Polo-like kinases (Plks) represent a family of highly conserved serine–threonine kinases that play essential roles in cell cycle progression. Plk3 plays contradictory roles in different cancers. However, the roles of Plk3 in PCa remain largely unexplored. Methods: Kaplan–Meier analysis and Cox regression analysis were performed to evaluate the relationship between Plk3 and prognosis of patients with PCa. Gene set enrichment analysis (GSEA) was conducted to evaluate proliferation and metastasis gene sets using The Cancer Genome Atlas Dataset. MTS assay, clone formation assay, cell migration, and wound healing assay were carried out to investigate biological functions of Plk3. Results: We found that high Plk3 expression was closely correlated with poor prognosis. GSEA revealed that Plk3 was involved in proliferation and metastasis. Loss-of-function assays demonstrated that Plk3 promoted proliferation and metastasis in PCa cells in vitro. Conclusion: We discovered that Plk3 plays a critical role in PCa, indicating that it may be a potential prognostic marker and help predict the progression, especially recurrence of PCa. 87. Photochemical graft of γ-cyclodextrin’s interior leading to in-situcharge-transfer complexes with unusual regioselectivity and its application in 3D photo-printing

Abstract

Photoirradiation of anthraquinone (AQ) derivatives with γ-cyclodextrin (γ-CD) in aqueous solution led to a γ-CD derivative with a 9,10-dihydroxylanthracene grafting unprecedentedly at the 5′-position of one glucoside of γ-CD in high efficiency. Meanwhile, another AQ enters into the remaining space of the 9,10-dihydroxylanthracene-occupied γ-CD cavity to in-situform a self-included quinhydrone charge transfer (CT) complex. The structure of the supramolecular CT complex was unambiguously demonstrated by 1D and 2D NMR (1H NMR, 13C NMR, DEPT, 1H-1H COSY, HSQC, HMBC, TOCSY, and ROESY) spectroscopies, which revealed a highly stereoselective stacking of the quinhydrone pairs in the γ-CD cavity. The supramolecular CT complex showed an unusually high binding affinity and improved oxidation resistance Polyacrylamides bearing γ-CD or anthraquinone were respectively synthesized by co-polymerization of acrylamide with the corresponding monomer. Photo-irradiation of aqueous mixtures containing these two types of polymers led to gelation, which was applied to 3D photo-printing of soft materials.

88. Supramolecular Enantiodifferentiating Photocyclodimerization of 2‐Anthracenecarboxylic Acid Mediated by Bridged β‐Cyclodextrins: Critical Effects of the Host Structure, pH and Co‐Solvents.

Abstract

Several sulfoxide‐ and sulfone‐bridged β‐cyclodextrin (CD) dimers were synthesized for mediating the enantiodifferentiating [4+4] photocyclodimerization of 2‐anthracenecarboxylic acid (AC). The complexation behavior of these chiral hosts with AC was investigated by UV‐vis, circular dichroism, fluorescence, and NMR spectroscopies and certified the formation of 1 : 1 and 1 : 2 host‐guest complexes. The product distribution and enantioselectivity of the photoreaction turned out to be a critical function of the chemical structure of bridged CDs. Comparing to the sulfur‐bridged 2AX−3GX β‐CD dimer 7, the conversion of the photolyzes with sulfoxide‐bridged was significantly improved, and the ee of cyclodimer 2 was remarkably increased from −82.8% with 7 to −96.7% with the sulfoxide‐bridged 2AX−3GX β‐CD dimer 8. The relative yields and ee values of the slipped cyclodimers 5 and 6 were greatly enhanced in the presence of 6 M CsCl. The reaction selectivity is susceptible to the pH variation of the aqueous buffer solution, demonstrating that the supramolecular photochirogenesis is controlled by multidimensional factors, including the chemical structure of the chiral host, solvent, and pH conditions.
