54. Cancer‐associated fibroblasts promote tumor progression by lncRNA‐mediated RUNX2/GDF10 signaling in oral squamous cell carcinoma.

Abstract

Cancer‐associated fibroblasts (CAF) are the most abundant stromal cells in tumor and exert a pro‐tumoral effect in cancer progression. Numerous evidence shows long non‐coding RNA (lncRNA) abnormally regulates gene expression in various cancers. However, little is known about the role of lncRNA in the interaction between CAF and cancer cells. Here, we first identify an uncharacterized lncRNA, LOC100506114, which is significantly upregulated in CAF and is involved in the functional transformation of normal fibroblasts (NF) and CAF. Expression of LOC100506114 enhances the expression of fibroblast activation protein alpha and α‐smooth muscle actin in NF and promotes malignant characteristics of NF and CAF in vivo and in vitro. The profile of gene co‐expression analysis shows that growth differentiation factor 10 (GDF10) is positively correlated with the expression of LOC100506114. CAF promote stromal fibroblast activation and the proliferation and migration of tumor cells by secreting GDF10. Our data demonstrate that lncRNA plays a critical role in the interplay of stromal fibroblasts and tumor cells in oral squamous cell carcinoma. [ABSTRACT FROM AUTHOR]

Abstract

Aiming at the problem that the location and capacity of distributed generation(DG) will have a great impact on power flow distribution of distribution network, a new method of location and capacity optimization of DG access to distribution network is proposed. A planning scheme with distribution network revenue, node voltage deviation and microgrid permeability as objective functions is established. Particle swarm optimization (PSO) algorithm is adopted, that is, the simplex method is used to initialize the PSO algorithm; a control factor is introduced to dynamically adjust the inertia weight to improve the convergence ability of the algorithm; and mutation factor is introduced to enable the algorithm to effectively jump out of the local optimum to find the global optimum solution in the later stage of convergence. An example shows that this method can effectively reduce the network loss and voltage deviation when DG is connected into the distribution network.

Abstract

The dry‐wet climate conditions of an area are mainly determined by the water input (e.g., precipitation, P) and output (e.g., potential evapotranspiration, PET). Based on three dry‐wet indices and the differential equation, this study designed a new attribution method to quantify the contributions of P and PET to the dry‐wet change, so as to determine the dominant factors of the dry‐wet change over whole China (CHINA) and various hydroclimate regions (i.e., arid region, AR; semi‐arid region, SAR; semi‐humid region, SHR; and humid region, HR) during 1960–2018 and six interdecadal periods. Results showed that the overall weak drying in CHINA was dominated by an increase in PET, while climate humidification in AR and HR was dominated by an increase in P, climate drying in SHR was dominated by the reduced P, and the weak drying in SAR was co‐dominated by P and PET during 1960–2018. The dominant factors of dry‐wet change over CHINA and each hydroclimate region varied in different decades. Briefly, climate humidification in CHINA and various hydroclimate regions was basically dominated by P in different decades, and the drying was mainly dominated by P in the 1960s–1980s, but the contribution of PET to climate drying began to become prominent after the 1980s. Besides, we found that P contributed more to dry‐wet change than PET on the station and regional scales, but the contribution of P to climate drying on a national scale was underestimated to some extent due to the regional offset effect of the increase/decrease in P during 1960–2018, as well as in the 1990s. These findings implied that the importance of research on the regional or local scale, and equal attention should be paid to the effects of P and PET changes in the future drought or dry‐wet change research. Abstract

Accumulating evidence has shown that mammalian target of rapamycin (mTOR) pathway and myeloid‐derived suppressor cells (MDSCs) are involved in pathogenesis of inflammatory bowel diseases (IBDs). INK128 is a novel mTOR kinase inhibitor in clinical development. However, the exact roles of MDSCs and INK128 in IBD are unclear. Here, we showed that the INK128 treatment enhanced the resistance of mice to dextran sodium sulfate (DSS)–induced colitis and inhibited the differentiation of MDSCs into macrophages. Moreover, interferon (IFN)‐α level was elevated in INK128‐treated colitis mice. When stimulated with IFN‐α in vitro, MDSCs showed a superior immunosuppression activity. Of note, the regulatory T cells (Tregs) increased but Th1 cells decreased in INK128‐treated colitis mice. These results indicate that mTOR inhibitor INK128 attenuates DSS‐induced colitis via Treg expansion promoted by MDSCs. Our work provides a new evidence that INK128 is potential to be a therapeutic drug on DSS‐induced colitis via regulating MDSCs as well as maintaining Treg expansion. 88. An Effective Vaccination Approach Augments Anti-HIV Systemic and Vaginal Immunity in Mice with Decreased HIV-1 Susceptible 47high CD4 T Cells

Abstract

HIV-1 preferentially infects activated CD4+ T cells expressing 47 integrin and conventional vaccination approaches non-selectively induce immune responses including 47high CD4+ T cells, suggesting that current candidate AIDS vaccines may produce more target cells for HIV-1 and paradoxically enhance HIV-1 infection. Thus it remains a challenge to selectively induce robust anti-HIV immunity without the unwanted HIV-1 susceptible 477high CD4 T cells. Here we describe a vaccination strategy that targets ALDH1a2, a retinoic acid producing enzyme in dendritic cells (DCs). Silencing ALDH1a2 in DCs enhanced the maturation and production of proinflammatory cytokines of DCs and promoted Th1/Th2 differentiation while suppressing Treg. ALDH1a2-silenced DCs effectively downregulated the expression of guthoming receptors 477 and CCR9 on activated T and B lymphocytes. Consequently, intranasal immunization of a lentiviral vaccine encoding ALDH1a2 shRNA and HIV-1 gp140 redirected gp140-specific mucosal T cell and antibody responses from the gut to the vaginal tract, while dramatically enhancing systemic gp140-specific immune responses. We further demonstrated that silencing ALDH1a2 in human DCs resulted in downregulation of 7 expression on activated autologous CD4 T cells. Hence this study provides a unique and effective strategy to induce 47low anti-HIV immune responses.
