiO2 layer wraps the AuNRs, providing light absorption in the second near-infrared (NIR-II) region. The SiO2 layer also adsorbs the MnO2 nanosheets, which have Fenton-like activity, resulting in a fluorescent sensing platform based on the fluorescence quenching properties of MnO2 for rhodamine B dye. The fluorescence can be recovered by the consumption of MnO2 by glutathione, which simultaneously produces Mn2+ in the tumor region. The recovery of fluorescence reflects the consumption of glutathione and the increase in Mn2+, which produces hydroxyl radicals via Fenton-like reaction in the tumor microenvironment to realize chemodynamic therapy. Meanwhile, the AuNRs are a good photothermal reagent that can effectively absorb NIR-II light and convert it into heat energy to kill tumor cells via photothermal therapy. The NIR-II absorption performance of the AuNRs provides good photoacoustic imaging and deep photothermal performance, which is favorable for efficient NIR-II photoacoustic imaging-guided photothermal therapy. As a result, the AuNRs@SiO2–RB@MnO2 nanotheranostic exhibits outstanding imaging and synergistic chemodynamic/photothermal therapeutic performance for tumor imaging and treatment. Abstract

Surface‐enhanced Raman scattering (SERS) has received considerable attention from researchers due to its high molecular specificity, high sensitivity, non‐invasiveness and multiplexing. Recently, various metal substrates have been exploited for SERS analysis and imaging. Among them, gold nanomaterials are important SERS substrates with outstanding surface plasmon resonance effects, structural adjustability and good biocompatibility, making them widely used in biomedical diagnosis and clinical fields. In this minireview, we discuss the latest progress about the application of gold‐based nanomaterials as SERS probes in biomedical research, primarily for in vivo disease diagnosis and imaging. This review mainly includes the basic shapes and morphologies of gold based SERS probes, such as gold nanoparticles (AuNPs), gold nanorods (AuNRs), gold nanostars (AuNSs), as well as other gold nanostructures. Finally, a brief outlook for the future development of SERS technique in the context of efficient diagnostics and therapy guidance is provided. Abstract

Herein, we describe an energy balance strategy between fluorescence and photoacoustic effects by sulfur substitution to transform existing hemicyanine dyes (Cy) into optimized NIRF/PA dual ratiometric scaffolds. Based on this optimized scaffold, we reported the first dual‐ratio response of nitroreductase probe AS‐Cy‐NO2, which allows quantitative visualization of tumor hypoxia in vivo. AS‐Cy‐NO2, composed of a new NIRF/PA scaffold thioxanthene‐hemicyanine (AS‐Cy‐1) and a 4‐nitrobenzene moiety, showed a 10‐fold ratiometric NIRF enhancement (I773/I733) and 2.4‐fold ratiometric PA enhancement (PA730/PA670) upon activation by a biomarker (nitroreductase, NTR) associated with tumor hypoxia. Moreover, the dual ratiometric NIRF/PA imaging accurately quantified the hypoxia extent with high sensitivity and high imaging depth in xenograft breast cancer models. More importantly, the 3D maximal intensity projection (MIP) PA images of the probe can precisely differentiate the highly heterogeneous oxygen distribution in solid tumor. Thus, this study provides a promising NIRF/PA scaffold that may be generalized for the dual ratiometric imaging of other disease‐relevant biomarkers. [ABSTRACT FROM AUTHOR]
