Abstract: Full-dimensional quantum calculations for the thermal rate constant of the OH+HCl→H2O+Cl reaction rigorously accounting for the total angular momentum are presented in this work. The dynamics calculation is performed on the Clary, Hernandez, and Nyman potential energy surface and employs the flux correlation function formalism and the multiconfigurational time-dependent Hartree scheme. Comparison of the theoretical k(T) values with experiment reveals deficiencies of the potential energy surface. In order to test the validity of different approximations often imposed on the overall rotation, quantum dynamics simulations have also been carried out within the coupled states (CS) and J-shifting approximations. No significant discrepancies are found between CS and accurate results as was to be expected from previous experience. In contrast to previous results for the OH+H2 reaction, the J-shifting approximation yields a reasonable agreement as well.

Abstract: The recently proposed self-consistent hybrid method is applied to study photoinduced electron transfer (ET) reactions in mixed-valence compounds (NH3)5RuIIINCRuII(CN)5- and (NH3)5RuIIINCFeII(CN)5- in solution. To describe these ET processes in the condensed phase, both intramolecular modes (inner sphere) and the solvent environment (outer sphere) are taken into account using a model that is based on the analysis of experimental optical line shapes. The dynamics of the back ET process after photoexcitation is studied in detail. In particular we investigate the effects of intramolecular vibrational modes and solvent dynamics, as well as the influence of important physical parameters such as the electronic coupling and the temperature. In qualitative agreement with results of time-resolved optical experiments, the simulations predict an ultrafast decay of the population of the charge-transfer state. Abstract: International audience; To speed up and reduce power consumption per cycle in robotic manipulation, one option is to exploit intentional collisions with the surrounding environment and objects, an approach referred to as impact-aware manipulation. Within this context, this paper focuses on developing an online collision monitoring framework for distinguishing between expected and unexpected post-impact behaviors. The classification is based on a desired post-impact motion created via an idealized rigid robot-object-environment model. To generate a classification error bound, it employs a causal envelop filter that is needed due to the unavoidable joint and environment flexibility. In this way, it becomes possible to compare a desired idealized rigid response, which is straightforward to obtain with existing tools, with a measured impact response, which is affected by difficult-to-model post-impact oscillations. The classifier can be used for single-contact as well as multi-contact impact scenarios, such as those occurring in surface-to-surface impacts, and allows for tuning of the sensitivity between expected and unexpected postimpact behaviors. The monitoring framework fuses a (bandpass) momentum observer with impact-aware control to extend the classical collision event pipeline. As a proof of concept, we show the effectiveness of the approach through numerical simulations as well as with preliminary experimental results.

Abstract: International audience; Background and aims: A single hepatitis B virus (HBV) particle is sufficient to establish chronic infection of the liver after intravenous injection, suggesting that the virus targets hepatocytes via a highly efficient transport pathway. We therefore investigated whether HBV utilizes a physiological liver-directed pathway that supports specific host-cell targeting in vivo.Methods: We established an ex vivo system of perfused human liver tissue that recapitulates the liver physiology to investigate HBV liver targeting. This model allowed us to investigate virus-host cell interactions in a cellular microenvironment mimicking the in vivo situation. Results: HBV was rapidly sequestered by liver macrophages within one hour after a virus pulse, but was detected in hepatocytes only after 16 hours. We found that HBV associates with lipoproteins. Electron- and immunofluorescence microscopy corroborated a co-localization in recycling endosomes within peripheral and liver macrophages. Importantly, recycling endosomes accumulated HBV and cholesterol, followed by transport of HBV back to the cell surface along the cholesterol efflux pathway. To reach hepatocytes as final target cells, HBV was able to utilise the hepatocyte-directed cholesterol transport machinery of macrophages.Conclusions: Our results propose that HBV by binding to liver targeted lipoproteins and utilizing the reverse cholesterol transport pathway of macrophages hijacks the physiological lipid transport pathways to the liver to most efficiently reach its target organ. This may involve trans-infection of liver macrophages and result in deposition of HBV in the perisinusoidal space from where HBV can bind its receptor on hepatocytes

Abstract: OBJECTIVES: To characterize phenotypes of T cells that accumulated in multiple sclerosis (MS) lesions, to compare the lesional T-cell receptor (TCR) repertoire of T-cell subsets to peripheral blood, and to identify paired alpha and beta chains from single CD8(+) T cells from an index patient who we followed for 18 years.; METHODS: We combined immunohistochemistry, laser microdissection, and single-cell multiplex PCR to characterize T-cell subtypes and identify paired TCRalpha and TCRbeta chains from individual brain-infiltrating T cells in frozen brain sections. The lesional and peripheral TCR repertoires were analyzed by pyrosequencing.; RESULTS: We found that a TCR Vbeta1(+) T-cell population that was strikingly expanded in active brain lesions at clinical onset comprises several subclones expressing distinct yet closely related Valpha7.2(+) alpha chains, including a canonical Valpha7.2-Jalpha33 chain of mucosal-associated invariant T (MAIT) cells. Three other alpha chains bear striking similarities in their antigen-recognizing, hypervariable complementarity determining region 3. Longitudinal repertoire studies revealed that the TCR chains that were massively expanded in brain at onset persisted for several years in blood or CSF but subsequently disappeared except for the canonical Valpha7.2(+) MAIT cell and a few other TCR sequences that were still detectable in blood after 18 years.; CONCLUSIONS: Our observation that a massively expanded TCR Vbeta1-Jbeta2.3 chain paired with distinct yet closely related canonical or atypical MAIT cell-related alpha chains strongly points to an antigen-driven process in early active MS brain lesions.

Abstract: HD~106906AB is so far the only young binary system around which a planet has been imaged and a debris disk evidenced thanks to a strong IR excess. As such, it represents a unique opportunity to study the dynamics of young planetary systems. We aim at further investigating the close (tens of au scales) environment of the HD~106906AB system. We used the extreme AO fed, high contrast imager SPHERE recently installed on the VLT to observe HD~106906. Both the IRDIS imager and the Integral Field Spectrometer were used. We discovered a very inclined, ring-like disk at a distance of 65~au from the star. The disk shows a strong brightness asymmetry with respect to its semi-major axis. It shows a smooth outer edge, compatible with ejection of small grains by the stellar radiation pressure. We show furthermore that the planet's projected position is significantly above the disk's PA. Given the determined disk inclination, it is not excluded though that the planet could still orbit within the disk plane if at a large separation (2000--3000 au). We identified several additional point sources in the SPHERE/IRDIS field-of-view, that appear to be background objects. We compare this system with other debris disks sharing similarities, and we briefly discuss the present results in the framework of dynamical evolution., Comment: 7 pages, 6 figures, accepted by Astronomy & Astrophysics

Abstract: International audience; For automotive field, the vehiclevibrations induce movements of connector systems,composed of a female part system (springs) which isconnected and tighten up with the male part system(pin). There is relative movement of the contact zonebetween spring and pin which can lead to the frettingcorrosion phenomena.Several studies have been performed to investigatevibration consequences on electrical connectors. Thiswork aims to address the influence of a variety of factorssuch as fretting amplitude, frequency and current on theperformance of automotive connectors. An originalbench has been designed in order to measure the threedimensional displacements of the connector and theelectric contact characteristics at the same time. Thevelocity is measured with a laser Doppler vibrometer within the excitation-axis and the displacements within the two other axes are measured with laser displacement while a vibration is applied to the system. In this study, a measurement of induced relative displacements for different types of connector systems is undertaken as an analysis of the influence of the connector shapes and the dynamic parameters on the contact area damage and on the connector threshold of use.

Abstract: International audience; The extreme AO system, SAXO (SPHERE AO for eXoplanet Observation), is the heart of the SPHERE system, feeding the scientific instruments with flat wave front corrected from all the atmospheric turbulence and internal defects. We will present the final performance of SAXO obtained during the instrument AIT in Europe as well as the very first on-sky results. The main requirements and system characteristics will be recalled and the full AO loop performance will be quantified and compared to original specifications. It will be demonstrated that SAXO meets or even exceeds (especially its limit magnitude and its jitter residuals) its challenging requirements (more than 90% of SR in H band and a 3 mas residual jitter). Finally, after 10 years of AO developments, from early design to final on-sky implementations, some critical system aspects as well as some important lesson-learned will be presented in the perspective of the future generation of complex AO systems for VLTs and ELTs.

Abstract: International audience; Respiratory chain deficiencies exhibit a wide variety of clinical phenotypes resulting from defective mitochondrial energy production through oxidative phosphorylation. These defects can be caused by either mutations in the mtDNA or mutations in nuclear genes coding for mitochondrial proteins. The underlying pathomechanisms can affect numerous pathways involved in mitochondrial physiology. By whole-exome and candidate gene sequencing, we identified 11 individuals from 9 families carrying compound heterozygous or homozygous mutations in GTPBP3, encoding the mitochondrial GTP-binding protein 3. Affected individuals from eight out of nine families presented with combined respiratory chain complex deficiencies in skeletal muscle. Mutations in GTPBP3 are associated with a severe mitochondrial translation defect, consistent with the predicted function of the protein in catalyzing the formation of 5-taurinomethyluridine (τm(5)U) in the anticodon wobble position of five mitochondrial tRNAs. All case subjects presented with lactic acidosis and nine developed hypertrophic cardiomyopathy. In contrast to individuals with mutations in MTO1, the protein product of which is predicted to participate in the generation of the same modification, most individuals with GTPBP3 mutations developed neurological symptoms and MRI involvement of thalamus, putamen, and brainstem resembling Leigh syndrome. Our study of a mitochondrial translation disorder points toward the importance of posttranscriptional modification of mitochondrial tRNAs for proper mitochondrial function.

Abstract: International audience; The SPHERE (Spectro-Polarimetry High-contrast Exoplanet Research) instrument is an ESO project aiming at the direct detection of extra-solar planets. SPHERE has been successfully integrated and tested in Europe end 2013 and has been re-integrated at Paranal in Chile early 2014 for a first light at the beginning of May. The heart of the SPHERE instrument is its eXtreme Adaptive Optics (XAO) SAXO (SPHERE AO for eXoplanet Observation) subsystem that provides extremely high correction of turbulence and very accurate stabilization of images for coronagraphic purpose. However, SAXO, as well as the overall instrument, must also provide constant operability overnights, ensuring robustness and autonomy. An original control scheme has been developed to satisfy this challenging dichotomy. It includes in particular both an Optimized Modal Gain Integrator (OMGI) to control the Deformable Mirror (DM) and a Linear Quadratic Gaussian (LQG) control law to manage the tip-tilt (TT) mirror. LQG allows optimal estimation and prediction of turbulent angle of arrival but also of possible vibrations. A specific and unprecedented control scheme has been developed to continuously adapt and optimize LQG control ensuring a constant match to turbulence and vibrations characteristics. SPHERE is thus the first operational system implementing LQG, with automatic adjustment of its models. SAXO has demonstrated performance beyond expectations during tests in Europe, in spite of internal limitations. Very first results have been obtained on sky last May. We thus come back to SAXO control scheme, focusing in particular on the LQG based TT control and the various upgrades that have been made to enhance further the performance ensuring constant operability and robustness. We finally propose performance assessment based on in lab performance and first on sky results and discuss further possible improvements.

Abstract: KAI2 are plant α/β hydrolase receptors, which perceive smoke-derived butenolide signals (karrikins) and putative endogenous, yet unidentified phytohormones (KAI2-ligands, KLs). The number of functional KAI2 receptors varies among plant species. It has been suggested that KAI2 gene duplication and sub-functionalization plays an adaptative role for diverse environments or ligand diversification by altering the receptor responsiveness to specific KLs. Legumes represent one of the largest families of flowering plants and contain many essential agronomic crops. Prior to legume diversification, KAI2 underwent duplication, resulting in KAI2A and KAI2B. Integrating plant genetics, ligand perception and enzymatic assays, and protein crystallography, we demonstrate that Pisum sativum KAI2A and KAI2B act as receptors and enzymes with divergent ligand stereoselectivity. KAI2B has a stronger affinity than KAI2A towards the KAI2-ligand (-)-GR24 and remarkably hydrolyses a broader range of substrates including the strigolactone-like isomer (+)-GR24. We determine the crystal structures of PsKAI2B in apo and butenolide-bound states. The biochemical and structural analyses as well as recorded mass spectra of KAI2s reveal a transient intermediate on the catalytic serine and a stable adduct on the catalytic histidine, further illuminating the role of KAI2 not only as receptors but also as bona fide enzymes. Our work uncovers the stereoselectivity of ligand perception and catalysis by evolutionarily diverged KAI2 receptors in KAR/KL signaling pathways and proposes adaptive sensitivity to KAR/KL and strigolactone phytohormones by KAI2B.

Abstract: Context: Type II supernovae provide a direct way to estimate distances through the expanding photosphere method, which is independent of the cosmic distance ladder. A recently introduced Gaussian process-based method allows for a fast and precise modelling of spectral time series, which puts accurate and computationally cheap Type II-based absolute distance determinations within reach. Aims: The goal of the paper is to assess the internal consistency of this new modelling technique coupled with the distance estimation empirically, using the spectral time series of supernova siblings, i.e. supernovae that exploded in the same host galaxy. Methods: We use a recently developed spectral emulator code, which is trained on \textsc{Tardis} radiative transfer models and is capable of a fast maximum likelihood parameter estimation and spectral fitting. After calculating the relevant physical parameters of supernovae we apply the expanding photosphere method to estimate their distances. Finally, we test the consistency of the obtained values by applying the formalism of Bayes factors. Results: The distances to four different host galaxies were estimated based on two supernovae in each. The distance estimates are not only consistent within the errors for each of the supernova sibling pairs, but in the case of two hosts they are precise to better than 5\%. Conclusions: Even though the literature data we used was not tailored for the requirements of our analysis, the agreement of the final estimates shows that the method is robust and is capable of inferring both precise and consistent distances. Abstract: International audience; Objectives A major goal of curative hepatitis B virus (HBV) treatments is the reduction or inactivation of intrahepatic viral covalently closed circular DNA (cccDNA). Hence, precise cccDNA quantification is essential in preclinical and clinical studies. Southern blot (SB) permits cccDNA visualisation but lacks sensitivity and is very laborious. Quantitative PCR (qPCR) has no such limitations but inaccurate quantification due to codetection of viral replicative intermediates (RI) can occur. The use of different samples, preservation conditions, DNA extraction, nuclease digestion methods and qPCR strategies has hindered standardisation. Within the ICE-HBV consortium, available and novel protocols for cccDNA isolation and qPCR quantification in liver tissues and cell cultures were compared in six laboratories to develop evidence-based guidance for best practices. Design Reference material (HBV-infected humanised mouse livers and HepG2-NTCP cells) was exchanged for cross-validation. Each group compared different DNA extraction methods (Hirt extraction, total DNA extraction with or without proteinase K treatment (+PK/−PK)) and nuclease digestion protocols (plasmid-safe ATP-dependent DNase (PSD), T5 exonuclease, exonucleases I/III). Samples were analysed by qPCR and SB. Results Hirt and −PK extraction reduced coexisting RI forms. However, both cccDNA and the protein-free relaxed circular HBV DNA (pf-rcDNA) form were detected by qPCR. T5 and Exo I/III nucleases efficiently removed all RI forms. In contrast, PSD did not digest pf-rcDNA, but was less prone to induce cccDNA overdigestion. In stabilised tissues (eg, Allprotect), nucleases had detrimental effects on cccDNA. Conclusions We present here a comprehensive evidence-based guidance for optimising, controlling and validating cccDNA measurements using available qPCR assays.

Abstract: We report a huge organic diversity in the Tissint Mars meteorite and the sampling of several mineralogical lithologies, which revealed that the organic molecules were nonuniformly distributed in functionality and abundance. The range of organics in Tissint meteorite were abundant C 3-7 aliphatic branched carboxylic acids and aldehydes, olefins, and polyaromatics with and without heteroatoms in a homologous oxidation structural continuum. Organomagnesium compounds were extremely abundant in olivine macrocrystals and in the melt veins, reflecting specific organo-synsthesis processes in close interaction with the magnesium silicates and temperature stresses, as previously observed. The diverse chemistry and abundance in complex molecules reveal heterogeneity in organic speciation within the minerals grown in the martian mantle and crust that may have evolved over geological time.

Abstract: International audience; In an embedded real-time system (ERTS), real-time tasks (software) are typically executed on a multicore shared-memory platform (hardware). The number of cores is usually small, contrasted with a larger number of complex tasks that share data to collaborate. Since most ERTSs are safety-critical, it is crucial to rigorously verify their software against various real-time requirements under the actual hardware constraints (concurrent access to data, number of cores). Both the real-time systems and the formal methods communities provide elegant techniques to realize such verification, which nevertheless face major challenges. For instance, model checking (formal methods) suffers from the state-space explosion problem, whereas schedulability analysis (real-time systems) is pessimistic and restricted to simple task models and schedulability properties. In this paper, we propose a scalable and generic approach to formally verify ERTSs. The core contribution is enabling, through joining the forces of both communities, compositional verification to tame the state-space size. To that end, we formalize a realistic ERTS model where tasks are complex with an arbitrary number of jobs and job segments, then show that compositional verification of such model is possible, using a hybrid approach (from both communities), under the state-of-the-art partitioned fixed-priority (P-FP) with limited preemption scheduling algorithm. The approach consists of the following steps, given the above ERTS model and scheduling algorithm. First, we compute fine-grained data sharing overheads for each job segment that reads or writes some data from the shared memory. Second, we generalize an algorithm that, aware of the data sharing overheads, computes an affinity (task-core allocation) guaranteeing the schedulability of hard-real-time (HRT) tasks. Third, we devise a timed automata (TA) model of the ERTS, that takes into account the affinity, the data sharing overheads and the scheduling algorithm, on which we demonstrate that various properties can be verified compositionally, i.e., on a subset of cores instead of the whole ERTS, therefore reducing the state-space size. In particular, we enable the scalable computation of tight worst-case response times (WCRTs) and other tight bounds separating events on different cores, thus overcoming the pessimism of schedulability analysis techniques. We fully automate our approach and show its benefits on three real-world complex ERTSs, namely two autonomous robots and an automotive case study from the WATERS 2017 industrial challenge.

Abstract: Strawberry (Fragaria × ananassa) fruits are an excellent source of L-ascorbic acid (AsA), a powerful antioxidant for plants and humans. Identifying the genetic components underlying AsA accumulation is crucial for enhancing strawberry nutritional quality. Here, we unravel the genetic architecture of AsA accumulation using an F1 population derived from parental lines ‘Candonga’ and ‘Senga Sengana’, adapted to distinct Southern and Northern European areas. To account for environmental effects, the F1 and parental lines were grown and phenotyped in five locations across Europe (France, Germany, Italy, Poland and Spain). Fruit AsA content displayed normal distribution typical of quantitative traits and ranged five-fold, with significant differences among genotypes and environments. AsA content in each country and the average in all of them was used in combination with 6,974 markers for quantitative trait locus (QTL) analysis. Environmentally stable QTLs for AsA content were detected in linkage group (LG) 3A, LG 5A, LG 5B, LG 6B and LG 7C. Candidate genes were identified within stable QTL intervals and expression analysis in lines with contrasting AsA content suggested that GDP-L-Galactose Phosphorylase FaGGP(3A), and the chloroplast-located AsA transporter gene FaPHT4;4(7C) might be the underlying genetic factors for QTLs on LG 3A and 7C, respectively. We show that recessive alleles of FaGGP(3A) inherited from both parental lines increase fruit AsA content. Furthermore, expression of FaGGP(3A) was two-fold higher in lines with high AsA. Markers here identified represent a useful resource for efficient selection of new strawberry cultivars with increased AsA content.

Abstract: The Asymptotic Equipartition Property (AEP) in information theory establishes that independent and identically distributed (i.i.d.) states behave in a way that is similar to uniform states. In particular, with appropriate smoothing, for such states both the min and the max relative entropy asymptotically coincide with the relative entropy. In this paper, we generalize several such equipartition properties to states on general von Neumann algebras. First, we show that the smooth max relative entropy of i.i.d. states on a von Neumann algebra has an asymptotic rate given by the quantum relative entropy. In fact, our AEP not only applies to states, but also to quantum channels with appropriate restrictions. In addition, going beyond the i.i.d. assumption, we show that for states that are produced by a sequential process of quantum channels, the smooth max relative entropy can be upper bounded by the sum of appropriate channel relative entropies. Our main technical contributions are to extend to the context of general von Neumann algebras a chain rule for quantum channels, as well as an additivity result for the channel relative entropy with a replacer channel., updated version with a new section on application

Abstract: We establish existence and uniqueness of the solution to the Dyson equation for the density-density response function in time-dependent density functional theory (TDDFT) in the random phase approximation (RPA). We show that the poles of the RPA density-density response function are forward-shifted with respect to those of the non-interacting response function, thereby explaining mathematically the well known empirical fact that the non-interacting poles (given by the spectral gaps of the time-independent Kohn-Sham equations) underestimate the true transition frequencies. Abstract: This open access book offers a multidisciplinary and comprehensive perspective regarding the immediate and long-term effects of the Covid-19 pandemic on coworking spaces in the European Region. The current pandemic has imposed several effects on work and spaces for work. Some are immediate effects and will last for a short time (such as the closing down of the space), some will last longer (namely, the reorganisation of the space to meet the physical distancing), and some will stay for a long time (remote working and hybrid working). Although the literature on coworking spaces and the effects of the pandemic is growing fast, empirical studies are yet limited. Within this context, this book seeks a twofold aim: (i) to contribute to the fast-growing literature on coworking space and their effects at different scales; (ii) to present a multidisciplinary perspective about the effects of the yet-lasting Corona-pandemic effects on the patterns of remote working and consequently on coworking spaces, as the most diffused form of new working spaces.

Abstract: Wheat is the most widely grown food crop, with 761 Mt produced globally in 2020. To meet the expected grain demand by mid-century, wheat breeding strategies must continue to improve upon yield-advancing physiological traits, regardless of climate change impacts. Here, the best performing doubled haploid (DH) crosses with an increased canopy photosynthesis from wheat field experiments in the literature were extrapolated to the global scale with a multi-model ensemble of process-based wheat crop models to estimate global wheat production. The DH field experiments were also used to determine a quantitative relationship between wheat production and solar radiation to estimate genetic yield potential. The multi-model ensemble projected a global annual wheat production of 1050 ± 145 Mt due to the improved canopy photosynthesis, a 37% increase, without expanding cropping area. Achieving this genetic yield potential would meet the lower estimate of the projected grain demand in 2050, albeit with considerable challenges., This study was a part of the Agricultural Model Intercomparison and Improvement Project (AgMIP) Wheat Phase 4. Abstract: Liquid metal catalysts have recently attracted attention for synthesizing high-quality two-dimensional (2D) materials facilitated via the catalysts’ perfectly smooth surface. However, the microscopic catalytic processes occurring at the surface are still largely unclear because liquid metals escape the accessibility of traditional experimental and computational surface science approaches. For this reason, numerous controversies are found regarding different applications, with graphene (Gr) growth on liquid copper (Cu) as a prominent prototype. In this work, we employ novel in situ and in silico techniques to achieve an atomic-level characterization of the graphene adsorption height above liquid Cu, reaching a quantitative agreement within 0.1 Å between experiment and theory. Our results are obtained via in situ synchrotron X-ray reflectivity (XRR) measurements over wide-range q-vectors and large-scale molecular dynamics simulations based on efficient machine-learning (ML) potentials trained to first-principles density-functional theory (DFT) data. We demonstrate our computational insight to be robust against inherent DFT errors and reveal the nature of graphene binding to be highly comparable at liquid Cu and solid Cu(111). Transporting the predictive first-principles quality via ML potentials to the scales and sampling required for liquid metal catalysis thus provides a powerful approach to reach microscopic understanding, analogous to the established computational approaches for catalysis at solid surfaces.

Abstract: International audience; This paper presents an improved identification method of the constitutive properties of lung parenchyma. We aim to determine the non-linear viscoelastic behavior of lung parenchyma with a particular focus on the compressible properties - i.e. the ability to change volume. Uniaxial tensile tests are performed on living precision-cut rat lung slices. Image registration is used to compute the displacement field at the surface of the sample. The constitutive model consists of a hyperelastic potential split into volumetric and isochoric contributions and a viscous contribution. This allows for the description of the experimentally observed hysteresis loop. The identification is performed numerically: each test is simulated using the realistic geometry of the sample; the difference between the measured and computed displacements is minimized with an optimization algorithm. We compare several hyperelastic potentials and we can determine the most suitable law for rat lung parenchyma. An exponential potential or a polynomial potential with a first order term and a third or higher order term give similarly satisfactory results. The identified parameters are: for the volumetric contribution: κ =7.25e4 Pa, for the exponential form: k1=4.34e3 Pa, k2=5.92, for the polynomial form: C1=2.87e3 Pa, C3=3.83e4 Pa. The identification of the time parameter for the viscous contribution shows that it depends on the loading frequency (0.2 Hz: τ =0.257 s, 0.4 Hz: τ=0.123 s, 0.8 Hz: τ=0.050 s). Adding a viscous contribution significantly increases the accuracy of the identification.

Abstract: International audience; Infections, microbe sampling and occasional leakage of commensal microbiota and their products across the intestinal epithelial cell layer represent a permanent challenge to the intestinal immune system. The production of reactive oxygen species by NADPH oxidase is thought to be a key element of defense. Patients suffering from chronic granulomatous disease are deficient in one of the subunits of NADPH oxidase. They display a high incidence of Crohn's disease-like intestinal inflammation and are hyper-susceptible to infection with fungi and bacteria, including a 10-fold increased risk of Salmonellosis. It is not completely understood which steps of the infection process are affected by the NADPH oxidase deficiency. We employed a mouse model for Salmonella diarrhea to study how NADPH oxidase deficiency (Cybb (-/-)) affects microbe handling by the large intestinal mucosa. In this animal model, wild type S. Typhimurium causes pronounced enteropathy in wild type mice. In contrast, an avirulent S. Typhimurium mutant (S.Tm(avir); invGsseD), which lacks virulence factors boosting trans-epithelial penetration and growth in the lamina propria, cannot cause enteropathy in wild type mice. We found that Cybb (-/-) mice are efficiently infected by S.Tm(avir) and develop enteropathy by day 4 post infection. Cell depletion experiments and infections in Cybb (-/-) Myd88 (-/-) mice indicated that the S.Tm(avir)-inflicted disease in Cybb (-/-) mice hinges on CD11c(+)CX3CR1(+) monocytic phagocytes mediating colonization of the cecal lamina propria and on Myd88-dependent proinflammatory immune responses. Interestingly, in mixed bone marrow chimeras a partial reconstitution of Cybb-proficiency in the bone marrow derived compartment was sufficient to ameliorate disease severity. Our data indicate that NADPH oxidase expression is of key importance for restricting the growth of S.Tm(avir) in the mucosal lamina propria. This provides important insights into microbe handling by the large intestinal mucosa and the role of NADPH oxidase in maintaining microbe-host mutualism at this exposed body surface.

Abstract: International audience; This paper is concerned with a new control method for path tracking of autonomous ground vehicles. We exploit the fuzzy model-based control framework to deal with the time-varying feature of the vehicle speed and the highly uncertain behaviors of the tire-road forces involved in the nonlinear vehicle dynamics. To avoid using costly vehicle sensors for feedback control while favoring the simplest control structure for real-time implementation, a new fuzzy static output feedback (SOF) scheme is proposed. In particular, though the robust set-invariance property and Lyapunov-based arguments, the physical constraints on the steering input saturation and the vehicle state can be taken into account in the control design to improve the driving safety and comfort. The theoretical development relies on the use of fuzzy Lyapunov functions and the non-parallel distributed compensation control concept to reduce the design conservatism. Exploiting some specific convexification techniques, the control design is reformulated as an optimization problem under linear matrix inequalities with a single line search, which are efficiently solved via semidefinite programming techniques. The proposed fuzzy path tracking controller is evaluated through various dynamic driving tests conducted with high-fidelity CarSim/Matlab co-simulations. Moreover, to emphasize the advantages of the new fuzzy SOF controller, a performance comparison with the CarSim driver model is also performed.

Abstract: International audience; PURPOSE: Optoacoustic imaging relies on the detection of ultrasonic waves induced by laser pulse excitations to map optical absorption in biological tissue. A tomographic geometry employing a conventional ultrasound linear detector array for volumetric optoacoustic imaging is reported. The geometry is based on a translate-rotate scanning motion of the detector array, and capitalizes on the geometrical characteristics of the transducer assembly to provide a large solid angular detection aperture. A system for three-dimensional whole-body optoacoustic tomography of small animals is implemented. METHODS: The detection geometry was tested using a 128-element linear array (5.0∕7.0 MHz, Acuson L7, Siemens), moved by steps with a rotation∕translation stage assembly. Translation and rotation range of 13.5 mm and 180°, respectively, were implemented. Optoacoustic emissions were induced in tissue-mimicking phantoms and ex vivo mice using a pulsed laser operating in the near-IR spectral range at 760 nm. Volumetric images were formed using a filtered backprojection algorithm. RESULTS: The resolution of the optoacoustic tomography system was measured to be better than 130 μm in-plane and 330 μm in elevation (full width half maximum), and to be homogenous along a 15 mm diameter cross section due to the translate-rotate scanning geometry. Whole-body volumetric optoacoustic images of mice were performed ex vivo, and imaged organs and blood vessels through the intact abdominal and head regions were correlated to the mouse anatomy. CONCLUSIONS: Overall, the feasibility of three-dimensional and high-resolution whole-body optoacoustic imaging of small animal using a conventional linear array was demonstrated. Furthermore, the scanning geometry may be used for other linear arrays and is therefore expected to be of great interest for optoacoustic tomography at macroscopic and mesoscopic scale. Specifically, conventional detector arrays with higher central frequencies may be investigated.

Abstract: Histone variants are non-allelic protein isoforms that play key roles in diversifying chromatin structure. The known number of such variants has greatly increased in recent years, but the lack of naming conventions for them has led to a variety of naming styles, multiple synonyms and misleading homographs that obscure variant relationships and complicate database searches. We propose here a unified nomenclature for variants of all five classes of histones that uses consistent but flexible naming conventions to produce names that are informative and readily searchable. The nomenclature builds on historical usage and incorporates phylogenetic relationships, which are strong predictors of structure and function. A key feature is the consistent use of punctuation to represent phylogenetic divergence, making explicit the relationships among variant subtypes that have previously been implicit or unclear. We recommend that by default new histone variants be named with organism-specific paralog-number suffixes that lack phylogenetic implication, while letter suffixes be reserved for structurally distinct clades of variants. For clarity and searchability, we encourage the use of descriptors that are separate from the phylogeny-based variant name to indicate developmental and other properties of variants that may be independent of structure.

Abstract: International audience; Frictional dynamic contact problems with complex geometries are a challenging task from the compu tational as well as from the analytical point of view since they generally involve space and time multi scale aspects. To be able to reduce the complexity of this kind of contact problem, we employ a non conforming domain decomposition method in space, consisting of a coarse global mesh not resolving the local struc ture and an overlapping fine patch for the contact computation. This leads to several benefits: First, we resolve the details of the surface only where it is needed, i.e., in the vicinity of the actual contact zone. Second, the subproblems can be discretized independently of each other which enables us to choose a much finer time scale on the contact zone than on the coarse domain. Here, we propose a set of interface conditions that yield optimal a priori error estimates on the fine meshed subdomain without any artificial dissipation. Further, we develop an efficient iterative solution scheme for the coupled problem that is robust with respect to jumps in the material parameters. Several complex numerical examples illustrate the performance of the new scheme.

Abstract: Advances in the time propagation of multidimensional wave packets are exploited to present the A‐band photodissociation dynamics of methyl iodide for five active vibrational modes on the three relevant excited ab initio potential surfaces. The five modes considered represent all of the experimentally observed dynamical activity. The only modes neglected are the asymmetric C–H stretch and the asymmetric deformation of the methyl group. The kinetic energy operator corresponding to these five degrees of freedom is derived. The fully quantum mechanical calculation was implemented upon grids using 2880 distinct time‐dependent configurations, determined by the multiconfigurational time‐dependent Hartree algorithm, for each electronic state. All of the currently known experimental results regarding the umbrella vibration, symmetric C–H stretching vibration, perpendicular rotation, and parallel rotation of the photodissociated methyl radical fragment are well reproduced. Abstract: Restless legs syndrome (RLS) is a sensorimotor disorder with an age-dependent prevalence of up to 10% in the general population above 65 years of age. Affected individuals suffer from uncomfortable sensations and an urge to move in the lower limbs that occurs mainly in resting situations during the evening or at night. Moving the legs or walking leads to an improvement of symptoms. Concomitantly, patients report sleep disturbances with consequences such as reduced daytime functioning. We conducted a genome-wide association study (GWA) for RLS in 922 cases and 1,526 controls (using 301,406 SNPs) followed by a replication of 76 candidate SNPs in 3,935 cases and 5,754 controls, all of European ancestry. Herein, we identified six RLS susceptibility loci of genome-wide significance, two of them novel: an intergenic region on chromosome 2p14 (rs6747972, P = 9.03 × 10−11, OR = 1.23) and a locus on 16q12.1 (rs3104767, P = 9.4 × 10−19, OR = 1.35) in a linkage disequilibrium block of 140 kb containing the 5′-end of TOX3 and the adjacent non-coding RNA BC034767., Author Summary Restless legs syndrome (RLS) is one of the most common neurological disorders. Patients with RLS suffer from an urge to move the legs and unpleasant sensations located mostly deep in the calf. Symptoms mainly occur in resting situations in the evening or at night. As a consequence, initiation and maintenance of sleep become defective. Here, we performed a genome-wide association study to identify common genetic variants increasing the risk for disease. The genome-wide phase included 922 cases and 1,526 controls, and candidate SNPs were replicated in 3,935 cases and 5,754 controls, all of European ancestry. We identified two new RLS–associated loci: an intergenic region on chromosome 2p14 and a locus on 16q12.1 in a linkage disequilibrium block containing the 5′-end of TOX3 and the adjacent non-coding RNA BC034767. TOX3 has been implicated in the development of breast cancer. The physiologic role of TOX3 and BC034767 in the central nervous system and a possible involvement of these two genes in RLS pathogenesis remain to be established.

Both obesity and being underweight have been associated with increased mortality. Underweight, defined as a body mass index (BMI) ≤ 18.5 kg per m(2) in adults and ≤ -2 standard deviations from the mean in children, is the main sign of a series of heterogeneous clinical conditions including failure to thrive, feeding and eating disorder and/or anorexia nervosa. In contrast to obesity, few genetic variants underlying these clinical conditions have been reported. We previously showed that hemizygosity of a ∼600-kilobase (kb) region on the short arm of chromosome 16 causes a highly penetrant form of obesity that is often associated with hyperphagia and intellectual disabilities. Here we show that the corresponding reciprocal duplication is associated with being underweight. We identified 138 duplication carriers (including 132 novel cases and 108 unrelated carriers) from individuals clinically referred for developmental or intellectual disabilities (DD/ID) or psychiatric disorders, or recruited from population-based cohorts. These carriers show significantly reduced postnatal weight and BMI. Half of the boys younger than five years are underweight with a probable diagnosis of failure to thrive, whereas adult duplication carriers have an 8.3-fold increased risk of being clinically underweight. We observe a trend towards increased severity in males, as well as a depletion of male carriers among non-medically ascertained cases. These features are associated with an unusually high frequency of selective and restrictive eating behaviours and a significant reduction in head circumference. Each of the observed phenotypes is the converse of one reported in carriers of deletions at this locus. The phenotypes correlate with changes in transcript levels for genes mapping within the duplication but not in flanking regions. The reciprocal impact of these 16p11.2 copy-number variants indicates that severe obesity and being underweight could have mirror aetiologies, possibly through contrasting effects on energy balance. Abstract: Comparison-based learning addresses the problem of learning when, instead of explicit features or pairwise similarities, one only has access to comparisons of the form: \emph{Object $A$ is more similar to $B$ than to $C$.} Recently, it has been shown that, in Hierarchical Clustering, single and complete linkage can be directly implemented using only such comparisons while several algorithms have been proposed to emulate the behaviour of average linkage. Hence, finding hierarchies (or dendrograms) using only comparisons is a well understood problem. However, evaluating their meaningfulness when no ground-truth nor explicit similarities are available remains an open question. In this paper, we bridge this gap by proposing a new revenue function that allows one to measure the goodness of dendrograms using only comparisons. We show that this function is closely related to Dasgupta's cost for hierarchical clustering that uses pairwise similarities. On the theoretical side, we use the proposed revenue function to resolve the open problem of whether one can approximately recover a latent hierarchy using few triplet comparisons. On the practical side, we present principled algorithms for comparison-based hierarchical clustering based on the maximisation of the revenue and we empirically compare them with existing methods., 26 pages, 6 figures, 5 tables. Abstract: ObjectiveIt is believed that intestinal recruitment of monocytes from Crohn’s Disease (CD) patients who carry NOD2 risk alleles may repeatedly give rise to recruitment of pathogenic macrophages. We investigated an alternative possibility that NOD2 may rather inhibit their differentiation from intravasating monocytes.DesignThe monocyte fate decision was examined by using germ-free mice, mixed bone marrow chimeras and a culture system yielding macrophages and monocyte-derived dendritic cells (mo-DCs). We next asked whether Nod2 in either monocytes or tissue macrophages have distinct resolving properties in colitis.ResultsDespite a similar abundance of monocytes, the intestinal frequency of mo-DCs from Nod2-deficient mice was lowered independently of the changes in the gut microbiota that are caused by Nod2 deficiency. Similarly, the pool of mo-DCs was poorly reconstituted with mobilized bone marrow Nod2-deficient cells. The use of pharmacological inhibitors revealed that activated NOD2 at an early stage of development dominantly inhibits mTOR-mediated macrophage differentiation in a TNFalpha-dependent manner. These observations were supported by the identification of a TNFalpha-dependent response to MDP that is specifically lost in CD14-expressing blood cells bearing the frameshift mutation in NOD2. Accordingly, loss of NOD2 in monocytes lowers glycolytic reserve, CD115 expression and pro-resolving features. It may thereby prevent the accumulation of pathogenic macrophages in patients with loss-of-function NOD2 alleles, which fail to respond to anti-TNF and are at greater risk of developing stricturing disease.

Abstract: International audience; The absence of stray fields, their insensitivity to external magnetic fields, and ultrafast dynamics make antiferromagnets promising candidates for active elements in spintronic devices. Here, we demonstrate manipulation of the Néel vector in the metallic collinear antiferromagnet Mn2Au by combining strain and femtosecond laser excitation. Applying tensile strain along either of the two in-plane easy axes and locally exciting the sample by a train of femtosecond pulses, we align the Néel vector along the direction controlled by the applied strain. The dependence on the laser fluence and strain suggests the alignment is a result of optically-triggered depinning of 90o domain walls and their sliding in the direction of the free energy gradient, governed by the magneto-elastic coupling. The resulting, switchable, state is stable at room temperature and insensitive to magnetic fields. Such an approach may provide ways to realize robust high-density memory device with switching timescales in the picosecond range.

Abstract: International audience; Evaluating natural frequencies for damage detection and localization is a standard procedure in non-destructive testing (NDT) and structural health monitoring (SHM). In particular, for cables and other prestressed tendons, natural frequencies exhibit high sensitivities to structural changes, which is often demonstrated by comparing the frequencies before and after damage occurred, e.g., using probability of detection (POD) or probability of localization (POL) curves. In this paper, an approach is developed to predict POD/POL curves without any data from the damaged state. The method is based on data from the undamaged state and a numerical model, which is utilized to obtain sensitivity vectors that linearize the relation between frequency changes and changes in structural parameters. At the same time, statistical uncertainties in the frequency estimation process are considered. Based on this method, a cable monitoring strategy is suggested, where sensitivity vectors are first used to calibrate decisive cable properties using model updating, followed by a performance assessment through POD/POL curves, and a continuous damage detection and localization based on statistical tests. The synergic effects of the presented methods are demonstrated by means of a numerical case study on a prestressed cable, where data from a single uni-axial vibration sensor is evaluated. The results indicate that changes in stiffness and axial forces can be distinguished using localization tests, but additional sensors and features are necessary for damage localization along the cable.

Abstract: International audience; Job management software on peta- and exascale supercomputers continues to provide static resource allocations, from a program’s start until its end. Dynamic resource allocation and management is a research direction that has the potential to improve the efficiency of HPC systems and applications by dynamically adapting the resources of an application during its runtime. Resources can be adapted based on past, current or even future system conditions and matching optimization targets. However, the implementation of dynamic resource management is challenging as it requires support across many layers of the software stack, including the programming model.In this paper, we focus on the latter and present our approach to extend MPI Sessions to support dynamic resource allocations within MPI applications. While some forms of dynamicity already exist in MPI, it is currently limited by requiring global synchronization, being application or application-domain specific, or by suffering from limited support in current HPC system software stacks.We overcome these limitations with a simple, yet powerful abstraction: resources as process sets, and changes of resources as set operations leading to a graph-based perspective on resource changes. As the main contribution of this work, we provide an implementation of this approach based on MPI Sessions and PMIx. In addition, an illustration of its usage is provided, as well as a discussion about the required extensions of the PMIx standard. We report results based on a prototype implementation with Open MPI using a synthetic application, as well as a PDE solver benchmark on up to four nodes and a total of 112 cores. Overall, our results show the feasibility of our approach, which has only very moderate overheads. We see this first proof-of-concept as an important step towards resource adaptivity based on MPI Sessions.

Abstract: International audience; Importance Albuminuria, routinely assessed as spot urine albumin-to-creatinine ratio (UACR), indicates structural damage of the glomerular filtration barrier and is associated with poor kidney and cardiovascular outcomes. Sodium-glucose cotransporter-2 (SGLT2) inhibitors have been found to reduce UACR in patients with type 2 diabetes, but its use in patients with heart failure (HF) is less well studied. Objective To analyze the association of empagliflozin with study outcomes across baseline levels of albuminuria and change in albuminuria in patients with HF across a wide range of ejection fraction levels. Design, Setting, and Participants This post hoc analysis included all patients with HF from the EMPEROR-Pooled analysis using combined individual patient data from the international multicenter randomized double-blind parallel-group, placebo-controlled EMPEROR-Reduced and EMPEROR-Preserved trials. Participants in the original trials were excluded from this analysis if they were missing baseline UACR data. EMPEROR-Preserved was conducted from March 27, 2017, to April 26, 2021, and EMPEROR-Reduced was conducted from April 6, 2017, to May 28, 2020. Data were analyzed from January to June 2022. Interventions Randomization to empagliflozin or placebo. Main Outcomes and Measures New-onset macroalbuminuria and regression to normoalbuminuria and microalbuminuria. Results A total of 9673 patients were included (mean [SD] age, 69.9 [10.4] years; 3551 [36.7%] female and 6122 [63.3%] male). Of these, 5552 patients had normoalbuminuria (UACR 300 mg/g). Compared with normoalbuminuria, macroalbuminuria was associated with younger age, races other than White, obesity, male sex, site region other than Europe, higher levels of N-terminal pro–hormone brain natriuretic peptide and high-sensitivity troponin T, higher blood pressure, higher New York Heart Association class, greater HF duration, more frequent previous HF hospitalizations, diabetes, hypertension, lower eGFR, and less frequent use of angiotensin-converting enzyme inhibitors or angiotensin receptor blockers and mineralocorticoid receptor antagonists. An increase in events was observed in individuals with higher UACR levels. The association of empagliflozin with cardiovascular mortality or HF hospitalization was consistent across UACR categories (hazard ratio [HR], 0.80; 95% CI, 0.69-0.92 for normoalbuminuria; HR, 0.74; 95% CI, 0.63-0.86 for microalbuminuria; HR, 0.78; 95% CI, 0.63-0.98 for macroalbuminuria; interaction P trend = .71). Treatment with empagliflozin was associated with lower incidence of new macroalbuminuria (HR, 0.81; 95% CI, 0.70-0.94; P = .005) and an increase in rate of remission to sustained normoalbuminuria or microalbuminuria (HR, 1.31; 95% CI, 1.07-1.59; P = .009) but not with a reduction in UACR in the overall population; however, UACR was reduced in patients with diabetes, who had higher UACR levels than patients without diabetes (geometric mean for diabetes at baseline, 0.91; 95% CI, 0.85-0.98 and for no diabetes at baseline, 1.08; 95% CI, 1.01-1.16; interaction P = .008). Conclusions and Relevance In this post hoc analysis of a randomized clinical trial, compared with placebo, empagliflozin was associated with reduced HF hospitalizations or cardiovascular death irrespective of albuminuria levels at baseline, reduced progression to macroalbuminuria, and reversion of macroalbuminuria. Abstract: Let There Be Light Retinitis pigmentosa, a disease that can result from a wide variety of genetic defects, causes degeneration of photoreceptor cells in the retina and leads to blindness. In the course of the disease, it is generally the rod photoreceptor cells that degenerate first. Cone photoreceptor cells may persist, but in a damaged and nonfunctional state. Busskamp et al. (p. 413 , published online 24 June; see the cover; see the Perspective by Cepko ) have now applied a gene therapy approach to mouse models of retinitis pigmentosa. Inducing expression of a bacterial light-activated ion pump, halorho dopsin, in the damaged cone cells improved visual responses in the diseased mouse retinas. Thus, it may be possible to rescue cone photoreceptors therapeutically, even after they have already been damaged.

Abstract: In this paper, we derive a new generalisation of the strong subadditivity of the entropy to the setting of general conditional expectations onto arbitrary finite-dimensional von Neumann algebras. The latter inequality, which we call approximate tensorization of the relative entropy, can be expressed as a lower bound for the sum of relative entropies between a given density and its respective projections onto two intersecting von Neumann algebras in terms of the relative entropy between the same density and its projection onto an algebra in the intersection, up to multiplicative and additive constants. In particular, our inequality reduces to the so-called quasi-factorization of the entropy for commuting algebras, which is a key step in modern proofs of the logarithmic Sobolev inequality for classical lattice spin systems. Abstract: International audience; This paper presents an adaptive impedance control architecture for robotic teleoperation of contact tasks featuring continuous interaction with the environment. We use Learning from Demonstration (LfD) as a framework to learn variable stiffness control policies. Then, the learnt state-varying stiffness is used to command the remote manipulator, so as to adapt its interaction with the environment based on the sensed forces. Our system only relies on the on-board torque sensors of a commercial robotic manipulator and it does not require any additional hardware or user input for the estimation of the required stiffness. We also provide a passivity analysis of our system, where the concept of energy tanks is used to guarantee a stable behavior. Finally, the system is evaluated in a representative teleoperated cutting application. Results show that the proposed variablestiffness approach outperforms two standard constant-stiffness approaches in terms of safety and robot tracking performance.

Abstract: We assess the crucial role of tetrapyrrole flexibility in the CO ligation to distinct Ru‐porphyrins supported on an atomistically well‐defined Ag(111) substrate. Our systematic real‐space visualisation and manipulation experiments with scanning tunnelling microscopy directly probe the ligation, while bond‐resolving atomic force microscopy and X‐ray standing‐wave measurements characterise the geometry, X‐ray and ultraviolet photoelectron spectroscopy the electronic structure, and temperature‐programmed desorption the binding strength. Density‐functional‐theory calculations provide additional insight into the functional interface. We unambiguously demonstrate that the substituents regulate the interfacial conformational adaptability, either promoting or obstructing the uptake of axial CO adducts., The CO ligation to two different ruthenium porphyrins on the Ag(111) surface under vacuum was investigated experimentally and theoretically. The electronic and geometric changes upon ligation as well as the energetics of the process were elucidated. The surface adaptability of the porphyrin macrocycle conformation proves crucial for CO binding.

Abstract: The heterochromatin-enriched HP1 proteins play a critical role in regulation of transcription. These proteins contain two related domains known as the chromo- and the chromoshadow-domain. The chromo-domain binds histone H3 tails methylated on lysine 9. However, in vivo and in vitro experiments have shown that the affinity of HP1 proteins to native methylated chromatin is relatively poor and that the opening of chromatin occurring during DNA replication facilitates their binding to nucleosomes. These observations prompted us to investigate whether HP1 proteins have additional histone binding activities, envisioning also affinity for regions potentially occluded by the nucleosome structure. We find that the chromoshadow-domain interacts with histone H3 in a region located partially inside the nucleosomal barrel at the entry/exit point of the nucleosome. Interestingly, this region is also contacted by the catalytic subunits of the human SWI/SNF complex. In vitro, efficient SWI/SNF remodeling requires this contact and is inhibited in the presence of HP1 proteins. The antagonism between SWI/SNF and HP1 proteins is also observed in vivo on a series of interferon-regulated genes. Finally, we show that SWI/SNF activity favors loading of HP1 proteins to chromatin both in vivo and in vitro. Altogether, our data suggest that HP1 chromoshadow-domains can benefit from the opening of nucleosomal structures to bind chromatin and that HP1 proteins use this property to detect and arrest unwanted chromatin remodeling., Author Summary HP1 proteins are transcriptional regulators frequently associated with gene silencing, a phenomenon involving masking of promoter DNA by dense chromatin. Owing to their chromo-domain, these proteins can read and bind an epigenetic mark that on many non-expressed genes is present on histone H3 at the surface of the nucleosome (the fundamental packing unit of chromatin). However, the binding to this mark does not explain the repressing activity of HP1 proteins. Here, we show that these proteins can establish a second contact with histone H3, independently of the epigenetic mark. This second contact site is located inside the nucleosome, in a position likely to be inaccessible. Interestingly, this site is also contacted by a subunit of the SWI/SNF complex and this contact is required for the ATP-dependent chromatin remodeling catalyzed by SWI/SNF. We provide evidence suggesting that HP1 proteins use the SWI/SNF chromatin remodeling to gain access to the contact site inside the nucleosome and to prevent further remodeling by competing with SWI/SNF for binding at this position. These observations lead us to suggest that HP1 proteins function as gatekeepers on promoters, detecting and stopping unwanted exposure of internal nucleosomal sites.

Abstract: International audience; Experimental validation is presented for a new type of microwave diagnostic, first introduced in the theoretical study in M. Usoltceva et al., Rev. Sci. Instrum. 93, 013,502 (2022). A new term is adopted for this technique to highlight its difference from interferometry: intensity refractometry. The diagnostic allows measuring electron density, and in this work, it is applied at the edge of a tokamak. The implementation of this technique at ASDEX Upgrade, called Microwave Intensity refractometer in the Limiter Shadow (MILS), provides the first experimental proof of the diagnostic concept. Densities predicted by MILS are compared to several other diagnostics. The agreement and discrepancy in various radial regions of the density profile are analyzed and possible reasons are discussed. A wide density coverage is shown in the example discharges with densities from 2 × 10 17 m − 3 to 2 × 10 19 m − 3 at the limiter position. In these experiments, the radial location of the measurements varied from 5 cm in front of the limiter (up to 1 cm inside the separatrix was measured) to 3 cm in the limiter shadow. Experimental challenges of MILS operation and data processing are presented.

Abstract: The original publication is available at www.springerlink.com; International audience; As demands on hospital efficiency increase, there is a stronger need for automatic analysis, recovery, and modification of surgical workflows. Even though most of the previous work has dealt with higher level and hospital-wide workflow including issues like document management, workflow is also an important issue within the surgery room. Its study has a high potential, e.g., for building context-sensitive operating rooms, evaluating and training surgical staff, optimizing surgeries and generating automatic reports. In this paper we propose an approach to segment the surgical work- flow into phases based on temporal synchronization of multidimensional state vectors. Our method is evaluated on the example of laparoscopic cholecystectomy with state vectors representing tool usage during the surgeries. The discriminative power of each instrument in regard to each phase is estimated using AdaBoost. A boosted version of the Dynamic Time Warping (DTW) algorithm is used to create a surgical reference model and to segment a newly observed surgery. Full cross-validation on ten surgeries is performed and the method is compared to standard DTW and to Hidden Markov Models.
