Abstract

Food fraud, even when not in the news, is ubiquitous and demands the development of innovative strategies to combat it. A new non-targeted method (NTM) for distinguishing spelt and wheat is described, which aids in food fraud detection and authenticity testing. A highly resolved fingerprint in the form of spectra is obtained for several cultivars of spelt and wheat using liquid chromatography coupled high-resolution mass spectrometry (LC-HRMS). Convolutional neural network (CNN) models are built using a nested cross validation (NCV) approach by appropriately training them using a calibration set comprising duplicate measurements of eleven cultivars of wheat and spelt, each. The results reveal that the CNNs automatically learn patterns and representations to best discriminate tested samples into spelt or wheat. This is further investigated using an external validation set comprising artificially mixed spectra, samples for processed goods (spelt bread and flour), eleven untypical spelt, and six old wheat cultivars. These cultivars were not part of model building. We introduce a metric called the D score to quantitatively evaluate and compare the classification decisions. Our results demonstrate that NTMs based on NCV and CNNs trained using appropriately chosen spectral data can be reliable enough to be used on a wider range of cultivars and their mixes. Abstract

Campylobacter jejuni is the leading bacterial food-borne pathogen in Europe. Despite the accepted limits of cultural detection of the fastidious bacterium, the “gold standard” in food microbiology is still the determination of colony-forming units (CFU). As an alternative, a live/dead differentiating qPCR has been established, using propidium monoazide (PMA) as DNA-intercalating crosslink agent for inactivating DNA from dead, membrane-compromised cells. The PMA treatment was combined with the addition of an internal sample process control (ISPC), i.e. a known number of dead C. sputorum cells to the samples. The ISPC enables i), monitoring the effective reduction of dead cell signal by the light-activated DNA-intercalating dye PMA, and ii), compensation for potential DNA losses during processing. Here, we optimized the method for routine application and performed a full validation of the method according to ISO 16140-2:2016(E) for the quantification of live thermophilic Campylobacter spp. in meat rinses against the classical enumeration method ISO 10272-2:2017. In order to render the method applicable and cost-effective for practical application, the ISPC was lyophilized to be distributable to routine laboratories. In addition, a triplex qPCR was established to simultaneously quantify thermophilic Campylobacter, the ISPC and an internal amplification control (IAC). Its performance was statistically similar to the two duplex qPCRs up to a contamination level of 4.7 log10 Campylobacter per ml of meat rinse. The limit of quantification (LOQ) of the alternative method was around 20 genomic equivalents per PCR reaction, i.e. 2.3 log10 live Campylobacter per ml of sample. The alternative method passed a relative trueness study, confirming the robustness against different meat rinses, and displayed sufficient accuracy within the limits set in ISO 16140-2:2016(E). Dies ist ein Beitrag für das 15. BAM-PTB-Kolloquium zur chemischen und physikalischen Sicherheitstechnik am 21. und 22. Mai 2019 bei der PTB in Braunschweig., Prävention beginnt im Prüflabor, wenn Chemikalien auf ihre gefährlichen Eigenschaften getestet werden. Dazu sind Prüfmethoden entwickelt und veröffentlicht worden, die heute weltweit Anwendung finden. Auf die Validität der Prüfmethode und richtige Durchführung der Prüfung im Laboratorium müssen sich Sicherheitsfachkräfte, Transporteure oder Händler verlassen können. Anhand der in den letzten 10 Jahren im Rahmen des CEQAT-DGHS von BAM und PTB durchgeführten Ringversuche (RV) wird gezeigt, dass bei allen bisher untersuchten Prüfmethoden ein Verbesserungsbedarf besteht. Die RV müssen daher zunächst auf die Methodenverbesserung und -validierung abzielen und nicht auf Leistungstests. Das Labormanagement und die praktische Durchführung der Prüfung sind in vielen Laboratorien verbesserungsbedürftig. Der Begriff „Erfahrung der Prüfer“ ist kritisch zu sehen: Eine „lange Erfahrung mit vielen Prüfungen“ ist nicht unbedingt ein Garant für richtige Ergebnisse. Bei der Prüfung der gefährlichen Eigenschaften von Chemikalien sind Referenzmaterialen auf Grund der Instabilität in der Regel nicht verfügbar. Für Prüflaboratorien sind RV daher eine Alternative bei der Qualitätssicherung. RV sind jedoch aufwendig und können nur in relativ großen Zeitabständen durchgeführt werden. Es sind deshalb Verfahren zur Verifizierung z. B. der in den Laboratorien verwendeten Prüfapparaturen zu entwickeln. Die Entwicklung von Verifizierungsverfahren wird am Bespiel der Prüfmethode UN Test N.5 demonstriert.

89. Diagnostic relevance of a novel multiplex immunoassay panel in breast cancer

Abstract

Multiple factors contribute to the development and progression of breast cancer. Markers of tumor growth and invasion, cell death, immune activation, and angiogenesis can be assessed in parallel by a novel multiplex immunoassay panel. The diagnostic performance of a multiplex cancer biomarker magnetic bead panel comprising 24 tumor associated parameters was evaluated in sera of 154 women including 77 patients with breast cancer, 10 with precancerous lesions, 31 with benign breast diseases, and 36 healthy controls. Marker levels were log-transformed for variance stabilization. Significance testing was done using t-test or Wilcoxon rank-sum test with correction of p values for multiple testing. Furthermore, receiver operating characteristic analyses were performed. Serum levels of several biomarkers were significantly (p0.001) higher in cancer patients than in healthy controls, particularly alpha-fetoprotein, cancer antigen 15-3, cancer antigen 19-9, migration inhibitory factor, carcinoembryonic antigen, cancer antigen 125, hepatocyte growth factor, soluble Fas, tumor necrosis factor-, stem cell factor, and osteopontin. As most markers were also elevated in benign breast diseases, only cancer antigen 15-3 showed significant differences to cancer patients (p0.001). The resulting areas under the curve in receiver operating characteristic curves for discrimination between benign and malignant breast diseases achieved 0.71 with a sensitivity of 33.8% at 95% specificity. Multiplexing enables parallel analysis of different biomarker classes for cancer detection. Established cancer antigen 15-3 proved to be most relevant for differential diagnosis.

Abstract

The article focuses on the Sun Protection Facilitator (SPF) determination began more than 50 years ago as an outdoor method, using erythema as the biological endpoint. It mentions the in vitro transmission approach was the first attempt to find an alternative to the in vivo method; and also mentions that under the leadership of Cosmetics Europe (CE), a comparison of in vivo versus in vitro SPF test methods was conducted using 24 commercial sunscreens.

Abstract

Various standardised test methods are used by laboratories to determine the hazardous properties and safety characteristics of chemical substances and mixtures, e.g. test methods published by the European Union (2008) and by the United Nations (2015). The test results are the basis (amongst others) for the correct classification and labelling of chemicals as hazardous according to GHS/CLP or as dangerous goods according to Transport Regulations. The REACH Regulation obliges all manufacturers and importers of chemicals to identify and manage risks associated with the substances they manufacture and market. Via the eChemPortal (www.echemportal.org) many data from the REACH registration dossiers are available. However, the quality and correctness of the information remains in the responsibility of the data submitter. Unfortunately, the quality of information on physicochemical properties is not always adequate, so the quality and adequacy of the data submitted shall be improved. This paper focusses on the quality of test results obtained by laboratories and the validation of the test methods used e.g. for classification of dangerous goods and indicates recent developments as well as steps to be taken for further improvement. In particular, the importance of interlaboratory tests and the need to validate test methods are presented in this publication. Interlaboratory tests play a decisive role in assessing the reliability of laboratory test results. Participation in interlaboratory tests is not only a crucial element of the quality assurance of laboratories; as such it is explicitly recommended in ISO/IEC 17025, ISO (2017). In addition, interlaboratory tests are also used to improve and validate test methods and can be used for the determination of the measurement uncertainty, Hässelbarth (2004), ISO (2004). This paper specially addresses interlaboratory tests for the validation of test methods performed by CEQATDGHS (Centre for quality assurance for testing of dangerous goods and hazardous substances, www.ceqatdghs. bam.de) and outlines typical results and general conclusions and steps to be taken to guarantee that laboratory test results are fit for the purpose and of high quality.
