Abstract

The role of mRNA in the coding of human plasma cholesteryl ester transfer protein (CETP) was investigated due to the discovery of a variant CETP cDNA. The exclusion of a complete sequence otherwise coded by exon nine implied a gene transcription change brought about by alternative RNA splicing confirmed by RNase protection analysis. Such mRNA splicing is present in varying proportions in different human tissues. Proteins encoded from exon nine depleted cDNA showed marked differences when compared to similar proteins encoded by full-length DNA thus showing the effects of alternative splicing of the ninth exon in CETP gene expression.

59. Forming Cooperative Programs for Mainstreaming Preschool Children with Severe Behavior Disorders: Policy, Programmatic, and Procedural Issues.

Abstract

The paper presents an organizational framework to guide the design and implementation of integrated services for preschool children with severe behavior disorders, particularly those programs where cooperative agreements are formed between separate service agencies. A project that mainstreams preschool children with severe behavior disorders into a local day care center is described to illustrate planning decisions made at the interdependent levels of (1) policy, (2) setting, (3) service provider, and (4) child. Suggestions are made and supported by the literature for considering the policy issues of legal mandates, attitudes toward integration, types of integrated experiences, the setting components of philosophies, organizational structures and needs, service provider decisions on program implementation, staff development, parent participation, and child concerns of selection and placement, instructional targets, intervention strategies, and progress evaluation. Abstract

The C19 diterpenoid alkaloids (C19 DTAs) are a large family of natural products, many of which modulate the activity of ion channels in vivoand are therefore of interest for the study of neurological and cardiovascular diseases. The complex architectures of these molecules continue to challenge the state-of-the art in chemical synthesis, particularly with respect to efficient assembly of their polcyclic ring systems. Here, we report the total syntheses of (−)-talatisamine, (−)-liljestrandisine, and (−)-liljestrandinine, three aconitine-type C19 DTAs, using a fragment coupling strategy. Key to this approach is a 1,2-addition/semipinacol rearrangement sequence which efficiently joins two complex fragments and sets an all-carbon quaternary center.

71. Radial polydactyly: putting together evolution, development and clinical anatomy.

Abstract

Evolutionary developmental pathology, a new biological field, connects the study of evolution, development and human pathologies. In radial polydactyly, traditional studies have focused mainly on skeletal anomalies. This study examines anatomical and operative records of 54 consecutive cases of radial polydactyly to investigate whether there is a consistent spatial correlation between muscles, tendons and bones and whether this reflects a link between the mechanisms that generate these structures. The data are explored in the context of two current models of limb development: the modularity and topology models. Autopod (hand) tendons and muscles are more predictable in terms of insertion site, supporting both topology and modularity models. Zeugopod (forearm) tendons are less predictable. Neither model universally predicts the anatomy in radial polydactyly. These observations provide evidence for the complexity of anatomy in radial polydactyly and the difficulty in predicting operative findings based on the level of skeletal duplication alone. Estrogens have significant effects on skin physiology and modulate epidermal keratinocytes, dermal fibroblasts and melanocytes, in addition to skin appendages including the hair follicle and the sebaceous gland. Importantly, skin aging can be significantly delayed by the administration of estrogen. This paper reviews the effects of estrogens on skin and the mechanisms by which estrogens can alleviate the changes due to aging that occur in human skin. The relevance of estrogen replacement therapy (HRT) in postmenopausal women and the potential value of selective estrogen receptor modulators (SERMs) as a therapy for diminishing skin aging are also highlighted.Keywords: estrogen receptors, skin, menopause, SERMs, HRT
