Abstract

We propose a model for a magnetic recording channel to estimate the influences of the nonlinear distortion of an MR head on the performance of the PRML system, using a superposition of a magnetic field and a transfer curve obtained by a micro-magnetic model. A readback signal was simulated using a nonlinear model. The simulated signal was compared with an actual readback signal in the analysis of the isolated pulse, frequency response, and extracted dipulse response. We found that the nonlinear model represents actual MR head output satisfactorily, and that the performance of the PR4ML system was affected by the nonlinear distortion of the MR head for a 5% loss of the amplitude margin when the asymmetry was within 10%.

The current study assessed attitudes toward metabolic adverse events among patients with schizophrenia. Methods: A brief questionnaire was constructed to investigate patient recognition of the following broad areas: dietary habits, lifestyle, self-monitoring, knowledge, and medical practice. Between January 2012 and June 2013, questionnaires were sent to patients associated with 520 outpatient facilities and 247 inpatient facilities belonging to the Japan Psychiatric Hospital Association. All of the participants (n=22,072; inpatients =15,170, outpatients =6,902) were diagnosed with schizophrenia based on the Diagnostic and Statistical Manual of Mental Disorders, fourth edition, or the International Classification of Diseases, tenth revision. Results: Approximately 55.0% (8,069/14,669) of inpatients and 44.8% of outpatients (2,978/6,649) reported that they did not exercise at all. Although 60.9% (4,116/6,760) of outpatients reported that they felt obese, only 35.6% (5,261/14,794) of inpatients felt obese. More than half of the inpatients (51.2%; 7,514/14,690) and outpatients (60.8%; 4,086/6,721) hoped to receive regular blood tests to prevent weight gain and diseases such as diabetes. Conclusion: Although more than half of patients hoped to prevent weight gain and diabetes, only a minority of patients were mindful of eating balanced meals and having physical exercise. Educational efforts and the promotion of the best pharmacotherapy and monitoring practices are needed for patients with schizophrenia. Keywords: metabolic syndrome, schizophrenia, attitude, Japan

Abstract

Bipolar disorder (BD) is a common, highly heritable neuropsychiatric disease characterized by recurrent episodes of mania and depression. Lithium is the best-established long-term treatment for BD, even though individual response is highly variable. Evidence suggests that some of this variability has a genetic basis. This is supported by the largest genome-wide association study (GWAS) of lithium response to date conducted by the International Consortium on Lithium Genetics (ConLiGen). Recently, we performed the first genome-wide analysis of the involvement of miRNAs in BD and identified nine BD-associated miRNAs. However, it is unknown whether these miRNAs are also associated with lithium response in BD. In the present study, we therefore tested whether common variants at these nine candidate miRNAs contribute to the variance in lithium response in BD. Furthermore, we systematically analyzed whether any other miRNA in the genome is implicated in the response to lithium. For this purpose, we performed gene-based tests for all known miRNA coding genes in the ConLiGen GWAS dataset (n = 2,563 patients) using a set-based testing approach adapted from the versatile gene-based test for GWAS (VEGAS2). In the candidate approach, miR-499a showed a nominally significant association with lithium response, providing some evidence for involvement in both development and treatment of BD. In the genome-wide miRNA analysis, 71 miRNAs showed nominally significant associations with the dichotomous phenotype and 106 with the continuous trait for treatment response. A total of 15 miRNAs revealed nominal significance in both phenotypes with miR-633 showing the strongest association with the continuous trait (p = 9.80E-04) and miR-607 with the dichotomous phenotype (p = 5.79E-04). No association between miRNAs and treatment response to lithium in BD in either of the tested conditions withstood multiple testing correction. Abstract

IMPORTANCE Lithium is a first-line mood stabilizer for the treatment of bipolar affective disorder (BPAD). However, the efficacy of lithium varies widely, with a nonresponse rate of up to 30%. Biological response markers are lacking. Genetic factors are thought to mediate treatment response to lithium, and there is a previously reported genetic overlap between BPAD and schizophrenia (SCZ). OBJECTIVES To test whether a polygenic score for SCZ is associated with treatment response to lithium in BPAD and to explore the potential molecular underpinnings of this association. DESIGN, SETTING, AND PARTICIPANTS A total of 2586 patients with BPAD who had undergone lithium treatment were genotyped and assessed for long-term response to treatment between 2008 and 2013.Weighted SCZ polygenic scores were computed at different P value thresholds using summary statistics from an international multicenter genome-wide association study (GWAS) of 36 989 individuals with SCZ and genotype data from patients with BPAD from the Consortium on Lithium Genetics. For functional exploration, a cross-trait meta-GWAS and pathway analysis was performed, combining GWAS summary statistics on SCZ and response to treatment with lithium. Data analysis was performed from September 2016 to February 2017. MAIN OUTCOMES AND MEASURES Treatment response to lithiumwas defined on both the categorical and continuous scales using the Retrospective Criteria of Long-Term Treatment Response in Research Subjects with Bipolar Disorder score. The effect measures include odds ratios and the proportion of variance explained. RESULTS Of the 2586 patients in the study (mean [SD] age, 47.2 [13.9] years), 1478 were women and 1108 were men. The polygenic score for SCZ was inversely associated with lithium treatment response in the categorical outcome, at a threshold P < 5 ? 10-2. We have studied the ATV-VCR tape format, which realizes the multi-speed mode, based on the 6mm VCR. Larger data amount are assigned for the slower trick play to realize picture quality in accordance to the human visual characteristic. The tape format and data placement require easy tracking and flexibility of various head structures. The tape format is based on 4-track sequence for easy tracking. Each trick play data has its own data recording area.

70. Characterization of Bulk and Surface Chemical States on Electrochemically Cycled LiFePO4: A Solid State NMR Study

Abstract

Bulk and surface chemical states were both investigated for electrochemically delithiated LixFePO4 using {7}Li and {31}P MAS NMR spectroscopy. The quantitative lithium extraction/insertion from LiFePO4 and the reversible two-phase reaction behavior between LiFePO4 and FePO4 were confirmed on electrochemical operation. The {7}Li and {31}P NMR spectra of the fully charged LixFePO4 evidenced a single Li-poor phase Li0.05FePO4 instead of a biphasic mixture of 0.05LiFePO4 and 0.95FePO4. Simultaneous growth of LixPOyFz was also shown as a surface film component on the charged electrodes, which dynamically increased and decreased in intensity during charging and discharging reactions, respectively. The electrode surface was further characterized with XPS to discuss the surface film formation during electrochemical cycles. Combined chemical state analyzes by NMR and XPS spectroscopies suggested that the degradation of LiPF6 salt occurred from the initial redox cycles, but that of solvent occurred after multiple cycling. The deposition of carbonaceous species would be related to a small capacity fading observed after the multiple charge-discharge cycles.

Abstract

In this work, we consider narrow band partial response (NBPR) systems for (1, 7) RLL code. As a new type of (1, 7) PRML system, a PR(1, 1.5, 0, -1.5, -1) system called a modified MEEPR4 (MMEEPR4) which contains a spectral null below the Nyquist frequency is proposed. We compared the performances of 8/9 (0, 4, 4) PR4, (1, 7) EEPR4, (1, 7) MEEPR4 and (1, 7) MMEEPR4 in recording systems with both electronic and medium noise characterized by Thurlings' model. Medium noise power was corrected using the code magnetic transition probability. Thus the average medium noise power for code-2/3 (1, 7) was at a 20% smaller than that of code-8/9 (0, 4, 4). To verify the practical use of this system, we defined the broader constraints and calculated the error propagation. It is clear that the (1, 7) MEEPR4 system does not satisfy the broader constraints because it generates no quasi-catastrophic trellis sequence. Simulation results indicate that the narrow-band (1, 7) MMEEPR4 system provides a significant improvement over 8/9 (0, 4, 4) PR4 and (1, 7) EEPR4 and avoids the signal band extension for a 2/3 code-rate without a loss in performance.

Abstract

We developed an integrated-Viterbi-detector which can handle PRML and EPRML. Both PR4 and EPR4 equalization can be accepted according to the recording density. The EPRML performance is superior to that of PRML for a normalized recording density above 1.8, but inferior at lower densities. Our experimental results agree with theoretical values and prove that the performance of this Viterbi detector is superior, by 8 dB, to that of conventional peak detectors when the normalized recording density is 2.7.

Abstract

Background Lithium is a first-line treatment in bipolar disorder, but individual response is variable. Previous studies have suggested that lithium response is a heritable trait. However, no genetic markers of treatment response have been reproducibly identified. Methods Here, we report the results of a genome-wide association study of lithium response in 2563 patients collected by 22 participating sites from the International Consortium on Lithium Genetics (ConLiGen). Data from common single nucleotide polymorphisms (SNPs) were tested for association with categorical and continuous ratings of lithium response. Lithium response was measured using a well established scale (Alda scale). Genotyped SNPs were used to generate data at more than 6 million sites, using standard genomic imputation methods. Traits were regressed against genotype dosage. Results were combined across two batches by meta-analysis. Findings A single locus of four linked SNPs on chromosome 21 met genome-wide significance criteria for association with lithium response (rs79663003, p=1·37×10-8; rs78015114, p=1·31×10-8; rs74795342, p=3·31×10-9; and rs75222709, p=3·50×10-9). In an independent, prospective study of 73 patients treated with lithium monotherapy for a period of up to 2 years, carriers of the response-associated alleles had a significantly lower rate of relapse than carriers of the alternate alleles (p=0·03268, hazard ratio 3·8, 95% CI 1·1-13·0). Interpretation The response-associated region contains two genes for long, non-coding RNAs (lncRNAs), AL157359.3 and AL157359.4. LncRNAs are increasingly appreciated as important regulators of gene expression, particularly in the CNS. Confirmed biomarkers of lithium response would constitute an important step forward in the clinical management of bipolar disorder. Abstract

Signal transducer and activator of transcription 3 (STAT3) play key roles in the intracellular signaling pathways of the interleukin (IL)-6 family of cytokines, which exhibit a diverse set of cellular responses, including cell proliferation and differentiation. Dysregulated IL-6/STAT3 signaling is involved in the pathogenesis of several diseases, for example autoimmune diseases and tumors. Type I interferon (IFN) induces the expression of proapoptotic genes and has been used in the clinical treatment of several tumors. In the present study, we found that type I IFN suppressed IL-6/STAT3-mediated transcription and gene expression. Furthermore, a type I IFN-induced protein, Daxx, also suppressed STAT3-mediated transcriptional activation, while overexpression of Daxx inhibited IL-6/STAT3-mediated gene expression. Importantly, small-interfering RNA-mediated reduction of Daxx expression enhanced IL-6/leukemia inhibitory factor (LIF)-induced STAT3-dependent transcription. Co-immunoprecipitation studies revealed a physical interaction between Daxx and STAT3 in transiently transfected 293T cells. We further found that Daxx and STAT3 were co-localized in the nucleus. These results indicate that Daxx may serve as a transcriptional regulator of type I IFN-mediated suppression of the IL-6/STAT3 signaling pathway.
