Abstract

Monitoring drug release in vitro and in vivo is of paramount importance to accurately locate diseased tissues, avoid inappropriate drug dosage, and improve therapeutic efficiency. In this regard, it is promising to develop strategies for real-time monitoring of drug release inside targeted cells or even in living bodies. Thus far, many multi-functional drug delivery systems constructed by a variety of building blocks, such as organic molecules, polymeric nanoparticles, micelles, and inorganic nanoparticles, have been developed for drug release monitoring. Especially, with the advancements in imaging modalities relating to nanomaterials, there has been an increasing focus on the use of non-invasive imaging techniques for monitoring drug release and drug efficacy in recent years. In this review, we introduce the application of fluorescence imaging, magnetic resonance imaging (MRI), surface-enhanced Raman scattering (SERS), and multi-mode imaging in monitoring drug release, involving a variety of nanomaterials such as organic or inorganic nanoparticles as imaging agents; their design principles are also elaborated. Among these, a special emphasis is placed on fluorescence-based drug release monitoring strategies, followed by a brief overview of MRI, SERS, and multi-mode imaging-based strategies. In the end, the challenges and prospects of drug release monitoring are also discussed.

80. A new method for predicting the shale distribution of the Wufeng Formation in the Upper Yangtze Region, China

Abstract

Taking the Late Ordovician Wufeng Formation (WFF) shale in the Upper Yangtze region as an example, we conducted a lithofacies distribution, thickness quantification, and paleo-topographic reconstruction of the Late Ordovician graptolite zones. Specifically, we focused on the Late Katian Dicellograptus complexus and the Early Hirnantian Metabolograptus extraordinarius within a chronostratigraphic framework, using the Geographic Information System (GIS) and 310 stratigraphic sections (incl. drilling) obtained from the Geobiodiversity Database (GBDB). Reconstruction of the geographic distribution indicates that the WFF and the synchronous sediments in the Upper Yangtze region contain 8 litho-stratigraphic units, which are geographically distributed across 7 provinces/municipalities and do not exhibit significant variations in lithofacies. The black graptolite shale extends in a broad swath from east to west within the basin, while the other lithofacies deposited during the same period are present on the periphery of the basin. All these strata were deposited in a normal neritic epicontinental sea environment, except for the flysch sediments in the southern Hunan area. The thickness reconstruction involves a comparison of three spatial interpolation methods, including Inverse Distance Weighting (IDW), Kriging, and the Radial Basis Function (RBF). Based on a general verification, IDW is considered to be the optimal method since it has the minimum standard deviation and variance. Based on the contours obtained from the IDW model, the WFF black shale is estimated to have an overall area of 0.67 × 106 km2, an average thickness of 6.2 m, and a total volume of 3902 km3. This shale was deposited over a 2.83 Ma period. Therefore, the volume of shale deposited per million years is estimated to be 1379 km3/my and the average thickness of shale deposited per million years is 2.37 m/my. The Hirnantian paleo-water-depth values obtained using 275 sections were used to infer the Late Katian paleo-topography. These results suggest that the Yangtze platform was surrounded by ancient highlands to the west, south, and north, exhibiting a paleo-geographic framework characterized by one uplift and four depressions. This setting blocked water circulation, causing the water to be contained and forming a closed and restricted marine environment, which was one of the major factors controlling the deposition of the organic-rich WFF shale. With the advent of the big data era of geology, the methodology of GIS-based technology is readily exportable to any resource play having spatial distribution pattern. Results can be provided rapidly and efficiently generated from geological data.

Abstract

Dynamically monitoring intracellular glutathione (GSH), a crucial biomarker of oxidative stress, is of significance for the diagnosis and treatment of certain diseases. Although manganese dioxide (MnO2) based GSH fluorescent sensors have exhibited high sensitivity and good selectivity owing to the specific reactivity between GSH and MnO2, near-infrared (NIR) MnO2 based nanoprobes for GSH detection are scarce. Herein, we have developed a NIR activatable fluorescence nanoprobe for the imaging and determination of intracellular GSH based on a core-shell nanoparticle, consisting of NIR emitted gold nanocluster doped silica as the fluorescent core and manganese dioxide as the GSH-responsive shell (named AuNCs@MnO2). Due to the absorption competition mechanism, the outer MnO2 shell rather than the inner AuNCs core preferentially absorbed the excitation light, thus leading to fluorescence quenching of the inner AuNCs core. Upon addition of GSH, the fluorescence of the nanoprobe restored along with the reduction of MnO2 to Mn2+ because of the absorption competition disappearance-induced emission. The activatable fluorescence linearly increased upon changing the GSH concentration in the range of 2 to 5000 μM with a detection limit of 0.67 μM. The cytotoxicity test shows that the AuNCs@MnO2 nanoprobes have a good biocompatibility. After entering the cancer cells, the intracellular GSH degraded the outermost MnO2 shell and initiated the NIR fluorescence restoration of AuNCs, which can be used to monitor the dynamic change of intracellular GSH. This strategy provides an NIR-activatable way to detect GSH levels in living cells and offers a promising platform for the diagnosis and treatment of GSH-related diseases.

Abstract

The toxicity and cost involved in the synthesis of nanoparticles using physical and chemical methods have led to the emergence of biological techniques being employed by scientists. Hence the present study employed an eco-friendly method by using Parkia biglobosa leaves extract as a reducing and stabilizing agent of chloroauric gold (HAuCl4) to gold nanoparticle (AuNPs). Direct Analysis in Real-Time (DART-MS) was employed for phytochemical screening. However, the AuNPs were characterized by Ultraviolet-Visible spectroscopy (UV/Vis), Transmission Electron Microscopy (TEM), Energy Dispersive X-ray spectroscopy, Fourier Transmission Infrared Spectroscopy, and X-ray diffraction spectroscopy, before the antibacterial and photocatalysis study. DART-MS showed more than 20 phytochemicals, including phenolic compounds. The FT-IR revealed the existence of flavonoids that we hypothesized are responsible for gold chloride reduction reactions. The optical properties of the gold nanoparticles by UV/Vis spectra registered plasmon surface resonance at a peak of 530 nm, whiles TEM revealed the AuNPs to be mono-dispersed and shapes were spherical and triangular with sizes ranging from 3.4 nm to 39.7 nm. XRD also recorded face-centered cubic AuNPs with 20.94 nm as the average crystallite size. The synthesized AuNPs were able to elucidate their anti-bacterial efficacy on Gram-positive bacteria Staphylococcus aureus, enterococcus faecalis and Gram-negative bacteria Escherichia coli, pseudomonas aeruginosa. The photocatalytic activities of AuNPs under UV/Vis irradiation exhibited a substantial degradation effect on rhodamine b (RhB) within 75 minutes. Due to their non-toxic nature, plant-mediated biosynthesized AuNPs, would be valuable in enhancing their biomedical applications and in wastewater treatment.
